Context The rearing system is a critical non-genetic factor that can considerably affect poultry production. Aims This study examined the effects of floor rearing systems (FRS), net rearing systems and cage rearing systems (CRS) on growth performance, carcass traits, meat quality and serum biochemical parameters of Gaoyou ducks. Methods A total of 450 healthy 22-day-old male Gaoyou ducks with similar average bodyweight were randomly divided into the FRS, net rearing systems and CRS groups. It is becoming increasingly attractive to execute workflows in the cloud, as the cloud environment enables scientific applications to utilize elastic computing resources on demand. However, despite being a key to efficiently managing application execution in the cloud, traditional workflow scheduling algorithms face significant challenges in the cloud environment. The gradient-based optimizer (GBO) is a newly proposed evolutionary algorithm with a search engine based on the Newton’s method. It employs a set of vectors to search in the solution space. This study designs a gradient-based scheduler by using GBO for workflow scheduling to minimize the usage costs of workflows under given deadline constraints. Extensive experiments are conducted on well-known scientific workflows of different sizes and types using WorkflowSim. The experimental results show that the proposed scheduling algorithm outperforms five other state-of-the-art algorithms in terms of both the constraint satisfiability and cost optimization, thereby verifying its advantages in addressing workflow scheduling problems.

CCR5 levels on the surface of monocyte/lymphocyte and plasma RANTES levels were detected by flow cytometry. TaqMan real-time fluorescent quantitative PCR was used to detect genetic polymorphisms of RANTES rs2280788 and CCR5 rs1799987.Results: There were no significant differences in frequencies of CCR5 rs1799987 genotype and A/G allele and frequencies of RANTES rs2280788 genotype and C/G allele, between subjects in NC and DM group (P > 0.05). Plasma RANTES level in DM group was significantly lower than NC group (P < 0.05), and difference came from patients with T2DM using insulin and subjects with normal blood glucose. CCR5 levels on the surface of monocytes and lymphocytes of patients in DM group were higher than NC group (P < 0.05). There was no significant difference in CCR5 level on the surface of monocytes and lymphocytes (or plasma RANTES level) among different genotypes of CCR5 rs1799987 (or RANTES rs2280788) (P > 0.05). RANTES level was positively correlated with age and TC and negatively correlated with diabetes course and HbA1c. CCR5 level on the surface of monocytes was positively correlated with drinking years, HbA1c, course of diabetes, and negatively correlated with TC. CCR5 on lymphocyte surface was positively correlated with diabetes course, smoking years, HbA1c, and negatively correlated with LDL, TC, HDL (P < 0.05).Conclusion: RANTES -28 (rs2280788) C/G polymorphism or CCR5 59029 (rs1799987) A/G polymorphism may not be associated with T2DM of Han nationality in Kunming and cannot affect RANTES and CCR5 expression. The design options of sensing-communication coexistence (SCC) vs. integrated sensing and communications (ISAC) relying on non-orthogonal downlink transmission (NO-DLT) and transmit antenna selection are investigated. To be practical, both realistic residual hardware impairments (RHI) and imperfect successive interference cancellation are considered. The performance of the SCC and ISAC NO-DLT frameworks is characterized by deriving both the exact and the asymptotic outage probabilities (OPs) as well as the probability of successful target detection (PoD). Our numerical results demonstrate the accuracy of the theoretical analysis and confirm that: 1) The ISAC NO-DLT framework is superior to the SCC NO-DLT framework both in terms of its OP and PoD; 2) The realistic RHI degrades the S&C performance of both the SCC and ISAC NO-DLT frameworks.

PurposeIn the sharing economy (SE), consumer misbehaviour is an operational challenge for platforms due to its negative outcomes. The psychological mechanism behind consumer misbehaviour remains unclear. As such, this research aims to investigate how consumers' sharing motivations affect their misbehaviours in the SE.Design/methodology/approachDrawing on motivated cognition theory, the authors establish a research model explaining the effects of consumers' sharing motivations on their cost–benefit analysis of misbehaviour and misbehaviour intention. A scenario-based online experiment is used to test the research hypotheses.FindingsThe results indicate that consumers' extrinsic and intrinsic motivations to share have different impacts on their perceived benefits and costs of potential misbehaviour, thereby influencing their misbehaviour intention.Originality/valueThis study reveals consumers' psychological mechanism underlying their misbehaviours in the SE and provides operational implications for platforms to help them effectively reduce consumer misbehaviours through preventive measures.

Postoperative pain after video-assisted thoracoscopic surgery (VATS) is common in lung cancer patients, and it is unclear whether cancer itself participates in pain regulation. Programmed cell death ligand-1 (PD-L1) expressed by tumours may be analgesic. Our study aimed to detect the association between PD-L1 and acute postoperative pain.We reviewed patients who underwent VATS for lung cancer with tumour PD-L1 expression analysed in our centre from Jan 2017 to Jul 2020. They were divided into PD-L1 (-) group and PD-L1 (+) group and were further divided into four subgroups according to opioids for postoperative analgesia: sufentanil PD-L1 (-), sufentanil PD-L1 (+), oxycodone PD-L1 (-), and oxycodone PD-L1 (+). We compared the numeric rating scale (NRS) for the first three postoperative days at rest or cough between groups.A total of 369 patients (167 in PD-L1 (-) vs. 202 in PD-L1 (+)) were included. On the first postoperative day, NRS at cough in the PD-L1 (+) patients were higher than those in the PD-L1 (-) patients (2.91 ± 1.07 vs. 2.66 ± 1.01, p = 0.018). On the third day, NRS at cough in PD-L1 (+) patients was higher (2.50 ± 1.02 vs. 2.26 ± 1.09, p = 0.043). For patients with oxycodone, NRS was higher in PD-L1 (+) than in PD-L1 (-) (p = 0.041) at cough after surgery. In contrast, those with sufentanil did not significantly differ in NRS between groups.Patients with PD-L1 (+) suffered graver pain in the early postoperative period after VATS for lung cancer compared with PD-L1 (-) on tumours. Analgesia with sufentanil seemed to overcome this effect better than oxycodone.We demonstrated that patients with positive programmed cell death ligand-1 (PD-L1) on tumours suffered graver pain in the early postoperative period after video-assisted thoracoscopic surgery for lung cancer and reacted differently with opioids. It might be beneficial to adjust analgesic protocols according to tumour PD-L1 expression for individualized postoperative pain management.

Objective: This study was undertaken to evaluate the clinical efficacy of Western medicine combined with Chinese medicine for pelvic inflammatory disease (damp-heat and stasis type). Methods: Seventy-four patients who were diagnosed with pelvic inflammatory disease (damp-heat and stasis type) by our hospital during July 2021 to July 2022 were randomized into two groups: the participants in the control group received conventional Western medicine treatment, and the participants in the study group received Western medicine combined with Chinese medicine. Results: After treatment, the total effectiveness of the control group (72.98%) was significantly lower than that of the study group (94.59%), (P < 0.05); the whole blood viscosity high cut, whole blood viscosity low cut, fibrinogen and plasma viscosity of the control group were all lower than those of the study group (P < 0.05); the levels of CRP, IL-6, and TNF-? in the control group were higher and IL-2 levels in the control group were lower than those in the study group (P < 0.05). Conclusion: Western medicine combined with Chinese medicine is more effective in curing damp-heat and stasis-type pelvic inflammatory disease by improving the blood rheological indexes and lowering the level of inflammatory factors.

Introduction: Cannabinoids including cannabidiol (CBD) have attracted enormous interest as bioactive ingredients for various dermatological and/or cosmeceutical uses. However, topical applications of cannabinoids might be limited without a fundamental understanding of their skin permeability. Herein, we aimed to evaluate the skin permeability of CBD and its topical formulations using artificial skin membrane assays. The solubility and stability of CBD in various surfactants that are commonly used in topical applications were also evaluated. Methods: CBD and two CBD-incorporated topical formulations (cream and gel) were prepared for this study. Computational predictions (SwissADME and DERMWIN™) and the parallel artificial membrane permeability assay (PAMPA) were used to evaluate the skin permeability of CBD isolate. The Franz cell diffusion (in vitro release testing) assay was used to evaluate the skin permeability of CBD formulations. The solubility and stability of CBD in surfactants were assessed by high-performance liquid chromatography and mass spectrometry analysis. Results: CBD isolate showed favorable skin permeability in the SwissADME and DERMWIN™ predictions (−Log Kp of 3.6 and 5.7 cm/s, respectively) and PAMPA (−LogPe value of 5.0 at pH of 6.5 and 7.4). In addition, CBD had higher solubility (378.4 μg/mL) in surfactant Tween 20 as compared to its solubility in polyisobutene. In an acidic environment (pH 5 and 6), Tween 20 maintained the CBD content at 81% and 70% over 30 days, respectively. CBD in the formulations of cream and gel also had moderate skin permeability in the Franz cell diffusion assay. Conclusion: Data from artificial membrane-based assays support that CBD is a skin permeable cannabinoid and the permeability and stability of its formulations may be influenced by several factors such as surfactant and pH environment. Findings from our study suggest that CBD may have suitable skin permeability for the development of dermatological and/or cosmeceutical applications but further studies using in vivo models are warranted to confirm this.

Objective: To investigate the clinical efficacy of the combination of enhanced “Shou Tai Wan” soup and progesterone in the treatment of early pre-eclampsia miscarriage. Methods: 86 patients with early pre-eclampsia miscarriage in our hospital from July 2021 to July 2022 were selected and randomly divided into two groups of 43 cases each. The control group was treated with progesterone, while the study group was treated with an addition of enhanced “Shou Tai Wan” soup. The treatment effects of the two groups and the changes in the inflammatory factor levels and sex hormone levels before and after the treatment were observed. Results: INF-? and Il-2 were lower and IL-4 was higher in the study group than in the control group after treatment (p-value < 0.05); the total effective rate of treatment was higher in the study group (95.35%) than in the control group (74.42%), (p-value < 0.05); the levels of P, E2 and ?-hCG were higher in the study group than in the control group after treatment (p-value < 0.05). Conclusion: The combined application of enhanced “Shou Tai Wan” soup and progesterone in patients with early preterm abortion is more effective by improving the patients’ sex hormone levels and reducing the level of inflammatory factors, which is conducive to the improvement of the treatment effect and worthy of promotion.

Many scientific applications can be well modeled as large-scale workflows. Cloud computing has become a suitable platform for hosting and executing them. Workflow scheduling has gained much attention in recent years. However, since cloud service providers must offer services for multiple users with various QoS demands, scheduling multiple applications with different QoS requirements is highly challenging. This work proposes a Multi-swarm Co-evolutionary-based Hybrid Optimization (MCHO) algorithm for multiple-workflow scheduling to minimize total makespan and cost while workflow deadline constraints. First, we design a multi-swarm co-evolutionary mechanism where three swarms are adopted to sufficiently search for various elite solutions. Second, to improving global search and convergence performance, we embed local and global guiding information into the updating process of a Particle Swarm Optimizer, and develop a swarm cooperation technique. Third, we propose a Genetic Algorithm-based elite enhancement strategy to exploit more non-dominated individuals, and apply the Metropolis Acceptance rule of Simulated Annealing to update the local guiding solution for each swarm so as to prevent it from being stuck into a local optimum at an early stage. Extensive experimental results demonstrate that MCHO outperforms the state-of-art scheduling algorithms with better distributed non-dominated solutions.

Zingiberis Rhizoma and Ginseng Radix et Rhizoma are usually used together for the treatment of ulcerative colitis in clinical practices. However, their compatibility mechanism remains unclear. In this study, a rapid and sensitive liquid chromatography with tandem mass spectrometry method was developed for simultaneous quantification of ginsenoside Re, ginsenoside Rg1, ginsenoside Rb1, and 6-gingerol in rat plasma after oral administration of Zingiberis Rhizoma-Ginseng Radix et Rhizoma herb pair and its single herb extracts. The calibration curves exhibited good linearity, with correlation coefficients of more than 0.993. The precision deviations of intra- and interday analysis were within 10.66%, and accuracy error ranged from -12.74 to 11.56%. The average recoveries of analytes were higher than 76.60% and the matrix effects were minimal. Thus, the validated method was successfully applied to a pharmacokinetic study of four ingredients in normal and ulcerative colitis rat plasma. The human androgen receptor plays a major role in the development and progression of prostate cancer and represents a well-established drug target. All clinically approved androgen receptor antagonists possess similar chemical structures and exhibit the same mode of action on the androgen receptor. Although initially effective, resistance to these androgen receptor antagonists usually develops and the cancer quickly progresses to castration-resistant and metastatic states. Yet even in these late-stage patients, the androgen receptor is critical for the progression of the disease. Thus, there is a continuing need for novel chemical classes of androgen receptor antagonists that could help overcome the problem of resistance. In this study, we implemented and used the synergetic combination of virtual and experimental screening to discover a number of new 10-benzylidene-10H-anthracen-9-ones that not only effectively inhibit androgen receptor transcriptional activity, but also induce almost complete degradation of the androgen receptor. Of these 10-benzylidene-10H-anthracen-9-one analogues, a lead compound (VPC-3033) was identified that showed strong androgen displacement potency, effectively inhibited androgen receptor transcriptional activity, and possesses a profound ability to cause degradation of androgen receptor. Notably, VPC-3033 exhibited significant activity against prostate cancer cells that have already developed resistance to the second-generation antiandrogen enzalutamide (formerly known as MDV3100). VPC-3033 also showed strong antiandrogen receptor activity in the LNCaP in vivo xenograft model. These results provide a foundation for the development of a new class of androgen receptor antagonists that can help address the problem of antiandrogen resistance in prostate cancer. Purpose: Malignant astrocytomas (MA) are aggressive central nervous system tumors with poor prognosis. Activating mutation of BRAF (BRAFV600E) has been reported in a subset of these tumors, especially in children. We have investigated the incidence of BRAFV600E in additional pediatric patient cohorts and examined the effects of BRAF blockade in preclinical models of BRAFV600E and wild-type BRAF MA.Experimental Design:BRAFV600E mutation status was examined in two pediatric MA patient cohorts. For functional studies, BRAFV600E MA cell lines were used to investigate the effects of BRAF shRNA knockdown in vitro, and to investigate BRAF pharmacologic inhibition in vitro and in vivo.Results:BRAFV600E mutations were identified in 11 and 10% of MAs from two distinct series of tumors (six of 58 cases total). BRAF was expressed in all MA cell lines examined, among which BRAFV600E was identified in four instances. Using the BRAFV600E-specific inhibitor PLX4720, pharmacologic blockade of BRAF revealed preferential antiproliferative activity against BRAFV600E mutant cells in vitro, in contrast to the use of shRNA-mediated knockdown of BRAF, which inhibited cell growth of glioma cell lines regardless of BRAF mutation status. Using orthotopic MA xenografts, we show that PLX4720 treatment decreases tumor growth and increases overall survival in mice-bearing BRAFV600E mutant xenografts, while being ineffective, and possibly tumor promoting, against xenografts with wild-type BRAF.Conclusions: Our results indicate a 10% incidence of activating BRAFV600E among pediatric MAs.
