Abstract

Knowing the cause of kidney stone formation is crucial to establish treatments that prevent recurrence. There are currently different approaches for determining the kidney stone type. However, the reference ex-vivo identification procedure can take up to several weeks, while an in-vivo visual recognition requires highly trained specialists. Machine learning models have been developed to provide urologists with an automated classification of kidney stones during an ureteroscopy; however, there is a general lack in terms of quality of the training data and methods. In this work, a two-step transfer learning approach is used to train the kidney stone classifier. The proposed approach transfers knowledge learned on a set of images of kidney stones acquired with a CCD camera (ex-vivo dataset) to a final model that classifies images from endoscopic images (ex-vivo dataset). The results show that learning features from different domains with similar information helps to improve the performance of a model that performs classification in real conditions (for instance, uncontrolled lighting conditions and blur). Finally, in comparison to models that are trained from scratch or by initializing ImageNet weights, the obtained results suggest that the two-step approach extracts features improving the identification of kidney stones in endoscopic images., Comment: This work has been submitted to the IEEE for possible publication. Abstract

Particle physics has an ambitious and broad experimental programme for the coming decades. This programme requires large investments in detector hardware, either to build new facilities and experiments, or to upgrade existing ones. Similarly, it requires commensurate investment in the R&D of software to acquire, manage, process, and analyse the shear amounts of data to be recorded. In planning for the HL-LHC in particular, it is critical that all of the collaborating stakeholders agree on the software goals and priorities, and that the efforts complement each other. In this spirit, this white paper describes the R&D activities required to prepare for this software upgrade.

19. Solid-State Fermentation-Assisted Extraction of Flavonoids from Grape Pomace Using Co-Cultures.

Abstract

Eighty percent of grape production is destined for the wine industry, which generates various types of waste, of which grape pomace is the main one, accounting for 50–60% of waste created during processing. This waste could be a promising source of bioactive compounds (e.g., flavonoids and tannin), which are known for their antioxidant properties. Although these byproducts pose disposal challenges, they can be utilized as a substrate for solid-state fermentation bioprocess using co-cultures, where different microorganisms can interact and complement each other, improving the efficiency of metabolite production or substrate degradation. This study investigates the extraction of phenolic compounds and the antioxidant activity of the compounds from grape pomace in the solid-state fermentation bioprocess, comparing fungal and yeast monocultures, and then exploring the use of two co-cultures (P. stipites/A. niger GH1 and S. cerevisiae/A. niger) on the flavonoid extractive process. Fermentation kinetics were evaluated over 120 h, with sampling done every 12 h. Initially, yeasts were used to reduce the content of simple sugars in the medium, and fungus was added at 24 h into the process due to its ability to produce a broad spectrum of extracellular enzymes, allowing a higher efficiency in substrate degradation. Competition or antagonism during co-culture leads to significantly higher production of compounds, which are recovered using different solvents. The evaluation included phenolic compounds (total polyphenols, condensed tannins, and total flavonoids), antioxidant activity (DPPH●/FRAP), molecular characterization (HPLC-MS), and structural microscopy during the bioprocess. The highest titers obtained were 62.46 g/L for total flavonoids and 32.04 g/L for condensed tannins, using acetone as the solvent in co-culture with P. stipitis after 120 h of fermentation. Characterization identified 38 compounds, highlighting families of flavonols, hydroxybenzoic acids, and hydroxycinnamic acids. The co-culture of P. stipitis and A. niger GH1 significantly improved the extraction yield of bioactive compounds through solid-state fermentation. [ABSTRACT FROM AUTHOR]

Abstract

A swarm of Pantala flavescens (Fabricius, 1798) observed in a section of the Barbate River in Cádiz is described. In addition, the reproduction of the species is confirmed in the La Breña and Marismas del Barbate Natural Park. These are the first records of migratory and reproductive behaviour of the species since its presence was first detected in the Iberian Peninsula (BERNAL-SÁNCHEZ & CONESA-GARCÍA, 2021). This study introduces a paradigm-shifting approach to optimize mitochondrial targeting. Employing a new fluorescent probe strategy, we unravel a combined influence of both Nernst potential (ψ) and partitioning (P) contributions. Through the synthesis of new benz[e]indolinium-derived probes, our findings redefine the landscape of mitochondrial localization by optimizing the efficacy of mitochondrial probe retention in primary cortical neurons undergoing normoxia and oxygen--glucose deprivation. This methodology not only advances our understanding of subcellular dynamics, but also holds promise for transformative applications in biomedical research and therapeutic development.
