Dealuminated Beta zeolite has a large amount of silanol defects on its interface, which provides an ideal place for embedding metal species and creating metal active sites in a confined microenvironment. The confined metal sites as well as their surroundings are closely related to the reactant activation and transient state achievement. Hence, unraveling the confined metal sites is of great significance for the catalytic reaction process. Herein, niobium species were incorporated into the silanol defects over dealuminated Beta zeolite with a facile dry impregnation method, co-grinding the niobium precursor with dealuminated Beta zeolite support. The successful incorporation of niobium into the silanol defects for 30Nb-Beta zeolite was verified by DRIFT, 1H MAS NMR, UV–Vis and UV-Raman characterizations. XAS characterization and DFT calculations further disclosed that the confined Nb species existed as (SiO) 2 Nb(OH)(=O), containing two Si−O−Nb bonds, one Nb=O bond as well as one Nb−OH bond. The synthesized 30Nb-Beta zeolite catalyst displayed a superior cyclohexene conversion of 51.1%, cyclohexene oxide selectivity of 83.1% as well as TOF value of 188.2 h−1 ascribed to the inherent electrophilicity of Nb(V) for confined (SiO) 2 Nb(OH)(=O) species as well as the low oxygen transfer energy barrier for NbV−OOH species. Investigation of photoluminescence (PL) and fracture-induced triboluminescence (TL) is necessary for the development of both fundamental theories and practical applications in mechanical energy conversion; however, most known PL/TL-emitting materials are confined to inorganic systems. In this study, a novel lanthanide-based crystalline complex (LnCC), Eu(DBM) 3 DETA was synthesized via the synergistic coordination of Eu3+ with DBM (Dibenzoylmethane) and DETA (Diethylenetriamine) units, leading to the formation of brighter LnCC with bright red emission, high PL quantum yields (57.19 %) and unique TL characteristics. The key to success in obtaining Eu(DBM) 3 DETA is the utilization of DETA molecule as synergistic ligand, presenting block crystals with higher coordination number of Eu3+ ions via recrystallization. Due to the dense accumulation of cross-linked three-dimensional frameworks through van der Waals interactions, the fracture-induced piezoelectric effect results in charge separation and excitation through the resultant electric field and discharge, triggering a fast TL response of Eu(DBM) 3 DETA and expanding the possibilities of the quantitative stress sensing. Importantly, amorphous powders can still recover to their original PL and TL emission intensities after recrystallization in cyclic crystal-to-amorphous phase transitions. The unique PL and TL characteristics of Eu(DBM) 3 DETA provide promising opportunities to display stress visualization differences of electronic signatures under different forces. HSA (human serum albumin), a most abundant protein in blood serum, plays a key role in maintaining human health. Abnormal HSA level is correlated with many diseases, and thus has been used as an essential biomarker for therapeutic monitoring and biomedical diagnosis. Development of small-molecule fluorescent probes allowing the selective and sensitive recognition of HSA in in vitro and in vivo is of fundamental importance in basic biological research as well as medical diagnosis. Herein, we reported a series of new synthesized fluorescent dyes containing D-π-A constitution, which exhibited different optical properties in solution and solid state. Among them, dye M−H−SO 3 with a hydrophilic sulfonate group at electron-acceptor part displayed selectivity for discrimination of HSA from BSA and other enzymes. Upon binding of dye M−H−SO 3 with HSA, a significant fluorescence enhancement with a turn-on ratio about 96-fold was triggered. The detection limit was estimated to be ∼ 40 nM. Studies on the interaction mechanism revealed that dye M−H−SO 3 could bind to site III of HSA with a 1:1 binding stoichiometry. Understanding microglia development could improve our understanding of the central nervous system (CNS) and neurological diseases. To explore the immune phenotypic changes that occur in microglia during development, we studied the morphology, inflammatory response, and expression of several important immune-related proteins in normal microglia from the embryonic, neonatal (postnatal day 3), and adult stages. Results showed that implantation of microglia into the CNS until adulthood resulted in dynamic changes in the expression levels of CD11b (α chain of complement receptor 3) and CX3CR1 (a chemokine receptor), which were consistent and correlated. Expression of proinflammatory cytokines in microglia during development is dynamic and highest in perinatal period. The inflammatory response of microglia was more vigorous and intense in the neonatal microglia than in the adult microglia. Furthermore, the morphology and function of neonatal and adult microglia differed, and thus neonatal microglia cannot be used in lieu of adult microglia for functional studies. In this paper, we provide a mathematical framework in studying the wave propagation with the annihilation phenomenon in excitable media. We deal with the existence and uniqueness of solutions to a one-dimensional free boundary problem (called a reaction–interface system) arising from the singular limit of a FitzHugh–Nagumo type reaction–diffusion system. Because of the presence of the annihilation, interfaces may intersect each other. We introduce the notion of weak solutions to study the continuation of solutions beyond the annihilation time. [Display omitted] • Senescent astrocytes are distributed in the glymphatic and meningeal lymphatic routes. • Meningeal lymphatic routes contribute to clearance of senescent astrocytes from brain parenchyma. • Clearance of senescent astrocytes through meningeal lymphatic routes depends on VEGF-C/CCL21 pathway. Recent progress on the central lymphatic system has greatly increased our understanding of how the brain maintains its own waste homeostasis. Here, we showed that perivascular spaces and meningeal lymphatic vessels form a functional route for clearance of senescent astrocytes from the aging brain. Blocking meningeal lymphatic drainage by ligation of the deep cervical lymph nodes impaired clearance of senescent astrocytes from brain parenchyma, subsequently increasing neuroinflammation in aged mice. By contrast, enhancing meningeal lymphatic vessel diameter by a recombinant adeno-associated virus encoding mouse vascular endothelial growth factor-C (VEGF-C) improved clearance of senescent astrocytes and mitigated neuroinflammation. Mechanistically, VEGF-C was highly expressed in senescent astrocytes, contributing themselves to migrate across lymphatic vessels along C–C motif chemokine ligand 21 (CCL21) gradient by interacting with VEGF receptor 3. Moreover, intra-cisternal injection of antibody against CCL21 hampered senescent astrocytes into the lymphatic vessels and exacerbated short memory defects of aged mice. Together, these findings reveal a new perspective for the meningeal lymphatics in the removal of senescent astrocytes, thus offering a valuable target for therapeutic intervention. [Display omitted] Transition metals with catalytic properties were expected to improve the fire resistance of the polymer. In this work, NiMoO 4 nanorods decorated with Co-Ni layered double hydroxide (LDH) were fabricated and incorporated into polyurea (PUA) to improve the flame retardancy during combustion. Thermogravimetric analysis (TGA) showed that the addition of NiMoO 4 @Co-Ni LDH slightly decreased the initial degradation and the maximum degradation temperature. With increasing loading of NiMoO 4 @Co-Ni LDH, the char residue increased obviously. The combustion performance of PUA with 3.0 wt% NiMoO 4 @Co-Ni LDH investigated by cone calorimeter (CCT) showed that the peak heat release rate (PHRR), total heat release (THR) and smoke factor (SF) decreased by 34.41%, 12.68% and 48.65%, respectively. To efficiently remove high concentrations of hexavalent chromium (Cr(VI)), calcium–aluminum-layered double hydroxide (CaAl-LDH, denoted as CAL), and polypyrrole-modified CAL (CAL-PPy) were prepared by hydrothermal and in situ polymerization methods, respectively. The chemical structure, morphology, and elemental results indicated that the chain-like polypyrrole was decorated with hexagonal CAL. The specific surface area of CAL-PPy increased from 8.746 m2/g to 24.24 m2/g. The adsorption performances of CAL and CAL-PPy for aqueous Cr(VI) were investigated using batch equilibrium experiments. The decontamination process of aqueous Cr(VI) (100 mg/L) reached the equilibrium state within 50 min, and the kinetic data met the pseudo-second-order kinetic equation. The Langmuir model described the isothermal data properly, and the obtained theoretical adsorption capacity of CAL for Cr(VI) at 318 K was 34.06 mg/g, while that of CAL-PPy was 66.14 mg/g. The removal mechanisms involved electrostatic attraction, surface complexation, anion exchange, and reduction to low-toxicity Cr(III). Current Environmental, Social, and Governance (ESG) rating systems encounter challenges due to their subjectivity, time lag, and inconsistent coverage, which present significant obstacles for investors in the stock market. In contrast, news reporting data, characterized by its objectivity, timeliness, publicity, and sociality, has been a crucial source of quality market information for investors. The potential of ESG-related news as a robust complement to the data sources used in existing ESG rating systems merits further exploration. This study investigates the impact of ESG-related news on the Chinese stock market, with the goal of providing compelling evidence for integrating news media into ESG assessments. By analyzing 32,296 ESG-related news articles covering 3643 companies in the Chinese market from 2015 to 2021, we utilize event study methodology to assess the market's abnormal returns in response to ESG-related news. Our findings suggest a significant market impact from ESG-related news, indicating that investors are increasingly attentive to information pertaining to ESG issues. The study also reveals that different attributes of ESG-related news have diverse effects on the market. Specifically, news with negative sentiments has the most significant impact, and investors demonstrate a stronger reaction to ESG-related news disseminated through unofficial media. Furthermore, the study highlights that enterprise heterogeneity affects the impact of ESG-related news, with companies in the secondary and tertiary industries showing higher sensitivity. Privately-owned enterprises are found to be more influenced by such news compared to state-owned enterprises. Enhancing the green economy efficiency (GEE) is crucial for building a sustainable economy. How can the rapidly advancing digital transformation contribute to this process? The paper empirically examines the direct and spatial spillover effects of digital transformation on cities' GEE in China. This study utilizes the National E-commerce Pilot City (NEPC) policy as a quasi-natural experiment of regional digital transformation and employs the staggered difference-in-differences (DID) method with heterogeneous effects. The findings reveal that (i) implementing the NEPC policy significantly increases urban GEE by 2.6%, corresponding to a 16% increase in the mean of GEE. This effect is particularly pronounced in non-resource-based cities and cities with high Internet penetration. (ii) The mechanism test shows that the pilot policy positively affects GEE by promoting green structural transformation, enhancing green innovation, and strengthening public environmental concerns. (iii) The study highlights a positive spatial spillover effect of the NEPC policy on the GEE of nonpilot cities. (iv) The adoption of the NEPC plays a pivotal role in advancing energy use and carbon emission efficiency. We previously demonstrated that angiopoietin-like 8 (ANGPTL8) forms a localized complex with ANGPTL4 to reduce its lipoprotein lipase (LPL)-inhibitory activity and enable increased postprandial uptake of fatty acids (FA) into adipose tissue. Because prolonged cold exposure may increase adipose tissue FA uptake and decrease circulating triglycerides (TG) by reducing ANGPTL4 expression and inducing ANGPTL8 expression (and thus ANGPTL4/8 expression), we investigated the effect of temperature on ANGPTL4 and ANGPTL4/8 LPL-inhibitory activities in vitro. As the ANGPTL4(E40K) mutation results in decreased TG, we also characterized ANGPTL4(E40K) and ANGPTL4(E40K)/8 complex LPL-inhibitory activities. Interestingly, while ANGPTL3, ANGPTL3/8, and ANGPTL4 showed similar LPL inhibition at 37 °C and 22 °C, the already reduced LPL-inhibitory activity of ANGPTL4/8 at 37 °C was even more decreased at 22 °C. At 37 °C, ANGPTL4(E40K) manifested decreased LPL-inhibitory activity compared to ANGPTL4/8, while ANGPTL4(E40K)/8 had even further reduced potency. Remarkably, ANGPTL4/8, ANGPTL4(E40K), and ANGPTL4(E40K)/8 were each actually capable of stimulating LPL activity at 22 °C. Together, these results indicate that ANGPTL4/8 stimulation of LPL activity at low temperatures may represent an additional mechanism for further increasing adipose tissue FA uptake during cold exposure, beyond that already occurring due to decreased ANGPTL4 expression and increased ANGPTL8 expression. Ischemia-reperfusion injury (IRI) is a major cause of acute kidney injury (AKI), which can lead to poor outcome and increased risk of mortality. Dabrafenib (DAB) is an approved cancer treatment. Little is known about the effect of DAB in prevention or treatment of renal IRI. For in vivo experiments, C57BL/6 mice were divided into four groups: sham (no IRI, no DAB), IRI, DAB, and DAB + IRI. IRI was induced by clamping of bilateral renal pedicles for 30 min. For in vitro experiments, HK-2 cells were used to establish the hypoxia/reoxygenation (H/R) injury model, with four groups: control (no H/R, no DAB), H/R, DAB, and DAB + H/R. Renal function and renal histological changes were recorded. Expression of NGAL and KIM-1 proteins and mRNAs were determined by western blotting and qRT-PCR; secretion of inflammatory cytokines (IL-6 and TNF- α) was determined by qRT-PCR; Cell death was determined using the TUNEL assay, measurement of cleaved caspase-3, and flow cytometry. Necroptosis-related proteins were determined by western blotting. In mice, DAB pretreatment improved renal function and also reduced histological injury, inflammation, cell death, and expression of necroptosis-associated proteins. In HK-2 cells, DAB significantly decreased the levels of NGAL and KIM-1, inflammatory cytokines, cell death, and necroptosis-related proteins. Our in vitro and in vivo experiments indicated that DAB appears to alleviate renal IRI by suppressing cell death and inhibiting inflammatory responses. DAB has potential use for the clinical prevention and treatment of AKI-induced IRI. • We clarified dabrafenib could attenuate renal IRI in vivo and in vitro , which may by inhibiting RIP3 mediated necroptosis. • First, we found that dabrafenib could alleviate mouse renal IRI and supress the expression of necroptotic related proteins. • Then, we established the model of renal ischemia reperfusion injury by using HK-2 cells in vitro. • Our data showed that dabrafenib could decrease cell death and downregulate the level of necroptotic related proteins. • Our study suggests that DAB should be examined as a potential therapeutic option for the prevention or treatment of AKI. Cardiotoxicity is a well-known adverse effect of doxorubicin (Dox) administration, but the underlying molecular mechanism of this effect is not fully understood. Over the past two decades, considerable efforts have focused on the potential molecular targets of cardiotoxicity in the hope that novel targeted therapies will be generated to attenuate Dox-induced cardiotoxicity. Here, we provide a comprehensive overview of genetically modified animals that show enhanced or reduced susceptibility to the cardiotoxic effects of Dox. We focused on the process by which the molecules involved in DNA damage, oxidative stress, apoptosis, autophagy and necrosis are affected in the presence of Dox. We also present a protein-protein interaction network and explain the contribution of the components to the process of Dox-induced cardiotoxicity. More importantly, data from the literature have indicated that PI3Kγ and Rac1 are potential targets with therapeutic advantages in cancer therapy; molecules that target these proteins can simultaneously attenuate Dox-induced cardiotoxicity and enhance its anticancer activity. Organic pollutants are important harmful components in atmospheric fine particulate matters (PM 2.5), health risks of which varied with temporal and spatial distributions. To clarify the characteristics of atmospheric organic pollution, the concentrations, sources, and human health risks of typical organic compositions in PM 2.5 samples from both industrial and urban areas of Nanjing in eastern China were investigated monthly for a year. Results showed that, the concentrations of PM 2.5 -bound polycyclic aromatic hydrocarbons (PAHs) and n-alkanes were higher in winter and spring than those in summer and autumn. The organic pollution was slightly higher in industrial than urban area, though the PAHs in autumn and the n-alkanes in warm season (summer and autumn) were higher in urban area. With regards to the pollutant sources, the atmospheric PAHs were dominated by motor vehicle exhaust in the urban area, and combined with coal combustion emission in the industrial area. Airborne n-alkanes were mainly from biological source accompanied by fossil fuel combustion in industrial area. The PM 2.5 -bound PAHs indicated higher risks to adults in industrial area than in urban area with the seasonal patterns: winter > spring > autumn > summer. More attention should be paid to the health risks of exposure to organic pollutants accumulated in PM 2.5 during cold season. Since 2001, many Chinese provinces have transitioned from a "sequential" to a "parallel" school choice or college admissions mechanism. Inspired by this natural experiment, we evaluate the sequential (immediate acceptance, IA), parallel (PA), and deferred acceptance (DA) mechanisms in the laboratory. We find that participants are most likely to reveal their preferences truthfully under DA, followed by PA and then DA. While stability comparisons also follow the same order, efficiency comparisons vary across environments. Regardless of the metrics, the performance of PA is robustly sandwiched between IA and DA. Furthermore, 53% of our subjects adopt an insurance strategy under PA, making them at least as well off as what they could guarantee themselves under IA. Abstract Constant light exposure is widespread in the intensive care unit (ICU) and could increase the rate of brain dysfunction as delirium and sleep disorders in critical patients. And the activation of hypothalamic neuropeptides is proved to play a crucial role in regulating hypercatabolism, especially skeletal muscle wasting in critical patients, which could lead to serious complications and poor prognosis. Here we investigated the hypothesis that constant light exposure could aggravate skeletal muscle wasting in endotoxemia rats and whether it was associated with alterations of circadian clock and hypothalamic proopiomelanocortin(POMC) expression. Fifty-four adult male Sprague-Dawley rats were intraperitoneally injected with lipopolysaccharide(LPS) or saline, subjected to constant light or a 12:12 h light-dark cycle for 7 days. On day 8, rats were sacrificed across six time points in 24 h and hypothalamus tissues and skeletal muscle were obtained. Rates of muscle wasting were measured by 3-methylhistidine(3-MH) and tyrosine release as well as expression of two muscle atrophic genes, muscle ring finger 1(MuRF-1) and muscle atrophy F-box(MAFbx). The expression of circadian clock genes, silent information regulator 1(SIRT1), POMC and hypothalamic inflammatory cytokines were also detected. Results showed that LPS administration significantly increased hypothalamic POMC expression, inflammatory cytokine levels and muscle wasting rates. Meanwhile constant light exposure disrupted the circadian rhythm, declined the expression of SIRT1 as well as aggravated hypothalamic POMC overexpression and skeletal muscle wasting in rats with endotoxemia. The results indicated that the surface groups of the alginate and LDH were retained and so was the crystal structure in the alginate microsphere. The adsorption performance of the Fe 3 O 4 /LDH-AM for Cd2+, Pb2+, and Cu2+ in aqueous solutions was evaluated by batch and column adsorption experiments. The effects of adsorption conditions, kinetics, isotherms, mechanisms, and potential applications were investigated. The adsorption kinetic data conformed to the pseudo-second-order kinetic equation, and the isotherm data fit well with the Freundlich and Langmuir isotherm models. The adsorption mechanism of Cd2+, Pb2+, and Cu2+ by the Fe 3 O 4 /LDH-AM entailed complexation and precipitation. The experimental breakthrough curves were correlated with the Thomas model. Moreover, the Fe 3 O 4 /LDH-AM displayed superior regeneration and reusability. Abstract Objectives Aquaporin-2 (AQP2) is an important water channel protein that is expressed in the renal collecting duct and plays a key role in urine concentration and body water homeostasis. It has been demonstrated that the urinary excretion of AQP2 correlates strongly with its expression in the kidney in adult humans and rats. However, there have been no studies on the urinary excretion of AQP2 in human fetuses during development. Fetal urine is the main source of the amniotic fluid; we speculate that the level of AQP2 in the amniotic fluid could reflect the expression level of the AQP2 protein in the fetal kidney. The purpose of the present study was to explore the relationship between AQP2 in the amniotic fluid and that in the fetal kidney. Methods In the present study, the concentration of the AQP2 protein in human amniotic fluid was measured by Enzyme-linked immunosorbent assay (ELISA) and its expression level in human fetal kidneys were examined by wastern blot and immunohistochemistry. Results Both the expression level of AQP2 in the fetal kidney (F = 195.9, P < 0.001) and the concentration of AQP2 in the amniotic fluid increased with gestational age (F = 1098, P < 0.001). Moreover, the concentration of AQP2 in the amniotic fluid was positively correlated with its expression level in the fetal kidney (r = 0.872, P < 0.0001). Abstract In this work, Zn 1-x Sn x O nanostructures with diverse morphological characters were prepared by hydrothermal technique. The morphology, structure, crystallization and the chemical states of the samples were characterized by scanning electron microscopy (SEM), X-ray diffraction (XRD) and X-ray photoelectron spectroscopy (XPS). The characterization of UV–Vis spectra indicates that the optical transmittance of Zn 1-x Sn x O nanostructure is above 90% in the visible range. The optical absorption edge of the samples was determined according to the Tauc relationship, and a remarkable bule shift of the band-gap appeared for the Zn 1-x Sn x O nanomaterials. The fluorescence spectra were measured by using photoluminescence (PL) spectrometer. The pure ZnO nanostructure exhibits a strong yellow emission and a weak near-ultraviolet emission. The effect of CTAB is mainly on the photoluminescence intensity rather than the peak position. However, the introduction of stannum has an interesting effect on the PL of ZnO. The Zn 1-x Sn x O nanostructure exhibits a remarkable near-band-edge (NBE) emission with two peaks. The dependence of NBE emission spectra on excitation power was analyzed. Two radiative recombination channels of the NBE emission have been discussed in detail. Glioblastoma multiforme (GBM) is the most common and aggressive brain tumor with a poor prognosis. The growth of GBM cells depends on the core transcriptional apparatus, thus rendering RNA polymerase (RNA pol) complex as a candidate therapeutic target. The RNA pol II subunit B (POLR2B) gene encodes the second largest subunit of the RNA pol II (RPB2); however, its genomic status and function in GBM remain unclear. Certain GBM data sets in cBioPortal were used for investigating the genomic status and expression of POLR2B in GBM. The function of RPB2 was analyzed following knockdown of POLR2B expression by shRNA in GBM cells. The cell counting kit-8 assay and PI staining were used for cell proliferation and cell cycle analysis. A xenograft mouse model was established to analyze the function of RPB2 in vivo. RNA sequencing was performed to analyze the RPB2-regulated genes. GO and GSEA analyses were applied to investigate the RPB2-regulated gene function and associated pathways. In the present study, the genomic alteration and overexpression of the POLR2B gene was described in glioblastoma. The data indicated that knockdown of POLR2B expression suppressed tumor cell growth of glioblastoma in vitro and in vivo. The analysis further demonstrated the identification of the RPB2-regulated gene sets and highlighted the DNA damage-inducible transcript 4 gene as the downstream target of the POLR2B gene. We compare the performance of the Boston Immediate Acceptance (IA) and Gale–Shapley Deferred Acceptance (DA) mechanisms in a laboratory setting where we increase the number of participants per match. In our experiment, we first increase the number of students per match from 4 to 40; when we do so, participant truth-telling increases under DA but decreases under IA, leading to a decrease in efficiency under both mechanisms. Furthermore, we find that DA remains more stable than IA, regardless of scale. We then further increase the number of participants per match to 4,000 through the introduction of robots. When robots report their preferences truthfully, we find that scale has no effect on human best response behavior. By contrast, when we program the robots to draw their strategies from the distribution of empirical human strategies, we find that our increase in scale increases human ex-post best responses under both mechanisms. The Dio2 / 3 gene is related to the photoperiodic response in mammals and plays an important role in the development of gonadal organs and seasonal breeding. Our previous studies have reported synchronous variations in the gonadal mass and photoperiodical transition around the summer solstice in a wild Brandt’s vole population, a species with striking seasonal breeding. To investigate the role of the Dio2 / 3 gene in the control of seasonal breeding in this species, we cloned and characterized its expression levels by high-throughput Real-Time PCR during the period around the summer solstice. We selected a domesticated strain to ensure similar development of samples. The synchronous variation pattern between the Dio2 / 3 expression levels and gonadal mass around the summer solstice supports the prediction that the Dio2 / 3 gene plays an important role in the seasonal transition in this species. We suggest that the observed photoperiod response may be triggered by differences in the day length rather than the absolute daylength in this species. However, the similar Dio2 / 3 gene expression patterns but inconsistent gonadal mass patterns between the domesticated strain and the wild strain in the samples collected on Sep 8th, an absolute nonbreeding stage in the wild, lead us to speculate that the core function of the Dio2 / 3 gene should be restricted in response to the photoperiod rather than factors directly regulating gonadal development, and this laboratory strain could be used as an animal model to test the mechanism of environmental adaptation. Objective To establish a high throughput, low cost, and simple nanotechnology-based method for the detection of single nucleotide polymorphism (SNP) loci in type 2 diabetes mellitus (T2DM). Methods Multiplex ligase detection reaction (LDR) amplification was performed using fluorescently labeled magnetic nanosphere-bound upstream LDR probes and downstream probes labeled with a unique fluorescent group for each SNP locus. The amplified LDR products were separated by magnetic nanospheres and then scanned by fluorescence spectroscopy. Four SNP loci associated with T2DM were detected, including the rs13866634 locus in SLC30A8, rs10811661in CDKN2A/2B, rs1111875 in the HHEX gene, and rs7903146 in the TCF7L2 gene. The SNP genotype was also determined by DNA sequencing as a control. Results The SNP genotypes of the four gene loci determined by the nanosphere-based multiplex LDR method were consistent with the DNA sequencing results. The accuracy rate was 100%. Myostatin (Mstn) is a negative regulator of muscle development in vertebrates. Although its function in muscle growth has been well studied in mammals and fish, it remains unclear whether or how mstn functions in the immune system. In this study, mstna −/- and mstnb −/- homozygous zebrafish were firstly generated using CRISPR/Cas9 (Clustered regularly interspaced short palindromic repeats/CRISPR associated protein 9). Deletion of mstnb but not mstna enhanced growth performance. Although survival rates under normal conditions were slightly decreased in both strains, mortality after dexamethasone-induced stress was increased by ∼30%. Furthermore, transcriptional levels of several critical immune-related genes were decreased, and the ability to withstand exposure to pathogenic E. tarda was decreased, compared with that of controls. In mstnb −/- but not mstna −/- zebrafish, expression of NF-κB subunits and several pro-inflammatory cytokines failed to respond to E. tarda exposure except nfkb1 , c-rel and tnfα . Taken together, these results indicate that mstnb but not mstna plays a key role in zebrafish muscle growth. While each paralogue contributes to the response to bacterial insult, mstnb affects the immune system through activation of the NF-κB pathway, and mstna is likely to act upstream of NF-κB at some as yet unidentified target. Current strategies for influenza virus vaccines primarily aim to elicit immune responses towards the globular head domain of the hemagglutinin (HA) protein so that binding of the virus to membrane receptors on the host cells is inhibited. In the present study, we show a novel strategy to generate immunity against the highly conserved region of the influenza virus. The globular head domain was replaced by different linkers to generate a headless HA (stalk domain) and then coexpressed with influenza M1 proteinin Tni insect cells. The expression was validated by western blot analysis, and stalk domain with peptides (GGGGS)4 linkers was identified to anchor in a stable way to the cell membrane. An immunoelectron microscope showed that stalk domain with (GGGGS)4 linkers were steadily incorporated to the surface of influenza virus-like particles (VLPs). Mice immunized with these VLPs exhibited enhanced systemic antibody responses with increased binding avidity and study found high titers of ADCC antibodies to the influenza virus, these VLPs also induced mucosal immune responses and produced antigen-specific IgG and IgA in nasal and lung washes. In addition, antigen-specific IgG antibody-secreting cells (ASCs) increased significantly in the spleen and lymph node. A great deal of evidence highlighted the pathophysiologic importance of TGF-β1/Smad3 pathway in masangial extracellular matrix (ECM) accumulation, but some alternative signaling pathways are also involved. TGF-β was shown recently to induce rapid and transient epidermal-like growth factor receptor (EGFR) transactivation and subsequent fibronectin expression via heparin-binding epidermal-like growth factors (HB-EGF) release and binding in mesangial cells, which is independent of Smad2 activation. However, whether TGF-β could induce persistent EGFR transactivation remains to be identified. The present study demonstrates that in addition to transient EGFR transactivation, TGF-β1 can also induce continuous EGFR transactivation by a non-ligand–dependent pathway in rat mesangial cells. This sustained EGFR transactivation is mainly due to Src kinase-mediated persistent EGFR tyrosine phosphorylation at Y845 rather than Y1173. TGF-β1–induced early Smad3 phosphorylation is independent of transient EGFR transactivation and ERK1/2 activation initiated by HB-EGF release, whereas Src–mediated chronic EGFR transactivation and ERK1/2 activation participate in Smad3 activation in a relatively modest and delayed manner. Therefore, the present study further clarifies the mechanisms of EGFR transactivation in the TGF-β–initiated ECM upregulation and raises the possibility that targeting EGFR may provide a viable alternative strategy for inhibiting TGF-β in chronic kidney disease. Extranodal NK/T-cell lymphoma (ENKTL), nasal type is an aggressive lymphoma characterized by rapid clinical progression, an unfavorable prognosis, and a short survival time. This study aimed to investigate the correlations of c-MYC protein expression, gene rearrangement, and gene copy number with the prognosis of ENKTL. Immunohistochemistry for CD20, CD3, CD56, TIA-1, Ki-67, and c-MYC were performed using tissue sections from 68 patients diagnosed with ENKTL. In situ hybridization was performed to detect Epstein-Barr virus (EBV)-encoded small RNA (EBER). c-MYC genetic alterations (located on chromosome 8q24) were detected using fluorescence in situ hybridization. Sixty patients with nasopharyngeal mucosa lymphadenosis were selected as a normal control group for the c-MYC gene and protein. Immunophenotypically, CD3 and TIA-1 were positive in all patients, CD56 was positive in 54 patients (79.4%), and CD20 was negative in all patients. A Ki-67 proliferation index of > 50% was observed in 94.1% of patients (64/68). In situ hybridization for EBER was positive in all patients. The c-MYC positive expression rate (> 40% of tumor cells with nuclear staining regarded as positive) in patients with ENKTL was 50% (34/68), which was significantly higher than that in patients with nasopharyngeal mucosa lymphadenosis (0%, 0/60; p < 0.05). Treatment efficacy was poor in patients with high c-MYC protein expression ( p < 0.05). Genetically, c-MYC rearrangements were not detected in any patients, but 26.5% (18/68) had increased c-MYC copy numbers. The gain of c-MYC copy numbers was positively correlated with c-MYC protein expression ( p < 0.05). Kaplan-Meier analysis illustrated that overall survival was significantly shorter in the c-MYC-positive group than in the c-MYC-negative group ( p < 0.05). Gain of c-MYC copy numbers was not associated with prognosis ( p > 0.05). Multivariate Cox regression analysis further confirmed that clinical stage and c-MYC positivity were independent prognostic factors in patients with ENKTL. We investigate the effects of team competition on pro-social lending activity on Kiva.org , the first microlending website to match lenders with entrepreneurs in developing countries. Using naturally occurring field data, we find that lenders who join teams contribute 1.2 more loans ($30–$42) per month than those who do not. To further explore factors that differentiate successful teams from dormant ones, we run a large-scale randomized field experiment ( n = 22 , 233 ) by posting forum messages. Compared to the control, we find that lenders make significantly more loans when exposed to a goal-setting and coordination message, whereas goal-setting alone significantly increases lending activities of previously inactive teams. We study a diffusive predator–prey model of Lotka–Volterra type functional response in which both species obey the logistic growth such that the carrying capacity of the predator is proportional to the prey population and the one for prey is a constant. Both continuous and discrete diffusion are addressed. Our aim is to see whether both species can survive eventually, if an alien invading predator is introduced to the habitat of an existing prey. The answer to this question is positive under certain restriction on the parameter. Applying Schauder's fixed point theory with the help of suitable upper and lower solutions, the existence of traveling wave solutions for this model is proven. Furthermore, by deriving the non-existence of traveling wave solutions, we also determine the minimal speed of traveling waves for this model. This provides an estimation of the invasion speed. Decades of intensifying human activities have caused dramatic changes in land use and land cover (LULC) in the ecologically fragile areas of the Qinghai-Tibet Plateau, which have led to significant changes in ecosystem service value (ESV). Taking the ecologically fragile Sanjiangyuan region of the Qinghai-Tibet Plateau as the research object, we focused on understanding the impact of LULC changes on the Sanjiangyuan's landscape pattern and its corresponding ESV, which was combined with a Markov-Plus model to predict LULC changes in 2030. The results showed: (1) from 2000 to 2020, the LULC of Sanjiangyuan has changed to varying degrees, respectively. In the central and southern regions where animal husbandry is the mainstay activity, the area of grass land converted to bareland had expanded; (2) from 2000 to 2010, the total regional ESV increased sharply. However, the total amount of ESV decreased from 2010 to 2020; (3) the overall ESV in the study area was observed to be trending down and is expected to decrease by approximately 4.25 billion CNY by 2030; (4) the fragmentation and complexity of regional landscape patterns will negatively affect local ecosystem stability and biodiversity. Overall, there is a strong temporal and spatial correlation between LULC and ESV. The alkaline-earth silicate Sr 3 MgSi 2 O 8 :Ce 3+ ,Tb 3+ phosphors have been synthesized via conventional high temperature solid-state method. The crystal structure, luminescence properties, morphology of phosphors are investigated systematically. The thermal quenching and concentration quenching mechanisms of phosphors are also discussed. The phosphors can be excited under near ultraviolet light ranging from 200 nm to 450 nm. The calcination temperature and calcination time are significant factors for luminescence property of phosphors. Increasing co-doping Tb 3+ makes the emission intensity of Ce 3+ decreases proportionally while the emission of Tb 3+ increases. It indicates that it exists energy transfer between Ce 3+ and Tb 3+ . The emission wavelength can be tuned from bluish violet region to blue region. The results imply Sr 3 MgSi 2 O 8 :Ce 3+ ,Tb 3+ phosphors can be a potential candidate for solid-state lighting. The crapemyrtle bark scale (CMBS), Acanthococcus lagerstroemiae, is an important pest of crapemyrtles, Lagerstroemia spp. Biopesticides are frequently used to control soft-bodied insects; however, there is no information on their efficacy against CMBS. The objectives of this study were to test the use of selected biopesticides against CMBS under different seasons, evaluate their pathogenicity to CMBS, and determine optimum temperature for spore germination. Commercial formulations of the fungal biopesticides Isaria fumosorosea PFR97, Beauveria bassiana ANT-03, and the bacterial biopesticides Burkholderia rinojensis A396 and Chromobacterium subtsugae PRAA4-1 T were tested in greenhouse settings and did not have significant treatment effect on immature mortality. Biopesticides containing I. fumosorosea PFR97, B. bassiana ANT-03, and B. bassiana GHA were applied to mature crapemyrtles grown in the landscape and naturally infested with CMBS, and the scale survivorship was determined in two trials at different seasons. When treatments were applied in winter, scale survivorship was significantly lower in the B. bassiana ANT-03 treatment compared to the control. When treatments were applied in spring, no treatment effect was observed. During another trial conducted in autumn using container-grown crapemyrtles, B. bassiana GHA treatment reduced scales survivorship when compared to the control. Pathogen re-isolation assays confirmed that scales collected in the spring trial were had all pathogens delivered; in addition, B. bassiana ANT-03 was the most recovered pathogen. In a temperature-dependent spore germination assay, the optimum temperature range for I. fumosorosea PFR97, B. bassiana ANT-03, and B. bassiana GHA was between 22.5 and 30 °C, with peak spore germination observed at 28 °C for all three pathogens. Results from this project suggest that biopesticides containing strains of B. bassiana can be included in the toolbox for CMBS control, provided that climate and product coverage are considered. A high-resolution nutrient emission inventory can provide reliable and accurate identification of priority control areas, which is crucial for efficient decisions on water quality restoration. However, the inventories widely used in large-scale modeling are usually based on provincial inputs, which induce the challenges of lacking localized parameters and missing localized characteristic when provincial scale inputs are converted to finer scales with the down-scale methods. Based on elaborate investigations and statistical data at the county scale with multi-scale data conversion, the China Emission Inventory of Nutrients (CEIN) was developed with a spatial resolution of a 0.1° grid and sub-basin scales. The Yangtze River Basin was used as a case study to illustrate the potential applications of CEIN. The emissions of total nitrogen (TN) and total phosphorus (TP) of Yangtze River Basin is 0.43 Mt and 0.04 Mt for point sources, 11.09 Mt and 4.64 Mt for diffuse sources in 2017. The hotspot analysis for 2606 sub-basins indicated that cropland is the key source of nutrient emissions, accounting for 58.88% and 79.15% of TN and TP, respectively. Industrial sewage and freshwater aquaculture accounted for 27.39% (TN) and 21.98% (TP) of the point sources, which is substantial due to their direct discharge into surface waters. The current results also reveal that, in contrast to CEIN, the previously used common emission factors based on GDP per capita produced considerable overestimations of 2.37 and 2.65 times the actual TN and TP emissions, respectively. Additional advantages of the CEIN have been demonstrated in identifying priority control areas more accurately with reduced bias and quantifying the effects of policies at much smaller scales. For example, the CEIN helps to distinguish hotspots, which was neglected when identifying sources at the level-III sub-basin scale, and indicates that the management of fractional areas (TN: 16.97%; TP: 13.44%) provides the highest nutrient emissions control (TN: 44.34%; TP: 48.65%) for the entire basin. The evaluation of China's toilet revolution policy demonstrates that achieving equitable access to safe sanitation has resulted in a reduction of 7240 t of TN and 833 t of TP, which is extremely critical for rural water quality and health. Objective: By analyzing the cortical activation and functional connectivity of the prefrontal cortex (PFC) during dual-task obstacle negotiation in the older adults, cognitive resources allocation and neural regulatory mechanisms of aging brain were shed light on in complex walking conditions.Methods: Twenty-eight healthy right-handed subjects participated in the study, including 15 men and 13 women (age: 68.6 ± 4.1 years, height: 162.96 ± 6.05 cm, weight: 63.63 ± 9.64 kg). There were four tasks: Normal Walk (NW), Obstacle Negotiation during Normal Walk (NW + ON), Walk while performing Cognitive Task (WCT), and Obstacle Negotiation during Walk while performing Cognitive Task (WCT + ON). Participants wore functional near-infrared spectroscopy (fNIRS) to collect hemodynamic signals from various regions of interest (ROIs) in the PFC, while the three-dimensional motion capture system was used to test the gait velocity. Cognitive task data was recorded.Results: In WCT + ON, the HbO2 concentration change value (△HbO2) of the left dorsolateral prefrontal cortex was significantly greater than that in the other three tasks (p < 0.05), and the△HbO2 of the right dorsolateral prefrontal cortex was significantly greater than that in NW + ON (p < 0.05). The gait velocities in the four tasks were significantly different (p < 0.05) (NW > WCT > NW + ON > WCT + ON). There was no significant difference in cognitive performance between in the WCT and WCT + ON (p > 0.05). In WCT + ON, the left and right dorsolateral prefrontal areas had strong functional connectivity and the left frontal pole was most widely connected to the other ROIs. Compared to that in NW, the functional connectivity of the left prefrontal lobe was significantly enhanced in WCT + ON (p < 0.05).Conclusions: As walking difficulty increased, the PFC activation in the older adults changed from right-sided to bilateral activation, indicating that the left PFC cognitive resources compensated for the right PFC in dual-task obstacle negotiation. The cognitive resources recruitment in dual-task obstacle negotiation might be achieved by synchronization and coordination of associated brain areas in the PFC, primarily to maintain dynamic postural balance when walking. Bronchopulmonary dysplasia (BPD) is a common chronic lung disease in premature infants, and its pathogenesis has not been clarified. Long non-coding RNAs (lncRNA) have important functions in cell bioactivity. However, their role in developmental lung disease remains unclear. The aim of this study was to demonstrate the role of lncRNA SNHG6 (SNHG6) in BPD and its underlying mechanisms. The blood of patients with BPD were collected, and BPD model of BEAS-2B cells was established by hyperoxia method. SNHG6, miR-335 and KLF5 mRNA expression were detected by RT-qPCR. Western blot was conducted to measure the levels of apoptosis-related proteins' expression and NF-κB pathway related proteins. BEAS-2B cell viability and apoptosis were assessed by CCK-8 and flow cytometry, respectively. Assay Kit was applied to detect ROS, MDA and SOD levels, respectively. ELISA was performed to assess the levels of inflammatory factors. The binding site of miR-335 with SNHG6 or KLF5 were predicted by using DIANA or TargetScan, and which was verified by double luciferase reporter assay. Firstly, SNHG6 was highly expressed and miR-335 was lowly expressed in BPD model, SNHG6 knockdown and miR-335 mimics both alleviated hyperoxia-induced lung cell injury, and SNHG6 targeted miR-335. Subsequently, KLF5 was targeted by miR-335, and KLF5 promoted lung cell injury via activating NF-κB pathway. Furthermore, SNHG6 mediated lung cell injury via regulating the miR-335/KLF5/NF-κB pathway. Our research confirmed that SNHG6 mediated hyperoxia-induced lung cell injury via regulating the miR-335/KLF5/NF-κB pathway. Insect predators are the most important natural enemies of the crapemyrtle bark scale (CMBS) in the USA. Mycopesticides (biocontrol fungi) are considered an IPM tool to increase CMBS mortality; however, their impacts on CMBS predators are unknown. The objectives of this study were to assess the abundance and diversity of CMBS natural enemies in Louisiana; evaluate the impacts of mycopesticides on survival of CMBS predators by life stage; and determine if entomopathogenic spores delivered to crapemyrtles are transferred to predators under field conditions. The mycopesticides Ancora® (Isaria fumosorosea PFR97), BioCeres® (Beauveria bassiana ANT-03), and BotaniGard® (B. bassiana GHA) were tested against the coccinellids Chilocorus spp. and Hyperaspis bigeminata under laboratory and field conditions. Adults and larvae of the coccinellids were treated with each mycopesticide and survival recorded over a 14-day period. The most common natural enemies on CMBS infested trees were the coccinellids Chilocorus cacti, C. stigma , and Hyperaspis bigeminata (Coleoptera: Coccinellidae). In laboratory bioassays BotaniGard® reduced survival of adults and larvae of both genera by at least 57%. BioCeres® reduced the survival of Chilocorus spp. adults by 40% and Ancora® reduced survival of H. bigeminata larvae by 69%. Under field conditions, CMBS infestations were sprayed with the mycopesticides and coccinellids were collected every other day for a two-week period. Spores of the applied mycopesticides were recovered from the coccinellids; however, it is not known if infection occurred in the field trial or spores were delivered to CMBS infestations by the coccinellids. One of the key limitations of the many existing visual tracking method is that they are built upon low-level visual features and have limited predictability power of data semantics. To effectively fill the semantic gap of visual data in visual tracking with little supervision, we propose a tracking method which constructs a robust object appearance model via learning and transferring mid-level image representations using a deep network, i.e., Network in Network (NIN). First, we design a simple yet effective method to transfer the mid-level features learned from NIN on the source tasks with large scale training data to the tracking tasks with limited training data. Then, to address the drifting problem, we simultaneously utilize the samples collected in the initial and most previous frames. The LIM-homeodomain (LIM-HD) family member Lmx1a has been successfully used to induce dopaminergic neurons from other cell types, thus showing significant implications in replacement therapies of Parkinson's disease, but the underlying mechanism remains elusive. In this study, we used Drosophila eye as a model system to investigate how forced expression of dLmx1a , the fly homolog of human Lmx1a , alters cell identify. We found that ectopic expression of dLmx1a suppresses the formation of Drosophila eye tissue and identified the LIM and HD as two essential domains. dLmx1a requires and physically binds to Chip, a well-known cofactor of LIM-HD proteins. Chip connects two dLmx1a proteins to form a functional tetrameric complex. In addition, we provide evidence showing that dLmx1a expression results in the suppression of two retina determination gene eyes absent ( eya ) and string ( stg ). Taken together, our findings identified Chip as a novel partner of dLmx1a to alter cell differentiation in Drosophila eye through repressing eya and stg expression, and provide an animal model for further understanding the molecular mechanism whereby Lmx1a determines cell fate. Social segregation is a ubiquitous feature of human life. People segregate along the lines of income, religion, ethnicity, language, and other characteristics. This study provides the first experimental examination of decentralized matching with search frictions and institutionalized segregation. The findings indicate that, without a segregation institution, high types over-segregate relative to the equilibrium prediction. We observe segregation attempts even when equilibrium suggests that everyone should accept everyone else. In the presence of a segregation institution, we find that, while the symmetric segregation institution increases matching success rate and efficiency in one environment, it has weak or no effect in a steep-incentive environment. By adding an entry cost to a flat-incentive market, however, the asymmetric segregation institution leads to an increased matching success rate and efficiency in both environments, which underscores the importance of a coordination device. Transcription factor FOXM1 plays a critical role in maintenance of stem cell pluripotency through stimulating the transcription of pluripotency-related genes in mouse pluripotent stem cells. In this study, we have found that the repression of FOXM1 expression is mediated by FOXM1 3′UTR during retinoic acid-induced differentiation of human pluripotent NT2/D1 embryonal carcinoma cells. FOXM1 3′UTR contains a microRNA response element (MRE) for miR-134, which has been shown to attenuate the expression of pluripotency-related genes post-transcriptionally during mouse embryonic stem cell differentiation. We have determined that miR-134 is induced during RA-induced differentiation of NT2/D1 cells and the overexpression of miR-134 represses the expression of FOXM1 protein but not FOXM1 mRNA. Furthermore, the expression of OCT4 is diminished by FOXM1 knockdown and the OCT4 promoter is regulated directly by FOXM1, suggesting that FOXM1 is required for maintaining the expression of OCT4 in NT2/D1 cells. Background : Tongue diagnosis, as a unique method of traditional Chinese medicine (TCM), discriminates physiological functions and pathological conditions by observing the changes of the tongue coating. Aims : To evaluate the differences of tongue images and tongue coating microbiome between patients with colorectal cancer and healthy people. Methods : The tongue diagnostic information acquisition system was used to photograph the tongue images and analyze the thickness of the tongue coatings in patients with colorectal cancer and healthy people. The next-generation sequencing technology was used to determine the V2–V4 hypervariable region of 16S rDNA to investigate the microbial community structure and diversity on the tongue coating. Results : The tongue coatings in patients with colorectal cancer were obvious thickening compared with tongue images in healthy people. The microbial community structure on the tongue coating was different between patients with colorectal cancer and healthy people. Conclusion : Tongue diagnosis may provide important leads towards novel microbiome-related diagnostic tools and tongue coating microbiome may be a novel biomarker for characterizing patient with colorectal cancer. Our previous study identified five new heat shock factor 4 (HSF4) mutations in 150 age-related cataract (ARC) patients which indicated that HSF4 mutations may be associated with this disease. Hypoxia-inducible factor (Hif1α) is an important downstream target of HSF4b. It has been found that Hif1α play also important roles in cataract development. To identify if HSF4b play it role in cataract development through HIF1α, we transfected SRA01/04 lens epithelial cells with small hairpin RNA of HSF4b and measured expressions Hif1α after transfection. Then, we perform chromatin immunoprecipitation quantitative PCR to see the relationship between HSF4b and HIF1α. We found that HSF4 downregulation led to decrease of HIF1α mRNA expression. Furthermore, we demonstrated by ChIP followed by quantitative PCR (ChIP-qPCR) that these HIF-1α is bound by HSF4b near promoters, not gene bodies. [ABSTRACT FROM AUTHOR]

Cylindromatosis (CYLD), a deubiquitinase and regulator of microtubule dynamics, has important roles in the regulation of inflammation, immune response, apoptosis, mitosis, cell migration and tumorigenesis. Although great progress has been made in the biochemical and cellular functions of CYLD, its role in animal development remains elusive. In this study, we identified Drosophila CYLD (dCYLD) as a negative regulator of the Hippo pathway in vivo. dCYLD associates and colocalizes with Hpo, a core component of the Hippo pathway, in the cytoplasm, and decreases Hpo activity through limiting its phosphorylation at T195. We also showed that dCYLD limits Hippo signal transduction as evidenced by decreasing phosphorylation and thereby increasing activity of Yki, the key downstream effector of the Hippo pathway. Background Oxidative stress and inflammation play important roles in the development of diabetes. Metformin (MET) is considered as the first-line therapy for patients with type 2 diabetes (T2D). Hypothalamic paraventricular nucleus (PVN) and hypothalamic arcuate nucleus (ARC) are vital in obesity and diabetes. However, there have been few studies on the effects of MET on inflammatory reaction and oxidative stress in the PVN and ARC of T2D diabetic rats. Methods Male Sprague-Dawley (SD) rats were fed with high-fat diet (HFD), and intraperitoneally injected with low-dose streptozotocin (STZ, 30 mg/kg) at 6th week to induce T2D diabetes. After injection of STZ, they were fed with HFD continually. Starting from the 8th week of HFD feeding, T2D rats received intragastrical administration of MET (150 mg/kg/day) in addition to the HFD for another 8 weeks. At the end of the 15th week, the rats were anaesthetized to record the sympathetic nerve activity and collect blood and tissue samples. Results In comparison with control rats, T2D diabetic rats had higher levels of pro-inflammatory cytokines (PICs) and excessive oxidative stress in the PVN and ARC, accompanied with more activated astrocytes. The renal sympathetic nerve activity (RSNA) and the plasma norepinephrine (NE) increased in T2D diabetic rats. The expression of tyrosine hydroxylase (TH) increased and the expression of 67-kDa isoform of glutamate decarboxylase (GAD67) decreased in T2D diabetic rats. Supplementation of MET decreased blood glucose, suppressed RSNA, decreased PICs (TNF-α, IL-1β and IL-6) in PVN and ARC, attenuated oxidative stress and activation of astrocytes in ARC and PVN of T2D diabetic rats, as well as restored the balance of neurotransmitter synthetase. The number of Fra-LI (chronic neuronal excitation marker) positive neurons in the ARC and PVN of T2D diabetic rats increased. Chronic supplementation of MET also decreased the number of Fra-LI positive neurons in the ARC and PVN of T2D diabetic rats. Abstract: APOBEC3G belongs to a family of DNA cytosine deaminases that are involved in the restriction of a broad number of retroviruses including human immunodeficiency virus type 1 (HIV-1). Prior studies have identified two distinct mechanistic steps in Vif-deficient HIV-1 restriction: packaging into virions and deaminating viral cDNA. APOBEC3A, for example, although highly active, is not packaged and is therefore not restrictive. APOBEC3G, on the other hand, although having weaker enzymatic activity, is packaged into virions and is strongly restrictive. Although a number of studies have described the propensity for APOBEC3 oligomerization, its relevance to HIV-1 restriction remains unclear. Here, we address this problem by examining APOBEC3 oligomerization in living cells using molecular brightness analysis. We find that APOBEC3G forms high-order multimers as a function of protein concentration. In contrast, APOBEC3A, APOBEC3C and APOBEC2 are monomers at all tested concentrations. Among other members of the APOBEC3 family, we show that the multimerization propensities of APOBEC3B, APOBEC3D, APOBEC3F and APOBEC3H (haplotype II) bear more resemblance to APOBEC3G than to APOBEC3A/3C/2. Prior studies have shown that all of these multimerizing APOBEC3 proteins, but not the monomeric family members, have the capacity to package into HIV-1 particles and restrict viral infectivity. This correlation between oligomerization and restriction is further evidenced by two different APOBEC3G mutants, which are each compromised for multimerization, packaging and HIV-1 restriction. Overall, our results imply that multimerization of APOBEC3 proteins may be related to the packaging mechanism and ultimately to virus restriction. [Copyright &y& Elsevier]
