Abstract

A labeling system for food allergenic ingredients was established in Japan in April 2002. To monitor the labeling, the Japanese government announced official methods for detecting allergens in processed foods in November 2002. The official methods consist of quantitative screening tests using enzyme-linked immunosorbent assays (ELISAs) and qualitative confirmation tests using Western blotting or polymerase chain reactions (PCR). In addition, the Japanese government designated 10 μg protein/g food (the corresponding allergenic ingredient soluble protein weight/food weight), determined by ELISA, as the labeling threshold. To standardize the official methods, the criteria for the validation protocol were described in the official guidelines. This paper, which was presented at the Advances in Food Allergen Detection Symposium, ACS National Meeting and Expo, San Diego, CA, Spring 2012, describes the validation protocol outlined in the official Japanese guidelines, the results of interlaboratory studies for the quantitative detection method (ELISA for crustacean proteins) and the qualitative detection method (PCR for shrimp and crab DNAs), and the reliability of the detection methods.

Abstract

Maillard-type glycosylation was applied to preparation of hypoallergenic agents from a major buckwheat allergen, Fag e 1. Conjugation with arabinogalactan (AG), xyloglucan (XG), or yeast glucomannan (YGM) successfully decreased in vitro allergenicity of Fag e 1. Determination of IgE titer in the tested allergic mice revealed that YGM was the most effective for in vivo allergenicity of Fag e 1 among these water-soluble polysaccharides. Real-time PCR analysis using a set of primer for IL-4 (a typical Th2 cytokine) or IFN-gamma (a typical Th1 cytokine) showed that expressed mRNA for IL-4 in splenocytes drastically decreased with increasing with Fag e 1-YGM conjugate feeding. In addition, based on a flow-cytometric analysis of T cell subsets in the splenocytes, it was confirmed that the feeding led to an improvement of Th1/Th2 balance in the allergic mice where population of Th1 increased from 2.91% to 4.02%, while that of Th2 decreased from 3.75% to 2.72%. Furthermore, it was revealed that differentiation ratio of regulatory T cell (Treg) in the splenocytes increased from 14.5% to 18.7% by the oral administration. These results indicated that Fag e 1-YGM conjugate can be available for an immunomodulating agent for buckwheat allergy.

Abstract

We developed a novel type of real-time polymerase chain reaction (PCR) array with TaqMan chemistry as a platform for the comprehensive and semiquantitative detection of genetically modified (GM) crops. Thirty primer-probe sets for the specific detection of GM lines, recombinant DNA (r-DNA) segments, endogenous reference genes, and donor organisms were synthesized, and a 96-well PCR plate was prepared with a different primer-probe in each well as the real-time PCR array. The specificity and sensitivity of the array were evaluated. A comparative analysis with the data and publicly available information on GM crops approved in Japan allowed us to assume the possibility of unapproved GM crop contamination. Furthermore, we designed a Microsoft Excel spreadsheet application, Unapproved GMO Checker version 2.01, which helps process all the data of real-time PCR arrays for the easy assumption of unapproved GM crop contamination.
