HESS J1809$-$193 is one of the unidentified very-high-energy gamma-ray sources in the H.E.S.S. Galactic Plane Survey (HGPS). It is located in a rich environment, with an energetic pulsar and associated X-ray pulsar wind nebula, several supernova remnants, and molecular clouds in the vicinity. Furthermore, HESS J1809$-$193 was recently detected at energies above 56 TeV with HAWC, which makes it a PeVatron candidate, that is, a source capable of accelerating cosmic rays up to PeV energies. We present a new analysis of the TeV gamma-ray emission of HESS J1809$-$193 with H.E.S.S., based on improved analysis techniques. We find that the emission is best described by two components with distinct morphologies and energy spectra. We complement this study with an analysis of Fermi-LAT data in the same region. Finally, taking into account further multi-wavelength data, we interpret our results both in a hadronic and leptonic framework., Comment: 6 pages, 5 figures. 4. Comparison of fentanyl and nalbuphine in obtunding stressor response to laryngoscopy and endotracheal intubation

Objective: Fentanyl has evinced its property of being a potent opioid and benumbing stress response to laryngoscopy and intubation (L&I). However respiratory depression and chest wall rigidity limits its utility at high doses. Nalbuphine, reported to have a ceiling effect for respiratory depression, can serve as an alternative.We studied and compared the efficacy of fentanyl and nalbuphine in preventing the rise in heart rate and blood pressure after L&I. Materials and Methods: A randomized double blind study was conducted in 60 subjects divided in two groups (Group F n=30 and Group N n=30) was done. Group F received 2µg kg-1 fentanyl intravenously while group N received nalbuphine 0.2mg kg-1 intravenously. Hemodynamic variables including heart rate (HR), systolic blood pressure (SBP), diastolic blood pressure (DBP) and mean arterial pressure (MAP) were measured at baseline and at intervals of five minutes of administering test drug, each minute up to five minutes after intubation, 10 minutes and 30 minutes after intubation. Complications, if any, were recorded after surgery. Change in HR, SBP, DBP and MAP between the two groups was compared using independent t-test. Results: The two drugs were equally effective in preventing HR rise at all-time points except after 30 minutes where fentanyl performed better. The change in SBP, DBP and MAP from baseline in both the groups was comparable. Conclusions: Nalbuphine can serve as an alternative to fentanyl in obtunding stress response when given five minutes before intubation. Keywords: Nalbuphine, Fentanyl, Intubation.

Nowadays the implementation of artificial neural networks in high-energy physics has obtained excellent results on improving signal detection. In this work we propose to use neural networks (NNs) for event discrimination in HAWC. This observatory is a water Cherenkov gamma-ray detector that in recent years has implemented algorithms to identify horizontal muon tracks. However, these algorithms are not very efficient. In this work we describe the implementation of three NNs: two based on image classification and one based on object detection. Using these algorithms we obtain an increase in the number of identified tracks. The results of this study could be used in the future to improve the performance of the Earth-skimming technique for the indirect measurement of neutrinos with HAWC., Comment: 8 pages, 6 figures, Proceedings of the 37th International Cosmic Ray Conference (ICRC 2021)

The realtime follow-up of neutrino events is a promising approach to search for astrophysical neutrino sources. It has so far provided compelling evidence for a neutrino point source: the flaring gamma-ray blazar TXS 0506+056 observed in coincidence with the high-energy neutrino IceCube-170922A detected by IceCube. The detection of very-high-energy gamma rays (VHE, $\mathrm{E} > 100\,\mathrm{GeV}$) from this source helped establish the coincidence and constrained the modeling of the blazar emission at the time of the IceCube event. The four major imaging atmospheric Cherenkov telescope arrays (IACTs) - FACT, H.E.S.S., MAGIC, and VERITAS - operate an active follow-up program of target-of-opportunity observations of neutrino alerts sent by IceCube. This program has two main components. One are the observations of known gamma-ray sources around which a cluster of candidate neutrino events has been identified by IceCube (Gamma-ray Follow-Up, GFU). Second one is the follow-up of single high-energy neutrino candidate events of potential astrophysical origin such as IceCube-170922A. GFU has been recently upgraded by IceCube in collaboration with the IACT groups. In this research work, the problem is to reduce the surface roughness of the Jacquard body which is a casting part to optimize the effects of different process parameters of surface roughness & to reduce surface roughness for milling operation by using the Taguchi method. Milling is better than other machines in the surface finish and accuracy. Greater quality product and higher productivity is the main vision of every entrepreneur. To achieve this, the process parameters of each process need to be optimized. In a milling machine, some of the important parameters like feed rate, depth of cut, speed, etc, play an important role in achieving the required quality product. To optimize these parameters, the most suitable and widely used method is the Taguchi method. The most significant objective of this research is to obtain effective quality parameters. Experimentation is done on the Jacquard body by varying the milling machine parameters. The experimental results indicate that the minimum surface roughness is obtained for the process parameters feed 0.7 mm per revolution, speed 225 rpm and 1.5 mm depth of cut. This study recommended that more surface finish be obtained by increasing speed, low feed rate, and depth of cut.

Gamma-ray bursts (GRBs) are among the most luminous sources in the universe. The nature of their emission at TeV energies is one of the most relevant open issues related to these events. The temporal and spectral features inferred from the early and late emissions usually known as prompt and afterglow, respectively, can be interpreted within the context of the fireball model. The synchrotron self-Compton process is expected during the afterglow phase. We explain how the theoretical SSC light curves can be compared with hypothetical upper limit located at z=0.3. We show the allowed parameter space of the microphysical parameters and density of the circumburst medium. The majority of the active galactic nuclei (AGN) detected at very-high-energies above 100 GeV belong to the class of blazars with a small angle between the jet-axis and the line-of-sight. Only about 10 percent of the gamma-ray AGN are objects with a larger viewing angle resulting in a smaller Doppler boosting of the emission. Originally, it was believed that gamma-ray emission can only be observed from blazars and those are variable in its brightness. Instead, the last years have shown that non-blazar active galaxies also show a fascinating variability behaviour which provide important new insights into the physical processes responsible for the gamma-ray production and especially for flaring events. Here, we report on the observation of gamma-ray variability of the active galaxy PKS 0625−354 detected with the H.E.S.S. telescopes in November 2018. The classification of PKS 0625−354 is a still matter of debate. The H.E.S.S. measurements were performed as part of a flux observing program and showed in the first night of the observation a detection of the object with > 5σ. A denser observation campaign followed for the next nine nights resulting in a decrease of the gamma-ray flux. Those observations were accompanied with Swift in the X-ray and UV/optical band allowing for the reconstruction of a multi-band broad-band spectral energy distribution. We will discuss the implications of the gamma-ray variability of the object.

Studies of radio galaxies at TeV energies are fascinating because their jets are misaligned concerning our sightline. Thus, it provides us with a unique opportunity to study the structure of their jets, the radiative processes, and the acceleration mechanisms involved in them. In addition, some radio galaxies have presented variability in their emission, like the giant radio galaxy M87, which has reported several activity periods. Due to its duty cycle >95% and instantaneous field of view of 2 sr, HAWC provides daily monitoring of variable sources visible from the Northern Hemisphere. In this work, we show the results of monitoring M87 between January 2015 and December 2018. HAWC's observations are consistent with the low activity state reported by other instruments (like H.E.S.S and MAGIC). The presence of dark matter (DM) is suggested by a wealth of astrophysical and cosmological measurements. However, its underlying nature is yet unknown. Among the most promising candidates are weakly interacting massive particles (WIMPs): particles with mass and coupling strength at the electroweak scale and thermally produced in the early universe have a present relic density consistent with that observed today. WIMP self-annihilation would produce Standard Model particles including gamma-rays, which have been long-time recognized as a prime messenger to indirectly detect dark matter signals. The centre of the Milky Way is predicted as the brightest source of DM annihilations. The H.E.S.S. collaboration is currently performing a survey of the inner region of the Milky Way, the Inner Galaxy Survey (IGS), intended to achieve the best sensitivity to faint and diffuse emissions in a region of several degrees around the Galactic Centre. We analyzed 2014-2020 observations taken with the five-telescope array to search for a DM annihilation signal. With the current dataset of about 550 hours, we found no significant excess and therefore derived strong constraints on the velocity-weighted annihilation cross-section. TeV thermal WIMPs can be probed in different annihilation channels.

M87 is a giant radio galaxy located in the Virgo Cluster, known to be a very high energy (VHE) gamma-ray source. As radio galaxies are considered the misaligned low-redshift counterparts of blazars, they are excellent laboratories for testing AGN emission models. M87 has been detected and monitored by Fermi-LAT and several atmospheric Cherenkov telescopes. Recently, the HAWC Collaboration presented marginal evidence of 4.5 year TeV gamma-ray emission from this object. The HAWC data has the potential to constrain the average VHE emission of sources of complex behavior, like M87, for which the physical origin of the VHE gamma-ray emission is still uncertain. We fitted a lepto-hadronic scenario to the broadband spectral energy distribution of M87 to model its non-flaring VHE emission using HAWC data.

eHWC J1825-134 is one of the brightest Galactic 𝛾-ray sources above 50 TeV observed by High Altitude Water Cherenkov 𝛾-Ray Observatory (HAWC). Detailed morphological studies have revealed a new point-like source inside this region with a spectral energy distribution extending beyond 200 TeV without any cutoff. These very-high-energy 𝛾 rays emission can originate from leptonic or hadronic processes. The new point-like source is located in a region containing PWNe and a high-density giant molecular cloud [MML2017]99. If the source emission is associated with the hadronic scenario the TeV 𝛾 rays may have been produced by cosmic rays colliding with ambient gas. If this were the case, eHWC J1825-134 is an indicator of the existence of a galactic PeVatron in the region that accelerates particles up to PeV energies.

We present the observation of an anomalous increment of the galactic cosmic ray (GCR) flux observed by the HAWC array during October 2016. We propose that this anisotropy of the GCR flux was caused by a magnetic flux rope (MFR) i. e., by the helicoidal magnetic field of an interplanetary coronal mass ejection observed at 1 AU at the same time. We computed the trajectory of protons with energy in the range of 10 to 60 GeV, traveling inside the helicoidal magnetic field observed in situ. The direction of these particles changes towards the axis of the MFR resulting in an anisotropy of the GCR flux along this axis. This model shows that the alignment between the MFR axis and the HAWC s asymptotic direction, combined with the high sensitivity of HAWC, allowed us to observe the effect of the passage of the MFR on the GCR flux. We present the HAWC observation associated with the passage of the MFR as well as the helio- spheric circumstances around such phenomenon.

The Cygnus Cocoon region is a complex region containing an OB star cluster that is prominent in the TeV energy range. Located in this region is 3HWC J2031+415, a significant TeV gamma-ray source whose emission is possibly associated with 2 components, the Cygnus OB2 star cluster and a pulsar wind nebula (PWN). In this work, several modelling methods are presented to best describe the emission. These models disentangle emission believed to be from the Cocoon and isolate the component emitted by the probable PWN. I will present several spectral models to describe the emission of the probable PWN using the latest data set from the High-Altitude Water Cherenkov (HAWC) observatory. Furthermore, I will present an energy morphology study of the PWN component of 3HWC J2031+415 in distinct energy bands.

The High Altitude Water Cherenkov (HAWC) Observatory is a wide-field-of-view gamma-ray observatory that is optimized to detect gamma rays between ~300 GeV and several hundred TeV. The HAWC Collaboration recently released their third source catalog (3HWC), which contains 65 sources. One of these sources, the ultra-high-energy gamma-ray source 3HWC J1908+063, may exhibit a hardening of the spectral index at the highest energies (above 56 TeV). At least two populations of particles are needed to satisfactorily explain the highest energy emission. This second component could be leptonic or hadronic in origin. If it is hadronic in origin, it would imply the presence of protons with energies up to $\sim$1 PeV near the source. We have searched other 3HWC sources for the presence of this spectral hardening feature. If observed, this would imply that the sources could make good PeVatron candidates.

The TeV gamma-ray source MGRO J1908+06 is one of the highest-energy sources known, with observed emission by the High Altitude Water Cherenkov (HAWC) Observatory extending well past 100 TeV. The source exhibits both energy-dependent morphology and a spatially-dependent spectral index. The emission is likely to be dominantly leptonic, and associated with the radio-quiet PSR J1907+0602. However, one-population models do not describe the data well; a second particle population is needed to explain the shape of the spectral energy distribution at the highest energies. This component can be well-described by either leptonic or hadronic hypotheses. We discuss this feature and implications for detection by multi-wavelength and multi-messenger experiments.

Objective Minor surgical procedures under general anaesthesia require a patent airway without the use of muscle relaxant. Supraglottic airway devices have been widely used for airway management. A study was undertaken to compare first-time insertion success rate, insertion time, sealing pressure and complications between the Baska® mask and I-gel. Methods After approval from the institutional ethical committee, a randomised single-blinded study was conducted on 50 American Society of Anesthesiologists' physical status I and II female patients aged 18-40 years who underwent minor surgical procedures under general anaesthesia. Patients were randomly categorized into two groups of 25 each; group Baska® mask and group I-gel, and the first-time success rate, mean insertion time and sealing pressure were measured. Background showers triggered by hadrons represent over 99.9% of all particles arriving at ground-based gamma-ray observatories. An important stage in the data analysis of these observatories, therefore, is the removal of hadron-triggered showers. Currently, the High-Altitude Water Cherenkov (HAWC) gamma-ray observatory employs an algorithm based on a single cut in two variables, unlike other ground-based gamma-ray observatories (e.g. H.E.S.S., VERITAS), which employ a large number of variables to separate the primary particles. In this work, we explore machine learning techniques (Boosted Decision Trees and Neural Networks) to identify the primary particles detected by HAWC. The paper also highlights performing of optimization using various performance indices. Another issue that is discussed in the paper is decentralized control.

A wide range of data formats and proprietary software have traditionally been used in gamma-ray astronomy, usually developed for a single specific mission or experiment. However, in recent years there has been an increasing effort towards making astronomical data open and easily accessible. Within the gamma-ray community this has translated to the creation of a common data format across different gamma-ray observatories: the "gamma-astro-data-format" (GADF). Based on a similar premise, open-source analysis packages, such as Gammapy, are being developed and aim to provide a single, robust tool which suits the needs of many experiments at once. In this contribution we show that data from the High-Altitude Water Cherenkov (HAWC) observatory can be made compatible with the GADF and present the first GADF-based production of event lists and instrument response functions for a ground-based wide-field instrument. We use these data products to reproduce with excellent agreement the published HAWC Crab spectrum using Gammapy. Having a common data format and analysis tools facilitates joint analysis between different experiments and effective data sharing. The present article reviews the literature pertaining to the control system analysis of multivariable process. MIMO control system analysis techniques like relative gain array (RGA), Neiderlinski Index , Morari Resiliency Index , Condition Number (CN), Singular Value Decomposition and robustness analysis (RA) using model mismatch are shown with the help of flow chart. In multivariable control system analysis, the important terms include interaction analysis, stability analysis, controllability test, observability test, singular value decomposition, resiliency and conditionality. Also, the capabilities such as tracking of set-point level, rejection of disturbance signals and robustness are explored for the controllers of MIMO processes.

The economy of the paper industry largely depends on the quality of the paper manufactured and the quality depends on the precise operation of Headbox, which is the key element of the paper machine and is highly responsible for the quality of the paper manufactured. Being a nonlinear multivariable system, a headbox needs to be controlled efficiently to have the desired response which in turn reflects in the quality of the paper. The headbox is subjected to various disturbances and thus it is required not only to suppress the effect of disturbances but also to ensure the smooth start of the headbox and fast response to the set-point changes. A control technique is proposed in this paper for a paper machine headbox based on the determination of ultimate gain and frequency using the fractional-order integrator (FOI). The PID tuning technique proposed has been applied on paper machine headbox and the responses are compared with the existing techniques.

Background:Intussusception is a condition in which one part of gut enters into another part of gut known as telescoping. Ultrasonography of abdomen is highly specific and sensitive for diagnosis. Most of the ileo-ileal and fewer ileo-colic intussusceptions may reduce spontaneously. Ultrasound guided reduction is an alternative to surgery. If intussusception is not treated timely it may cause intestinal ischemia, gangrene, perforation peritonitis, shock and death. Method:This is a hospital based prospective study including 96 patients over a period of 18 months. Detailed proforma was used, including name, age, sex, weight, presenting complaints, duration of complaints, abdominal sign & symptoms, signs of the sepsis & shock, peritonitis, perforation, USG finding, and type of surgery performed. The presence of the complications also noted. Results:Maximum patients (63.5%) of intussusception diagnosed were between 6 months to two years. Out of total 96 patients, 21 were treated by pneumatic reduction successfully. In 46 patient exploratory laparotomy and intra-operative reduction done, while in 29 children required resection anastomosis. Most common complaint was abdominal pain (100%) followed by Excessive cry (92.7%). Most common type of intussusception was Ileo-colic(88.5%). Respiratory distress (23.9%) followed by septicemia (20.8%) was the most common complications. Conclusions:Intussusception is a common surgical problem in childhood. Children of age 6 month to 2 years of age are affected most commonly. Ileo-colic is the most common type. Ultrasonography is diagnostic as well as therapeutic tool for intussusception. USG guided pneumatic reduction is a simple and good technique, if patient comes to hospital earlier. Early diagnosis of signs and symptoms may help clinician to take early interventions to save the gut as well life of the patient.

Present paper attempts to review the literature related to various strategies to design the control system for the Lime kiln industrial process. Lime Kiln is multivariable process with complex interactions of variables. It has higher degree of coupling, severe non linearities, and frequent disturbances. This paper covers the modeling and control aspects of Lime Kiln process. The lime kiln control in recovery section of paper mill along with heat and mass transfer in reaction, radiation, and convection in limekiln is also presented. This paper also explores the possibilities of improving the control of the lime kiln by reviewing the literature of various control techniques including both the conventional and the modern ones.

The mixture of solid particles and gases is air pollution. Sot, smog, mercury, plumage, dioxins, carbon dioxide etc, emissions of gas / carbon, vehicle fuels, industrial emissions could be the pollutants of air. These pollutants are the main cause for lung damage, kidneys, brain inhalation, eyes and throat irritation. If a significant amount of carbon dioxide plays a role in air pollution, it can seriously damage life. It is a greenhouse gas that allows the atmosphere to burn. This affects most people who work outside. Based on the reasons explained, air pollution must be controlled and air quality improved. However, in reality, we can not stop plying vehicles, industry and air pollution completely, but we can try to reduce emissions and therefore reduce the impact to make things better. Air pollution reached critical levels at the end of the 20th century and continues to grow even in the 21st century. Although the governments have taken several steps to reduce air pollution, a great deal still needs to be done to monitor contaminants. In this paper, we have attempted to classify the top 10 countries that emit large amounts of carbon dioxide from different sources. These include both developed and developing countries.

Transfer learning (TL) is widely used in conventional hybrid automatic speech recognition (ASR) system, to transfer the knowledge from source to target language. TL can be applied to end-to-end (E2E) ASR system such as recurrent neural network transducer (RNN-T) models, by initializing the encoder and/or prediction network of the target language with the pre-trained models from source language. In the hybrid ASR system, transfer learning is typically done by initializing the target language acoustic model (AM) with source language AM. Several transfer learning strategies exist in the case of the RNN-T framework, depending upon the choice of the initialization model for encoder and prediction networks. This paper presents a comparative study of four different TL methods for RNN-T framework. We show ~10%-17% relative word error rate reduction with different TL methods over randomly initialized RNN-T model. We also study the impact of TL with varying amount of training data ranging from 50 hours to 1000 hours and show the efficacy of TL for languages with a very small amount of training data.

Background: Vaso-occlusive crises (VOCs) are the hallmark of SCD. VOCs and silent vaso-occlusion can lead to complications (eg acute chest syndrome, hepatic/renal dysfunction, chronic pain, multi-organ failure) and premature death. Crizanlizumab, an anti-P-selectin monoclonal antibody (mAb), is authorized in >40 countries to prevent/reduce VOCs in SCD pts aged ≥16 yrs. IRRs are defined as any signs/symptoms (S/S) experienced by pts during/within 24 hrs of infusion of a pharmacologic/biologic agent. IRRs are quite common with mAbs (frequency 1.6‒99%; Rombouts et al Anticancer Res 2020). S/S of IRRs vary; pain events, such as headache, back pain, myalgia, chest pain and joint pain, have been described as S/S of IRRs. Although pain events are known adverse drug reactions in the crizanlizumab label (eg arthralgia, myalgia, pain at various locations), due to data limitations and confounding manifestations of SCD, pain events occurring during/within 24 hrs of crizanlizumab infusion in SUSTAIN were not identified as potential IRRs (Ataga et al N Engl J Med 2017). For pts receiving crizanlizumab, IRR-related pain events may differ in location, severity and/or nature from a pt's usual SCD/VOC pain. Aim: To review data on IRRs presenting as pain events in SCD pts treated with crizanlizumab via reports received by Novartis since approval in Nov 2019. Methods: Data sources included PM reports from providers (spontaneous) and reports from the managed access/pt orientation program. To obtain the reports (which could include ≥1 event), a cumulative custom search of the Novartis safety database was performed up to Jun 2021, using ~111 MedDRA terms associated with potential S/S of IRRs presenting as pain events. IRRs must have occurred during/within 24 hrs of the most recent crizanlizumab infusion, and pain could differ from a pt's usual SCD/VOC, with/without other S/S. IRR incidence was measured by the reporting rate (RR). Reports were not gathered via a uniform data collection system, so there are limitations, including potential underreporting, incompletely documented cases, or bias towards reporting severe events. Results: IRRs presenting as pain events were experienced by 28 pts (Table 1); the most common S/S were back pain, pain in extremity, arthralgia, musculoskeletal chest pain and headache. RR was 1.67 cases per 100 pt-yrs (95% CI 1.11‒2.42). Most pts (n=24) initially experienced IRR at the 1st or 2nd infusion, and the majority recovered within 3 days. IRR recurred on subsequent infusion(s) in 6 pts. Of the 28 pts, 20 (71%) were hospitalized for further treatment, including analgesics, antihistamines, IV fluids and/or steroids. Nine pts (32%) reported SCD complications after IRR (Table 2). Crizanlizumab was discontinued in 23 pts (82%) after their most recent IRR occurrence, including all pts who experienced secondary SCD complications. Discussion: Comprehensive investigation identified 28 pts with reported IRRs presenting as pain events that had a potential causal relationship with crizanlizumab infusion based on temporality. All pts recovered or are recovering, except 1 who had SCD complications and refused blood transfusions for personal reasons. Most pts had initial IRR at the 1st or 2nd infusion and discontinued crizanlizumab after initial IRR; 6 experienced recurrent IRRs on subsequent infusion(s). Resolution time was prolonged for pts who reported known SCD complications following IRR. Causal association of complications following IRR was confounded by the underlying disease and use of steroids to treat IRRs. Systemic corticosteroid exposure in SCD pts has been associated with pain and other complications, from severe VOCs to hemorrhagic stroke and death. No data are available regarding whether the 28 pts had an active VOC or other SCD complications prior to receiving crizanlizumab. Conclusions: Although rare, based on review of PM data, healthcare professionals should be aware of the possibility of IRRs presenting as pain events during or after any crizanlizumab infusion. Crizanlizumab labels have been/are being updated by Novartis to provide information on monitoring for S/S of IRRs presenting as pain events, and guidance on management/prevention of subsequent IRRs, including a statement recommending caution when using corticosteroids in SCD pts. Given the limited data regarding IRRs and predictability of complications, Novartis is committed to further understanding these events. Figure 1 Figure 1. In this work, various algorithms of PID (Proportional-Integral-Derivative) controllers such as series, parallel, series together with derivative filter and parallel together with derivative filter form are implemented for the comparative analysis of the closed loop responses of an important parameter in head box of a paper machine in a paper making process industry called consistency, which has FOPDT (First Order Plus Dead Time) dynamics. Also the comparison is made between the closed loop responses achieved from different values of derivative filter factor for non-ideal situations. The steady state and dynamic characteristics are analyzed and compared and important inferences are derived.

There is a growing interest in the development of chameleon textiles using chromic dyes (inks) for several uses, such as brand protection, responsive camouflage, ultraviolet (UV) sensors, and fashion apparel. However, their use is limited to very few colors and responsivity to color change in the spectral range. This investigation used two photochromic inks: magenta and turquoise. These inks change color when exposed to certain wavelengths of light. The magenta and turquoise photochromic inks belong to the naphthopyran and spironaphthoxazine classifications of dye, respectively. These plastisol inks were applied on the nylon and polyester fabrics using a screen-printing technique. The color-changing behavior of the printed fabrics was studied using a Hunter Lab Color Spectrophotometer after exposure to tungsten, fluorescent, UV, and sunlight. Remogliflozin etabonate is an orally available prodrug of remogliflozin, an inhibitor of renal sodium glucose co-transporter-2 (SGLT2) with antihyperglycemic activity. The present study was conducted to characterize the pharmacokinetic and safety profile of remogliflozin etabonate under fasting and fed conditions at single oral doses of 100 and 250 mg in healthy Asian Indian adults. Sixty-five healthy, adult Asian Indian male subjects were enrolled in an open-label, two-stage, single-period pharmacokinetic study. Remogliflozin was given under fasting and/or fed conditions as a single oral dose of 100 or 250 mg. The plasma concentrations of remogliflozin etabonate, remogliflozin, and the metabolite GSK279782 were quantified by validated liquid chromatography–tandem mass spectrometry (LC–MS/MS) method. Pharmacokinetic parameters were determined from the plasma concentration–time profile by non-compartmental analysis. Safety was assessed through monitoring of adverse events. Descriptive statistics were calculated and reported for all parameters. The plasma concentration profiles showed rapid absorption of the prodrug remogliflozin etabonate and rapid conversion to the active moiety, remogliflozin, which is then further metabolized to another active metabolite, GSK279782. The geometric mean maximum concentration (Cmax) and area under the plasma concentration–time curve (AUC) were comparable for all three analytes between the fasted and fed state. The fed/fasted ratio for Cmax ranged from 0.77 to 1.44 at the 100 mg dose, and from 0.80 to 1.12 at the 250 mg dose. The fed/fasted ratio for AUC was 1.22 and 1.35 at 100 and 250 mg, respectively. An early time to Cmax (tmax) was observed for all three analytes while being administered in the fasted state. Both the Cmax and AUClast of all the three analytes increased in a dose-proportional manner under the fasted and fed states. The terminal half-life for remogliflozin ranged from 1.53 to 2.07 h. All three analytes had comparable terminal half-lives irrespective of dose levels or dietary conditions. Following single oral administration at 100 and 250 mg, remogliflozin etabonate showed favorable, linear pharmacokinetics. There were no clinically relevant food effects on the pharmacokinetics at both the 100 and 250 mg dose levels. Remogliflozin etabonate was well-tolerated without any safety concerns or hypoglycemic events. Clinical trial registration: Clinical Trial Registry–India identifier number CTRI/2017/10/010043.
