Comparison with conventional reservoirs, the prediction of pressure in ultra-deep tight sandstone formations is difficult. The prediction of seismic pressure is more challenging than well logging pressure prediction. The main methods for seismic pressure prediction include equivalent depth method, Eaton method, Fillippone formula, and its modified versions. Among them, the Eaton method is widely used and has good effectiveness. However, this method relies on difficult-to-obtain normal compaction trend line, which leads to low prediction accuracy in space. To address this issue, a method combining the Eaton method and Collocated cokriging is proposed. The Eaton formula is used to predict formation pressure at the well, with compressional wave velocity as the covariate for predicting the main variable, formation pressure. By simulating the shear wave velocity based on seismic compressional wave velocity, the influence of various parameters on the prediction results is analyzed, and the accuracy of this method is verified by comparing it with other methods. The proposed method is then applied to predict formation pressure in the ultra-deep formations of the Junggar Basin. The simulation results show that the Collocated cokriging method achieves higher planar accuracy and better matches the experimental expectations in terms of prediction results. The application results also demonstrate the scientific effectiveness of the combined method, which has achieved good results in practical production applications.

Apurinic/apyrimidinic endonuclease 1 (APE1) exonuclease plays a vital proofreading role in the base excision repair pathway by removing 3’end group, including oxidatively damaged DNA bases, chain terminating nucleotide analog drugs, terminal blocking groups and mismatched bases. Herein, a simple and real time fluorometric method was developed for detecting APE1 exonuclease activity using molecular beacon as substrate. Experimental results demonstrated that the detection limit of this method was 0.01 U/μL for APE1, and the Km value was (0.27 ± 0.10) μmol/L. Moreover, the method was used to screen antibiotics and the results showed that streptomycin sulfate and gentamycin sulfate had a significant inhibitory effect on APE1 exonuclease activity in a concentration-dependent manner. Meanwhile, the assay was used for investigating the inhibitory effects of heavy metal ions on APE1 exonuclease activity. Finally, the method was further used to monitor the effect of heavy metal ions treatment of the APE1 exonuclease activity of ramie, and the results indicated that APE1 exonuclease activity was inhibited in a concentration-dependent manner after treatment with Pb2+ and Cd2+, which was consistent with heavy metal ions assay in vitro. In summary, this method provided an alternative tool for the biochemical analysis for APE1 exonuclease activity, which was hopeful for the assessment for heavy metal ions adsorption according to the enzymatic activity change in phytoremediation.

Lymphatic vascular invasion (LVI) is a poor prognostic factor for hypopharyngeal squamous cell carcinoma (HPSCC), but the risk factors of LVI and its relationship with clinicopathological of HPSCC remain unclear. This study aims to explore these issues. We retrospectively analyzed the clinicopathological data of 170 patients with HPSCC from January 2011 to December 2015. The relationship between LVI and clinicopathologic was analyzed by Chi-square test or Fisher’s exact test. The risk factors of LVI were examined using a logistic regression model, while risk factors of survival rate were carried out using the Cox regression model. LVI occurred in 59 cases (34.7%). In multivariate analysis, T3–4 stage (HR = 2.877; 95% CI: 1.379–6.004; p = 0.005), N2–3 stage (HR = 2.325; 95% CI: 1.120–4.824; p = 0.024), and poor differentiation (HR = 2.983; 95% CI: 1.229–7.242; p = 0.016) were independent risk factors for LVI; positive LVI was an independent risk factor for local recurrence (HR = 2.488; 95% CI: 1.150–5.383; p = 0.021), poor 5-year OS (HR = 0.375; 95% CI: 0.232–0.606; p

TGA is one of the members of TGACG sequence-specific binding protein family, which plays a crucial role in the regulated course of hormone synthesis as a stress-responsive transcription factor (TF). Little is known, however, about its implication in response to bacterial wilt disease in potato (Solanum tuberosum) caused by Ralstonia solanacearum. Here, we performed an in silico identification and analysis of the members of the TGA family based on the whole genome data of potato. In total, 42 StTGAs were predicted to be distributed on four chromosomes in potato genome. Phylogenetic analysis showed that the proteins of StTGAs could be divided into six sub-families. We found that many of these genes have more than one exon according to the conserved motif and gene structure analysis. The heat map inferred that StTGAs are generally expressed in different tissues which are at different stages of development. Genomic collinear analysis showed that there are homologous relationships among potato, tomato, pepper, Arabidopsis, and tobacco TGA genes. Cis-element in silico analysis predicted that there may be many cis-acting elements related to abiotic and biotic stress upstream of StTGA promoter including plant hormone response elements. A representative member StTGA39 was selected to investigate the potential function of the StTGA genes for further analysis. Quantitative real-time polymerase chain reaction (qRT-PCR) assays indicated that the expression of the StTGAs was significantly induced by R. solanacearum infection and upregulated by exogenous salicylic acid (SA), abscisic acid (ABA), gibberellin 3 (GA3), and methyl jasmonate (MeJA). The results of yeast one-hybrid (Y1H) assay showed that StTGA39 regulates S. tuberosum BRI1-associated receptor kinase 1 (StBAK1) expression. Thus, our study provides a theoretical basis for further research of the molecular mechanism of the StTGA gene of potato tolerance to bacterial wilt.

The clinical diagnosis of gastrointestinal stromal tumors (GISTs) requires time-consuming tumor localization by physicians, while automated detection of GIST can help physicians develop timely treatment plans. Existing GIST detection methods based on fully supervised deep learning require a large amount of labeled data for the model training, but the acquisition of labeled data is often time-consuming and labor-intensive, hindering the optimization of the model. However, the semi-supervised learning method can perform better than the fully supervised learning method with only a small amount of labeled data because of the full use of unlabeled data, which effectively compensates for the lack of labeled data. Therefore, we propose a semi-supervised gastrointestinal stromal tumor (GIST) detection method based on self-training using the new selection criterion to guarantee the quality of pseudo-labels and adding the pseudo-labeled data to the training set together with the labeled data after linear mixing. In addition, we introduce the improved Faster RCNN with the multiscale module and the feature enhancement module (FEM) for semi-supervised GIST detection. The multiscale module and the FEM can better fit the characteristics of GIST and obtain better detection results. The experiment results showed that our approach achieved the best performance on our GIST image dataset with the joint optimization of the self-training framework, the multiscale module, and the FEM.

Background: The characterization of gene families especially MYB being the largest transcription factor (TFs) families in plants is a crucial step for functional studies. The completion of ramie genome sequencing provides a great opportunity to investigate the organization and evolutionary traits of ramie MYB genes at the genome-wide level. Results: A total of 105 BnGR2R3-MYB genes were identified in ramie and divided into 35 distinct subfamilies according to phylogeny divergence and sequences similarity. These genes were unevenly distributed among 14 chromosomes. Collinearity analysis showed that the segmental and tandem duplication events is the dominant form of the gene family expansion, and duplications prominent in distal telomeric regions. 88 BnGR2R3-MYB genes showed syntenic relationship with those in Apocynum venetum, followed by Cannabis sativa (58), Arabidopsis (53), maize (8) and rice (4). 103 of the 105 BnGMYB proteins were predicted nuclear subcellular, the remaining two were in either chloroplast or cytoplasm. The combination of transcriptome data and phylogenetic tree allows us to propose some powerful MYB candidates that might be involved in the regulation of secondary wall-associated cellulose synthases (BnGMYB14) secondary cell wall thickening (BnGMYB66/67) and flavonoids synthesis (BnGMYB60). The MYBs subgroups involve in regulating anthocyanin were different from arabidopsis and tomato pointing that BnGMYB in other groups play a role in regulating anthocyanin synthesis. qPCR results revealed 8 MYB TFs candidate genes for cadmium stress in ramie. There was an increased in synthesis of procyanidins under the cadmium stress, which suggest a new regulatory pathway in response to the stress. The predicted network identifies the interface between flavonoid metabolic pathways and adversity stress, and found evidence for the involvement of flavonoid synthetic pathways in the stress regulation.Conclusions: This work provides a basic understanding of BnGR2R3-MYB gene family characteristics and provides valuable information that may contribute in improving the tolerance of ramie to cadmium stress and fiber quality.

Al2O3/cerium oxide composite films were prepared on the surface of aluminum by an anodic oxidation and chemical conversion method. The aluminum was firstly anodized in sulfuric acid, and then the anodized aluminum was treated with the Ce(NO3)3 solution. The effect of different concentrations of 1H-Benzotriazole (BTA) on the corrosion resistance of Al2O3/cerium oxide composite films was investigated. X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), scanning electron microscope (SEM) with energy dispersive spectrometer (EDS), and electrochemical workstation were used to characterize the properties of the composite films. The results show that the deposition effect of cerium ions is optimal in 0.5 g/L BTA. At the same time, the surface uniformity and smoothness of the composite films are both enhanced. And the corrosion resistance of the composite film is strengthened.

In this paper, we present the transcriptome profiles of the A. venetum L. by RNA-Seq approach. A total of 6.57 Gb raw data were obtained, and 52,983 unigenes with an average length of 1009 bp and N50 of 1632 bp were annotated with the 7 databases. The unigenes annotated to KEGG database were divided into 21 categories from 6 main groups. Among these, 4952 (22.21%) unigenes were clustered to “Global and overview maps”, and 1834 (8.23%) unigenes were clustered to “Carbohydrate metabolism”. In addition, 6340 unigenes containing 7579 SSRs were identified and the mononucleotide, dinucleotide, trinucleotide motifs were the most common motif type (95.59%), accounting for 39.62%, 36.02%, and 19.95%, respectively.

Abscisic acid (ABA) is known as an important hormone regulating plant stress resistance, such as salt, drought and heavy metal resistance. However, the relationship between ABA and cadmium (Cd) enrichment in ramie (Boehmeria nivea L.) is still unclear to date. This study aimed to reveal the effect of ABA on Cd enrichment in ramie, and we received the following results: (1) Under Cd treatment, the Cd uptake of ramie increased with the increase of Cd concentration, but the chlorophyll content decreased. Under Cd treatment, the ABA content was highest in roots of ramie, followed by that in old leaves, and lowest in new leaves. Long-time treatment of high Cd concentration reduced the ability of endogenous ABA biosynthesis. (2) Spraying ABA on ramie plants (SORP) and adding ABA directly to the culture solution (ADCS) with low concentration can promote the growth of ramie and increase the amount of Cd uptake, and the effect of SORP is better. (3) The molecular reason for the decrease of chlorophyll content due to Cd stress, may be resulted from the down-regulated expression of the chlorophyll synthesis genes (BnPAO and BnNYC1) and the up-regulated expression of the chlorophyll degradation genes (BnCHLH, BnCHLG, BnHAP3A and BnPPR1). The elevated ABA content in ramie plants may due to the up-regulated expression of the ABA synthesis related genes (BnABA1, BnNCED3, and BnNCED5) and the genes (BnABCG40, BnNFXL2, BnPYL9, BnGCR2, BnGTG1, BnBGLU1, BnUTG1, BnVHAG1 and BnABI5) that encoding ABA transport and response proteins, which was consistent with the enhance the Cd uptake in ramie. Our study revealed the relationship between ABA and Cd uptake in ramie, which provided a reference for improving the enrichment of Cd in ramie.

Flavonoids contained in ramie (Boehmeria nivea) root are important resources for officinal use. The plant seedling can grow optimally and develop an easy-harvesting adventitious root system in a hydroponic system. This research was carried out to unravel the regulation mechanism of flavonoid biosynthesis under hydroponic culturing condition, which hitherto remained unknown. Root samples of the ramie plants grown in soil and hydroponic conditions at different developmental stages were collected and pooled for transcriptome sequencing, and the expression levels of flavonoid biosynthesis-related genes and total flavonoid content were quantified. A total of 43,541 unigenes were finally annotated and the expression of 9 key unigenes were confirmed by qRT-PCR. The flavonoid biosynthesis-related gene BnF3H was significantly up-regulated in the roots of hydroponic ramie, and higher total flavonoids content in the hydroponic ramie roots was also observed. The large dataset of transcripts and unigenes provided abundant genetic information for identifying flavonoid biosynthesis genes in ramie roots. Our results suggest flavonoid biosynthesis in ramie roots is positively affected in aquatic environment, and hydroponic culturing could have a potential for enhancing total flavonoids content in ramie roots.

In the current study, the active flavonoids from Apocynum venetum and Apocynum hendersonii leaf were efficiently characterized using UPLC-ESI-MS/MS, and yielding the highest content of 15.35 mg/g (A. venetum) and 13.28 mg/g (A. hendersonii) respectively. The antioxidant assay in vitro showed that the isolated flavonoid ingredient groups exhibited free radical scavenging activities to DPPH, ABTS and linoleic acid. The antimicrobial assay revealed the isolated flavonoid ingredient from both A. venetum and A. hendorsonii have exerted anti-MRSA and anti-P. aeruginosa effect through disrupting cell integrity and declining ATP. In vivo assay demonstrated that these flavonoid ingredients effectively accelerated MRSA-infected and P. aeruginosa-infected Balb/c mice wound healing. In summary, these results showed that the characterized flavonoid ingredients exhibited great potential as natural antioxidant and antimicrobial agents, and shed light into future potential applications of Apocynum spp.

Xylose utilization is one of the key issues in lignocellulose bioconversion. Because of glucose repression, in most engineered yeast with heterogeneous xylose metabolic pathway, xylose is not consumed until glucose is completely utilized. Although simultaneous glucose and xylose utilization have been achieved in yeast by RPE1 deletion, we regulated ZWF1 and PGI1 transcription to improve simultaneous xylose and glucose utilization by controlling the metabolic flux from glucose into the PP pathway. Xylose and glucose consumption increased by approximately 80 and 72%, respectively, whereas ZWF1 was overexpressed by multi-copy plasmids with a strong transcriptional promoter. PGI1 expression was knocked down by promoter replacement; the glucose and xylose metabolism increased when PGI1p was replaced by weak promoters, SSA1p and PDA1p. ZWF1 overexpression decreased while PGI1 down-regulation increased the ethanol yield to some extent in the recombinant strains.

It is an increasing interest in developing a hydroponic culture system for ramie propagation, which provide a promising method for blocking-up the soilborne diseases. However, there is an urgent need for better understanding of ramie adaptation mechanism during hydroponic culturing under low-oxygen condition. In this study, a genome-wide study plus transcriptomic and proteomic analysis was conducted to profile the molecular responses related to ramie oxidative adaption. The results showed that aldehyde dehydrogenases (ALDHs) played important roles in molecular oxygen deficiency and anaerobic metabolism in ramie. Totally, 24 ALDH genes in ramie genome were identified and grouped into 8 superfamily. All of these 24 BnALDH genes, which are unevenly distributed on the 12 chromosomes, exhibited differential expression patterns under hydroponic and soiled culturing. The expression level of almost all the BnALDHs in leaves are lower than in the roots. The BnALDHs classified into family 2 and family 10 were up-regulated in the hydroponic cultured ramie roots. Especially, BnALDH18 belonging to family 10 showed the strongest response to low-oxygen stress. In addition, heterologous overexpression analysis of BnALDH18 in Escherichia coli showed its function for elevating tolerance to oxidative stress. The genome-wide identification and analysis of evolution and expression manner demonstrated that BnALDHs are promising and untapped genetic resources for improving oxygen deficiency tolerance of ramie under hydroponic culturing.

The proteolytic activation of prophenoloxidase (proPO) is a humoral defense mechanism in insects and crustaceans. Phenoloxidase (PO) is produced as an inactive precursor namely, proPO and is activated via specific proteolytic cleavage by proPO-activating proteinase. The current research reports two novel serine proteinase genes (PxSP1-768 bp and PxSP2-816 bp) from Plutella xylostella, encoding 255 and 271 amino acid residues, respectively. Tissue distribution analyses by semiquantitative reverse transcription-PCR (RT-PCR) revealed the resultant genes to be primarily expressed in the hemocytes, while quantitative-RT-PCR (qRT-PCR) assay showed that transcription level of PxSP1 and PxSP2 increased significantly after injection of the fungal pathogen Beauveria bassiana. Purified recombinant fusion proteins of PxSP2 and PxSP1 were injected to New Zealand white rabbits and polyclonal antibodies were generated with the titers of 1:12,800. After silencing the expression of PxSP2 by RNAi, the PO activity decreased significantly. The results show that PxSP2 is involved in prophenoloxidase activation in P. xylostella.

Nitrogen-use efficiency (NUE) has significant impacts on plant growth and development. NUE in plants differs substantially in physiological resilience to nitrogen stress, however, the molecular mechanisms underlying enhanced resilience of high-NUE plants to nitrogen deficiency remains unclear. We compared transcriptome-wide gene expression between high-NUE and low-NUE ramie (Boehmeria nivea (L.) Gaudich) genotypes under nitrogen (N)-deficient and normal conditions to identify the transcriptomic expression patterns that contribute to ramie resilience to nitrogen deficiency. Two ramie genotypes with contrasting NUE were used in the study, including T29 (NUE = 46.01%) and T13 (NUE = 15.81%). Our results showed that high-NUE genotypes had higher gene expression under the control condition across 94 genes, including frontloaded genes such as GDSL esterase and lipase, gibberellin, UDP-glycosyltransferase, and omega-6 fatty acid desaturase. Seventeen stress-tolerance genes showed lower expression levels and varied little in response to N-deficiency stress in high-NUE genotypes. In contrast, 170 genes were upregulated under N deficiency in high-NUE genotypes but downregulated in low-NUE genotypes compared with the controls. Furthermore, we identified the potential key genes that enable ramie to maintain physiological resilience under N-deficiency stress, and categorized these genes into three groups based on the transcriptome and their expression patterns. The transcriptomic and clustering analysis of these nitrogen-utilization-related genes could provide insight to better understand the mechanism of linking among the three gene classes that enhance resilience in high-NUE ramie genotypes.

Background MicroRNAs (miRNAs) regulate numerous crucial abiotic stress processes in plants. However, information is limited on their involvement in cadmium (Cd) stress response and tolerance mechanisms in plants, including ramie (Boehmeria nivea L.) that produces a number of economic valuable as an important natural fibre crop and an ideal crop for Cd pollution remediation. Results Four small RNA libraries of Cd-stressed and non-stressed leaves and roots of ramie were constructed. Using small RNA-sequencing, 73 novel miRNAs were identified. Genome-wide expression analysis revealed that a set of miRNAs was differentially regulated in response to Cd stress. In silico target prediction identified 426 potential miRNA targets that include several uptake or transport factors for heavy metal ions. The reliability of small RNA sequencing and the relationship between the expression levels of miRNAs and their target genes were confirmed by quantitative PCR (q-PCR). We showed that the expression patterns of miRNAs obtained by q-PCR were consistent with those obtained from small RNA sequencing. Moreover, we demonstrated that the expression of six randomly selected target genes was inversely related to that of their corresponding miRNAs, indicating that the miRNAs regulate Cd stress response in ramie. Conclusions This study enriches the number of Cd-responsive miRNAs and lays a foundation for the elucidation of the miRNA-mediated regulatory mechanism in ramie during Cd stress. By using a hydroponic culture system, the terrestrial fiber crop ramie can growth optimally in aquatic environment and enhance exponentially quantities of high quality seedlings for subsequent field cultivation. In this study, the survival rate of ramie seedling was more than 97% when cultured using the novel hydroponic method. Further physiological analysis of the hydroponic ramie to different concentration of livestock wastewater demonstrated that all of these ramies can survival in livestock wastewater, but the 4 times diluted livestock wastewater (total N: 100.9 mg L−1, total P: 2.69 mg L−1) was more appropriate for ramie growth. The nutrients N and P in livestock wastewater were significantly decreased by the growth of ramie, and the removal efficiency of total N and total P in the 4 times diluted livestock wastewater achieved 78.1% and 43.1% respectively within 5 weeks. In conclusion, our studies highlight that the combination of ramie and the hydroponic technology resulted to be effective in the phytoremediation of livestock wastewater.

Ramie, Boehmeria nivea (L.) Gaudich, family Urticaceae, is a plant native to eastern Asia, and one of the world's oldest fibre crops. It is also used as animal feed and for the phytoremediation of heavy metal-contaminated farmlands. Thus, the genome sequence of ramie was determined to explore the molecular basis of its fibre quality, protein content and phytoremediation. For further understanding ramie genome, different paired-end and mate-pair libraries were combined to generate 134.31 Gb of raw DNA sequences using the Illumina whole-genome shotgun sequencing approach. The highly heterozygous B. nivea genome was assembled using the Platanus Genome Assembler, which is an effective tool for the assembly of highly heterozygous genome sequences. The final length of the draft genome of this species was approximately 341.9 Mb (contig N50 = 22.62 kb, scaffold N50 = 1,126.36 kb). Based on ramie genome annotations, 30,237 protein-coding genes were predicted, and the repetitive element content was 46.3%. The completeness of the final assembly was evaluated by benchmarking universal single-copy orthologous genes (BUSCO); 90.5% of the 1,440 expected embryophytic genes were identified as complete, and 4.9% were identified as fragmented. Phylogenetic analysis based on single-copy gene families and one-to-one orthologous genes placed ramie with mulberry and cannabis, within the clade of urticalean rosids. Genome information of ramie will be a valuable resource for the conservation of endangered Boehmeria species and for future studies on the biogeography and characteristic evolution of members of Urticaceae.

Spodoptera litura is one of the most destructive phytophagous pest infesting cotton, vegetable, oilseed and fiber crops around the world. Destruxin A (DA), is a one of a kind microbial insecticide, which has potent toxins with bioactivity against S. litura larvae. An abnormal wing disc (AWD) protein was identified as a DA toxic effect protein in S. litura SL-1 cells. To better understand the role of the AWD gene of S. litura (SLAWD) it was purified and characterized. The entire coding region of the SLAWD gene was cloned into a pET-32a (+) expression vector and transformed into competent Escherichia coli BL21 (DE3) cells. SDS-PAGE and western blotting analysis showed that the best induction conditions were 1 mmol mL−1 isopropyl-β-D-thiogalactopyranoside (IPTG) for 6 h at 37°C; the molecular weight of the fusion protein was 35.0 kDa. The production of polyclonal antibodies and an enzyme-linked immunosorbent assay (ELISA) showed that the titer of antiserum was 1: 25600; western blotting analysis showed that the recombinant SLAWD was recognized by the anti-SLAWD polyclonal antibody. AWD is a key protein involved in wing development in insects. These tools will assist in the further characterization of SLAWD and studies on the mechanism of action of destruxin A.

Ramie (Boehmeria nivea (L.) Gaud.) is a traditionally terrestrial fiber crop. However, hydroponic technology can enhance the quantity and quality of disease free Ramie plant seedlings for field cultivation. To date, few studies have attempted to examine the hydroponic induction of ramie roots and the molecular responses of ramie roots to aquatic environment. In this study, ramie tender stems was grown in the soil or in a hydroponic water solution, and cultured in the same environmental conditions. Root samples of terrestrial ramie, and different developmental stages of hydroponic ramie (5 days, 30 days), were firstly pooled for reference transcriptome sequencing by Illumina Hiseq 2000. Gene expression levels of each samples were quantified using the BGISEQ500 platform to help understand the distribution of aquatic root development related genes at the macro level (GSE98903). Our data resources provided an opportunity to elucidate the adaptation mechanisms of ramie seedlings roots in aquatic environment.

This study explored the effects of long-term bathing in radon hot springs on oxidative damage and antioxidation function in humans. In this study, blood was collected from residents in the Pingshan radon hot spring area (RHSA), Jiangzha RHSA, and control area (CA). 8-Hydroxydeoxyguanosine (8-OHdG) and thioredoxin reductase (TrxR), representing oxidation and antioxidant levels, respectively, were analyzed as indices. Compared to the CA group, the RHSA group in the Pingshan and Jiangzha areas showed significantly decreased 8-OHdG levels ( Z = −3.350, −3.316, respectively, P < .05) and increased TrxR levels ( Z = 2.394, 3.773, respectively, P < .05). The RHSA and CA groups in Jiangzha had lower levels of TrxR and 8-OHdG compared to those in Pingshan. This finding may be related to the different radon concentration levels, bathing time and other factors. Results suggested that long-term bathing in radon hot spring may activate antioxidant function and reduce oxidative damage in the body.

Cecropins are linear cationic antibacterial peptides that have potent activities against microorganisms. In the present study, a 480bp full-length cDNA encoding diamondback moth (Plutella xylostella) cecropin 1 (designated as Px-cec1) was obtained using RT-PCR. A Northern blot analysis showed that the Px-cec1 transcript was predominantly expressed in fat bodies, hemocytes, midgut and epidermis with the highest expression level in fat bodies. The expression of Px-cec1 mRNA in fat bodies was significantly increased 24h after microbial challenge, with the highest induced expression by Staphylococcus aureus. A circular dichroism (CD) analysis revealed that the recombinant Px-cec1 mainly contained α-helixes. Antimicrobial assays demonstrated that recombinant Px-cec1 exhibited a broad spectrum of anti-microbial properties against fungi, Gram-positive and Gram-negative bacteria, but it did not exhibit hemolytic activity against human erythrocytes. Furthermore, Px-cec1 caused significant morphological alterations of S. aureus, as shown by scanning electron microscopy and transmission electron microscopy. These results demonstrated that Px-cec1 exerts its antibacterial activity by acting on the cell membrane to disrupt bacterial cell structures.

The endoglucanase gene endo753 from Aspergillus flavus NRRL3357 strains was cloned, and the recombinant Endo753 was displayed on the cell surface of Saccharomyces cerevisiae EBY100 strain by the C-terminal fusion using Aga2p protein as anchor attachment tag. The results of indirect immunofluorescence and Western blot confirmed the expression and localization of Endo753 on the yeast cell surface. The hydrolytic activity test of the whole-cell enzyme revealed that Endo753 immobilized on the yeast cell surface had high endoglucanase activity. The functional characterization of the whole-cell enzyme was investigated, and the whole-cell enzyme displayed the maximum activity at pH 8 and 50 °C. The enzyme was stable in a pH range of 7.0–10.0. Furthermore, the whole-cell enzyme displayed high thermostability below 50 °C and moderate stability between 50 and 70 °C. These properties make endo753 a good candidate in bioethanol production from lignocellulosic materials after displaying on the yeast cell surface.

Network measurement is an important approach to understand network behaviors, which has been widely studied. Both TCP and ICMP are applied in network measurement, while investigating the differences between the measured results of these two protocols is an important topic that has been less addressed. In this paper, to compare the differences between TCP and ICMP when they are used in measuring host connectivity, RTT, and packet loss rate, we designed two groups of comparison programs, and after careful evaluating of the program parameters, we executed a lot of comparison experiments on the Internet. The experimental results show that, there are significant differences between the host connectivity measured using TCP or ICMP; in general, the accuracy of connectivity measured using TCP is 20-30% higher than that measured using ICMP. The case of RTT and packet loss rate is complicated, which are related to path loads and destination host loads. While commonly, the RTT and packet loss rate measured using TCP or ICMP are very close. According to the experimental results, we also give some advices on protocol selection for conducting accurate connectivity, RTT and packet loss rate measurements.

Functional screening of a metagenomic library of termite hindgut identified an overlapping gene cluster encoding the phosphotransferase system (PTS) components, which consisted of a glucoside specific PTS enzyme II gene (glu1923) and a glycoside hydrolase gene (glu1392). Hydrolytic experiments revealed that the combined effect of Glu1923 and Glu1392 was responsible for the utilization of glucosidic substrates in recombinant Escherichia coli (E. coli) strains. Yeast two hybrid analysis suggested that there was an interaction between Glu1923 and Glu1392, and the domain EIIA of Glu1923 played an important role for the interaction. In addition, the hydrolase Glu1392 displayed hydrolysis ability toward cellobiose and maltose, and had a very high tolerance to glucose with a Ki value of 2.25M. These properties make Glu1392 an interesting candidate in biotechnology applications after further study.

With the basis of classification characteristics of sports tourism of Xinjiang Mongolian, this paper tries to carry out comparative analysis with the emphasis on the "Naadam" festival in Xinjiang and Inner Mongolia. This paper approves that Xinjang has its merits in the development of sports tourism with Mongolian characteristics by combining the local characteristics of physical aspects while there also exists some problems. Hence this paper puts forward some strategies of the development of sports tourism project in Xinjiang Mongolian.

Simplified FE (finite element) model about heavy commercial vehicle cab and acoustic cavity are built in this paper. They are verified through the modal analysis and modal test. Then a acoustic-structure coupled model is formed. Modal analysis is performed using MSC. Nastran. It is shown from the simulation result that the modes of coupled system reflect both the characteristics of structural and cave model. Meanwhile, excitations received from road test are acted to the acoustic-structure coupled model to make a frequency response analysis. The interior noise of response is correlative to the measured values well, so the established acoustic-structure coupled model can achieve better simulation of interior noise, and it is benefit for the later optimal design of acoustic.

Arterial stiffness has a strong relationship with cardiovascular disease. Pulse wave velocity (PWV) is increasingly used as a measure of arterial stiffness. The calculation of PWV requires accurate measurement of blood flow velocity and aortic length. Using magnetic resonance imaging (MRI), we are able to accurately acquire blood flow velocity and to virtually measure the aortic length inside the central aorta. Manual measurement of aorta length is inaccurate and subjective. In this study, we set out to develop a method which automatically locates the thoracic aorta and measures aortic length, allowing noninvasive measurement of PWV derived from the aortic flow. Our method is novel, efficient and robust. This offers a reliable and convenient tool for PWV measurement allowing detailed non invasive assessment of cardiovascular risk.
