The halfway point for the implementation of the United Nations Sustainable Development Goals (SDGs) was marked in 2023, as set forth in the 2030 Agenda. Geospatial technologies have proven indispensable in assessing and tracking fundamental components of each of the 17 SDGs, including climatological and ecological trends, and changes and humanitarian crises and socio-economic impacts. However, gaps remain in the capacity for geospatial and related digital technologies, like AI, to provide a deeper, more comprehensive understanding of the complex and multi-factorial challenges delineated in the SDGs. Lack of progress toward these goals, and the immense implementation challenges that remain, call for inclusive and holistic approaches, coupled with transformative uses of digital technologies. This paper reviews transdisciplinary, holistic, and participatory approaches to address gaps in ethics and diversity in geospatial and related technologies and to meet the pressing need for bottom-up, community-driven initiatives. Small-scale, community-based initiatives are known to have a systemic and aggregate effect toward macro-economic and global environmental goals. Cybernetic systems thinking approaches are the conceptual framework investigated in this study, as these approaches suggest that a decentralized, polycentric system—for example, each community acting as one node in a larger, global system—has the resilience and capacity to create and sustain positive change, even if it is counter to top-down decisions and mechanisms. Thus, this paper will discuss how holistic systems thinking—societal, political, environmental, and economic choices considered in an interrelated context—may be central to building true resilience to climate change and creating sustainable development pathways. Traditional and Indigenous knowledge (IK) systems around the world hold holistic awareness of human-ecological interactions—practicable, reciprocal relationships developed over time as a cultural approach. This cultural holistic approach is also known as Systemic Literacy, which considers how systems function beyond "mechanical" aspects and include political, philosophical, psychological, emotional, relational, anthropological, and ecological dimensions. When Indigenous-led, these dimensions can be unified into participatory, community-centered conservation practices that support long-term human and environmental well-being. There is a growing recognition of the criticality of Indigenous leadership in sustainability practices, as well as that partnerships with Indigenous peoples and weaving knowledge systems, as a missing link to approaching global ecological crises. This review investigates the inequality in technological systems—the "digital divide" that further inhibits participation by communities and groups that retain knowledge of "place" and may offer the most transformative solutions. Following the review and synthesis, this study presents cybernetics as a bridge of understanding to Indigenous systems thinking. As non-Indigenous scholars, we hope that this study serves to foster informed, productive, and respectful dialogues so that the strength of diverse knowledges might offer whole-systems approaches to decision making that tackle wicked problems. Lastly, we discuss use cases of community-based processes and co-developed geospatial technologies, along with ethical considerations, as avenues toward enhancing equity and making advances in democratizing and decolonizing technology. This study examines the influence of the El Niño Southern Oscillation (ENSO) on Jamaica's rainfall patterns, leveraging CHIRPS data from 1981 to 2021 in 370 locations. Our analysis reveals a distinct ENSO imprint on rainfall, with La Niña phases showing a consistently higher probability of exceeding various rainfall thresholds compared to El Niño. Notably, La Niña increases the likelihood of heavier rainfall, particularly in the wet seasons, with probabilities of exceeding 200 mm reaching up to 50% during wet season II. Spatially, the probability of total monthly rainfall (TMR) during La Niña is elevated in the northeastern regions, suggesting regional vulnerability to excess rainfall. Additionally, during El Niño, the correlation between TMR and the maximum air temperature (Tmax) is significantly stronger, indicating a positive and more pronounced relationship between higher temperatures and rainfall, with correlation coefficients ranging from 0.39 to 0.80. Wind speed and evapotranspiration show a negligible influence on TMR during both ENSO phases, maintaining stable correlation patterns with only slight variations. The results of this study underscore the necessity for differentiated regional strategies in water resource management and disaster preparedness, tailored to the unique climatic characteristics imposed by ENSO variability. These insights contribute to a refined understanding of climate impacts, essential for enhancing resilience and adaptive capacity in Jamaica and other small island developing states. The Caribbean region is highly vulnerable to multiple hazards. Resultant impacts may be derived from single or multiple cascading risks caused by hydrological-meteorological, seismic, geologic, or anthropological triggers, disturbances, or events. Studies suggest that event records and data related to hazards, risk, damage, and loss are limited in this region. National Disaster Risk Reduction (DRR) planning and response require data of sufficient quantity and quality to generate actionable information, statistical inferences, and insights to guide continual policy improvements for effective DRR, national preparedness, and response in both time and space. To address this knowledge gap, we review the current state of knowledge, data, models, and tools, identifying potential opportunities, capacity needs, and long-term benefits for integrating Earth Observation (EO) understanding, data, models, and tools to further enhance and strengthen the national DRR framework using two common disasters in Jamaica: floods and landslides. This review serves as an analysis of the current state of DRR management and assess future opportunities. Equally, to illustrate and guide other United Nations Disaster Risk Reduction (UNDRR) priority countries in the Pacific region, known as Small Island Developing States (SIDS), to grapple with threats of multiple and compounding hazards in the face of increasing frequency, intensity, and duration of extreme weather events, and climate change impact. The impact melt‐bearing breccias at the Ries impact structure, Germany, host degassing pipes: vertical structures that are inferred to represent conduits along which gases and fluids escaped to the surface, consistent with hydrothermal activity that occurs soon after an impact event. Although the presence of degassing pipes has been recognized within the well‐preserved and long‐studied ejecta deposits at the Ries, a detailed mineralogical study of their alteration mineralogy, as an avenue to elucidate their origins, has not been conducted to date. Through the application of high‐resolution in situ reflectance imaging spectroscopy and X‐ray diffraction, this study shows for the first time that the degassing pipe interiors and associated alteration are comprised of hydrated and hydroxylated silicates (i.e., Fe/Mg smectitic clay minerals with chloritic or other hydroxy‐interlayered material) as secondary hydrothermal mineral phases. This study spatially extends the known effects of impact hydrothermal activity into the ejecta deposits, beyond the crater rim. It has been suggested that the degassing pipes at the Ries are analogous to crater‐related pit clusters observed in impact melt‐bearing deposits on Mars, Ceres, and Vesta. The results of this work may inform on the presence of crustal volatiles and their interaction during the impact process on rocky bodies throughout the solar system. The Mars 2020 Perseverance rover may have the opportunity to investigate impact‐related features in situ; if so, this work suggests that such investigations may provide key information on the origin and formation of clay minerals on Mars as well as hold exciting implications for future Mars exploration. Perseverance's Mastcam-Z instrument provides high-resolution stereo and multispectral images with a unique combination of spatial resolution, spatial coverage, and wavelength coverage along the rover's traverse in Jezero crater, Mars. Images reveal rocks consistent with an igneous (including volcanic and/or volcaniclastic) and/or impactite origin and limited aqueous alteration, including polygonally fractured rocks with weathered coatings; massive boulder-forming bedrock consisting of mafic silicates, ferric oxides, and/or iron-bearing alteration minerals; and coarsely layered outcrops dominated by olivine. Pyroxene dominates the iron-bearing mineralogy in the fine-grained regolith, while olivine dominates the coarse-grained regolith. Solar and atmospheric imaging observations show significant intra- and intersol variations in dust optical depth and water ice clouds, as well as unique examples of boundary layer vortex action from both natural (dust devil) and Ingenuity helicopter-induced dust lifting. High-resolution stereo imaging also provides geologic context for rover operations, other instrument observations, and sample selection, characterization, and confirmation.

Immunoproteasomes comprise a specialized subset of proteasomes that is defined by the presence of three catalytic immunosubunits: LMP2, MECL-1 (LMP10), and LMP7. Proteasomes in general serve many cellular functions through protein degradation, whereas the specific function of immunoproteasomes has been thought to be largely, if not exclusively, optimization of MHC class I Ag processing. In this report, we demonstrate that T cells from double knockout mice lacking two of the immunosubunits, MECL-1 and LMP7, hyperproliferate in vitro in response to various polyclonal mitogens. We observe hyperproliferation of both CD4(+) and CD8(+) T cell subsets and demonstrate accelerated cell cycling. We do not observe hyperproliferation of T cells lacking only one of these subunits, and thus hyperproliferation is independent of either reduced MHC class I expression in LMP7(-/-) mice or reduced CD8(+) T cell numbers in MECL-1(-/-) mice. We observe both of these latter two phenotypes in MECL-1/LMP7(-/-) mice, which indicates that they also are independent of each other. Finally, we provide evidence of in vivo T cell dysfunction by demonstrating increased numbers of central memory phenotype CD8(+) T cells in MECL-1/LMP7(-/-) mice. In summary, this novel phenotype of hyperproliferation of T cells lacking both MECL-1 and LMP7 implicates a specific role for immunoproteasomes in T cell proliferation that is not obviously connected to MHC class I Ag processing.

Ionizing radiation is a well-known risk factor for thyroid cancer in human populations. Chromosomal rearrangements involving the RET gene, known as RET/PTC, are prevalent in thyroid papillary carcinomas from patients with radiation history. We studied the generation of RET/PTC in HTori-3 immortalized human thyroid cells exposed to a range of doses of gamma-radiation and harvested 2, 5-6, and 9 d later. RET/PTC1 and RET/PTC3 were detected by RT-PCR followed by Southern blotting and hybridization with internal oligonucleotide probes. No RET/PTC was found in cells harvested 2 and 5-6 d after irradiation, whereas 59 RET/PTC events were detected in cells collected 9 d after exposure. The average rate of RET/PTC induction was 0.1 x 10(-6) after exposure to 0.1 Gy, 1.6 x 10(-6) after 1 Gy, 3.0 x 10(-6) after 5 Gy, and 0.9 x 10(-6) after 10 Gy. When adjusted for cell survival, the rate after 10 Gy was comparable with those after 5 Gy. RET/PTC1 was more common than RET/PTC3 after each dose, comprising 80% of all rearrangements. In this study, we demonstrate a dose-dependent induction of RET/PTC rearrangements in human thyroid cells after exposure to 0.1-10 Gy gamma-radiation. This provides additional evidence for a direct link between this genetic event and radiation exposure and offers a powerful experimental system for studying radiation-induced carcinogenesis in the thyroid gland.

A PAX8-PPARgamma rearrangement has been recently identified in follicular thyroid carcinomas, but not in follicular adenomas or other thyroid tumors. We report here the analyses of PAX8-PPARgamma in a series of 118 thyroid tumors using a newly developed RT-PCR assay to detect this rearrangement in frozen and paraffin-embedded tissues and using immunostaining with a PPARgamma antibody. PAX8-PPARgamma was detected by RT-PCR in eight of 15 (53%) follicular carcinomas and two of 25 (8%) follicular adenomas but not in 35 papillary carcinomas (including 12 follicular variants), 12 Hurthle cell carcinomas, 12 Hurthle cell adenomas, two anaplastic carcinomas, one poorly differentiated carcinoma, or 16 hyperplastic nodules. The prevalence was higher in follicular carcinomas from patients with a history of radiation exposure (three of three). Strong, diffuse nuclear immunostaining with the PPARgamma antibody correlated with the presence of PAX8-PPARgamma detected by RT-PCR. Most sporadic follicular carcinomas positive for PAX8-PPARgamma were overtly invasive, whereas tumors lacking the rearrangement were predominantly minimally invasive. The two follicular adenomas positive for PAX8-PPARgamma had trabecular growth pattern and thick capsule, but no invasion, and thus may represent "pre-invasive" follicular carcinomas. The absence of PAX8-PPARgamma rearrangements in Hurthle cell tumors and papillary thyroid carcinomas highlights the differences in the molecular pathogenesis of these thyroid tumors.
