Abstract Background The purpose of this study was to investigate the photoprotection effect of peroxiredoxin 1 (PRDX1) protein in ultraviolet B (UVB) irradiation-induced damage of retinal pigment epithelium (RPE) and its possible molecular mechanism. Methods ARPE-19 cell viability and apoptosis were assessed by MTT assay and flow cytometry, respectively. Real-time quantitative reverse transcription polymerase chain reaction (qRT-PCR) was used to detect the PRDX1 expression. The corresponding kits were employed to measure the levels or activities of lactate dehydrogenase (LDH), 8-hydroxy-2-deoxyguanosine (8-OHdG), reactive oxygen species (ROS), malondialdehyde (MDA), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD). Western blotting was applied to examine PRDX1 expression and mitogen-activated protein kinase (MAPK) signaling pathway-related proteins. Results After exposure to 20 mJ/cm2 intensity of UVB irradiation for 24 h, ARPE-19 cells viability was decreased, the leakage degree of LDH and 8-OHdG were increased, and cell apoptosis was elevated. The expression of PRDX1 was significantly down-regulated in UVB-induced ARPE-19 cells. The low expression of PRDX1 was involved in high irradiation intensity. Overexpression of PRDX1 increased cell activity, decreased cell apoptosis, and LDH as well as 8-OHdG leakage in UVB-induced ARPE-19 cells. In addition to alleviating UVB-induced cell damage, PRDX1 overexpression also inhibited UVB-induced oxidative stress (down-regulation of ROS and MDA levels, up-regulation of GSH-Px and SOD activities) and the activation of MAPK signaling pathway in ARPE-19 cells. Conclusion PRDX1 exerts a photoprotection effect on RPE by attenuating UVB-induced cell damage and inhibiting oxidative stress, which can be attributed to the inhibition of MAPK signaling pathway activation.

IntroductionImmunotherapies targeting T cells in solid cancers are revolutionizing clinical treatment. Novel immunotherapies have had extremely limited benefit for acute myeloid leukemia (AML). Here, we characterized the immune microenvironment of t(8;21) AML patients to determine how immune cell infiltration status influenced prognosis.MethodsThrough multi-omics studies of primary and longitudinal t(8;21) AML samples, we characterized the heterogeneous immune cell infiltration in the tumor microenvironment and their immune checkpoint gene expression. Further external cohorts were also included in this research.ResultsCD8+ T cells were enriched and HAVCR2 and TIGIT were upregulated in the CD34+CD117dim%-High group; these features are known to be associated with immune exhaustion. Data integration analysis of single-cell dynamics revealed that a subset of T cells (cluster_2) (highly expressing GZMB, NKG7, PRF1 and GNLY) evolved and expanded markedly in the drug-resistant stage after relapse. External cohort analysis confirmed that the cluster_2 T-cell signature could be utilized to stratify patients by overall survival outcome.DiscussionIn conclusion, we discovered a distinct T-cell signature by scRNA-seq that was correlated with disease progression and drug resistance. Our research provides a novel system for classifying patients based on their immune microenvironment.

It is challenging to reveal the catalytic mechanism of Cu/ZnO catalysts with multivalent copper and improve the performance of CO2 hydrogenation to methanol. We synthesized Cu/ZnO catalysts with different Cu:Zn ratios using acetylacetone salts as the precursors, in which Cu, Cu2O, CuO coexist. The catalytic activity was significantly associated with the abundance of Cu+ active sites on the surface and the formation of more Cu+-ZnO interfaces. When the ratio of Cu:Zn = 5:4, the catalyst without reduction pretreatment showed good performance, with a CO2 conversion rate of 12.9%, methanol selectivity of 85.1%, and the space-time yield of 0.238 gMeOH gcat−1 h−1. It was suggested that Cu+ tends to closely bind with ZnO, and the preparation method leads to more Cu+-ZnO interfaces. The synergistic interaction between Cu+ and ZnO promoted the adsorption and dissociation of CO2 and H2, verifying its dual effect on CO2 and H2 and promoting CO2 hydrogenation to methanol.

Background: Inflammatory bowel disease (IBD; mainly ulcerative colitis and Crohn’s disease) is associated with the development of colorectal cancer (CRC) referred to as colitis-associated colorectal cancer (CAC). In inflammatory flares of IBD, the production of luminal nitric oxide (NO) increases due to the increased inducible nitric oxide synthase (iNOS) activity in inflamed tissue. It is believed that iNOS parallels pro-inflammatory interleukin-1β (IL-1β). How these biomarkers relate to CAC pathogenesis or survival is unknown. Aim: The primary aim of this study was to investigate iNOS and IL-1β immunoreactivity in CAC tumors in comparison with CRC and normal colonic mucosa, and the secondary aim was to determine if immunoreactivity correlates with 5-year survival of CAC. Methods: Immunohistochemistry was performed on tissue sections as follows: CAC (n = 59); sporadic CRC (sCRC) (n = 12); colonic mucosa >2 cm outside sCRC margin (normal mucosa) (n = 22); paracancerous IBD (pIBD) (n = 12). The expression of iNOS and IL-1β was quantified separately for epithelium and stroma. Data were evaluated using the Mann-Whitney U-test and the log-rank test for 5-year Kaplan-Meier survival curves. Results were compared with online mRNA databases. Results: Immunoreactivity occurred predominantly in epithelial cells and to lesser extent in stroma. Compared with normal mucosa, immunoreactivity for iNOS (P < 0.01) and IL-1β (P < 0.005) was higher in CAC epithelium. In CAC stroma, iNOS immunoreactivity was lower than normal mucosa (P < 0.001), whereas IL-1β was higher (P < 0.05). Immunoreactivity differences of iNOS or IL-1β among CAC patients failed to correlate with 5-year survival. These findings were supported by online mRNA databases. Conclusion: Consistent with high NO production in IBD, there is more iNOS in CAC epithelium, albeit not in stroma. This immunoreactivity difference exists for IL-1β in both epithelium and stroma. The intervention of arginine or iNOS activity for CAC chemotherapy is not straightforward.

The clinical applications of acrosin activity are limited. We analyzed 61 578 male partners in infertile couples who visited the outpatient department of the Reproductive and Genetic Hospital of CITIC-Xiangya (Changsha, China) between August 2014 and December 2019 to determine the reference ranges and thresholds for acrosin activity in infertile Chinese men; to determine whether correlations exist between acrosin activity and age, sperm concentration, sperm morphology, or sperm motility; and to evaluate whether acrosin activity could serve as an effective prognostic indicator for choosing between in vitro fertilization (IVF) and intracytoplasmic sperm injection (ICSI) in the clinic. The cut-off value for the normal reference range of acrosin activity for male partners in infertile couples was 24.78 μIU per 10[6] sperm. There was no significant association between acrosin activity and age, sperm concentration, semen volume, total sperm count, progressive motility, or total motile spermatozoa. A weak positive correlation was found between acrosin activity and normal sperm morphology. The crystallography of transformation products with different cooling rates has been investigated in a typical low-carbon alloy steel, and its effect on low-temperature impact toughness uniformity in the thickness direction of heavy steel plates has been elucidated. The results show that both the full lath martenite (LM) microstructure transformed at the cooling rate of 45 K s−1 and granular bainite (GB) + lath bainite (LB) + LM microstructure transformed at the cooling rates of 3∼6 K s−1 have higher density of high-angle grain boundaries (HAGBs). The density of HAGBs in the full GB microstructure transformed at the cooling rate of 0.5 K s−1 is the lowest, and that in the LB + LM microstructure transformed at the cooling rates of 15∼22 K s−1 is the middle. In the GB + LB + LM microstructure, the division of austenite by multi-stage transformation reduces the LB and LM transformation temperatures, contributing to a weakening of variant selection and a tendency to close-packed plane grouping. Therefore, the GB + LB + LM microstructure achieves the refinement degree similar to the full LM microstructure, but slightly higher than that of the LB + LM microstructure. For the 60 mm thick heavy steel plate with ultra-fast cooling, the cooling rate decreases from 41 K s−1 at the subsurface to 14 K s−1 at the 1/4-thickness and then to 4 K s−1 at the 1/2-thickness. The full LM microstructure, LB + LM microstructure, and GB + LB + LM microstructure are obtained in sequence, resulting in a more uniform density of HAGBs. Consequently, the relatively uniform impact toughness at 143 K in the thickness direction is achieved with a small difference of ∼30 J.

Background Acute ischemic stroke is a major cause of mortality and disability worldwide, with approximately 7.4% to 7.7% recurrence within the first 3 months. This study aimed to identify potential biomarkers for predicting stroke recurrence. Methods and Results We conducted a nested case–control study using a hospital‐based cohort from the Third China National Stroke Registry selecting 214 age‐ and sex‐matched patients with ischemic stroke with hypertension and no history of diabetes or heart disease. Using data‐independent acquisition for discovery and multiple reaction monitoring for quantitative validation, we identified 26 differentially expressed proteins in large‐artery atherosclerosis (Causative Classification of Ischemic Stroke [CCS]1), 16 in small‐artery occlusion (CCS3), and 25 in undetermined causes (CCS5) among patients with recurrent stroke. In the CCS1 and CCS3 subgroups, differentially expressed proteins were associated with platelet aggregation, neuronal death/cerebroprotection, and immune response, whereas differentially expressed proteins in the CCS5 subgroup were linked to altered metabolic functions. Validated recurrence predictors included proteins associated with neutrophil activity and vascular inflammation (TAGLN2 [transgelin 2], ITGAM [integrin subunit α M]/TAGLN2 ratio, ITGAM/MYL9 [myosin light chain 9] ratio, TAGLN2/RSU1 [Ras suppressor protein 1] ratio) in the CCS3 subgroup and proteins associated with endothelial plasticity and blood–brain barrier integrity (ITGAM/MYL9 ratio and COL1A2 [collagen type I α 2 chain]/MYL9 ratio) in the CCS3 and CCS5 subgroups, respectively. Conclusions These findings provide a foundation for developing a blood‐based biomarker panel, using causative classifications, which may be used in routine clinical practice to predict stroke recurrence.

In this paper, in the context of rural revitalization + data platform, a grassroots employment platform for college students is designed according to the data architecture, and the data is processed hierarchically to achieve the organization and knowledge accumulation of cross-domain employment data, which makes multiple employment data standardized. Using data analysis parallelization algorithm and dimensionality reduction operation method, the training data are read and loaded into the distributed memory of the employment data platform to select the best employment features. According to the design platform of this paper, the employment rate for those employed in the eastern region and 80.16% in the central region has reached 91.15%. This paper's design model can improve employee efficiency and expand the grassroots employment space of college students, as indicated.

This paper performs feature extraction based on the Bayesian nonparametric model for smart sports courses to analyze the Gaussian process principle for composing random variables. The probability distribution of the parameters is estimated based on the known data set, given the input data vector to decompose the likelihood function. To calculate the posterior probabilities of the parameters, the kernel function features are analyzed to give the expressions of eigenvalues and eigenvectors, and data normalization is performed. According to the results, the experimental group had higher sports test scores than the control group, and the third test score of high-speed running reached 9 points. Therefore, colleges and universities should focus on cultivating students' innovative thinking abilities and helping them master sports knowledge and skills to achieve their educational goals.

Hainan yellow cattle are indigenous Zebu cattle from southern China known for their tolerance of heat and strong resistance to disease. Generations of adaptation to the tropical environment of southern China and decades of artificial breeding have left identifiable selection signals in their genomic makeup. However, information on the selection signatures of Hainan yellow cattle is scarce. Herein, we compared the genomes of Hainan yellow cattle with those of Zebu, Qinchuan, Nanyang, and Yanbian cattle breeds by the composite likelihood ratio method (CLR), Tajima’s D method, and identifying runs of homozygosity (ROHs), each of which may provide evidence of the genes responsible for heat tolerance in Hainan yellow cattle. The results showed that 5210, 1972, and 1290 single nucleotide polymorphisms (SNPs) were screened by the CLR method, Tajima’s D method, and ROH method, respectively. A total of 453, 450, and 325 genes, respectively, were identified near these SNPs. These genes were significantly enriched in 65 Gene Ontology (GO) functional terms and 11 Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways (corrected p < 0.05). Five genes—Adenosylhomocysteinase-like 2, DnaJ heat shock protein family (Hsp40) member C3, heat shock protein family A (Hsp70) member 1A, CD53 molecule, and zinc finger and BTB domain containing 12—were recognized as candidate genes associated with heat tolerance. After further functional verification of these genes, the research results may benefit the understanding of the genetic mechanism of the heat tolerance in Hainan yellow cattle, which lay the foundation for subsequent studies on heat stress in this breed.

Hydrate formation in foam drainage gas recovery wells and the shut in accidents caused by plugging have become an important problem that restricts the safe production of natural gas. The blockage and accumulation of hydrates is a gradual problem. This research goes beyond predicting the formation of hydrates and delves deeper into examining the rate of hydrate formation and the degree of pipeline blockage at different wellbore locations. First, the temperature model, pressure model, multiphase flow model, and hydrate plugging model of hydrate formation process are established from the equations of mass conservation, energy conservation, and momentum conservation. Second, an iterative approach is employed to solve the model, with a maximum error of 6.86% in model validation. Finally, sensitivity analysis shows that wellhead temperature, wellhead pressure, and foam viscosity have different effects on hydrate formation, maximum plugging position, and plugging degree. At the same time, combined with the actual drainage and gas production process, and the characteristics of hydrate blockage, proposed hydrate prevention measures can be taken to achieve safe production of natural gas. The research results indicate that a decrease in temperature signifies an increase in undercooling, resulting in an accelerated rate of hydrate formation and an elevated risk of hydrate blockage. The decrease in wellhead pressure leads to a decrease in the rate of hydrate formation and an increase in production, which is beneficial for the hydrate prevention. However, larger pressure differences and gas production rates will put higher requirements on equipment such as well control devices. An increase in foam viscosity will lead to increased pressure, foam compression, reduced drainage capacity, and intensified hydrate generation. Phenotypic variation analysis and comprehensive evaluation are important tools for selecting provenances of forest trees, which, in turn, is crucial for genetic improvement of forest trees. The study analyzed eight needle-leaf and fourteen tracheid trait indexes of ten provenances of Qinghai spruce forest stands. Multiple comparisons, correlation analysis, cluster analysis, and principal component analysis were used to evaluate the differences in needle-leaf and tracheid traits among the different provenances. The study found significant differences in the needle-leaf and tracheid traits among the provenances. All traits included 50 pairs of positive and 26 pairs of negative correlations. The coefficient of variation for the tracheid trait (18.86%) was higher than that for the needle-leaf trait (17.20%). A comprehensive evaluation of needle-leaf and tracheid traits was conducted using principal component analysis. The study demonstrated that the degree of variation in needle-leaf and tracheid traits among the provenances of Qinghai spruce was extensive, with a multitude of sources of variation. Therefore, it is crucial to strengthen the selection of good provenances in the early seed selection process. Background: Verrucas that occur on the soles of the feet are called plantar warts, most of which can recur repeatedly and are difficult to eradicate. Hypertrophic and refractory plantar warts are often accompanied by pain and discomfort, which cause many inconveniences in patients' daily lives. Aim: This study aimed to analyze the therapeutic effect of superficial radiotherapy (SRT‐100) on refractory plantar warts and further create favorable conditions for the subsequent treatment of this disease with a high recurrence rate. Methods: A retrospective analysis was conducted for refractory plantar warts treated with superficial radiotherapy in our outpatient department from January to June 2023. Results: A total of 30 patients were included in our study (median age, 33 years). The female‐to‐male ratio was 1:3.29. Two to six months after radiotherapy, all of the warts subsided in 23 (76.67%) patients, most of the warts subsided in 4 (13.33%) patients, 3 (10%) patients did not respond to treatment, and 7 (23.33%) patients had recurrent or new warts after their warts subsided. Conclusions: Most patients with refractory plantar warts improved after superficial radiotherapy. Our study presented that men are more susceptible to plantar warts than women, and young and middle‐aged people are the main population affected by the disease. Superficial radiotherapy is an effective treatment for refractory plantar warts, which can quickly remove the warts in a short period. It is safe and noninvasive, with minimal adverse reactions. Some patients relapse after the lesion is clear, and superficial radiotherapy can create favorable conditions for the subsequent treatment of viral warts in clinical practice. [ABSTRACT FROM AUTHOR]

Sporadic or late-onset Alzheimer's disease (LOAD) accounts for more than 95% of Alzheimer's disease (AD) cases without any family history. Although genome-wide association studies have identified associated risk genes and loci for LOAD, numerous studies suggest that many adverse environmental factors, such as social isolation, are associated with an increased risk of dementia. However, the underlying mechanisms of social isolation in AD progression remain elusive. In the current study, we found that 7 days of social isolation could trigger pattern separation impairments and presynaptic abnormalities of the mossy fibre-CA3 circuit in AD mice. We also revealed that social isolation disrupted histone acetylation and resulted in the downregulation of 2 dentate gyrus (DG)-enriched miRNAs, which simultaneously target reticulon 3 (RTN3), an endoplasmic reticulum protein that aggregates in presynaptic regions to disturb the formation of functional mossy fibre boutons (MFBs) by recruiting multiple mitochondrial and vesicle-related proteins. Interestingly, the aggregation of RTN3 also recruits the PP2A B subunits to suppress PP2A activity and induce tau hyperphosphorylation, which, in turn, further elevates RTN3 and forms a vicious cycle. Finally, using an artificial intelligence-assisted molecular docking approach, we determined that senktide, a selective agonist of neurokinin3 receptors (NK3R), could reduce the binding of RTN3 with its partners. Moreover, application of senktide in vivo effectively restored DG circuit disorders in socially isolated AD mice. Taken together, our findings not only demonstrate the epigenetic regulatory mechanism underlying mossy fibre synaptic disorders orchestrated by social isolation and tau pathology but also reveal a novel potential therapeutic strategy for AD. Objective To understand the prevalence and annual change trend of Internet addiction among students aged 11 and 22 in Inner Mongolia in 2019 and 2021, and to explore the influencing factors of Internet addiction behavior, so as to provide reference basis for the prevention of Internet addiction among students. Methods Students were selected from 12 cities of Inner Mongolia Autonomous Region by stratified cluster random sampling. Internet addiction disorder was determined according to the items of "surveillance and intervention of common diseases and health influencing factors of students in China". Logistic regression method was used to analyze the relationship between Internet addiction and behavioral factors. Results The prevalence of Internet addiction among students aged 11 to 22 years in Inner Mongolia in 2019 was 1.92%, 2.50%, and 2.21%, respectively. Logistic regression analysis showed that vocational high school (OR=1.347, 95%CI: 1.083-1.675), single parent family (OR=1.279, 95% CI: 1.090-1.500), reconstituted family (OR=1326, 95% CI: 1.041-1.690), bullying (OR=1.755, 95% CI: 1.443-2.135), depression (OR=3.557, 95% CI: 3.146-4.021), fighting (OR=3.374, 95% CI: 2.512-4.531), sugary drinks (OR=2.182. 95% CI:1.753-2.716), and eating fried food (OR=2.398, 95% CI: 1.908-3.014) were the risk factor of Internet addiction. Outdoor activities for 2 hours (OR= 0.620, 95% CI: 0.530-0.726), non-smoking (OR=0.553, 95% CI: 0.476-0.643), and non-drinking (OR=0.581, 95% CI: 0.507-0.666) were the protective factors of Internet addiction. Conclusion The current situation of Internet addiction among students in Inner Mongolia is not optimistic. Attention should be paid to bullying, depression, reconstituted families, fighting students, and vocational high school students. Outdoor activities for 1 to 2 hours a day, reducing the intake of sugary drinks and fried foods, and no smoking and drinking can effectively reduce the incidence of Internet addiction. Flax is the main oil crop in China, and has been affected by flax wilt for a long time. The catalytic subunit of β-1, 3-glucan synthase (FKS/GLS) is necessary for cell wall synthesis, the biological function of which was analyzed in the study through cloning the gene, constructing FolGLS2 gene deletion mutant, observing phenotype and assaying the virulence of the gene disruption mutants. A full-length of 5947bp DNA sequence was obtained, which contained an open reading frame 5847bp, encoding a total of 1948 amino acids. Observation of the phenotype of the FolGLS2 gene deletion mutant showed that the colony of the mutant was small and compact, its growth rate decreased obviously, the mycelium was short, thick and no longer produced conidia. The cell wall of the mutant mycelium changed somehow, which was strongly resistant to the protoplasting buffer commonly used for wild type and was unable to release protoplasts. Infection assay to the flax seedling demonstrated that pathogenicity of deletion mutant was significantly decreased. Conclusively, FolGLS2 gene, which encodes catalytic subunit of β-1, 3- glucan synthase, plays a regulatory role in conidiogenesis, mycelial vegetative growth, cell wall properties and pathogenicity of Fusarium oxysporum f.sp.lini. Pollinator sharing often leads to receipt of heterospecific pollen (HP) along with conspecific pollen. As a result, flowering plants can accumulate diverse communities of HP on stigmas. While variation in HP diversity is an important selective force contributing to flowering plant fitness, evolution and community assembly, our understanding of the extent and drivers of heterogeneity of HP diversity is limited.In this study, we examined the species compositions and abundances of ~1000 HP communities across 59 co‐flowering plant species in three serpentine seep communities in California, USA. We evaluated the variation in HP diversity (γ diversity) across plant species in each seep and asked whether the variation in HP γ diversity was caused by variation in HP diversity within stigmas (α diversity) or HP compositional variation among stigmas (β diversity) due to the replacement of HP species (turnover) or their loss (nestedness) from one stigma to another. We further evaluated the potential drivers of variation in HP α and β diversity using phylogenetic structural equation models.We found that variation in HP γ diversity across plant species was driven strongly by differences among species in HP α diversity and to a lesser extent by HP β diversity. HP community turnover contributed more to HP β diversity than nestedness consistently across plant species and seeps, suggesting a general pattern of HP compositional heterogeneity from stigma to stigma. The phylogenetic structural equation models further revealed that floral traits (e.g., stigma area, stigma‐anther distance, stigma exposure) and floral abundance were key in determining HP α diversity by influencing HP abundance (load size), while floral traits and abundance showed variable impact on HP β diversity (turnover and nestedness). Pollination generalism contributed relatively less to HP‐α and β diversity.These findings disentangle the heterogeneity in HP diversity at different levels, which is essential for understanding the process underlying patterns of HP receipt in plant communities.
