Paper has been widely employed as cheap material for the development of a great number of sensors such as pregnancy tests, strips to measure blood sugar, and COVID-19 rapid tests. The need for new low-cost analytical devices is growing, and consequently the use of these platforms will be extended to different assays, both for the final consumer and within laboratories. This work describes a paper-based electrochemical sensing platform that uses a paper disc conveniently modified with recognition molecules and a screen-printed carbon electrode (SPCE) to achieve the detection of gluten in a deep eutectic solvent (DES). This is the first method coupling a paper biosensor based on aptamers and antibodies with the DES ethaline. Ethaline proved to be an excellent extraction medium allowing the determination of very low gluten concentrations. The biosensor is appropriate for the determination of gluten with a limit of detection (LOD) of 0.2 mg L−1 of sample; it can detect gluten extracted in DES with a dynamic range between 0.2 and 20 mg L−1 and an intra-assay coefficient of 10.69%. This approach can be of great interest for highly gluten-sensitive people, who suffer from ingestion of gluten quantities well below the legal limit, which is 20 parts per million in foods labeled gluten-free and for which highly sensitive devices are essential. Urea is one of the most widely used nitrogen fertilizers. However, it is lost to the environment via processes such as denitrification, surface runoff, volatilization, and leaching. In this paper, a novel material is reported on, with low production cost and avoiding the use of harmful solvents, with a pastille morphology developed by a simple method from a mixture of wheat gluten and urea, with potential use as a prolonged-release system of urea (PRSU). The PRSU obtained was characterized by scanning electron microscopy, kinetics of water absorption, equilibrium water content (EWC), Fourier-transform infrared (FT-IR) spectroscopy analysis, and release kinetics. The PRSU diameter was 2.46 cm, and its thickness was 0.17 cm. The PRSU showed physical and structural characteristics such as micropores and hollow fractions in its structure. In addition, the wheat gluten pastille is classified as a swelling material and demonstrated an EWC of 58.47 ± 1.50%. FT-IR analysis of the samples showed hydrogen-bond interactions between the amino and carbonyl groups in the urea and the wheat gluten proteins. Laboratory tests showed that the system can release 97% of the urea within 8–10 h. These results showed that the PRSU presents suitable characteristics for its application as a fertilization alternative for carrying out better agronomic practices. [ABSTRACT FROM AUTHOR]

Celiac disease is caused by an abnormal intestinal T cell response to cereal gluten proteins. The disease has a strong human leukocyte antigen (HLA) association, and CD4+ T cells recognizing gluten epitopes presented by disease-associated HLA-DQ allotypes are considered to be drivers of the disease. The availability of analytical methods to detect and determine levels of markers of priority allergens in foods is of the utmost importance to support standard setting initiatives, the development of compliance and enforcement activities, as well as to provide guidance to industry on implementation of quality control practices, ensuring the effectiveness of allergen-related sanitation techniques. This paper describes the development and implementation of a mass-spectrometry-based technique to determine markers for individual sources of gluten in beer products. This methodology was shown to answer the requirements of Health Canada’s proposed labeling standard for individual gluten source declaration, in order to achieve its policy objectives (i.e., protection of sensitive consumers, while promoting choice). Minimal sample work-up was required and the results obtained by ELISA were further complemented using the LC-MS/MS method. Celiac disease is a chronic inflammatory condition of the small intestine caused by aberrant adaptive immune response to gluten protein from wheat and related cereal plants. Over 90% of celiac disease patients carry the HLA-DQ2.5 allotype and HLA-DQ2.5 presents gluten peptides to gluten-reactive CD4+ T cells in celiac disease patients. A large number of HLA-DQ2.5-restricted gluten T cell epitopes have been identified over the years. These epitopes are in general proline-rich and contain at least one glutamic acid residue that is generated from glutamine in the native gluten protein by deamidation. The deamidation is mediated by the enzyme transglutaminase 2 (TG2). It has been shown that the same T cell could recognize several different HLA-DQ2.5-restricted gluten T cell epitopes due to sequence similarities. In this paper, we demonstrate that three T cell clones derived from duodenal biopsies of different celiac disease patients are able to respond to at least five different gluten T cell epitopes within the DQ2.5-glia-γ4 epitope family, including two novel epitopes. Retrogradation is the process of starch recrystallization, and it profoundly affects the quality, acceptability and shelf-life of starch-containing foods. The influence of glutelin on the wheat starch retrogradation was studied in this paper. Glutenin was isolated from wheat flour, and its effect on retrogradation of wheat starch, amylose and amylopectin was investigated with UV-Vis (starch-iodine), IR and C NMR. The results showed that glutenin probably interacted with amylose during gelation and retrogradation of starch. The results of IR showed that the addition of glutenin to wheat starch reduced the number of hydrogen bonds formed between amyloses during retrogradation. The C NMR results suggested that tyrosine (Tyr) of glutenin might combine with amylose at the first carbon atom when they were mixed homogeneously and such combination was strengthened during retrogradation. Glutenin and amylose formed double helix with each other completely and hindered amylose-amylose short-term retrogradation of wheat starch, and glutenin would no longer inhibit the retrogradation of starch when all of the Tyr formed hydrogen bond with amylose. During wheat digestion, gluten-derived proteolytic resistant peptides are generated, some of them involved in celiac disease. In vitro digestion models able to mimic the peptides generated in the human gastrointestinal tract are extremely useful to assess the pathogenicity of wheat-derived products. In this paper, samples belonging to three different durum wheat varieties were taken at six different steps of the pasta production chain and two different digestion models present in the literature were assessed on the different samples: a more complex one using artificial fluids simulating the exact composition of digestive juices, and a simplified method based on a peptic-tryptic/chymotryptic treatment of wheat ethanolic extract. An extensive characterization of the peptides generated using two in vitro digestion models was performed through LC-MS/MS techniques and the two methods were compared in order to evaluate qualitative and quantitative differences and their possible implications for varietal screening. Strong differences in the type of peptides produced with the two methods were detected, indicating that the simplified method can still be used for a varietal screening but is not representative of the peptides really generated after physiological human digestion. This paper reports the glutenin diversity of a collection of 23 wild populations of the grass Brachypodium distachyon collected in the Mediterranean and southern areas of the Iberian Peninsula. The plant material studied included the three different cytotypes of this species: 2 n = 10, 2 n = 20 and 2 n = 30. A specific method of extraction was used to isolate the glutenin subunits from the caryopsis. Separation by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) showed them to correspond to wheat low-molecular-weight glutenin subunits (LMW-GS). Twenty-two LMW-GS-like were identified that showed great diversity within and between populations. All the populations investigated were polymorphic for the endosperm proteins studied. The 2 n = 30 forms had the largest number of subunits; these were also more diverse than those of the 2 n = 10 or 2 n = 20 forms. The 2 n = 10 forms, the most common in the higher, interior areas of the Iberian Peninsula, showed the smallest subunit variation. In fact, negative correlations were found between subunit diversity and altitude and longitude. In contrast, a positive correlation was detected with the annual average temperature and the indices of thermicity and Mediterraneity. The similarity between the populations was estimated using the Sorensen-Dice coefficient, calculated on the basis of the presence/absence of the 22 LMW-GS proteins. The similarity indices were used to produce a dendrogram using the unweighted pair-group method with arithmetic means (UPGMA). This method produced three main groups corresponding to the three cytotypes. An analysis was made of the possible correlation between eco-geographic and climatic factors and the gene diversity and polymorphism shown in the populations. The diversity correlated positively with the number of chromosomes (2 n), annual mean temperature, index of thermicity and index of Mediterraneity. In contrast, diversity correlated negatively with altitude, longitude and index of precipitation during the summer. The number of chromosomes correlated negatively with altitude, longitude and precipitation during summer and positively with all the other climatic indices. The celiac disease (CD) is an inflammatory condition characterized by injury to the lining of the small-intestine on exposure to the gluten of wheat, barley and rye. The involvement of gluten in the CD syndrome has been studied in detail in bread wheat, where a set of “toxic” and “immunogenic” peptides has been defined. For wheat diploid species, information on CD epitopes is poor. In the present paper, we have adopted a genomic approach in order to understand the potential CD danger represented by storage proteins in diploid wheat and sequenced a sufficiently large number of cDNA clones related to storage protein genes of Triticum monococcum. Four bona fide toxic peptides and 13 immunogenic peptides were found. All the classes of storage proteins were shown to contain harmful sequences. The major conclusion is that einkorn has the full potential to induce the CD syndrome, as already evident for polyploid wheats. In addition, a complete overview of the storage protein gene arsenal in T. monococcum is provided, including a full-length HMW x-type sequence and two partial HMW y-type sequences. This paper reports the correlation between the rheological properties of bread wheat dough and the types and quantities of endosperm proteins in 28 common wheat cultivars. Different methods were used to analyse the allelic composition of these cultivars and the relative quantities of the different proteins contributing to the gluten structure. Neither dough strength ( W) nor tenacity/extensibility ( P/ L) correlated with allelic composition. Different wheats with the same allelic composition (i.e., with respect to glutenins) showed different rheological properties. The glutenins were the most influential components of W and P/ L, especially the high molecular weight (HMW) glutenin subunits and in particular the type x form. These proteins seem to increase W and are the main constituents of the gluten network. The gliadins and low molecular weight (LMW) glutenin subunits appear to act as a “solvent”, and thus modify the rheological properties of the dough by either interfering with the polymerisation of the HMW glutenin subunits, or by altering the relative amounts of the different types of glutenin available. Thus, the protein subunits coded for by the alleles Glu-B1x7 and Glu-D1x5 stabilised the gluten network, whereas those coded for by Glu-B1x17 and Glu-D1x2 had the opposite effect. Dough properties therefore appear to depend on the glutenin/gliadins balance, and on the ratio of the type x and type y HMW proteins.
