Abstract

Berries contain powerful antioxidants, potential allergens, and other bioactive compounds. Genetic and environmental factors affect production and storage of such compounds. For this reason breeding and biotechnological approaches are currently used to control or to increase the content of specific health-related compounds in fruits. This work reviews the main bioactive compounds determining the nutritional quality of berries, the major factors affecting their content and activity, and the genetic options currently available to achieve new genotypes able to provide, under controlled cultivation conditions, berries with the proper balance of bioactive compounds for improving consumer health.

3. Comparative genetic mapping between octoploid and diploid Fragaria species reveals a high level of colinearity between their genomes and the essentially disomic behavior of the cultivated octoploid strawberry

Abstract

Macrosynteny and colineality between Fragaria (strawberry) species showing extreme levels of ploidy have been studied through comparative genetic mapping between the octoploid cultivated strawberry (F. X ananassa) and its diploid relatives. A comprehensive map of the octoploid strawberry, in which almost all linkage groups are ranged into the seven expected homoeologous groups was obtained, thus providing the first reference map for the octoploid Fragaria. High levels of conserved macrosynteny and colinearity were observed between homo(eo)logous linkage groups and between the octoploid homoeologous groups and their corresponding diploid linkage groups. These results reveal that the polyploidization events that took place along the evolution of the Fragaria genus and the more recent juxtaposition of two octoploid strawberry genomes in the cultivated strawberry did not trigger any major chromosomal rearrangements in genomes involved in F. Xananassa. They further suggest the existence of a close relationship between the diploid Fragaria genomes. In addition, despite the possible existence of residual levels of polysomic segregation suggested by the observation of large linkage groups in coupling phase only, the prevalence of linkage groups in coupling/ repulsion phase clearly demonstrates that the meiotic behavior is mainly disomic in the cultivated strawberry.

Abstract

Flowering is a key event in plant life, and is finely tuned by environmental and endogenous signals to adapt to different environments. In horticulture, continuous flowering (CF) is a popular trait introduced in a wide range of cultivated varieties. It played an essential role in the tremendous success of modern roses and woodland strawberries in gardens. CF genotypes flower during all favourable seasons, whereas once-flowering (OF) genotypes only flower in spring. Here we show that in rose and strawberry continuous flowering is controlled by orthologous genes of the TERMINAL FLOWER 1 (TFL1) family. In rose, six independent pairs of CF/OF mutants differ in the presence of a retrotransposon in the second intron of the TFL1 homologue. Because of an insertion of the retrotransposon, transcription of the gene is blocked in CF roses and the absence of the floral repressor provokes continuous blooming. In OF-climbing mutants, the retrotransposon has recombined to give an allele bearing only the long terminal repeat element, thus restoring a functional allele. In OF roses, seasonal regulation of the TFL1 homologue may explain the seasonal flowering, with low expression in spring to allow the first bloom. In woodland strawberry, Fragaria vesca, a 2-bp deletion in the coding region of the TFL1 homologue introduces a frame shift and is responsible for CF behaviour. A diversity analysis has revealed that this deletion is always associated with the CF phenotype. Our results demonstrate a new role of TFL1 in perennial plants in maintaining vegetative growth and modifying flowering seasonality

International audience; The Rosodae supertribe is composed of economically important species (strawberry, rose and raspberry). In order to extend the repertoire of markers for optimizing genetic mapping and synteny within this supertribe, we developed 20 functional markers from two Fragaria cDNA libraries enriched in AG/AC microsatellite motifs. We tested their transferability within the Rosodae supertribe, including two reference diploid and octoploid strawberry segregating populations. Out of these 20 functional markers, 70% revealed polymorphism in both diploid and octoploid Fragaria segregating populations and were mapped on the diploid genetic map to provide useful information for further marker-assisted selection. These 20 EST-SSRs were also tested in 17 wild Fragaria species and displayed a high cross-species transferability (mean of 95%), providing new markers for further diversity studies within the genus. These markers were also transferable to the phylogenetically close Potentilla genus, whereas their transferability was lower for the polyploid Duchesnea indica species and for the more distant Rosa genus. Their use for genetic mapping within the Rosodae will further permit a comparison of genetic information among the different genera.

Abstract

International audience; A comparative mapping study has been done among two closely related genera in the Rosoideaetribe. Our objective was to use a common set of markersto show macrosynteny and macrocolinearity across the two generaFragaria(strawberry:2n=2x= 14) and Rosa (rose:2n=2x= 14). We used anonymous SSRs derived from a genomic library (55 primer pairs fromF. x ananassa) and from Expressed Sequence Tag (70 new primer pairs derived from Rosa(30), F.vesca (18 )and F.x ananassa (22) ESTs). In addition, we have tested 24 primer pairs derived from gene sequences involved in Rosa floral initiation. The anonymous SSRs and the SSR-ESTwere tested on denaturating polyacrylamide gel with silver staining. On the total of 125 primer pairs, only twelve (10%) were polymorphic in both populations. In order to reveal more polymorphism, we tested all primers pairs derived from ESTs with the Single-Strainded Conformation Polymorphism approach. Using this technique, more polymorphic SSRs were detected. Furthermore, primer pairs derived from floral initiation gene sequences in Rosawere tested on SSCP and 25 % were polymorphic in both populations. At thistime, 24 markers will provide anchor points to link the 7 homologous linkage groups .In order to have a true comparison of the homologous linkage groups, we shall verify the orthology of alleles by sequencing them. This comparative mapping study will provide useful information regarding the evolution of the two genomes.

Abstract

The study aimed at determining the vitamin C and total anthocyanin content in fruits of different strawberry cultivars in several harvest times in a particular harvest season. The trial was planted in 2003 in the north-western part of Croatia (vicinity of Zagreb) with the following cultivars that were harvested three times in the 2004 season: 'Elsanta', 'Marmolada', 'Madeleine', 'Miss', 'Miranda', 'Pegasus' and 'Honeoye'. Chemical analysis shows significant differences in the vitamin C and total anthocyanin content as a function of cultivar and harvest date. 'Marmolada', 'Madeleine’ and 'Miss’ had significantly higher vitamin C content with regard to 'Miranda', 'Pegasus', 'Honeoye’ and 'Elsanta'. The cultivars 'Madeleine', 'Honeoye', 'Marmolada’ and 'Pegasus’ had significantly higher total anthocyanin content in comparison with other cultivars. The vitamin C content showed the peak in the first harvest and than progressively decreased towards the last harvest, while the anthocyanin showed reverse trend with the highest content in the last harvest. The results obtained confirm that both cultivar and harvest date significantly influence the vitamin C and total anthocyanin content in strawberry.

Abstract

Three different strawberry cultivars Marmolada, Arosa and Raurica were harvested in different maturity stages (100, 85 and 70 % of red colour covered fruits). The aim of study was to investigate changes of internal and external quality parameters in relation to genotype and maturity stage. At these samples are done objective measurements of their quality: total soluble solids (TSS), and titratable acidity (TA), and fruit colour (CieLAB). Fruits of less coloured phase had higher percentage of TA. The proportion value TSS/TA was increased according to maturity stage. The lowest TSS/TA ratio had cultivar Arosa. The cultivar Raurica had higher L, a, b and C in comparison to others cultivars. Colour values L, a and C were also higher if fruits were harvested in phase 70 % of red colour. Our results confirm that genotype and maturity phases are important factors in determining of strawberry fruit quality.

Abstract

The woodland strawberry, Fragaria vesca (2n = 2x = 14), is a versatile experimental plant system. This diminutive herbaceous perennial has a small genome (240 Mb), is amenable to genetic transformation and shares substantial sequence identity with the cultivated strawberry (Fragaria × ananassa) and other economically important rosaceous plants. Here we report the draft F. vesca genome, which was sequenced to ×39 coverage using second-generation technology, assembled de novo and then anchored to the genetic linkage map into seven pseudochromosomes. This diploid strawberry sequence lacks the large genome duplications seen in other rosids. Gene prediction modeling identified 34,809 genes, with most being supported by transcriptome mapping. Genes critical to valuable horticultural traits including flavor, nutritional value and flowering time were identified. Macrosyntenic relationships between Fragaria and Prunus predict a hypothetical ancestral Rosaceae genome that had nine chromosomes. New phylogenetic analysis of 154 protein-coding genes suggests that assignment of Populus to Malvidae, rather than Fabidae, is warranted.
