Abstract

National audience; Developmental transitions, such as sexual maturation, demand considerable energy: this suggests diet might be an important external factor controling them. The male Agrotis ipsilon, goes through a sexual maturation between the third and the fifth day of adult life. Morphological and functional shifts in the reproductive system occur during this period. Moreover, moths obtain nutrients (water, amino acids and soluble carbohydrates like saccharose, fructose and glucose) within the nectar. Some of them practice puddling, a behavior allowing to sodium intake from excrements and puddles of brackish water. All of these nutrients are essentials for the accomplishment of the reproductive function in butterflies. In this study, we assessed the impact of diet on sexual maturation in the male moth Agrotis ipsilon.

28. Exposure to Conspecific and Heterospecific Sex-Pheromones Modulates Gustatory Habituation in the Moth Agrotis ipsilon

Abstract

In several insects, sex-pheromones are essential for reproduction and reproductive isolation. Pheromones generally elicit stereotyped behaviors. In moths, these are attraction to conspecific sex-pheromone sources and deterrence for heterospecific sex-pheromone. Contrasting with these innate behaviors, some results in social insects point toward effects of non-sex-pheromones on perception and learning. We report the effects of sex-pheromone pre-exposure on gustatory perception and habituation (a non-associative learning) in male Agrotis ipsilon moths, a non-social insect. We also studied the effect of Z5-decenyl acetate (Z5), a compound of the sex-pheromone of the related species Agrotis segetum. We hypothesized that conspecific sex-pheromone and Z5 would have opposite effects. Pre-exposure to either the conspecific sex-pheromone or Z5 lasted 15 min and was done either immediately or 24 h before the experiments, using their solvent alone (hexane) as control. In a sucrose responsiveness assay, pre-exposure to the conspecific sex-pheromone had no effect on the dose-response curve at either delays. By contrast, Z5 slightly improved sucrose responsiveness 15 min but not 24 h after pre-exposure. Interestingly, the conspecific sex-pheromone and Z5 had time-dependent effects on gustatory habituation: pre-exposing the moths with Z5 hindered learning after immediate but not 24-h pre-exposure, whereas pre-exposure to the conspecific sex-pheromone hindered learning at 24-h but not immediate pre-exposure. They did not have opposite effects. This is the first time a sex-pheromone is reported to affect learning in a non-social insect. The difference in modulation between conspecific sex-pheromone and Z5 suggests that con- and hetero-specific sex-pheromones act on plasticity through different cerebral pathways.

Abstract

Restrained honeybees can easily associate a sucrose reward to a metal plate. To study the role of nicotinic receptors in this learning, nicotinic antagonists (mecamylamine, MLA or α-bungarotoxin) or a NO-synthase inhibitor (L-NAME) were injected in haemolymph using doses leaving intact perception or motor functions. Mecamylamine impaired memory retrieval and single-trial learning without affecting long-term memory (LTM) formation that specifically occurs during multiple-trial learning. On the other hand, L-NAME, α- bungarotoxin and MLA only blocked LTM formation. This pharmacological dissociation indicates the existence of various functional types of nicotinic receptors. Those sensible to MLA and α-bungarotoxin could be coupled to NO-synthase., L'abeille en contention peut associer une récompense sucrée au scan antennaire d'une plaque métallique. Pour étudier le rôle des récepteurs nicotiniques dans cet apprentissage, des antagonistes nicotiniques (mécamylamine, MLA ou α-bungarotoxine) ou un inhibiteur de la NO-synthase (le L-NAME) ont été injectés dans l'hémolymphe à des doses sans effet sur la perception ou la motricité. La mécamylamine a bloqué le rappel de la trace mnésique et l'apprentissage en un essai sans affecter la formation de la mémoire à long terme (MLT) qui se produit spécifiquement durant l'apprentissage en plusieurs essais. Par contre, le L-NAME, l'α- bungarotoxine et le MLA ne bloquent que la formation de la MLT. Cette dissociation pharmacologique indique l'existence de plusieurs types fonctionnels de récepteurs nicotiniques. Ceux qui sont sensibles au MLA et à l'α-bungarotoxine seraient couplés à la NO-synthase.

Abstract

The postsynaptic scaffolding A-kinase anchoring protein 79/150 (AKAP79/150) signaling complex regulates excitatory synaptic transmission and strength through tethering protein kinase A (PKA), PKC, and calcineurin (CaN) to the postsynaptic densities of neurons (Sanderson and Dell'Acqua, 2011), but its role in inhibitory synaptictransmission and plasticity is unknown. Using immunofluorescence and whole-cell patch-clamp recording in rat midbrain slices, we show that activation of postsynaptic D2 -like family of dopamine (DA) receptor in the ventral tegmental area (VTA) induces long-term depression (LTD) of GABAergic synapses on DA neurons through an inositol triphosphate receptor-mediated local rise in postsynaptic Ca2 and CaN activation accompanied by PKA inhibition, which requires AKAP150 as a bridging signaling molecule. Our data also illuminate a requirement for a clathrin-mediated internalization of GABAA receptors in expression of LTDGABA. Moreover, disruption of AKAP-PKA anchoring does not affect glutamatergic synapses onto DA neurons, suggesting that the PKA-AKAP-CaN complex is uniquely situated at GABAA receptor synapses in VTA DA neurons to regulate plasticity associated with GABAA receptors. Drug-induced modulation of GABAergic plasticity in the VTA through such novel signaling mechanisms has the potential to persistently alter the output of individual DA neurons and of the VTA, which may contribute to the reinforcing or addictive properties of drugs of abuse.

Abstract

Age-related memory impairment (AMI) is a common feature and a debilitating phenotype of brain aging in many animals. However, the molecular mechanisms underlying AMI are still largely unknown. The cricket Gryllus bimaculatus is a useful experimental animal for studying age-related changes in learning and memory capability; because the cricket has relatively short life-cycle and a high capability of olfactory learning and memory. Moreover, the molecular mechanisms underlying memory formation in crickets have been examined in detail. In the present study, we trained male crickets of different ages by multiple-trial olfactory conditioning to determine whether AMI occurs in crickets. Crickets 3 weeks after the final molt (3-week-old crickets) exhibited levels of retention similar to those of 1-week-old crickets at 30 min or 2 h after training; however they showed significantly decreased levels of 1-day retention, indicating AMI in long-term memory (LTM) but not in anesthesia-resistant memory (ARM) in olfactory learning of crickets. Furthermore, 3-week-old crickets injected with a nitric oxide (NO) donor, a cyclic GMP (cGMP) analog or a cyclic AMP (cAMP) analog into the hemolymph before conditioning exhibited a normal level of LTM, the same level as that in 1-week-old crickets. The rescue effect by NO donor or cGMP analog injection was absent when the crickets were injected after the conditioning. For the first time, an NO donor and a cGMP analog were found to antagonize the age-related impairment of LTM formation, suggesting that deterioration of NO synthase (NOS) or molecules upstream of NOS activation is involved in brain-aging processes. Abstract

During an infection, animals suffer several changes in their normal physiology and behavior which may include lethargy, appetite loss, and reduction in grooming and general movements. This set of alterations is known as sickness behavior and although it has been extensively believed to be orchestrated primarily by the immune system, a relevant role for the central nervous system has also been established. The aim of the present work is to develop a simple animal model to allow studying how the immune and the nervous systems interact coordinately during an infection. We administered a bacterial lipopolysaccharide (LPS) into the thorax of honey bees to mimic a bacterial infection, and then we evaluated a set of stereotyped behaviors of the animals that might be indicative of sickness behavior. First, we show that this immune challenge reduces the locomotor activity of the animals in a narrow time window after LPS injection. Furthermore, bees exhibit a loss of appetite 60 and 90 min after injection, but not 15 h later. We also demonstrate that LPS injection reduces spontaneous antennal movements in harnessed animals, which suggests a reduction in the motivational state of the bees. Finally, we show that the LPS injection diminishes the interaction between animals, a crucial behavior in social insects. To our knowledge these results represent the first systematic description of sickness behavior in honey bees and provide important groundwork for the study of the interaction between the immune and the neural systems in an insect model. Insecticides have long been used as the main method in limiting agricultural pests, but their widespread use has resulted in environmental pollution, development of resistances, and biodiversity reduction. The effects of insecticides at low residual doses on both the targeted crop pest species and beneficial insects have become a major concern. In particular, these low doses can induce unexpected positive (hormetic) effects on pest insects, such as surges in population growth exceeding what would have been observed without pesticide application. Methomyl and chlorpyrifos are two insecticides commonly used to control the population levels of the cotton leafworm Spodoptera littoralis, a major pest moth. The aim of the present study was to examine the effects of sublethal doses of these two pesticides, known to present a residual activity and persistence in the environment, on the moth physiology. Using a metabolomic approach, we showed that sublethal doses of methomyl and chlorpyrifos have a systemic effect on the treated insects. We also demonstrated a behavioral disruption of S. littoralis larvae exposed to sublethal doses of methomyl, whereas no effects were observed for the same doses of chlorpyrifos. Interestingly, we highlighted that sublethal doses of both pesticides did not induce a change in acetylcholinesterase activity in head of exposed larvae. [ABSTRACT FROM AUTHOR]

Abstract

In many animals, drastic changes are observed during sexual maturation characterized by the reproductive system development concomitantly to the sexual behavior ontogenesis. These modifications are under the control of internal and external factors such as food. Sexual maturation requires considerable energetic investment, and diet has been shown to affect reproductive activities in many taxonomic groups, especially in insects and vertebrates. By contrast, diet effects on sexual behavior development remain largely unexplored. To elucidate this aspect, we used the male moth Agrotis ipsilon which undergoes sexual maturation occurring between the third and the fifth day postemergence. During this period, males are sensitive to female sex pheromones and a stereotypical sexual behavior characterized by female‐oriented flight takes place. In our study, we compared (1) sex pheromone detection by electroantennography recordings and (2) behavioral response in wind tunnel assays between males fed with different diets found in nature. Compared to standard sucrose diet, males fed with sucrose, fructose, and glucose supplemented with sodium (a mineral element necessary for the locomotor activity in several moths) did not respond better to female sex pheromones but clearly exhibited an earlier behavioral response. Thus, such a diet accelerates the development of sex pheromone‐mediated oriented flight, probably by facilitating the central processing of sex pheromone information in male A. ipsilon moths. Abstract

International audience; Invertebrate Physiology, a section of the journal Frontiers in Physiology Adult moths need energy and nutrients for reproducing and obtain them mainly by consuming flower nectar (a solution of sugars and other compounds). Gustatory perception gives them information on the plants they feed on. Feeding and food perception are integrated in the proboscis extension response, which occurs when their antennae touch a sugar solution. We took advantage of this reflex to explore moth sugar responsiveness depending on different parameters (i.e., sex, age, satiety, site of presentation, and composition of the solution). We observed that starvation but not age induced higher response rates to sucrose. Presentation of sucrose solutions in a randomized order confirmed that repeated sugar stimulations did not affect the response rate; however, animals were sometimes sensitized to water, indicating sucrose presentation might induce non-associative plasticity. Leg stimulation was much less efficient than antennal stimulation to elicit a response. Quinine prevented and terminated sucrose-elicited proboscis extension. Males but not females responded slightly more to sucrose than to fructose. Animals of either sex rarely reacted to glucose, but curiously, mixtures in which half sucrose or fructose were replaced by glucose elicited the same response rate than sucrose or fructose alone. Fructose synergized the response when mixed with sucrose in male but not female moths. This is consistent with the fact that nectars consumed by moths in nature are mixtures of these three sugars, which suggests an adaptation to nectar perception.

43. Insights on the nutritional ecology of a nocturnal pollinating insect.

Abstract

Nutritional ecology examines the environmental effects on nutritional needs, food intake and foraging behaviors, and the use of nutrients ingested by animals. Adults of many insects' species feed on nectars rich in sugars allowing them to match the nutritional needs necessary for reproduction. Among insects, Lepidoptera are often considered opportunistic foragers that visit a wide variety of available flowers, although with some preferences. While nutritional ecology of diurnal Lepidoptera is beginning to be explored, very little work focuses on nocturnal species because they are complicated to study in the wild. To address this, we used new laboratory approaches to study feeding behaviors (number and duration of visits to artificial flowers, food preferences associated with the texture and odors of the flowers) as well as gustatory detection by antennae (proboscis extension reflex) in the male crop pest moth Agrotis ipsilon. We showed that (i) food responsiveness is age-dependent and increases mainly with sugar quantity and marginally with sugar quality, (ii) diet quality impacts feeding behaviors in the first days of adulthood, and (iii) male moths choose their food through floral cues. Taken together, these data allow to define this species as a generalist forager with a preference for flowers with sugary nectars rich in sucrose, fructose, and glucose. Our results thus provide considerable information on the close links between food sources and nutritional ecology in this species, which is important for guiding future studies on their behavioral ecology, population dynamics, as well as for population monitoring and for regional pest management., (© 2025 The Author(s). As in other animals, diet is known to influence insect reproduction, and its impact has been intensively investigated in females. In our study, we examined the effects of various diets on male reproductive success in the moth Agrotis ipsilon, a pest of many crops. Our experiments showed an increase in the rates of fertilization and hatching when males fed with various sugars (sucrose, fructose, and glucose) supplemented with sodium. Abstract

The postsynaptic scaffolding A-kinase anchoring protein 79/150 (AKAP79/150) signaling complex regulates excitatory synaptic transmission and strength through tethering protein kinase A (PKA), PKC, and calcineurin (CaN) to the postsynaptic densities of neurons (Sanderson and Dell'Acqua, 2011), but its role in inhibitory synaptic transmission and plasticity is unknown. Using immunofluorescence and whole-cell patch-clamp recording in rat midbrain slices, we show that activation of postsynaptic D(2)-like family of dopamine (DA) receptor in the ventral tegmental area (VTA) induces long-term depression (LTD) of GABAergic synapses on DA neurons through an inositol triphosphate receptor-mediated local rise in postsynaptic Ca(2+) and CaN activation accompanied by PKA inhibition, which requires AKAP150 as a bridging signaling molecule. Our data also illuminate a requirement for a clathrin-mediated internalization of GABA(A) receptors in expression of LTD(GABA). Moreover, disruption of AKAP-PKA anchoring does not affect glutamatergic synapses onto DA neurons, suggesting that the PKA-AKAP-CaN complex is uniquely situated at GABA(A) receptor synapses in VTA DA neurons to regulate plasticity associated with GABA(A) receptors. Drug-induced modulation of GABAergic plasticity in the VTA through such novel signaling mechanisms has the potential to persistently alter the output of individual DA neurons and of the VTA, which may contribute to the reinforcing or addictive properties of drugs of abuse.
