Agar substrates forin vitroculture are well adapted to plant micropropagation, but not to plant rooting and acclimatization. Conversely, paper-pulp-based substrates appear as potentially well adapted forin vitroculture and functional root production. To reinforce this hypothesis, this study comparesin vitrodevelopment of nemesia on several substrates. Strong differences between nemesia roots growing in agar or in paper-pulp substrates were evidenced through scanning electron microscopy. Roots developed in agar have shorter hairs, larger rhizodermal cells, and less organized root caps than those growing on paper pulp. In conclusion, it should be noted that in this study,in vitromicroporous substrates such as paper pulp lead to the production of similar root hairs to those found in greenhouse peat substrates. Consequently, if agar could be used for micropropagation, rooting, and plant acclimatization, enhancement could be achieved if rooting stage was performed on micro-porous substrates such as paper pulp.

Background Most studies on the paper mulberry are mainly focused on the medicated and pharmacology, fiber quality, leaves feed development, little is known about its mechanism of adaptability to abiotic stress. Physiological measurement, transcriptomics and proteomic analysis were employed to understand its response to cold stress in this study. Methods The second to fourth fully expanded leaves from up to down were harvested at different stress time points forthe transmission electron microscope (TEM) observation. Physiological characteristics measurement included the relative electrolyte leakage (REL), SOD activity assay, soluble sugar content, and Chlorophyll fluorescence parameter measurement. For screening of differentially expressed genes, the expression level of every transcript in each sample was calculated by quantifying the number of Illumina reads. To identify the differentially expressed protein, leaves of plants under 0, 6, 12, 24, 48 and 72 h cold stress wereharvested for proteomic analysis. Finally, real time PCR was used to verify the DEG results of the RNA-seq and the proteomics data. Results Results showed that at the beginning of cold stress, respiratory metabolism was decreased and the transportation and hydrolysis of photosynthetic products was inhibited, leading to an accumulation of starch in the chloroplasts. Total of 5800 unigenes and 38 proteins were affected, including the repressed expression of photosynthesis and the enhanced expression in signal transduction, stress defense pathway as well as secondary metabolism. Although the transcriptional level of a large number of genes has been restored after 12 h, sustained cold stress brought more serious injury to the leaf cells, including the sharp rise of the relative electrolyte leakage, the declined Fv/Fm value, swelled chloroplast and the disintegrated membrane system. Conclusion The starch accumulation and the photoinhibition might be the main adaptive mechanism of the paper mulberry responded to cold stress. Most of important, enhancing the transport and hydrolysis of photosynthetic products could be the potential targets for improving the cold tolerance of the paper mulberry. SummaryClimate change-driven extreme weather events, combined with increasing temperatures, harsh soil conditions, low water availability and quality, and the introduction of many man-made pollutants, pose a unique challenge to plants. Although our knowledge of the response of plants to each of these individual conditions is vast, we know very little about how a combination of many of these factors, occurring simultaneously, i.e., multifactorial stress combination, impacts plants.Seedlings of wild type and different mutants of Arabidopsis thaliana plants were subjected to a multifactorial stress combination of six different stresses, each applied at a low level, and their survival, physiological and molecular responses determined.Our findings reveal that while each of the different stresses, applied individually, had a negligible effect on plant growth and survival, the accumulated impact of multifactorial stress combination on plants was detrimental. We further show that the response of plants to multifactorial stress combination is unique and that specific pathways and processes play a critical role in the acclimation of plants to multifactorial stress combination.Taken together our findings reveal that further polluting our environment could result in higher complexities of multifactorial stress combinations that in turn could drive a critical decline in plant growth and survival.Plain Language SummaryThe effects of multiple stress conditions occurring simultaneously, i.e., multifactorial stress combination, on plants is currently unknown. Here we show that different co-occurring stresses can interact to negatively impact plant growth and survival, even if the effect of each individual stress is negligible. We further identify several key pathways essential for plant acclimation to multifactorial stress combination.

Summary Tropical climates are getting warmer, with pronounced dry periods in large areas. The productivity and climate feedbacks of future tropical forests depend on the ability of trees to acclimate their physiological processes, such as leaf dark respiration (R d), to these new conditions. However, knowledge on this is currently limited due to data scarcity.We studied the impact of growth temperature on R d and its dependency on net photosynthesis (A n), leaf nitrogen (N) and phosphorus (P) contents, and leaf mass per unit area (LMA) in 16 early‐successional (ES) and late‐successional (LS) tropical tree species in multispecies plantations along an elevation gradient (Rwanda TREE project). Moreover, we explored the effect of drought on R d in one ES and one LS species.Leaf R d at 20°C decreased at warmer sites, regardless if it was expressed per unit leaf area, mass, N or P. This acclimation resulted in an 8% and a 28% decrease in R d at prevailing nighttime temperatures in trees at the intermediate and warmest sites, respectively. Moreover, drought reduced R d, particularly in the ES species and at the coolest site.Thermal acclimation of R d is complete or overcompensatory and independent of changes in leaf nutrients or LMA in African tropical trees.

To maintain constant body temperature (Tb) over a wide range of ambient temperatures (Ta) endothermic animals require large amounts of energy and water. In hot environments, the main threat to endothermic homeotherms is insufficient water to supply that necessary for thermoregulation. We investigated flexible adjustment of traits related to thermoregulation and water conservation during acclimation to hot conditions or restricted water availability, or both, in the zebra finch, Taeniopygia guttata a small arid-zone passerine. Using indirect calorimetry, we measured changes in whole animal metabolic rate (MR), evaporative heat loss (EHL) and Tb before and after acclimation to 23 or 40 °C, with different availability of water. Additionally, we quantified changes in partitioning of EHL into respiratory and cutaneous avenues in birds exposed to 25 and 40 °C. In response to heat and water restriction zebra finches decreased MR, which together with unchanged EHL resulted in increased efficiency of evaporative heat loss. This facilitated more precise Tb regulation in heat-acclimated birds. Acclimation temperature and water availability had no effect on the partitioning of EHL into cutaneous or respiratory avenues. At 25 °C, cutaneous EHL accounted for ~ 60% of total EHL, while at 40 °C, its contribution decreased to ~ 20%. Consistent among-individual differences in MR and EHL suggest that these traits, provided that they are heritable, may be a subject to natural selection. We conclude that phenotypic flexibility in metabolic heat production associated with acclimation to hot, water-scarce conditions is crucial in response to changing environmental conditions, especially in the face of current and predicted climate change.

The process of local adaptation involves differential changes in fitness over time across different environments. Although experimental evolution studies have extensively tested for patterns of local adaptation at a single time point, there is relatively little research that examines fitness more than once during the time course of adaptation. We allowed replicate populations of the fruit pest Drosophila suzukii to evolve in one of eight different fruit media. After five generations, populations with the highest initial levels of maladaptation had mostly gone extinct, whereas experimental populations evolving on cherry, strawberry and cranberry media had survived. We measured the fitness of each surviving population in each of the three fruit media after five and after 26 generations of evolution. After five generations, adaptation to each medium was associated with increased fitness in the two other media. This was also true after 26 generations, except when populations that evolved on cranberry medium developed on cherry medium. These results suggest that, in the theoretical framework of a fitness landscape, the fitness optima of cherry and cranberry media are the furthest apart. Our results show that studying how fitness changes across several environments and across multiple generations provides insights into the dynamics of local adaptation that would not be evident if fitness were analysed at a single point in time. By allowing a qualitative mapping of an experimental fitness landscape, our approach will improve our understanding of the ecological factors that drive the evolution of local adaptation in D. suzukii., Using fitness landscape theory to study adaptation and correlated fitness responses over two time scales in Drosophila suzukii populations evolving in different fruit environments.

Storage of meristematic tissue at ultra-low temperatures offers a mean to maintain valuable genetic resources from vegetatively reproduced plants. To reveal the biology underlying cryo-stress, shoot tips of the model plant Arabidopsis thaliana were subjected to a standard preservation procedure. A transcriptomic approach was taken to describe the subsequent cellular events which occurred. The cryoprotectant treatment induced the changes in the transcript levels of genes associated with RNA processing and primary metabolism. Explants of a mutant lacking a functional copy of the transcription factor WRKY22 were compromised for recovery. A number of putative downstream targets of WRKY22 were identified, some related to phytohormone-mediated defense, to the osmotic stress response, and to development. There were also alterations in the abundance of transcript produced by genes encoding photosynthesis-related proteins. The wrky22 mutant plants developed an open stomata phenotype in response to their exposure to the cryoprotectant solution. WRKY22 probably regulates a transcriptional network during cryo-stress, linking the explant’s defense and osmotic stress responses to changes in its primary metabolism. A model is proposed linking WRKY53 and WRKY70 downstream of the action of WRKY22., The transcription factor WRKY22 links the explant’s defense and osmotic stress response during Arabidopsis shoot tip cryo-stress.

Tuberose (Polianthes tuberosa L.) is one of the most important tropical bulbous-ornamental cultivated for production of long-lasting flowers spikes. Adaptation and acclimatization of different cultivars under humid agro-climatic conditions of Goa are to be confirmed for their better performance. The present investigation was conducted to evaluate the performance of tuberose cultivars under agro-climatic conditions of Goa during 2014- 2017. Five single and six double cultivars of tuberose were evaluated during the study period. All the cultivars differed in their growth and flowering behaviour. Among the single cultivars, evaluated, maximum number of florets per spike (47.00) was observed in Pune local whereas spike-length (75.59 cm) was maximum in Mexican Single. Among the double cultivars, evaluated, maximum plant height (52.21 cm) and maximum number of leaves per plant (59.63) were recorded with cultivar Arka Suvasini. Leaf length was significantly higher (52.93 cm) in Pearl double whereas leaf width (2.04 cm) was maximum in Calcutta Double. Days to appearance of flower spike were earlier in Arka Suvasini. Minimum days taken for opening of basal floret (84.88 days) were recorded with cultivar Arka Suvasini. Spike girth (0.68 cm), Spike fresh-weight (69.06 cm), floret stalk-length (3.6 cm), floret diameter (5.24 cm), weight of individual floret (3.49 g) and vase life (7.93 days) was significantly maximum in Cv. Arka Suvasini followed by Pearl Double. Based on the performance evaluation cv. Mexican Single among single types and cv. Arka Suvasini and Pearl Double among double types could be recommended for commercial cultivation under agro climatic conditions of Goa.

Gene regulation is central for growth, development, and adaptation to environmental changes in all living organisms. Many genes are induced by environmental cues, and the expression of these inducible genes is often repressed under normal conditions. Here, we show that the SHINY2 (SHI2) gene is important for repressing salt-inducible genes and also plays a role in cold response. The shi2 mutant displayed hypersensitivity to cold, abscisic acid (ABA), and LiCl. Map-based cloning demonstrates that SHI2 encodes a DEAD- (Asp-Glu-Ala-Asp) box RNA helicase with similarity to a yeast splicing factor. Transcriptomic analysis of the shi2 mutant in response to cold revealed that the shi2 mutation decreased the number of cold-responsive genes and the magnitude of their response, and resulted in the mis-splicing of some cold-responsive genes. Under salt stress, however, the shi2 mutation increased the number of salt-responsive genes but had a negligible effect on mRNA splicing. Our results suggest that SHI2 is a component in a ready-for-transcription repressor complex important for gene repression under normal conditions, and for gene activation and transcription under stress conditions. In addition, SHI2 also serves as a splicing factor required for proper splicing of cold-responsive genes and affects 5' capping and polyadenylation site selection., The DEAD-box RNA helix SHI2 represses expression of salt-inducible genes and modulates splicing of cold-responsive genes. SHI2 participates in transcription and post-transcriptional processes in response to stress conditions.

The positive effects of high atmospheric CO2 concentrations [CO2] decrease over time in most C3 plants because of down-regulation of photosynthesis. A notable exception to this trend is plants hosting N-fixing bacteria. The decrease in photosynthetic capacity associated with an extended exposure to high [CO2] was therefore studied in non-nodulating rice that can establish endophytic interactions. Rice plants were inoculated with diazotrophic endophytes isolated from the Salicaceae and CO2 response curves of photosynthesis were determined in the absence or presence of endophytes at the panicle initiation stage. Non-inoculated plants grown under elevated [CO2] showed a down-regulation of photosynthesis compared to those grown under ambient [CO2]. In contrast, the endophyte-inoculated plants did not show a decrease in photosynthesis associated with high [CO2], and they exhibited higher photosynthetic electron transport and mesophyll conductance rates than non-inoculated plants under high [CO2]. The endophyte-dependent alleviation of decreases in photosynthesis under high [CO2] led to an increase in water-use efficiency. These effects were most pronounced when the N supply was limited. The results suggest that inoculation with N-fixing endophytes could be an effective means of improving plant growth under high [CO2] by alleviating N limitations., Symbiotic endophytes facilitate electron transport and CO2 diffusion in leaves of rice grown under elevated CO2, and alleviate photosynthetic down-regulation and improve water-use efficiency of the plants.

Bio-filtration technology is considered as one of prevention solutions for declining raw water quality. The research purpose was to identify biofilm growth formation as viability cell in acclimatization period that can be used for ammonia elimination by using various indigenous chemo-heterotroph bacteria isolated from bio-filter media specimen and moving bed bio-filter reactor performance. Reactor was configured within aerobic system comparing two biofilm–attached support–medium filters (i.e. drinking-water-bottle-PET-chips and PVC). Bacteria potency on ammonia removal in kinetics modeling was advance investigated. Kinetics determination was based on empirical approach in first order by using formula of Cout = Cin exp(-k.EBRT), where Cout is ammonia concentration in outlet, Cin is ammonia concentration in inlet, k is first-order reaction rate constant and Empty Bed Retention Time (EBRT) can be formulated about setting comparison the discharge ammonia load (Q) and volume reactor (V). Total thirteen potential biofilm bacterias were successfully isolated in 24–hours–incubation at 37oC and by using Bergey’s method successfully identified prime biofilm isolates of Bacillus sp and Pseudomonas sp, then re-selected to determine one with the best ammonia removal ability. Plants are constantly exposed to temperature fluctuations, which have direct effects on all cellular reactions because temperature influences reaction likelihood and speed. Chloroplasts are crucial to temperature acclimation responses of plants, due to their photosynthetic reactions whose products play a central role in plant metabolism. Consequently, chloroplasts serve as sensors of temperature changes and are simultaneously major targets of temperature acclimation. The core subunits of the complexes involved in the light reactions of photosynthesis are encoded in the chloroplast. As a result, it is assumed that temperature acclimation in plants requires regulatory responses in chloroplast gene expression and protein turnover. We conducted western blot experiments to assess changes in the accumulation of two photosynthetic complexes (PSII, and Cytb6f complex) and the ATP synthase in tobacco plants over two days of acclimation to low temperature. Surprisingly, the concentration of proteins within the chloroplast varied negligibly compared to controls. To explain this observation, we used a simplified Ordinary Differential Equation (ODE) model of transcription, translation, mRNA degradation and protein degradation to explain how the protein concentration can be kept constant. This model takes into account temperature effects on these processes. Through simulations of the ODE model, we show that mRNA and protein degradation are possible targets for control during temperature acclimation. Our model provides a basis for future directions in research and the analysis of future results. The aim of the work on sulphates was to investigate their impact on UASB performance, including the response to step changes in influent concentration. It has been shown that UASB reactors treating municipal-type wastewaters at 20 o C can remove sulphate effectively even at relatively low COD/SO4 ratios and/or high concentrations. In this experiment, therefore the UASB reactors were fed on influent with different sulphate concentrations to establish the effects on key performance parameters. It was used eight 4-litre continuously fed UASB reactors. The effect of sulphate addition on methane production was much greater than on COD removal, the specific methane production and the ratio of actual to theoretical methane produced per g of COD removed showed relatively little effect from influent sulphate concentrations below ~120 mg SO 4 l -1 (COD/SO 4 ratio 5.8), although this may in part have reflected a period of acclimatisation while the population of sulphate-reducing bacteria (SRB) increased.

Maize is a cold-sensitive species, but selective breeding programs have recently succeeded in producing plants strikingly well adapted to the cold springs of a temperate climate, showing the potential for improved cold tolerance. The aim of the present study was to determine whether the adaptation of some inbred lines to spring chills is due to their increased true cold tolerance or whether it only represents an avoidance mechanism, which was the sole mode of adaptation during early stages of agricultural dispersal of maize towards higher latitudes. By characterizing numerous physiological features of several lines of different cold sensitivity, we show that a combination of both avoidance and tolerance is involved. A novel avoidance mechanism was found that favored unhindered development of the photosynthetic apparatus through protection of the shoot apex below soil level due to a shortened mesocotyl. It seems to be mediated by increased seedling photosensitivity at early growth stages. True tolerance involved improved protection of the cell membrane against cold injury at temperatures close to 0 °C and stimulation of light-induced processes (accumulation of anthocyanins, carotenoids, and chlorophyll, proper development of chloroplasts) at temperatures in the range of 10–14 °C, likely also related to the increased photosensitivity and mediated by gibberellin signaling., High photosensitivity of seedlings of some modern maize inbred lines keeps the growing shoot below soil level and protects it against cold injury allowing for unhindered development at low temperatures.

Few studies have addressed how reduced water salinity affects cardiovascular and metabolic function in marine euryhaline fishes, despite its relevance for predicting impacts of natural salinity variations and ongoing climate change on marine fish populations. Here, shorthorn sculpin (Myoxocephalus scorpius) were subjected to different durations of reduced water salinity from 33 to 15 ppt. Routine metabolic rate decreased after short-term acclimation (4–9 days) to 15 ppt, which corresponded with similar reductions in cardiac output. Likewise, standard metabolic rate decreased after acute transition (3 h) from 33 to 15 ppt, suggesting a reduced energetic cost of osmoregulation at 15 ppt. Interestingly, gut blood flow remained unchanged across salinities, which contrasts with previous findings in freshwater euryhaline teleosts (e.g., rainbow trout) exposed to different salinities. Although plasma osmolality, [Na+], [Cl−] and [Ca2+] decreased in 15 ppt, there were no signs of cellular osmotic stress as plasma [K+], [hemoglobin] and hematocrit remained unchanged. Taken together, our data suggest that shorthorn sculpin are relatively weak plasma osmoregulators that apply a strategy whereby epithelial ion transport mechanisms are partially maintained across salinities, while plasma composition is allowed to fluctuate within certain ranges. This may have energetic benefits in environments where salinity naturally fluctuates, and could provide shorthorn sculpin with competitive advantages if salinity fluctuations intensify with climate change in the future.

The purpose of this study was to investigate local sweat rate (LSR) and sweat composition before and after active or passive heat re-acclimation (HRA). Fifteen participants completed four standardized heat stress tests (HST): before and after ten days of controlled hyperthermia (CH) heat acclimation (HA), and before and after five days of HRA. Each HST consisted of 35 min of cycling at 1.5W·kg−1 body mass (33°C and 65% relative humidity), followed by a graded exercise test. For HRA, participants were re-exposed to either CH (CH-CH, n = 6), hot water immersion (water temperature ~40°C for 40 min; CH-HWI, n = 5) or control (CH-CON, n = 4). LSR, sweat sodium, chloride, lactate and potassium concentrations were determined on the arm and back. LSR increased following HA (arm +18%; back +41%, P ≤ 0.03) and HRA (CH-CH: arm +31%; back +45%; CH-HWI: arm +65%; back +49%; CH-CON arm +11%; back +11%, P ≤ 0.021). The objective was to determine the optimal dehydrated passion fruit pulp replacing ground corn in diets for goat kids. We used 24 crossbred castrated goat kids with initial body weight of 26.6±0.3 kg. The experimental period was 23 days, with 15 days of adjustment and acclimatization (mostly to the installation and diet). Kids were distributed in a randomized experimental design with four treatments of 0, 80, 160, or 240 g/kg of dehydrated passion fruit residue as a supplementary inclusion in the dry matter (DM) content of the diet. The statistical model considered the fixed effect of the included amount of residue from dehydrated passion fruit. Analysis of variance was conducted using the PROC GLM of SAS. Intakes of DM and total digestible nutrients (TDN) were not affected by residue inclusion. However, neutral detergent fiber (NDF) intake increased linearly. The inclusion linearly decreased intake of ether extract (EE) and non-fibrous carbohydrates (NFC). Crude protein (CP) intake was quadratically affected, with a greater value under treatment of 160 g/kg passion fruit. Dry matter and NFC digestibility decreased linearly with the amount of passion fruit included in diet. Fecal N and total N excretion values (in g/d and %N intake) increased linearly. Nitrogen balance and N retained values showed a quadratic increase, with greater values under the 160 g/kg inclusion. Time spent ruminating, DM and NDF eating efficiency, and g DM/bolus chewed were affected quadratically by inclusion of passion fruit residue. The replacement of ground corn with dehydrated passion fruit residue at a level of 160 g/kg is recommended for diets of goat kids to improve their NDF and CP intake and their N balance without affecting DM and TDN intakes.

We discuss the contribution of the versatile and atypical plant histone deacetylase, HDA9, to plant growth, development, and acclimation to the environment, and propose novel leads for future research., Plants tightly control gene transcription to adapt to environmental conditions and steer growth and development. Different types of epigenetic modifications are instrumental in these processes. In recent years, an important role for the chromatin-modifying RPD3/HDA1 class I HDAC HISTONE DEACETYLASE 9 (HDA9) emerged in the regulation of a multitude of plant traits and responses. HDACs are widely considered transcriptional repressors and are typically part of multiprotein complexes containing co-repressors, DNA, and histone-binding proteins. By catalyzing the removal of acetyl groups from lysine residues of histone protein tails, HDA9 negatively controls gene expression in many cases, in concert with interacting proteins such as POWERDRESS (PWR), HIGH EXPRESSION OF OSMOTICALLY RESPONSIVE GENES 15 (HOS15), WRKY53, ELONGATED HYPOCOTYL 5 (HY5), ABA INSENSITIVE 4 (ABI4), and EARLY FLOWERING 3 (ELF3). However, HDA9 activity has also been directly linked to transcriptional activation. In addition, following the recent breakthrough discovery of mutual negative feedback regulation between HDA9 and its interacting WRKY-domain transcription factor WRKY53, swift progress in gaining understanding of the biology of HDA9 is expected. In this review, we summarize knowledge on this intriguing versatile—and long under-rated—protein and propose novel leads to further unravel HDA9-governed molecular networks underlying plant development and environmental biology.

The pig is a powerful model for intestinal barrier studies, and it is important to carefully plan animal care and handling for optimal study design as psychological and physiological stressors significantly impact intestinal mucosal barrier function. Here, we report the effects of a period of environmental acclimation versus acute transport stress on mucosal barrier repair after intestinal ischemic injury. Jejunal ischemia was induced in young pigs which had been allowed to acclimate to a biomedical research housing environment or had been transported immediately prior to experimental injury (non-acclimated). Mucosa was then incubated ex vivo on Ussing chambers. In uninjured mucosa, there was no difference in transepithelial electrical resistance (TEER) or epithelial integrity between groups. However, acclimated pigs had increased macromolecular flux as compared to non-acclimated pigs during the first hour of ex vivo incubation. Ischemia induced greater epithelial loss in non-acclimated pigs as compared to acclimated pigs, yet this group achieved greater wound healing during recovery. Non-acclimated pigs had more robust TEER recovery ex vivo following injury versus acclimated pigs. The expression pattern of the tight junction protein claudin-4 was disrupted in acclimated pigs following recovery but showed enhanced localization to the apical membrane in non-acclimated pigs following recovery. Acute transport stress increases mucosal susceptibility to epithelial loss but also primes the tissue for a more robust barrier repair response. Alternatively, environmental acclimation increases leak pathway and diminishes barrier repair responses after ischemic injury.

A one-stage anaerobic ammonium oxidation (ANAMMOX) reactor can be quickly started within 40 days by mixing partial nitrifying sludge with ANAMMOX granular sludge with an average temperature of 30 &deg, C. After 70 days of nitrogen load acclimation, Acinetobacter, including Candidatus Kuenenia, became the dominant strain of the system within the reactor, which exhibited high efficiency and a stable nitrogen removal performance. At an influent chemical oxygen demand (COD), NH4+-N content, total nitrogen (TN) content, hydraulic retention time (HRT), temperature, and reactor dissolved oxygen (DO) content of 100, 60, and 70 mg/L, 6 h, 30 &plusmn, 1 &deg, C, and below 0.6 mg/L, respectively, the one-stage ANAMMOX reactor could effectively treat domestic sewage on campus. The removal rates of COD, NH4+-N, and TN were approximately 89%, 96.7%, and 70%, respectively.

The anammox process is an energy efficient promising alternative to biologically remove the nitrogen. Thus, a 5-L anammox granular reactor was inoculated with sludge coming from a sidestream partial nitritation and anammox reactor (>200 mg TN/L and 30 °C) and it was directly subjected to 15 ± 1 °C treating mimicked municipal wastewater (50 mg TN/L). Results indicated that an acclimation period (commonly used) to progressive reach the mainstream conditions is not needed, shortening the start-up periods. The long-term anammox process stability was proved to treat synthetic wastewater with decreasing alkalinities and nitritified primary settled municipal wastewater. The low pH values (6.2 ± 0.1) of the municipal wastewater fed did not affect the process stability. Residual organic matter concentrations augmented the nitrogen removal efficiency from 80 % (with the synthetic medium) to 92 % achieving effluent concentrations below 10 mg TN/L. Acclimation, via phenotypic flexibility, is a potential means for a fast response to climate change. Understanding the molecular mechanisms underpinning phenotypic flexibility can provide a fine-scale cellular understanding of how organisms acclimate. In the last 30 years, Mya truncata populations around the UK have faced an average increase in sea surface temperature of 0.7 °C and further warming of between 1.5 and 4 °C, in all marine regions adjacent to the UK, is predicted by the end of the century. Hence, data are required on the ability of M. truncata to acclimate to physiological stresses, and most notably, chronic increases in temperature. Animals in the present study were exposed to chronic heat-stress for 2 months prior to shell damage and subsequently, only 3, out of 20 damaged individuals, were able to repair their shells within 2 weeks. Differentially expressed genes (between control and damaged animals) were functionally enriched with processes relating to cellular stress, the immune response and biomineralisation. Comparative transcriptomics highlighted genes, and more broadly molecular mechanisms, that are likely to be pivotal in this lack of acclimation. This study demonstrates that discovery-led transcriptomic profiling of animals during stress-response experiments can shed light on the complexity of biological processes and changes within organisms that can be more difficult to detect at higher levels of biological organisation. This study addressed whether competition under different light environments was reflected by changes in leaf absorbed light energy partitioning, photosynthetic efficiency, relative growth rate and biomass allocation in invasive and native competitors. Additionally, a potential allelopathic effect of mulching with invasive Prunus serotina leaves on native Quercus petraea growth and photosynthesis was tested. The effect of light environment on leaf absorbed light energy partitioning and photosynthetic characteristics was more pronounced than the effects of interspecific competition and allelopathy. The quantum yield of PSII of invasive P. serotina increased in the presence of a competitor, indicating a higher plasticity in energy partitioning for the invasive over the native Q. petraea, giving it a competitive advantage. The most striking difference between the two study species was the higher crown-level net CO2 assimilation rates (Acrown) of P. serotina compared with Q. petraea. At the juvenile life stage, higher relative growth rate and higher biomass allocation to foliage allowed P. serotina to absorb and use light energy for photosynthesis more efficiently than Q. petraea. Species-specific strategies of growth, biomass allocation, light energy partitioning and photosynthetic efficiency varied with the light environment and gave an advantage to the invader over its native competitor in competition for light. However, higher biomass allocation to roots in Q. petraea allows for greater belowground competition for water and nutrients as compared to P. serotina. This niche differentiation may compensate for the lower aboveground competitiveness of the native species and explain its ability to co-occur with the invasive competitor in natural forest settings. Changes in the apoplast associated with cell wall modifications and anti-freeze proteins in the shoot apical meristem and vascular transition zone suggest differential acclimation responses within the winter wheat crown., The wheat (Triticum aestivum L.) crown is the critical organ of low temperature stress survival over winter. In cold-acclimated crowns, ice formation in the apoplast causes severe tissue disruption as it grows at the expense of intracellular water. While previous crown studies have shown the vascular transition zone (VTZ) to have a higher freezing sensitivity than the shoot apical meristem (SAM), the mechanism behind the differential freezing response is not fully understood. Cooling cold-acclimated crowns to –10 °C resulted in an absence of VTZ tetrazolium chloride staining, whereas the temperatures at which 50% of the SAM stained positive and 50% of plants recovered (LT50) were similar after cold acclimation for 21 (–16 °C) and 42 d (–20 °C) at 4 °C. Proteomic analysis of the apoplastic fluids identified dehydrins, vernalization-responsive proteins, and cold shock proteins preferentially accumulated in the SAM. In contrast, modifications to the VTZ centered on increases in pathogenesis-related proteins, anti-freeze proteins, and sugar hydrolyzing enzymes. Fourier transform infrared spectroscopy focal plane array analysis identified the biochemical modification of the cell wall to enhance methyl-esterified cross-linking of glucuronoarabinoxylans in the VTZ. These findings indicate that the SAM and VTZ express two distinct tissue-specific apoplastic responses during cold acclimation.

Hypobaric hypoxia elicits several patho-physiological manifestations, some of which are known to be lethal. Among various molecular mechanisms proposed so far, perturbation in redox state due to imbalance between radical generation and antioxidant defence is promising. These molecular events are also related to hypoxic status of cancer cells and therefore its understanding has extended clinical advantage beyond high altitude hypoxia. In present study, however, the focus was to understand and propose a model for rapid acclimatization of high altitude visitors to enhance their performance based on molecular changes. We considered using simulated hypobaric hypoxia at some established thresholds of high altitude stratification based on known physiological effects. Previous studies have focused on the temporal aspect while overlooking the effects of varying pO2 levels during exposure to hypobaric hypoxia. The pO2 levels, indicative of altitude, are crucial to redox homeostasis and can be the limiting factor during acclimatization to hypobaric hypoxia. In this study we present the effects of acute (24 h) exposure to high (3049 m; pO2: 71 kPa), very high (4573 m; pO2: 59 kPa) and extreme altitude (7620 m; pO2: 40 kPa) zones on lung and plasma using semi-quantitative redox specific transcripts and quantitative proteo-bioinformatics workflow in conjunction with redox stress assays. It was observed that direct exposure to extreme altitude caused 100% mortality, which turned into high survival rate after pre-exposure to 59 kPa, for which molecular explanation were also found. The pO2 of 59 kPa (very high altitude zone) elicits systemic energy and redox homeostatic processes by modulating the STAT3-RXR-Nrf2 trio.
