Changes in lighting accompany modern urbanization trends and can lead to various pathologies based on circadian disturbances. In this study, we assessed the changes in the circadian rhythm of core body temperature (Tcore) and locomotor activity of Wistar-Kyoto rats (WKY) and spontaneously hypertensive rats (SHR) following exposure to different lighting conditions: extended light phase of the day (16 h–8 h, 20 h–4 h, 24 h–0 h), light pollution, monochromatic light, and bright light therapy. The telemetry data was collected after experimental lighting conditions during periods with standard lighting (12 h of light and 12 h of darkness) and was processed using linear and cosinor analysis. The daily rhythms of rats’ parameters persisted in accordance with the standard lighting regime. Tcore changes were observed in both groups compared to the initial period: in WKY, a decrease in Tcore during the darkness and an increase during the light; in SHR, the opposite trend, with Tcore increased during the darkness and decreased during the light phase of the day. A relationship between Tcore and activity was observed with weak correlation. WKY exhibited more pronounced signs of adaptive variation and desynchronization compared to SHR, which could be associated with a wider range of functional capabilities of the organism without cardiovascular pathology.

Machine vision is a field of artificial intelligence that deals with processing images and videos using special algorithms. This allows devices to analyze visual information. Machine vision helps with tasks such as pattern recognition, image segmentation, object detection, and tracking. In microscopy, machine vision plays an important role, particularly in laser scanning microscopy (LSM). Laser scanning microscopy, laser scribing, and laser correction of the topology of semiconductor crystals are important technological processes in the production, control and adjustment of semiconductor crystals both on debug samples and as part of a test batch on a wafer. Laser exposure allows not only mechanically separate crystals, but also carry out more delicate and minimally invasive actions, in particular, adjustments to the values of thin-film resistors or burning of bridges necessary to adjust the circuit and disable unused blocks of the crystal. The paper lists the main parameters of the positioning system as part of the laser scanning setup, their impact on the quality of scanning and exposure to focused radiation at control points. The principles of machine vision algorithms when working with the image of the scanned object and the results of testing in the task of automated laser burning of bridges on a semiconductor wafer are described. Since the number of bridges can reach tens of thousands, and positioning systems have significant errors, machine vision allows you to correct the area and accuracy of laser exposure at any stage of scanning, which significantly improves the quality and efficiency of the process.

The development and widespread of high-tech microelectronic products impose increased requirements on the quality and reliability of microcircuits. The most effective methods for reliability improvement of electronic systems include diagnostic non-destructive testing (NDT) methods and selective destructive testing in special cases. Studies using visual inspection and electrical testing, consisting of functional and parametric testing, do not provide enough information to detect latent defects (for example, macro-defects in SiO 2 layers in CMOS chips) and to detect fakes and counterfeits. A fake integrated circuit (IC) may contain an undeclared malicious modification of the circuit, called hardware bugs. The common ICs studying tools are systems based on microfocus X-ray sources, scanning acoustic microscopes, optical and scanning electron microscopes, and X-ray fluorescence spectroscopes. Products destruction avoidance is a fundamental point, for example, for the technological process control in crystal manufacturing. Investigation of ICs using a light microscope is one of the most accessible and widespread method of microchip NDT. Semiconductor ICs structure scanning from the side of the device layer is limited by the shielding effect of metallization, since the metal is opaque for light. This limitation can be overcome by an alternative approach to microchip scanning based on irradiating the IC from the side of the substrate with laser sources in the near-IR range. This paper provides a brief overview of the major methods used in laser scanning microscopy to analyze the structures, responses, and features of the operating modes of semiconductor circuits. The main advantages and limitations in the use of optical methods are described, as well as what information about the product can be obtained as a result of laser scanning.

This article presents the prospect of using the process of deep hydrodesulfurization and hydrodearomatization of secondary gas oils using highly active NiMo catalysts to obtain hydrocarbon bases for drilling fluids. Catalysts were synthesized using PMo heteropolyanions, citric acid, and diethylene glycol on alumina carriers with different pore volumes. This study showed that the concentration of the impregnating solution affects the composition and morphology of the active phase particles of the prepared catalyst, while the textural characteristics of the carrier influence the physicochemical properties and catalytic activity of the NiMo/Al2O3 catalysts. The catalyst that was synthesized using a carrier with the largest pore volume and an effective diameter of more than 7 nm exhibited the highest activity. It was demonstrated that the use of such a catalyst allows for the procurement of hydrocarbon bases for drilling fluids from mixtures of secondary gas oils at a hydrogen pressure of 15–20 MPa. This study has practical significance for the development of sustainable and economically efficient methods for the utilization of low-quality petroleum gas oils to produce high-margin environmentally friendly non-fuel petroleum products, as well as contributes to the development of economically efficient technologies for the utilization of petroleum raw materials.

Background: The insulin receptor-related receptor (IRR) was originally discovered due to its high homology to the other family members (insulin receptor and insulin-like growth factor 1 receptor). We determined that IRR can be activated by mildly alkaline extracellular media and has typical features of the ligand-receptor interaction, including its specificity and dose-dependence. Since pH-sensitive properties of IRR are determined by its ectodomain; therefore, we chose as an option to study the soluble extracellular domain IRR. Methods: The investigation carried out in Titan Krios 60-300 TEM/STEM (FEI, USA) CryoEM, equipped with direct electron detector Falcon II (FEI, USA) and Cs image corrector (CEOS, Germany), at an accelerating voltage of 300 kV. Data processing and 3D reconstruction were carried out using computing resources of the Federal Collective Usage Center Complex for Simulation and Data Processing for Mega-Science Facilities at NRC “Kurchatov Institute.” Results: The obtained 2D classifications of particles of the ectodomain IRR at a neutral pH form several 3D models. This indicates that the ectodomain has several possible conformations, which is consistent with our previously obtained data using SEC-SAXS and AFM. In the future, additional careful data processing is required, as well as studies of the IRR ectodomain in mildly alkaline pH. Conclusion: In this study, we presented the structural characteristics of the IRR ectodomain obtained by CryoEM. These results are an important step towards understanding the mechanism of functioning of the IRR.

YBa2Cu3Ox (YBCO) thin films were deposited on 14°–22° tilted-axes NdGaO3 (NGO) substrates with pulsed laser deposition. Two orientations of the film are possible due to the presence of symmetrically equivalent (110) and (1 − 1 0) facets on the substrate surface. At high deposition rates, the pseudo-a grains, (100)YBCO||(110)NGO, gradually overgrow the pseudo-c grains, (001)YBCO||(110)NGO, forming high-tilted (tilt angle of the c axis 69°–74°) YBCO films. The substrate tilt angles in the vicinity of 18.43° promote formation of high-tilted films as a result of graphoepitaxial seeding of the pseudo-a grains on the (110) facets on the substrate surface due to the matching of the c lattice constant of the film with the length of the (110) terraces on the substrate surface. The effect of graphoepitaxial seeding was observed for the substrate tilt angles in the range 16°–21°, probably due to undulating edges of steps on the substrate surface. An increase of the deposition rate above a threshold level of ~ 3 nm/s results in a change of the growth mode with the formation of films with smooth surface morphology. The probable reason is a transition from vapor phase epitaxy to vapor–liquid–solid crystallization mechanism. The tilted-axes YBCO films demonstrate superconducting transition temperature superior to standard samples fabricated on (110) NGO substrates. The width of the superconducting transition for tilted-axes films increases due to the suppression of the critical current in the direction normal to the tilt axis. [ABSTRACT FROM AUTHOR]

The determination of pH in live cells and tissues is of high importance in physiology and cell biology. In this report, we outline the process of the creation of SypHerExtra, a genetically encoded fluorescent sensor that is capable of measuring extracellular media pH in a mildly alkaline range. SypHerExtra is a protein created by fusing the previously described pH sensor SypHer3s with the neurexin transmembrane domain that targets its expression to the cytoplasmic membrane. We showed that with excitation at 445 nm, the fluorescence lifetime of both SypHer3s and SypHerExtra strongly depend on pH. Using FLIM microscopy in live eukaryotic cells, we demonstrated that SypHerExtra can be successfully used to determine extracellular pH, while SypHer3s can be applied to measure intracellular pH. Thus, these two sensors are suitable for quantitative measurements using the FLIM method, to determine intracellular and extracellular pH in a range from pH 7.5 to 9.5 in different biological systems.

Background: It was shown that when exposed to extreme environment Dps-DNA co-crystallization takes place in Escherichia coli as a protective mechanism. However, to this date most of the proposed models of co-crystals were not backed up with any direct methods. Recent developments in Cryo Electron Tomography allow to visualize co-crystals with high spacial resolution in nearly native state. Methods: The investigations were carried out in a Titan Krios 60-300 TEM/STEM (FEI, USA) CryoEM, equipped with direct electron detector Falcon II (FEI, USA) and Cs image corrector (CEOS, Germany), at accelerating voltage of 300 kV. Tilt series were collected automatically using FEI Tomography software in low-dose mode which allowed to minimize the radiation damage. Subtomogram averaging was performed in Relion2 software package. Data processing and 3D reconstruction were carried out using computing resources of the Federal Collective Usage Center Complex for Simulation and Data Processing for Mega-Science Facilities at NRC “Kurchatov Institute”. Results: In this study, different types of Dps-DNA co-crystals were observed in vitro. Structural characteristics of the co-crystals were revealed for the first time by Cryo Electron Tomography. Subtomogram averaging allowed the direct visualization of Dps and DNA molecules in co-crystals. It was shown that the first type of co-crystals exhibits lamellar morphology and is formed by Dps layers alternating with DNA strands. The co-crystals adopt central symmetric triclinic crystal structure. The second type adopts body-centered cubic crystal lattice with mutually perpendicular DNA strands packed between Dps molecules. We assume that co-crystals’ structure could be dependent on the parameters of the utilized Dps and DNA solutions. Conclusion: In this study, we established structural characteristics of different types of Dps-DNA co-crystals. The obtained results are a vital step towards understanding the mechanism of protective Dps-DNA co-crystallization process.
