4. Whey protein concentrate and skimmed milk powder as encapsulation agents for coffee silverskin extracts processed by spray drying.

- Abstract
We tested the hypothesis that milk proteins, through microencapsulation, guarantee protection against bioactive substances in coffee silverskin extracts. Therefore, the aim of this study was to carry out technological, nutritional and physicochemical characterisation of a coffee silverskin extract microencapsulated using instant skim milk powder and whey protein concentrate as wall materials. The aqueous extract of coffee silverskin was spray-dried using 10% (w/v) skim milk powder and whey protein concentrate. The samples were characterised by determining the water content, water activity, particle size distribution, colour analysis and total phenolic compound content as well as antioxidant activity using 2,2-diphenyl-radical 1-picrylhydrazyl scavenging methods, nitric oxide radical inhibition and morphological analysis. The product showed water activity within a range that ensured greater stability, and the reduced degradation of the dried coffee silverskin extract with whey protein concentrate resulted in better rehydration ability. The luminosity parameter was higher and the browning index was lower for the encapsulated samples than for the pure coffee silverskin extract. The phenolic compound content (29.23 ± 8.39 and 34.00 ± 8.38 mg gallic acid equivalents/g for the coffee silverskin extract using skimmed milk powder and whey protein concentrate, respectively) and the antioxidant activity of the new product confirmed its potential as a natural source of antioxidant phenolic compounds. We conclude that the dairy matrices associated with spray drying preserved the bioactive and antioxidant activities of coffee silverskin extracts. [ABSTRACT FROM AUTHOR]

5. Expanding the molecular toolkit for the new gas-converting Acetobacterium wieringae strain JM

- Abstract
Gas fermentation is a promising way to convert CO-rich gases to chemicals. In this work we aimed to develop an electrotransformation protocol for the promising gas-converting Acetobacterium wieringae strain JM (sJM) [1], to further implement heterologous pathways for the production of new compounds from gas streams. In this matter, Clostridial species are the most genetic manipulated homoacetogens, and within the Acetobacterium genus, only A. woodii has been used. Therefore, there is a strong need and opportunity to expand the molecular toolkit for sJM and other Acetobacterium species. To achieve this, we tested reported electrotransformation procedures for A. woodii, using available molecular tools from Clostridial species, namely the plasmid vectors pMTL82151, pMTL83151, pMTL84151, and pMTL84151 containing the Gram-positive replicons pBP1, pCB102, pCD6, and pIM13 with the thiamphenicol resistance gene (catP). The utilization reported electrotransformation procedures did not result in sJM transformants. Therefore, we developed a new protocol which we report to be applicable to sJM, A. wieringae DSM 1911 and A. woodii DSM 1030. The most important changes for higher expeditiously and reproducibility were the use of fixed cell density of competent cells resulting in electroporation cell-plasmid ratios of 2 to 3 (OD600.mL/g), the use of an electric field strength of 10 kV/cm, and the selection of transformants in serum anaerobic bottles with molten agar of our optimized basal media. All plasmid vectors were transformed, having pMTL83151 yielded the highest transformation efficiency in all tested Acetobacterium strains, reaching efficiencies up to 5.0 × 102, 2.0 × 103, and 5.1 × 103 transformants g-1 DNA for sJM, A. wieringae DSM 1911 and A. woodii DSM 1030, respectively. Key factors affecting the electrotransformation efficiency include cell-wallweakening using D-threonine, pH of wash buffer, field strength of the electric pulse, plasmid amount, and sucrose osmoprotection. A demanda por produtos alimentícios com alegações funcionais é crescente nos últimos anos. No entanto, muitas substâncias bioativas apresentam limitações tecnológicas para utilização como ingredientes na indústria de alimentos. Uma das formas de otimizar essa aplicação é a utilização da técnica de microencapsulação por spray drying das substâncias de interesse. Entre os diversos carreadores e agentes de encapsulamento usados atualmente, destacam-se as proteínas lácteas. Devido ao seu alto valor nutricional, as proteínas também agregam valor a esses produtos. O objetivo desta revisão é apresentar as principais pesquisas dos últimos anos que utilizaram essas fontes proteicas como protetores de compostos fenólicos naturais. Para isso, foi realizada uma busca de artigos dos últimos cinco anos nos sites PubMed, Web of Sciences e Scopus. Essa revisão apresenta os principais conceitos envolvendo proteínas lácteas e microencapsulação de compostos fenólicos. Todo o processo de elaboração das micropartículas, desde a extração de fenólicos até a sua liberação em condições gastrointestinais é discutido neste trabalho. De acordo com os trabalhos avaliados, os produtos derivados de proteínas lácteas, usados isoladamente ou combinados com outros polímeros, são opções viáveis e promissoras na proteção de compostos fenólicos naturais. The demand for food products with functional claims has been growing in recent years. However, many bioactive substances have technological limitations for use as ingredients in the food industry. One of the ways to optimize this application is the use of the microencapsulation technique by spray drying the substances of interest. Among the various carriers and encapsulation agents currently used, dairy proteins stand out. Due to their high nutritional value, proteins also add value to these products. This work aims to summarize recent findings regarding the application of these protein sources as protectors of natural phenolic compounds. For this purpose, a search for articles from the last five years was carried out on the PubMed, Web of Sciences and Scopus websites. This review presents the main concepts involving milk proteins and microencapsulation of phenolic compounds. Additionally, the discussion addresses the whole process of microparticles production, from the extraction of phenolics to their release under gastrointestinal conditions. According to the studies evaluated, products derived from dairy proteins, used alone or combined with other polymers, are viable and promising options to protect natural phenolic compounds.

Com o objetivo primordial de descrever as atividades desenvolvidas ao longo do estágio curricular, foi elaborado o seguinte relatório, com foco nas atividades principais realizadas no grupo Moretextile. Durante seis meses foram desenvolvidas atividades na área do custeio e controlo de gestão numa empresa têxtil, com maior incidência no setor do custeio, que proporcionaram o desenvolvimento do mestrando enquanto profissional. O foco teórico centrou-se principalmente nas áreas: contabilidade gestão, controlo interno e orçamentação. Com este estágio, o mestrando teve a possibilidade de presenciar a aplicação de todos os temas mencionados no mercado laboral. Adicionalmente, o dia-a-dia laboral foi desafiante, enriquecedor e permitiu ao mestrando a aquisição e desenvolvimento de soft-skills, tais como, a gestão de tempo, comunicação, mediação de conflitos, trabalho de equipa, resolução de problemas e pensamento criativo.

8. Physical literacy: validation of tests for assessment of motor competence, motivation and knowledge of brazilian children

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior O letramento corporal vem ganhando popularidade como objetivo central da educação física em diversos países, embora ainda seja pouco difundido no Brasil. Pode ser definido como a motivação, confiança, competência física, conhecimento e entendimento para valorizar e assumir a responsabilidade pelo envolvimento em atividades físicas ao longo da vida. Instrumentos para a avaliação da jornada de letramento corporal têm sido desenvolvidos por pesquisadores, com destaque para o Canadian Assessment of Physical Literacy - 2 nd ed. (CAPL- 2). Integram esta bateria o CAMSA, que avalia a competência motora e o questionário CAPL- 2, que avalia a motivação e o conhecimento. O CAMSA propõe avaliar a competência motora, assim como outros dois testes consolidados na literatura: o KTK e o TGMD-3. Contudo, apenas o TGMD-3 encontra-se validado no Brasil. Em relação ao questionário CAPL-2, o mesmo ainda não foi traduzido, adaptado culturalmente e validado para o Brasil. Desta forma, o presente trabalho foi dividido em três etapas, apresentadas em artigos científicos. O artigo 1 investigou a estrutura fatorial do KTK e comparou quatro métodos para o cálculo do escore fatorial. Os resultados mostraram que o KTK apresenta uma boa estrutura fatorial e que o método mais simples da “soma dos escores brutos” pode ser utilizado para interpretar os resultados. O artigo 2 verificou a validade concorrente do CAMSA, utilizando o KTK, o TGMD-3 e a combinação dos dois testes como medidas de referência. Os resultados mostraram correlações positivas, de moderada a forte, confirmando o teste como alternativa para a avaliação da competência motora O artigo 3 realizou a tradução e adaptação transcultural do questionário CAPL-2 no contexto brasileiro. As análises fatoriais confirmatórias e os índices de ajuste dos modelos confirmaram que o questionário CAPL-2-Br é um instrumento eficiente para avaliar os domínios “motivação e confiança” e “conhecimento e compreensão” do letramento corporal. Physical literacy has been gaining popularity as a central objective of physical education in several countries, although it is still not widespread in Brazil. It is defined as motivation, confidence, physical competence, knowledge, and understanding to value and take responsibility for engaging in physical activities throughout life. Instruments for the assessment of the physical literacy journey have been developed by researchers, with emphasis on the Canadian Assessment of Physical Literacy - 2nd ed. (CAPL-2). This battery includes CAMSA, which assesses motor skills, and the CAPL-2 questionnaire, which assesses motivation and knowledge. CAMSA proposes to assess motor competence, as well as two other tests consolidated in the literature: KTK and TGMD-3. However, only TGMD-3 was validated in Brazil. Regarding the CAPL-2 questionnaire, it has not yet been translated, culturally adapted, and validated for Brazil. Thus, the present work has three stages, presented in scientific articles. Article 1 investigated the factorial structure of the KTK and compared four methods for calculating the factor score. The results showed that the KTK has a good factorial structure and that the simplest “sum of the raw scores” method can be used to interpret the results. Article 2 verified the concurrent validity of CAMSA, using KTK, TGMD-3, and the combination of the two tests as reference measures. The results showed positive correlations, from moderate to substantial, confirming the test as an alternative for the assessment of motor competence. Article 3 performed the translation and cross-cultural adaptation of the CAPL-2 questionnaire in the Brazilian context. The confirmatory factor analysis and the adjustment indexes of the models confirmed that the CAPL-2-Br questionnaire is an efficient instrument to assess the domains "motivation and confidence" and "knowledge and understanding" of physical literacy. O foco principal que este trabalho apresenta é desmistificar a imagem pejorativa que as pessoas têm sobre os lares de idosos, demonstrando que no Lar Nossa Senhora da Luz os idosos não são abandonados e mantém vivo o convívio com suas famílias, assim criando vínculos uns com os outros. Também busca mostrar a infraestrutura e preparos necessários que o lar possui para atender as necessidades dos idosos da melhor maneira possível. Neste contexto foi realizado uma pesquisa de forma online com a ferramenta do Google Docs com homens e mulheres de 25 a 35 anos de idade, a fim de entender de maneira ampla qual a percepção que eles têm sobres os lares de idosos. De acordo com as respostas e interpretações das mesmas, o grupo obteve ideias centrais, fundamentais para o planejamento e a criação da campanha. The main objective of this work is to demystify the pejorative image that people have about the homes of the elderly, demonstrating that in the Home of Our Lady of Light old people are not abandoned and keep alive the conviviality with their families, thus creating bonds with each other. It also seeks to show the infrastructure and necessary preparation that the home has to meet the needs of the elderly in the best possible way. In this context, an online search was done with the Google Docs tool with men and women between 25 and 35 years of age, in order to understand in a broad way the perception they have about the homes of the elderly. According to the responses and interpretations of these, the group got central ideas, fundamental for the planning and the creation of the campaign.

- Abstract
Genetic profiles of SalmonellaMinnesota isolates were analyzed using pulsed-field gel electrophoresis (PFGE). In total, 13 isolates obtained from the broiler industry collected in the states of Minas Gerais (11) and São Paulo (2), as well as five recovered from cases of foodborne infections in humans in the states of Minas Gerais (2), Santa Catarina (1), and Rio Grande do Sul (2), were submitted to PFGE. These 18 S. Minnesota isolates together with other 12 of poultry origin were also subjected to antimicrobial susceptibility testing. The PFGE analysis of 18 strains of S. Minnesota generated a dendrogram that grouped the isolates with 83–90% similarity into four main clusters. Among them, cluster “A” grouped the majority of isolates (13), including two of human origin that showed 90% similarity with a broiler isolate, both recovered in Minas Gerais. The S. Minnesota isolates showed resistance to tetracycline (80%), cefoxitin (80%), ceftazidime (46.7%), nalidixic acid (23.3%), ciprofloxacin (13.3%), and streptomycin (10%). No resistance to gentamicin, chloramphenicol, meropenem, nitrofurantoin, and sulfamethoxazole-trimethoprim was found. Moreover, 23.3% of the evaluated isolates presented multi-resistance profile, all from Minas Gerais. The results highlight the importance of further studies involving S. Minnesota, which is prevalent in the Brazilian broiler flocks and could provoke foodborne infection in humans.

Dissertação de mestrado em Biotecnologia, O monóxido de carbono (CO) é um gás poluente produzido por vários setores industriais (por exemplo, energia e indústria química). É também um componente importante, habitualmente o mais abundante do gás síntese (syngas), que é gerado a partir da gaseificação de materiais contendo carbono. O glicerol é um inevitável subproduto gerado em grandes quantidades durante processos industriais de produção de bioetanol e de biodiesel. Syngas/CO e glicerol podem ser usados por alguns microrganismos anaeróbios para produzir combustíveis ou produtos químicos de elevado valor económico. O objetivo deste trabalho foi construir um consórcio microbiano capaz de converter CO/Syngas e/ou glycerol em propionato. O propionato é um composto de elevado valor com inúmeras aplicações industriais: aplicações variadas na indústria farmacêutica, agente antifúngico na indústria alimentar e funciona também como um percursor para a construção química para produzir plásticos e herbicidas. Um enriquecimento mesofílico (37 ºC) capaz de produzir propionato foi obtido a partir biomassa anaeróbia (biomassa granular) através de transferências sucessivas em CO (≈1.0 bar de pressão parcial) mais acetato (20 mM). A diversidade microbiana desta cultura foi estudada por análise do gene 16S rRNA, sendo identificados como predominantes os microrganismos pertencentes ao gênero Acetobacterium. Microrganismos relacionados com Clostridium propionicum e C. neopropionicum (identidade do gene 16S rRNA> 98%) e com Pelobacter sp. (93% de identidade do gene 16S rRNA) também foram abundantes. Uma nova estirpe, designada strain JM, 99% de identidade do gene 16S rRNA Acetobacterium sp. foi isolada de um enriquecimento microbiano usando técnicas de diluição em série, em meio líquido e sólido, e usando CO como única fonte de carbono e energia. Este organismo consegue converter CO em acetato e / ou etanol, e é também capaz de converter glicerol em 1,3-propanodiol. Foi construído um consórcio sintético (co-cultura) composto por Acetobacterium sp. estirpe JM e por Clostridium neopropionicum, que tem a capacidade de produzir propionato a partir de CO (0,1 mol de propionato/ mol de CO convertido). Com o objetivo de obter informações sobre a fisiologia e bioquímica da conversão biológica de CO a propionato e as potenciais interações microbianas entre os dois microrganismos no consórcio, foi aplicada a técnica de proteómica., Carbon monoxide (CO) is a polluting gas produced by several industrial sectors (e.g. energy, chemical industry). It is also a major component of syngas generated from the gasification of carbon-containing materials. Glycerol is an inevitable byproduct generated in large amounts during the production processes of both bioethanol and biodiesel industries. Syngas/CO and glycerol can be used by some anaerobes to produce fuels or valuable chemicals. The aim of this work was to construct a microbial consortium capable of converting CO/ Syngas and/or glycerol to propionate. Propionate is a value-added compound with numerous industrial applications, e.g. as an antifungal agent in foods and feeds, and as a building block to produce plastics and herbicides. A mesophilic (37 ºC) propionate-producing enrichment was obtained from anaerobic sludge by successive transfers on CO (≈1.0 bar of partial pressure) plus acetate (20 mM). Microbial diversity of this culture was studied by 16S rRNA gene analysis, and microorganisms belonging to Acetobacterium genus were identified as predominant. Microorganisms related to Clostridium propionicum and C. neopropionicum (16S rRNA gene identity > 98%) and to Pelobacter species (93% identity) were also abundant. A new Acetobacterium sp. strain JM was isolated from the enrichment culture using dilution series technique and solid medium using CO as sole carbon and energy source. This organism was able to convert CO to acetate and/or ethanol, and able to convert glycerol into 1,3-propanediol. A synthetic consortium (co-culture) composed of Acetobacterium sp. strain JM and Clostridium neopropionicum was constructed and was capable of producing propionate from CO (0.1 mol propionate/mol CO converted). O quadro atual aponta para um decréscimo dos níveis de competência motora em crianças, com consequente diminuição da prática de atividades físicas, queda nos níveis de aptidão física e prejuízos à saúde e qualidade de vida dessa população. Assim, parece importante investir em programas que busquem ampliar tal competência desde a infância, especialmente nas aulas de Educação Física. Para isso, além de estratégias de intervenção, é necessária a realização de testes motores para medir os níveis de competência motora, a fim de monitorar o processo. Neste sentido, é preciso um teste motor válido, confiável, objetivo, de baixo custo, que possa ser facilmente aplicado e analisado. O Teste de Coordenação Corporal para Crianças - KTK (Körperkoordinationstest Für Kinder) parece ser uma excelente opção. Composto por quatro tarefas: Equilíbrio na Trave, Saltos Laterais, Transferência sobre Plataformas e Saltos Monopedais, propõe medir o fator competência motora. Os objetivos da pesquisa foram testar a validade do KTK para uma amostra brasileira composta por 295 crianças, de 5 e 10 anos de idade, além de propor uma equação para o cálculo do seu quociente motor geral, que leve em consideração a influência específica de cada uma das tarefas do teste para medir a competência motora, o que não é considerado em seu formato original. Verificou-se também a influência do sexo e da idade das crianças nas pontuações obtidas por elas nos itens testados. Os resultados demonstraram que as propriedades psicométricas e a estrutura fatorial do KTK foram bem ajustadas ao modelo, que se mostrou adequado, confirmando a competência motora como seu único fator, além de mostrar que as cargas fatoriais dos itens da bateria são diferentes. Por meio da abordagem da regressão dos mínimos quadrados para obtenção do escore fatorial, chegou- se à equação para o cálculo do novo quociente motor do KTK, que leva em consideração a influência que cada sub-teste tem no resultado final da bateria e permite a comparação dos resultados entre grupos distintos. Os resultados também sugerem que a idade interfere na pontuação das quatro tarefas do KTK e do novo quociente motor e que o sexo interfere nas tarefas Transferência sobre Plataformas e Saltos Monopedais, e também no novo quociente motor. Pode-se concluir que o KTK é um teste válido e confiável para medir a competência motora de crianças brasileiras, sensível ao sexo e à idade, de simples execução, com resultados objetivos, de baixo custo, aspectos que o qualificam como um bom instrumento a ser conduzido nas escolas brasileiras. Além disso, o novo quociente motor, que ressalta o peso de cada tarefa no resultado final do teste, irá oferecer novas formas de interpretação desse resultado e novas possibilidades para a pesquisa. The current situation points towards a reduction in motor competence levels in children, with a consequent decline in their practice of physical activities, a drop in their levels of physical aptitude, and damages to the health and quality of life of such population. Therefore, it seems important to invest in programs which aim to broaden such competence from as early as childhood, especially in physical education classes. In addition to well-planned and coherent interventions, it is necessary to carry out systematic motor tests to measure the levels of motor competence, in order to monitor the whole process. It is therefore necessary to have a motor test which is valid, trustworthy, objective and low-cost, and which may be easily administered and interpreted. For such purposes, the Körperkoordinationstest Für Kinder – KTK seems to be an excellent option. The KTK is a German test, established for over four decades, widely mentioned in the literature and much used in Brazil. Composed of four tasks – Walking Backwards, Jumping Sideways, Moving Sideways and Hopping for Height – the test aims at measuring the motor competence factor. The objectives of this study were to test the validity of the KTK to a sample of 295 Brazilian children, aged 5 to 10 years, as well as to suggest an equation to calculate a general motor quotient that takes into consideration the specific influence to each of the four motor-competence tests, which is not considered in the KTK’s original format. The influence of gender and age in the scores obtained by the children in each test was also analyzed. The results showed that the psychometric properties and the KTK structure were well-adjusted to the model (x² = 3,972; p = 0,137; CFI = 0,997; TLI = 0,990; RMSEA = 0,058; SRMR = 0,012), and confirmed the motor competence as its only latent factor, as well as the fact that the factorial charges of the battery items are different. By using the least squares regression approach to obtain the factorial score, it was possible to reach an equation to calculate the KTK new motor quotient, which takes into consideration the influence of each subtest in the battery final result, and allows the results of distinct groups to be compared. The results also suggested that age interferes in the scores of all four KTK and new motor quotient tasks and that gender interferes in the Moving Sideways and Hopping for Height, and also in the new motor quotient. It was possible to conclude that the KTK is a valid, low-cost and trustworthy test to measure the motor competence of Brazilian children, sensitive to gender and age, of simple execution and objective results. Such aspects qualify the KTK as a good instrument to be used by Brazilian schools. In addition, the new motor quotient, which highlights the weight of each task in the test’s final result, will offer new ways to interpret such result and new possibilities of research.

The aim of this study was to verify the influence of the technical skills level on the decision making of novice volleyball players on service, reception and set actions. The sample was composed of 80 novice athletes of school volleyball teams of both sexes. Based on the results from the Volleyball Skills Test Battery, the sample was divided into three groups: low, intermediate and high technical level, by means of One-Way ANOVA. The intermediate group was excluded from further analyses. Decision making was assessed with the Game Performance Assessment Instrument (GPAI). For data analysis purposes, the independent student-t test was used, adopting significance level of p≤0.05. The results showed that players with low skill level in service and set actions also had lower performance on the decision-making test when compared to players with high technical level. On the other hand, in the reception action, no differences were found among groups. It was therefore concluded that the technical level seems to influence the decision making in service and set actions in novice volleyball players. Resumo O estudo objetivou verificar a influência do nível da habilidade técnica na tomada de decisão de jogadores iniciantes de voleibol nas ações de saque, recepção e levantamento. A amostra foi composta por 80 atletas iniciantes de equipes escolares de voleibol de ambos os sexos. Com base nos resultados na Bateria de Testes de Habilidades de Voleibol, a amostra foi dividida em três grupos: baixo, intermediário e alto nível técnico, por meio da ANOVA One-Way. O grupo intermediário foi excluído das análises posteriores. A tomada de decisão foi avaliada por meio do Game Performance Assessment Instrument (GPAI). Utilizou-se o teste t student independente para análise dos dados, adotando um nível de significância de p≤0,05. Os resultados encontrados demonstraram que os jogadores com baixo nível de habilidade nas ações de saque e levantamento, também obtiveram um pior desempenho no teste de tomada de decisão quando comparado aos jogadores de alto nível técnico. Por outro lado, na ação de recepção, não foram encontradas diferenças significativas entre os grupos. Conclui-se que o nível técnico parece influenciar na tomada de decisão na ação de saque e levantamento dos jogadores iniciantes de voleibol.

This paper presents the design of a right knee joint prototype, from a 3D scanner, and then obtaining an output STL file for the 3D printer. Two different types of plastic (ABS and PLA) and two models of 3D printer (MakerBot Replicator 2X and Flashforge Creator Pro) were used in achieving the goal of reproducing as accurate as possible and cheaper than the original knee prototype, purchased on the internet, made of polyvinyl chloride (PVC) plastic by a German factory specialized in making scientific prototypes. Visual quality, production time, and weight of the printed parts were compared with reference to the machinery, material types, quantity, and printing parameters. Through this hands-on project, the students were trained in emerging manufacturing technologies such as 3D scanning, 3D printing and rapid prototyping, and additive manufacturing. Relative Age Effect (RAE) and Constituent Year Effect (CYE) seem to influence the process of training young tennis players. The aim of this study was to investigate the presence of these effects in the ranking of junior tennis players of the International Tennis Federation (ITF) and its influence on the score obtained by such tennis players. The results show the presence of the RAE and the CYE (p = 0.05). The regression model identified that the variables "year" and "month" of birth predict the scores of athletes in the ITF ranking with an explained variance of 19%. In conclusion, the ranking composed of various age groups does not seem to be a classification strategy sensitive enough to prevent the RAE and the CYE among junior tennis players. Background Schistosomiasis constitutes a major public health problem, and 200 million people are estimated to be infected with schistosomiasis worldwide. In Brazil, schistosomiasis has been reported in 19 states, showing areas of high and medium endemicity and a wide range of areas of low endemicity (ALE). Barra Mansa in Rio de Janeiro state has an estimated prevalence of 1%. ALE represent a new challenge for the helminth control because about 75% of infected individuals are asymptomatic and infections occur with a low parasite load (<100 eggs per gram of feces), causing a decrease in sensitivity of stool parasitological techniques, which are a reference for the laboratory diagnosis of this helminth. The objective of this study was to evaluate the performance of a TaqMan quantitative polymerase chain reaction (qPCR) technique in serum and feces DNA samples using the techniques of Kato- Katz (KK), Hoffman, Pons and Janer (HH) as references, during an epidemiological survey using fecal samples and sera from randomized residents from an ALE. Methods A cross-sectional study conducted from April to December 2011 using a probabilistic sampling that collected 572 fecal and serum samples. The laboratory diagnostic techniques used were: KK, HH and qPCR (feces and serum). Results We obtained the following results using the different diagnostic techniques: KK and HH, 0.9% (n =5); qPCR-feces, 9.6% (n =55); and qPCR-serum, 1.4% (n =8). The qPCR-feces presented the highest positivity, whereas the techniques of HH and KK were the least sensitive to detect infections (0.8%). Compared to HH and KK, qPCR-feces showed a statistically significant difference in positivity (p <0.05), although with poor agreement. Conclusion The positivity rate presented by the qPCR approach was far higher than that obtained by parasitological techniques. The lack of adequate surveillance in ALE of schistosomiasis indicates a high possibility of these areas being actually of medium and high endemicity. This study presents a control perspective, pointing to the possibility of using combined laboratory tools in the diagnosis of schistosomiasis in ALE. [ABSTRACT FROM AUTHOR]

- Abstract
Lipid droplets (LDs) are organelles involved in lipid storage, maintenance of energy homeostasis, protein sequestration, signaling events and inter-organelle interactions. Recently, LDs have been shown to favor the replication of members from different viral families, such as the Flaviviridae and Coronaviridae. In this work, we show that LDs are essential organelles for members of the Arenaviridae family. A virus-driven reduction of LD number was observed in cultures infected with Junín mammarenavirus (JUNV), caused in part by action of the viral nucleoprotein. Notably, we identified a new pool of nucleoprotein and viral RNA that localizes in the vicinity of LDs, suggesting that LDs play a role during the viral replication cycle. Regarding the mechanism behind LD exhaustion, we found evidence that lipophagy is involved in LD degradation with the resulting fatty acids being substrates of fatty acid β-oxidation, which fuels viral multiplication.
