Metabolic dysfunction-associated steatohepatitis (MASH) is a major cause of liver-related morbidity and mortality, yet treatment options are limited. Manual scoring of liver biopsies, currently the gold standard for clinical trial enrollment and endpoint assessment, suffers from high reader variability. This study represents the most comprehensive multisite analytical and clinical validation of an artificial intelligence (AI)-based pathology system, AI-based measurement of metabolic dysfunction-associated steatohepatitis (AIM-MASH), to assist pathologists in MASH trial histology scoring. AIM-MASH demonstrated high repeatability and reproducibility compared to manual scoring. AIM-MASH-assisted reads by expert MASH pathologists were superior to unassisted reads in accurately assessing inflammation, ballooning, MAS ≥ 4 with ≥1 in each score category and MASH resolution, while maintaining non-inferiority in steatosis and fibrosis assessment. These findings suggest that AIM-MASH could mitigate reader variability, providing a more reliable assessment of therapeutics in MASH clinical trials.

In this paper, the authors present the results of a qualitative case-study seeking to characterize data discovery needs and barriers of principal investigators and research support staff in clinical translational science. Several implications for designing and implementing transnational research systems have emerged through the authors' analysis. The results also illustrate the benefits of forming early partnerships with scientists to better understand their workflow processes and end-user computing practices in accessing data for research. The authors use this user-centered, iterative development approach to guide the implementation and extension of i2b2, a system they have adapted to support cross-institutional aggregate anonymized clinical data querying. With ongoing evaluation, the goal is to maximize the utility and extension of this system and develop an Interface that appropriately fits the swiftly evolving needs of clinical transnational scientists. A major challenge to implementing precision medicine is the need for an efficient and cost-effective strategy for returning individual genomic test results that is easily scalable and can be incorporated into multiple models of clinical practice. My46 is a Web-based tool for managing the return of genetic results that was designed and developed to support a wide range of approaches to disclosing results, ranging from traditional face-to-face disclosure to self-guided models. My46 has five key functions: set and modify results-return preferences, return results, educate, manage the return of results, and assess the return of results. These key functions are supported by six distinct modules and a suite of features that enhance the user experience, ease site navigation, facilitate knowledge sharing, and enable results-return tracking. A computer model in FORTRAN for the growth of the giant kelp,Macrocystis, which inhabits the coastal waters of southern California, has been developed. The present model considers only physical factors, water temperature and submarine light, as the factors that control the growth of single fronds in large kelp beds.The model has been used to examine two aspects of kelp growth in water of 20 meters depth. The opti mum time of the year for the start of new growth of fronds arising from the bottom in an established kelp bed was found to be early June. The optimum time of the year for regrowth of harvested canopy was found to be early July. In each case, the optimum time is interpreted as providing the shortest time for the frond to reach "maturity" (a length of 40 meters).The penetration of sunlight into the hydrosphere is adversely affected by particulate matter in the water such as results from the introduction of sewage ef fluent. The simulation identifies the critical values for turbidity which will prevent new growth from occurring. At a depth of 20 meters, light absorbency of 18% per meter reduces the possible growing time by only 20%. But when absorbency is increased to only 24%, the growing time becomes zero. Simulation runs for other kelp-bed water depths revealed similar critical values of absorbency resulting in the restric tion of growth to shallower waters. This type of analysis to resource-management efforts is obvious.The paper concludes with suggestions for modifying the integration techniques used and for incorporating additional quantifiable values to refine the model.

Moving through the many phases of drug development, from the identification of a candidate molecule for development to the filing and approval of a license to market a product, analytical and microbiological control methods and the associated technology for both drug substance and product may undergo many changes. In a world where globalization leads to this process and to the subsequent commercialization of product being executed on a number of different sites worldwide, the effective and efficient transfer of analytical and control technology is a critical factor in ensuring consistent quality standards.In order to maximize the efficiency of the analytical technology transfer, it has been necessary to reconsider some of Sterling Winthrop's traditional ways of doing business, and in collaboration with colleagues in both research and manufacturing, additional changes are to be introduced to further streamline the process. Demonstrating the effectiveness of the technology transfer process, a matter of some importance at the time of Food and Drug Administration (FDA) preapproval inspections, is an area where further opportunities for improvement are still available.

FLT3-ITD mutations are seen in 20-30% of adult and 15% of pediatric acute myeloid leukemia (AML) patients and are associated with poor prognosis with higher rates of relapse and therapy resistance. FLT3 tyrosine kinase inhibitors (TKIs) have shown some success in treatment of FLT3-ITD AML, but responses are often not complete or sustained. Multiple potential mechanisms of drug resistance could be active in patients with FLT3-ITD AML including mutations in the FLT3 kinase, exogenous signaling from the bone marrow microenvironment, and altered epigenetic regulation. Mutations involving epigenetic modifiers such as DNMT3A, IDH1/2, TET2, ASXL1 and MLL1 are often co-expressed with the FLT3-ITD mutation. In addition, over-expression of EZH2, DOT1L and protein arginine methyl-transferase (PRMT) have also been reported. However, the role of different epigenetic mechanisms in regulating growth of FLT3-ITD AML cells and to modulate ability of TKIs to target FLT3-ITD AML cells has not been systematically studied. The objective of our study was to identify epigenetic targets in FLT3-ITD AML by screening a panel of epigenetic modifiers for their ability to inhibit growth of FLT3-ITD AML cells, and to enhance growth inhibition in combination with a FLT3 TKI.
