Distiller's grains, byproducts of the brewing process, represent a valuable resource for extracting natural phenolic compounds due to their significant global production. This study presents the first evidence of the protective effects of Moutai distiller's grain polyphenol extract (MDGP) on dextran sulfate sodium (DSS)-induced colitis in mice. These protective effects manifest predominantly through the amelioration of general colitis indices and histopathological improvements. Utilizing liquid chromatography-high-resolution electrospray ionization mass spectrometry (LC-HR-ESI-MS), the main components of MDGP were identified as rutin, quercetin, naringenin, and dihydroquercetin. Moreover, a novel mechanism was elucidated by which rutin, the primary active component of MDGP, alleviates DSS-induced colitis. Assessment of intestinal barrier function, microbial sequencing, fecal transplantation, and antibiotic depletion experiments revealed that rutin suppresses the abundance of pathogenic bacteria (Helicobacter, Klebsiella, and Veillonella) while promoting the proliferation of beneficial bacteria (Ruminococcus_torques_group, Lachnoclostridium, and norank_f__Muribaculaceae). This modulation culminates in elevated butyric acid concentrations within short-chain fatty acids (SCFAs), amplified integrity of tight (ZO-1, occludin) and adherent (E-cadherin, β-catenin) junctional complexes, fortified intestinal barrier function, and diminished intestinal inflammation.This investigation accentuates the innovative therapeutic potential of MDGP and its main active component, rutin, in assuaging DSS-induced intestinal inflammation and fortifying the intestinal barrier through a mechanism predominantly mediated by the intestinal microbiota. Such insights potentially elevate the prominence of distiller's grains in the realm of functional food development.

Armillariella tabescens is a traditional Chinese medicinal and edible fungus which holds importance for treatment of gastrointestinal disorders. This study was aimed to evaluate the effect of methanol extract of A. tabescens (AME) in dextran sulfate sodium (DSS) induced ulcerative colitis (UC). The results showed that AME significantly attenuated UC symptoms, suppressed the activity of myeloperoxidase (MPO), decreased the levels of LPS, TNF-α, IL-1β, IL-18, and increased the level of IL-10 in serum. Besides, AME could improve the expression of tight junction proteins ZO-1, occludin and claudin-1. Moreover, AME inhibited the activation of TLR4/NF-κB and NLRP3 signaling pathways in colon tissue. In addition, eight compounds were identification in AME by high performance liquid chromatography (HPLC)-high resolution mass spectroscopy (HRMS). Therefore, this study indicated that AME was potential to be further developed into a suitable functional food additive or promising therapeutic agent for UC treatment.

A new peroxide of xanthone (1), together with five analogues (2–6), was obtained from horseradish peroxidase (HRP) catalysed biotransformation of α-mangostin. In order to increase the yield of compound 1, the environmental pH was succinctly adjusted, and as a result, compound 1 was formed with considerable selectivity. The anticancer potential of compound 1, a pentacyclic xanthone with a 1,2-dioxolane ring, was investigated due to its potent cytotoxic activity. The results showed that apoptosis induced by compound 1 in human hepatocellular carcinoma (HepG2) cell was associated with activation of caspase-dependent apoptotic pathway, the generation of reactive oxygen species (ROS) and the activation of c-Jun N-terminal kinases (JNK). In summary, compound 1 with a peroxide group was biosynthesized in considerable yield through pH-switched biotransformation catalysed by HRP, which could be further developed as a promising candidate in the treatment of liver cancer.

Genus Alpinia consists of over 250 species, which are widely distributed in south and southeast Asia. Many plants of genus Alpinia have been used for thousands of years to treat digestive system diseases and as anti-inflammatory drugs. Phytochemical research on this genus has led to the isolation of different kinds of diarylheptanoids, terpenes triterpenoids, phenylbutanoids, lignans, and flavonoids. Experimental evidences revealed that both the crude extracts and pure constituents isolated from the genus Alpinia exhibit a wide range of bioactivities such as anti-cancer, anti-oxidant, anti-bacterial, anti-viral, cardiovascular, and digestive system protective effects. Here, we summarize the phytochemistry and pharmacology investigation of the genus Alpinia, which can provide reference for further research and drug development.

Water spinach (I. aquatica) is one of the common green leafy vegetables used in Asia. In a continuing chemical investigation on the cytotoxic resin glycosides from water spinach, eight new resin glycosides, Aquaterins XII–XIX (1–8), were isolated, among which 1, 2 and 4 exhibited potent cytotoxic activities. To further characterise the anticancer potential of the resin glycosides from water spinach, Aquaterin II obtained previously was investigated on HepG2 cells. The results showed that Aquaterin II induced G0/G1 arrest regulated by related proteins CDK4/6, cyclin D/E and p21, and caused mitochondria-mediated apoptosis featured by MMP decrease, ROS accumulation, caspase cascade activation and Bax/Bcl-2 alteration. Additionally, the suppression on PI3K/Akt signalling pathway was observed, suggesting that it participated in the Aquaterin II-induced cell growth inhibition. Our results are the first to demonstrate the antiproliferative mechanism of resin glycosides. This investigation also advanced the potential application of water spinach as a functional food.

The bulbs of Lilium brownii var. viridulum (LB) are commonly used as both traditional Chinese medicines and popular functional food for many centuries in China. Previous studies reported that the extract of lily bulbs exhibited anti-inflammatory activity both in vivo and in vitro, but its active components and associated molecular mechanisms remain elusive. In the present study, using bioassay-guided isolation method, two phenylpropenoid acylglycerols, 1-O-feruloyl-2-O-p-coumaroylglycerol (1) and 1,3-O-diferuloylglycerol (2), were obtained and identified from the chloroform fraction of LB. Both compounds 1 and 2 significantly decreased the production of nitrite oxide (NO) in lipopolysaccharide (LPS)-stimulated mouse macrophage RAW264.7 cells in a dose-dependent manner with half maximal inhibitory concentration (IC50) values of 9.12 ± 0.72 μM and 12.01 ± 1.07 μM, respectively. They also inhibited the production of prostaglandin E2 (PGE2) and several other pro-inflammatory cytokines, such as interleukin-1β (IL-1β), interleukin-6 (IL-6), and tumor necrosis factor-α (TNF-α). Furthermore, compounds 1 and 2 downregulated the protein levels of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2). They also inhibited the nuclear translocation of nuclear factor-κB (NF-κB) p65 subunit and suppressed mitogen-activated protein kinases (MAPKs) pathway. Taken these data together, compounds 1 and 2 exhibited anti-inflammatory activities through acting on the NF-κB and MAPKs pathway. This research provides the first evidence on the major bioactive constituents and related molecular mechanisms of LB as an anti-inflammatory agent. Ten new sucrose esters, physakengoses A-J (1 - 10), were isolated from the aerial parts of Physalis alkekengi var. franchetii under the guidance of ¹H NMR spectroscopy. Their structures were determined by spectroscopic analyses (HRESIMS, 1D and 2D NMR, and ESIMS) and chemical methods. These new compounds were tested for antibacterial activities against Staphylococcus aureus, Bacillus subtilis, Pseudomonas aeruginosa and Escherichia coli. Among them, compounds 2 and 5-8 showed potent inhibitory effects against test strains with MIC values ranging from 3.5 to 14.9 μg/mL. The fingerprints of Rhizoma coptidis (Huanglian) collected in different growing periods were established by high-performance liquid chromatography (HPLC), and the seasonal variations of six alkaloids, namely berberine, palmatine, jateorrhizine, coptisine, epiberberine and columbamine were assessed. In order to highlight the differences of their HPLC fingerprints, three chemometrics methods including Spearman's rank correlation, hierarchical clustering analysis (HCA) and principal component analysis were applied. The anti-inflammatory activity of R. coptidis collected in different developing stages was measured by NO inhibition assay. The relationship between HPLC fingerprints and anti-inflammatory activity named spectrum- effect relationship was investigated by partial least squares regression. The anti-inflammatory activity was positively correlated with the alkaloid contents. Berberine, palmatine, coptisine, jateorrhizine and epiberberine were the main anti-inflammatory components on the basis of the quantitative determination of the six alkaloids and spectrum–effect investigation. A rare labdane diterpene γ-lactam, 1, and two new labdane diterpenoids, 2 and 3, along with two known diterpenes, 4 and 5, were isolated from the roots of Amomum maximum. Their structures were established on the basis of spectroscopic data. All of the isolates were evaluated for their cytotoxic activities, and compound 3 was found to exhibit an inhibitory activity against MG-63 cell lines with an IC50 value of 3.3±0.8 mg/ml. Peer-to-peer (P2P) networks have been adopted for Internet live video-streaming service, and several practical systems have been deployed in past years due to the inherent scalability and ease of deployment. However, most of these systems are commercial and proprietary, and hence little research was done in the area of characterizing practical system performance properties. In this article, we mainly present our experience on a practical P2P-based live video-streaming system called GridMedia, which was employed to broadcast live the Chinese Spring Festival Gala show over the Internet. Benefiting from two sets of flush-crowd traces with about 15,239 and 224,453 concurrent users in a 300 kb/s streaming session in 2005 and 2006, we perform a trace study to understand the service capacity, quality of streaming service, connection heterogeneity, user geographic distribution, and request and online duration characteristics. Aim: To investigate apparent diffusion coefficient (ADC) values as an indication of reconditioning of acute hepatic injury (AHI) after allogeneic mononuclear bone marrow cell (MBMC) transplantation., Methods: Three groups were used in our study: a cell transplantation group (n = 21), transplantation control group (n = 21) and normal control group (n = 10). AHI model rabbits in the cell transplantation group were injected with 5 mL of MBMC suspension at multiple sites in the liver and the transplantation controls were injected with 5 mL D-Hanks solution. At the end of the 1st, 2nd and 4th wk, 7 rabbits were randomly selected from the cell transplantation group and transplantation control group for magnetic resonance diffusion-weighted imaging (MR-DWI) and measurement of the mean ADC values of injured livers. After MR-DWI examination, the rabbits were sacrificed and the livers subjected to pathological examination. Ten healthy rabbits from the normal control group were used for MR-DWI examination and measurement of the mean ADC value of normal liver., Results: At all time points, the liver pathological scores from the cell transplantation group were significantly lower than those in the transplantation control group (27.14 ± 1.46 vs 69.29 ± 6.16, 22.29 ± 2.29 vs 57.00 ± 1.53, 19.00 ± 2.31 vs 51.86 ± 6.04, P = 0.000). The mean ADC values of the cell transplantation group were significantly higher than the transplantation control group ((1.07 ± 0.07) × 10⁻³ mm²/s vs (0.69 ± 0.05) × 10⁻³ mm²/s, (1.41 ± 0.04) × 10⁻³ mm²/s vs (0.84 ± 0.06) × 10⁻³ mm²/s, (1.68 ± 0.04) × 10⁻³ mm²/s vs (0.86 ± 0.04) × 10⁻³ mm²/s, P = 0.000). The pathological scores of the cell transplantation group and transplantation control group gradually decreased. However, their mean ADC values gradually increased to near that of the normal control. At the end of the 1st wk, the mean ADC values of the cell transplantation group and transplantation control group were significantly lower than those of the normal control group [(1.07 ± 0.07) × 10⁻³ mm²/s vs (1.76 ± 0.03) × 10⁻³ mm²/s, (0.69 ± 0.05) × 10⁻³ mm²/s vs (1.76 ± 0.03) × 10⁻³ mm²/s, P = 0.000]. At any 2 time points, the pathological scores and the mean ADC values of the cell transplantation group were significantly different (P = 0.000). At the end of the 1st wk, the pathological scores and the mean ADC values of the transplantation control group were significantly different from those at the end of the 2nd and 4th wk (P = 0.000). However, there was no significant difference between the 2nd and 4th wk (P = 0.073 and 0.473, respectively). The coefficient of correlation between the pathological score and the mean ADC value in the cell transplantation group was -0.883 (P = 0.000) and -0.762 (P = 0.000) in the transplantation control group., Conclusion: Tracking the longitudinally dynamic change in the mean ADC value of the AHI liver may reflect hepatic injury reconditioning after allogeneic MBMC transplantation.
