We present the results of a study of the connectivity between Pacific cod spawning and nursery areas, and settlement of Pacific cod in the Gulf of Alaska. This work was conducted to address the hypothesis that spatial and temporal patterns of recruitment are related to variability in connectivity between spawning and nursery areas. To examine this hypothesis, we developed a Lagrangian, biophysical, individual-based model of Pacific cod early life history and dispersal using the Dispersal Model for Early Life Stages (DisMELS) framework. This model is driven by currents and scalars such as temperature from a version of the Regional Oceanographic Model System (ROMS) developed for the Gulf of Alaska. Results of our study show connectivity patterns predicted by the model that agree with our understanding (based on genetic analyses) that there is a high degree of localized retention in Pacific cod. The results indicate that the Shumagin Islands and Prince William Sound regions may serve as important collectors of Pacific cod recruits from upstream spawning areas. We also find correlations between individual-based model outputs and several large-scale climate indicators that appear to show settlement in several important nursery areas, and recruitment overall, are positively affected by slower gyre circulation in the Gulf of Alaska. We hypothesize that this is due to enhancement of retention, settlement in the Shumagin Island region, and reduction of transport of young cod out of the Gulf of Alaska to the southwest. Abstract: Modeling the coastal Gulf of Alaska (CGOA) is complicated by the highly diverse physical and biological features influencing productivity and energy flow through the region. The GOA consists of the offshore oceanic environment, characterized by iron limitation, high-nutrients and low-chlorophyll. The coastal environment is consistently downwelling, with high iron levels from glacial melt water and runoff, but lower concentrations of macronutrients, and with a spring bloom, nutrient depletion cycle (low-nutrient, high-chlorophyll). Cross-shelf movement of water masses mixes coastal and oceanic ecosystem elements. Simulations and field data indicate that the minimum model complexity necessary to characterize lower trophic-level production and biomass in the offshore and coastal regions includes 10 boxes: iron, nitrate, ammonium, small phytoplankton, large phytoplankton, small microzooplankton, large microzooplankton, small copepods, large oceanic copepods and detritus, with copepod mortality as a model closure term. We present the model structure, equations required (and initial parameters used) to simulate onshore and offshore lower trophic-level production in the Gulf of Alaska, along with the information from field data and simulations used to construct the model. We show the results of simulations with and without iron, and with and without two size classes of phytoplankton. These simulations indicate that our method of inclusion of iron works well to distinguish the coastal and the oceanic ecosystems, and that the inclusion of two size categories of phytoplankton is also necessary to generate the differences between these two ecosystems. A three-dimensional biophysical nutrient–phytoplankton–zooplankton model was used to investigate the spatial and temporal dynamics of food resources for young walleye pollock in the western Gulf of Alaska, to further understanding of recruitment processes for pollock. We modeled nitrogen, phytoplankton, a large herbivorous grazer parameterized as Neocalanus spp. (the biomass dominant copepod in the Gulf), and the 13 stages (egg, naupliar and copepodite) of Pseudocalanus spp. (a major constituent of the diet of pollock) so that the appropriate size class of food for each size of larval pollock was represented. Model results identified an area between the Semidi and Shumagin Islands that may not be suitable as a nursery area early in the year due to low prey abundance. Modeled mesoscale eddies, previously hypothesized to be important for larval pollock retention in Shelikof Strait, contained higher prey concentrations than the surrounding waters when they were cyclonic. This work also help to understand the consistency of pollock spawning in time and space in Shelikof Strait, by examining the timing and location of prey availability which, along with transport, narrows the window for optimal spawning. This paper gives a review of acoustic and ultrasonic optical fiber sensors (OFSs). The review covers optical fiber sensing methods for detecting dynamic strain signals, including general sound and acoustic signals, high-frequency signals, i.e., ultrasonic/ultrasound, and other signals such as acoustic emissions, and impact induced dynamic strain. Several optical fiber sensing methods are included, in an attempted to summarize the majority of optical fiber sensing methods used to date. The OFS include single fiber sensors and optical fiber devices, fiber-optic interferometers, and fiber Bragg gratings (FBGs). The single fiber and fiber device sensors include optical fiber couplers, microbend sensors, refraction-based sensors, and other extrinsic intensity sensors. The optical fiber interferometers include Michelson, Mach-Zehnder, Fabry-Perot, Sagnac interferometers, as well as polarization and model interference. The specific applications addressed in this review include optical fiber hydrophones, biomedical sensors, and sensors for nondestructive evaluation and structural health monitoring. Polycrystalline CdS films, with thicknesses typically 20–180 nm, have been chemically deposited on glass substrates using an ammonia–cadmium–thiourea reaction solution. Film elemental composition, thickness and microstructure have been examined using proton-induced X-ray emission, Rutherford backscattering and atomic force microscopy. Analysis indicates that the stability of the deposition temperature plays a critical role in CdS film growth and composition. Films deposited with high temperature stability (60±0.5 °C) show a consistent 1:1 Cd:S atomic ratio for all stages of film growth, and have good substrate adhesion. Films deposited with lower temperature stability (60±4 °C) show initial high S concentrations, followed by a rapid increase in Cd concentration, until a final 1.2:1 Cd:S ratio is achieved. Little is known about the mechanism of transport that enables age-0 sablefish (Anoplopoma fimbria) to reach suitable nursery sites from spawning locations far offshore, or the strength of the connection between individual spawning sites and nursery areas, or how variability in the strength of these connections may impact recruitment success. Using a model for the early life stages of sablefish, we explored the variability in connectivity between spawning and recruitment sites that can arise solely from interannual variability in environmental forcing and its impact on transport. Our major findings are that 1) the model indicates young sablefish settling in nursery areas in the Gulf of Alaska were most likely spawned in the eastern Gulf; 2) sablefish spawned in the western Gulf of Alaska are unlikely to settle anywhere in the Gulf, and are more likely to be advected farther west, perhaps to settle in the Aleutian islands or Bering Sea (to contribute to the Alaska population, they would have to undergo an active return migration as they mature); 3) total connectivity between all spawning sites and nursery areas showed stronger correlation with recruitment estimates than the strength of connections to or from specific regions; and 4) transport to St. John Baptist Bay, a known sablefish nursery area, was not the most probable end point for sablefish spawned throughout our Gulf of Alaska model domain. This suggests that young individuals arrive at this persistent nursery area due to directional swimming behavior, highly localized spawning, or small-scale currents not captured in the hydrographic model. The fact that no single correlate in our analysis had a very strong relationship to sablefish recruitment indicates that recruitment variability arises from complex interactions between the environment and the individual, and a possible disconnect in spatial scales between the Gulf of Alaska sablefish IBM and the broader sablefish stock assessment, which includes both the GOA and the Eastern Bering Sea, as well as possible contributions from Canadian stocks to the south. Our analyses determined that although the timing and extent of this transport shows significant interannual variability, both the location of likely sablefish spawning (source) areas and the comparative strength of connectivity between spawning and nursery sites appear to be relatively consistent year-to-year. Backgrounds: Milk of magnesia (MoM) has been reported to accelerate return of bowel function following surgery. However, there is insufficient evidence regarding the impact of MoM on postoperative recovery after colorectal surgery. We aimed to determine the impact of MoM on postoperative length of stay in patients undergoing colorectal surgery., Methods: All patients who underwent colorectal resection without an ileostomy between 2015 and 2018 were included. Patients were divided into two groups based on whether postoperative MoM (MoM) was administered or not, according to surgeons' prescribing preferences. Consecutive patients of surgeons who prescribe MoM were included in the MoM group, while consecutive patients of surgeons who prefer not to prescribe MoM served as the control group. Age, gender, preoperative comorbidities, surgical approach, length of stay, readmission, postoperative complications and mortality were evaluated and compared between the groups., Results: A total of 3292 patients were included; 523 (15.9%) patients were prescribed MoM. Patients in the MoM group were found to be significantly older, with higher BMI and ASA class, and more often undergoing operations for cancer resection or colostomy creations, than the control group. Postoperative complications were comparable between the groups. On multivariable linear regression, MoM use was associated with a 14.1% reduction in length of stay (MoM group 4 (2; 8), control group 5 (3; 8 P = 0.006))., Conclusion: MoM as adjunct medication in the postoperative period following colorectal surgery is associated with reduced length of stay, without an increase in postoperative complications., (© 2022 The Authors. Impairment of long-term potentiation (LTP) is a common feature of many pre-clinical models of neurological disorders; however, studies in humans are limited by the inaccessibility of the brain. Human induced pluripotent stem cells (hiPSCs) provide a unique opportunity to study LTP in disease-specific genetic backgrounds. Here we describe a multi-electrode array (MEA)-based assay to investigate chemically induced LTP (cLTP) across entire networks of hiPSC-derived midbrain dopaminergic (DA) and cortical neuronal populations that lasts for days, allowing studies of the late phases of LTP and enabling detection of associated molecular changes. We show that cLTP on midbrain DA neuronal networks is largely independent of the N-methyl-D-aspartate receptor (NMDAR) and partially dependent on brain-derived neurotrophic factor (BDNF). Finally, we describe activity-regulated gene expression induced by cLTP. In the 1990, radon and radon progeny was reported to contribute approximately 70% of the average 1.4 + 1.0-mSv annual dose across 26 Western Australian underground non-uranium mines. The Western Australian underground mining workforce has expanded significantly, and parameters used to calculate doses have changed significantly, warranting a review of the 1990's data. The review concludes that doses received by the contemporary mining workforce has increased on average, by 5.4%, with annual dose estimates ranging from 0.53 to 3.56 mSv, with a mean of 1.33 mSv. Doses in 12 of the 23 underground mines exceed 1 mSv and are required to comply with radiation safety legislation. It is estimated that 5400 underground workers will fall into the greater than 1-mSv category. A direct scattering optical coherence tomography forward model was developed to simulate A-scans for both idealized and real light sources on an arbitrary given sample structure. Previous models neglected absorption, scattering, and multiple reflections at interfacial layers, and so two extended models were developed to investigate the impact of these processes. The first model uses the Beer-Lambert law to incorporate both absorption and scattering optical processes, and the second model uses a recursive form to model multiple reflections. These models were tested on a structure representative of a multilayered skin sample. The results show that the absorption and scattering processes have significant impact on the height of the peaks in the simulated A-scans. Conversely, the incorporation of multiple reflections has very little impact on the height of these peaks. Neither of the above processes has any impact on the locations of the A-scan peaks, which are associated with the sample interfaces between layers.

We report a new hybrid integration scheme that offers for the first time a nanowire-on-lead approach, which enables independent electrical addressability, is scalable, and has superior spatial resolution in vertical nanowire arrays. The fabrication of these nanowire arrays is demonstrated to be scalable down to submicrometer site-to-site spacing and can be combined with standard integrated circuit fabrication technologies. We utilize these arrays to perform electrophysiological recordings from mouse and rat primary neurons and human induced pluripotent stem cell (hiPSC)-derived neurons, which revealed high signal-to-noise ratios and sensitivity to subthreshold postsynaptic potentials (PSPs). We measured electrical activity from rodent neurons from 8 days in vitro (DIV) to 14 DIV and from hiPSC-derived neurons at 6 weeks in vitro post culture with signal amplitudes up to 99 mV. Overall, our platform paves the way for longitudinal electrophysiological experiments on synaptic activity in human iPSC based disease models of neuronal networks, critical for understanding the mechanisms of neurological diseases and for developing drugs to treat them.

The axial resolution is a critical parameter in determining whether optical coherent tomography (OCT) can be used to resolve specific features in a sample image. Typically, measures of resolution have been attributed to the light source characteristics only, including the coherence length and the point spread function (PSF) width of the OCT light sources. The need to cost effectively visualize the generated PSF and OCT cross-correlated interferogram (A-scan) using many OCT light sources have led to the extrinsic evolution of the OCT simulation model presented. This research indicated that empirical resolution in vivo, as well as depending on the light source's spectral characteristics, is also strongly dependent on the optical characteristics of the tissue, including surface reflection. This research showed that this reflection could be digitally removed from the A-scan of an epithelial model, enhancing the stratum depth resolution limit (SDRL) of the subsurface tissue. Specifically, the A-scan portion above the surface, the front surface interferogram, could be digitally subtracted, rather than deconvolved, from the subsurface part of each A-scan. This front surface interferogram subtraction resulted in considerably reduced empirical SDRLs being much closer to the superluminescent diodes' resolution limits, compared to the untreated A-scan results.

A Fibre Bragg Grating (FBG) acoustic sensor is used as a receiver for acoustic communications signals. Acoustic transmissions were generated in aluminium and Carbon Fibre Composite (CFC) panels. The FBG receiver was coupled to the bottom surface opposite a piezoelectric transmitter. For the CFC, a second FBG was embedded within the layup for comparison. We show the transfer function, frequency response, and transient response of the acoustic communications channels. In addition, the FBG receiver was used to detect Phase Shift Keying (PSK) communications signals, which was shown to be the most robust method in a highly resonant communications channel.

The fusion-active conformation of the envelope protein gp41 of HIV-1 consists of an N-terminal trimeric alpha-helical coiled-coil domain and three anti-parallel C-terminal helices that fold down the grooves of the coiled-coil to form a six-helix bundle. Disruption of the six-helix bundle is considered to be a key component of an effective non-peptide fusion inhibitor. In the current study, a fluorescence resonance energy transfer (FRET) experiment for the detection of inhibitor binding to the gp41 N-peptide coiled-coil of HIV-1 was performed, utilizing peptide inhibitors derived from the gp41 C-terminal helical region. The FRET acceptor is a 31-residue N-peptide containing a known deep hydrophobic pocket, stabilized into a trimer by ferrous ion ligation. The FRET donor is a 16-18-residue fluorophore-labeled C-peptide, designed to test the specificity of the N-C interaction. Low microM dissociation constants were observed, correlated to the correct sequence and helical propensity of the C-peptides. Competitive inhibition was demonstrated using the assay, allowing for rank ordering of peptide inhibitors according to their affinity in the 1-20 microM range. The assay was conducted by measuring fluorescence intensity in 384-well plates. The rapid detection of inhibitor binding may permit identification of novel drug classes from a library.

The function of the corpus luteum (CL) is a key element in many reproductive processes including ovulation, length of the estrous cycle, recognition of pregnancy and embryo survival in all mammalian species. The main function of the CL is to produce progesterone which acts on its tissues to prepare them for successful pregnancy. The CL is controlled by numerous biological compounds which provide luteotropic support during the estrous cycle and pregnancy and for inducing luteolysis at the end of the cycle The purpose of this paper is to review the mechansims responsible for controlling the endocrine function of this tissue in the bovine ovary.
