Given the persistently low adoption rate of improved sorghum varieties over decades, it is relevant to assess whether it is profitable or not to grow these varieties in Mali. Over the past years, little evidence has demonstrated the profitability analysis as decision-support information regarding the adoption of improved sorghum varieties. This study used cost-benefit analysis to assess the profitability of two Improved and Climate-Resilient Sorghum Varieties (ICRSVs), "Soubatimi and Tiandougou-coura" compared to the "Local" ones, using three years of average yield data (2017, 2018, and 2020) in Sikasso region, Mali. The objective was to perform a consistent cost-benefit analysis through net income, cost-benefit ratio, and gross profit margin. The study used a farm partial budget framework, sensitivity analysis, and stochastic dominance analysis methods. A final sample of 31 farmers' on-farm trials under the fertiliser package of "100 kg complex cereal and 50 kg urea" per hectare was held as the agronomic package. The key findings showed that both varieties were profitable, with 79,661 CFA (123.56 USD) and 45,073 CFA (69.91 USD) average net incomes corresponding to 1.54 and 1.32 CBR, and 34 and 24 percents average gross profit margins, respectively, while growing the "local" varieties was not profitable, with an average loss of 12,113 CFA (18.79 USD) with 0.91 CBR and 10 percent average gross profit margin. In light of these results, the study suggests a large dissemination of the ICRSVs in Mali. Policy-makers should facilitate the implementation of outreach programs to inform smallholder sorghum farmers on the ICRSVs' traits and profitability information as decision support tool for a larger adoption. A series of novel 3-benzimidazolyl retrochalcones (6a–g) was synthesized and evaluated for their anticancer activities against breast (MDA-MB-231, MCF-7) and human colon (Caco-2, HCT-116) cancer cell lines. The compounds demonstrated significant anticancer potential, with compound 6d exhibiting the most potent activity (IC50 < 1.56 μM) across all tested cell lines. Most compounds were more active than Roscovitine but less potent than Paclitaxel (Taxol®). Notably, compound 6e was inactive against both Caco-2 and MDA-MB-231 cell lines. The presence of electron-donating methoxy groups enhanced anticancer efficacy, while electron-withdrawing nitro groups had a detrimental effect. The high amplitude values (70%–98%) observed for the compounds indicate effective targeting of cancer cells, while lower amplitudes in fibroblasts (27%–49%) suggest selective antimitotic effects. These findings highlight the potential of benzimidazole-supported retrochalcones as promising candidates for further development as broad anticancer agents. [ABSTRACT FROM AUTHOR]

This study analyzes the relationships between the onset, the duration, and the maximum extent of the Atlantic Cold Tongue (ACT) using ERA5 reanalysis data from the European Centre for Medium-Range Weather Forecasts (ECMWF) over the period 1979–2019. After calculating the start and end dates of the ACT each year, this study investigates potential relationships between early or late onset that may be linked to the maximum duration and extent of the ACT, which is known to influence weather patterns and precipitation in surrounding regions and the West African Monsoon System. Results show that 68% of years with a short ACT duration are associated with a late-onset ACT, while 70% of years with a long ACT duration are associated with early ACT onset years. In addition, 63% of years with a short duration of ACT have a cold tongue with a low maximum extent, while 83% of years with a long duration of ACT have a cold tongue with a greater maximum extent. Finally, 78% of early ACT onset years are associated with the coldest SST tongue in the eastern equatorial Atlantic Ocean. A comparison of the last 20 years (1999–2019) with the previous 20 years (1979–1998) shows a cooling trend in SST, with ACT occurring and ending earlier in recent years than in the past. However, as the changes in the end date are greater than those in the onset date, the duration of the ACT has been 5–12 days shorter in the last 20 years than in the previous 20 years. Knowledge of these ACT characteristics and their interrelations and drivers is crucial for understanding the West African Monsoon System and for improving climate models and seasonal forecasts. The aim of this study is to characterize the compound extremes of rainfall and temperature in Côte d'Ivoire. For this purpose, we analyzed the outputs of fourteen (14) climate models from the CORDEX-Africa project. Results show an increase (approximately 4.3 °C) in the surface temperature and a decrease (5.90%) of the mean rainfall in the near (2036–2065) and far futures (2071–2100) over Côte d'Ivoire during the January–February–March (JFM) period. The analysis of the compound extremes of the wet/warm type highlights an increase in the frequency of this climatic hazard in the northern and central parts of the country during the January–March (JFM) season in the near and far futures. The dry/warm mode will increase in the central and southern parts of the country in the near future and in the whole country in the far future. These increases in compound extremes could lead to an increase in droughts and natural disasters across the country and could have a negative impact on socio-economic activities, such as transportation and agricultural production. This work could provide decision support for political decision-makers in formulating future public policies for managing agricultural production, food security, and natural disasters. Background: Germline RUNX1 haplodeficiency (RHD) is associated with thrombocytopenia, platelet dysfunction, and predisposition to myeloid malignancies. Platelet expression profiling of an RHD patient showed decreased F13A1, encoding for the A subunit of factor (F)XIII, a transglutaminase that cross-links fibrin and induces clot stabilization. FXIII-A is synthesized by hematopoietic cells, megakaryocytes, and monocytes. Objectives: To understand RUNX1 regulation of F13A1 expression in platelets/megakaryocytes and the mechanisms and consequences of decreased F13A1 in RHD. Methods: We performed studies in platelets, human erythroleukemia (HEL) cells, and human CD34+ cell-derived megakaryocytes including on clot contraction in cells following small inhibitor RNA knockdown (KD) of RUNX1 or F13A1. Results: Platelet F13A1 mRNA and protein were decreased in our index patient and in 2 siblings from an unrelated family with RHD. Platelet-driven clot contraction was decreased in the patient and affected daughter. Promoter studies in HEL cells showed that RUNX1 regulates F13A1 transcription; RUNX1 overexpression increased, and small inhibitor RNA RUNX1 KD reduced F13A1 promoter activity and protein. Following RUNX1 or F13A1 KD, clot contraction by HEL cells was decreased, as were FXIII-A surface expression, myosin light chain phosphorylation, and PAC1 antibody binding upon activation. F13A1 expression and clot contraction were impaired in RUNX1 downregulation in human megakaryocytes. Conclusion: RUNX1 regulates platelet-megakaryocyte F13A1 expression, which is decreased in RHD, reflecting regulation of a coagulation protein by a hematopoietic transcription factor. Platelet and megakaryocyte clot contraction is decreased in RHD, related to multiple impaired mechanisms including F13A1 expression, myosin phosphorylation, and αIIbβ3 activation.

La enfermedad de Kawasaki, bien conocida por los pediatras, es una vasculitis multisistémica aguda que afecta a los vasos de mediano calibre, con predilección por las arterias coronarias. En los países desarrollados, esta enfermedad es la principal causa de cardiopatía adquirida en niños. La fisiopatología de esta enfermedad todavía se desconoce, pero suele admitirse que uno o varios agentes infecciosos inducen una respuesta inflamatoria inapropiada en una persona con predisposición genética. El diagnóstico es clínico y debe sospecharse en los niños que presenten fiebre prolongada de más de 5 días con un síndrome inflamatorio en las pruebas de laboratorio. La American Heart Association definió unos criterios clínicos de inflamación en 2004 para establecer el diagnóstico. Sin embargo, algunos niños pueden presentar formas incompletas de la enfermedad, sobre todo cuando aparece a una edad inusual. Los síntomas se parecen a los de algunas patologías infecciosas, como la escarlatina o una infección viral, lo que dificulta el diagnóstico. Además, un retraso del diagnóstico o la ausencia de tratamiento aumentan el riesgo de desarrollar aneurismas coronarios. Las inmunoglobulinas polivalentes constituyen el tratamiento de primera línea, con una tasa de éxito del 80%. Si no hay respuesta a esta primera línea de tratamiento, se puede proponer un segundo ciclo de inmunoglobulinas, asociadas a corticoides, en las formas de mal pronóstico. Otros tratamientos, como el antifactor de necrosis tumoral alfa o algunos inmunosupresores, también pueden utilizarse en caso de resistencia a las inmunoglobulinas, pero deben comentarse con un equipo especializado.

Les pneumopathies aiguës bactériennes (PAB) occupent une place importante en pédiatrie. Elles représentent la première cause infectieuse d’hospitalisation chez l’enfant. L’OMS dénombre environ 2 millions de décès par an chez l’enfant de moins de 5 ans dont 70 % dans les pays en développement. L’objectif principal était d’étudier les profils épidémio-cliniques et thérapeutiques des PAB chez les enfants de moins de 16 ans.

Résumé : Inscrite dans le champ des littéracies universitaires, l’étude a pour objet les pratiques d’évaluation d’un genre de l’écrit à l’université : le mémoire de master dans sa version provisoire. Elle interroge, d’une part, ce qui est évalué à travers les interventions des enseignants sur les textes en devenir des étudiants et, d’autre part, de quelle manière ces interventions construisent un dialogue pédagogique avec les scripteurs. D’un point de vue méthodologique, l’analyse s’appuie sur un corpus de commentaires évaluatifs prélevés sur une trentaine de versions intermédiaires de mémoire. Les résultats montrent que les objets pris en compte dans les interventions correctives sur les textes des étudiants relèvent de trois dimensions de l’écrit, avec une attention soutenue pour la dimension formelle et ses catégories linguistiques (orthographe, syntaxe, ponctuation). Ils mettent également en évidence, à travers l’examen de leurs modalités énonciatives, la prépondérance du caractère sommatif des interventions au détriment de leur finalité formative. L’étude conclut en conséquence à la nécessité de construire à l’université une culture évaluative de l’écrit partagée, nécessaire au développement de pratiques outillées et permettant de faire de l’activité corrective un lieu de réduction des inégalités d’apprentissage de l’écriture de recherche. 32. A cost–benefit analysis of the production system with improved and climate-resilient sorghum varieties in southern Mali

Given the persistently low adoption rate of improved sorghum varieties over decades, it is relevant to assess whether it is profitable or not to grow these varieties in Mali. Over the past years, little evidence has demonstrated the profitability analysis as decision-support information regarding the adoption of improved sorghum varieties. This study used cost-benefit analysis to assess the profitability of two Improved and Climate-Resilient Sorghum Varieties (ICRSVs), “Soubatimi and Tiandougou-coura” compared to the “Local” ones, using three years of average yield data (2017, 2018, and 2020) in Sikasso region, Mali. The objective was to perform a consistent cost-benefit analysis through net income, cost-benefit ratio, and gross profit margin. The study used a farm partial budget framework, sensitivity analysis, and stochastic dominance analysis methods. A final sample of 31 farmers’ on-farm trials under the fertiliser package of “100 kg complex cereal and 50 kg urea” per hectare was held as the agronomic package. The key findings showed that both varieties were profitable, with 79,661 CFA (123.56 USD) and 45,073 CFA (69.91 USD) average net incomes corresponding to 1.54 and 1.32 CBR, and 34 and 24 percents average gross profit margins, respectively, while growing the “local” varieties was not profitable, with an average loss of 12,113 CFA (18.79 USD) with 0.91 CBR and 10 percent average gross profit margin. In light of these results, the study suggests a large dissemination of the ICRSVs in Mali. Policy-makers should facilitate the implementation of outreach programs to inform smallholder sorghum farmers on the ICRSVs’ traits and profitability information as decision support tool for a larger adoption.
