Abstract Background In the current study, we evaluated whether atopic dermatitis (AD) affects the entire body rather than being limited to skin barrier damage and inflammation. We hypothesized that medium-term exposure of distant organs to systemic inflammatory cytokines in sub-chronic inflammatory skin diseases has detrimental effects on distant tissues. Results Our findings demonstrated the dysregulation of genes and pathways associated with inflammation and the skin barrier, as well as genes and pathways involved in muscle development that respond to chemicals or stress in muscle tissues, all of which were reversed by hydrocortisone (Hc) administration. The expression of Ces1d showed significant differences during disease onset and after treatment in both skin and skeletal muscle, suggesting that Ces1d is likely responsible for the alleviation of subchronic AD. Conclusions Using NC/Nga mice with AD-like symptoms, we compared the transcriptomes of the skeletal muscle (a tissue that is relatively distant from the skin) with those of the skin (the lesion induction site) before and after disease induction, after which Hc was administered. Although further study is needed to better understand the effects of Ces1d on AD, skeletal muscle was associated with AD pathogenesis, and AD-like symptoms appeared to affect the body in a systemic manner. Given the importance of evidence-based medicine and the development of precision medicine, our findings provide insights into the mechanisms of AD onset and progression.

This study aims to improve the microclimate conditions in a mechanically ventilated broiler house by proposing and evaluating a ventilation-control algorithm based on heat-energy balance analysis. The new algorithm is designed to optimise the ventilation-rate requirement and thereby improve control of the indoor temperature. The analysis of one year of operational data collected at the experimental farm indicates that the current ventilation-control system successfully maintained optimal indoor temperatures for 74% of the time. In contrast, the proposed algorithm has the potential to improve this number significantly (up to 92%). The new algorithm was implemented and evaluated at two broiler houses (control and experimental) starting from day 20 to day 34 during one rearing period under high-temperature conditions. The results confirm that the new algorithm effectively reduced indoor temperatures by 1.5–2 °C during the day, which reduces heat stress significantly. Even though cooling pad usage increased to about eight times, the reduction in tunnel fan usage (to about 52%) led to significant energy savings. Furthermore, broiler mortality was reduced by 16.5%, which means there is also potential for improved productivity. The proposed ventilation control algorithm can effectively enhance microclimate conditions and energy efficiency in broiler production, though longer-term studies are required to fully assess its impact on growth performance.

Maintaining the optimal microclimate in broiler houses is crucial for bird productivity, yet enabling efficient temperature control remains a significant challenge. This study developed and validated a computational fluid dynamics (CFD) model to predict temporal changes in indoor air temperature in response to variable ventilation operations in a commercial broiler house. The model accurately simulated air velocity and airflow distribution for different numbers of tunnel fans in operation, with air-velocity errors ranging from −0.22 to 0.32 m s−1. The predicted airflow rates through inlets and cooling pads showed good agreement with measured values with an accuracy of up to 108.1%. Additionally, the CFD model effectively predicted temperature dynamics, accounting for chicken heat production and ventilation effect. The model successfully predicted the longitudinal temperature gradients and their variations during ventilation cycles, validating its reliability through comparison with experimental data. This study also explored different variable inlet configurations to mitigate the temperature gradient. The variable inlet adjustment showed the potential to relieve the high temperatures but may reduce overall ventilation efficiency or intensify temperature gradients, which confirms the importance of optimising ventilation strategies. This CFD model provides a valuable tool for evaluating and improving ventilation systems and contributes to enhanced indoor microclimates and productivity in poultry houses.

Quantifying emission factors of ammonia and particulate matter (PM) in livestock production systems is crucial for assessing and mitigating the environmental impact of animal production and for ensuring industry sustainability. This study aimed to determine emission factors of ammonia, total suspended particles (TSPs), PM10, and PM2.5 for piglets and growing–finishing pigs at a commercial pig farm in Korea. It also sought to identify factors influencing these emission factors. The research found that the emission factors measured were generally lower than those currently used in Korea, but were consistent with findings from individual research studies in the literature. Seasonal variations were observed, with ammonia emissions peaking in spring and autumn, and PM emissions rising in summer. Correlation analyses indicated that the number of animals and their average age correlated positively with both ammonia and PM emission factors. Ventilation rate was also positively correlated with PM emissions. Future extended field measurements across diverse pig farms will offer deeper insights into the emission factors of pig farms in Korea, guiding the development of sustainable livestock management practices.

Efficient collection of airborne spray is crucial to reduce environmental contamination and ensure effective pesticide application in agriculture. This study explored the efficacy of passive spray drift samplers, focusing on string collectors for capturing airborne spray droplets. String collectors were assessed in laboratory experiments using a spray drift tunnel. A notable average recovery rate of 82% was observed when string collectors were examined immediately after pesticide capture. Collection efficiency was found to increase with wind speed. Of all the string collectors, string #5, a yarn type, demonstrated consistent collection efficiency, meeting the criteria for passive samplers. This includes effective droplet capture at low wind speeds, a high recovery rate of 93.31%, and suitability for field experiments. Field evaluations further underlined the efficiency of string #5, showcasing its ability to capture spray drift across a wider area and varied heights with less effort and manpower compared to traditional nylon screens.

Accurate ventilation control is crucial for maintaining a healthy and productive environment in research-specialized pig facilities. This study aimed to evaluate actual ventilation rates and ventilation efficiency by investigating different inlet and exhaust configurations. The research was conducted in two pig rooms, namely pig room A and pig room B, in the absence of animals and workers to focus solely on evaluating the ventilation system’s performance. Actual ventilation rates were measured using hood-type anemometers, and the local air change per hour was analyzed at various measurement points via tracer gas decay experiments. The results demonstrated that specific inlet and exhaust combinations, such as side inlet/chimney outlet and ceiling inlet/side outlet, exhibited higher ventilation rates. However, the measured ventilation rates were much lower than the manufacturer’s specifications. The side exhaust fan closer to the pig activity space demonstrated better ventilation effectiveness for the animals than the chimney exhaust fan. Additionally, the ceiling inlet exhibited superior air distribution and uniformity. Lower ventilation rates and higher infiltration ratios resulted in reduced ventilation efficiency, with the difference between pig and worker activity spaces being pronounced. This study emphasizes the importance of selecting optimal inlet and exhaust configurations to achieve efficient ventilation and create a healthy environment for both pigs and workers.

Pesticide application is essential for improving crop productivity; however, undesirable pesticide drift must be mitigated because of its adverse impacts on humans, the environment and ecosystems. The collection and accurate quantification of airborne droplets are key elements involved in identifying the spatial and temporal dispersion of off-target spray movement. Various types of passive and active collectors have been deployed to measure airborne spray drift; however, the collection efficiencies of only a few samplers have been verified. This study evaluated the collection efficiency of two airborne-spray-drift collectors using an experimental drift wind tunnel. The airborne spray drifts were quantified by a total organic carbon analyser and validated by comparison to measurements using liquid chromatography with tandem mass spectrometry. Computational fluid dynamics (CFD) simulations were used to explore the effects of droplet size and wind speed on the collection performance. It was found that nylon screens, passive samplers, captured 57.9–88.1% of the airborne spray drift. These results are considered reliable and are comparable to those found in the literature. Additionally, the CFD results demonstrated that the collection efficiency increased with droplet diameter. An increase in wind speed improved the collection efficiency of fine droplets (≤100 μm diameter); however, wind speed had no significant influence on the collection of coarse droplets. These measurements, alongside the aerodynamic approach adopted in this study, can provide a comprehensive understanding of the collection performance of nylon screens.

BACKGROUND: Low survival rate of transplanted cells compromises the efficacy of cell therapy. Hexokinase II (HKII) is known to have anti-apoptotic activity through its interaction with mitochondria. The objective was to identify miRNAs targeting HKII and investigate whether miRNA-mediated modulation of HKII could improve the survival of mesenchymal stem cells (MSCs) exposed to H2O2. The expression of HKII in MSCs exposed to H2O2 was evaluated, and HKII-targeting miRNA was screened based on miRNA-target prediction databases. The effect of H2O2 on the expression of the selected HKII-targeting miRNA was examined and the effect of modulation of the selected HKII-targeting miRNA using anti-miRNA on H2O2-induced apoptosis of MSC was evaluated. RESULTS: H2O2 (600 μΜ) induced cell death of MSCs and decreased mitochondrial HKII expression. We have identified miR-181a as a HKII-targeting miRNA and H2O2 increased the expression of miR-181a in MSCs. Delivery of anti-miR-181a, which neutralizes endogenous miR-181a, significantly attenuated H2O2-induced decrease of HKII expression and disruption of mitochondrial membrane potential, improving the survival of MSCs exposed to H2O2. CONCLUSIONS: These findings suggest that H2O2-induced up-regulation of miR-181a contributes to the cell death of MSCs by down-regulating HKII. Neutralizing miR-181a can be an effective way to prime MSCs for transplantation into ischemic tissues.

Mesenchymal stem cells (MSCs) have the potential to repair and regenerate ischemic heart tissue; however, the poor viability of transplanted MSCs in the ischemic region is a major obstacle to their therapeutic use. This cell death is caused by Fas and Fas ligand (FasL) interactions under harsh conditions. To investigate improving the survival and therapeutic effects of MSCs, we focused our research on Fas–FasL-mediated cell death. In this study, we found that the poor viability of transplanted MSCs was caused by Fas–FasL interactions between host ischemic myocardial cells and implanted MSCs. In addition, we found that increased Fas expression and the corresponding decrease of cell survival were in close relation to hypoxic MSCs treated with FasL and H 2 O 2 . When MSCs were treated with a recombinant Fas/Fc chimera (Fas/Fc) inhibiting Fas–FasL interactions, the expressions of proapoptotic proteins including caspase-8, caspase-3, Bax, and cytochrome-c were attenuated, and the survival of MSCs was recovered. In ischemia–reperfusion injury models, the interaction between FasL in ischemic heart and Fas in implanted MSCs caused a loss of transplanted MSCs, whereas the inhibition of this interaction by Fas/Fc treatment improved cell survival and restored heart function. Thus, our study suggests that Fas–FasL interactions are responsible for activating cell death signaling in implanted stem cells and could be a potential target for improving therapeutic efficacy of stem cells in treating ischemic heart diseases.

Emerging evidence suggests that cell therapy with mesenchymal stem cells (MSCs) has beneficial effects on the injured heart. However, the decreased survival and/or adhesion of MSCs under ischemic conditions limits the application of cell transplantation as a therapeutic modality. We investigated a potential method of increasing the adhesion ability of MSCs to improve their efficacy in the ischemic heart. Treatment of MSCs with PKC activator, phorbol 12-myristate 13-acetate (PMA), increased cell adhesion and spreading in a dose-dependent method and significantly decreased detachment. When MSCs were treated with PKC inhibitor, that is, rottlerin, adhesion of MSCs was slightly diminished, and detachment was also decreased compared to the treatment with PMA. MSCs treated with both PMA and rottlerin behaved similarly to normal controls. In 3D matrix cardiogel, treatment with PMA increased the number of MSCs compared to the control group and MSCs treated with rottlerin. Expressions of focal adhesion kinase, cytoskeleton-associated proteins, and integrin subunits were clearly demonstrated in PMA-treated MSCs by immunoblotting and/or immunocytochemistry. The effect of PKC activator treatment on MSCs was validated in vivo. Following injection into rat hearts, the PMA-treated MSCs exhibited significantly higher retention in infarcted myocardium compared to the MSC group. Infarct size, fibrosis area, and apoptotic cells were reduced, and cardiac function was improved in rat hearts injected with PMA-treated MSCs compared to sham and/or MSC-implanted group. These results indicate that PKC activator is a potential target for niche manipulation to enhance adhesion of MSCs for cardiac regeneration.

Background: Fibrous dysplasia (FD) is a localized bone disorder in which fibro-osseous tissue replaces the normal bone structure. Patients with craniofacial FD often present with gradual swelling, deformity, and compromised vision or hearing. We previously introduced "the core extirpation method," a novel surgical technique that is minimally invasive like traditional bone shaving but has longer-lasting effects. This study presents the long-term outcomes of our core extirpation method. Methods: We conducted a retrospective analysis of patients who underwent core extirpation for FD of the zygomaticomaxillary region from 2012 through 2021. Computed tomography (CT) scans were performed 6 to 12 months before the operation, immediately before and after the operation, and during follow-up visits. We performed all operations using the upper gingivobuccal approach, and we extirpated the core of the lesion while preserving the cortical structures of the zygoma and the maxilla to maintain symmetrical facial contour. Results: In 12 patients with lesions in the growth phase, anteroposterior/mediolateral (AP/ML) length discrepancies and the volume increased between preoperative and immediate postoperative CT scans. All patients' immediate postoperative AP/ML discrepancies were stable up to 12-17 months postoperatively. Postoperative volume showed continuous lesion growth; the median volume growth rate was 0.61 cc per month. Conclusion: In this article, we present our experiences managing FD using the minimally invasive core extirpation technique, which entails small expected blood loss and can be performed as day surgery. It provides similar cosmetic outcomes as traditional bone shaving but with longer-lasting results. Although there are some limitations with the study's retrospective nature and small sample size, our 4-year follow- up results show promising results of the core extirpation method in well-indicated patients. Purpose: Epidermoid cysts are common benign skin neoplasms derived from the pilosebaceous apparatus that usually develop in hair-bearing regions such as the head and neck. Epidermal cysts rarely occur in the extremities, especially in the palmoplantar region. Therefore, they can be easily misdiagnosed as warts or calluses. Here, we present our experience treating epidermal cysts in the extremities, including a very rare case of malignant transformation into squamous cell carcinoma. Methods: This retrospective study enrolled all patients who underwent excision of epidermoid cysts in the upper and lower extremities from March 2006 to April 2021. Results: Among 249 patients, there were 10 (seven male and three female patients) who had epidermal cysts in the extremities (4.0%). All four plantar cysts were located in weight-bearing areas. One palmar epidermal cyst occurred 33 years after the trauma. There was one case of a highly recurrent epidermal cyst on the heel that was eventually diagnosed as squamous cell carcinoma arising from the cyst lining on excisional biopsy. After wide excision with a margin of 2 cm, the resulting defect was reconstructed using a free thoracodorsal artery perforator flap. Conclusion: Epidermoid cysts in the extremities, especially the palmoplantar region, are rare. Detailed history taking, including underlying diseases and trauma history, is helpful for diagnosis. Complete excision is necessary to avoid relapse and to confirm the final diagnosis, especially in cases suspected of malignant transformation arising from epidermoid cysts. [ABSTRACT FROM AUTHOR]

Purpose: We prospectively assessed the early radiographic and clinical outcomes (minimum follow-up of 2 years) of robot-assisted pedicle screw fixation (Robot-PSF) and conventional freehand pedicle screw fixation (Conv-PSF). Materials and Methods: Patients were randomly assigned to Robot-PSF (37 patients) or Conv-PSF (41 patients) for posterior interbody fusion surgery. The Robot-PSF group underwent minimally invasive pedicle screw fixation using a pre-planned robotguided screw trajectory. The Conv-PSF underwent screw fixation using the freehand technique. Radiographic adjacent segment degeneration (ASD) was measured on plain radiographs, and clinical outcomes were measured using visual analogue scale (VAS) and Oswestry disability index (ODI) scores regularly after surgery. Results: The two groups had similar values for radiographic ASD, including University California at Los Angeles grade, vertebral translation, angular motion, and loss of disc height (p=0.320). At final follow-up, both groups had experienced significant improvements in back VAS, leg VAS, and ODI scores after surgery (p<0.001), although inter-group differences were not significant for back VAS (p=0.876), leg VAS (p=0.429), and ODI scores (p=0.952). In the Conv-PSF group, revision surgery was required for two of the 25 patients (8%), compared to no patients in the Robot-PSF group. Conclusion: There were no significant differences in radiographic ASD and clinical outcomes between Robot-PSF and Conv-PSF. Thus, the advantages of robot-assisted surgery (accurate pedicle screw insertion and minimal facet joint violation) do not appear to be clinically significant. [ABSTRACT FROM AUTHOR]

The article discusses the use of an elasto-conductive epicardial mesh made for electromechanical cardioplasty. The mesh is designed to resemble cardiac tissue and confer the function of cardiac conduction system. It is noted capable in the detection of electrical signals on the heart of moving rat without impeding diastolic function following induced myocardial infarction and in the delivery of an electrical shock to end a ventricular tachyarrhythmia.

Introduction: Despite the success of interventional processes such as drug-eluting stents, complete prevention of restenosis is still hindered by impaired or delayed endothelialization or both. Here, we report that 1H-pyrrole-2,5-dione-based small molecule-generated mesenchymal stem cell-derived functional endothelial cells (MDFECs) facilitated rapid transmural coverage of injured blood vessels. Methods: Small molecules that induced CD31 expression were screened by principal component analysis (PCA). Rat mesenchymal stem cells (MSCs) were treated with selected small molecules for up to 16 days, and the expression levels of CD90 and CD31 were examined by immunocytochemistry. In vitro functional assays of MDFECs, including tube formation assays and nitric oxide production assays, were performed. After MDFECs (intravenous, 3×106 cells per animal) were injected into balloon-injured rats, neointima formation was monitored for up to 21 days. The endothelial coverage of denuded blood vessels was evaluated by Evans Blue staining. The functionality of repaired blood vessels was evaluated by measuring vasorelaxation and hemodynamic changes. Additionally, derivatives of the selected small molecules were examined for their ability to induce endothelial markers. Results: PCA indicated that 3-(2,4-dichlorophenyl)-4-(1-methyl-1H-indol-3-yl)-1H-pyrrole-2,5-dione effectively induced MDFECs. MDFECs inhibited the neointima formation of denuded blood vessels by facilitating more rapid endothelialization. Further examination indicated that derivatives with a 1H-pyrrole-2,5-dione moiety are important for initiating the endothelial cell differentiation of MSCs. Conclusions: Small molecules with 1H-pyrrole-2,5-dione as a core structure have great potential to improve the efficacy of MSC-based cell therapy for vascular diseases, such as atherosclerosis and restenosis. Introduction: Mesenchymal stem cells (MSCs) have therapeutic potential for the repair of myocardial injury. The efficacy of MSC therapy for myocardial regeneration mainly depends on the survival of cells after transplantation into the infarcted heart. In the transplanted regions, reactive oxygen species (ROS) can cause cell death, and this process depends on caspase activation and autophagosome formation. Methods: A Software TargetScan was utilized to search for microRNAs (miRNAs) that target caspase-3 mRNA. Six candidate miRNAs including let-7b were selected and transfected into human MSCs in vitro. Expression of MEK-EKR signal pathways and autophagy-related genes were detected. Using ischemia/reperfusion model (I/R), the effect of MSCs enriched with let-7b was determined after transplantation into infarcted heart area. Miller catheter was used to evaluate cardiac function. Results: Here, we report that let-7b targets caspase-3 to regulate apoptosis and autophagy in MSCs exposed to ROS. Let-7b-transfected MSCs (let-7b-MSCs) showed high expression of survival-related proteins, including p-MEK, p-ERK and Bcl-2, leading to a decrease in Annexin V/PI- and TUNEL-positive cells under ROS-rich conditions. Moreover, autophagy-related genes, including Atg5, Atg7, Atg12 and beclin-1, were significantly downregulated in let-7b-MSCs. Using a rat model of acute myocardial infarction, we found that intramyocardial injection of let-7b-MSCs markedly enhanced left ventricular (LV) function and microvessel density, in accordance with a reduced infarct size and the expression of caspase-3. Conclusions: Taken together, these data indicate that let-7b may protect MSCs implanted into infarcted myocardium from apoptosis and autophagy by directly targeting caspase-3 signaling. Despite the safety and feasibility of mesenchymal stem cell (MSC) therapy, an optimal cell type has not yet emerged in terms of electromechanical integration in infarcted myocardium. We found that poor to moderate survival benefits of MSC-implanted rats were caused by incomplete electromechanical integration induced by tissue heterogeneity between myocytes and engrafted MSCs in the infarcted myocardium. Here, we report the development of cardiogenic cells from rat MSCs activated by phorbol myristate acetate, a PKC activator, that exhibited high expressions of cardiac-specific markers and Ca2+ homeostasis-related proteins and showed adrenergic receptor signaling by norepinephrine. Histological analysis showed high connexin 43 coupling, few inflammatory cells, and low fibrotic markers in myocardium implanted with these phorbol myristate acetate-activated MSCs. Infarct hearts implanted with these cells exhibited restoration of conduction velocity through ciacreased tissue heterogeneity and improved myocardial contractility. The integrity of transplanted mesenchymal stem cells (MSCs) for cardiac regeneration is dependent on cell-cell or cell-matrix adhesion, which is inhibited by reactive oxygen species (ROS) generated in ischemic surroundings after myocardial infarction. Intracellular ROS play a key role in the regulation of cell adhesion, migration, and proliferation. This study was designed to investigate the role of ROS on MSC adhesion. In H2O2 treated MSCs, adhesion and spreading were inhibited and detachment was increased in a dose-dependent manner, and these effects were significantly rescued by co-treatment with the free radical scavenger, N-acetyl-L-cysteine (NAC, 1 mM). A similar pattern was observed on plates coated with different matrices such as fibronectin and cardiogel. Hydrogen peroxide treatment resulted in a marked decrease in the level of focal adhesion-related molecules, such as phospho-FAK and p-Src in MSCs. We also observed a significant decrease in the integrin-related adhesion molecules, αV and α1, in H2O2 treated MSCs. When injected into infarcted hearts, the adhesion of MSCs co-injected with NAC to the border region was significantly improved. Consequently, we observed that fibrosis and infarct size were reduced in MSC and NAC-injected rat hearts compared to in MSC-only injected hearts. These results indicate that ROS inhibit cellular adhesion of engrafted MSCs and provide evidence that the elimination of ROS might be a novel strategy for improving the survival of engrafted MSCs. A complete finite-element analysis method for corona discharge process, which is governed and coupled by charge transport equation and electric field equation, was presented. The charge transport equation of first order was transformed into second order by utilizing the artificial diffusion scheme. The two second-order equations were analyzed by the finite-element formulation which is well-developed for second-order equations. The Fowler-Nordheim injection boundary condition was adopted for charge transport equation. After verifying the numerical results by comparing to the analytic solutions using parallel plane electrodes with one carrier system, we extended the result to blade-plane electrodes in 2-D xy geometry with three carriers system. Radius of the sharp tip was taken to be 50 μm. When this sharp geometry was solved by utilizing the space discretizing methods, the very sharp tip was found to cause a singularity in electric field and space charge distribution around the tip. To avoid these numerical difficulties in the finite-element method, finer meshes, a higher-order shape function, and artificial diffusion scheme were employed. [ABSTRACT FROM AUTHOR]

Abstract Background Giant congenital melanocytic nevi (GCMN) are usually defined as nevi that exceed 20 cm in maximal diameter or 15% of the total body surface area. There have been reports of life-long malignant change risks arising from GCMN, leading to surgical excision of GCMN. This study aims to evaluate the thickness of melanocytes based on clinical factors in order to provide objective information for the complete resection of the lesion. Methods Overall, 75 patients diagnosed with GCMN between 2000 and 2021 were included, and their clinical records were collected retrospectively. 117 pathologic slides obtained during excision were reviewed to measure nevus thickness. Clinical factors were assessed with a generalized estimated equation model for association with nevus thickness. Abstract Despite the aid of tissue expansion, the ideal design of random pattern flap is not always available in patients with extensive skin lesions. We investigated the effectiveness of surgical delay on expanded flaps in pediatric patients. Retrospective cohort study was performed on patients who underwent tissue expansion surgery for extensive skin lesions at Seoul National University Children’s Hospital. The surgical delay technique was employed for patients with unfavorable flap conditions related to location or transposition angles. The dimensions of skin lesions and flaps were measured based on medical photographs. Dynamic mechanical analysis was performed to analyse the mechanical behaviours of the two materials at 37 °C and 80 °C, respectively. Furthermore, the shape memory property of the two materials was evaluated using a U-shape bending test, and the shape recovery ratio for 60 min at 37 °C was calculated. The results indicate that TC-85 can constantly apply a light force to the teeth when used for the 3D printed clear aligners, owing to its flexibility and viscoelastic properties. In addition, it is expected that the force decay induced by repeated insertion of the clear aligners will be reduced and a constant orthodontic force will be maintained. Furthermore, its geometric stability at high temperatures and the shape memory properties provide advantages for the clinical application.

Many edible mushrooms have been used by ancient cultures to increase human longevity, as well as for good foods, while being devoid of undesirable side effects. Presently, mushrooms are appreciated and consumed primarily for their nutritional properties; however they are also increasingly used in medicine. Therefore, the food industry is especially interested in cultivated and wild mushrooms. The nutritional value of mushrooms is attributed to high levels of protein, fiber, carbohydrates, vitamins, and minerals and low fat levels. As cardiovascular disease is a major cause of morbidity and mortality in our society, mushrooms are currently investigated for their therapeutic properties. In in vitro and in vivo investigations, mushrooms were shown to reduce risk factors of cardiovascular disease such as high total cholesterol and low-density lipoprotein (LDL) cholesterol, low high-density lipoprotein (HDL) cholesterol, high blood pressure, atherosclerosis, and oxidative and inflammatory damage. In this review, we summarize the current understanding of mushroom effects on cardiovascular disease, and discuss the therapeutic applications of mushrooms for heart disease. [ABSTRACT FROM AUTHOR]

Under distinct pathological heart conditions, the expression of a single miRNA can display completely opposite patterns. However, the mechanism underlying the bidirectional regulation of a single miRNA and the clinical implications of this regulation remain largely unknown. To address this issue, we examined the regulation of miR-1, one of the most abundant miRNAs in the heart, during cardiac hypertrophy and ischemia/reperfusion (I/R). Our data indicated that different magnitudes and chronicities of ROS levels in cardiomyocytes resulted in differential expression of miR-1, subsequently altering the expression of myocardin. In animal models, the administration of a miR-1 mimic attenuated cardiac hypertrophy by suppressing the transverse aortic constriction-induced increase in myocardin expression, whereas the administration of anti-miR-1 ameliorated I/R-induced cardiac apoptosis and deterioration of heart function. Our findings indicated that a pathologic stimulus such as ROS can bidirectionally alter the expression of miRNA to contribute to the development of pathological conditions exhibiting distinct phenotypes and that the meticulous adjustment of the pathological miRNA levels is required to improve clinical outcomes.
