Abstract The Venus flytrap (Dionaea muscipula J. Ellis) is a carnivorous plant able to synthesize large amounts of phenolic compounds, such as phenylpropanoids, flavonoids, phenolic acids, and 1,4-naphtoquinones. In this study, the first genetic transformation of D. muscipula tissues is presented. Two wild-type Rhizobium rhizogenes strains (LBA 9402 and ATCC 15834) were suitable vector organisms in the transformation process. Transformation led to the formation of teratoma (transformed shoot) cultures with the bacterial rolB gene incorporated into the plant genome in a single copy. Using high-pressure liquid chromatography, we demonstrated that transgenic plants were characterized by an increased quantity of phenolic compounds, including 1,4-naphtoquinone derivative, plumbagin (up to 106.63 mg × g−1 DW), and phenolic acids (including salicylic, caffeic, and ellagic acid), in comparison to non-transformed plants. Moreover, Rhizobium-mediated transformation highly increased the bactericidal properties of teratoma-derived extracts. The antibacterial properties of transformed plants were increased up to 33% against Staphylococcus aureus, Enterococcus faecalis, and Escherichia coli and up to 7% against Pseudomonas aeruginosa. For the first time, we prove the possibility of D. muscipula transformation. Moreover, we propose that transformation may be a valuable tool for enhancing secondary metabolite production in D. muscipula tissue and to increase bactericidal properties against human antibiotic-resistant bacteria. Key points • Rhizobium-mediated transformation created Dionaea muscipula teratomas. • Transformed plants had highly increased synthesis of phenolic compounds. • The MBC value was connected with plumbagin and phenolic acid concentrations.

Thermal imaging was used to study the early stage response to light-induced heating of Arabidopsis thaliana leaves. Time-series thermograms provided a spatial and temporal characterization of temperature changes in Arabidopsis wild type and the ost1-2 mutant rosettes exposed to excessive illumination. The initial response to high light, defined by the exponential increase in leaf temperature of ost1-2 gave an increased thermal time constant compared to wild type plants. The inability to regulate stomata in ost1-2 resulted in enhanced stomatal conductance and transpiration rate. Under strong irradiation, a significant decline in the efficiency of photosystem II was observed. This study evaluates infrared thermography kinetics and determines thermal time constants in particular, as an early and rapid method for diagnosing the prime indicators of light stress in plants under excessive light conditions.

Key message Defence responses of cyst nematode and/or wheat curl mite infested barley engage the altered reactive oxygen species production, antioxidant machinery, carbon dioxide assimilation and photosynthesis efficiency. Abstract The primary aim of this study was to determine how barley responds to two pests infesting separately or at once; thus barley was inoculated with Heterodera filipjevi (Madzhidov) Stelter (cereal cyst nematode; CCN) and Aceria tosichella Keifer (wheat curl mite; WCM). To verify hypothesis about the involvement of redox metabolism and photosynthesis in barley defence responses, biochemical, photosynthesis efficiency and chlorophyll a fluorescence measurements as well as transmission electron microscopy were implemented. Inoculation with WCM (apart from or with CCN) brought about a significant suppression in the efficiency of electron transport outside photosystem II reaction centres. This limitation was an effect of diminished pool of rapidly reducing plastoquinone and decreased total electron carriers. Infestation with WCM (apart from or with CCN) also significantly restricted the electron transport on the photosystem I acceptor side, therefore produced reactive oxygen species oxidized lipids in cells of WCM and double infested plants and proteins in cells of WCM-infested plants. The level of hydrogen peroxide was significantly decreased in double infested plants because of glutathione–ascorbate cycle involvement. The inhibition of nitrosoglutathione reductase promoted the accumulation of S-nitrosoglutathione increasing antioxidant capacity in cells of double infested plants. Moreover, enhanced arginase activity in WCM-infested plants could stimulate synthesis of polyamines participating in plant antioxidant response. Infestation with WCM (apart from or with CCN) significantly reduced the efficiency of carbon dioxide assimilation by barley leaves, whereas infection only with CCN expanded photosynthesis efficiency. These were accompanied with the ultrastructural changes in chloroplasts during CCN and WCM infestation.

Background Plant transformation with rol oncogenes derived from wild strains of Rhizobium rhizogenes is a popular biotechnology tool. Transformation effects depend on the type of rol gene, expression level, and the number of gene copies incorporated into the plant’s genomic DNA. Although rol oncogenes are known as inducers of plant secondary metabolism, little is known about the physiological response of plants subjected to transformation. Results In this study, the physiological consequences of rolB oncogene incorporation into the DNA of Dionaea muscipula J. Ellis was evaluated at the level of primary and secondary metabolism. Examination of the teratoma (transformed shoots) cultures of two different clones (K and L) showed two different strategies for dealing with the presence of the rolB gene. Clone K showed an increased ratio of free fatty acids to lipids, superoxide dismutase activity, synthesis of the oxidised form of glutathione, and total pool of glutathione and carotenoids, in comparison to non-transformed plants (control). Clone L was characterised by increased accumulation of malondialdehyde, proline, activity of superoxide dismutase and catalase, total pool of glutathione, ratio of reduced form of glutathione to oxidised form, and accumulation of selected phenolic acids. Moreover, clone L had an enhanced ratio of total triglycerides to lipids and accumulated saccharose, fructose, glucose, and tyrosine. Conclusions This study showed that plant transformation with the rolB oncogene derived from R. rhizogenes induces a pleiotropic effect in plant tissue after transformation. Examination of D. muscipula plant in the context of transformation with wild strains of R. rhizogenes can be a new source of knowledge about primary and secondary metabolites in transgenic organisms.

An energy-saving light emitting diode (LED) system allows for adjustment of light quality, which affects plant development and metabolic processes in in vitro cultures. The study investigated the content of endogenous carbohydrates and the condition of the photosynthetic apparatus of Gerbera jamesonii Bolus ex Hook. f. Our aim was to analyze the effects of different LED light qualities—100% red light (R LED), 100% blue (B LED), a mixture of red and blue (7:3) (RB LED), and a fluorescent lamp as a control (Fl)—during the multiplication of axillary shoots. After 40 days, the culture measurements were performed using a non-invasive pulse amplitude modulation (PAM) fluorimeter. Sugar content was assessed with high performance liquid chromatography (HPLC). Two forms of free monosaccharides (glucose and fructose), two sugar alcohol derivatives (inositol and glycerol), and seven forms of free oligosaccharides were identified. Of those, glucose content was the highest. LEDs did not disturb the sugar metabolism in multiplied shoots. Their monosaccharides were three times more abundant than oligosaccharides, the same results were found in plants grown under control light. R light depleted the performance of the photosynthetic apparatus and caused its permanent damage. The RB LED spectrum ensured the most efficient non-photochemical quenching of the photosystem II (PS II) excitation state and high shoot quality.

We compared the effectiveness of three different trap colours (blue, yellow and white) to identify the optimal trap colour for the monitoring of the thripsFrankliniella intonsa,Thrips tabaci,Thrips fuscipennisandAeolothrips intermediusin the three pea cultivars, namely Polar, Izolda and Tarchalska. The number of captured thrips specimens was significantly affected by trap colour, pea cultivar and interaction between these factors. The two most attractive trap base colours for thrips were blue (peak at 450 nm) and yellow (peak at 550 nm). Irrespective of the pea cultivar, mostF. intonsawere captured on the blue and yellow traps. Blue traps were the most attractive toT. fuscipennisandT. tabaci, followed by yellow and white ones, and yellow traps were most attractive toA. intermedius. More thrips were caught on the traps located in the plots with cultivars characterised by a longer growing season Tarchalska and Izolda, which were inhabited by large numbers of thrips. There was a significant interaction effect between trap colour and pea cultivar on the number of caughtF. intonsaandT. fuscipennisin both years and forT. tabaciin 2010. Overall, among the tested trap colours, blue traps were the most effective ones for monitoring thrips in pea fields and could be used as an early detection tool. Yellow traps may be risky because they reduce population densities of the predaceousA. intermediusin pea fields, thus leading to an increase in pest numbers.

A non-destructive thermal imaging method was used to study the stomatal response of salt-treated Arabidopsis thaliana plants to excessive light. The plants were exposed to different levels of salt concentrations (0, 75, 150, and 220 mM NaCl). Time-dependent thermograms showed the changes in the temperature distribution over the lamina and provided new insights into the acute light-induced temporary response of Arabidopsis under short-term salinity. The initial response of plants, which was associated with stomatal aperture, revealed an exponential growth in temperature kinetics. Using a single-exponential function, we estimated the time constants of thermal courses of plants exposed to acute high light. The saline-induced impairment in stomatal movement caused the reduced stomatal conductance and transpiration rate. Limited transpiration of NaCl-treated plants resulted in an increased rosette temperature and decreased thermal time constants as compared to the controls. The net CO2 assimilation rate decreased for plants exposed to 220 mM NaCl, in the case of 75 mM NaCl treatment, an increase was observed. A significant decline in the maximal quantum yield of photosystem II under excessive light was noticeable for the control and NaCl-treated plants. This study provides evidence that thermal imaging as a highly sensitive technique may be useful for analyzing the stomatal aperture and movement under dynamic environmental conditions.

The present study investigated the response of non-metallicolous (NM) and metallicolous (M) Alyssum montanum shoots cultured in vitro on a medium supplemented simultaneously with heavy metals (HMs) to identify mechanisms involved in alleviating metal-induced damage. Plant status in respect to photosynthetic apparatus efficiency was determined and linked with changes in biochemical composition of shoots, namely phenolic acids’ and stress-related phytohormones. Results showed the considerable inter-ecotype differences in (1) the photosynthetic pigments’ amount, (2) the functioning of membrane electron transporters as well as (3) the linear and alternative electron transport pathways, whose lower values were reported in NM than in M HM-treated culture. Photosynthetic apparatus protection in M specimens was assured by the activation of cinnamic acid synthesis (by phenylalanine ammonia lyase) and its further transformations to benzoic acid derivatives with high ability to counteract oxidative stress, that was accompanied by the overexpression of jasmonic acid stimulating antioxidant machinery. In turn, detrimental HM effects on NM shoots could result from the diminution of most phenolics’ accumulation, and only the content of coumarate (produced by bifunctional phenylalanine/tyrosine ammonia lyase) and rosmarinic acid increased. All these together with an enhanced concentration of abscisic acid might suggest that NM strategy to cope with HMs is based mostly on a restriction of metal movement with transpiration flow and their limited distribution in leaves. Summarizing, our findings for the first time point out the physiological and metabolic adaptation of pseudometallophyte A. montanum to adverse conditions.

Grass pea (Lathyrus sativus) is a leguminous plant of outstanding tolerance to abiotic stress. The aim of the presented study was to describe the mechanism of grass pea (Lathyrus sativus L.) photosynthetic apparatus acclimatisation strategies to salinity stress. The seedlings were cultivated in a hydroponic system in media containing various concentrations of NaCl (0, 50, and 100 mM), imitating none, moderate, and severe salinity, respectively, for three weeks. In order to characterise the function and structure of the photosynthetic apparatus, Chl a fluorescence, gas exchange measurements, proteome analysis, and Fourier-transform infrared spectroscopy (FT-IR) analysis were done inter alia. Significant differences in the response of the leaf and stem photosynthetic apparatus to severe salt stress were observed. Leaves became the place of harmful ion (Na+) accumulation, and the efficiency of their carboxylation decreased sharply. In turn, in stems, the reconstruction of the photosynthetic apparatus (antenna and photosystem complexes) activated alternative electron transport pathways, leading to effective ATP synthesis, which is required for the efficient translocation of Na+ to leaves. These changes enabled efficient stem carboxylation and made them the main source of assimilates. The observed changes indicate the high plasticity of grass pea photosynthetic apparatus, providing an effective mechanism of tolerance to salinity stress.

Understanding the mechanisms of plant tolerance to osmotic and chemical stress is fundamental to maintaining high crop productivity. Soil drought often occurs in combination with physiological drought, which causes chemical stress due to high concentrations of ions. Hence, it is often assumed that the acclimatization of plants to salinity and drought follows the same mechanisms. Grass pea (Lathyrus sativus L.) is a legume plant with extraordinary tolerance to severe drought and moderate salinity. The aim of the presented study was to compare acclimatization strategies of grass pea seedlings to osmotic (PEG) and chemical (NaCl) stress on a physiological level. Concentrations of NaCl and PEG were adjusted to create an osmotic potential of a medium at the level of 0.0, &minus, 0.45 and &minus, 0.65 MPa. The seedlings on the media with PEG were much smaller than those growing in the presence of NaCl, but had a significantly higher content percentage of dry weight. Moreover, the stressors triggered different accumulation patterns of phenolic compounds, soluble and insoluble sugars, proline and &beta, N-oxalyl-L-&alpha, &beta, diamino propionic acid, as well as peroxidase and catalase activity. Our results showed that drought stress induced a resistance mechanism consisting of growth rate limitation in favor of osmotic adjustment, while salinity stress induced primarily the mechanisms of efficient compartmentation of harmful ions in the roots and shoots. Furthermore, our results indicated that grass pea plants differed in their response to drought and salinity from the very beginning of stress occurrence.

The role of an endophytic Zygomycete Mucor sp. in growth promotion and adaptation of the photosynthetic apparatus to increased energy demands of its hosts Arabidopsis arenosa and Arabidopsis thaliana was evaluated. Inoculation with the fungus improved the water use efficiency of the plants and allowed for them to utilize incident light for photochemistry more effectively by upregulating the expression of several photosystem I- and II-related genes and their respective proteins, proteins involved in light harvesting in PSII and PSI and carbon assimilation. This effect was independent of the ability of the plants to acquire nutrients from the soil. We hypothesize that the accelerated growth of the symbiotic plants resulted from an increase in their demand for carbohydrates and carbohydrate turnover (sink strength) that triggered a simultaneous upregulation of carbon assimilation. Arabidopsis plants inoculated with Mucor sp. exhibited upregulated expression in several genes encoding proteins involved in carbohydrate catabolism, sugar transport, and smaller starch grains that indicate a significant upregulation of carbohydrate metabolism.

Kidney vetch (Anthyllis vulneraria L.) is a well-known Zn hyperaccumulator. Zn often occurs with Pb in one ore; thus, plants inhabiting waste dumps are exposed not only to Zn but also to Pb toxicity. While the response of kidney vetch to Zn toxicity is relatively well known, the Pb survival strategy of Anthyllis vulneraria has not been the subject of investigations. The aim of presented research was to determine the survival strategy of kidney vetch exposed to high lead concentrations. Shoot explants of a calamine kidney vetch ecotype were placed on agar media containing 0.0, 0.5, 1.0, and 1.5 mM Pb. Morphological, physiological, and biochemical responses, in particular photosynthetic apparatus of plantlets, were examined. The most pronounced changes were observed in plants grown on media supplemented with 1.5 mM Pb after 8 weeks of culture. Increased dry weight and high lead accumulation were observed in roots. Similarly, in shoots, increased dry weight and a decreased number of newly formed shoots were recorded. The accumulation of lead was many times lower in shoots than in roots. In leaf cells’ ultra-structure, looser arrangement of chloroplast thylakoid grana was observed. Despite the decrease in chlorophyll a and carotenoid content, the photosynthetic apparatus remained efficient due to the lack of photoinhibition and increased electron transport rate beyond photosystem II (PSII). For the first time, an acclimatization mechanism based on maintaining the high efficiency of photosynthetic apparatus resulting from increasing of electron transport rate was described.

The endophytic fungus Mucor sp. was isolated from Arabidopsis arenosa inhabiting post mining wastes lands. Its role in plant adaptation to toxic metal enriched environment was evaluated. Plants inoculated with the fungus yielded significantly more biomass. Their growth response was correlated with significant elongation of root hairs, an improved water and P status and a significant upregulation of the expression of genes associated with nutrient uptake. The mechanism of root hair elongation was investigated with auxin and ethylene insensitive Arabidopsis thaliana mutants. The results clearly indicate that the root hair elongation phenotype results from fungi induced alterations in ethylene metabolism. The upregulation of close to 50 genes associated with ethylene biosynthesis and signaling confirm these results. The accumulation of Zn and Fe was lower in endophyte inoculated plants. Additionally, root to shoot translocation of Fe, Cd and Zn was improved. The expression of metal transporters associated with metal influx, efflux and distribution within the plant corresponded with altered metal homeostasis. The results of this study clearly show that the endophytic fungus plays an important role in the adaptation of the non-mycorrhizal A. arenosa to metal toxicity.

Stress response of Dionaea muscipula J. Ellis to various light quality and quantity regimes, in the context of secondary metabolites production was determined. Venus Flytrap plants were cultivated in vitro in four different light regimes: fluorescence (50 µmol × m−2 × s−1 PPFD—control), fluorescence (120 µmol × m−2 × s−1 PPFD) and white LED (120 µmol × m−2 × s−1 PPFD) and in the darkness. Higher light intensity caused significant increase of membrane lipid peroxidation during the first 4 weeks of experiment. Whereas, light spectrum caused activation of defensive mechanisms. In the case of white LED, this mechanism was associated with the increased catalase activity and elevated synthesis of cinnamic acid derivatives, total phenols and plumbagin. While, in higher light intensity of fluorescence lamp, a reduction of chlorophyll content, and an increase amount of phenylopropanoids and plumbagin was observed. In turn, Venus Flytrap plants showed the highest activity of antioxidant enzymes, especially peroxidase, in the darkness. Higher concentration of plumbagin observed for the first time in response to different light quality and quantity, suggests that plumbagin is involved in photoprotection and/or in the antioxidant activity in Venus Flytrap tissue. Consequently, light might be used as an inorganic elicitator of 1,4-naphthoquinone derivatives and other phenolic compounds.

This work presents method of identification of the functional parameters of a voltage-controlled oscillator (VCO) using artificial neural network (ANN). The VCO under test is excited with a selected stimuli and the ANN is trained by the VCO response in time-domain. Investigated VCO is a mixed-signal circuit, therefore spread of circuit parameters (tolerance) is modelled by Monte-Carlo analysis. Existence of a circuit failures, modelled as spot defects, is also taken into consideration. Additionally, a genetic algorithm (GA) is used to minimize number of circuit response samples. The goal is shortening of data acquisition and processing time during circuit testing, as having significant influence on final manufacturing cost. The proposed method enables shortening test time of the VCO, together with high efficiency of functional parameters identification.

Lead is one of the most toxic trace elements influencing plant growth and development processes including photosynthesis. Although lead is characterized by low mobility, it can get into the plant and after destroying physical barrier (Casparian strip), enters the xylem and translocates to the aerial parts of a plant affecting photosynthetic apparatus in both indirect and direct ways.

Plants growing under natural conditions are constantly exposed to ultraviolet (UV), primarily UV-A, radiation. Grass pea (Lathyrus sativus L.) is a legume species resistant to harsh growing conditions, such as drought, salinity or periodic flooding. Due to the advantageous composition of seeds, it is used for consumption in such regions as South Asia or East Africa where high intensity of UV radiation occurs. Absorption of this spectral range causes changes in the photosynthetic apparatus of plants, including damage to the photosystem II (PSII) reaction centres. The aim of the work was to examine whether the use of the combination UV-A: red light as a source of radiation would enable quick acclimatization of the photosynthetic apparatus of grass pea to the negative effect of UV-A radiation. 14-day-old plants were exposed to UV-A:red radiation for 48 h. The plants exposed to UV-A:red radiation showed enhanced effective efficiency of PSII and increased total electron carriers, which enabled more effective photosynthesis at higher values of radiation intensity in comparison with control plants, kept under white LED light. At the same time, there were no statistically significant differences in both the photosynthetic pigment contents and the level of lipid peroxidation. The obtained results indicate that the observed increase in the efficiency of CO2 carboxylation after short-term UV-A:red radiation has resulted from the efficient linear electron transport due to maintaining the effective oxygen evolving complex (OEC) and increased total electron carriers.

This paper presents the results of consolidated and undrained (CU) triaxial cyclic tests related to the influence of tire waste addition on the strength characteristics of two different soils from Southern Poland: unswelling kaolin and swelling red clay. The test procedure included the normally consolidated remolded specimens prepared from pure red clay (RC) and kaolin (K) and their mixtures with two different fractions of shredded rubber powder (P) and granulate (G) in 5%, 10%, and 25% mass proportions. All samples were subjected to low-frequency cyclic loading carried out with a constant stress amplitude. Analysis of the results includes consideration of the effect of rubber additive and number of load cycles on the development of excess pore pressure and axial strain during the cyclic load operation and on the maximum stress deviator value. A general decrease in the shear strength due to the cyclic load operation was observed, and various effects of shear strength depended on the mixture content and size of the rubber waste particles. In general, the use of soil–rubber mixtures, especially for expansive soils and powder, should be treated with caution for cyclic loading.

The carnivorous plant Dionaea muscipula J. Ellis (Venus flytrap) is a widely known medical herb, capable of producing various phenolic compounds known for their strong antioxidant and antibacterial properties. In the pharmaceutical industry, Venus flytrap is grown in tissue cultures, as the natural population of D. muscipula is very limited. Here, we describe an improved method to increase the quantity and quality of phenolic compounds produced in D. muscipula. This is achieved by combining biotic elicitation (using Cronobacter sakazakii bacteria lysate) of D. muscipula cultured with rotary shaking (hydromechanical stress), which we describe here for the first time. The antibacterial activity and the antioxidant properties of the obtained compounds were studied on two antibiotic-resistant human pathogenic bacteria. The proposed plant culture conditions resulted in an increase in fresh weight, as well as a higher total phenolic content, in comparison to traditional tissue cultures on agar-solidified medium. With the use of high-performance liquid chromatography, we demonstrated that the described elicitation strategy leads to an increased synthesis of myricetin, caffeic acid, ellagic acid and plumbagin in D. muscipula tissue. We also found that a higher level of antioxidant activity, exhibited by the plant extract, corresponded with its higher phenylpropanoid content. The bactericidal activity of the extract against Staphylococcus aureus was dependent on the duration of plant culture under described elicitation conditions, whereas neither elicitation condition (duration or elicitor concentration) seemed relevant for the bactericidal activity of the extract towards Escherichia coli. This suggest that Gram-negative bacteria are less sensitive to compounds derived from Venus flytrap tissue.

Physiological and molecular mechanisms of adaptation to abiotic stresses of grass pea (Lathyrus sativus L.) are still poorly understood. Responses of four genotypes of grass pea to salinity stress in tissue culture conditions were investigated at early seedling growth stages. Salinity stress was induced in the agar media by adding 0, 50, 100 and 200 mM of NaCl. Germination and seedling emergence percentage was not significantly affected by 50 and 100 mM of NaCl. However, NaCl in 200 mM concentration lowered level of these parameters. Generally, exposure to NaCl stress significantly reduced length of grass pea seedling organs (root and shoot) but did not influence the content of dry weight in shoots and increased it in the roots in two cases. Increasing salt concentration decreased integrity of cellular membranes both in root and shoot tissues. Higher accumulation of phenolic compounds and significant changes in activity of antioxidant enzymes (peroxidase and catalase) were observed in the roots but not in the shoots. Similarly, the content of proline increased mostly in the roots from moderate (100 mM) salinity conditions. Adverse conditions did not resulted in alterations in photosynthetic pigments content of any tested genotypes. The better performance of shoots than roots may result from in vitro conditions in which experiments were conducted.

In stems of Clusia, CO 2 concentrated in the xylem sap in CAM trees can be fixed by PEPC and Rubisco, while in C 3 trees only Rubisco is engaged. The photosynthetic characteristics of 7–8-year-old stems of two tropical trees representing the Clusiaceae family were compared: Clusia multiflora Kunth. described as an obligate C3 and Clusia rosea Jacq. as an obligate CAM plant. Photosynthetic gas exchange, xylem CO2 concentration, chlorophyll distribution, 13C discrimination, daily malate and citrate fluctuations and the abundance of Rubisco (ribulose-1,5-bisphosphate carboxylase/oxygenase) and PEPC (phosphoenolpyruvate carboxylase) proteins were measured in leaves and stems. In stems of both species a low CO2 efflux (in the range of 0.05–0.1 µmol m−2 s−1) was observed as a result of extremely low cork conductance for water vapor (0.15–0.2 mmol m−2 s−1). This led to the CO2 concentration in xylem sap reaching 5.2 (CO2*) mmol l−1. The substantial amount of chlorophyll in the outer part of the bark and light-induced decrease of CO2 concentration within the xylem can be explained by photosynthetic activity in this tissue. Moreover, Western blotting analyses proved the presence of Rubisco in the stems of both Clusia species; however, PEPC was only found in C. rosea. Additionally, daily fluctuations in the concentration of citrate and malate (higher than in leaves) and significant enrichment in 13C in C. rosea stems were observed. These facts allow us to conclude that the examined stems of C. rosea and C. multiflora represent specific types of photosynthetic metabolism.

Ceylon leadwort (Plumbago zeylanica) is ornamental plant known for its pharmacological properties arising from the abundant production of various secondary metabolites. It often grows in lead polluted areas. The aim of presented study was to evaluate the survival strategy of P. zeylanica to lead toxicity via photosynthetic apparatus acclimatization. Shoots of P. zeylanica were cultivated on media with different Pb concentrations (0.0, 0.05, and 0.1 g Pb∙l&minus, 1). After a four-week culture, the efficiency of the photosynthetic apparatus of plants was evaluated by Chl a fluorescence measurement, photosynthetic pigment, and Lhcb1, PsbA, PsbO, and RuBisCo protein accumulation, antioxidant enzymes activity, and chloroplast ultrastructure observation. Plants from lower Pb concentration revealed no changes in photosynthetic pigments content and light-harvesting complex (LHCII) size, as well as no limitation on the donor side of Photosystem II Reaction Centre (PSII RC). However, the activity and content of antioxidant enzymes indicated a high risk of limitation on the acceptor side of Photosystem I. In turn, plants from 0.1 g Pb∙l&minus, 1 showed a significant decrease in pigments content, LHCII size, the amount of active PSII RC, oxygen-evolving complex activity, and significant remodeling of chloroplast ultrastructure indicated limitation of PSII RC donor side. Obtained results indicate that P. zeylanica plants acclimate to lead toxicity by Pb accumulation in roots and, depending on Pb concentration, by adjusting their photosynthetic apparatus via the activation of alternative (cyclic and pseudocyclic) electron transport pathways.
