Musculoskeletal ultrasound is a safe, reliable, and inexpensive alternative to other imaging modalities. Ultrasound is utilized by clinicians both diagnostically and therapeutically. It can be used to assess inflammatory and non-inflammatory conditions of the joints and peri-articular structures. Utilizing musculoskeletal ultrasound for invasive procedures, such as aspiration and injection of joints, bursa, and tendon sheaths, allows for improved precision and increases patient satisfaction.

Purpose: Advanced-stage mucinous ovarian carcinoma (MOC) has poor chemotherapy response and prognosis and lacks biomarkers to aid stage I adjuvant treatment. Differentiating primary MOC from gastrointestinal (GI) metastases to the ovary is also challenging due to phenotypic similarities. Clinicopathologic and gene-expression data were analyzed to identify prognostic and diagnostic features. Experimental Design: Discovery analyses selected 19 genes with prognostic/diagnostic potential. Validation was performed through the Ovarian Tumor Tissue Analysis consortium and GI cancer biobanks comprising 604 patients with MOC (n = 333), mucinous borderline ovarian tumors (MBOT, n = 151), and upper GI (n = 65) and lower GI tumors (n = 55). Results: Infiltrative pattern of invasion was associated with decreased overall survival (OS) within 2 years from diagnosis, compared with expansile pattern in stage I MOC [hazard ratio (HR), 2.77; 95% confidence interval (CI), 1.04–7.41, P = 0.042]. Increased expression of THBS2 and TAGLN was associated with shorter OS in MOC patients (HR, 1.25; 95% CI, 1.04–1.51, P = 0.016) and (HR, 1.21; 95% CI, 1.01–1.45, P = 0.043), respectively. ERBB2 (HER2) amplification or high mRNA expression was evident in 64 of 243 (26%) of MOCs, but only 8 of 243 (3%) were also infiltrative (4/39, 10%) or stage III/IV (4/31, 13%). Conclusions: An infiltrative growth pattern infers poor prognosis within 2 years from diagnosis and may help select stage I patients for adjuvant therapy. High expression of THBS2 and TAGLN in MOC confers an adverse prognosis and is upregulated in the infiltrative subtype, which warrants further investigation. Anti-HER2 therapy should be investigated in a subset of patients. MOC samples clustered with upper GI, yet markers to differentiate these entities remain elusive, suggesting similar underlying biology and shared treatment strategies.

Clinical guidelines for obtaining accurate scan data during the intraoral scanning of inlay cavities with various configurations and interproximal distances are lacking.The purpose of this in vitro study was to evaluate the effect of interproximal distance and cavity type on the accuracy of digital scans for inlay cavities, including proximal cavities.Four artificial teeth with 4 types of inlay cavities designed based on the buccolingual width and gingival level of the proximal box were installed in a mannequin at distances of 0.6, 0.8, and 1.0 mm from the adjacent teeth. Reference scans of the 4 artificial teeth were obtained by using a laboratory scanner. The CEREC Primescan AC was used to acquire digital scan data (each n=10). Standard tessellation language (STL) files were analyzed with a 3-dimensional analysis software program. The mean deviation values were measured with a 3-dimensional best-fit alignment method to evaluate the accuracy of the digital scan data. Statistical analyses were performed by using 2-way ANOVA and the Bonferroni multiple comparison test (α=.05).As per the interproximal distance, the 1.0-mm group showed significantly higher trueness than the 0.6-mm group (P.05). As the interproximal distance increased, the maximum positive deviation significantly decreased (P.05). Maximum negative deviation and precision of the scan data were not significantly different among the distance groups (P.05). Cavity type had a significant influence on the trueness and precision of the scan data (P.05). In particular, the narrow long cavity type had an adverse effect on the precision and maximum positive deviation of scan data.During the intraoral scanning of class II inlay restoration, interproximal distance and cavity type affected the accuracy of an intraoral scan. As the interproximal distance increased, the trueness of the acquired digital images increased and the maximum positive deviation significantly decreased. The narrow long cavity type negatively affected the mean maximum positive deviation and precision of scan data.

In this paper, we present BodyTrak, an intelligent sensing technology that can estimate full body poses on a wristband. It only requires one miniature RGB camera to capture the body silhouettes, which are learned by a customized deep learning model to estimate the 3D positions of 14 joints on arms, legs, torso, and head. We conducted a user study with 9 participants in which each participant performed 12 daily activities such as walking, sitting, or exercising, in varying scenarios (wearing different clothes, outdoors/indoors) with a different number of camera settings on the wrist. The results show that our system can infer the full body pose (3D positions of 14 joints) with an average error of 6.9 cm using only one miniature RGB camera (11.5mm x 9.5mm) on the wrist pointing towards the body. Based on the results, we disscuss the possible application, challenges, and limitations to deploy our system in real-world scenarios.

This study aimed to evaluate the stress distribution that results from temperature changes in teeth restored with gold inlay and to predict the fatigue life of the teeth. A thermal load regimen for finite element analysis (FEA) was established in an in vivo experiment, in which participants with gold inlays drank hot and cold water. An extracted human maxillary molar was converted into a 3D FEA model. The tooth models with Class I and II preparations were restored with gold inlay in the experimental groups. The stress distribution was analyzed under the thermal loads. Also, lifetime prediction was conducted using Basquin's formulation. The gold inlay groups showed a higher stress distribution than the preparation-only groups regardless of the type of preparation. The phenolics concentrations of rapeseed are important because they are antinutritional com-pound or directly positive related to antioxidant actives. In this study, single nucleotide poly-morphisms (SNPs) identified from genotyping-by-sequencing (GBS), and genome-wide associa-tion study of phenolic contents were investigated in 95 rapeseed mutant lines derived from gamma rays and their original cultivar. A total of 3,196,318 SNPs were detected, identifying a set of 70,208 union SNPs used to perform association studies. Six phenolics and glucosinolate (pro-goitrin) were identified, sinapine was the major phenolic compound (91.2–99.2%) in all geno-types. The association study was conducted for six phenolic compounds, progoitrin, total sinap-ine content (TSC), total phenolic content (TPC) and seed yield. The association study identified 241 SNPs that were significantly associated with six phenolic compounds, progoitrin, TSC and TPC. Based on the detected SNP markers, BnaA06g31740D, a gene associated progoitrin, BnaC-nng39930D, a gene associated trans-sinapine 1, BnaA07g31720D, a gene associated trans-sinapine 2, BnaC03g31950D and BnaA06g09180D, a gene associated TSC, and BnaAnng27700D, a gene associ-ated TPC, were selected as candidate genes. The SNP makers associated major phenolic com-pounds identified and could be used as targets for breeding programs aimed at improving anti-oxidant contents and reducing antinutritional components.

Background: During the cementation of indirect restorations, the conditions of the dentin surface and dentin adhesive's curing technique that affect the adhesion between dentin and an indirect restoration are essential issues for clinicians. This study investigates the effects of dentin's wetness, roughness, and curing modes of resin cement on bond strength when using universal adhesives for the cementation of indirect restorations. Methods: Forty human teeth were divided into eight groups according to three experimental factors: 1) dentin's roughness achieved by 250-or 600-grit Silicon-carbide (SiC) paper (coarse or fine), 2) dentin wetness with an air-drying time (5-s or 10-s), and 3) Single Bond Universal adhesive's curing mode by co-curing with RelyX Ultimate resin cement or light-curing separately after applying the adhesive(co-curing or light-curing). After resin discs were cemented with RelyX Ultimate, the microtensile bond strength (μTBS) was measured (n = 15 each). The failure mode and adhesive layers were observed using stereoscopic and confocal laser scanning microscopy (CLSM). Results: The roughness of the dentin surface did not significantly affect the bond strength (p > 0.05), and the curing mode of dentin adhesive and the 5-s drying group of dentin had significant effects on bond strength (p < 0.05). Light-curing groups and 5-s drying groups showed significantly higher bond strength than co-curing groups (p < 0.05), and 10-s drying groups (p < 0.05), respectively. Conclusions: When using Single Bond Universal adhesive and RelyX Ultimate resin cement, the light curing mode, in which the adhesive and resin cement is light-cured, showed higher bond strength than the co-curing mode. The bond strength between the resin cement and dentin was improved in the 5-s drying groups than in the 10-s drying groups.

Time reversal symmetry (TRS) breaking often generates exotic quantum phases in condensed matter. In superconductors, TRS breaking by an external magnetic field not only suppresses superconductivity but also leads to a novel quantum state called the gapless superconducting state. Here we show that magneto-terahertz spectroscopy provides us with a rare opportunity to access and explore the gapless superconducting state of Nb thin films. We present the complete functional form of the superconducting order parameter for an arbitrary magnetic field, for which a fully self-consistent theory is, surprisingly, yet unavailable. We observe a Lifshitz topological phase transition with a vanishing quasiparticle gap everywhere on the Fermi surface, whereas the superconducting order parameter smoothly crosses over from the gapped to the gapless regime. Our observation of the magnetic pair-breaking effects in Nb challenges traditional perturbative theories and opens a pathway to further exploring and manipulating the exotic state of gapless superconductivity.

Cisplatin resistance along with chemotherapy-induced neuropathic pain is an important cause of treatment failure for many cancer types and represents an unmet clinical need. Therefore, future studies should provide evidence regarding the mechanisms of potential targets that can overcome the resistance as well as alleviate pain. Here, we show that the emergence of cisplatin resistance is highly associated with EGFR hyperactivation, and that EGFR hyperactivation is arisen by a transcriptional increase in the pain-generating channel, TRPV1, via NANOG. Furthermore, TRPV1 promotes autophagy-mediated EGF secretion via Ca2+ influx, which activates the EGFR-AKT signaling and, consequentially, the acquisition of cisplatin resistance. Importantly, TRPV1 inhibition renders tumors susceptible to cisplatin. Thus, our findings indicate a link among cisplatin resistance, EGFR hyperactivation, and TRPV1-mediated autophagic secretion, and implicate that TRPV1 could be a crucial drug target that could not only overcome cisplatin resistance but also alleviate pain in NANOG+ cisplatin-resistant cancer.

Through the wide adaptation of next-generation sequencing (NGS) technology within clinical practice, molecular profiling of the tumor has been the principal component of personalized treatment. In our study, we have generated a large collection of cancer genomes on East Asian epithelial ovarian carcinoma (EOC) patients and demonstrate the feasibility and utility of NGS platforms to explore the dynamic interrelations of major cancer driver alterations and their impacts on clinical prognosis and management. A total of 652 EOC patients have undergone clinical NGS panels to determine the prevalence of germline and somatic mutations. Notably, TP53 was the most frequently altered event (73%), followed by both BRCA1 and BRCA2 (22% each) and MYC (19%) through pan-EOC analysis. When analyzed based on individual histopathological levels, TP53 mutation was highly dominant in high-grade serous and mucinous histology, whereas mutations in PIK3CA and ARID1A were mostly observed in clear cell carcinoma, and KRAS, BRAF, and CDKN2A mutations were enriched in endometrioid, low-grade serous, and mucinous tumors, respectively. The network-based probabilistic model showed significant co-occurrences of TP53 with BRCA1 and ALK with BRCA2, NOTCH1, and ROS1, whereas mutual exclusivity of TP53 with KRAS and PIK3CA was evident. Furthermore, we utilized machine-learning algorithms to identify molecular correlates that conferred increased sensitivity to platinum and olaparib treatments including somatic mutations in BRCA1, ATM, and MYC. Conversely, patients with ALK mutation were considerably resistant to both treatment modalities. Collectively, our results demonstrate the clinical feasibility of prospective genetic sequencing to facilitate personalized treatment opportunities for patients with EOC.

Aims: To evaluate BRAK and APRIL in serum samples from healthy patients and an ovarian tumor group and analyze their effective value as biomarkers. Materials & methods: BRAK and APRIL were measured in 197 serum samples including 34 healthy controls, 48 patients with benign ovarian cysts and 115 patients with ovarian cancer, and the best statistical cut-off values were calculated. Then, the sensitivity, specificity, accuracy, positive predictive value and negative predictive value for selected cut-off points were assessed. Results: The healthy control group had statistically significant higher BRAK and lower APRIL than the ovarian tumor group. BRAK was excellent for differentiating healthy patients from patients with ovarian tumors, showing area under the receiver operating characteristic curve 0.983, 98.16% sensitivity and 100% specificity. When BRAK was combined with APRIL and CA-125, it also played a role in distinguishing benign cysts from malignancies with area under the curve 0.864, 81.74% sensitivity and 79.17% specificity. Conclusions: BRAK and APRIL are good candidates for ovarian tumor biomarkers.

Digital feedback for tooth preparation can provide dental practitioners with more objective and accurate evaluations compared to conventional methods. This study aimed to evaluate the educational effect and satisfaction of digital feedback compared with those of the conventional putty index method for tooth preparation.Forty-eight third-grade dental students were selected. All students performed a full-coverage preparation on a right mandibular first molar resin tooth. They were randomly divided into four groups (n = 12)-no guide (control), putty index, digital feedback, and digital and putty index assessment. Three-dimensional analysis was performed using an analysis software (GomInspect 2018, Gom) to evaluate the amount of tooth structure removed. At the end of the practice, the students completed a questionnaire to evaluate the educational satisfaction of the respective methods.There was no statistical significance of the amount of preparation amongst groups in most of the measured areas only except for several specific points. Overall occlusal surface showed 0.99 ± 0.27 mm in the N group (no guide) and 1.15 ± 0.31 mm in the D group (digital feedback), and overall axial surface showed no statistical differences (p .05). The groups that used digital assessment showed a high level of satisfaction compared with conventional assessment.It was difficult to confirm that the digital-based feedback promotes accurate tooth preparation compared to conventional feedback within the limitation of this study. However, it improved educational satisfaction and permitted objective evaluation.

Cancer immunoediting drives the adaptation of tumor cells to host immune surveillance. Immunoediting driven by antigen (Ag)-specific T cells enriches NANOG expression in tumor cells, resulting in a stem-like phenotype and immune resistance. Here, we identify HDAC1 as a key mediator of the NANOG-associated phenotype. NANOG upregulated HDAC1 through promoter occupancy, thereby decreasing histone H3 acetylation on K14 and K27. NANOG-dependent, HDAC1-driven epigenetic silencing of cell-cycle inhibitors CDKN2D and CDKN1B induced stem-like features. Silencing of TRIM17 and NOXA induced immune and drug resistance in tumor cells by increasing antiapoptotic MCL1. Importantly, HDAC inhibition synergized with Ag-specific adoptive T-cell therapy to control immune refractory cancers. Our results reveal that NANOG influences the epigenetic state of tumor cells via HDAC1, and they encourage a rational application of epigenetic modulators and immunotherapy in treatment of NANOG+ refractory cancer types.
