Das Killertoxin K28 ist ein viral codiertes A/B-Toxin der Hefe Saccharomyces cerevisiae, das nach endozytotischer Aufnahme und retrogradem Transport zum ER gelangt. Nach Retrotranslokation äußert sich der letale Effekt in Abhängigkeit zur applizierten Konzentration entweder in einem G1/S-Zellzyklusarrest oder der Apoptose. Im Rahmen dieser Arbeit wurde gezeigt, dass die ER-Zytosol-Dislokation des Dimers durch das Chaperon BiP sowie die Isomerase-Aktivität der Protein-Disulfidisomerase und eine ausgeglichene Ca2+-Konzentration katalysiert wird. Die Überwindung der ER-Membran wird vermutlich durch Sec61p vermittelt und findet unabhängig von ERAD-Komponenten, UPR sowie Autophagozytose oder der Synthese von „lipid droplets“ (LD) statt. Die Expression der α Untereinheit führt ebenfalls zu einem G1/S-Arrest der Zielzelle. Auch hier werden Kar2p sowie eine intakte Ca2+-Homöostase zur effizienten Retrotranslokation in das Zytosol benötigt. Sec61p sowie Der1p des ERAD-L-Pathways bilden wahrscheinlich die potentielle Dislokationspore, wobei der ER-Export auch mit der LD-Synthese in Zusammenhang gebracht werden kann. Die gängigen zellulären Degradations-Prozesse wie ERAD, HiP und Autophagie tragen jedoch ausschließlich zum Abbau des Toxins bei. Die Wirkung der K28α-Untereinheit auf höhere Eukaryonten resultiert in der Induktion der Apoptose. UPS scheint im Säuger den ER-Export von K28α zu unterstützen, da die Substitution interner Lysinreste mit einer Reduktion der Toxizität einhergeht. The killer toxin K28 is a viral encoded A/B toxin of the yeast Saccharomyces cerevisiae which is taken up by endocytosis and is transported retrogradely to the ER. After retrotranslocation the lethal effect culminates either in a G1/S cell cycle arrest or in apoptosis depending on the toxin concentration. In this work it was shown that ER-cytosol dislocation of the α/β dimer is catalyzed by the chaperone BiP and the isomerase activity of protein disulfide isomerase Pdi1p and requires balanced intracellular Ca2+ levels. Passing the ER-membrane is assumed to be mediated by Sec61p and is independent of ERAD components, UPR, autophagy or the synthesis of "lipid droplets" (LDs). The expression of the plasmid-driven K28α-subunit also leads to a G1/S-arrest of the target cell. Kar2p and an intact Ca2+ homeostasis are also required for efficient retrotranslocation into the cytosol. As part of the ERAD-L pathway, Sec61p and Der1p are likely to form the potential dislocation pore whereupon ER export of K28α can also be associated with LD-synthesis. However, all major cellular degradation processes such as ERAD, HiP and autophagy contribute exclusively to the degradation of the toxin. The effect of K28α expression in higher eukaryotes results in the induction of apoptosis. In this case, the UPS seems to support ER export of K28α since the substitution of internal lysine residues is accompanied by a reduction of toxicity.

Das virale A/B-Toxin K28 aus Saccharomyces cerevisiae tötet sensitive Hefen in einem rezeptorvermittelten Prozess durch Induktion eines Zellzyklusarrestes. Nach endozytotischer Internalisierung durchläuft das Toxin den Sekretionsweg retrograd über Golgi und ER. In dieser Arbeit wurde der Mechanismus der ER/Cytosol-Retrotranslokation von K28 im Detail untersucht. Durch die Expression einer letalen K28alpha-Variante im ER-Lumen ausgewählter Deletionsmutanten konnte nachgewiesen werden, dass K28alpha nach Eintritt in den Sekretionsweg mit Hilfe von Erv29p und Bst1p in COPII-Vesikel verpackt und zum Golgi transportiert wird, wo die pro-Region durch die Aktivität der Endoprotease Kex2p entfernt wird. Dieser Schritt ist zwingend notwendig für die in vivo Toxizität von alpha. Im Gegensatz zu exogen appliziertem A/B-Toxin nutzt K28alpha den zellulären ERAD-Qualitätskontrollmechanismus, um aus dem ER in das Cytosol zu gelangen. Es konnte gezeigt werden, dass die Funktion des HRD1-Komplexes (im Unterschied zu CDC48) von essenzieller Bedeutung für die Toxizität von alpha ist. Wurde eine Polyubiquitinierung von K28alpha durch Austausch der internen Lysinreste verhindert oder eine Ubiquitinmutante, die keine Polyubiquitinketten mehr bilden kann, simultan überexprimiert, so kam es in beiden Fällen zu einem Verlust der Toxizität. Die Ubiquitinierung von K28alpha scheint somit für die Toxin-Retrotranslokation wichtig zu sein; ein direkter biochemischer Nachweis hierfür konnte bislang jedoch noch nicht erbracht werden. After endocytotic uptake the toxin passes the secretory pathway in reverse via Golgi and ER. In this study, toxin retrotranslocation from the ER into the cytosol has been investigated in detail by regulated expression of a lethal K28alpha variant in the ER of selected deletion mutants. By this means it became evident that K28alpha is packaged into COPII vesicles and transported to the Golgi by the help of Erv29p and Bst1p which normally recruit lumenal ERAD substrates. Within a late Golgi removal of the pro-region by endopeptidase Kex2p cleavage is crucial for the in vivo toxicity of K28alpha. In contrast to exogenously applied A/B toxin, K28alpha reaches the cytosol by abusing components of the ER quality control pathway ERAD. Sensitivity assays further indicated that HRD1 (in contrast to CDC48) complex components play a central role in K28alpha in vivo toxicity. Preventing polyubiquitination of K28alpha either by substitution of all internal lysine residues or by simultaneous overexpression of mutant ubiquitin, unable to form polyubiquitin chains, severely impaired in vivo toxicity. Thus, ubiquitination of K28alpha seems to be essential for its retrotranslocation, although a direct biochemical proof is still missing.
