Treatment of bone defects resulting after tumor surgeries, accidents, or non-unions is an actual problem linked to morbidity and the necessity of a second surgery and often requires a critical healthcare cost. Although the surgical technique has changed in a modern way, the treatment outcome is still influenced by patient age, localization of the bone defect, associated comorbidities, the surgeon approach, and systemic disorders. Three-dimensional magnesium-based scaffolds are considered an important step because they can have precise bone defect geometry, high porosity grade, anatomical pore shape, and mechanical properties close to the human bone. In addition, magnesium has been proven in in vitro and in vivo studies to influence bone regeneration and new blood vessel formation positively. In this review paper, we describe the magnesium alloy's effect on bone regenerative processes, starting with a short description of magnesium's role in the bone healing process, host immune response modulation, and finishing with the primary biological mechanism of magnesium ions in angiogenesis and osteogenesis by presenting a detailed analysis based on a literature review. A strategy that must be followed when a patient-adapted scaffold dedicated to bone tissue engineering is proposed and the main fabrication technologies are combined, in some cases with artificial intelligence for Mg alloy scaffolds, are presented with examples. We emphasized the microstructure, mechanical properties, corrosion behavior, and biocompatibility of each study and made a basis for the researchers who want to start to apply the regenerative potential of magnesium-based scaffolds in clinical practice. Extrusion-based 4D-printing, which is an emerging field within additive manufacturing, has enabled the technical transfer of bioinspired self-shaping mechanisms by emulating the functional morphology of motile plant structures (e.g., leaves, petals, capsules). However, restricted by the layer-by-layer extrusion process, much of the resulting works are simplified abstractions of the pinecone scale's bilayer structure. This paper presents a new method of 4D-printing by rotating the printed axis of the bilayers, which enables the design and fabrication of self-shaping monomaterial systems in cross sections. This research introduces a computational workflow for programming, simulating, and 4D-printing differentiated cross sections with multilayered mechanical properties. Taking inspiration from the large-flowered butterwort (Pinguicula grandiflora), which shows the formation of depressions on its trap leaves upon contact with prey, we investigate the depression formation of bioinspired 4D-printed test structures by varying each depth layer. Cross-sectional 4D-printing expands the design space of bioinspired bilayer mechanisms beyond the XY plane, allows more control in tuning their self-shaping properties, and paves the way toward large-scale 4D-printed structures with high-resolution programmability. In an increasing effort to address the environmental challenges caused by the currently linear economic paradigm of "produce, use, and discard", the construction industry has been shifting towards a more circular model. A circular economy requires closing of the loops, where the end-of-life of a building is considered more carefully, and waste is used as a resource. In comparison to traditional building materials such as timber, steel and concrete, mycelium-based materials are renewable alternatives that use organic agricultural and industrial waste as a key ingredient for production, and do not rely on mass extraction or exploitation of valuable finite or non-finite resources. Mycelium-based materials have shown their potential as a more circular and economically competitive alternative to conventional synthetic materials in numerous industries ranging from packaging, electronic prototyping, furniture, fashion to architecture. However, application of mycelium-based materials in the construction industry has been limited to small-scale prototypes and architectural installations due to low mechanical properties, lack of standardisation in production methods and material characterisation. This paper aims to review the current state of the art in research and applications of mycelium-based materials across disciplines, with a particular focus on digital methods of fabrication, production, and design. The information gathered from this review will be synthesised to identify key challenges in scaling up applications of mycelium-based materials as load-bearing structural elements in architecture and suggest opportunities and directions for future research. The complex structure, chemical composition, and biomechanical properties of craniofacial cartilaginous structures make them challenging to reconstruct. Autologous grafts have limited tissue availability and can cause significant donor-site morbidity, homologous grafts often require immunosuppression, and alloplastic grafts may have high rates of infection or displacement. Furthermore, all these grafting techniques require a high level of surgical skill to ensure that the reconstruction matches the original structure. Current research indicates that additive manufacturing shows promise in overcoming these limitations. Autologous stem cells have been developed into cartilage when exposed to the appropriate growth factors and culture conditions, such as mechanical stress and oxygen deprivation. Additive manufacturing allows for increased precision when engineering scaffolds for stem cell cultures. Fine control over the porosity and structure of a material ensures adequate cell adhesion and fit between the graft and the defect. Several recent tissue engineering studies have focused on the trachea, nose, and ear, as these structures are often damaged by congenital conditions, trauma, and malignancy. Triply periodic minimal surfaces (TPMSs) are found in many natural objects including butterfly wings, sea urchins, and biological membranes. They simultaneously have zero mean curvature at every point and a crystallographic group symmetry. A metamaterial can be created from such periodic surfaces or used as a reinforcement of a composite material. While a TPMS as a mathematical object has been known since 1865, only novel additive manufacturing (AM) technology made it possible to fabricate cellular materials with complex TPMS shapes. Cellular TPMS-based metamaterials have remarkable properties related to wetting/liquid penetration, shock absorption, and the absence of stress concentrators. Recent studies showed that TPMSs are also found in natural crystals when electron surfaces are considered. Artificial crystal-inspired metamaterials mimic such crystals including zeolites and schwarzites. These metamaterials are used for shock, acoustic waves, and vibration absorption, and as structural materials, heat exchangers, and for other applications. The choice of the crystalline cell of a material, as well as its microstructure, plays a decisive role in its properties. The new area of crystal-inspired materials has many common features with traditional biomimetics with models being borrowed from nature and adjusted for engineering applications. The demand for building materials has been constantly increasing, which leads to excessive energy consumption for their provision. The looming environmental consequences have triggered the search for sustainable alternatives. Mycelium, as a rapidly renewable, low-carbon natural material that can withstand compressive forces and has inherent acoustic and fire-resistance properties, could be a potential solution to this problem. However, due to its low tensile, flexural and shear strength, mycelium is not currently widely used commercially in the construction industry. Therefore, this research focuses on improving the structural performance of mycelium composites for interior use through custom robotic additive manufacturing processes that integrate continuous wood fibers into the mycelial matrix as reinforcement. This creates a novel, 100% bio-based, wood-veneer-reinforced mycelium composite. As base materials, Ganoderma lucidum and hemp hurds for mycelium growth and maple veneer for reinforcement were pre-selected for this study. Compression, pull-out, and three-point bending tests comparing the unreinforced samples to the veneer-reinforced samples were performed, revealing improvements on the bending resistance of the reinforced samples. Additionally, the tensile strength of the reinforcement joints was examined and proved to be stronger than the material itself. The paper presents preliminary experiment results showing the effect of veneer reinforcements on increasing bending resistance, discusses the potential benefits of combining wood veneer and mycelium's distinct material properties, and highlights methods for the design and production of architectural components. We overview recent findings achieved in the field of model-driven development of additively manufactured porous materials for the development of a new generation of bioactive implants for orthopedic applications. Porous structures produced from biocompatible titanium alloys using selective laser melting can present a promising material to design scaffolds with regulated mechanical properties and with the capacity to be loaded with pharmaceutical products. Adjusting pore geometry, one could control elastic modulus and strength/fatigue properties of the engineered structures to be compatible with bone tissues, thus preventing the stress shield effect when replacing a diseased bone fragment. Adsorption of medicals by internal spaces would make it possible to emit the antibiotic and anti-tumor agents into surrounding tissues. The developed internal porosity and surface roughness can provide the desired vascularization and osteointegration. We critically analyze the recent advances in the field featuring model design approaches, virtual testing of the designed structures, capabilities of additive printing of porous structures, biomedical issues of the engineered scaffolds, and so on. Nature provides a considerable number of good examples for simple and very efficient joint assemblies. One example is the enormously flexible cervical spine of American barn owls, which consists of 14 cervical vertebrae. Each pair of vertebrae produces a comparatively small individual movement in order to provide a large overall movement of the entire cervical spine. The biomimetic replication of such joints is difficult due to the delicate and geometric unrestricted joint shapes as well as the muscles that have to be mimicked. Using X-ray as well as micro-computed tomography images and with the utilisation of additive manufacturing, it was possible to produce the owl neck vertebrae in scaled-up form, to analyse them and then to transfer them into technically usable joint assemblies. The muscle substitution of these joints was realised by smart materials actuators in the form of shape memory alloy wire actuators. This actuator technology is outstanding for its muscle-like movement and for its high-energy density. The disadvantage of this wire actuator technology is the low rate of contraction, which means that a large length of wire has to be installed to generate adequate movement. For this reason, the actuator wires were integrated into additively manufactured carrier components to mimic biological joints. This resulted in joint designs that compensate for the disadvantages of the small contraction of the actuators by intelligently installing large wire lengths on comparatively small installation spaces, while also providing a sufficient force output. Whether it is a plant- or animal-based bio-inspiration design, it has always been able to address one or more product/component optimisation issues. Today's scientists or engineers look to nature for an optimal, economically viable, long-term solution. Similarly, a proposal is made in this current work to use seven different bio-inspired structures for automotive impact resistance. All seven of these structures are derived from plant and animal species and are intended to be tested for compressive loading to achieve load-bearing capacity. The work may even cater to optimisation techniques to solve the real-time problem using algorithm-based generative shape designs built using CATIA V6 in unit dimension. The samples were optimised with Rhino 7 software and then simulated with ANSYS workbench. To carry out the comparative study, an experimental work of bioprinting in fused deposition modelling (3D printing) was carried out. The goal is to compare the results across all formats and choose the best-performing concept. The results were obtained for compressive load, flexural load, and fatigue load conditions, particularly the number of life cycles, safety factor, damage tolerance, and bi-axiality indicator. When compared to previous research, the results are in good agreement. Because of their multifunctional properties combining soft and high stiffness and lightweight properties of novel materials, novel materials have many potential applications in the medical, aerospace, and automotive sectors. The industrial revolution 4.0 has led to a burst in the development of robotic automation and platforms to increase productivity in the industrial and health domains. Hence, there is a necessity for the design and production of smart and multi-functional tools, which combine several cutting-edge technologies, including additive manufacturing and smart control systems. In the current article, a novel multi-functional biomimetic soft actuator with a pneumatic motion system was designed and fabricated by combining different additive manufacturing techniques. The developed actuator was bioinspired by the natural kinematics, namely the motion mechanism of worms, and was designed to imitate the movement of a human finger. Furthermore, due to its modular design and the ability to adapt the actuator's external covers depending on the requested task, this actuator is suitable for a wide range of applications, from soft (i.e., fruit grasping) or industrial grippers to medical exoskeletons for patients with mobility difficulties and neurological disorders. In detail, the motion system operates with two pneumatic chambers bonded to each other and fabricated from silicone rubber compounds molded with additively manufactured dies made of polymers. Moreover, the pneumatic system offers multiple-degrees-of-freedom motion and it is capable of bending in the range of −180° to 180°. The overall pneumatic system is protected by external covers made of 3D printed components whose material could be changed from rigid polymer for industrial applications to thermoplastic elastomer for complete soft robotic applications. In addition, these 3D printed parts control the angular range of the actuator in order to avoid the reaching of extreme configurations. Finally, the bio-robotic actuator is electronically controlled by PID controllers and its real-time position is monitored by a one-axis soft flex sensor which is embedded in the actuator's configuration.
