In the present study, we synthesized a novel near-infrared turn-on BODIPY probe and a new norbornene-modified glucosamine derivative. The probe exhibits a significant NIR fluorescence emission with a turn-on response and can perform tumour-specific imaging in tumour-bearing mice. The non-natural glucosamine provides metabolic glycoengineering labelling. It can be expressed on cells as chemical tags and further reacted with fluorescence dyes for cell labelling. We developed a dual-responsive fluorescent probe MC-V-P for the simultaneous detection of ONOO− and viscosity by different imaging channels. MC-V-P has high sensitivity and selectivity, and shows good stability at different pH levels. Notably, the probe has two independent fluorescence signals toward ONOO− and viscosity changes at 580 nm and 740 nm, respectively. A series of naphthalimide – tetrazines were developed as bioorthogonal fluorogenic probes, which could produce significant fluorescence enhancement, notable aggregation-induced emission (AIE) characters and multicolor emissions after bioorthogonal reaction with strained dienophiles. Manipulating the π-bridge in the fluorophore skeleton allows fine-tuning of the emission wavelength and influences the AIE-active properties. In this work, by installing a free-rotating benzene ring to suppress the intermolecular π–π stacking effect in the aggregated state, a naphthalimide-derived hypochlorite (ClO−) fluorescent probe, Probe A, with the aggregation-caused quenching (ACQ) effect was successfully transformed into Probe B possessing typical aggregation-induced emission (AIE) characteristics. The experimental results indicated that Probe B with good selectivity and a low detection limit (LOD) of 0.02 µM can also exhibit a significant ratiometric fluorescence color change from cyan to dark blue within 2 min in a nearly pure water solvent system after the addition of ClO−. Transferrin-modified AuNCs (Tf-AuNCs) with two photon-near infrared (TP-NIR) fluorescence were prepared. For the first time, a novel nanoprobe platform, Tf-AuNCs@MnO2, was developed for the TP-NIR fluorescence imaging and magnetic resonance imaging of living cells and tissues. A new fluorescent probe (Q-lipo) was developed by conjugating a xanthene scaffold with a quinoline moiety for activatable imaging of lipophagy. Q-lipo with acidic pH activated near infrared fluorescence and the lipid droplet targeting ability allowed activatable fluorescence imaging and flow cytometry detection of lipophagy in live cells with high contrast. We developed a new method for the de novo formation of fluorophores based on citrate (DNFC) in biological samples. Use of an amide coupling reagent and microwave irradiation greatly facilitates the fluorophore formation on peptides and proteins with N-terminal cysteine or serine. Since N-terminal cysteine and serine can form thiazolopyridone- or oxazolopyridone-based fluorophores emitting blue and green fluorescence, respectively, by the DNFC staining, each organelle, cell and tissue exhibited a characteristic fluorescence distribution. A small series of fluorescent lysosome-targeting probes based on the BODIPY fluorophore and containing morpholine and nitro groups were rationally designed. These probes emitted light from green to NIR wavelengths, and provided specificity for imaging the lysosomes of hypoxic cells. We report the first dual-responsive 19F MRI and fluorescence imaging probe for cellular hypoxia. The Cu2+-based probe exhibits no 19F MR signal and reduced fluorescence signal due to paramagnetic quenching; however, the probe turns-on in both modes following reduction to Cu+. Small fluorescent molecules have been an important tool for fluorescence-based imaging, thanks to their technical simplicity, sensitivity and structural flexibility. However, probing specific intracellular systems with a small fluorescent molecule is not an easy task owing to the intricate nature of the cells. Ideal imaging probes should be highly permeable and be without background noise to allow undisrupted observations. These probes will unquestionably be more advantageous for interrogating intracellular architectures and dynamics without compromising the cellular integrity, and could therefore be applied for applications in the native cellular environment. This review will highlight the advances in design strategies for cell-permeable fluorescent probes through a diversity-oriented fluorescence library approach and rational design via computational-aided predictive models. The first hyperbranched BODIPY-based conjugated polymer dots (Pdots) were reported. The Pdots showed quantum yields of as high as 22%, which is 40% higher than their linear counterparts. We report the design and synthesis of a photoactivatable azido-acyl oxazine fluorophore. Photoactivation is achieved cleanly and rapidly with UV light, producing a single fluorescent oxazine photoproduct. A novel terpyridine derivative formed stable aggregates in aqueous media (DMSO/H2O = 1/99) with dramatically enhanced fluorescence compared to its organic solution. Moreover, the ultra-violet absorption spectra also demonstrated specific responses to the incorporation of water. The yellow emission at 557 nm changed to a solution with intense greenish luminescence only in the presence of protons and it conformed to a molecular logic gate with a two-input INHIBIT function. A novel “turn-on” fluorescent probe HP for hypoxia imaging was designed and synthesized based on rhodamine B and a naphthalimide fluorophore. The fluorescence of HP is very weak owing to the FRET effect from rhodamine B to the azo-naphthalimide unit. Under hypoxia conditions, the azo-bond is reduced and the fluorescence at 581 nm enhances dramatically as a result of disintegration of the quencher structure. Verified by the cyclic voltammetry reduction potential and proposed product HPN, the probe HP could undergo the chemical and cytochrome P450 enzymatic reduction quickly. When cultured with HeLa cells, HP showed remarkable fluorescence differences at various oxygen concentrations, and the ratio of fluorescence intensity between hypoxic and normoxic cells could reach 9 fold. The design, synthesis and in vitro assessment of a bifunctional imaging probe for dual fluorine (19F) magnetic resonance spectroscopy (19F-MRS) and fluorescence detection is reported. Naphthalene–salisaldehyde conjugate (NAPSAL) is established as a novel arsenate (H2AsO4−) selective ‘turn-on’ fluorescence probe.
