Abstract

Metastasis and recurrence are the main challenges in cancer treatment. Among various therapeutic approaches, immunotherapy holds promise for preventing metastasis and recurrence. In this study, we evaluated the efficacy of treating primary cancer and blocking metastasis and recurrence with photo-immunotherapeutic nanoparticles, which were synthesized using two types of charged polysaccharides. Codium fragile polysaccharide (CFP), which exhibits immune-stimulating properties and carries a negative charge, was combined with positively charged chitosan to synthesize nanoparticles. Additionally, indocyanine green (ICG), a photosensitizer, was loaded inside these particles and was referred to as chitosan-CFP-ICG (CC-ICG). Murine colon cancer cells (CT-26) internalized CC-ICG, and subsequent 808-nanometer laser irradiation promoted apoptotic/necrotic cell death. Moreover, intratumoral injection of CC-ICG, with 808-nanometer laser irradiation eliminated CT-26 tumors in mice. Rechallenged lung metastases of CT-26 cancer were inhibited by dendritic cell activation-mediated cytotoxic T lymphocyte stimulation in mice cured by CC-ICG. These results demonstrated that CC-ICG is a natural tumor therapeutic with the potential to treat primary tumors and suppress metastasis and recurrence. [ABSTRACT FROM AUTHOR]

13. Anti-Inflammatory Effect of Fucoidan from Costaria costata Inhibited Lipopolysaccharide-Induced Inflammation in Mice.

Abstract

Seaweed extracts, especially fucoidan, are well known for their immune-modulating abilities. In this current study, we extracted fucoidan from Costaria costata, a seaweed commonly found in coastal Asia, and examined its anti-inflammatory effect. Fucoidan was extracted from dried C. costata (FCC) using an alcohol extraction method at an extraction rate of 4.5 ± 0.21%. The extracted FCC comprised the highest proportion of carbohydrates, along with sulfate and uronic acid. The immune regulatory effect of FCC was examined using bone marrow-derived dendritic cells (BMDCs). Pretreatment with FCC dose-dependently decreased the lipopolysaccharide (LPS)-induced upregulation of co-stimulatory molecules and major histocompatibility complex. In addition, FCC prevented morphological changes in LPS-induced BMDCs. Moreover, treatment of LPS-induced BMDCs with FCC suppressed the secretion of pro-inflammatory cytokines. In C57BL/6 mice, oral administration of FCC suppressed LPS-induced lung inflammation, reducing the secretion of pro-inflammatory cytokines in the bronchoalveolar lavage fluid. Finally, the administration of FCC suppressed LPS-induced sepsis. Therefore, FCC could be developed as a health supplement based on the observed anti-inflammatory effects. [ABSTRACT FROM AUTHOR]

Abstract

A new class of near‐infrared (NIR) fluorophores, PAI, is obtained by consecutive C−N/C−C bond formation between diphenylamines and 9,10‐dibromoperylenecarboximide. Owing to the rigid structure, extended π‐conjugation and pronounced push‐pull substitution, these fluorophores show emission maxima up to 804 nm and large Stokes shifts. The extraordinarily high fluorescence quantum yields from 47 % to 70 % are attributed to chloro substitution in the bay positions of the perylene core. These characteristics, together with high photostability, qualify them as useful NIR emitters for applications as biomarkers and security inks. [ABSTRACT FROM AUTHOR]

47. Immunomodulatory effects of polysaccharides from marine algae for treating cancer, infectious disease, and inflammation.

Abstract

A significant rise in the occurrence and severity of adverse reactions to several synthetic drugs has fueled considerable interest in natural product‐based therapeutics. In humans and animals, polysaccharides from marine microalgae and seaweeds have immunomodulatory effects. In addition, these polysaccharides may possess antiviral, anticancer, hypoglycemic, anticoagulant, and antioxidant properties. During inflammatory diseases, such as autoimmune diseases and sepsis, immunosuppressive molecules can serve as therapeutic agents. Similarly, molecules that participate in immune activation can induce immune responses against cancer and infectious diseases. We aim to discuss the chemical composition of the algal polysaccharides, namely alginate, fucoidan, ascophyllan, and porphyran. We also summarize their applications in the treatment of cancer, infectious disease, and inflammation. Recent applications of nanoparticles that are based on algal polysaccharides for the treatment of cancer and inflammatory diseases have also been addressed. In conclusion, these applications of marine algal polysaccharides could provide novel therapeutic alternatives for several diseases. [ABSTRACT FROM AUTHOR]

48. Escherichia coliMimetic Gold Nanorod-Mediated Photo- and Immunotherapy for Treating Cancer and Its Metastasis

Abstract

Most cancer-related deaths are due to metastasis or recurrence. Therefore, the ultimate goal of cancer therapy will be to treat metastatic and recurrent cancers. Combination therapy for cancer will be one of trial for effective treating metastasis and recurrence. In this study, Escherichia coli-mimetic nanomaterials are synthesized using Escherichia colimembrane proteins, adhesion proteins, and gold nanorods, which are named E. colimimetic AuNRs (ECA), for combination therapy against cancer and its recurrence. ECA treatment with 808 nm laser irradiation eliminates CT-26 or 4T1 tumors viaa photothermal effect. ECA with laser irradiation induces activation of immune cells in the tumor-draining lymph nodes. The mice cured from CT-26 or 4T1 tumor by ECA are rechallenged with those cancer in the lung metastatic form, and the results showed that ECA treatment for the first CT-26 or 4T1 tumor challenge prevents cancer infiltration to the lung in the second challenge. This preventive effect of ECA against tumor growth in the second challenge is aided by cancer antigen-specific T cell immunity. Overall, these findings show that ECA is a nanomaterial with dual functions as a photothermal therapy for treating primary cancers and as immunotherapy for preventing recurrence and metastasis.

Breast Cancer (BC) has the highest incidence among all forms of malignancies detectedin women globally. The therapeutic approaches available for BC include chemotherapy, radiationtherapy, hormonal therapy, and surgery. Recently, advanced immunology-based therapeutics withpotential for BC treatment, including immune checkpoint blockades, vaccines, and combinationswith other treatment strategies, have emerged. Although commonly used treatments such as trastuzumab/pertuzumab for human epidermal growth factor receptor 2-positive BC and hormone therapyfor estrogen receptor-positive and/or progesterone receptor-positive BC are specific, triple-negativeBC cases remain a great challenge for treatment measures. Immune checkpoint inhibitors (anti-PD-1/anti-CTLA-4) and anti-cancer vaccines (NeuVax, MUC-1, AVX901, INO-1400, andCEA), either alone or in combination with other therapies, represent a new paradigm in cancer therapeutics.In this review, we highlight the current immunotherapeutic aspects and ongoing trialsaimed at the development of better treatment regimens for BC.

Abstract

We elucidated the proper routes for injecting lipid-DNA micelles formulated with an oligonucleotide immune adjuvant in naïve and tumor-bearing mice. We report herein that after assessing six ways of administering an immunostimulatory nanoparticle (INP), several of the injection routes effectively induced dendritic cell-mediated immune-stimulation in vivo, making them amenable to cancer immunotherapies and vaccines using such DNA materials.
