43. Kinetics of Activation of Nicotinamide Adenine Dinucleotide Kinase by Phytochrome-Far Red-absorbing Form.

Abstract

The effects of exogenous redox cofactors and purine analogues on the activation of the NAD kinase by Pfr were examined. Addition of phenazine methosulfate, flavin mononucleotide, or methylene blue increased the activation of NAD kinase by red light in a partially purified preparation of phytochrome. Phenazine methosulfate and flavin mononucleotide do not absorb light in the red or far red region, so they do not act as light receptors in this activation. Thus they probably intervene in electron transfer between phytochrome and NAD kinase. Addition of kinetin with these compounds increased photopotentiation further. In the presence of phenazine methosulfate and kinetin, the activation of NAD kinase by red light was counteracted by illumination with far red light immediately after red light.The relation between the dose of illumination with red light and the degree of activation of NAD kinase was measured. Illumination for 5 minutes with 800 ergs cm(-2) sec(-1) of red light was sufficient to obtain maximal activation. In the dark, the activated state of NAD kinase was maintained for about 10 min and then rapidly lost. The time of preservation of the activated state was greatly shortened by illumination with far red light, indicating the participation of phytochrome.

Abstract

A dinucleoside monophosphate, 6,2'-anhydro-6-oxy-1-beta-D-arabinofuranosyluracil-phosphoryl- (3'-5')-6,2'-anhydro-6-oxy-1-beta-D-arabinofuranosyluracil (I) was synthesized by the condensation reaction using DCC from 5'-monomethoxytrityl derivative(VII) and 3'-acetyl-5'-phosphate(X) of the monomer units. Yield was ca. 70%. UV of compound I showed no hypochromicity and its CD spectrum showed only a trough at 252 nm. No temperature dependency of epsilon and [Q] values was observed. These results suggested an unstacked random conformation for I. Complex formation of I with cycloadenosine oligomer could not be detected under the experimental condition.

Abstract

Poly(formycin phosphate) and poly(laurusin phosphate) were synthesized by polymerizing formycin and laurusin 5'-diphosphate by means of E. coli polynucleotide phosphorylase. The complex formation of these polynucleotides with cyclonucleoside polynucleotides were investigated. While poly(formycin phosphate) did not form the complex with an octanucleotide of 6,2'-anhydro-6-oxy-1-beta-D-arabinofuranosyluracil, poly(laurusin phosphate) did form a 1: 1 complex with octanucleotide of 8,2'-anhydro-8-mercapto-9-beta-D-arabinofuranosyladenine in the presence of 0.15M Na ion at neutrality and 3(o). CD spectrum of this complex showed a couple of a trough at 286 nm and a peak at 262 nm. This fact suggests that the complex has a left-handed helical conformation, which is opposite to the natural double helical polynucleotides. The cause of this phenomenon was discussed in connection with the complex of cyclonucleoside oligonucleotides.
