Microbial communities that inhabit the host's intestine influence many aspects of the host's health and bear the adaptive potential to alterations in harsh environments and diets. The Qinghai‐Tibet Plateau represents one of the harshest environments in the world. Preliminary progress has been made in identifying the communities of gut microbes in Indigenous Tibetans and non‐human animals. However, due to the complexity of microbial communities, the effects of gut microbes on the host's health and high‐plateau adaptation remain unexplained. Herein, we review the latest progress in identifying factors affecting the gut microbiota of native Tibetans and non‐human animals and highlight the complex interactions between the gut microbiota, health and highland adaptation, which provides a basis for exploring the correlations between the gut microbiota and clinical indexes in native highland residents and travellers, as well as developing microbiota‐based strategies to mitigate health risks for tourists and treatments for mountain sickness during high‐altitude travel in the future. Influence of water content on the expression of lignocellulolytic enzymes by Phanerochaete chrysosporium remains unclear. This work compares the enzyme production profiles of P. chrysosporium during solid-state and submerged fermentation. There were 110 and 64 extracellular carbohydrate-active enzymes identified in solid-state and submerged fermentation respectively, among which 57 enzymes were common to both of the secretomes. P. chrysosporium secreted more cellulases (especially lytic polysaccharide monooxygenase) and hemicellulases during solid-state fermentation while the proportion of enzyme containing carbohydrate-binding module was higher for submerged fermentation. Although its activities were weaker, the enzyme cocktail from submerged fermentation was surprisingly more effective in hydrolysis at low substrate loading. This advantage of enzymes from submerged fermentation was mainly attributed to carbohydrate-binding module because more xylanases bound with substrate at the beginning of hydrolysis. A comparative experimental and numerical study of the impact behaviour of carbon-fiber-reinforced thermoplastic (TP) and thermoset (TS) composites has been carried out. On the one hand, low velocity impact (LVI) tests were performed on TP and TS composites with different lay-up sequences at different energy levels, and the damage modes and microscopic damage mechanisms after impact were investigated using macroscale inspection, C-scan inspection, and X-ray-computed tomography. The comparative results show that the initial damage valve force under LVI depends not only on the material, but also on the layup sequence. The initial valve force of the P2 soft layer with lower stiffness is about 11% lower than that of the P1 quasi-isotropic layer under the same material, while the initial valve force of thermoplastic composites is about 28% lower than that of thermoset composites under the same stacking order. Under the same stacking order and impact energy level, the damage area and depth of TP composites are smaller than those of TS composites; while under the same material and impact energy level, the indentation depth of P2 plies is greater than that of P1 plies, and the damage area of P2 plies is smaller than that of P1 plies, but the change of thermoplastic composites is not as obvious as that of thermoset composites. This indicates that TP composites have a higher initial damage threshold energy and impact resistance at the same lay-up order, while increasing the lay-up ratio of the same material by 45° improves the impact resistance of the structure. In addition, a damage model based on continuum damage mechanics (CDM) was developed to predict different damage modes of thermoplastic composites during low velocity impact, and the analytical results were compared with the experimental results. At an impact energy of 4.45 J/mm, the error of the initial damage valve force is 5.26% and the error of the maximum impact force is 4.36%. Herein, we report the secretome of Penicillium chrysogenum P33 under induction of lignocellulose for the first time. A total of 356 proteins were identified, including complete cellulases and numerous hemicellulases. Supplementing a commercial cellulase with increasing dosage of P33 enzyme cocktail from 1 to 5 mg/g substrate increased the release of reducing sugars from delignified corn stover by 21.4% to 106.8%. When 50% cellulase was replaced by P33 enzyme cocktail, release of reducing sugars was 78.6% higher than with cellulase alone. Meanwhile, glucan and xylan conversion was increased by 37% and 106%, respectively. P33 enzyme cocktail also enhanced commercial cellulase hydrolysis against four different delignified lignocellulosic biomass. These findings demonstrate that mixing appropriate amount of P33 cocktail with cellulase improves polysaccharide hydrolysis, suggesting P33 enzymes have great potential for industrial applications. In this study, lignite was degraded using a purified esterase from Penicillium decumbens P6 for the first time. The esterase was purified using ammonium sulfate precipitation, anion exchange and gel filtration chromatography. The recovery and purification yield of the enzyme were 15% and 83 folds, respectively. The molecular weight of the purified enzyme was about 45 kDa. Both crude and purified esterases were studied for lignite depolymerization. The tendency of depolymerization by crude enzyme was consistent with the enzyme secreted in the medium. Along with the increased purified esterase concentration from 8 to 50 mg/ml, A 450 value increased from 0.38 to 2.08. The contribution of esterase to the depolymerization was about 40% in the crude supernatant. Compared with aHA (crude lignite humic acid), bHA (esterase degraded lignite humic acid) has a lower percentage of aromatic carbon and ester groups, but a higher percentage of aliphatic carbon. bHA can promote the growth of asparagus lettuce. The results demonstrated that lignite was depolymerized by the purified esterase and evidenced the potential of esterase application in conversion of lignite into compounds with high bioactivities. Stevioside is a non-caloric, natural, high-intensity sweetener. However, the bitter aftertaste of stevioside restricts its utilization for human consumption and limits its application in the food industry. In this study, a high efficiency enzymatic modification system was investigated to improve stevioside taste quality. A cyclodextrin glucanotransferase (CGTase) producing strain Paenibacillus sp. CGMCC 5316 was isolated from Stevia planting soil. With starch as glycosyl donor, this CGTase can transform stevioside into a single specific product which is an isomer of rebaudioside A and identified as mono-glycosylated stevioside . The taste of stevioside is improved noticeably by generating mono-glycosylated stevioside, which possesses a sucrose-like taste and has sweetness increased significantly by 35.4 %. Next, the parameters influencing CGTase production were optimized. Compared to initial conditions, CGTase activity increased by 214.7 % under optimum conditions of 3.9 g/L starch, 17.9 g/L tryptone, and 67.6 h of culture time, and the transglycosylation rate of stevioside was remarkably increased by 284.8 %, reaching 85.6 %. This CGTase modification system provides a promising solution for improving the sweetness and taste quality of stevioside. Microalgae are the largest CO 2 fixer and O 2 producer on the earth and occupy an increasingly important position in human life and production. Various environmental factors have a significant impact on the growth and metabolism of microalgae. As global warming intensifies, heat stress has become a crucial factor affecting the microalgae industry. However, till now, it has not been clear how microalgae sensed the temperature stress, transmitted stress signals and adjusted in intracellular metabolic pathways. In this study, the growth of microalgae Auxenochlorella protothecoides UTEX2341 was inhibited at 32 °C, but the single cell dry weight increased. The cell component analyses showed that both the carbohydrate and total protein content decreased significantly, while the lipid content increased by 158%. Meanwhile, the intracellular Ca2+ concentration increased continuously, with a maximum increase of 1.65 times. According to the transcriptome analyses, the up-regulation of Ca2+ influx channel protein mid1-complementing activity 1 (MCA1) gene and the down-regulation of efflux channel protein cation exchanger 1(CAX) and autoinhibited Ca2+-ATPase 1 (ACA1) genes in cytoplasmic membrane jointly facilitated the increase of Ca2+ in the cytoplasm. Coexpression network analysis indicated that the fluctuation of Ca2+ in the cytoplasm could activate the expression of transcription factors MYB3 and AP2-4 through calmodulin (CAM) and calcium-dependent protein kinase (CDPK), and then regulate glycerol-3-phosphate acyltransferases (GPAT) at the beginning of TAG synthesis and diacylglycerol acyltransferase (DGAT)/phospholipid: diacylglycerol acyltransferase (PDAT) in the last step of TAG synthesis. Furthermore, the addition of Ca2+ specific chelator BAPTA-AM inhibited the expression of GPAT , which was consistent with the decrease in microalgae lipid content. The results proved that Ca2+ participated in the regulation of microalgae TAG synthesis under heat stress, which provided a new view for the understanding of the microalgae lipid accumulation mechanism. Phosphate accumulating bacterium Pseudomonas stutzeri YG-24 exhibited efficient heterotrophic nitrification and aerobic denitrification ability. Single factor experiments showed that both heterotrophic nitrification and aerobic denitrification occurred with sodium citrate as carbon source and lower C/N ratio of 8. High average NH 4 + -N, NO 2 − -N and NO 3 − -N removal rates of 8.75, 7.51 and 7.73 mg L −1 h −1 were achieved. The application of strain YG-24 in wastewater samples resulted in TN, NH 4 + -N, NO 2 − -N, NO 3 − -N and P removal efficiencies of 85.28%, 88.13%, 86.15%, 70.83% and 51.21%. Sequencing and quantitative amplification by real-time PCR of napA , nirS and ppk showed that nitrogen removal pathway of strain YG-24 was achieved through heterotrophic ammonium nitrification coupled with fast nitrite denitrification (NH 4 + -N to NO 2 − -N and then to gaseous nitrogen) directly. Algae lipid production combined with heavy metal removal is a cost-effective and environment-friendly method for algae biofuel production and hazardous waste treatment. Chlorella minutissima UTEX 2341 had strong resistance to cadmium, copper, manganese and zinc ions under heterotrophic culture condition and could efficiently remove them through intracellular accumulation and extracellular immobilization. Meanwhile, lipid accumulation was not inhibited by heavy metals. Instead, the algae lipid content significantly increased by 21.07% and 93.90%, respectively with the addition of cadmium and copper. Furthermore, the heavy metal residue in lipid was within μg range and satisfied the commercial standard. This artificial wastewater-algae biofuel-heavy-metal integrated utilization technology offered a new alternative solution to the problems of energy shortage and environmental pollution. The vibration and damping characteristics of free-free composite sandwich cylindrical shell with pyramidal truss-like cores have been conducted using the Rayleigh-Ritz model and finite element method. The predictions for the modal properties of composite sandwich cylindrical shell with pyramidal truss-like cores showed good agreement with the experimental tests. The influences of fiber ply angles on the natural frequency and damping loss factor were investigated. Three types of such composite sandwich cylindrical shells were manufactured using a hot press molding method and the relevant modal characteristics of various sandwich cylindrical shells could be obtained by modal tests. It can be found that modal strain energy approach was effective to estimate the structural damping loss factors and the variation of damping for each vibration modes could be explained suitably through the contributions of each strain energy components. Results showed that the structural damping loss factors not only depended on inherent material damping, but also relied on the vibration modes. The natural frequencies of composite sandwich cylindrical shell increased with the increasing of the ply angle of the inner and outer curve face sheets, whereas the damping loss factors of present shells did not increase monotonically. The economical production of 5‐aminolevulinic acid (ALA) has recently received increasing attention for its extensive use in agriculture. In this study, a strain of Bacillus cereus PT1 could initially produce ALA at a titre of 251.72 mg/L by using a hydrolysate mixture of low‐cost cassava residue and fish waste. The integration of endogenous hemA encoding glutamyl‐tRNA reductase led to a 39.30% increase in ALA production. Moreover, improving cell permeability by deletion of the LytR‐CpsA‐Psr (LCP) family gene tagU led to a further increase of 59.73% in ALA production. Finally, the engineered strain B. cereus PT1‐hemA‐ΔtagU produced 2.62 g/L of ALA from the previously mentioned hydrolysate mixture in a 7‐L bioreactor. In a pot experiment, foliar spray of the ALA produced by B. cereus PT1‐hemA‐ΔtagU from the hydrolysates increased salt tolerance of cucumber by improving chlorophyll content and catalase activity, while decreasing malondialdehyde content. SUMMARY The diversity in alternative splicing of long noncoding RNAs (lncRNAs) poses a challenge for functional annotation of lncRNAs. Moreover, little is known on the effects of alternatively spliced lncRNAs on crop yield. In this study, we cloned nine isoforms resulting from the alternative splicing of the lncRNA LAIR in rice. The LAIR isoforms are generated via alternative 5′/3′ splice sites and different combinations of specific introns. All LAIR isoforms activate the expression of the neighboring LRK1 gene and enhance yield‐related rice traits. In addition, there are slight differences in the binding ability of LAIR isoforms to the epigenetic modification‐related proteins OsMOF and OsWDR5, which affect the enrichment of H4K16ac and H3K4me3 at the LRK1 locus, and consequently fine‐tune the regulation of LRK1 expression and yield‐related traits. These differences in binding may be caused by polymorphic changes to the RNA secondary structure resulting from alternative splicing. It was also observed that the composition of LAIR isoforms was sensitive to abiotic stress. These findings suggest that the alternative splicing of LAIR leads to the formation of a functional transcript population that precisely regulates yield‐related gene expression, which may be relevant for phenotypic polymorphism‐based crop breeding under changing environmental conditions. Background: Yield and quality are the two most important traits in crop breeding. Exploring the regulatory mechanisms that affect both yield and quality traits is of great significance for understanding the molecular genetic networks controlling these key crop attributes. Expansins are cell wall loosening proteins that play important roles in regulating rice grain size. Results: We investigated the effect of OsEXPA7, encoding an expansin, on rice grain size and quality. OsEXPA7 overexpression resulted in increased plant height, panicle length, grain length, and thousand-grain weight in rice. OsEXPA7 overexpression also affected gel consistency and amylose content in rice grains, thus affecting rice quality. Subcellular localization and tissue expression analyses showed that OsEXPA7 is localized on the cell wall and is highly expressed in the panicle. Hormone treatment experiments revealed that OsEXPA7 expression mainly responds to methyl jasmonate, brassinolide, and gibberellin. Transcriptome analysis and RT-qPCR experiments showed that overexpression of OsEXPA7 affects the expression of OsJAZs in the jasmonic acid pathway and BZR1 and GE in the brassinosteroid pathway. In addition, OsEXPA7 regulates the expression of key quantitative trait loci related to yield traits, as well as regulates the expression levels of BIP1 and bZIP50 involved in the seed storage protein biosynthesis pathway. Conclusions: These results reveal that OsEXPA7 positively regulates rice yield traits and negatively regulates grain quality traits by involving plant hormone pathways and other trait-related pathway genes. These findings increase our understanding of the potential mechanism of expansins in regulating rice yield and quality traits and will be useful for breeding high-yielding and high-quality rice cultivars. Liberating high value-added compounds ferulic acid (FA) and xylo-oligosaccharides (XOSs) from agricultural residues is a promising strategy for the utilization of lignocellulose. In this study, a bifunctional xylanase/feruloyl esterase from bacterial consortium EMSD5 was heterogeneously expressed in Escherichia coli. Depending on the inter-domain synergism of the recombinant enzyme rXyn10A/Fae1A, high yields of FA (2.78, 1.82, 1.15 and 7.31 mg/g substrate, respectively) were obtained from 20 mg in-soluble wheat arabinoxylan, de-starched wheat bran, ultrafine-grinding corn stover and steam-exploded corncob. Meanwhile, 3.210, 1.235, 1.215 and 0.823 mg xylose/XOSs were also released. For cost-saving enzyme production, we firstly constructed a recombinant E. coli , which could secrete the bifunctional xylanase/feruloyl esterase out of cells. When the recombinant E. coli was cultured in medium containing 200 mg de-starched wheat bran, 474 μg FA and 18.2 mg xylose/XOSs were also detected. • Bacterial consortium EMSD5 produced isopropanol from xylans efficiently. • 7.00 g/L isopropanol was produced from unpretreated corncob by consortia-based CBP. • Arabinose residues of xylan could enhance the CBP performance. • Clostridium beijerinckii and lignocellulose degraders performed CBP synergistically. Consolidated bioprocessing (CBP) of lignocellulose by bacterial consortium for isopropanol production is considered as the most promising strategy. However, low utilization of xylan caused by the complex sidechain structure remains inhibit the conversion of full-biomass. In this study, isopropanol production from different lignocelluloses by the consortium EMSD5 through CBP was performed. A total of 7.00 g/L of isopropanol was obtained from corncob by optimizing fermentation conditions. Isopropanol production by EMSD5 was mainly based on utilizing xylan in corncob and isopropanol titer was increased by 47.71% and reached up to 8.39 g/L using arabinoxylan compared with linear xylan. The analysis of community structures and the isolation of key strain confirmed the enrichment of the isopropanol producer, Clostridium beijierinckii , in the bacterial community when it was incubated with corn glucuronoarabinoxylan and the cooperation between C. beijerinckii and lignocellulose degraders. Background: Xylanases randomly cleave the internal β-1,4-glycosidic bonds in the xylan backbone and are grouped into different families in the carbohydrate-active enzyme (CAZy) database. Although multiple xylanases are detected in single strains of many filamentous fungi, no study has been reported on the composition, synergistic effect, and mode of action in a complete set of xylanases secreted by the same microorganism. Results: All three xylanases secreted by Penicillium chrysogenum P33 were expressed and characterized. The enzymes Xyl1 and Xyl3 belong to the GH10 family and Xyl3 contains a CBM1 domain at its C-terminal, whereas Xyl2 belongs to the GH11 family. The optimal temperature/pH values were 35 °C/6.0, 50 °C/5.0 and 55 °C/6.0 for Xyl1, Xyl2, and Xyl3, respectively. The three xylanases exhibited synergistic effects, with the maximum synergy observed between Xyl3 and Xyl2, which are from different families. The synergy between xylanases could also improve the hydrolysis of cellulase (C), with the maximum amount of reducing sugars (5.68 mg/mL) observed using the combination of C + Xyl2 + Xyl3. Although the enzymatic activity of Xyl1 toward xylan was low, it was shown to be capable of hydrolyzing xylooligosaccharides into xylose. Xyl2 was shown to hydrolyze xylan to long-chain xylooligosaccharides, whereas Xyl3 hydrolyzed xylan to xylooligosaccharides with a lower degree of polymerization. Conclusions: Synergistic effect exists among different xylanases, and it was higher between xylanases from different families. The cooperation of hydrolysis modes comprised the primary mechanism for the observed synergy between different xylanases. This study demonstrated, for the first time, that the hydrolysates of GH11 xylanases can be further hydrolyzed by GH10 xylanases, but not vice versa. [ABSTRACT FROM AUTHOR]

Symbiotic nitrogen fixation (SNF) is a crucial process for nitrogen geochemical cycling and plant‐microbe interactions. Water‐soluble humic acid (WSHM), an active component of soil humus, has been shown to promote SNF in the legume‐rhizobial symbiosis, but its molecular mechanism remains largely unknown. To reveal the SNF‐promoting mechanism, we conducted transcriptomic analysis on soybean treated with WSHM. Our findings revealed that up‐ and downregulated differentially expressed genes (DEGs) were mainly involved in plant cell‐wall/membrane formation and plant defence/immunity in the early stage, while the late stage was marked by the flavonoid synthesis and ethylene biosynthetic process. Further study on representative DEGs showed that WSHM could inhibit GmBAK1d‐mediated immunity and BR signalling, thereby promoting rhizobial colonisation, infection, and nodulation, while not favoring pathogenic bacteria colonisation on the host plant. Additionally, we also found that the ethylene pathway is necessary for promoting the soybean nodulation by WSHM. This study not only provides a significant advance in our understanding of the molecular mechanism of WSHM in promoting SNF, but also provides evidence of the beneficial interactions among the biostimulator, host plant, and soil microbes, which have not been previously reported. Background: As part of the plant microbiome, endophytic bacteria play an essential role in plant growth and resistance to stress. Water-soluble humic materials (WSHM) is widely used in sustainable agriculture as a natural and non-polluting plant growth regulator to promote the growth of plants and beneficial bacteria. However, the mechanisms of WSHM to promote plant growth and the evidence for commensal endophytic bacteria interaction with their host remain largely unknown. Here, 16S rRNA gene sequencing, transcriptomic analysis, and culture-based methods were used to reveal the underlying mechanisms. Results: WSHM reduced the alpha diversity of soybean endophytic bacteria, but increased the bacterial interactions and further selectively enriched the potentially beneficial bacteria. Meanwhile, WSHM regulated the expression of various genes related to the MAPK signaling pathway, plant-pathogen interaction, hormone signal transduction, and synthetic pathways in soybean root. Omics integration analysis showed that Sphingobium was the genus closest to the significantly changed genes in WSHM treatment. The inoculation of endophytic Sphingobium sp. TBBS4 isolated from soybean significantly improved soybean nodulation and growth by increasing della gene expression and reducing ethylene release. Conclusion: All the results revealed that WSHM promotes soybean nodulation and growth by selectively regulating soybean gene expression and regulating the endophytic bacterial community, Sphingobium was the key bacterium involved in plant-microbe interaction. RNA‐binding proteins (RBPs) are components of the post‐transcriptional regulatory system, but their regulatory effects on complex traits remain unknown. Using an integrated strategy involving map‐based cloning, functional characterizations, and transcriptomic and population genomic analyses, we revealed that RBP‐K (LOC_Os08g23120), RBP‐A (LOC_Os11g41890), and RBP‐J (LOC_Os10g33230) encode proteins that form an RBP‐A‐J‐K complex that negatively regulates rice yield‐related traits. Examinations of the RBP‐A‐J‐K complex indicated RBP‐K functions as a relatively non‐specific RBP chaperone that enables RBP‐A and RBP‐J to function normally. Additionally, RBP‐J most likely affects GA pathways, resulting in considerable increases in grain and panicle lengths, but decreases in grain width and thickness. In contrast, RBP‐A negatively regulates the expression of genes most likely involved in auxin‐regulated pathways controlling cell wall elongation and carbohydrate transport, with substantial effects on the rice grain filling process as well as grain length and weight. Evolutionarily, RBP‐K is relatively ancient and highly conserved, whereas RBP‐J and RBP‐A are more diverse. Thus, the RBP‐A‐J‐K complex may represent a typical functional model for many RBPs and protein complexes that function at transcriptional and post‐transcriptional levels in plants and animals for increased functional consistency, efficiency, and versatility, as well as increased evolutionary potential. Our results clearly demonstrate the importance of RBP‐mediated post‐transcriptional regulation for the diversity of complex traits. As the third pole of the world and Asia's water tower, the Tibetan Plateau experiences daily release of pharmaceutical and personal care products (PPCPs) due to increasing human activity. This study aimed to explore the potential relationship between the concentration and composition of PPCPs and human activity, by assessing the occurrence of PPCPs in areas of typical human activity on the Qinghai-Tibet Plateau and evaluating their ecological risk. The results indicate that 28 out of 30 substances were detected in concentrations ranging from less than 1 ng/L to hundreds of ng/L, with the average concentration of most PPCPs in the Tibet Autonomous Region being higher than that in Qinghai Province. Among the detected substances, CAF, NOR, CTC, CIP, TCN, OTC, AZN, and DOX accounted for over 90% of the total concentration. The emission sources of PPCPs were identified by analyzing the correlation coefficients of soil and water samples, with excess PPCPs used by livestock breeding discharged directly into soil and then into surface water through leaching or runoff. By comparing the concentration and composition of PPCPs with those in other regions, this study found that CIP, ENR, LOM, NOR, CTC, DOX, OTC, and TCN were the most commonly used PPCPs in the Qinghai-Tibet Plateau. To assess the ecological risk of PPCPs, organisms at different trophic levels, including algae, crustaceans, fish, and insects, were selected. The prediction of the no effect concentration of each PPCP showed that NOR, CTC, TCN, CAF, and CBZ may have deleterious effects on water biota. This study can assist in identifying the emission characteristics of PPCPs from different types and intensities of human activities, as well as their occurrence and fate during the natural decay of aquatic systems. Most widely studied Structural Health Monitoring methods need baseline data from the undamaged state and a high number of sensors attached to the structure, which makes these methods difficult to be applied in real structures for on-line monitoring. In order to deal with that, the NOde DISplacement (NODIS) method is proposed by considering the structural damage indicator. The idea of the present work is to develop a baseline-free damage location and quantification method based on the node displacement of structural mode shapes. The damage location procedure is developed by combining the nodes from different mode shapes and the damage quantification is achieved by the proposed Transfer-Function-based Damage Indicator (TFDI). The advantages of this method are: (1) baseline-free, in terms of the comparison with a reference signal is not needed, (2) only a relatively small number of sensors is required for real-time damage location and (3) with the proposed TFDI, the damage detection result can be quantified. In the present paper, the baseline-free damage location algorithm is proposed and validated on a steel beam with an analytical model, a numerical model and experiments. In addition, the influence of temperature, the additional weight of the sensor and the error in sensor location on the NODIS method are studied. After that, the TFDI is developed and improved for the experiment. Amylose content (AC) is the main factor determining the palatability, viscosity, transparency, and digestibility of rice (Oryza sativa) grains. AC in rice grains is mainly controlled by different alleles of the Waxy (Wx) gene. The AP2/EREBP transcription factor OsEBP89 interacts with the MYC‐like protein OsBP5 to synergistically regulate the expression of Wx. Here, we determined that the GLYCOGEN SYNTHASE KINASE 5 (OsGSK5, also named SHAGGY‐like kinase 41 [OsSK41]) inhibits the transcriptional activation activity of OsEBP89 in rice grains during amylose biosynthesis. The loss of OsSK41 function enhanced Wx expression and increased AC in rice grains. By contrast, the loss of function of OsEBP89 reduced Wx expression and decreased AC in rice grains. OsSK41 interacts with OsEBP89 and phosphorylates four of its sites (Thr‐28, Thr‐30, Ser‐238, and Thr‐257), which makes OsEBP89 unstable and attenuates its interaction with OsBP5. Wx promoter activity was relatively weak when regulated by the phosphomimic variant OsEBP89E–OsBP5 but relatively strong when regulated by the nonphosphorylatable variant OsEBP89A–OsBP5. Therefore, OsSK41‐mediated phosphorylation of OsEBP89 represents an additional layer of complexity in the regulation of amylose biosynthesis during rice grain development. Auxin is an important phytohormone in plants, and auxin signaling pathways in rice play key roles in regulating its growth, development, and productivity. To investigate how rice grain yield traits are regulated by auxin signaling pathways and to facilitate their application in rice improvement, we validated the functional relationships among regulatory genes such as OsIAA10, OsSK41, and OsARF21 that are involved in one of the auxin (OsIAA10) signaling pathways. We assessed the phenotypic effects of these genes on several grain yield traits across two environments using knockout and/or overexpression transgenic lines. Based on the results, we constructed a model that showed how grain yield traits were regulated by OsIAA10 and OsTIR1, OsAFB2, and OsSK41 and OsmiR393 in the OsSK41‐OsIAA10‐OsARF module and by OsARF21 in the transcriptional regulation of downstream auxin response genes in the OsSK41‐OsIAA10‐OsARF module. The population genomic analyses revealed rich genetic diversity and the presence of major functional alleles at most of these loci in rice populations. The strong differentiation of many major alleles between Xian/indica and Geng/japonica subspecies and/or among modern varieties and landraces suggested that they contributed to improved productivity during evolution and breeding. We identified several important aspects associated with the genetic and molecular bases of rice grain and yield traits that were regulated by auxin signaling pathways. We also suggested rice auxin response factor (OsARF) activators as candidate target genes for improving specific target traits by overexpression and/or editing subspecies‐specific alleles and by searching and pyramiding the 'best' gene allelic combinations at multiple regulatory genes in auxin signaling pathways in rice breeding programs. Xylanolytic enzymes, with both endo‐xylanase and arabinoxylan arabinofuranohydrolase (AXH) activities, are attractive for the economically feasible conversion of recalcitrant arabinoxylan. However, their characterization and utilization of these enzymes in biotechnological applications have been limited. Here, we characterize a novel bifunctional enzyme, rAbf43A, cloned from a bacterial consortium that exhibits AXH and endo‐xylanase activities. Hydrolytic pattern analyses revealed that the AXH activity belongs to AXHd3 because it attacked only the C(O)‐3‐linked arabinofuranosyl residues of double‐substituted xylopyranosyl units of arabinoxylan and arabinoxylan‐derived oligosaccharides, which are usually resistant to hydrolysis. The enzyme rAbf43A also liberated a series of xylo‐oligosaccharides (XOSs) from beechwood xylan, xylohexaose and xylopentaose, indicating that rAbf43A exhibited endo‐xylanase activity. Homology modelling based on AlphaFold2 and site‐directed mutagenesis identified three non‐catalytic residues (H161, A270 and L505) located in the substrate‐binding pocket essential for its dual‐functionality, while the mutation of A117 located in the −1 subsite to the proline residue only affected its endo‐xylanase activity. Additionally, rAbf43A showed significant synergistic action with the bifunctional xylanase/feruloyl esterase rXyn10A/Fae1A from the same bacterial consortium on insoluble wheat arabinoxylan and de‐starched wheat bran degradation. When rXyn10A/Fae1A was added to the rAbf43A pre‐hydrolyzed reactions, the amount of released reducing sugars, xylose and ferulic acid increased by 9.43% and 25.16%, 189.37% and 93.54%, 31.39% and 32.30%, respectively, in comparison with the sum of hydrolysis products released by each enzyme alone. The unique characteristics of rAbf43A position it as a promising candidate not only for designing high‐performance enzyme cocktails but also for investigating the structure–function relationship of GH43 multifunctional enzymes. • A compact acoustic wave amplifier based on non-adiabatic compression of sound in metamaterial waveguides is proposed. • It is found that the combined effects of structural discontinuity and thermal viscous dissipations in the metamaterial waveguides is the key to reduce the size of metamaterial device and optimize its sound amplification capability. • The idea of achieving miniaturized metamaterial devices based on non-adiabatic fast wave compression and amplification provides a novel technique to develop acoustic wave amplifiers, which could be practically used in many applications. Inspired by the optical non-adiabatic wave propagation effect in plasmonic waveguides, we explore the acoustic non-adiabatic wave compression effect in acoustic metamaterial waveguides, and demonstrate its potential for the development of compact metamaterial devices with significant sound amplification capability. In particular, our numerical and experimental results have shown that there exists combined effects of structure discontinuity and thermal viscous dissipations in the metamaterial waveguides, which is essential to reduce the size of metamaterial device and optimize its field enhancement capability. Background: After centuries of heavy reliance on fossil fuel energy, the world suffers from an energy crisis and global warming, calling for carbon emission reduction and a transition to clean energy. Microalgae have attracted much attention as a potential feedstock for biofuel production due to their high triacylglycerol content and CO2 sequestration ability. Many diacylglycerol acyltransferases (DGAT) species have been characterized, which catalyze the final committed step in triacylglycerol biosynthesis. However, the detailed structure–function features of DGATs and the role of the interactions among DGAT proteins in lipid metabolism remained largely unknown. Results: In this study, the three characterized DGATs of Auxenochlorella protothecoides 2341 showed distinct structural and functional conservation. Functional complementation analyses showed that ApDGAT1 had higher activity than ApDGAT2b in yeast and model microalgae, and ApDGAT2a had no activity in yeast. The N-terminus was not essential to the catalysis function of ApDGAT1 but was crucial to ApDGAT2b as its enzyme activity was sensitive to any N-terminus modifications. Similarly, when acyl-CoA binding proteins (ACBPs) were fused to the N-terminus of ApDGAT1 and ApDGAT2b, zero and significant activity changes were observed, respectively. Interestingly, the ApACBP3 + ApDGAT1 variant contributed to higher oil accumulation than the original DGAT1, and ApACBP1 + ApDGAT1 fusion boosted oleic acid content in yeast. Overexpression of the three DGATs and the variation of ApACBP3 + ApDGAT1 increased the content of C18:1 of Chlamydomonas reinhardtii CC-5235. Significantly, ApDGAT1 interacted with itself, ApDGAT2b, and ApACBP1, which indicated that these three lipid metabolic proteins might have been a part of a dynamic protein interactome that facilitated the enrichment of oleic acid. Conclusions: This study provided new insights into the functional and structural characteristics of DGATs and elucidated the importance of these physical interactions in potential lipid channeling. Optimization of the composition of enzyme cocktail is an effective way to improve the hydrolytic efficiency and reduce the cost during enzymatic hydrolysis of lignocellulose. Herein, the optimal enzymes ratios of enzyme cocktails for efficient hydrolysis of corncobs with distinct pretreatments were quantitatively formulated. As the importance of pretreatment type on xylan extraction, the optimized cellulase/hemicellulase ratios for NaClO 2 , H 2 SO 4 and steam explosion treated corncob were 1:1.5, 1:0.15 and 1:0.75, respectively. Meanwhile, different requirements for types of cellulases are also determined: endoglucanase and cellobiohydrolase were key enzymes in the hydrolysis of NaClO 2 and H 2 SO 4 treated corncobs, respectively, while the synergy of three kinds of cellulases was preferred for steam-exploded corncob. Furthermore, based on the optimized cellulase/hemicellulase ratios, cellulases proportions for hydrolyzing the corncobs pretreated with distinctive methods were determined. Notably, by the enzymatic formulation strategy, 100% yields of glucose and xylose were obtained simultaneously from NaClO 2 treated corncob, which was up to 70.1% and 80.1% at high solid loading (20%), respectively. Without detoxification of hydrolysate, high isopropanol yield of 0.45 g/g (of dry matter) was achieved. Polyvinyl chloride (PVC) foam sandwich panels have a wide application prospect in the field of warship impact protection due to the low moisture absorption, low density, excellent energy consumption and chemical resistance of PVC foam. The dynamic response of layered density-graded PVC foam sandwich panel under impact load was experimentally and numerically investigated here. A three-layer PVC foam sandwich specimen was designed and fabricated. The graded strategy of foam core was achieved by setting various foam layers with different densities. A large-diameter split-Hopkinson pressure bar device was used to perform the experimental study. The effect of density-graded strategy of foam core on the transmitted force, transmitted impulse and energy absorption were discussed. The software ANSYS/LS-DYNA was used to simulate the dynamic impact test of the foam sandwich structure. The simulation results are in good agreement with the experimental results. The results reveal that, the order of the core layer of density-graded sandwich panel has little effect on the impact resistance of the structure, while the impact resistance can be improved by increasing the density difference between foam layers. The uniform configuration UD has the largest impact force and the most specific energy absorption, while the large gradient configuration large gradient configuration has the smallest transmitted impulse under various impact velocities of the projectile. Magnetorheological elastomer (MRE) is a novel intelligent material, which shows excellent potential in vibration control applications. Previous researches have fully demonstrated that the magneto-induced shear storage modulus of MRE largely determines the vibration control effect. However, both existing theoretical and experimental ways to measure the magneto-induced shear storage modulus of MRE face their own shortage. Therefore, a novel physics-guided deep learning model is proposed to efficient predict the magneto-induced mechanical properties of MRE based on Magnetic Dipole theory and data-driven methods. A small database is built by collecting the magneto-induced shear storage modulus of MRE with different material ratios tested on a special shear rheometer. The proposed model trained with small training samples and its prediction results fit well with experimental values (average R2 of 0.99) which is superior to existing constitutive models. The training only takes 25 s, which significantly shortens the time compared to the experiment. Furthermore, the proposed model effectively predicts the magneto-induced storage modulus of MRE and has good generalization and superior transfer performance. The pulse force generated by the propeller excites the hull through the propulsion shafting system, which is the primary source of vibration and noise of ships. In order to control the transmission of longitudinal vibration of ship shafting, a multimodal dynamic vibration absorber (MDVA) with broadband vibration control performance is proposed. A magnetorheological elastomer (MRE) was prepared and used as an elastic element to construct a semi-active dynamic vibration absorber (SDVA), designed spiral flexible springs and embedded SDVA into the proposed MDVA. An equivalent propeller propulsion shaft system model was established, and its first three longitudinal vibrations were controlled based on the MDVA. Furthermore, the stiffness and damping of the MDVA were optimized by fixed point theory. A design scheme for the MDVA considering the changes in oil film stiffness of thrust bearings under different propeller speed conditions was proposed. The results indicate that the MDVA has excellent vibration transmission control performance at the first three natural frequencies. The proposed MDVA also has a broader frequency band for controlling vibration near first-order resonance than the traditional single degree of freedom DVA, which has excellent vibration control performance under variable speed conditions. It is urgent to explore new ways to protect endangered wild animals and develop sustainable animal husbandry on the Qinghai–Tibet Plateau due to its fragile ecological environment. Ruminants, raised in captivity and free-range, have important niches in the Plateau and are the best models to analyze the effects of different feeding modes on their health. In this study, two ruminants, yaks and goats in free-range and captive modes, respectively, were selected to study the relationship between gut microbes and ruminant health. The results showed that the gut microbial diversity of free-range ruminants was higher than those of captive ruminants. Principal co-ordinates analysis (PCoA) showed that there were significant differences in the gut microbial communities in different breeding modes. Both the captive ruminants enriched the Succinivibrionaceae family, which had a strong potential to synthesize lipopolysaccharide, and the low exercise amount of the captive animals was significantly related to this function. Meanwhile, free-range ruminants enriched Oscillospiraceae, which had the potential to degrade benzoic acid, and this potential had a significant positive correlation with resistance to parasitic infections. A new type of composite armor, which is composed of the front steel plate, medium dual-layer piecewise ceramic tiles separated by steel grid and the Kevlar back plate, was designed based on the bio-inspiration. The failure mechanism, energy absorption, and the effect of the geometric parameters on ballistic response of composite armors were analyzed. The results show that the damage area of ceramic layer can be remarkably decreased by replacing a monolithic ceramic plate by the piecewise ceramic tiles, and the ballistic properties of armors can be enhanced by using multiple ceramic tiles separated by metal grids in composite armors. Summary: Gamma‐aminobutyric acid (GABA) and delta‐aminolevulinic acid (ALA), playing important roles in agriculture, medicine and other fields, are multifunctional non‐protein amino acids with similar and comparable properties and biosynthesis pathways. Recently, microbial synthesis has become an inevitable trend to produce GABA and ALA due to its green and sustainable characteristics. In addition, the development of metabolic engineering and synthetic biology has continuously accelerated and increased the GABA and ALA yield in microorganisms. Here, focusing on the current trends in metabolic engineering strategies for microbial synthesis of GABA and ALA, we analysed and compared the efficiency of various metabolic strategies in detail. Under ultra-high cadmium (Cd) stress, large amounts of glutathione are produced in Auxenochlorella protothecoides UTEX 2341, and the lipid content increases significantly. Glutathione is the best reductant that can effectively remove Cd, but the relationship between lipid accumulation and the cellular response to Cd stress has not been ascertained. Integrating analyses of the transcriptomes and lipidomes, the mechanism of lipid accumulation to Cd tolerance were studied from the perspectives of metabolism, transcriptional regulation and protein glutathionylation. Under Cd stress, basic metabolic pathways, such as purine metabolism, translation and pre-mRNA splicing process, were inhibited, while the lipid accumulation pathway was significantly activated. Further analysis revealed that the transcription factors (TFs) and genes related to lipid accumulation were also activated. Analysis of the TF interaction sites showed that ABI5, MYB_rel and NF-YB could further regulate the expression of diacylglycerol acyltransferase through glutathionylation/deglutathionylation, which led to increase of the triacylglycerol (TAG) content. Lipidomes analysis showed that TAG could help maintain lipid homeostasis by adjusting its saturation/unsaturation levels. This study for the first time indicated that glutathione could activate TAG synthesis in microalga A. protothecoides , leading to TAG accumulation and glutathione accumulation under Cd stress. Therefore, the accumulation of TAG and glutathione can confer resistance to high Cd stress. This study provided insights into a new operation mode of TAG accumulation under heavy metal stress. Heading date is an important agronomic trait in rice (Oryza sativa L.). We previously fine‐mapped two complementary genes, Late Heading Date 1 and Late Heading Date 2 (LH2), that control the late heading trait in rice. Here, we cloned and analyzed the function of LH2. LH2 was fine‐mapped to a 53‐kb genomic region. Sequencing analysis revealed that there were differences in the coding sequence of LOC_Os08g07740 between the varieties 'Bo B' and 'Yuefeng B'. Therefore, LOC_Os08g07740 was identified as a candidate gene for LH2 and subsequently determined that LH2 is a Type 3 allele of Days To Heading 8. Transgenic plants of Yuefeng B carrying LH2 from Bo B significantly delayed the heading date under short‐day and long‐day conditions. A yeast two‐hybrid analysis revealed that LH2 physically interacted with Heme Activator Protein (HAP) 5A, ‐H, ‐I, ‐L, ‐K, and HAP5‐LIKE. The HAP5 domain of HAP5 family members has various functions. Further analysis revealed that LH2 represses photoperiodic flowering by controlling the expression of Early Heading Date 1, Heading date 3a, and Rice Flowering Locus T1 in rice. Background: Both APETALA2/Ethylene Responsive Factor (AP2/ERF) superfamily and R2R3-MYB family were from one of the largest diverse families of transcription factors (TFs) in plants, and played important roles in plant development and responses to various stresses. However, no systematic analysis of these TFs had been conducted in the green algae A. protothecoides heretofore. Temperature was a critical factor affecting growth and lipid metabolism of A. protothecoides. It also remained largely unknown whether these TFs would respond to temperature stress and be involved in controlling lipid metabolism process. Results: Hereby, a total of six AP2 TFs, six ERF TFs and six R2R3-MYB TFs were identified and their expression profiles were also analyzed under low-temperature (LT) and high-temperature (HT) stresses. Meanwhile, differential adjustments of lipid pathways were triggered, with enhanced triacylglycerol accumulation. A co-expression network was built between these 18 TFs and 32 lipid-metabolism-related genes, suggesting intrinsic associations between TFs and the regulatory mechanism of lipid metabolism. Conclusions: This study represented an important first step towards identifying functions and roles of AP2 superfamily and R2R3-MYB family in lipid adjustments and response to temperature stress. These findings would facilitate the biotechnological development in microalgae-based biofuel production and the better understanding of photosynthetic organisms' adaptive mechanism to temperature stress. Leaf‐color mutants have been extensively studied in rice (Oryza sativa L.) and many corresponding genes have been identified and cloned. However, the mechanism of complex leaf‐color mutations requires further study. In this study, we obtained an introgression line (TIL22) from a set of introgression lines raised using African cultivated rice (O. glaberrima Steud.) as the donor parent and O. sativa subsp. indica Teqing as the recipient. Compared with Teqing, TIL22 showed a yellow‐green leaf phenotype at the seedling stage, but the leaves gradually changed to green after the five‐leaf stage. The photosynthetic pigment contents of yellow‐green leaves at the seedling stage were significantly reduced and chloroplast development was retarded compared with those of Teqing. Genetic analysis indicated that the introgression line phenotype was controlled by a single nuclear gene, temporarily designated YGL22. Map‐based cloning and sequence analysis suggested that the candidate gene was likely to be LOC_Os01g15390, which encodes a chloroplast protein. Real‐time quantitative polymerase chain reaction (qPCR) analysis revealed that the YGL22 transcript level was significantly lower in TIL22 than that of Teqing at the seedling stage. We generated the ygl22 knockout mutant using the CRISPR/Cas9 system. The ygl22 mutants showed a similar phenotype to that of TIL22 at the seedling stage, suggesting that YGL22 may be involved in the early development of chloroplasts in rice. In addition, plant height, number of panicles per plant, and grain yield per plant of ygl22 were not significantly affected. This research focuses on the dynamic response of sandwich panels with multilayered graded hourglass lattice core subjected to blast loading. A three-layer lattice core configuration is proposed to improve the absorption efficient of kinetic energy resulted from blast shock wave. The relative density of each core layer is changed with sectional dimension of core truss members to regulate the energy absorption of each core layer. Three-dimensional numerical simulation analyses of dynamic response are carried out, and the applied impulsive pressure distribution on the surface of the panels is calculated using the CONWEP code. The panels are made of stainless steel AL6XN, which is assumed to follow bilinear strain hardening and strain rate-dependence. Peak back sheet deflection and energy absorption of core layers for four types of hourglass lattice panels are comparatively analyzed and the effects of load intensity on the peak deflection are discussed. The modification of tRNA is important for accurate, efficient protein translation. A number of tRNA-modifying enzymes were found to influence various developmental processes in distinct organisms. However, few genetic or molecular studies have focused on genes encoding tRNA-modifying enzymes in green plant organelles. Here, we discovered that PDD OL , a natural variation allele of PLEIOTROPIC DEVELOPMENTAL DEFECTS (PDD), leads to pleiotropic developmental defects in a near-isogenic line (NIL) generated by introgressing the wild rice Oryza longistaminata into the rice (Oryza sativa) cv 187R. Map-based cloning revealed that PDD encodes an evolutionarily conserved tRNA-modifying GTPase belonging to the tRNA modification E family. The function of PDD was further confirmed by genetic complementation experiments and mutant analysis. PDD mRNA is primarily expressed in leaves, and PDD is localized to chloroplasts. Biochemical analyses indicated that PDD187R forms homodimers and has strong GTPase activity, whereas PDDOL fails to form homodimers and has weak GTPase activity. Liquid chromatography–coupled tandem quadrupole mass spectrometry revealed that PDD is associated with the 5-methylaminomethyl-2-thiouridine modification of chloroplast tRNA. Furthermore, compared to 187R, NIL- PDD OL has severely reduced levels of proteins involved in photosynthesis and ribosome biogenesis but increased levels of plastid-encoded RNA polymerase subunits. Finally, we demonstrate that the defect due to PDD OL alters chloroplast gene expression, thereby affecting communication between the chloroplast and the nucleus. [ABSTRACT FROM AUTHOR]

Raffinose family oligosaccharides (RFOs) negatively affect nutritional value of legume-derived food and feed. It has been challenging to develop a high performance α-galactosidase excelled on catalytic efficiency, thermostability, pH stability and protease-resistance that could efficiently hydrolyze RFOs. In this study, the first GH family 27 α-galactosidase gene from Irpex lacteus was cloned. The gene had an open reading frame of 1314 bp interrupted by 12 introns. The recombinant α-galactosidase expressed in Pichia pastoris (rILgalA) had an apparent molecular mass of 64 kDa and was highly N-glycosylated. rILgalA was maximally active at pH 4.8 and 70 °C. It was stable over a broad pH range of 3–11, retained 90% of its activity after incubation at 60 °C for 10 h and exhibited strong resistance to digestive proteases. Unlike many other α-galactosidases, rILgalA was hyperactive on RFOs. Its specific activities toward melibiose, raffinose and stachyose were 644, 755 and 833 U mg−1, respectively. The corresponding K cat / K m values were 120, 130 and 180 mM−1 s−1, which were the highest among reported α-galactosidases. rILgalA almost completely hydrolyzed raffinose and stachyose in soymilk at 60 °C in 30 min. These superior properties would make rILgalA an ideal remover of RFOs in food and feed industries. Heading date is one of the most important traits in rice and regulated by multiple genes. Common wild rice is the ancestor of Asian cultivated rice and harbours abundant genetic diversity. To use wild rice resource in rice breeding, a set of 154 introgression lines (ILs) covering 93% of the genome of Jinghong common wild rice was constructed in the background 'Teqing', using 208 simple sequence repeat markers evenly distributed on 12 chromosomes. Among the ILs, the line JIL64 displayed late heading independent of photoperiod. Genetic analysis using the two F2 populations crossed ''Teqing'/JIL64 and JIL64/'Teqing' revealed that late flowering was controlled by a recessive gene on chromosome 8 (designated early heading date 8, ehd8), and ehd8 was fine mapped to the 50‐kb region flanked by markers RM22221 and 64Indel4. Sequencing and qRT‐PCR demonstrated that LOC_Os08g01410 and LOC_Os08g01420 were deleted in JIL64 and may be associated with the late heading of Jinghong common wild rice. Objectives. Ankylosing spondylitis (AS) is a chronic disease that decreases mobility, function, and quality of life. This study introduced the "Smart-phone SpondyloArthritis Management System" (SpAMS), an interactive mobile health (mHealth) tool designed for AS/spondyloarthritis (SpA) disease management and used SpAMS data to evaluate clinical characteristics of Chinese patients with AS. Methods. SpAMS integrates patient's and physician's portals in a smart phone application. The Chinese Ankylosing Spondylitis Prospective Imaging Cohort was launched using SpAMS in April 2016. Patient self-assessments were completed online at baseline and at every subsequent clinic visit. Physician-reported assessments and treatments were recorded by rheumatologists during each visit. Results. In total, 1201 patients with AS [mean (SD) age, 30.6 (8.7) years; male, 82.6%] were recruited. Mean (SD) disease duration was 8.4 (6.1) years. Past or current symptoms of acute anterior uveitis (AAU), psoriasis, and inflammatory bowel disease (IBD) were observed in 21.0%, 3.7%, and 9.4% of patients, respectively. AAU and IBD occurred significantly more in patients with symptom duration > 10 years. The most commonly used medications at baseline were nonsteroidal anti-inflammatory drugs (98.2%). Patients using tumour necrosis factor inhibitors accounted for 20.8%, and 66.4% of patients used conventional synthetic disease-modifying antirheumatic drugs. At baseline, 57.2% of patients had inactive disease (ID)/low disease activity (LDA); this rate significantly improved to 79.2% after a mean follow-up of 13.3 (5.9) months. Compared with relapsed patients, new achievers of ID/LDA underwent more online patient assessments (P < .001). Problems solved in SpAMS caused 29.1% of clinic visits to a tertiary hospital unnecessary. SpAMS saved an average of 5.3 hours and 327.4 RMB per person on traffic expenses; these expenses equalled 16% of the Chinese monthly disposable personal income. Conclusions. In recent years, MRI has been regarded as a major diagnostic tool for spondyloarthritis (SpA), and anti-TNF therapy has been widely confirmed as an effective treatment strategy. This study was designed to investigate the correlation between the secreted protein dickkopf-1 (Dkk-1) and abnormal findings on magnetic resonance imaging (MRI) through a prospective study of 30 cases of SpA. Thirty patients with active SpA were included, all treated with recombinant human tumor necrosis factor (TNF) receptor-antibody fusion protein (YiSaiPu) injection at 50 mg/week for 6 months. All patients were also examined for their clinical, serological, and imaging manifestations of the condition before and after treatment. In patients receiving TNF inhibitor treatment, the erythrocyte sedimentation rate (ESR), C-reactive protein (CRP), and clinical activity indices BASDAI, BASFI, BASMI, ASDAS-CRP were significantly decreased (p < 0.01). Serum Dkk-1 concentration was also significantly decreased (p < 0.05), as were the scores of bone marrow edema of the sacroiliac joints and the spine (p < 0.05). The score of sacroiliac joint backfill was significantly increased (p < 0.05), and the baseline and changes in the serum Dkk-1 concentration were significantly correlated with the baseline and changes in spinal bone marrow edema levels. Inhibition of the level of serum Dkk-1 by TNF inhibitors may be the molecular basis for inhibiting the formation of new bone in SpA patients. As results, ultrafine grinding or steam explosion pretreated substrates can induce T. harzianum EM0925 to secret holo lignocellulolytic enzymes; acid treated substrate can induce cellobiohydrolase; while alkali or sodium chlorite treated substrates can induce β-xylosidase specifically. Furthermore, the combination of enzyme cocktails with different hydrolysis characteristics can further improve the hydrolysis efficiency, since 100% yields of glucose and xylose were obtained simultaneously from ultrafine grinding treated corn stover at low enzyme dosage (1.2 mg proteins/g substrate). Grain size and shape are important determinants of grain weight and yield in rice. Here, we report a new major quantitative trait locus (QTL), q TGW3 , that controls grain size and weight in rice. This locus, q TGW3 , encodes OsSK41 (also known as OsGSK5), a member of the GLYCOGEN SYNTHASE KINASE 3/SHAGGY-like family. Rice near-isogenic lines carrying the loss-of-function allele of Os SK41 have increased grain length and weight. We demonstrate that OsSK41 interacts with and phosphorylates AUXIN RESPONSE FACTOR 4 (OsARF4). Co-expression of Os SK41 with Os ARF4 increases the accumulation of OsARF4 in rice protoplasts. Loss of function of Os ARF4 results in larger rice grains. RNA-sequencing analysis suggests that Os ARF4 and Os SK41 repress the expression of a common set of downstream genes, including some auxin-responsive genes, during rice grain development. The loss-of-function form of Os SK41 at q TGW3 represents a rare allele that has not been extensively utilized in rice breeding. Suppression of Os SK41 function by either targeted gene editing or QTL pyramiding enhances rice grain size and weight. Thus, our study reveals the important role of OsSK41 in rice grain development and provides new candidate genes for genetic improvement of grain yield in rice and perhaps in other cereal crops. Airborne bacteria were measured when a dust storm passed Beijing in spring 2012 with a focus on cell concentration, viability and TSA- and R2A-cultured strain composition. The concentration varied at an order of 10 7 cells/m 3 with dust loading (demonstrated with PM 10 ) and they had a very close correlation ( R T 2 = 0.91, p < 0.01). At the time of highest PM 10 of 652 μg/m 3 , the bacterial concentration reached 1.4 × 10 8 cells/m 3 , which was larger than that before and after the dust event by one order. Bacterial viability, the ratio of number concentration of viable cells to total cells, was 32%–64% and smaller in the dust plume than that before the dust arrival. Bacterial strains from the culture ranged between 2.5 × 10 4 and 4.6 × 10 5 CFU/m 3 and no correlation with PM 10 was determined. Their composition was different before and after the dust arrival according to 16S rRNA gene sequences and strains belong to Actinomycetes and Firmicutes were the majority in the dust samples. To maintain outstanding thermal stability, amino- and hydroxyl-containing phthalonitrile monomers, 4-(4-aminophenoxy)-phthalonitrile (APN) and 4-(4-hydroxyphenoxy)-phthalonitrile (HPN) were selected and synthesized. Their structures were confirmed by proton nuclear magnetic resonance spectroscopy. Their curing polymers were characterized by Fourier transform infrared spectroscopy. The self-catalytic curing behaviors of the monomers were investigated by differential scanning calorimetry (DSC) at different heating rates. From the results, APN exhibits a higher curing temperature, while HPN exhibits a longer curing time. Then, mixtures of these monomers were investigated by DSC. The result shows that the 50/50 mixture exhibits different autocatalytic behaviors: the curing temperature is lower than that of APN and the curing time of the mixture is shorter than that of HPN. Furthermore, thermogravimetric analysis shows that the polymer from the mixture exhibits higher temperature of 5% weight loss (T5%) and char yield value at 800°C than those of the polymers from each monomer. All these results indicate that the new mixture resin exhibits improved processability with excellent thermal stability, attributed to the synergistic effect between similar monomers; the synergistic effect optimizes the cure reaction kinetics and promotes cross-linking reactions, thereby producing an excellent resin; this approach is a new method for improving the processability without sacrificing thermal stability. Temperature is a crucial factor affecting microalgae CO 2 capture and utilization. However, an in-depth understanding of how microalgae respond to temperature stress is still unclear. In particular, the regulation mechanism under opposite temperature (heat and cold) stress had not yet been reported. In this study, the physicochemical properties and transcription level of related genes of microalgae Auxenochlorella protothecoides UTEX 2341 under heat and cold stress were investigated. Heat stress (Hs) caused a drastic increase of reactive oxygen species (ROS) in UTEX 2341. As key elements responded to Hs, superoxide dismutase (SOD) enzyme increased by 150%, 70%, and 30% in activity, and nitric oxide (NO) grew by 409.6%, 212.5%, and 990.4% in content compared with the control at 48 h, 96 h, 168 h. Under cold stress (Cs), ROS increased in the early stage and decreased in the later stage. As key factors responded to Cs, proline (Pro) increased respectively by 285%, 383%, and 81% in content, and heat shock transcriptional factor HSFA1d increased respectively by 161%, 71%, and 204% in transcript level compared with the control at 48 h, 96 h, 168 h. Furthermore, the transcript level of antioxidant enzymes or antioxidant coding genes was consistent with the changing trend of enzymes activity or antioxidant content. Notably, both glutathione (GSH) and heat shock protein 97 (hsp 97) were up-regulated in response to Hs and Cs. In conclusion, GSH and hsp 97 were the core elements of UTEX 2341 in response to both Hs and Cs. SOD and NO were the key elements that responded to Hs, while proline and HSFA1d were the key elements that responded to Cs. Paenibacillus sp. strain A2 is a Gram-negative rod-shaped bacterium isolated from a mixture of formation water and petroleum in Daqing oilfield, China. This facultative aerobic bacterium was found to have a broad capacity for metabolizing hydrocarbon and organosulfur compounds, which are the main reasons for the interest in sequencing its genome. Here we describe the features of Paenibacillus sp. strain A2, together with the genome sequence and its annotation. The 7,650,246 bp long genome (1 chromosome but no plasmid) exhibits a G+C content of 54.2 % and contains 7575 protein-coding and 49 RNA genes, including 3 rRNA genes. One putative alkane monooxygenase, one putative alkanesulfonate monooxygenase, one putative alkanesulfonate transporter and four putative sulfate transporters were found in the draft genome. Integrative sandwich panels with multifunction of load bearing, thermal protection, and heat insulation are manufactured through co-injection resin transfer molding process. These panels have a polymethacrylimide foam core for heat insulation and two composite layers made of carbon/epoxy for load bearing and carbon/phenolic for thermal protection. In co-injection resin transfer molding process, the epoxy resin and phenolic resin are synchronously injected and cured in order to make integrative structures and improve the properties of the interface between different layers. The influence of main factors in the process of co-injection is investigated: the vacuum assistance is indispensable to improve the efficiency of manufacture and guarantee the properties of the products, volume fraction of carbon fiber plays the main role in determining size accuracy of the product, adjusting injection pressure is a simple and efficient way to adjust the infusion time of resin, while injection temperature has smaller influence in the features of manufacture. The mechanical properties of sandwich panels manufactured through co-injection resin transfer molding process and multi-step process are tested and compared to validate the advantage of the co-injection resin transfer molding process. The test shows that the in-plane compressive strength, three-point flexural strength, and interface shear strength are all improved by the co-injection resin transfer molding process.
