The ligand design factors that may influence the isolation of metallosupramolecular helicates or mesocates still deserve to be investigated. In this sense, dinuclear nickel(II), copper(II) and zinc(II) compounds were obtained by electrochemical synthesis using a family of five Schiff base ligands, H2Ln (n = 1–5), derived from bisphenylmethane and functionalized with bulky tert-butyl groups in the periphery and ethyl groups in the spacer. Six of the new complexes were characterized by X-ray crystallography, thus demonstrating that the helicate structure is predominant in the solid state. 1H NMR studies were performed for the zinc complexes to analyze if the helical architecture of the metal complexes is retained in solution. These studies reveal that the presence of a tert-butyl group in the ortho position with respect to the OH group is an essential factor identified for the existence of a helicate conformation in solution. Manganosalen complexes are a class of catalytic antioxidants with beneficial effects against different neurological disorders according to various in vitro and in vivo studies. The interest in the factors that determine their antioxidant activity is based on the fact that they are key to achieving more efficient models. In this work, we report a set of new manganosalen complexes, thoroughly characterized in the solid state and in solution by different techniques. The chelating Schiff base ligands used were prepared from condensation of different substituted hydroxybenzaldehydes with 1,2-diaminoethane and 1,3-diaminopropane. The antioxidant activity of the new models was tested through superoxide dismutase and catalase probes in conjunction with the studies about their neuroprotective effects in human SH-SY5Y neuroblastoma cells in an oxidative stress model. The ability to scavenge excess reactive oxygen species (ROS) varied depending on the manganosalen models, which also yielded different improvements in cell survival. An assessment of the different factors that affect the oxidant activity for these complexes, and others previously reported, revealed the major influence of the structural factors versus the redox properties of the manganosalen complexes. In the present work, we report a neutral dinuclear copper(II) complex, [Cu2(L1)(OH)], derived from a new [N,O] donor Schiff base ligand L1 that was formed after the endogenous hydroxylation of an initial carbamate Schiff base H2L coordinated with copper ions in an electrochemical cell. The copper(II) complex has been fully characterized using different techniques, including X-ray diffraction. Direct current (DC) magnetic susceptibility measurements were also performed at variable temperatures, showing evidence of antiferromagnetic behavior. This review aims to analyze the emerging number of studies on biological media that describe the unexpected effects of different natural bioactive antioxidants. Hormetic effects, with a biphasic response depending on the dose, or activities that are apparently non-dose-dependent, have been described for compounds such as resveratrol, curcumin, ferulic acid or linoleic acid, among others. The analysis of the reported studies confirms the incidence of these types of effects, which should be taken into account by researchers, discarding initial interpretations of imprecise methodologies or measurements. The incidence of these types of effects should enhance research into the different mechanisms of action, particularly those studied in the field of basic research, that will help us understand the causes of these unusual behaviors, depending on the dose, such as the inactivation of the signaling pathways of the immune defense system. Antioxidative and anti-inflammatory activities in biological media should be addressed in ways that go beyond a mere statistical approach. In this work, some of the research pathways that may explain the understanding of these activities are revised, paying special attention to the ability of the selected bioactive compounds (curcumin, resveratrol, ferulic acid and linoleic acid) to form metal complexes and the activity of these complexes in biological media. The design of artificial helicoidal molecules derived from metal ions with biological properties is one of the objectives within metallosupramolecular chemistry. Herein, we report three zinc helicates derived from a family of bisthiosemicarbazone ligands with different terminal groups, Zn2(LMe)2∙2H2O 1, Zn2(LPh)2∙2H2O 2 and Zn2(LPhNO2)23, obtained by an electrochemical methodology. These helicates have been fully characterized by different techniques, including X-ray diffraction. Biological studies of the zinc(II) helicates such as toxicity assays with erythrocytes and interaction studies with proteins and oligonucleotides were performed, demonstrating in all cases low toxicity and an absence of covalent interaction with the proteins and oligonucleotides. The in vitro cytotoxicity of the helicates was tested against MCF-7 (human breast carcinoma), A2780 (human ovarian carcinoma cells), NCI-H460 (human lung carcinoma cells) and MRC-5 (normal human lung fibroblasts), comparing the IC50 values with cisplatin. The mechanism of carbamate activation promoted by different metal ions has been explored in this work. The reaction of the carbamate ligand H2L with chloride metal salts (M = Ni, Cu, Zn, Cd) leads to the coordination of the metal ions to the ligand, causing hydrolysis of the systems. This self-immolation process results in mononuclear dihydrazone complexes, carbon dioxide and the release of alcohol species from the pendant groups of the carbamate ligand. Featured Application: Kraft pulp bleaching and delignification. Lignin and other colored structures need to be bleached after the Kraft process in the pulp industry. Development of environmentally-safe bleaching catalysts or electrocatalysts constitutes an attractive strategy for selective removal of lignin. Seven manganese(III)-complexes with Schiff base ligands 1–7 were synthetized and characterized by different analytical and spectroscopic techniques. The tetragonally elongated octahedral geometry for the manganese coordination sphere and the global µ-aquo dimeric structure were revealed by X-ray diffraction (XRD) studies for 1, Mn2L12(H2O)2(N(CN)2)2 (N(CN)2 = dicyanamide). Complexes 1–4 behave as more efficient peroxidase mimics as compared to 5–7. The condensation of 3-methoxy-2-hydroxybenzaldehyde and the diamines 1,2-diphenylendiamine, 1,2-diamine-2-methylpropane and 1,3-propanediamine yielded the dianionic tetradentate Schiff base ligands N,N′-bis(2-hydroxy-4-methoxybenzylidene)-1,2-diphenylendiimine (H2L1), N,N′-bis(2-hydroxy-4-methoxybenzylidene)-1,2-diamino-2-methylpropane (H2L2) and N,N′-bis(2-hydroxy-4-methoxybenzylidene)-1,3-diaminopropane (H2L3) respectively. The organic compounds H2L1 and H2L2 have been characterized by elemental analysis, IR, 1H and 13C NMR spectroscopies and mass spectrometry electrospray (ES). The crystal structure of H2L2 in solid state, solved by X-ray crystallography, is highly conditioned in the solid state by two N-H•••N intramolecular interactions. The synthesis of three new manganese(III) complexes 1–3, incorporating these ligands, H2L1–H2L3, and dicyanamide (DCA), is reported. The complexes 1–3 have been physicochemically characterized by elemental analysis, IR and paramagnetic 1H NMR spectroscopy, ESI mass spectrometry, magnetic moment at room temperature and conductivity measurements. Complex 1 has been crystallographically characterized. The X-ray structure shows the self-assembly of the Mn(III)-Schiff base-DCA complex through µ-aquo bridges between neighbouring axial water molecules and also by π-π stacking interactions, establishing a dimeric structure. The manganese complexes were also tested as peroxidase mimics for the H2O2-mediated reaction with the water-soluble trap ABTS, showing complexes 1-2 relevant peroxidase activity in contrast with 3. The electrochemical synthesis has afforded the enantioselective formation of chirally pure cobalt helicates, and the ΛΛ isomer of a single enantiomer has been crystallized as only product for the cobalt methyl-substituted thiosemicarbazone helicate. Interestingly linkage isomers have been formed from zinc ethyl-substituted thiosemicarbazone helicate enantiomers for the first time. Four manganese(iii)-Schiff base complexes (1–4) of formula [MnLn(H2O)2]2(ClO4)2·mH2O (n = 1–4; m = 0, 1) have been prepared. The multidentate H2Ln Schiff base ligands consist of 3R,5R-substituted N,N′-bis(salicylidene)-1,2-diimino-2,2-dimethylethane, where R = OEt, OMe, Br or Cl. The complexes have been thoroughly characterized by elemental analysis, mass spectrometry, magnetic susceptibility measurements, IR, UV, paramagnetic 1H NMR and EPR spectroscopies. Other properties, including redox studies and molar conductivity measurements, have also been assessed. The crystal structure of 1 was solved by X-ray diffraction, which revealed the dimeric nature of the compound through μ-aqua bridges. The ability of these complexes to split water has been studied by water photolysis experiments, with the oxygen evolution measured in aqueous media in the presence of a hydrogen acceptor (p-benzoquinone), the reduction of which was followed by UV-spectroscopy. The discussion of the photolytic behaviour includes advances in the knowledge of the structural motifs and the chemical activity of this type of complex, as revealed by the development of several characterization techniques in the last decade. Manganosalen complexes are coordination compounds that possess a chelating salen-type ligand, a class of bis-Schiff bases obtained by condensation of salicylaldehyde and a diamine. They may act as catalytic antioxidants mimicking both the structure and the reactivity of the native antioxidant enzymes active site. Thus, manganosalen complexes have been shown to exhibit superoxide dismutase, catalase, and glutathione peroxidase activities, and they could potentially facilitate the scavenging of excess reactive oxygen species (ROS), thereby restoring the redox balance in damaged cells and organs. Initial catalytic studies compared the potency of these compounds as antioxidants in terms of rate constants of the chemical reactivity against ROS, giving catalytic values approaching and even exceeding that of the native antioxidative enzymes. Although most of these catalytic studies lack of biological relevance, subsequent in vitro studies have confirmed the efficiency of many manganosalen complexes in oxidative stress models. These synthetic catalytic scavengers, cheaper than natural antioxidants, have accordingly attracted intensive attention for the therapy of ROS-mediated injuries. The aim of this review is to focus on in vivo studies performed on manganosalen complexes and their activity on the treatment of several pathological disorders associated with oxidative damage. These disorders, ranging from the prevention of fetal malformations to the extension of lifespan, include neurodegenerative, inflammatory, and cardiovascular diseases; tissue injury; and other damages related to the liver, kidney, or lungs. Abstract: The first [5+5] coordination isomer of a zinc helical dimer derived from a pentadentate bisthiosemicarbazone Schiff base ligand was obtained by means of a new route. This compound was synthesised using a template procedure, spectroscopically characterised and recrystallised in a 1:1 v/v dichloromethane/acetonitrile mixture. The geometry showed by this compound is new for well-known dimer zinc complexes derived from pentadentate thiosemicarbazones and suggests that the experimental procedure could be an important factor in defining the stereochemistry of the metal atoms in such helical dimers. The electrochemical oxidation of a tin plate in the presence of an acetonitrile suspension of the ligand bis-(4-N-methylthiosemicarbazone)-2,6-diacetylpyridine, H4DAPTsz-Me, yields the complex Sn(H2DAPTsz-Me)(H2O)2 (1). Recrystallisation of 1 in a methanol/acetone solution leads us to isolate yellow crystals of [Sn(H2DAPTsz-Me)(CH3OH)2] · CH3OH (2), structurally characterised. To the best of our knowledge, 2 is the first neutral Sn(II) complex derived from a pentadentate thiosemicarbazone Schiff base ligand. This complex shows a distorted pentagonal bipyramidal geometry for Sn(II), being the metal surrounded by the N3S2 donor set of the bideprotonated ligand [H2DAPTsz-Me]2−. We have studied the factors that affect the formation of different metallosupramolecular architectures by metal direct self-assembly. A synthetic route has been developed to obtain mesocates or dihelicates selectively. For this purpose a series of five bisthiosemicarbazone ligands derived from 1,3-diacetylbenzene were designed and synthesised. Namely H(2)L(Me), H(2)L(Et), H(2)L(Ph), H(2)L(PhN) and H(2)L(PhOMe) with different substituents on the 4-N terminal position of the thiosemicarbazone strands. The Co(II), Zn(II) and Cd(II) complexes of these ligands were prepared by an electrochemical procedure. Crystallographic studies revealed the formation of mesocates for Co(II) and Zn(II) metal ions whereas the Cd(II) complexes gave a bishelical supramolecular structure. Therefore these bisthiosemicarbazone ligands enable the selective isolation of mesocate or dihelicate complexes by using a specific metal ion.

The coordinative chemistry of the tridentate half-salen ligand 5-(dimethylamino)-N-(2-((2-hydroxybenzylidene)amino)phenyl)naphthalene-1-sulfonamide (H(2)L, 1) has been studied by means of an electrochemical method. All of the complexes have been characterised using analytical and spectroscopic techniques. Ligand 1 and two nickel (6 and 7), copper (9), zinc (12) and cadmium (14) metal complexes have been studied by crystallography. Complexes 6 and 7 are octahedral and tetrahedral nickel(II) complexes, respectively, and both contain an [L](2-) molecule that behaves in an [N(2)O] tridentate manner. Nickel(II) completes its coordination kernel with three water molecules in complex 6, whereas in complex 7 the nickel ion is further bound to a molecule of dansylamine arising from a hydrolysis process. The copper(II) complex 9 is a monomeric compound that contains a bideprotonated ligand thread and a dimethylsulfoxide molecule coordinated through the sulfur atom. The zinc complex 12 is an unusual pentanuclear cluster compound whose structure consists of four anionic ligand units and two hydroxo anions bound to five zinc(II) centres. The appearance of the hydroxo anions in this complex provides new evidence for water reduction electrochemically promoted by zinc metal under mild conditions. The cadmium complex 14 is a dimeric compound that comprises two molecules of the anionic ligand and two dimethylsulfoxide molecules. The great structural variety exhibited by all these complexes demonstrates that the introduction of asymmetry in a salen skeleton by incorporating a dansyl pendant increases the versatility of the resulting ligand on coordination. All complexes are luminescent in solution at room temperature in acetonitrile solutions.

A novel neutral triple-stranded hexanuclear copper(I) cluster helicate [Cu(I)(6)L(3)]·2CH(3)CN derived from a thiosemicarbazone ligand could be synthesized and crystallographically characterized. The MALDI mass spectrum of this complex suggests that the tetranuclear copper(I) cluster helicate [Cu(I)(4)L(2)] is also present in solution. These copper(I) cluster helicates are capable, in the presence of O(2), of hydroxylating the arene linker of their supporting ligand strands. The resulting dinuclear complex [Cu(II)(2)L'(OH)] is formed by two copper(II) centers, a new ligand arising from the hydroxylation reaction, and one hydroxide group. The magnetic investigation of this compound shows a strong antiferromagnetic coupling between the two Cu(II) centers. The kinetic studies for the hydroxylation process show values of ΔH(≠)=-70 kJ mol(-1), similar to those mediated by the tyrosinase enzymes.

The coordinative chemistry of the tridentate thiosemicarbazone ligand 2-pyridinecarboxaldehyde 4-N-ethylthiosemicarbazone (HL(Et)) has been explored by using an electrochemical methodology. All the complexes have been characterized using analytical and spectroscopic techniques. In the case of copper we have isolated two different complexes, with Cu(L(Et))(2).4H(2)O (6) and [Cu(2)(L(Et))(2)(SO(4))] (7) formulae. The sulfate group coordinated to the copper atoms in was probably released to the media as a consequence of a desulfurization process. Single X-ray crystallography has been carried out for the ligand HL(Et) and the complexes , [Ag(6)(L(Et))(6)].CH(3)CN (9) and [Pb(L(Et))(2)] (2). The copper(ii) complex is a dimer compound in which two antiparallel monodeprotonated ligands are coordinated to two copper centres by establishment of mu(2)-thiolate bridges and the additional coordination of a sulfate group bridging the two metal atoms. The silver complex is an unusual hexanuclear cluster compound with a wheel-type conformation, while the lead complex is a monomer which exhibits the lone pair effect. A structural comparative study of the electrochemically obtained complexes derived from HL(X) ligands (X= Me, Et and Ph) have been performed. Finally, the solution behaviour of the complexes was checked by NMR, UV and fluorescence studies.

We have prepared some transition and post-transition metal complexes derived from the pentadentate thiosemicarbazone ligand bis(4-N-ethylthiosemicarbazone)-2,6-diacetylpyridine H(2)L(Et), by both chemical and electrochemical procedures. The complexes have been synthesised and fully characterised, including the crystal structures for the ligand H(2)L(Et) and its manganese, cadmium and lead complexes. We have also performed multinuclear (109)Ag, (113)Cd, (119)Sn and (207)Pb studies for silver, cadmium, tin and lead compounds, respectively. Moreover we present here a comparative study on the different structures found for pentadentate thiosemicarbazonate complexes, trying to check the influence of different factors, such as experimental procedure, size of metal, structure of the ligand, and metal oxidation state, on the final structure of the complex formed. Our aim is gaining a better insight into the coordination trends of pentadentate thiosemicarbazone ligands.

The influence of the metal size in the nuclearity of the complexes derived from the hydrazone ligand 2,6-bis(1-salicyloylhydrazonoethyl)pyridine [H(4)daps] has been investigated. We have synthesised a series of new complexes [M(H(x)daps)] x yH(2)O, (x = 2,3; y = 0-3) with M = Ag (1), Cd (2), Al (3), Sn (4) and Pb (6), using an electrochemical procedure. The crystal and molecular structures have been determined for the mononuclear complexes [Sn(H(2)daps)(H(2)O)(2)] x 4H(2)O (5) and [Pb(H(2)daps)(CN)][Et(4)N] (7). Complex is the first neutral Sn(II) complex derived from a pentadentate hydrazone Schiff base ligand. Complex shows the lead coordinated to the hydrazone donor set and a cyanide ligand, being the first reported complex with the lead atom coordinated to a monodentate cyanide group. Additionally, we have synthesised the lead complex using chemical conditions, in the presence of sodium cyanide which allowed us to isolate the neutral complex [Pb(H(2)daps)] (8). Evaporation of these mother liquors led the novel compound [Pb(Hdaphs)(CH(3)COO)] (9). Complex 9 shows the initial ligand hydrolysed in one of the imine bonds giving rise to a new tetradentate ligand [H(2)daphs] coordinated to the lead atom and a bidentate acetate group. Moreover, the solution behaviour of the complexes has been investigated by (1)H, (113)Cd, (117)Sn and (207)Pb NMR techniques. In particular multinuclear NMR has provided new useful data to correlate factors such as oxidation state, coordination number and nature of the kernel donor atoms due to the new coordination found in complexes 5 and 7. The comparative study of the structures of the complexes derived from this pentadentate [N(3)O(2)] hydrazone ligand let us to conclude that the metal size is a key factor to control the nuclearity of the complexes derived from the ligand [H(4)daps].

In this paper we describe the electrochemical synthesis and characterization of new neutral manganese, iron, cobalt, nickel, copper, zinc, cadmium and lead complexes with the ligand bis(4-N-methylthiosemicarbazone)-2,6-diacetylpyridine, H4DAPTsz-Me. X-Ray structures of [Mn(H2DAPTsz-Me)(EtOH)2] 1, [Pb(H2DAPTsz-Me)] 3 and [Zn(H2DAPTsz-Me)]2.EtOH.2H2O 4, were also determined. In these complexes the ligand behaves as bis-deprotonated and SNNNS pentadentate. In the manganese complex the metal is heptacoordinated, in a distorted pentagonal-bipyramidal environment, with the N3S2 donor set of the ligand in the pentagonal girdle and two solvent molecules occupying the axial positions. The bishelical zinc complex 4 shows each zinc atom with different coordination geometry, one octahedrally six-coordinate while the other is distorted tetrahedrally four-coordinate.
