Magonia pubescens is a natural species from the Brazilian cerrado biome. Its fruits and seeds are used in the treatment of seborrheic dermatitis, a common inflammatory skin disease. In this work, the known compounds lapachol, stigmasterol, maniladiol and scopoletin were isolated from hexane and dichloromethane extracts of M. pubescens branches. The aqueous extract of this material was fractioned through a liquid-liquid partition and the obtained fractions were analyzed by UHPLC-MS/MS. The results obtained were compared with data from three databases, leading to the putative identification of 51 compounds from different classes, including flavonoids, saponins and triterpenes. The cytotoxicity of aqueous fractions was assayed against breast cancer (MDA-MB-231) and leukemia (THP-1 and K562) cells. Botanical biopesticides have emerged as sustainable and eco-friendly alternatives to synthetic pesticides, whose indiscriminate use leads to several drawbacks to human and environmental health. To the best of our knowledge, there have been no reports on M. pubescens' bioactivity on phytopathogens affecting crops as a potential fungicide or antifeedant. This has encouraged us to investigate the potential of the roots of this plant as a source of biopesticides. The present study reports on the evaluation of the roots extract from Magonia pubescens A. St.-Hill., a species from the Cerrado (Brazilian savannah), on the phytopathogenic fungi Botrytis cinerea, Fusarium oxysporum, and Alternaria alternata. In addition, its insect antifeedant effect was assayed against Chrysodeixis chalcites. Thus, an in vitro test-assay was used to determine the fungicide potential (percentage growth inhibition, % GI) of the ethanolic extract of this plant species, whereas a leaf-disk bioassay on the 5th instar larvae of C. chalcites was performed to evaluate its insecticidal potential. The ethanolic extract was further fractionated by liquid–liquid partition using solvents of increasing polarity. The hexane/dichloromethane fraction exhibited a moderated potency and was similar to the ethanolic extract on the three assayed fungi (around % GI 30 at 1 mg/mL), whereas the n-butanol fraction showed a slight improvement of the fungicide effect against B. cinerea (% GI 39.18 at 1 mg/mL). Moreover, the ethanolic extract exhibited a strong antifeedant activity, with a refusal rate (FR) higher than 90% in both choice and non-choice assays against C. chalcites, while the ethyl acetate and n-butanol fractions behaved as appetite suppressors. Objective: To investigate the antinociceptive effect of tingenone on inflammatory pain, as well as and the involvement of the cannabinoid receptors type 2 (CB2) and spinal microglia in this process. Methods: Male Swiss mice were subjected to inflammatory pain induced by intraplantar injection of carrageenan. The nociceptive threshold was measured by von Frey filaments test. Tingenone was administered orally 60 min before carrageenan injection. To evaluate the involvement of CB2 receptor, endocannabinoids, and microglia, AM630 (a CB2 receptor antagonist), MAFP (an inhibitor of an enzyme that hydrolyses endocannabinoids), and minocycline (a microglial inhibitor) were given intrathecally 20 min before tingenone administration. In addition, an immunofluorescence assay was used to evaluate CB2 receptor and CD11B (a microglial marker) expression in the spinal cord dorsal horn. Results: Tingenone significantly reduced carrageenan-induced hyperalgesia, which was reversed by pretreatment with AM630. MAFP and minocycline potentiated and prolonged the tingenoneinduced antinociception. CD11B expression was increased in the spinal cord dorsal horn of mice with inflammatory pain pretreated with tingenone, which was reduced by AM630, MAFP, and minocycline. Friedelan-3-one (1) and friedelane-3,16-dione (2) isolated from leaves and branches of Maytenus robusta Reissek were subjected to structural modifications via nucleophilic addition to the carbonyl group and Baeyer-Villiger oxidation in order to synthesize potential cytotoxic compounds. The oximes friedelane-3-hydroxyimino (3) and 3-hydroxyiminofriedelan-16-one (4) together with the lactones friedelane-3,4-lactone (5) and 3,4-lactonefriedelan-16-one (6) were characterized by IR and NMR spectroscopic analyses. Compounds 4 and 6 are reported for the first time. Cytotoxic screening via MTT assay in human leukemia cell lines (THP-1 and K562) demonstrated no significant improvement of compounds 3-6 when compared to the starting materials. Only compounds 3 and 5 demonstrated an improvement against K562 cells. However, the same assay on ovarian and breast cancer cell lines (TOV-21G and MDA-MB-231) showed a reduction in the IC50 for compounds 4-6, indicating that ring A modifications may enhance the biological potential. Cyanobacteria proliferation represents a problem to water treatment for human consumption supply. Species of the genus Microcystis, commonly found in continental waters, can bloom and produce microcystine and other toxic compounds associated to human and animal poisoning, depending on the nutrient levels present in the water. The use of natural products to control cyanobacteria blooms is promising since they are less harmful to the environment than compounds obtained by organic syntheses. Triterpenes, which are associated with several biological activities, have been isolated from species of the genus Maytenus (Celastraceae), commonly found in Cerrado regions of Brazil. Tingenone is a pentacyclic triterpene that has cytotoxic properties and induces growth inhibition in some microorganisms. In this study, the effects of tingenone isolated from Maytenus gonoclada at distinct concentrations (e.g., 50, 150, 500, 1,500, and 4,500 μg  L −1) on the growth rates of Microcystis novacekii was investigated. The algicide activity was verified using M. novacekii cultures growing in ASM1 culture medium at 23 °C and continuous illumination for 96 h. Growth inhibition was monitored by light microscopy and optical density (OD 680 nm). The median effective concentration associated to the M. novacekii inhibition growth induced by tingenone was 12.2 μg  L −1. The result indicates that tingenone has algicide effect and can be potentially applied in water management for public supply, replacing synthetic algicides. [ABSTRACT FROM AUTHOR]

In continuation of our work on Maytenus salicifolia, we report herein the isolation and structural elucidation of two new compounds, salicassin ( 1), a diterpene-chalcone adduct with an unprecedented C-framework, and (16 β)-16-hydroxypristimerin ( 2), a quinone methide triterpenoid. Their structures were elucidated on the basis of spectroscopic analysis, including 1D- and 2D-NMR techniques (COSY, ROESY, HSQC, and HMBC). In addition, 22 known compounds were isolated and characterized by comparison of their spectra with reported data. Compound 2, structurally related to the well known cytotoxic quinone methide triterpenoids, exhibited an antiproliferative effect on HeLa, A-549, and HL-60 human cell lines, with IC50 values of 2.2, 3.2, and 2.7 μ M, respectively. Leaves of Maytenus robusta (Celastraceae) were subjected to phytochemical investigation mainly directed at the isolation of pentacyclic triterpenes. The compounds friedelin (1), β-friedelinol (2), 3-oxo-21β-H-hop-22(29)-ene (7), 3,4-seco-friedelan-3,11β- olide (8), 3β-hydroxy-21β-H-hop-22(29)-ene (9), 3,4-seco-21β-H-hop-22(29)-en-3-oic acid (10), 3,4-seco-friedelan-3-oic acid (11), and sitosterol were identified in the hexane extract of M. robusta leaves. Compounds 8 and 9 are described herein for the first time. The structure and stereochemistry of both compounds were experimentally established by IR, HRLC-MS, and 1D (1H, 13C, and DEPT 135) and 2D (HSQC, HMBC and COSY) NMR data and supported by correlations with carbon chemical shifts calculated using the DFT method (BLYP/6-31G" level). Compounds 7 and 10 are also described for the first time, and their chemical structures were established by comparison with NMR data of similar structures described in the literature and correlations with BLYP/6-31G" calculated carbon chemical shifts. Compound 9, a mixture of 11 and sitosterol, and 3β,11β-dihydroxyfriedelane (4) were evaluated by the Ellman's method and all these compounds showed acethylcholinesterase inhibitory properties. A new pentacyclic triterpene, 3β,11β-dihydroxyfriedelane was isolated from leaves of Maytenus robusta Reissek (Celastraceae) together with other four known compounds: 3-oxofriedelane (friedelin), 3β-hydroxyfriedelane, 3-oxo-29-hydroxyfriedelane and 3-oxo-11β-hydroxyfriedelane. A new friedelane type triterpene, 3,16-dioxo-12α-hydroxyfriedelane was isolated from the chloroform extract of branches of Maytenus gonoclada Martius (Celastraceae) together with nine known compounds: 3,11-dioxofriedelane, 3,16-dioxofriedelane, 3,12-dioxofriedelane, 3-oxo-12α,29-dihydroxyfriedelane, 3-oxofriedelane (friedelin), 3β-hydroxyfriedelane, 3-oxo-12α-hydroxyfriedelane and 3β-hydroxylup-20(29)-ene (lupeol) and a mixture of long chain hydrocarbons. The structure and stereochemistry of the new friedelane triterpene were established from the 1H NMR and 13C NMR spectra including two-dimensional experiments (HSQC, HMBC and NOESY) and high resolution mass spectrometry. Diarrhea is an infectious disease caused by bacterial, virus, or protozoan, and dengue is caused by virus, included among the neglected diseases in several underdeveloped and developing countries, with an urgent demand for new drugs. Considering the antidiarrheal potential of species of Maytenus genus, a phytochemical investigation followed by antibacterial activity test with extracts of branches and heartwood and bark of roots from Maytenus gonoclada were conducted. Moreover, due the frequency of isolation of lupeol from Maytenus genus the antiviral activity against Dengue virus and cytotoxicity of lupeol and its complex with β-cyclodextrins were also tested. The results indicated the bioactivity of ethyl acetate extract from branches and ethanol extract from heartwood of roots of M. gonoclada against diarrheagenic bacteria. The lupeol showed potent activity against Dengue virus and low cytotoxicity in LLC-MK2 cells, but its complex with β-cyclodextrin was inactive. Considering the importance of novel and selective antiviral drug candidates the results seem to be promising.

The phytochemical study of hexane, chloroform and methanol extracts from leaves of Psychotria viridis resulted in the identification of: the pentacyclic triterpenes, ursolic and oleanolic acid; the steroids, 24-methylene-cycloartanol, stigmasterol and β-sitosterol; the glycosylated steroids 3-O-β-D-glucosyl-β-sitosterol and 3-O-β-D-glucosyl-stigmasterol; a polyunsaturated triterpene, squalene; the esters of glycerol, 1-palmitoylglycerol and triacylglycerol; a mixture of long chain hydrocarbons; the aldehyde nonacosanal; the long chain fat acids hentriacontanoic, hexadecanoic and heptadenoic acid; the ester methyl heptadecanoate; the 4-methyl-epi-quinate and two indole alkaloids, N,N-dimethyltryptamine (DMT) and N-methyltryptamine. The chemical structures were determined by means of spectroscopic (IR, 1H and 13C NMR, HSQC, HMBC and NOESY) and spectrometric (CG-MS and LCMS-ESI-ITTOF) methods. The study of biologic properties of P. viridis consisted in the evaluation of the acetylcholinesterase inhibition and cytotoxic activities. The hexane, chloroform, ethyl acetate and methanol extracts, the substances 24-methylene-cycloartanol, DMT and a mixture of 3-O-β-D-glucosyl-β-sitosterol and 3-O-β-D-glucosyl-stigmasterol showed cholinesterase inhibiting activity. This activity induced by chloroform and ethyl acetate extracts was higher than 90%. The methanol and ethyl acetate extracts inhibit the growth and/or induce the death of the tumor cells strains B16F10 and 4T1, without damaging the integrity of the normal cells BHK and CHO. DMT also demonstrated a marked activity against tumor cell strains B16F10 and 4T1.

Background: Aiming the continuity of the studies of Austroplenckia populnea, Brazilian species of the Celastraceae family, in the present study, it was investigated the effect of crude extracts obtained with ethanol, ethyl acetate and chloroform and two purified constituents, proanthocyanidin A and 4'-O-methylepigallocatechin, both isolated from its samaras, on cancer cell proliferation assays., Materials and Methods: The human cancer cells lines MCF-7 (ductal breast carcinoma), A549 (lung cancer), HS578T (ductal breast carcinoma) and non-cancer HEK293 (embryonic kidney cells) were treated with different concentrations of extracts and constituents and the effect was observed through the acid phosphatase method. The chemical structures of the purified compounds were identified by the respective IR and (1)H and (13)C nuclear magnetic resonance spectral data., Results: While crude extracts from samaras of the folk medicine A. populnea can trigger cell proliferative effects in human cell lines, the purified compounds (proanthocyanidin A and 4'-O-methyl-epigallocatechin) isolated from the same extracts can have an opposite (anti-proliferative) effect., Conclusion: Based on the results, it was possible to suggest that extracts from samaras of A. populnea should be further investigated for possible cancer-promoting activities; and the active extracts can also represent a source of compounds that have anti-cancer properties.
