Rhizosphere microbiota are an important factor impacting plant uptake of pollutants. However, little is known about how microbial nitrogen (N) transformation in the rhizosphere affects the uptake and accumulation of antibiotics in plants. Here, we determined recruitment of N transformation functional bacteria upon ciprofloxacin (CIP) exposure, by comparing differences in assembly processes of both rhizospheric bacterial communities and N transformation between two choysum ( Brassica parachinensis ) varieties differing in CIP accumulation. The low accumulation variety (LAV) of CIP recruited more host bacteria (e.g., Nitrospiria and Nitrolancea ) carrying nitrification genes (mainly nxrA ) but fewer host bacteria carrying denitrification genes, especially narG , relative to the high accumulation variety (HAV) of CIP. The nxrA and narG abundance in the LAV rhizosphere were, respectively, 1.6-7.8 fold higher and 1.4-3.4 fold lower than those in the HAV rhizosphere. Considering that nitrate can decrease CIP uptake into choysum through competing for the proton motive force and energy, such specific bacteria recruitment in LAV favored the production and utilization of nitrate in its rhizosphere, thus limiting its CIP accumulation with 1.6-2.4 fold lower than the HAV. The findings give insight into the mechanism underlying low pollutant accumulation, filling the knowledge gap regarding the profound effects of rhizosphere microflora and N transformation processes on antibiotic accumulation in crops.

Microcystins (MCs) are hepatotoxic heptapeptides identified in cyanobacterial bloom-impacted waters and soils. However, their environmental fate in soils is poorly understood, preventing reliable site assessment. This study aims to clarify the variant-specific adsorption, desorption, and dissipation of MC-LR and MC-RR in agricultural soils. Results revealed that their adsorption isotherms followed the Freundlich model ( R 2 ≥ 0.96), exhibiting a higher nonlinear trend and lower adsorption capacity for MC-LR than for MC-RR. The soils had low desorption rates of 8.14-21.06% and 3.06-34.04%, respectively, following a 24 h desorption cycle. Pairwise comparison indicated that soil pH and clay played key roles in MC-LR adsorption and desorption, while organic matter and cation exchange capacity played key roles in those of MC-RR. MC-LR dissipation half-lives in soils were 27.18-42.52 days, compared with 35.19-43.87 days for MC-RR. Specifically, an appreciable decrease in MC concentration in sterile soils suggested the significant role of abiotic degradation. This study demonstrates that the minor structural changes in MCs might have major effects on their environmental fates in agricultural soil and indicates that the toxic effects of MCs should be of high concern due to high adsorption, low desorption, and slow dissipation.

Perfluorooctanesulfonate (PFOS) as an accumulative emerging persistent organic pollutant in crops poses severe threats to human health. Lettuce varieties that accumulate a lower amount of PFOS (low-accumulating crop variety, LACV) have been identified, but the regarding mechanisms remain unsolved. Here, rhizospheric activation, uptake, translocation, and compartmentalization of PFOS in LACV were investigated in comparison with those of high-accumulating crop variety (HACV) in terms of rhizospheric forms, transporters, and subcellular distributions of PFOS. The enhanced PFOS desorption from the rhizosphere soils by dissolved organic matter from root exudates was observed with weaker effect in LACV than in HACV. PFOS root uptake was controlled by a transporter-mediated passive process in which low activities of aquaporins and rapid-type anion channels were corrected with low expression levels of PIPs ( PIP1-1 and PIP2-2 ) and ALMTs ( ALMT10 and ALMT13 ) genes in LACV roots. Higher PFOS proportions in root cell walls and trophoplasts caused lower root-to-shoot transport in LACV. The ability to cope with PFOS toxicity to shoot cells was poorer in LACV relative to HACV since PFOS proportions were higher in chloroplasts but lower in vacuoles. Our findings provide novel insights into PFOS accumulation in lettuce and further understanding of multiprocess mechanisms of LACV.

Perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) are perfluorinated alkyl substances widely used in industrial and domestic products. The European Food Safety Authority and United States Environmental Protection Agency have recently lowered the reference doses (RfDs) for PFOA and PFOS 4-1800-fold. The recently lowered RfDs call for re-evaluation of potential human health risks from PFOA and PFOS via food consumption. Serious concerns arise because some intakes of PFOA and PFOS exceeded the RfDs. Innovative cultivation of low-accumulating crop varieties becomes an option to decrease human exposure. We present an up-to-date review on low-accumulating crop varieties for PFOA and PFOS in reference to toxic metals and other organic pollutants, including the variety identification, physiological-biochemical mechanisms, molecular uptake mechanisms, and molecular docking, to call for attention and research efforts to decrease human intakes of PFOA and PFOS via crop consumption.

The presence of chromium (Cr) in cultivated fields affects carbohydrate metabolism of rice ( Oryza sativa L.) and weakens its productivity. Little is known about the molecular mechanism of sucrose metabolism underlying Cr stress response in rice plants. In the present study, the transcriptome map of sucrose metabolism in rice seedlings exposed to both trivalent and hexavalent chromium was investigated using Agilent 4 × 44K rice microarray analysis. Results indicated that Cr exposure (3 days) significantly ( p < 0.05) improved sucrose accumulation, and altered the activities of sucrose synthetase, sucrose phosphate phosphatase, and amylosynthease in rice tissues. We identified 119 differentially regulated genes involved in 17 sucrose metabolizing enzymes and found that gene responses in roots were significantly ( p < 0.05) stronger than in shoots under both Cr(III) and Cr(VI) treatment. The network maps of gene regulation responsible for sucrose metabolism in rice plants provide a theoretical basis for further cultivating Cr-resistant rice cultivars through molecular genetic improvement.

This work developed a bioaugmentation strategy that simultaneously reduced soil di(2-ethylhexyl) phthalate (DEHP) pollution and its bioaccumulation in Brassica parachinensis by inoculating the isolated strain Rhodococcus sp. 2G. This strain could efficiently degrade DEHP at a wide concentration range from 50 to 1600 mg/L and transformed DEHP through a unique biochemical degradation pathway that distinguished it from other Rhodococcus species. Besides, strain 2G colonized well in the rhizosphere soil of the inoculated vegetable without competition with indigenous microbes, resulting in increased removal of DEHP from soil (∼95%) and reduced DEHP bioaccumulation in vegetables (∼75% in the edible part) synchronously. Improved enzyme activities and DOC content in the rhizosphere of the planting vegetable and inoculating strain 2G were responsible for the high efficiency in mitigating DEHP contamination to vegetable cultivation. This work demonstrated a great potential application to grow vegetables in contaminated soil for safe food production.

Di- n-butyl phthalate (DBP) is a prevalent pollutant in agricultural soils due to use of plastic film. This study focused on sorption mechanism, kinetics, and isotherms of DBP to six paddy soil particle-size fractions (i.e., coarse sand, fine sand, coarse silt, fine silt, clay, and humic acid fractions). DBP sorption involved in both boundary layer diffusion and intraparticle diffusion, following pseudo-second-order kinetics. DBP sorption was a spontaneous physical process, which fit the Freundlich model. Hydrophobic and ionic interaction relevant to the organic matter content, cation exchange capacity, surface area, and pore volume of soil fractions played key roles in DBP sorption. DBP was strongly adsorbed to humic acid and the sorption was reversely associated with soil particle sizes. DBP may exhibit higher mobility and bioavailability in a soil-crop system at lower temperature (15 °C), due to the lower log K oc values.

Di- n-butyl phthalate (DBP), as an endocrine-disrupting chemical that tends to be accumulated in crops, poses great risks to human health through the food chain. To identify the molecular mechanism underlying differences in their DBP accumulation, the root physiological and proteomic responses to DBP stress of two Brassica parachinensis cultivars, a high-DBP accumulator (Huaguan) and a low-DBP accumulator (Lvbao), were investigated. Root damage of greater severity and significantly greater ( p < 0.05) decreases in root protein content and root activity were detected in Lvbao than in Huaguan, suggesting that Lvbao had lower tolerance to DBP. In total, 52 DBP-responsive proteins were identified by two-dimensional electrophoresis and MALDI-TOF mass spectrometry. More proteins involved in basic metabolic processes, such as protein synthesis and energy metabolism, were downregulated in Lvbao, possibly explaining its lower tolerance and root damage. Several proteins involved in starch metabolism, cell-wall biosynthesis and modification, and stress response were activated in Huaguan, suggesting greater tolerance to DBP. Overall, differences in root proteome between the two cultivars might be responsible for the genotype-dependent DBP tolerance and accumulation in B. parachinensis.

Perfluorooctanesulfonate (PFOS) is a toxic and persistent organic pollutant that can be widely detected in agricultural soils. In this study, two lettuce cultivars with low PFOS accumulation were screened out to reduce the exposure of PFOS to the human body via vegetable consumption. The screened low-PFOS cultivars may help to ensure food safety, despite planting in highly PFOS-polluted soils (1.0 mg/kg), due to their high tolerance to PFOS and 4.4-5.7 times lower shoot PFOS concentration than the high-PFOS cultivars. Protein content and protein-mediated transpiration played key roles in regulating PFOS accumulation in the lettuce cultivars tested. Lower protein content, lower stomatal conductance, and lower transpiration rate resulted in low PFOS accumulation. This study reveals the mechanism of forming low-PFOS accumulation of lettuce cultivars at physiological and biochemical levels and lays a foundation for developing a cost-effective and safe approach to grow vegetables in PFOS-polluted soils.

Sorption of perfluorooctanoic acid (PFOA), a toxic and persistent organic pollutant, by various size fractions of an agricultural soil at environmentally relevant concentrations was evaluated. PFOA sorption to all fractions involved both film diffusion and intraparticle diffusion with the rate-limiting step by the latter. PFOA isotherm data fitted a linear model. Organic matter (OM), cation exchange capacity, pore volume, and the Brunauer-Emmett-Teller area played key roles in PFOA sorption. The sorption capacity followed the order of humic acid > clay (0.15-4.4 mm) > fine silt (1.9-39.8 mm) > coarse silt (17.3-79.4 mm) > fine sand (45.7-316.2 mm) > coarse sand (120-724.4 mm), opposite to their contributions to overall PFOA sorption due to the influence of their percentage weight in the original soil. Percentage OM content was the dominant factor controlling the fraction contributions to overall PFOA sorption, demonstrating influence of the hydrophobic force on sorption. PFOA should be highly mobile and bioavailable in soil-crop systems due to the low log K oc values.

iTRAQ analysis was used to map the proteomes of two Brassica parachinensis cultivars that differed in dibutyl phthalate (DBP) accumulation. A total of 5699 proteins were identified to obtain 152 differentially regulated proteins, of which 64 and 48 were specific to a high- and a low-DBP-accumulation cultivar, respectively. Genotype-specific biological processes were involved in coping with DBP stress, accounting for the variation in DBP tolerance and accumulation. Formation of high DBP accumulation in B. parachinensis might attribute to the more effective regulation of protein expression in physiology and metabolism, including (a) enhanced cell wall biosynthesis and modification, (b) better maintenance of photosynthesis and energy balance, (c) greatly improved total capacity for antioxidation and detoxification, and (d) enhanced cellular transport and signal transduction. Our novel findings contribute to a global picture of DBP-induced alterations of protein profiles in crops and provide valuable information for the development of molecular-assisted breeds of low-accumulation cultivars.

A robust method was developed for simultaneous determination of nine trace perfluoroalkyl carboxylic acids (PFCAs) in various edible crop matrices including cereal (grain), root vegetable (carrot), leafy vegetable (lettuce), and melon vegetable (pumpkin) using ultrasonic extraction followed by solid-phase extraction cleanup and high liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). The varieties of extractants and cleanup cartridges, the usage of Supelclean graphitized carbon, and the matrix effect and its potential influencing factors were estimated to gain an optimal extraction procedure. The developed method presented high sensitivity and accuracy with the method detection limits and the recoveries at four fortification levels in various matrices ranging from 0.017 to 0.180 ng/g (dry weight) and from 70% to 114%, respectively. The successful application of the developed method to determine PFCAs in various crops sampled from several farms demonstrated its practicability for regular monitoring of PFCAs in real crops.

A reliable, sensitive, and cost-effective method was developed for determining three quaternary ammonium compounds (QACs) including dodecyltrimethylammonium chloride, cetyltrimethylammonium chloride, and didodecyldimethylammonium chloride in various vegetables using ultrasonic-assisted extraction and gas chromatography-mass spectrometry. The variety and acidity of extraction solvents, extraction times, and cleanup efficiency of sorbents were estimated to obtain an optimized procedure for extraction of the QACs in nine vegetable matrices. Excellent linearities (R(2) > 0.992) were obtained for the analytes in the nine matrices. The limits of detection and quantitation were 0.7-6.0 and 2.3-20.0 μg/kg (dry weight, dw) in various matrices, respectively. The recoveries in the nine matrices ranged from 70.5% to 108.0% with relative standard deviations below 18.0%. The developed method was applied to determine the QACs in 27 vegetable samples collected from Guangzhou in southern China, showing very high detection frequency with a concentration of 23-180 μg/kg (dw).

A selective and sensitive method for the simultaneous detection of three common and hazardous microcystins (microcystins-LR, -RR, and -YR) in various vegetables was established using solid-phase extraction followed by high performance liquid chromatography coupled with mass spectrometry. The methanol-water proportion ratio of the extraction solvent and its acidity, as well as the efficiencies of solid-phase extraction, were evaluated to optimize a pretreatment procedure for extracting the microcystins from 10 vegetable matrices. The limits of detection and quantitation were below 7.5 μg/kg (dw) and 25 μg/kg (dw), respectively, in different vegetable matrices. The recoveries of the microcystins in the 10 vegetable matrices ranged from 61.3 to 117.3%, with RSDs of 0.2-18.3%. The established method was used to analyze 28 field vegetable samples collected from the sides of Lake Dianchi, and microcystin-RR was found in almost all samples at concentrations of 36.4-2352.2 μg/kg (dw).

Thirteen antibiotics in soil from vegetable farmlands of the Pearl River Delta, southern China, were investigated. At least three antibiotics were detected in each sample. Six antibiotics including four quinolones, tetracycline, and sulfamethoxazole were detected in >94% of the samples. The total contents of three tetracyclines, eight sulfonamides, and four quinolones were not detected-242.6, 33.3-321.4, and 27.8-1537.4 μg/kg, respectively. The highest antibiotic concentrations were observed mainly in vegetable farmlands affiliated with livestock farms. Chlortetracycline, sulfameter, and quinolones in some samples exceed the ecotoxic effect trigger value (100 μg/kg) set by the Steering Committee of Veterinary International Committee on Harmonization. The composition and concentration of antibiotics in soil were correlated with vegetable species. This study has revealed an alarming condition of antibiotics in vegetable farmland soil. Further investigation including environmental fate, plant uptake, and human exposure to antibiotics by plant-derived food should be conducted.
