Extracellular vesicles (EVs) secreted by human brain cells have great potential as cell‐free therapies in various diseases, including stroke. However, because of the significant amount of EVs needed in preclinical and clinical trials, EV application is still challenging. Vertical‐Wheel Bioreactors (VWBRs) have designed features that allow for scaling up the generation of human forebrain spheroid EVs under low shear stress. In this study, EV secretion by human forebrain spheroids derived from induced pluripotent stem cells as 3D aggregates and on Synthemax II microcarriers in VWBRs were investigated with static aggregate culture as a control. The spheroids were characterized by metabolite and transcriptome analysis. The isolated EVs were characterized by nanoparticle tracking analysis, electron microscopy, and Western blot. The EV cargo was analyzed using proteomics and miRNA sequencing. The in vitro functional assays of an oxygen and glucose‐deprived stroke model were conducted. Proof of concept in vivo study was performed, too. Human forebrain spheroid differentiated on microcarriers showed a higher growth rate than 3D aggregates. Microcarrier culture had lower glucose consumption per million cells and lower glycolysis gene expression but higher EV biogenesis genes. EVs from the three culture conditions showed no differences in size, but the yields from high to low were microcarrier cultures, dynamic aggregates, and static aggregates. The cargo is enriched with proteins (proteomics) and miRNAs (miRNA‐seq), promoting axon guidance, reducing apoptosis, scavenging reactive oxygen species, and regulating immune responses. Human forebrain spheroid EVs demonstrated the ability to improve recovery in an in vitro stroke model and in vivo. Human forebrain spheroid differentiation in VWBR significantly increased the EV yields (up to 240–750 fold) and EV biogenesis compared to static differentiation due to the dynamic microenvironment and metabolism change. The biomanufactured EVs from VWBRs have exosomal characteristics and more therapeutic cargo and are functional in in vitro assays, which paves the way for future in vivo stroke studies. High‐resolution seismic model is crucial for advancing our understandings on geological processes and enhancing seismic hazard mitigation programs. We construct a high‐resolution China Seismological Reference Model (CSRM‐1.0) in the top 100 km of the crust and uppermost mantle in continental China following a top‐down construction process. The employed seismic constraints include P‐wave polarization angle from tele‐seismic event, short‐period Rayleigh wave ellipticity from ambient noise, long‐period Rayleigh wave ellipticity from earthquake data, receiver function, empirical Green's function from ambient noise, Rayleigh wave phase/group velocity dispersion curves from regional earthquakes, and Pn‐wave travel time extracted from seismic data of 4,435 stations. CSRM‐1.0 has a spatial crustal resolution of ∼60 km beneath the north‐south seismic belt and trans‐North China orogen regions and ∼120 km beneath the rest of continental China, and a spatial mantle resolution of ∼300 km. CSRM‐1.0 exhibits prominent velocity heterogeneities in the crust and uppermost mantle and an eastward thinning of the crust, geographically correlating with geological settings. CSRM‐1.0 improvements include accurate estimation of shallow seismic structure, increased spatial resolution and improved model accuracy. Crustal composition inferred from CSRM‐1.0 exhibits a general transition from a felsic upper crust to a mafic lower crust. Mafic rocks in the lower crust are found predominantly along inter‐block boundaries and sporadically within the interiors of blocks, likely resulted from preferential inter‐block intrusions of magmas related to various oceanic plate subductions and the Emeishan mantle plume. This study contributes seismic constraints and CSRM‐1.0 to the CSRM product center (http://chinageorefmodel.org) as a backbone open‐access geophysical cyberinfrastructure. 4. Ultrasensitive Luminescence Switching of Zeolite Y Confined Silver Clusters for Dual‐Channel Oxygen Sensing.

Designing photoluminescence sensing devices for gaseous oxygen that combine ease of acquisition with efficient performance is a critical yet challenging task. Herein, a novel oxygen‐sensing probe based on zeolite Y confined silver clusters prepared via a simple approach, is proposed. The large Ag6 clusters formed after hydrogenation exhibit a bright orange emission. They are characterized by a high photoluminescence quantum yield (≈31%) and a long luminescence decay time (>500 µs). A possible formation mechanism of these clusters, which are for the first time observed in Faujasite zeolites, is proposed. Luminescence switching‐off, triggered by oxygen, can be detected in a dual‐channel combination of linear emission‐quenching and lifetime‐reduction, enabling a visual and ultrasensitive (limit of detection of 58 Pa) quantitative detection of oxygen in a wide pressure range (0–100 kPa) with fast response (<1 s), reversibility and reproducibility. A more detailed analysis shows that this high sensitivity is due to the long decay time of the orange emission and the high affinity of the zeolite for O2 adsorption rather than to a high rate constant for quenching. This work contributes to the development of optical oxygen sensing technology and lays the foundation for the design of silver cluster‐based zeolites for photo‐functional applications. 6. Clinicopathological features of hepatoid adenocarcinoma and non‐hepatoid adenocarcinoma of the stomach: A systematic review and meta‐analysis.

Background: Hepatoid adenocarcinoma of the stomach (HAS) is an extremely rare and unique malignant gastric tumor with a significantly worse prognosis than non‐hepatoid adenocarcinoma of the stomach (non‐HAS). The present study explored the clinicopathological features of HAS and non‐HAS patients to provide insights into HAS treatment strategies. Methods: From December 26, 2023, we performed a comprehensive search of the PubMed, Web of Science, Cochrane Library, and Embase.com databases for relevant studies. Two authors independently screened the studies, evaluated their quality, extracted data, and performed the analyses. This study was registered with PROSPERO on January 2, 2024. Results: Nine retrospective studies were included for analysis after screening 833 articles. A total of 350 and 924 patients were enrolled in the HAS and non‐HAS groups, respectively. While no significant differences were observed in age, sex, tumor size, T3 or T4 stage, and N2 or N3 stage between the two groups, the HAS group exhibited higher rates of lymph node metastasis (OR = 1.93, 95% CI: 1.19–3.13, p = 0.007), liver metastasis (OR = 3.45, 95% CI: 2.26–5.28, p < 0.001), and vascular invasion (OR = 2.76, 95% CI: 2.05–3.71, p < 0.001). Additionally, the HAS group had lower 3‐year survival rates (HR = 2.35, 95% CI: 1.70–3.25, p < 0.001) and 5‐year survival rates (HR = 3.63, 95% CI: 1.49–8.88, p = 0.005), but lower rates of lymphatic permeation (OR = 0.68, 95% CI: 0.47–0.99, p = 0.040). Conclusion: Based on the current clinical evidence, patients with HAS present distinct clinicopathological features, greater invasiveness, and poorer prognosis than non‐HAS patients. Recently, therapies utilizing extracellular vesicles (EVs) derived from mesenchymal stromal/stem cells (MSCs) have begun to show promise in clinical trials. However, EV therapeutic potential varies with MSC tissue source and in vitro expansion through passaging. To find the optimal MSC source for clinically translatable EV‐derived therapies, this study aims to compare the angiogenic and immunomodulatory potentials and the protein and miRNA cargo compositions of EVs isolated from the two most common clinical sources of adult MSCs, bone marrow and adipose tissue, across different passage numbers. Primary bone marrow‐derived MSCs (BMSCs) and adipose‐derived MSCs (ASCs) were isolated from adult female Lewis rats and expanded in vitro to the indicated passage numbers (P2, P4, and P8). EVs were isolated from the culture medium of P2, P4, and P8 BMSCs and ASCs and characterized for EV size, number, surface markers, protein content, and morphology. EVs isolated from different tissue sources showed different EV yields per cell, EV sizes, and protein yield per EV. Gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analyses of proteomics data and miRNA seq data identified key proteins and pathways associated with differences between BMSC‐EVs and ASC‐EVs, as well as differences due to passage number. In vitro tube formation assays employing human umbilical vein endothelial cells suggested that both tissue source and passage number had significant effects on the angiogenic capacity of EVs. With or without lipopolysaccharide (LPS) stimulation, EVs more significantly impacted expression of M2‐macrophage genes (IL‐10, Arg1, TGFβ) than M1‐macrophage genes (IL‐6, NOS2, TNFα). By correlating the proteomics analyses with the miRNA seq analysis and differences observed in our in vitro immunomodulatory, angiogenic, and proliferation assays, this study highlights the trade‐offs that may be necessary in selecting the optimal MSC source for development of clinical EV therapies. 8. Pharmacokinetic, Pharmacodynamic, and Safety Profiles of Proline Henagliflozin in Chinese Subjects with Varying Degrees of Liver Dysfunction.

Proline henagliflozin, a novel selective inhibitor of sodium glucose cotransporter 2, is a treatment for type 2 diabetes mellitus. We designed a parallel‐group, open‐label, and multicenter study to evaluate the pharmacokinetic (PK), pharmacodynamic (PD), and safety profiles of henagliflozin in Chinese subjects with varying degrees of liver dysfunction. Thirty‐two subjects were enrolled and divided into four groups based on liver function (normal liver function, mild, moderate, or severe liver dysfunction). The area under the plasma concentration from time zero to infinity of henagliflozin in subjects with mild liver dysfunction, moderate liver dysfunction, and severe liver dysfunction compared with normal liver function was increased by 137%, 197%, and 204%, respectively. The maximum plasma concentration was also increased by 123%, 129%, and 139%, respectively. PK parameters of three metabolites varied to different degrees in the liver dysfunction groups than in the normal liver function group. The mean accumulative excretion amounts and fraction of dose excreted in urine expressed as a percentage were all increased with the decrease of liver function. The PD parameters were significantly higher in liver dysfunction groups than those in the normal liver function group. However, the urine creatinine (UCr) was not significantly different among the groups. No notable adverse events or adverse drug reactions were observed. Due to the higher exposures in subjects with liver dysfunction, the benefit: risk ratio should be individually assessed because the long‐term safety profile and efficacy have not been specifically studied in this population. Background: Cardiovascular disease (CVD) is recognized as a primary and severe comorbidity in patients with type 2 diabetes mellitus (T2DM) and is also identified as a leading cause of mortality within this population. Consequently, the identification of novel biomarkers for the risk stratification and progression of CVD in individuals with T2DM is of critical importance. Methods: This retrospective cohort study encompassed 979 patients diagnosed with T2DM, of whom 116 experienced CVD events during the follow‐up period. Clinical assessments and comprehensive blood laboratory analyses were conducted. Age‐ and sex‐adjusted Cox proportional hazard regression analysis was utilized to evaluate the association between lipoprotein‐associated phospholipase A2 (Lp‐PLA2), C1q/tumor necrosis factor‐related protein 3 (CTRP‐3), and the incidence of CVD in T2DM. The diagnostic performance of these biomarkers was assessed through receiver operating characteristic (ROC) curve analysis and the computation of the area under the curve (AUC). Results: Over a median follow‐up of 84 months (interquartile range: 42 [32–54] months), both novel inflammatory markers, Lp‐PLA2 and CTRP‐3, and traditional lipid indices, such as low‐density lipoprotein cholesterol and apolipoprotein B, exhibited aberrant expression in the CVD‐afflicted subset of the T2DM cohort. Age‐ and sex‐adjusted Cox regression analysis delineated that Lp‐PLA2 (hazard ratio [HR] = 1.007 [95% confidence interval {CI}: 1.005–1.009], p < 0.001) and CTRP‐3 (HR = 0.943 [95% CI: 0.935–0.954], p < 0.001) were independently associated with the manifestation of CVD in T2DM. ROC curve analysis indicated a substantial predictive capacity for Lp‐PLA2 (AUC = 0.81 [95% CI: 0.77–0.85], p < 0.001) and CTRP‐3 (AUC = 0.91 [95% CI: 0.89–0.93], p < 0.001) in forecasting CVD occurrence in T2DM. The combined biomarker approach yielded an AUC of 0.94 (95% CI: 0.93–0.96), p < 0.001, indicating enhanced diagnostic accuracy. Conclusions: The findings suggest that the biomarkers Lp‐PLA2 and CTRP‐3 are dysregulated in patients with T2DM who develop CVD and that each biomarker is independently associated with the occurrence of CVD. The combined assessment of Lp‐PLA2 and CTRP‐3 may significantly augment the diagnostic precision for CVD in the T2DM demographic. [ABSTRACT FROM AUTHOR]

11. Clinicopathological characteristics and diagnostic accuracy of BRAF mutations in ameloblastoma: A Bayesian network analysis.

Objective: This Bayesian network meta‐analysis was performed to analyze the associations between clinicopathological characteristics and BRAF mutations in ameloblastoma (AM) patients and to evaluate the diagnostic accuracy. Materials and Methods: Four electronic databases were searched from 2010 to 2024. The search terms used were specific to BRAF and AM. Observational studies or randomized controlled trials were considered eligible. The incidence of BRAF mutation and corresponding clinicopathological features in AM patients were subjected to Bayesian network analyses and diagnostic accuracy evaluation. Results: A total of 937 AM patients from 20 studies were included. The pooled prevalence of BRAF mutations in AM patients was 72%. According to the Bayesian network analysis, BRAF mutations are more likely to occur in younger (odds ratio [OR], 2.3; credible interval [CrI]: 1.2–4.5), mandible site (OR, 3.6; 95% CrI: 2.7–5.2), and unicystic (OR, 1.6; 95% CrI: 1.1–2.4) AM patients. Similarly, higher diagnostic accuracy was found in the younger, mandible, and unicystic AM groups. Conclusions: The incidence, risk, and diagnostic accuracy of BRAF mutation in AM were greater in younger patients, those with mandible involvement, and those with unicystic AM than in patients with other clinicopathological features. In addition, there was a strong concordance in the diagnostic accuracy between molecular tests and immunohistochemical analysis. [ABSTRACT FROM AUTHOR]

Interactions with primary caregivers shape children's expectations of interactions with others, but the mechanism is not clear yet. We included Han Chinese children aged 5–6 (N = 121, 61 boys) from a city in East China and tested their support‐giving expectations of mothers, fathers and unfamiliar others after observing mother–child interactions with different levels of maternal responsiveness. Across two studies, we found that children's support‐giving behaviour and willingness expectations decreased significantly in the unresponsive condition for mothers and fathers but not for unfamiliar others; while these expectations did not change significantly for all partners in the responsive condition. The results suggest that relationship type between children and interaction partners moderates the effects of maternal responsiveness on children's expectations of partners' support‐giving. Chaihu‐jia‐Longgu‐Muli decoction (CLMD) has been proven clinically effective in treating vertigo with anxiety disorder. However, the mechanism is not clear. This study aimed to explore the mechanism of CLMD in treating vertigo with anxiety disorder based on ultra‐performance liquid chromatography–quadrupole time‐of‐flight/mass spectrometry (UPLC–Q‐TOF/MS) and network pharmacology. UPLC–Q‐TOF/MS was performed to identify the compounds in blood and the targets of compounds of CLMD in vertigo and anxiety were searched using databases. A protein–protein interaction network was built to screen the core targets. The core targets were analyzed by Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analyses. In addition, the vertigo with anxiety rat model was used to verify the results. A total of 22 compounds were absorbed into the blood. Eighty‐one potential targets associated with CLMD for vertigo with anxiety disorder were identified through network pharmacological analysis. Subsequently, GO and KEGG analysis showed that CLMD treatment for vertigo with anxiety disorder is associated with neurotransmitter levels and other pertinent physiological processes. The results of the animal experiments showed that CLMD decreased the levels of serotonin, norepinephrine and dopamine, alleviating the symptoms of vertigo and anxiety disorder in model rats. The study revealed CLMD could alleviate vertigo and anxiety symptoms through reducing the levels of neurotransmitters. [ABSTRACT FROM AUTHOR]

Summary: Real‐time estimation of crowd counting in underground metro systems, constrained by limited space, is crucial for managing heightened pedestrian volumes and responding promptly to emergencies. To address this challenge, we propose a passenger state transition‐based model, called STRmt, designed for the seamless and continuous monitoring of real‐time crowd movement within service areas of stations and trains, leveraging auto fare collection systems (AFC) as a comprehensive sensor network. Our innovation lies in modeling the dynamic movement of passengers within a metro system over time as a state transition process aligned with the train schedule. To achieve this, we introduce a spatio‐temporal deep learning framework, denoted as STnet, designed to dynamically predict these state transitions. The performance of our method is rigorously assessed through extensive experiments conducted spanning 2 years in Shenzhen, China, utilizing AFC data, train schedule data, and weather data. Although the tandem WOLED have the potential of high efficiency and long lifetime, the high voltage of tandem structure usually limits its application inAR/VR products. To achieve low voltage WOLED, it is necessary to develop single structure. Most of the reported single WOLED with the use of TADF materials show a relatively large CIE coordinates. In this work, we use the combination of blue fluorescence and yellow TADF materials with a 1‐2 nm p‐type interlayer material between them. The energy is transferred from blue fluorescence to TADF. And finally, the single WOLED with tunable color temperature was realized. The CIE coordinates can be tuned from the warm white point of (0.313, 0.323) to the cold white point of (0.285, 0.279). The roll‐off of efficiency from 240 nit to 3000 nit is lower than 10%, and the lifetime (LT95) can reach 1300 h at 1000 nit. [ABSTRACT FROM AUTHOR]

Due to its high quantum yield and unique spectral characteristics, quantum dots (QDs) color conversion materials have attracted great attention in the application of luminescent display products. However, there is a lack of effective optical model for QD color conversion materials, especially for the propagation and distribution of light, when the QD ink contains scattering particles. In this paper, a full‐view optical model of QD conversion and blue backlight device architecture is established. The model can not only output the spectral information in the front view, but also predict the spectral information of the QD color conversion device in other different views. At the same time, the effects of QD concentration, thickness, particle size and refractive index on the luminous efficiency of the device structure are also studied by using this optical model. On the other hand, the influence of conversion efficiency of QD and EL backlight on the power consumption and lifetime of QD products is also studied by using this optical model. The model can realize the output of the optical influence on the future products by the parameters related to QD color conversion materials and backlight without actual device fabrication, which can greatly reduce time and labor cost. We discuss the effects of different electron injection layers (EILs) (NaF/LiF/Liq/ETL:Li) on the electrical properties, lifetime and capacitance systematically base on the bottom emission blue OLED device in the paper. We can determine the aging speed and initial decay of the OLED device from the characteristics of the capacitance. In the Addition, the mechanism of diverse inorganic electron injection layers resulting in the different characteristics of OLED devices is deeply analyzed. Macrophage pyroptosis is of key importance to host defence against pathogen infections and may participate in the progression and recovery of periodontitis. However, the role of pyroptotic macrophages in regulating periodontal ligament stem cells (PDLSCs), the main cell source for periodontium renewal, remains unclear. First, we found that macrophage pyroptosis were enriched in gingiva tissues from periodontitis patients compared with those of healthy people through immunofluorescence. Then the effects of pyroptotic macrophages on the PDLSC osteogenic differentiation were investigated in a conditioned medium (CM)‐based coculture system in vitro. CM derived from pyroptotic macrophages inhibited the osteogenic differentiation‐related gene and protein levels, ALP activity and mineralized nodule formation of PDLSCs. The osteogenic inhibition of CM was alleviated when pyroptosis was inhibited by VX765. Further, untargeted metabolomics showed that glutamate limitation may be the underlying mechanism. However, exogenous glutamate supplementation aggravated the CM‐inhibited osteogenic differentiation of PDLSCs. Moreover, CM increased extracellular glutamate and decreased intracellular glutamate levels of PDLSCs, and enhanced the gene and protein expression levels of system xc− (a cystine/glutamate antiporter). After adding cystine to CM‐based incubation, the compromised osteogenic potency of PDLSCs was rescued. Our data suggest that macrophage pyroptosis is related to the inflammatory lesions of periodontitis. Either pharmacological inhibition of macrophage pyroptosis or nutritional supplements to PDLSCs, can rescue the compromised osteogenic potency caused by pyroptotic macrophages. [ABSTRACT FROM AUTHOR]

19. Sleep quality during pregnancy and fetal growth: A prospective cohort study.

Summary The aim of this study is to investigate the association between sleep quality during pregnancy and fetal growth. Pregnant women and their fetuses at 16–20 gestational weeks in Nantong Maternal and Child Health Hospital were recruited. Women were classified as having “good sleep quality” (Pittsburgh Sleep Quality Index score ≤ 5) and “poor sleep quality” (Pittsburgh Sleep Quality Index score > 5) according to the Pittsburgh Sleep Quality Index scores. The fetal growth was evaluated by three ultrasonographic examinations, birth weight and birth length. We used general linear model and multiple linear regression models to estimate the associations. A total of 386 pairs of mother and infant were included in the data analysis. After adjusting for gestational weight gain, anxiety and depression, fetuses in the good sleep quality group had greater abdominal circumference (p = 0.039 for 28–31+6 weeks gestation, p = 0.012 for 37–40+6 weeks gestation) and femur length (p = 0.014 for 28–31+6 weeks gestation, p = 0.041for 37–40+6 weeks gestation) at 28–31+6 weeks gestation and 37–40+6 weeks gestation, and increased femur length (p = 0.007) at 28–31+6 weeks gestation. Birth weights (p = 0.018) were positively associated with sleep quality. Poor sleep quality was associated with poor intrauterine physical development, decreased abdominal circumference and femur length, and lower birth weight after adjusting for confounding factors. Attention to the fetal growth of pregnant women with poor sleep quality has the potential to decrease the risk of adverse fetal outcomes. [ABSTRACT FROM AUTHOR]

20. A neural network copula function approach for solving joint basic probability assignment in structural reliability analysis.

- Abstract
Applying evidence theory to structural reliability analysis under epistemic uncertainty, it is necessary to consider the correlation of evidence variables. Among them, solving the joint basic probability assignment (BPA) of the evidence variables is a crucial link. In this study, a solution method of joint BPA based on neural network copula function is proposed. This method is to automatically construct copula function through neural network, which avoids the process of selecting the optimal copula function. Firstly, the neural network copula function is constructed based on the sample set of evidence variables. Then, the expression for solving the joint BPA using the neural network copula function is derived through vectors. Furthermore, the expression is used to map the marginal BPA of evidence variables to joint BPA, thus realizing the solution of joint BPA. Finally, the effectiveness of this method is verified by three examples. The results show that the neural network copula function describes the data distribution better than the optimal copula function selected by the traditional method. In addition, there is actually an error in solving the reliability intervals using the traditional optimal copula function method, whereas the results of this paper's neural network copula function method are more accurate and better for decision making. 21. Novel multidisciplinary cooperation model for obstetric medical quality control: A quasi‐experimental study.

Objective: To explore the impact of a novel multidisciplinary cooperation model in obstetric medical quality control. Methods: This quasi‐experimental study analyzed the quality indicators of full‐term pregnant women who underwent vaginal trial labor in Zibo Maternal and Child Health Hospital between July 2021 and June 2022. The pregnant women were divided into two groups based on implementation of novel multidisciplinary cooperation: multidisciplinary and non‐multidisciplinary. We compared the rate of labor analgesia, postpartum hemorrhage in vaginal delivery, transfer to cesarean section, and the 5‐min Apgar score ≤7 in full‐term neonates. Results: A total of 3751 pregnant women were enrolled into the study, of whom 2004 were included in the non‐multidisciplinary group and 1747 in the multidisciplinary group. The analgesic rate of delivery of the multidisciplinary group was higher than that of the non‐multidisciplinary group (P = 0.000). We established that the rate of postpartum bleeding (P = 0.040), transfer cesarean section (P = 0.003) and the incidence of Apgar score ≤7 in 5 min of full‐term neonates (P = 0.038) of the multidisciplinary group was lower than that of the non‐multidisciplinary group. There was no significant difference in the mean ages (29.40 ± 3.99 vs. 29.90 ± 4.27 years; P = 0.126), mean delivery gestational ages (39.65 ± 0.87 vs. 39.64 ± 1.06; P = 0.221), mean gravidity values (1.93 ± 1.09 vs. 2.00 ± 1.18; P = 0.586) and mean parity (1.40 ± 0.56 vs. 1.42 ± 0.59; P = 0.635) of the women in the two groups. Conclusion: Multidisciplinary cooperation in delivery management can significantly improve some quality indicators. We established the analgesic rate of delivery can be increased and the rate of postpartum bleeding, transfer cesarean section and the incidence of Apgar score ≤7 in 5 min of full‐term neonates can be decreased with the implementation of novel multidisciplinary cooperation. Background: Renal allograft interstitial fibrosis/tubular atrophy (IF/TA) constitutes the principal histopathological characteristic of chronic allograft dysfunction (CAD) in kidney‐transplanted patients. While renal vascular endothelial‐mesenchymal transition (EndMT) has been verified as an important contributing factor to IF/TA in CAD patients, its underlying mechanisms remain obscure. Through single‐cell transcriptomic analysis, we identified Rictor as a potential pivotal mediator for EndMT. This investigation sought to elucidate the role of Rictor/mTORC2 signalling in the pathogenesis of renal allograft interstitial fibrosis and the associated mechanisms. Methods: The influence of the Rictor/mTOR2 pathway on renal vascular EndMT and renal allograft fibrosis was investigated by cell experiments and Rictor depletion in renal allogeneic transplantation mice models. Subsequently, a series of assays were conducted to explore the underlying mechanisms of the enhanced mitophagy and the ameliorated EndMT resulting from Rictor knockout. Results: Our findings revealed a significant activation of the Rictor/mTORC2 signalling in CAD patients and allogeneic kidney transplanted mice. The suppression of Rictor/mTORC2 signalling alleviated TNFα‐induced EndMT in HUVECs. Moreover, Rictor knockout in endothelial cells remarkably ameliorated renal vascular EndMT and allograft interstitial fibrosis in allogeneic kidney transplanted mice. Mechanistically, Rictor knockout resulted in an augmented BNIP3‐mediated mitophagy in endothelial cells. Furthermore, Rictor/mTORC2 facilitated the MARCH5‐mediated degradation of BNIP3 at the K130 site through K48‐linked ubiquitination, thereby regulating mitophagy activity. Subsequent experiments also demonstrated that BNIP3 knockdown nearly reversed the enhanced mitophagy and mitigated EndMT and allograft interstitial fibrosis induced by Rictor knockout. Conclusions: Consequently, our study underscores Rictor/mTORC2 signalling as a critical mediator of renal vascular EndMT and allograft interstitial fibrosis progression, exerting its impact through regulating BNIP3‐mediated mitophagy. Tight junction (TJ) proteins establish a physical barrier between epithelial cells, playing a crucial role in maintaining tissue homeostasis by safeguarding host tissues against pathogens, allergens, antigens, irritants, etc. Recently, an increasing number of studies have demonstrated that abnormal expression of TJs plays an essential role in the development and progression of inflammatory airway diseases, including chronic obstructive pulmonary disease, asthma, allergic rhinitis, and chronic rhinosinusitis (CRS) with or without nasal polyps. Among them, CRS with nasal polyps is a prevalent chronic inflammatory disease that affects the nasal cavity and paranasal sinuses, leading to a poor prognosis and significantly impacting patients' quality of life. Its pathogenesis primarily involves dysfunction of the nasal epithelial barrier, impaired mucociliary clearance, disordered immune response, and excessive tissue remodeling. Numerous studies have elucidated the pivotal role of TJs in both the pathogenesis and response to traditional therapies in CRS. Objective: To evaluate patient preference for short (gist) or detailed/extensive decision aids (DA) for genetic testing at ovarian cancer (OC) diagnosis. Design: Cohort study set within recruitment to the Systematic Genetic Testing for Personalised Ovarian Cancer Therapy (SIGNPOST) study (ISRCTN: 16988857). Setting: North‐East London Cancer Network (NELCN) population. Population/Sample: Women with high‐grade non‐mucinous epithelial OC. Methods: A more detailed DA was developed using patient and stakeholder input following the principles/methodology of IPDAS (International Patients Decision Aids Standards). Unselected patients attending oncology clinics evaluated both a pre‐existing short and a new long DA version and then underwent mainstreaming genetic testing by a cancer clinician. Appropriate inferential descriptive and regression analyses were undertaken. Main outcome measures: Satisfaction, readability, understanding, emotional well‐being and preference for long/short DA. Results: The mean age of patients was 66 years (interquartile range 11), and 85% were White British ethnicity. Of the participants, 74% found DAs helpful/useful in decision‐making. Women reported higher levels of satisfaction (86% versus 58%, p < 0.001), right amount of information provided (76.79% versus49.12%, p < 0.001) and improved understanding (p < 0.001) with the long DA compared with the short DA. There was no statistically significant difference in emotional outcomes (feeling worried/concerned/reassured/upset) between 'short' and 'long' DA; 74% of patients preferred the long DA and 24% the short DA. Patients undergoing treatment (correlation coefficient (coef) = 0.603; 95% CI 0.165–1.041, p = 0.007), those with recurrence (coef = 0.493; 95% CI 0.065–0.92, p = 0.024) and older women (coef = 0.042; 95% CI 0.017–0.066, p = 0.001) preferred the short DA. Ethnicity did not affect outcomes or overall preference for long/short DA. Conclusions: A longer DA in OC patients has higher satisfaction without increasing emotional distress. Older women and those undergoing treatment/recurrence prefer less extensive information, whereas those in remission preferred a longer DA. [ABSTRACT FROM AUTHOR]

25. Validating the accuracy of the Hendrich II Fall Risk Model for hospitalized patients using the ROC curve analysis.

This retrospective study was conducted at a medical center in southern Taiwan to assess the accuracy of the Hendrich II Fall Risk Model (HIIFRM) in predicting falls. Sensitivity, specificity, accuracy, and optimal cutoff points were analyzed using receiver operating characteristic (ROC) curves. Data analysis was conducted using information from the electronic medical record and patient safety reporting systems, capturing 303 fall events and 47,146 non‐fall events. Results revealed that at the standard threshold of HIIFRM score ≥5, the median score in the fall group was significantly higher than in the non‐fall group. The top three units with HIIFRM scores exceeding 5 were the internal medicine (50.6%), surgical (26.5%), and oncology wards (14.1%), indicating a higher risk of falls in these areas. ROC analysis showed an HIIFRM sensitivity of 29.5% and specificity of 86.3%. The area under the curve (AUC) was 0.57, indicating limited discriminative ability in predicting falls. At a lower cutoff score (≥2), the AUC was 0.75 (95% confidence interval: 0.666–0.706; p < 0.0001), suggesting acceptable discriminative ability in predicting falls, with an additional identification of 101 fall events. This study emphasizes the importance of selecting an appropriate cutoff score when using the HIIFRM as a fall risk assessment tool. 26. Association of cardiovascular disease prevalence with BMD and fracture in men with T2DM.

Background: Patients with type 2 diabetes mellitus (T2DM) are predisposed to cardiovascular disease (CVD). Bone mineral density (BMD) is linked to CVD, but most studies focused on women. Our analysis aims to explore the association of BMD and fracture with the prevalence of CVD in men with T2DM. Methods: In this retrospective cross‐sectional study, 856 men with T2DM were enrolled. BMDs at the lumbar spine (L2‐4), femoral neck (FN), and total hip (TH) were measured by dual‐energy X‐ray absorptiometry (DXA). The CVD outcome was determined as the sum of the following conditions: congestive heart failure, coronary heart disease, angina pectoris, myocardial infarction, the requirement for coronary artery revascularization, and stroke. The relationship between BMDs and CVD was investigated by restricted cubic spline curves and logistic regression models. Results: A total of 163 (19.0%) patients developed CVD. The restricted cubic spline curve revealed a linear and negative association between FN‐BMD, TH‐BMD, and CVD. After full adjustments for confounding covariates, the odds ratios were 1.34 (95% confidence interval [CI] [1.11–1.61], p <.05), 1.3 (95% CI [1.05–1.60], p <.05), and 1.26 (95% CI [1.02–1.55], p <.05) for each 1‐SD decrease in BMDs of L2‐4, FN and TH, respectively. T‐scores of < −1 for BMD of L2‐4 and FN were independently associated with CVD (p <.05). Subgroup analyses further supported our findings. Conclusions: The prevalence of CVD was inversely correlated with BMD levels in men with T2DM, particularly at the FN. We hypothesized that monitoring FN‐BMD and early intervention would help reduce CVD risk in men with T2DM, especially those with hypertension. [ABSTRACT FROM AUTHOR]

Background: Diabetic nephropathy (DN) is a complication caused by diabetes. Circular RNAs (circRNAs) are a kind of RNA with a closed circular structure, which has high stability and is involved in many disease‐related processes. The mechanism of circRNA TAO kinase 1 (circTAOK1) in the pathogenesis and development of DN is unclear. Methods: CircTAOK1, microRNA (miR)‐142‐3p, and sex‐determining region Y‐box transcription factor 6 (SOX6) mRNA levels were analyzed by real‐time quantitative polymerase chain reaction (RT‐qPCR). Cell counting kit‐8 (CCK8) and 5‐ethynyl‐2′‐deoxyuridine (EdU) assays were used to analyze cell proliferation. Cell cycle distribution was detected by flow cytometry. Western blot assay was performed to test B‐cell lymphoma 2 (Bcl‐2), Bcl‐2 associated X (Bax), cleaved‐caspase 3, and fibronectin (FN), collagen I (Col I), and collagen IV (Col IV) protein levels. ELISA assay was used to measure interleukin 1β (IL‐1β), interleukin 6 (IL‐6), and tumor necrosis factor (TNF‐α) levels. The reactive oxygen species (ROS) and malondialdehyde (MDA) levels and the superoxide dismutase (SOD) activity were assessed by the corresponding kits. And the correlation between miR‐142‐3p and circTAOK1 or SOX6 was confirmed by dual luciferase reporter assay, RNA immunoprecipitation assay and RNA pull down assay. Results: CircTAOK1 and SOX6 expression levels were up‐regulated, while miR‐142‐3p expression was down‐regulated in DN serum and HG‐treated HK‐2 cells. Knockdown of circTAOK1 could inhibit cell injury of HG‐induced HK‐2 cells. The inhibitory effect of circTAOK1 knockdown on HG‐induced HK‐2 cell injury was restored by miR‐142‐3p downregulation. CircTAOK1 acted as a sponge for miR‐142‐3p, and SOX6 was targeted by miR‐142‐3p. The overexpression of SOX6 could recover the effect of miR‐142‐3p overexpression on HG‐induced HK‐2 cell injury. CircTAOK1 regulated the expression of SOX6 by targeting miR‐142‐3p. Conclusion: CircTAOK1 knockdown inhibited HG‐induced HK‐2 cell damage in DN by the miR‐142‐3p/SOX6 axis. [ABSTRACT FROM AUTHOR]

Activating cGAS‐STING pathway has great potential to achieve effective antitumor immunotherapy. However, mutant p53 (mutp53), a commonly observed genetic alteration in over 50% of human cancer, will impede the therapeutic performance of the cGAS‐STING pathway. Herein, multifunctional ZIF‐8@MnO2 nanoparticles are constructed to degrade mutp53 and facilitate the cGAS‐STING pathway. The synthesized ZIF‐8@MnO2 can release Zn2+ and Mn2+ in cancer cells to induce oxidative stress and cytoplasmic leakage of fragmented mitochondrial double‐stranded DNAs (dsDNAs). Importantly, the released Zn2+ induces variable degradation of multifarious p53 mutants through proteasome ubiquitination, which can alleviate the inhibitory effects of mutp53 on the cGAS‐STING pathway. In addition, the released Mn2+ further increases the sensitivity of cGAS to dsDNAs as immunostimulatory signals. Both in vitro and in vivo results demonstrate that ZIF‐8@MnO2 effectively promotes the cGAS‐STING pathway and synergizes with PD‐L1 checkpoint blockades, leading to remarkable regression of local tumors as well as distant metastases of breast cancer. This study proposes an inorganic metal ion‐based nanoplatform to enhance the cGAS‐STING‐mediated antitumor immunotherapy, especially to those tumors with mutp53 expression. [ABSTRACT FROM AUTHOR]

Small metal oligomers have gained significant attention due to their exceptional optoelectronic properties and versatile applications, ranging from sensors to imaging and catalysis. However, the need for matrices (silica, zeolites, or metal‐organic frameworks) to prevent aggregation into larger metallic particles reduces their potential applicability in light‐emitting devices (LEDs) due to their poor electrical properties. To address this issue, a novel semiconductor stabilizer, titanosilicate, is proposed as an innovative solution for Ag‐based luminescent materials. These results show that titanosilicate, characterized by high thermochemical stability and well‐defined semiconducting properties, is an ideal scaffold for Ag‐luminescent species. Optical spectroscopic techniques reveal that Ag‐titanosilicates feature an orange fluorescence with a photoluminescence quantum yield reaching 20%. In contrast, a combination of X‐ray diffraction, TEM, and XEOL‐X‐ray absorption spectroscopic techniques show that, unlike zeolites, the luminescent species consist of single Ag atoms. The electroluminescent properties of the proposed Ag‐titanosilicate are further investigated within a conventional LED architecture by using the new material as an emissive layer. The developed proof‐of‐concept LED exhibits a significant improvement of the ZEOLEDs, where a smaller low turn‐on voltage of 2 V (enabling energy‐efficient operation) and high color rendering index of 80 introduces exciting prospects for developing advanced Ag‐LED phosphors. Rapid advances in robotics have placed urgent demands on more intelligent human‐machine interaction technologies. Specifically, the way of establishing dual‐way intuitive communication with a consistent sensory system can greatly enhance efficiency and reliability. Here, a bi‐directional human‐machine interface (HMI) is designed by applying starch‐based hydrogel sensors. The whole system consists of a multi‐modal wearable sensory exoskeleton with a haptic feedback module and sensory robotic hand. The sensory exoskeleton with strain‐sensing glove and rotation‐sensing arm can capture and project the motion of the entire upper limb. The system offers object recognition functions by utilizing a sensing array on the robotic hands and machine learning algorithms, which can identify the shape and hardness information. The recognized results can be delivered back to the operator via vision and vibrational haptic feedback, respectively. Molecular diodes are of considerable interest for the increasing technical demands of device miniaturization. However, the molecular diode performance remains contact‐limited, which represents a major challenge for the advancement of rectification ratio and conductance. Here, it is demonstrated that high‐quality ultrathin organic semiconductors can be grown on several classes of metal substrates via solution‐shearing epitaxy, with a well‐controlled number of layers and monolayer single crystal over 1 mm. The crystals are atomically smooth and pinhole‐free, providing a native interface for high‐performance monolayer molecular diodes. As a result, the monolayer molecular diodes show record‐high rectification ratio up to 5 × 108, ideality factor close to unity, aggressive unit conductance over 103 S cm−2, ultrahigh breakdown electric field, excellent electrical stability, and well‐defined contact interface. Large‐area monolayer molecular diode arrays with 100% yield and excellent uniformity in the diode metrics are further fabricated. These results suggest that monolayer molecular crystals have great potential to build reliable, high‐performance molecular diodes and deeply understand their intrinsic electronic behavior. This research presents a method for the oxidative selenylation and thiolylation of Morita‐Baylis‐Hillman adducts through constant‐current electrolysis in an undivided cell at room temperature. The reaction enables direct production of trisubstituted (Z)‐allyl aryl selenides and sulfides with yields of 27–98%. A wide variety of different functionalities are well tolerated under these reaction conditions. This strategy could be carried out on gram scale and several synthetic transformations were accomplished for the construction of other allyl selenide derivatives. Poly (ethylene oxide) (PEO)‐based polymer electrolytes show the prospect in all‐solid‐state lithium metal batteries; however, they present limitations of low room‐temperature ionic conductivity, and interfacial incompatibility with high voltage cathodes. Therefore, a salt engineering of 1, 1, 2, 2, 3, 3‐hexafluoropropane‐1, 3‐disulfonimide lithium salt (LiHFDF)/LiTFSI system was developed in PEO‐based electrolyte, demonstrating to effectively regulate Li ion transport and improve the interfacial stability under high voltage. We show, by manipulating the interaction between PEO matrix and TFSI−‐HFDF−, the optimized solid‐state polymer electrolyte achieves maximum Li+ conduction of 1.24 × 10−4 S cm−1 at 40 °C, which is almost 3 times of the baseline. Also, the optimized polymer electrolyte demonstrates outstanding stable cycling in the LiFePO4/Li and LiNi0.8Mn0.1Co0.1O2/Li (3.0–4.4 V, 200 cycles) based all‐solid‐state lithium batteries at 40 °C. Under the fixed light source intensity (LSI), eggshell thickness difference results in excessive exposure or darkening in imaging, which affects the accuracy of crack identification. To optimize it, a solution involves the utilization of an innovative adaptive light source for optical imaging is proposed. A multi function station is adopted. Among them, one station serves as a reference workstation is utilized to obtain the average grayscale value of the eggs image. The other sub stations serve as detection workstations to acquire and detect the panoramic images of the eggshell. By using the red component average grayscale as a reference to adjust the LSI in the remaining sub stations, imaging consistency is effectively. Moreover, a dual threshold segmentation algorithm was developed to attenuate the dark spot, thereby improving crack detection. Experiments results demonstrating the proposed method significantly improving the detection accuracy of non microcracked eggs while achieving approximately 90% and 92% detection accuracy for intact and cracked eggs, respectively. Practical Applications: There are a few studies on the consistency of light for eggs, and in actual online detection, the light source is often set to a fixed light intensity value, which can lead to overexposure or over‐darkening of the egg image due to variations in the eggshell thickness, and both overexposed and over‐darkened images will result in decreased accuracy of crack recognition. This study designed and built an online detection system for eggshell cracks based on variable light intensity. Therefore, the system not only can help to improve the crack recognition rate of poultry eggs and improve their quality but also would help to promote the development of the commercialization of poultry eggs, which has a broad application prospect. Due to the absence of acquired immunity, insects primarily rely on their innate immune system to resist pathogenic microorganisms and parasitoids in natural habitats. This innate immune system can be classified into cellular immunity and humoral immunity. Cellular immunity is mediated by hemocytes, which perform phagocytosis, aggregation, and encapsulation to fight against invaders, whereas the humoral immunity primarily activates the immune signaling pathways and induces the generation of immune effectors. Existing studies have revealed that the hemipteran aphids lack some crucial immune genes compared to other insect species, indicating the different immune mechanisms in aphids. The current review summarizes the adverse impacts of pathogenic microorganisms and parasitoids on aphids, introduces the cellular and humoral immune systems in insects, and analyzes the differences between aphids and other insect species. Furthermore, our review also discussed the existing prospects and challenges in aphid immunity research, and proposed the potential application of immune genes in green pest management. Aims/Introduction: Previous studies have shown that circular ribonucleic acid mediates the occurrence of diabetic nephropathy. This study aimed to analyze the effects of circ_0068087 on high‐glucose (HG)‐induced human kidney 2 (HK2) cell dysfunction. Materials and Methods: Circ_0068087, miR‐580‐3p, and progestin and adipoQ receptor 3 (PAQR3) expression were detected by quantitative reverse transcription polymerase chain reaction. Cell viability and proliferation were investigated by Cell Counting Kit‐8 and EdU assays, respectively. The cell apoptotic rate was assessed by flow cytometry. Inflammatory response was assessed by enzyme‐linked immunoassays. Oxidative stress was evaluated by a superoxide dismutase activity assay kit and lipid peroxidation malondialdehyde assay kit. Molecular interaction was identified by dual‐luciferase reporter assay. Results: Circ_0068087 and PAQR3 expression were significantly upregulated in diabetic nephropathy patients. HG treatment inhibited HK2 cell proliferation, but induced cell apoptosis, inflammation, oxidative stress and epithelial–mesenchymal transition by regulating circ_0068087. Circ_0068087 acted as a microribonucleic acid‐580‐3p (miR‐580‐3p) sponge, and miR‐580‐3p targeted PAQR3. Furthermore, circ_0068087 depletion repressed PAQR3 expression through miR‐580‐3p. MiR‐580‐3p inhibitors or PAQR3 introduction attenuated circ_0068087 silencing mediated‐effects in HG‐treated HK2 cells. Conclusion: Circ_0068087 promoted HG‐induced HK2 cell injuries by the regulation of the miR‐580‐3p/PAQR3 pathway. [ABSTRACT FROM AUTHOR]

Objectives: As a critical component of the epithelial barrier, tight junctions (TJs) are essential in nasal mucosa against pathogen invasion. However, the function of TJs has rarely been reported in nasal inverted papilloma (NIP). This study aims to investigate the potential factors of TJs' abnormality in NIP. Methods: We assessed the expression of ZO‐1, occludin, claudin‐1, claudin‐3, and claudin‐7 in healthy controls and NIP by real‐time quantitative polymerase chain reaction and immunofluorescent staining. The correlation between TJs expression and neutrophil count, TH1/TH2/TH17 and regulatory T cell biomarkers, and the proportion of nasal epithelial cells was investigated. Results: Upregulation of ZO‐1, occludin, claudin‐1, and claudin‐7, along with downregulation of claudin‐3, was found in NIP compared to control (all p < 0.05). An abnormal proportion with a lower number of ciliated cells (control vs. NIP: 37.60 vs. 8.67) and goblet cells (12.52 vs. 0.33) together with a higher number of basal cells (45.58 vs. 124.00) in NIP. Meanwhile, claudin‐3 was positively correlated with ciliated and goblet cells (all p < 0.01). Additionally, neutrophils were excessively infiltrated in NIP, negatively correlated with ZO‐1, but positively with claudin‐3 (all p < 0.05). Furthermore, FOXP3, IL‐10, TGF‐β1, IL‐5, IL‐13, and IL‐22 levels were induced in NIP (all p < 0.01). Occludin level was negatively correlated with IL‐10, IL‐5, IL‐13, and IL‐22, whereas ZO‐1 was positively with TGF‐β1 (all p < 0.05). Conclusion: Nasal epithelial barrier dysfunction with TJs anomalies is commonly associated with abnormal proliferation and differentiation of epithelial cells and imbalance of immune and inflammatory patterns in NIP. Continuous 6‐mercaptopurine (6‐MP) dose titration is necessary because of its narrow therapeutic index and frequently encountered dose‐limiting hematopoietic toxicity. However, evidence‐based guidelines for gene‐based 6‐MP dosing have not been established for Chinese children with acute lymphoblastic leukemia (ALL). This multicenter, randomized, open‐label, active‐controlled clinical trial randomly assigned Chinese children with low‐ or intermediate‐risk ALL in a 1:1 ratio to receive TPMT–NUDT15 gene–based dosing of 6‐MP (N = 44, 10 to 50 mg/m2/day) or standard dosing (N = 44, 50 mg/m2/day) during maintenance therapy. The primary end point was the incidence of 6‐MP myelosuppression in both groups. Secondary end points included frequencies of 6‐MP hepatotoxicity, duration of myelosuppression and leukopenia, event‐free survival, and steady‐state concentrations of active metabolites (6‐thioguaninenucleotides and 6‐methylmercaptopurine nucleotides) in erythrocytes. A 2.2‐fold decrease in myelosuppression, the primary end point, was observed in the gene‐based–dose group using ~ 50% of the standard initial 6‐MP dose (odds ratio, 0.26, 95% confidence interval, 0.11 to 0.64, P = 0.003). Patients in the gene‐based–dose group had a significantly lower risk of developing thiopurine‐induced myelosuppression and leukopenia (P = 0.015 and P = 0.022, respectively). No significant differences were observed in the secondary end points of the incidence of hepatotoxicity and steady‐state concentrations of active metabolites in erythrocytes between the two groups. TPMT‐ and NUDT15‐based dosing of 6‐MP will significantly contribute toward further reducing the incidence of leukopenia in Chinese children with ALL. The direct epitaxial growth of graphene on semi‐insulating SiC presents significant potential for a variety of technologically important applications, including next‐generation electronics, photonics, and quantum metrology. However, this approach also poses a competitive disadvantage in terms of quality and cost, primarily due to the uncontrollable and time‐consuming nature of the annealing process. Herein, a thermal shock annealing (TSA) method is reported that enables kinetics‐controlled epitaxial growth of graphene on SiC within 10 s, which efficiently fulfills the requirements for producing high‐quality, few‐layer, and low‐cost graphene on SiC. The epitaxial graphene (EG) grown on both β‐SiC nanoparticles (SiC@EG NPs) and centimeter‐scale α‐SiC wafer (EG/SiC) exhibits mono‐ or bi‐layer features with negligible structural defects. Moreover, the findings indicate that the TSA method can efficiently mitigate the persistent issue of step bunching conundrum and improve the flatness of EG/SiC. As an application demonstration, the significant enhancement of surface‐enhanced infrared absorption (SEIRA) by SiC@EG NPs is exhibited. The graphene plasmon arising on SiC@EG NPs enables SEIRA detection sensitivity of up to a monolayer of p‐nitrobenzenethiol (p‐NTP). Consequently, the precise regulation and comprehensive comprehension of TSA afford an exceedingly desirable approach to produce cost‐effective, high‐quality EG growth on SiC for diverse emerging application scenarios. Postoperative cognitive dysfunction (POCD) is a prevalent central nervous system complication predominantly observed in elderly patients. Sevoflurane, a general anaesthetic agent, has been implicated in the development of POCD, yet the underlying regulatory mechanisms potentially involving Sestrin1 (SESN1), a stress‐responsive protein that plays a critical role in cellular homeostasis and protection against stress‐induced damage, including oxidative stress and DNA damage, remain elusive. This study endeavoured to elucidate the impact of SESN1 on sevoflurane‐induced cognitive impairment in rats. Employing a model in which SESN1 was transfected into SD male rats and cognitive dysfunction was induced by sevoflurane. The Morris Water Maze test was used for behavioural evaluation, Enzyme‐Linked Immunosorbent Assay, Western blotting and immunofluorescence were applied to assess the influence of SESN1 on the inflammatory response and mitophagy in the rat hippocampus. The study further aimed to uncover the putative mechanism by which SESN1, through SIRT1, might modulate cognitive function. Concurrently, levels of malondialdehyde, superoxide dismutase and mitochondrially produced ATP within the rat hippocampus were quantified. Experimental outcomes suggested that SESN1 overexpression significantly mitigated the deleterious effects of sevoflurane anaesthesia, ameliorated neuroinflammation and inflammasome activation, modified mitochondrial function and facilitated mitophagy. Additionally, SESN1, via the activation of SIRT1, may suppress inflammasome activation and mitochondrial dysfunction. Collectively, these findings underscore SESN1's integral role in counteracting sevoflurane‐induced cognitive impairment, impeding inflammasome activation, enhancing mitochondrial function and fostering mitophagy, which appear to be intricately linked to SESN1‐mediated SIRT1 activation. The aqueous micro batteries (AMBs) are expected to be one of the most promising micro energy storage devices for its safe operation and cost‐effectiveness. However, the performance of the AMBs is not satisfactory, which is attributed to strong interaction between metal ions and the electrode materials. Here, the first AMBs are developed with NH4+ as charge carrier. More importantly, to solve the low conductivity and the dissolution during the NH4+ intercalation/extraction problem of perylene material represented by perylene‐3,4,9,10‐tetracarboxylic dianhydride (PTCDA), the Ti3C2Tx MXene with high conductivity and polar surface terminals is introduced as a conductive skeleton (PTCDA/Ti3C2Tx MXene). Benefitting from this, the PTCDA/Ti3C2Tx MXene electrodes exhibit ultra‐high cycle life and rate capability (74.31% after 10 000 galvanostatic chargedischarge (GCD) cycles, and 91.67 mAh g−1 at 15.0 A g−1, i.e., capacity retention of 45.2% for a 30‐fold increase in current density). More significantly, the AMBs with NH4+ as charge carrier and PTCDA/Ti3C2Tx MXene anode provide excellent energy density and power density, cycle life, and flexibility. Extracellular vesicles (EVs) secreted by human‐induced pluripotent stem cells (hiPSCs) have great potential as cell‐free therapies in various diseases, including prevention of blood–brain barrier senescence and stroke. However, there are still challenges in pre‐clinical and clinical use of hiPSC‐EVs due to the need for large‐scale production of a large quantity. Vertical‐Wheel bioreactors (VWBRs) have design features that allow the biomanufacturing of hiPSC‐EVs using a scalable aggregate or microcarrier‐based culture system under low shear stress. EV secretion by undifferentiated hiPSCs expanded as 3‐D aggregates and on Synthemax II microcarriers in VWBRs were investigated. Additionally, two types of EV collection media, mTeSR and HBM, were compared. The hiPSCs were characterized by metabolite and transcriptome analysis as well as EV biogenesis markers. Protein and microRNA cargo were analysed by proteomics and microRNA‐seq, respectively. The in vitro functional assays of microglia stimulation and proliferation were conducted. HiPSCs expanded as 3‐D aggregates and on microcarriers had comparable cell number, while microcarrier culture had higher glucose consumption, higher glycolysis and lower autophagy gene expression based on mRNA‐seq. The microcarrier cultures had at least 17–23 fold higher EV secretion, and EV collection in mTeSR had 2.7–3.7 fold higher yield than HBM medium. Microcarrier culture with mTeSR EV collection had a smaller EV size than other groups, and the cargo was enriched with proteins (proteomics) and miRNAs (microRNA‐seq) reducing apoptosis and promoting cell proliferation (e.g. Wnt‐related pathways). hiPSC‐EVs demonstrated the ability of stimulating proliferation and M2 polarization of microglia in vitro. HiPSC expansion on microcarriers produces much higher yields of EVs than hiPSC aggregates in VWBRs. EV collection in mTeSR increases yield compared to HBM. The biomanufactured EVs from microcarrier culture in mTeSR have exosomal characteristics and are functional in microglia stimulation, which paves the ways for future in vivo anti‐aging study. Organic field‐effect transistors switched by insulated gates are the most essential building blocks, while usually plagued with degraded gate control arising from complicated dielectric engineering. Subsequently, the resulting large supply voltage and power consumption remain an essential issue for portable electronics driven by a single battery of only 1.5 V. Herein, wafer‐scale organic Schottky‐gate transistor arrays using inkjet‐printed few‐layer organic semiconducting crystals are reported. The transistors exhibit steep switching characteristics with an average subthreshold swing of 55 mV per dec and high signal amplification efficiency over 45 S A‐1, attributed to efficient Schottky gating and enhanced charge injection. Thereafter, high‐gain inverters are successfully demonstrated with an ultralow power consumption of only 800 pW; also, they are integrated as 1 V driven sequential logic circuits. A coplanar double‐gate geometry is also introduced for low‐voltage, single‐device AND logic. Therefore, the work opens new avenues toward the sustainable advancement in single‐battery‐driven, ultralow‐power organic electronics. [ABSTRACT FROM AUTHOR]
