Abstract Background Investigating the molecular mechanism of colorectal cancer (CRC), a common lethal malignancies worldwide, is of great clinical significance. Solute carrier family 25 member 19 (SLC25A19) is a member of the solute carrier family that contribute to cellular functions, including tumor biology. Recently, many studies have attention on uncovering the relationship of SLC25A19 with malignant cancers, but its precise involvement in the regulation of CRC has not been thoroughly understood. This study sought to uncover the role and mechanism of SLC25A19 in CRC development. Methods The GEPIA database and immunohistochemical staining were utilized to detect the expression of SLC25A19 in CRC tissues. The functional influences of SLC25A19 on CRC cell phenotypes were evaluated through a series of assays including celigo cell count, colony formation, CCK‐8, flow cytometry, wound healing, and transwell assays following knocking down SLC25A19. Subsequently, the xenograft tumor model was constructed to evaluate the effect of SLC25A19 on tumor growth in vivo. The underlying mechanisms of SLC25A19 silencing were investigated using the human phospho‐kinase array. Results This study demonstrated the upregulation of SLC25A19 in CRC and its significant correlation with unfavorable prognosis in CRC patients. Suppression of SLC25A19 resulted in significant inhibition of cell proliferation, colony formation, and cell migration, alongside a boost in cell apoptosis. In vivo experiments revealed that silenced SLC25A19 displayed reduced growth rates and formed smaller xenografts. Mechanistically, the p53 pathway was found to be upregulated by SLC25A19 knockdown and mediated the function of SLC25A19. Conclusions Consequently, SLC25A19 was identified as a novel molecule with key regulatory ability in CRC development.

Traditional exhaust-gas turbocharging exhibits hysteresis under variable working conditions. To achieve rapid-intake supercharging, this study investigates the synergistic coupling process between the detonation and diesel cycles using gasoline as fuel. A numerical simulation model is constructed to analyze the detonation characteristics of a pulse-detonation combustor (PDC), followed by experimental verification. The comprehensive process of the flame’s deflagration-to-detonation transition (DDT) and the formation of the detonation wave are discussed in detail. The airflow velocity, DDT time, and peak pressure of detonation tubes with five different blockage ratios (BR) are analyzed, with the results imported into a one-dimensional GT-POWER engine model. The results indicate that the generation of detonation waves is influenced by flame and compression wave interactions. Increasing the airflow does not shorten the DDT time, whereas increasing the BR causes the DDT time to decrease and then increase. Large BRs affect the initiation speed of detonation in the tube, while small BRs impact the DDT distance and peak pressure. Upon connection to the PDC, the transient response rate of the engine is slightly improved. These results can provide useful guidance for improving the transient response characteristics of engines.

Herein, a diesel engine jet disturbance combustion system was proposed to achieve efficient and clean combustion under heavy load conditions in heavy-duty diesel engines. The key components of the combustion system were designed, and a research platform was constructed. Focusing on the internal combustion conditions of the disturbance chamber and the developmental path of high-speed jets, the design and comprehensive optimization of the jet disturbance combustion system were carried out. Following optimization, the peak internal heat release rate increased from 86 J/deg to 269 J/deg, and the cumulative heat release increased by 112 J, significantly enhancing the energy of the disturbance chamber jet. Then, considering combustion optimization and the heat transfer loss from the piston, it was determined that the optimal configuration for the disturbance chamber jet channel angles was 60 deg inter-channel angle and 10 deg channel incidence angle. This configuration allowed the disturbance chamber jet to precisely disturb the concentrated mixture area in the middle and late stages of combustion. The intervention of the disturbance chamber jet provided sufficient energy for the fuel–air mixing process and complicated the gas flow state in the main combustion chamber. Despite its low-momentum density, the residual mixture in the cylinder maintained a high mixing rate after the end of the fuel injection process. Single-cylinder engine test results showed that a diesel engine using this jet disturbance system and a 180 MPa common rail pressure fuel system achieved 52.12% thermal efficiency.

Abstract Objective Our previous study reveals that proprotein convertase subtilisin/kexin type 9 (PCSK9) is positively related to inflammatory markers, T helper (Th)‐17 cells, and treatment response in ankylosing spondylitis (AS) patients. Subsequently, this study aimed to explore the effect of PCSK9 on Th cell differentiation and its potential molecular mechanism in AS. Methods Serum PCSK9 was determined by enzyme‐linked immunosorbent assay in 20 AS patients and 20 healthy controls (HCs). Then naïve CD4+ T cells were isolated from AS patients and infected with PCSK9 overexpression or knockdown adenovirus followed by polarization assay. Afterward, PMA (an NF‐κB activator) was administrated. Results PCSK9 was increased in AS patients compared to HCs (p .05); its knockdown displayed the opposite function on them. Excessive nitrogen fertilizer usage in agricultural often leads to negative ecological and production gains. Alterations in the physical and chemical properties and microbial community structure of agricultural soils are both the cause and consequence of this process. This study explored the perturbation of soil properties and microorganisms in agricultural soils by different nitrogen levels. Soil total nitrogen, total phosphorus, and total potassium decreased in the shallow soil layer with decreasing nitrogen. Changes in nitrogen affected soil organic matter, pH, bulk density, and water content. However, a moderate reduction in nitrogen did not cause significant yield loss; the increased nitrogen use efficiency was the main reason, attributed to the available phosphorus and potassium. Short-term changes in nitrogen had limited effects on soil microbial community structure. Bacteria were more susceptible to perturbation by nitrogen changes. Nitrogen reduction increased the relative abundance of MND1 (1.21%), RB41 (1.96%), and Sphingomonas (0.72%) and decreased Dongia (0.3%), Chaetomium (0.41%), and Penicillium (0.5%). Nitrogen reduction significantly increased the bacteria functional composition of aerobic ammonia oxidation (4.20%) and nitrification (4.10%) and reduced chemoheterotrophy (2.70%) and fermentation (4.08%). Available phosphorus specifically drove bacterial community structure variation in the shallow soil layers of moderate nitrogen reduction treatments. Steroidobacter, RB41, Gemmatimonas, Ellin6067, Haliangium, and Sphingomonas were the main component nodes in this community structure. These results provide insights into the study of nitrogen and microorganisms in agricultural soils.

Effects of dual-treatment on cellulose fiber quality, micro-structure appearance, crystalline structure, hydrogen bonds, and surface elements were analyzed using a fiber quality analyzer (FQA), scanning electron microscope (SEM), wide-angle X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FT-IR), and X-ray photoelectron spectroscopy (XPS), respectively. In comparison to the untreated pulp, the brightness of pulp increased by 51.71%, whereas the apparent density and burst strength index of the pulpboard decreased by 11.76% and 48.18%, respectively. The absorption time, absorbability, and bulk of the fiber obtained by defibering the pulpboard decreased by 47.40%, 8.33%, and 5.32%, respectively, when it had been subjected to supplementary alkali. With the analysis of FQA and SEM, fiber swelled and curled, its surface was relatively smooth, and filaments of its partial surface area were exposed as a result of the supplementary alkali. Additionally, XRD analysis, FT-IR analysis, and XPS scanning spectra all showed that the crystallinity degree of fiber decreased by 45.10%, indicating more crystal structures were converted into amorphous structures. The proportion of total intra-molecular hydrogen bond intensity to total hydrogen bonds increased. The –OH content on fiber surface only decreased by 4.15%, compared with those of the untreated pulp, when the fiber was subjected to the two-step chemical treatment.

For the composite glass box girder, the generalized Bayesian objective function of elastic constants of the structure was derived based on layered shell element theory. Mechanical performances of the composite glass box girder were solved by layered shell element method. Combined with quadratic parabolic interpolation search scheme of optimized step length, the adaptive Powell's optimization theory was taken to complete the stochastic identification of elastic constants of composite glass box girder. Then the adaptive Powell's identification steps of elastic constants of the structure were presented in detail and the adaptive Powell's identification procedure was accomplished. From some classic examples, it is finally achieved that the adaptive Powell's identification of elastic constants of composite glass box girder has perfect convergence and numerical stability, which testifies that the adaptive Powell's identification theory of elastic constants of composite glass box girder is correct and reliable. The stochastic characteristics of systematic responses and elastic constants are well deliberated in generalized Bayesian objective function. And in iterative processes, the adaptive Powell's identification is irrelevant with the complicated partial differentiation of the systematic responses from the layered shell element model to the elastic constants, which proves high computation efficiency. The present study aimed to evaluate the expression of vascular endothelial growth factor C (VEGF-C) in human cholangiocarcinoma tissues and its role in metastasis in vitro. A total of 65 biopsy samples of cholangiocarcinoma, plus the FRH-0201 cell line, were investigated. The expression of VEGF-C in the human cholangiocarcinoma specimens was evaluated by immunohistochemistry (IHC). The effect of VEGF-C on tumor cell migration and proliferation was measured by MTT and Transwell assays in the FRH-0201 cell line. According to the IHC results, the biopsies of human cholangiocarcinoma were stained positively for VEGF-C, with a positive rate of 75.4% (49/65). Moreover, VEGF-C was expressed at a higher level in the patients with lymph node metastasis than in those without lymph node metastasis. In vitro, VEGF-C exhibited marked growth stimulation below the concentration of 5 ng/ml and was able to promote cholangiocarcinoma cell migration significantly. These findings suggested that VEGF-C may be a useful factor to predict lymph node metastasis in cholangiocarcinoma tissues and indicates that VEGF-C plays a significant role in proliferation and migration in cholangiocarcinoma cells. Pancreatic duct stone is thought not only to be the cause of abdominal pain but also to be a risk factor for pancreatic cancer. Several treatment options have been implemented in the treatment of pancreatic duct stones. Stone location is a critical factor in selecting treatment. We present the results of 27 endoscopic treatments and 35 surgical treatments performed in three hospitals at a single university between January 2000 and January 2005. The results were compared retrospectively in terms of success rate of stone removal, length of hospital stay, complications, pain relief, and changes of pancreatic functions. In our study, endoscopy resulted in a similar success rate of stone removal and short-term pain relief rate as the surgical approach and with a shorter length of hospital stay. However, the surgical group had amore favorable long-term clinical result, as well as a lower number of hospital readmissions at the 5-year follow-up point. Background: Both phosphorylated signal transducer and activator of transcription 3(pStat-3) and integrin αvβ6 can play vital role in the development and progression of cancer. However, little is known about their expression correlation and clinical significance in gallbladder cancer(GBC)., Objective: The aim of our present study was to investigate the expression of pStat-3 and integrin αvβ6, two proteins' correlation and their clinical significance in GBC tissues., Results: The expression of pStat-3 and integrin αvβ6 were both significantly associated with T stage, lymph node metastasis status, TNM stage (P=0.008, P=0.000, P=0.000 and P=0.036, P=0.001,P=0.000,respectively). IHC and Western blot showed their expressions in GBC tissues were higher than that in paraneoplastic tissues. Moderate positive correlation existed between the two proteins (r =0.349, P <0.001). The survival analysis by Kaplan-Meier and Cox regression model showed that GBC patients with pStat-3 or integrin αvβ6 positive expression had a significantly poorer 2-year survival rate (P = 0.002 and 0.000, the log-rank test, respectively), and either marker could act as unfavorable independent prognostic factors(RR=1.907, P=0.021 and RR=2.046, P=0.038)., Materials and Methods: The expression levels of pStat-3 and integrin αvβ6 were analyzed in GBC cancerous and paraneoplastic tissues of 97 cases via immunohistochemistry(IHC) and further validated by western blot method. Besides, SPSS software was used to observe their clinical significance as well as the two proteins' correlation., Conclusion: pStat-3 and integrin αvβ6 were indicators of tumor's progression and poor prognosis of patients with GBC. And the further study involving them may provide a helpful therapeutic target in prevention and treatment of GBC patients.
