A resposta imune exacerbada do hospedeiro desencadeia grande produção de citocinas inflamatórias, como IL-1β e TNFα, contribuindo para o aparecimento dos sintomas da doença. Durante a malária murina, embora haja ativação de caspase-11, a via não canônica de ativação do inflamassoma parece não contribuir para a patogênese da doença, enquanto a caspase-1 desempenha papel importante na produção de IL-1β. Nossos resultados mostram que há ativação de caspase-8 em monócitos do baço de camundongos infectados e a caspase-8 participa da produção de IL-1β e TNFα. Além disso, camundongos Ripk3/Casp8-/- são parcialmente resistentes ao modelo de infecção e desafio com LPS e à malária cerebral experimental, enquanto os camundongos Ripk3/Casp8/1/11-/- e Ripk3/Casp8/Gsdmd-/- são altamente em ambos os modelos. Esses resultados demonstram um papel complementar da caspase-8, porém independente à função do inflamassoma, para mediar a patogênese da malária. Além disso, pacientes infectados por P. falciparum ou P. vivax possuem GSDMD ativa e ativação de caspase-4 e -8 em monócitos. Assim, esse trabalho sugere a importância da caspase-8 durante a malária e seu potencial como possível alvo de intervenção terapêutica. Malaria is a disease caused by Plasmodium spp. parasites which affects millions of people every year. Exacerbated immune response by the host triggers a large production of inflammatory cytokines such as IL-1β and TNFα, contributing to the symptoms of the disease. During mouse malaria, although there is caspase-11 activation, the non-canonical pathway of inflammasome does not seem to contribute to the pathogenesis of the disease, whereas caspase-1 plays an important role on IL-1β production. Caspase-8 is cleaved in splenic monocytes from infected mice and caspase-8 participates in IL-1β and TNFα production. Moreover, Ripk3/Casp8-/- mice are partially resistant to infection followed by LPS challenge and experimental cerebral malaria and Ripk3/Casp8/1/11-/- e Ripk3/Casp8/Gsdmd-/- mice highly resistant in both models. These results demonstrate a complementary, but independent, role of caspase-8 and inflammasomes in mediating malaria pathogenesis. Besides, P. vivax and P. falciparum infected patients display active GSDMD, caspase-4 and caspase-8 in monocytes. In conclusion, the present work suggests the importance of caspase-8 during malaria and its potential role as a possible therapeutical target.

Pain is a complex and multifaceted experience that can affect the orofacial region. The temporomandibular joint (TMJ) is one of the structures that can be affected, resulting in a clinical condition of the disabling course, such as temporomandibular disorders (TMD). Pharmacotherapy has been used in the treatment of TMDs, however, due to the limited effectiveness and important adverse effects, it makes possible the search for new drugs. Goat milk is a food matrix of high nutritional value and with properties that have been evaluated. The objective of the study was to investigate the antinociceptive and antiinflammatory effect of the casein fraction of goat milk (CNF) and two hydrolysates, from its hydrolysis with pepsin (HDP) and trypsin (HDT), in a model of hypernociception induced by formalin in the TMJ of rats. For this purpose, male Wistar rats (180-240 g, n = 5) were pre-treated (per os) with CNF (1 mg / kg), HDP (1 mg / kg) and HDT (1 mg / kg). After 60 minutes, an intraarticular injection (a.i.) of saline solution (50 μL, 0.9%) was applied in the control group or formalin (50 μL, 1.5%) in the left TMJ. The nociceptive response was measured by quantifying, in seconds, the act of the mouse scratching the left TMJ region and by the number of times to raise the head reflexively over the 45 minutes period. Then, the animals were perfused with general anesthesia, followed by the removal of the TMJ and the trig eminal ganglion (TG) for immunohistochemical analysis by TMA for cytokines TNFα, IL -1β and IL-10. In addition, we investigated the roles of hemeoxygenase-1 (HO-1) and nitric oxide (NO) pathways in the antinociceptive mechanism of action of hydrolysates. The results showed that the three samples studied showed antinociceptive effects by reducing the nociceptive behavioral response. HDP and HDT have an antinociceptive and anti-inflammatory effect independent of the HO -1 and NO pathways, but can reduce the cytokines TNFα, IL-1β and increase IL-10 in TMJ and TG. The HDP and HDT compounds, therefore, have antinociceptive and anti-inflammatory activity mediated by the cytokines TNFα, IL -1β and IL-10. A dor é uma experiência complexa e multifacetada que pode afetar a região orofacial. A articulação temporomandibular (ATM) é uma das estruturas que podem ser acometidas, resultando em condição clínica de curso incapacitante, as disfunções temporomandibulares (DTM). A farmacoterapia tem sido utilizada no tratamento das DTMs, porém, devido à efetividade limitada e efeitos adversos importantes, faz-se oportuno a busca por novos medicamentos. O leite caprino é uma matriz alimentar de alto valor nutritivo e com propriedades que vêm sendo avaliadas. O objetivo do estudo foi investigar o efeito antinociceptivo e anti-inflamatório da fração caseínica do leite caprino (FCN) e de dois hidrolisados, obtidos a partir da sua hidrólise com pepsina (HDP) e tripsina (HDT), em um modelo de hipernocicepção inflamatória induzida por formalina na ATM de ratos. Para tanto, ratos Wistar machos (180-240 g, n=5) foram pré-tratados (v.o.) com FCN (1 mg/kg), HDP (1 mg/kg) e HDT (1 mg/kg). Após 60 minutos, foi aplicada injeção intra-articular (i.a.) de solução salina (50 μL, 0,9%) no grupo controle ou formalina (50 μL, 1,5%) na ATM esquerda. A resposta nociceptiva foi mensurada pela quantificação, em segundos, do ato do rato coçar a região da ATM esquerda e pelo número de vezes de ergue r a cabeça de forma reflexiva por 45 minutos. Em seguida, os animais foram perfundidos com paraformaldeído sob anestesia, seguidos da retirada da ATM e do gânglio trigeminal (GT) para análise imunohistoquímica por TMA para as citocinas TNFα, IL-1β e IL-10. Além disso, investigaram-se os papeis das vias da hemeoxigenase-1 (HO-1) e do óxido nítrico (NO) no mecanismo de ação antinociceptivo dos hidrolisados. Os resultados mostraram que as três amostras estudadas apresentaram efeitos antinociceptivos através da redução da resposta comportamental nociceptiva. HDP e HDT mostraram efeito antinociceptivo e anti-inflamatório independente das vias da HO-1 e NO, porém foram capazes de reduzir as citocinas TNFα, IL-1β e aumentar IL-10 na ATM e no GT. Os compostos HDP e HDT apresentam, portanto, atividade antinociceptiva e anti-inflamatória mediadas pelas citocinas TNFα, IL -1β e IL-10.

Os organofosforados, como o Diclorvós (DDVP), são um dos mais utilizados devido à sua eficiência e relativa baixa toxicidade. O DDVP pode atuar como desregulador endócrino, contribuindo para o desenvolvimento de lesões na próstata. O câncer de próstata assumiu, nestes últimos anos, o primeiro lugar em incidência de neoplasias nos Estados Unidos, e no Brasil estima-se 66,12 casos novos a cada 100 mil homens, sendo o câncer mais comum entre os homens. O processo inflamatório predispõe os indivíduos a vários tipos de câncer, pois induz a produção de citocinas como o TNFα que está envolvida no desenvolvimento do câncer de próstata. A inflamação pode ser ativada pelos receptores Toll-Like que pode resultar na ativação de duas vias, NFκB e a JNK. A via de sinalização do NFκB desempenha papel importante nas respostas ao câncer, inflamação, estresse e diferenciação celular. Assim, o presente estudo teve como objetivo avaliar a influência do praguicida Diclorvós em concentrações permitidas na próstata ventral de ratos em situação controle e após indução química por MNU (N-metil-N-nitrosureia), sobre a inflamação mediada por TLR4. Foram utilizados 40 ratos da linhagem Fischer 344, com idade de 90 dias. Os ratos foram separados em quatro grupos experimentais: Sham (G1), Sham+DDVP (G2), MNU (G3), MNU+DDVP (G4). Foram feitas análises histopatológica e imunohistoquímica da próstata ventral dos ratos. Foram encontrados três tipos de lesões na próstata ventral de ratos: Displasia, Neoplasia Intraepitelial Prostática (PIN) e Neoplasia intraepitelial de alto grau (HGPIN).O TLR4 esteve presente nas células epiteliais luminais da próstata ventral de todos os grupos experimentais. Nos grupos G1, G2 e G3 houve marcação em todo citoplasma, sendo a marcação mais intensa na região do aparelho de Golgi e no grupo G4 essa marcação foi moderada na região do aparelho de Golgi e intensa na membrana apical das células. O TNFα foi detectado em todo citoplasma das células epiteliais luminais da próstata ventral de todos os grupos experimentais destacando-se intensamente na região apical e moderadamente na região do aparelho de Golgi. O NFkB foi imunolocalizado em todo citoplasma das células epiteliais luminais da próstata ventral em todos os grupos experimentais, sendo mais intenso na região apical. Os grupos G1 e G2 apresentaram imunorreatividade intensa na região apical e somente os grupos G3 e G4 apresentaram imunorreatividade no envoltório nuclear. Verificamos a expressão de NFkB no núcleo e a diminuição da expressão de TLR4 e TNFα principalmente na região do Golgi, bem como o aumento da expressão de TLR4 na membrana apical das células epiteliais luminais, que pode ser associado ao agravamento das lesões prostáticas, corroborando com o papel desses marcadores do processo inflamatório, nas etapas iniciais da carcinogênese. Assim, podemos concluir que o DDVP em concentrações permitidas altera a imunolocalização de marcadores moleculares da inflamação na próstata ventral de ratos. Além disso, o DDVP associado à indução química pode acentuar essas alterações dos marcadores. Brazil is the largest consumer of pesticides in the world. Organophosphates, such as Diclorvos (DDVP), are among the most used because of their efficiency and relatively low toxicity. DDVP can act as an endocrine disrupter, contributing to the development of prostate lesions. In recent years, prostate cancer has been the number one cancer incidence in the United States, and in Brazil it is estimated 66.12 new cases per 100,000 men, the most common cancer among men. The inflammatory process predisposes individuals to various types of cancer, as it induces the production of cytokines such as TNFα, which is involved in the development of prostate cancer. Inflammation can be activated by Toll-Like receptors which can result in the activation of two pathways, NFκB and JNK. The NFκB signaling pathway plays an important role in cancer responses, inflammation, stress, and cell differentiation. Thus, the present study aimed to evaluate the influence of the Diclorvos pesticide at permissible concentrations in the ventral prostate of rats under control and after chemical induction by MNU (N-methyl-N-nitrosourea) on TLR4-mediated inflammation. 40 rats of the Fischer 344 strain, aged 90 days, were used. The rats were separated into four experimental groups: Sham (G1), Sham + DDVP (G2), MNU (G3), MNU + DDVP (G4). Histopathological and immunohistochemical analyzes of the ventral prostate of rats were performed. Three types of lesions were found in the ventral prostate of rats: Dysplasia, Prostatic Intraepithelial Neoplasia (PIN) and High Grade Intraepithelial Neoplasia (HGPIN). TLR4 presented in luminal epithelial cells of the ventral prostate of all experimental groups. In groups G1, G2 and G3 there was staining throughout the cytoplasm, being the most intense staining in the region of the Golgi apparatus. In group G4 this staining was moderate in the region of the Golgi apparatus and intense in the apical membrane of the cells. TNFα was detected in all cytoplasms of the luminal epithelial cells of the ventral prostate of all experimental groups, standing out intensely in the apical region and moderately in the region of the Golgi apparatus. NFκB was immunolocalized in all cytoplasm of luminal epithelial cells of the ventral prostate in all experimental groups, being more intense in the apical region. Groups G1 and G2 presented intense immunoreactivity in the apical region and only the G3 and G4 groups showed immunoreactivity in the nuclear envelope. We verified the expression of NFκB in the nucleus and decreased expression of TLR4 and TNFα mainly in the Golgi region, as well as the increase in TLR4 expression in the apical membrane of the luminal epithelial cells, which may be associated with worsening of the prostatic lesions, corroborating with the role of these markers of the inflammatory process, in the early stages of carcinogenesis. Thus, we can conclude that DDVP at permissible concentrations alters the immunolocalization of molecular markers of inflammation in the ventral prostate of rats. É acompanhada por alterações no sistema imune com variações nos níveis séricos das citocinas pró-inflamatórias. O tratamento disponível no Brasil é o benzonidazol, com baixa eficácia na fase crônica e efeitos colaterais importantes. Sabe-se que os adipocitos são reservatórios de T. cruzi, aspecto importante que é a relação com as citocinas pro-inflamatórias e com o aumento de T.cruzi nos adipocitos interferindo na evolução da doença. Sendo assim, o objetivo do presente estudo foi avaliar os efeitos da obesidade e das citocinas pro inflamatórias em camundongos balb/c machos infectados com T. cruzi tratados ou não com benzonidazol. Foram utilizados 120 animais, divididos em seis grupos, sendo 3 com dieta normal e 3 com dieta hipercalórica. Esses foram avaliados em cinco momentos: M1-1°dia, M2-3°dia, M3-5°dia, M4-7°dia e M5-8°dia. Para infecção utilizou a cepa aguda Y, com pico de parasitemia no sétimo dia (M4). A eutanásia foi realizada em cinco animais de cada grupo, em cada momento. Para determinação das citocinas utilizou o Kits de CBA –Cytometric Beads Array, através de citômetro de fluxo. Além disso, foi verificado o peso da gordura. O medicamento foi ofertado na dosagem de 100 mg/Kg/dia. A análise estatística foi realizada por medidas descritivas com o cálculo de média, desvio padrao, minimo, maximo e mediana, testes ANOVA, Tukey, Wald e Qui-quadrado. Dentre os resultados observa-se que houve aumento do peso relativo e de gordura corporal entre os consumiram dieta hipercalórica, classificando-os com obesidade. Além disso, pode-se verificar que as alterações metabólicas e inflamatórias estão mais elevadas nos grupos obesos, sendo eles infectados ou não, com ou sem tratamento. Portanto o estado nutricional tem uma grande influencia no processo infeccioso pelo T. cruzi. O presente estudo revelou que a obesidade desencadeia alterações inflamatórias detectadas pelas citocinas pro inflamatórias e metabólicas, e que a associação com o processo infeccioso agravou esse processo. O tratamento com benzonidazol contribuiu para uma resposta melhor com menos alterações metabólicas e inflamatórias Chagas disease, caused by Trypanosoma cruzi (T. cruzi), is a chronic infectious disease that has a high incidence in Brazil and, in recent years, has become endemic in urban centers. It is accompanied by changes in the immune system with variations in serum levels of pro-inflammatory cytokines. The available treatment in Brazil is benzonidazole, with low efficacy in the chronic phase and important side effects. It is known that the adipocytes are reservoirs of T. cruzi, an important aspect that is the relation with the pro-inflammatory cytokines and with the increase of T. cruzi in the adipocytes interfering in the evolution of the disease. Therefore, the objective of the present study was to evaluate the effects of obesity and proinflammatory cytokines in T. cruzi infected or non-treated male balb / c mice with benzonidazole. A total of 120 animals were used, divided into six groups: 3 with normal diet and 3 with hypercaloric diet. These were evaluated in five moments: M1-1 day, M2-3 day, M3-5 day, M4-7 day and M5-8 day. For infection, the acute Y strain was used, with a peak of parasitemia on the seventh day (M4). Euthanasia was performed in five animals from each group, at each moment. In order to determine the cytokines, the CBA-Cytometric Beads Array Kits were used by flow cytometer. In addition, the fat weight was checked. The drug was offered in the dosage of 100 mg / kg / day. Statistical analysis was performed by descriptive measures with mean, standard deviation, minimum, maximum and median, ANOVA, Tukey, Wald and Chi-square tests. Among the results it was observed that there was an increase in relative weight and body fat among those consuming a hypercaloric diet, classifying them as obese. In addition, it can be verified that the metabolic and inflammatory alterations are higher in the obese groups, being they infected or not, with or without treatment. Therefore, the nutritional status has a great influence on the T. cruzi infectious process. The present study revealed that obesity triggers inflammatory changes detected by pro-inflammatory and metabolic cytokines, and that association with the infectious process aggravated this process. Benzonidazole treatment contributed to a better response with fewer metabolic and inflammatory changes

O aumento de atividade nervosa simpática (ANS) destaca-se como um dos principais agravantes para as doenças relacionadas ao sistema cardiovascular, como a hipertensão arterial (HA). De fato, diversos grupos de pesquisa têm direcionado seus estudos para o entendimento de como alterações de regiões e núcleos localizados no sistema nervoso central (SNC), poderiam resultar na hiperatividade simpática e, consequente, desenvolvimento e manutenção da HA. Regiões de controle cardiovascular como o núcleo pré-óptico mediano (MnPO) e o núcleo paraventricular (PVN) do hipotálamo tem sido foco de importantes investigações sobre a participação do SNC na fisiopatologia da HA. No entanto, as vias de sinalização que possam modular os neurônios desses núcleos ainda precisam ser esclarecidas. Assim, a primeira parte deste estudo teve como objetivo investigar envolvimento das neurotransmissões glutamatérgica e angiotensinérgica do MnPO nos aumentos da atividade simpática e pressão arterial observados em ratos hipertensos. Para tanto, ratos espontaneamente hipertensos (SHR) e ratos submetidos previamente ao protocolo de Goldblatt (dois rins; um clipe; 2R1C) foram utilizados. Os ratos de ambos os grupos (250 a 350g, n = 6) foram anestesiados com uretano (1,2g / kg, i.v.) e instrumentalizados para registros de pressão arterial média (PAM), frequência cardíaca (FC) e atividade nervosa simpática renal (ANSR). Nanoinjeções (100nl) de salina (NaCl, 150 mM), Losartan (antagonista do receptor AT1; 10mM) e ácido quinurênico (antagonista dos receptores de glutamato; 50mM), foram realizadas no MnPO. Em ratos 2R1C, o bloqueio glutamatérgico promoveu diminuição da PAM e ANSR (-19,1 ± 0,9 mmHg, -21,6 ± 2,8 %, p < 0,05) quando comparado com as nanoinjeções de salina (-0,4 ± 0,6 mmHg, 0,2 ± 0,7 %, p < 0,05). A inibição angiotensinérgica também reduziu esses parâmetros (-11,5 ± 1,2 mmHg, -10,5 ± 1,0 %, p < 0,05) em 2R1C. Em SHR, as nanoinjeções de ácido quinurênico produziram quedas da PAM e ANSR (-21,0 ± 2,5 mmHg, -24,7 ± 2,4 %, p < 0,05), bem como nanoinjeções de Losartan (-9,7 ± 1,2 mmHg; p < 0,05) e ANSR (-12,0 ± 2,4 %, p < 0,05). Os resultados observados suportam a conclusão de que uma neurotransmissão tônica excitatória exercida pela angiotensina II, e, principalmente, pelo glutamato no MnPO, estão envolvidos na elevação da pressão arterial e ANSR observados em modelos de hipertensão genética ou secundária à estenose na artéria renal. Na segunda parte do presente estudo, nós testamos a hipótese de que a citocina pró-inflamatória (CIP) fator de necrose tumoral alfa (TNFα) poderia ativar os neurônios do PVN de forma aguda para promover aumento da ANS. Para tanto, ratos Sprague-Dawley (350-550g) foram anestesiados com uma mistura dos anestésicos α-cloralose (80 mg / kg) e uretano (800 mg / kg) e instrumentalizados para os registros de PAM, FC, ANS esplâncnica (ANSS) e nanoinjeções unilaterais no PVN. Inicialmente os animais passaram por séries de experimentos com os seguintes pré-tratamentos no PVN: veículo (PBS, 0,5 nmol / 50 nl, n = 5), NBQX (antagonista do receptor AMPA; 5,2 nmol / 50 nl, n = 5) e AP5 (antagonista de receptor NMDA; 24 nmol / 50 nl, n = 5). Após cada pré-tratamento foi realizada nanoinjeção de TNFα (0,6 pmol / 50 nl) e as variáveis cardiovasculares e simpáticas foram avaliadas por 60 min. As nanoinjeções de TNFα no PVN promoveram progressivo aumento da ANSS (55 ± 5,5 %) quando previamente tratado com veículo. No entanto, não foram observadas alterações na PAM (95 ± 8 mmHg vs. 94,5 ± 6,2 mmHg) e na FC (400,2 ± 13,5 bpm vs. 377 ± 9 bpm) neste grupo. Por outro lado, o pré-bloqueio dos receptores ionotrópicos de glutamato, AMPAR e NMDAR promoveram reduções na resposta simpatoexcitatória (24 ± 5 % e 32 ± 6,5 %, respectivamente) induzida pela nanoinjeção de TNFα no PVN. Nesses grupos também não foram observadas alterações nos parâmetros cardiovasculares (NBQX: 93 ± 9 mmHg e 429 ± 10 bpm; AP5: 104 ± 3 mmHg e 439 ± 29 bpm). Os experimentos seguintes buscaram então investigar o papel dos receptores NMDA e AMPA na manutenção da simpatoexcitação esplâncnica causado pela nanoinjeção de TNFα no PVN. Assim, foram realizadas nanoinjeções de veículo (PBS, 0,5 nmol / 50 nl, n = 3), NBQX (5,2 nmol / 50 nl, n = 5) e AP5 (24 nmol / 50 nl, n = 4) após 60 min da nanoinjeção de TNFα no PVN. Os parâmetros avaliados foram então registrados por 60 min adicionais. Os pós-tratamentos não promoveram alterações significativas em nenhuma das variáveis avaliadas, o que sugere que esses receptores não possuem importância na manutenção da simpatoexcitação desencadeada pela administração do TNFα no PVN. Finalmente, avaliamos se a inibição do PVN poderia influenciar na manutenção da simpatoexcitação induzida por TNFα. Para isto, foram realizadas nanoinjeções unilaterais do agonista dos receptores GABAA, muscimol (1 nmol / 50 nl, n=5) após 60 min das nanoinjeções de TNFα no PVN. A ativação dos receptores GABAA no PVN, reverteu a simpatoexcitação esplâncnica induzida por TNFα de 173 ± 11 % para 106 ± 5 %, (p < 0,05). Os dados encontrados no presente estudo indicam que o desenvolvimento do aumento da ANSS promovido pelo TNFα no PVN é dependente da ativação dos receptores ionotrópicos de glutamato, AMPAR e NMDAR neste núcleo. Em conjunto estes achados indicam a importância da neurotransmissão glutamatérgica no PVN para a geração da resposta simpatoexcitatória desencadeada pela neuro-inflamação induzida por (TNFα) neste núcleo. The increase of sympathetic nervous activity (SNA) stands out as the main aggravating factor for diseases related to the cardiovascular system, as well arterial hypertension (AH). In fact, several research groups have directed their studies to the understanding of how alterations of regions and nuclei located in the central nervous system (CNS) could result in sympathetic hyperactivity and, consequently, the development and maintenance of AH. Cardiovascular control regions such as the median preoptic nucleus (MnPO) and the paraventricular nucleus (PVN) of the hypothalamus have been the focus of important investigations on the involvement of the CNS in the physiopathology of AH. However, signaling pathways that can modulate the neurons of these nuclei still need to be clarified. Therefore, the first part of this study aimed to investigate the involvement of angiotensinergic and glutamatergic neurotransmissions in the MnPO on the sympathetic activity and blood pressure increases observed in hypertensive rats. Spontaneously hypertensive rats (SHR) and rats submitted previously to the Goldblatt protocol (two kidneys; one clip; 2K1C) were used. Rats of both groups (250 to 350g, n=6) were anesthetized with urethane (1.2g/kg,i.v.) and instrumented to record mean arterial pressure (MAP), heart rate (HR) and renal sympathetic nerve activity (RSNA). Nanoinjection (100nl) of saline (NaCl, 150 mM), Losartan (AT1 receptor antagonist; 10mM) and kynurenic acid (glutamate receptor antagonist; 50mM) into the MnPO were performed. In 2K1C rats, glutamatergic blockade promoted decreases in MAP and RSNA (-19.1 ± 0.9 mmHg, -21.6 ± 2.8 %, p< 0.05) when compared to saline (-0.4 ± 0.6 mmHg, 0.2 ± 0.7 %, p < 0.05). Angiotensinergic inhibition also reduced these parameters (-11.5 ± 1.2 mmHg, -10.5 ± 1.0 %, p < 0.05) in 2K1C. In SHR, Kynurenic acid nanoinjections produced hypotension and sympathoinhibition (-21.0 ± 2.5 mmHg, -24.7 ± 2.4 %, p < 0.05), as well Losartan nanoinjections (-9.7 ± 1.2 mmHg; p < 0.05) and RSNA (-12.0 ± 2.4 %, p < 0.05). These findings support the conclusion that a tonic excitatory neurotransmission exerted by angiotensin II, and mostly by glutamate in the MnPO are involved in the elevation of blood pressure and RSNA observed in models of genetic hypertension or secondary to renal artery stenosis. In the second part of this study, we tested the hypothesis that the pro-inflammatory cytokine (PIC) tumor necrosis factor alpha (TNFα) could acutely activate the PVN neurons to promote SNA increased. The animals were submitted to series of experiments with the following pretreatments in the PVN: vehicle (PBS, 0.5 nmol / 50 nl, n = 5), NBQX (AMPA receptor antagonist; 5.2 nmol / 50 nl n = 5) and AP5 (NMDA receptor antagonist; 24 nmol / 50 nl, n = 5). After each pretreatment, nanoinjection of TNFα (0.6 pmol / 50 nl) was performed and cardiovascular and sympathetic variables, evaluated for 60 min. Considerando a alta herdabilidade da doença, vários polimorfismos genéticos vêm sendo estudados e associados com o surgimento da DAC e, entre eles, destacam-se variantes nos genes CD14, TLR4, NFKB1 e TNFα, os quais regulam a via de sinalização celular do sistema imune inato e desencadeiam o processo inflamatório na DAC. Objetivo: O objetivo principal deste estudo é verificar a possível associação de polimorfismos nos genes CD14 (rs2569190), TLR4 (rs4986790 e rs4986791), NFKB1 (rs28362491) e TNFα (rs1800629 e rs361525) com a DAC. Metodologia: A amostra foi composta por 707 indivíduos adultos submetidos ao exame de cateterismo cardíaco no Hospital Bruno Born, de Lajeado, RS. Todos os indivíduos assinaram um termo de consentimento livre e esclarecido e responderam a um questionário semiestruturado. Os indivíduos foram classificados entre casos e controles, por um médico cardiologista, com base no seguinte critério: presença de estenose, com comprometimento maior do que 50%, em pelo menos uma das artérias coronárias. Foram também coletadas amostras de sangue periférico para análises bioquímicas e moleculares. A extração de DNA foi realizada pelo método de salting out. Os polimorfismos dos genes CD14, TLR4 e TNFα foram genotipados pelo sistema de discriminação alélica TaqMan, em equipamento de reação em cadeia da polimerase (PCR) em Tempo Real (StepOnePlus®). O polimorfismo rs28362491, no gene NFKB1, foi amplificado através da técnica convencional de PCR. Resultados: Identificamos uma associação dos polimorfismos rs2569190, localizado no gene CD14, e rs28362491, no gene NFKB1, com a DAC. Além disso, foram detectados efeitos dos polimorfismos dos genes TLR4 e TNFα nos níveis glicêmicos e dos polimorfismos nos genes TLR4, NFKB1 e TNFα no perfil lipídico. Conclusão: Nossos achados sugerem a participação de polimorfismos nos genes CD14 e NFKB1 no desenvolvimento da DAC na nossa amostra, corroborando evidências prévias do envolvimento de genes do processo inflamatório nessa patologia. Introduction: Coronary artery disease (CAD) is the main cause of morbidity and mortality in the world, being characterized as a chronic, multifactorial inflammatory disease, whose pathophysiology is atherosclerosis. Considering the high heritability of the disease, several genetic polymorphisms have been investigated and associated with CAD and, among them, there are variants in the CD14, TLR4, NFKB1 and TNFα genes, which regulate cell signaling pathways of the innate immune system and inflammatory process in CAD. Objective: The main objective of this study is to verify the association beteween polymorphisms in the CD14 (rs2569190), TLR4 (rs4986790 and rs4986791), NFKB1 (rs28362491) and TNFα (rs1800629 and rs361525) genes and CAD. Methods: The sample group was composed of 707 adult individuals, recruited at the time when they were bought in for coronary angiography procedures at the Hemodynamic Center of the Hospital Bruno Born, City of Lajeado, Rio Grande do Sul. The individuals were classified between cases and controls by a cardiologist, based on the following criteria: presence of stenosis, greater than 50% of the luminal diameter, in at least one of the coronary arteries. Peripheral blood samples were also collected for biochemical and molecular analyzes. DNA extraction was performed using the salting out method. The polymorphisms in the CD14, TLR4 e TNFα genes were genotyped by Taqman® allelic discrimination assays. The rs28362491 polymorphism in the NFKB1 gene was amplified by polymerase chain reaction (PCR). Results: We identified an association between the polymorphisms rs2569190, located in the CD14 gene, and rs28362491, located in the NFKB1 gene, and CAD. In addition, there were detected significant effects of TLR4 and TNFα gene polymorphisms on glycemic levels and TLR4, NFKB1 and TNFα gene polymorphisms on the lipid profile. Conclusion: Our findings suggest a role for the polymorphisms in CD14 and NFKB1 genes in CAD susceptibility in our sample, corroborating previous evidence of the envolviment of inflamotory process genes in this patology.

Introdução: O Lúpus Eritematoso Sistémico (LES) é uma doença auto-imune crónica sistémica com múltiplas alterações imunológicas e que apresenta uma grande diversidade de manifestações clínicas. É caraterizada pela produção de auto-anticorpos e pela infiltração de células inflamatórias nos tecidos alvo. Os neutrófilos são células fagocíticas com um tempo de semi-vida curto, sendo das primeiras células a serem recrutadas do sangue periférico para o local de inflamação, contribuindo desta forma para o processo inflamatório. Embora a etiopatogenia do LES não esteja completamente compreendida, considera-se que as anormalidades do processo apoptótico possam estar envolvidas no desenvolvimento da doença. Apoptose é também essencial para o estabelecimento de tolerância imune. Neste sentido fomos quantificar a expressão genética de Fas-L, Bcl-2, TNFα e TNFR1 em neutrófilos do sangue periférico purificados de doentes com LES activo, inactivo e de um grupo controlo, correlacionando estes dados com parâmetros de atividade da doença. O grupo de doentes com LES era constituído por 45 mulheres e 4 homens com uma média de idades de 35,3±11,5 e 23,5±4,2 anos, respetivamente, o grupo controlo era constituído por 30 indivíduos saudáveis com uma média de idades de 30±6 anos. Métodos: Analisou-se a expressão do RNAm da Bcl-2, Fas-L, TNFR1 e TNFα em neutrófilos do sangue periférico, separados por cell sorting. A quantificação da expressão de mRNA foi efectuada através de PCR em tempo real. Resultados: Verificou-se um aumento da expressão genética de TNFR1 em doentes com a doença activa e de uma maneira geral, em LES, uma diminuição da expressão de Bcl-2, de TNFα e de Fas-L. Conclusão: Os resultados sugerem a presença de alterações na expressão genética de genes associados à morte celular por apoptose, em neutrófilos do sangue periférico de doentes com LES, o que pode contribuir para a neutropenia que ocorre nesta doença. Introduction: The Systemic Lupus Erythematosus (SLE) is a chronic systemic auto-immune disease with multiple immunologic abnormalities that presents a variety of clinical manifestations. It is characterized by the production of autoantibodies and the infiltration of inflammatory cells on target tissues. The neutrophils are phagocytes with short half-life, being the first to be recruited from the peripheral blood to the inflammation site, and therefore contributing to the inflammatory process. Even though the etiopathogeny of SLE is not fully understood, it is considered that abnormalities in the apoptosis process might be involved in the disease development. Apoptosis is also essential to the establishment of immune tolerance. Therefore our purpose was to quantify the genetic expression of Fas-L, Bcl-2, TNFα and TNFR1 in purified peripheral blood neutrophils from active SLE patients, inactive SLE patients, and from a control group, correlating the obtained results with activity parameters of the disease. The SLE patients group was composed by 45 women and 4 men with an average age of 35,3±11,5 e 23,5±4,2 years, and a control group formed by 30 healthy individuals with an average age of 30±6 years. Methods: We analyzed the mRNA expression of Bcl-2, Fas-L, TNFR1 and TNFα in peripheral blood purified neutrophils by real time PCR. Results: Our results suggest that patients with active SLE express higher amounts of TNFR1, and in general, we observed, in SLE patients, lower mRNA levels of Bcl-2, TNFα and Fas-L. Conclusion: Based in our results, we suggest that some modifications occurred in the mRNA expression of genes associated with apoptosis cell death in peripheral blood neutrophils from SLE patients,that can contribute, at least in part, to the neutropenia observed in the disease.

Mestrado em Biologia Molecular e Celular O conhecimento crescente sobre a biologia das Células Mesenquimais do Estroma (MSCs) tem proporcionado o desenvolvimento de novas terapias celulares, especialmente na área reumatológica de etiologia auto-imune. Estas células exibem propriedades imunossupressoras, tendo a capacidade de suprimir a inflamação local e o dano do tecido numa grande variedade de doenças, em particular na Artrite Reumatóide (AR). O objetivo do estudo foi analisar a ação imunomoduladora das MSCs derivadas da medula óssea na capacidade de produção de citocinas inflamatórias pelas células NK do sangue periférico em doentes com AR e comparar com os resultados obtidos nas mesmas células de indivíduos saudáveis. O estudo envolveu amostras de sangue periférico de oito controlos e de doze doentes com AR. Para determinar a frequência de células NK a expressar TNFα e IFNγ por citometria de fluxo, realizaram-se co-culturas com células mononucleares do sangue periférico e MSCs, com posterior ativação in vitro com PMA e Ionomicina. Na presença de MSCs observou-se, em ambos os grupos em estudo, uma diminuição estatisticamente significativa na percentagem de células NK a produzir TNFα e IFNγ, bem como na expressão destas proteínas por célula. Observou-se também uma correlação negativa entre a percentagem de inibição relativa à expressão de TNFα ou IFNγ nas células NK e o índice DAS da AR. As MSCs inibem de forma eficiente a capacidade de produção de citocinas, nomeadamente TNFα e IFNγ pelas células NK, podendo desta forma contribuir para uma diminuição da inflamação e dos sintomas na AR. The increasing knowledge about Mesenchymal Stromal Cells (MSCs) biology has provided the development of new cell therapies, particularly in Rheumatology area of auto-immune etiology. These cells exhibit immunosuppressive properties, having the ability to suppress local inflammation and tissue damage, in a wide variety of diseases, in particular, Rheumatoid Arthritis (AR). The objective of the study was to analyse the immunomodulatory action of bone marrow derived MSCs, in the ability of peripheral blood NK cells from patients with AR to produce inflammatory cytokines and compare the obtained results with those obtained from healthy individuals. Twelve patients with AR and eight controls were enrolled in this study. We performed co-cultures with peripheral blood mononuclear cells and MSCs during 24 hours. After this period mononuclear cells were activated in vitro with PMA and Ionomycin in order to determine the frequency of NK cells producing TNFα or IFNγ, by flow cytometry. In the presence of MSCs, it was observed, in both studied groups, a statistically significant decrease in the frequency of NK cells producing TNFα or IFNγ, as well as in the expression of both proteins at single cell level. It was also noticeable a negative correlation between the relative percentage of inhibition of TNFα or IFNγ expression in NK cells and the DAS index. MSCs inhibit, in an efficient way, the ability of cytokine production, namely TNFα and IFNγ by NK cells, and therefore contributing to the decrease of inflammation and AR symptoms.

Genetic polymorphisms of IL1 and TNFα have been associated with expression variation of these proteins, what could justify interindividual differences in the disease forms. The aim of this study was to investigate possible associations between IL1B, IL1RN and TNFA genes and the susceptibility to aggressive periodontitis and severe chronic periodontitis. We selected 145 patients from Rio de Janeiro state, 43 with aggressive periodontitis (PAgr) (33.1 ± 4.8 years old), 52 with severe chronic periodontitis (PCr) (50.6 ± 5.8 years old), and 50 controls (40.1 ± 7.8 years old). Genomic DNAs of patients of PAgr, PCr and control groups were obtained through the collection of oral epithelial cells scraped from the inside cheek with a swab. The SNPs IL1B -511C>(pT, and TNFA -1031T>C were analyzed by PCR-RFLP technique using the restriction enzymes Ava I, Taq I and Bpi I, respectively. The polymorphism of variable number of tandem repeats (VNTR) in intron2 of the IL1RN gene was done by direct analysis of amplicons. All polymorphisms were analyzed by 8% polyacrylamide gel electrophoresis. Allelic and genotypic frequencies of the IL1B +3954C>T polymorphism in the PCr group were significantly different as compared to those observed in the control group (p=0.003 and p=0.041, respectively). The frequency of A2 allele of IL1RN intron 2 VNTR polymorphism in the PAgr group was significantly higher than in the control group (p=0.035). There was no association between the polymorphisms IL1B -511C>T and TNFA -1031T>C and aggressive and chronic periodontitis. The presence of alleles 2 in combined genotypes of IL1RN intron 2VNTR and IL1B +3954 C>T in the PCr group was significantly higher as compared to the control group (p=0.045). However no associations between the genotypic combinations IL1B -511C>T IL1B +3954C>T and IL1RN VNTR / IL1B -511C>T and predisposition to periodontal disease were observed. According to our results one can suggest that for the studied population the IL1B +3954C>T polymorphism interferes in the development of chronic periodontitis, whereas the presence of the A2 allele of IL1RN intron 2 VNTR polymorphism may be considered as an indicator of risk for aggressive periodontitis. The present study also allows us to suggest that the absence of homozygous for alleles 1 in the combined genotypes of IL1RN intron 2 VNTR and IL1B +3954C>T may increase the susceptibility to severe chronic periodontitis. A periodontite é um processo inflamatório crônico de origem bacteriana mediado por citocinas, em especial, interleucina-1 (IL1) e fator de necrose tumoral (TNFα). Polimorfismos genéticos de IL1 e TNFA têm sido associados com a variação de expressão dessas proteínas, o que poderia justificar as diferenças interindividuais de manifestação da doença. O objetivo do presente estudo foi investigar possíveis associações entre os genes IL1B, IL1RN e TNFA e a suscetibilidade à periodontite agressiva e à periodontite crônica severa. Foram selecionados 145 pacientes do Estado do Rio de Janeiro, 43 com periodontite agressiva (PAgr) (33,1 ± 4,8 anos), 52 com periodontite crônica severa (PCr) (50,6 ± 5,8 anos) e 50 controles (40,1 ± 7,8 anos). Os DNAs genômicos dos integrantes dos grupos PAgr, PCr e controle foram obtidos através da coleta de células epiteliais bucais raspadas da parte interna da bochecha com cotonete. Os SNPs IL1B -511C>T, IL1B +3954C>T e TNFA -1031T>C foram analisados pela técnica de PCR-RFLP, utilizando as enzimas de restrição Ava I Taq I e Bpi I, respectivamente. O polimorfismo de número variável de repetições in tandem (VNTR) no intron 2 do gene IL1RN foi feita pela análise direta dos amplicons. Todos os polimorfismos foram analisados por eletroforese em gel de poliacrilamida 8%. As frequências alélica e genotípica do polimorfismo IL1B +3954C>T no grupo PCr foram significativamente diferentes das observadas no grupo controle (p=0,003 e p=0,041, respectivamente). A freqüência do alelo A2 do polimorfismo IL1RN VNTR intron2 no grupo PAgr foi significativamente maior do que no grupo controle (p=0,035). Não houve associação entre os polimorfismos IL1B -511C>T e TNFA -1031T>C e as periodontites agressiva e crônica. A presença dos alelos 2 nos genótipos combinados de IL1RN VNTR intron2 e IL1B +3954C>T no grupo PCr foi significativamente maior quando comparada ao grupo controle (p=0,045). Entretanto, não se observou associação entre as combinações genotípicas IL1B -511C>T / IL1B +3954C>T e IL1RN VNTR / IL1B -511C>T e a predisposição à doença periodontal. De acordo com os nossos resultados podemos sugerir que, para a população estudada, o polimorfismo IL1B +3954C>T interfere no desenvolvimento da periodontite crônica, enquanto a presença do alelo A2 do polimorfismo IL1RN VNTR intron2 pode ser considerado como indicador de risco para a periodontite agressiva. O presente estudo também nos permite sugerir que a ausência de homozigose dos alelos 1 nos genótipos combinados de IL1RN VNTR intron2 e IL1B +3954C>T pode representar maior suscetibilidade à periodontite crônica severa.
