Se evaluó la actividad antifúngica de ajoene sobre Histoplasma capsulatum, mediante los métodos de difusión en agar con discos (MDD) y pozos (MDP), usando agar Sabouraud dextrosa (SDA) y Müeller-Hinton modificado (MHM). El inóculo fúngico, obtenido de la fase exponencial y ajustado a 0,5 Mac-Farland, se distribuyó (0,2 ml) sobre las placas de agar. Para el MDD, se siguió el protocolo M44-A-NCCLS, con modificaciones; para el MDP, se abrieron pozos de 4 mm y se procedió como en MDD. Se usaron concentraciones de ajoene de 3,75 a 120 μg. Las placas se incubaron a temperatura ambiente y la inhibición se leyó al séptimo y al 16º día. Los ensayos se realizaron por duplicado. El MDD, al séptimo día, reveló inhibición del crecimiento desde 15 μg de ajoene en SDA y desde 30 μg en MHM. Con el MDP, se observó este efecto a partir de 7,5 y 30 μg en SDA y MHM, respectivamente. La disminución de las zonas de inhibición, al 16° día, en ambos métodos, evidenció efecto fungistático de ajoene sobre H. capsulatum. Con los resultados obtenidos, se podría considerar que H. capsulatum (fase micelial) es sensible a 15 μg de ajoene usando ambos métodos con SDA, y con MHM, a 30 (MDD) y 60 μg (MDP). Ambas metodologías demostraron ser válidas para realizar pruebas de susceptibilidad en H. capsulatum utilizando SDA o MHM. Estos resultados abren las puertas para estudios futuros con el fin de ratificar estos hallazgos. The sensibility of Histoplasma capsulatum to ajoene was evaluated through disk (DDM) and holes (HDM) diffusion methods, using Sabouraud dextrose (SDA) and modified Mueller-Hinton (MHM) agars. The fungal inoculum, obtained from the exponential growth phase, was adjusted to 0.5 Mc-Farland and seeded (0,2 ml) on the plates. For DDM, the protocol M44A-NCCLS was followed, with some modifications; for HDM, 4 mm-holes were punched and proceeded as for DDM. Ajoene was used at concentrations from 3.75 to 120 µg. The plates were kept at room temperature and the inhibition was recorded at days seven and 16. All the assays were run by duplicate. On day seven, the DDM using SDA revealed inhibition zones from 15 µg of ajoene and when using MHM, the inhibition was observed from 30µg. With HDM, ajoene inhibited fungal growth from 7.5 µg when using SDA and with MHM, this effect was noticed from 30 µg. Day 16 showed a considerably decrease of the inhibition zones, by both methods, with the two media, which demonstrates a fungistatic effect of ajoene on this fungus. Taking in account these results, it could be considered that H.capsulatum (mycelial phase) is susceptible to 15 µg of ajoene by both methods, with SDA and with MHM, to 30 (DDM) and 60 µg (HDM). Both methodologies demonstrated validity to perform sensibility tests with H.capsulatum, using both culture media.These promising results justify further research to confirm the findings of the present work.

Ajoene es un agente seguro y eficaz en el tratamiento de cromomicosis y algunas dermatofitosis; ha mostrado su actividad sobre aislados de Microsporum canis, perfilándose como una droga potencial para el tratamiento tópico de la tinea capitis. Por ello, consideramos relevante determinar la susceptibilidad in vitro de 6 aislados de Microsporum canis, obtenidos de pacientes con dicha patología, a los antifúngicos ajoene, terbinafina y griseofulvina, utilizando procedimientos establecidos por el NCCLS M38-A con algunas modificaciones Para el compuesto ajoene los valores de CMI y CI50 estuvieron entre 30-204 µM y 1,45-2,96 µM; las CMI y CI50 de terbinafina se localizaron entre 10-30 µM y 0,02-0,12 µM y para la griseofulvina entre 10-30 µM y 1,08-3,91 µM (0,38-1,38µg/ml) respectivamente. M. canis fue susceptible a ajoene de manera dosis-dependiente, pudiendo este compuesto constituir una alternativa eficiente para el tratamiento de la tinea capitis. Ajoene is a secure and effective agent in chromomycosis and some dermatophytosis treatments; it has shown its activity on Microsporum canis isolates, being a potential drug for topical treatment of tinea capitis. We consider outstanding to determine in vitro susceptibility to ajoene, terbinafine and griseofulvin of six Microsporum canis isolated of patients with this disease, following the guidelines of NCCLS M38-A for molds, with some modifications. The ajoene MIC range were 30-204µM and the IC50 values oscillated among 1,45-2,96 µM; terbinafine MIC and IC50values were among 10-30 µM and 0,02-0,12 µM; griseofulvin MIC and IC50 values were among 10-30µM and 1,08-3,91µM (0,38-1,38µg/ml) respectively. M. canis shows a dose-dependent susceptibility to ajoene, being able to constitute an efficient alternative for tinea capitis treatment.

Ajoene es un compuesto órganosulfurado obtenido originalmente de extractos alcohólicos del ajo y presenta una actividad antimicótica que ha sido ampliamente demostrada. En este trabajo estudiamos la susceptibilidad in vitro al ajoene, de levaduras aisladas de pacientes con onicomicosis, relacionándola con la actividad in vivo mostrada por este compuesto. Para ello fueron seleccionados 8 pacientes con diagnóstico clínico y micológico de onicomicosis. Seis recibieron ajoene en solución al 0,4% y dos fluconazol 150 mg/semanal por 4 meses, con controles clínicos y micológicos a los 30, 60 y 90 días. Se determinaron la CMI y CI50 de ajoene, fluconazol y terbinafina siguiendo las directrices del NCCLS M27-A con modificaciones (RPMI 2% G y uso de hemocitómetro) Los valores de CMI de ajoene oscilaron entre 2.34 - 70.2 µg/ml y la CI50 entre 0.26 - 7.08 µg/ml,. Todos los pacientes tratados con ajoene mostraron después de cuatro meses de tratamiento una importante mejoría de los síntomas y siete de los ocho tratados mostraron cura micológica. Ajoene is an organosulphur compound obtained from garlic alcoholic extracts, with demonstrated broadly antifungal activity both in vivo as in vitro. In this work we determine the susceptibility in vitro of yeast isolates of patients with onychomycosis and which were related with the in vivo activity shown by this compound. Eight patients were selected with clinical and mycological diagnostic of onychomycosis. Six received ajoene solution 0,4% and two fluconazol 150 mg/weekly for 4 months, with clinical and mycologycal controls on days 30, 60 and 90. The MIC and IC50 of ajoene, fluconazol and terbinafine were determined following the guidelines of NCCLS M27-A with modifications (RPMI 2% G and use of hemocytometer). The values of CMI oscillated among 2.34 - 70.2 µg/ml and the CI50 among 0.26 - 7.08 µg/ml. All the patients treated with ajoene showed improvement of symptoms and mycological cure after four months of treatment.
