Respiratory dysfunction is a major cause of death in people with spinal cord injury (SCI). A remaining unsolved problem in treating SCI is the intolerable side effects of the drugs to patients. In a significant departure from conventional targeted nanotherapeutics to overcome the blood-brain barrier (BBB), this work pursues a drug-delivery approach that uses neural tracing retrograde transport proteins to bypass the BBB and deliver an adenosine A1 receptor antagonist drug, 1,3-dipropyl-8-cyclopentyl xanthine, exclusively to the respiratory motoneurons in the spinal cord and the brainstem. A single intradiaphragmatic injection at one thousandth of the native drug dosage induces prolonged respiratory recovery in a hemisection animal model. To translate the discovery into new treatments for respiratory dysfunction, we carry out this study to characterize the purity and quality of synthesis, stability, and drug-release properties of the neural tracing protein (wheat germ agglutinin chemically conjugated to horseradish peroxidase)-coupled nanoconjugate. We show that the batch-to-batch particle size and drug dosage variations are less than 10%. We evaluate the nanoconjugate size against the spatial constraints imposed by transsynaptic transport from pre to postsynaptic neurons. We determine that the nanoconjugate formulation is capable of sustained drug release lasting for days at physiologic pH, a prerequisite for long-distance transport of the drug from the diaphragm muscle to the brainstem. We model the drug-release profiles using a first-order reaction model and the Noyes-Whitney diffusion model. We confirm via biological electron microscopy that the nanoconjugate particles do not accumulate in the tissues at the injection site. We define the nanoconjugate storage conditions after monitoring the solution dispersion stability under various conditions for 4 months. This study supports further development of neural tracing protein-enabled nanotherapeutics for treating respiratory problems associated with SCI.

In the past 50 years, isolated blood platelets have had restricted use in wound healing, cancer therapy, and organ and tissue transplant, to name a few. The major obstacle for its unrestricted use has been, among others, the presence of ultrahigh concentrations of growth factors and the presence of both pro-angiogenic and anti-angiogenic proteins. To overcome this problem requires the isolation and separation of the membrane bound secretory vesicles containing the different factors. In the current study, high-resolution imaging of isolated secretory vesicles from human platelets using atomic force microscopy (AFM) and mass spectrometry enabled characterization of the remaining vesicles size and composition following their immunoseparation. The remaining vesicles obtained following osmotic lysis, when subjected to immunoseparation employing antibody to different vesicle-associated membrane proteins (VAMPs), demonstrate for the first time that VAMP-3-, VAMP-7-, and VAMP-8-specific vesicles each possesses distinct size range and composition. These results provide a window into our understanding of the heterogeneous population of vesicles in human platelets and their stability following both physical manipulation using AFM and osmotic lysis of the platelet. This study further provides a platform for isolation and the detailed characterization of platelet granules, with promise for their future use in therapy. Additionally, results from the study demonstrate that secretory vesicles of different size found in cells reflect their unique and specialized composition and function.

Forces between colloidal particles determine the performances of many industrial processes and products. Colloidal force measurements conducted between a colloidal particle AFM probe and particles immobilized on a flat substrate are valuable in selecting appropriate surfactants for colloidal stabilization. One of the features of inorganic fillers and extenders is the prevalence of rough surfaces-even the polymer latex particles, often used as model colloidal systems including the current study, have rough surfaces albeit at a much smaller scale. Surface roughness is frequently cited as the reason for disparity between experimental observations and theoretical treatment but seldom verified by direct evidence. This work reports the effect of nanoscale surface roughness on colloidal force measurements carried out in the presence of surfactants. We applied a heating method to reduce the mean surface roughness of commercial latex particles from 30 to 1 nm. We conducted force measurements using the two types of particles at various salt and surfactant concentrations. The surfactants used were pentaethylene glycol monododecyl ether, Pluronic F108, and a styrene/acrylic copolymer, Joncryl 60. In the absence of the surfactant, nanometer surface roughness affects colloidal forces only in high salt conditions when the Debye length becomes smaller than the surface roughness. The adhesion is stronger between colloids with higher surface roughness and requires a higher surfactant concentration to be eliminated. The effect of surface roughness on colloidal forces was also investigated as a function of the adsorbed surfactant layer structure characterized by AFM indentation and dynamic light scattering. We found that when the layer thickness exceeds the surface roughness, the colloidal adhesion is less influenced by surfactant concentration variation. This study demonstrates that surface roughness at the nanoscale can influence colloidal forces significantly and should be taken into account in colloidal dispersion formulations.

This paper describes electrochemical synthesis of nanorods of tetrathiafulvalene (TTF) charge-transfer salt on gold nanoparticle (GNP) seeds. The seed-mediated process was monitored by cyclic voltammetry, AFM, and field-emission SEM. The electrodeposition of GNPs (nucleation seeds) on highly oriented pyrolytic graphite (HOPG) electrodes was studied as a function of the electrolytic conditions. The GNP size increases with increasing HAuCl4 concentration and decreasing applied overpotential. A morphological transition from quasi-spherical particles to dendritic aggregates occurs when the HAuCl4 concentration increases from 0.5 to 1 mM. The electrocrystallization of (TTF)Br0.76 on the GNP-decorated HOPG was investigated as a function of TTF concentration and GNP morphology. We observed a preferential nucleation of (TTF)Br0.76 on the GNP seed. The seed-mediated (TTF)Br0.76 crystals display a confined crystal morphology in comparison to those nucleated on bare HOPG. Nanopatterns have applications in many areas including sensors, optoelectronics, and crystallization screening. Particle lithography is a convenient method to manufacture nanoring nanopatterns based on organosilane surface chemistry. The pattern thickness is generally limited to the monolayer thickness. This work is focused on the chemical vapor deposition conditions that yield nanopatterns with multilayer thickness. The supra-monolayer n-octadecyltrichlorosilane (OTS) nanoring patterns are made using polystyrene particle lithography. The supra-monolayer nanopatterns are used as "nano-flasks" to deposit and nucleate nanoparticles of small organic molecules including n-docosane, aspirin, and clarithromycin. The supra-monolayer OTS nanopattern is an effective template for nanoparticle array deposition of all three chemicals with high degree of fidelity to the substrate pattern. The nanoparticle size is varied by solution concentration. The preferential deposition of the organic molecules inside the nanoring is attributed to the dewetting of the liquid film on the nanopattern. The dewetting process effectively distributes the liquid film among the "nano-flasks" so that millions of solution experiments can be carried out in isolated droplets with droplet volume as small as 1×10(-10) nL. The research demonstrates a method to manufacture "nano-flask" arrays for high-throughput nanoparticle deposition trials and manufacture of monodisperse organic/drug nanoparticles through self-assembly.

This paper describes nanorod formation induced by nanoparticle seeds. Carboxylic acids self-assemble on graphite into an epitaxial pattern. However, this persistent pattern is changed into nanorods by nanoparticles. Two control parameters have been identified, including nucleus/substrate contact angle and seed-to-critical nucleus size ratio. Effective carboxylic chain length and nanoparticle size range are identified, which can be related to the seed size effect. Some nanorods are detached when methyl-terminated nanoparticles are used, and the percentage of detached nanorods increases with increasing carbon chain length. In contrast, the nanorods do not detach from carboxyl-terminated nanoparticles. Micelle-based siRNA carriers (“micelleplexes”) were prepared from the A-B-C triblock copolymer, poly(ethylene glycol)-poly(n-butyl acrylate)-poly(2-(dimethylamino)ethyl methacrylate) (PEG-PnBA-PDMAEMA), and their in vitro performance and in vivo biodistribution properties were compared with the benchmark PEGylated and basic polycation systems, PEG-PDMAEMA and PDMAEMA, respectively. The micelle architecture, incorporating increased PEG shielding and a larger particle size (~50 nm) than polycation-based complexes (polyplexes; ~10 nm), enhances siRNA delivery performance in two important aspects: in vitro gene silencing efficiency, and in vivo tumor accumulation. The in vitro gene silencing efficiency of the micelleplexes (24% in HeLa cells) was significantly better than the statistically-insignificant levels observed for PDMAEMA and PEG-PDMAEMA polyplexes under identical conditions. This enhancement is linked to the different mechanisms by which micelleplexes are internalized (i.e., caveolar, etc.) compared to PDMAEMA and PEG-PDMAEMA polyplexes. Folate-functionalization significantly improved micelleplex uptake but had negligible influence on gene silencing efficiency, suggesting that this parameter is not limited by cellular internalization. In vivo biodistribution analysis revealed that siRNA delivered by micelleplexes was more effectively accumulated and retained in tumor tissues than that delivered by PEGylated polyplexes. Overall, the micelle particle size and architecture appear to improve in vitro and in vivo delivery characteristics without significantly changing other properties, such as cytotoxicity and resistance to enzymes and dissociation. The self-assembled nature of micelleplexes is expected to enable incorporation of imaging modalities inside the hydrophobic micelle core, thus combining therapeutic and diagnostic capabilities. The findings from the present study suggest that the micelleplex-type carrier architecture is a useful platform for potential theranostic and tumor-targeting applications.

The paper describes immobilization methods of bacteriophage P22 and tailspike gp9 proteins isolated from P22 on atomic force microscope (AFM) probes. The paper also reports single molecule force spectroscopy (SMFS) using AFM of the immobilized P22 (or gp9) interactions with substrate-supported O-antigenic lipopolysaccharides (LPS) bilayers. LPS covers the outer membrane of gram-negative bacteria, such as Salmonella typhimurium. Evidence from AFM imaging and SMFS shows that immobilized P22 (or gp9) are capable of strong and multivalent binding to supported LPS. The most common rupture forces between P22 and LPS were identified to be 72, 130, 206, and 279 pN at force loading rate of 12,000 pN/s. The quantized unbinding force was found to decrease with decreasing force loading rate as predicted by the Bell model. By fitting the force data with the Bell model, an energy barrier of 55 kJ/mol was obtained. Evidence is also provided that demonstrates the resilience of phage to pH and temperature fluctuation as well as dehydration/rehydration cycles. The biospecific interactions between P22 and the LPS are relevant to cell infection, inflammation, cancer progression and metastasis, food safety, pharmaceuticals, and biosensor development.

The effect of cross-linking layer-by-layer (LbL) films consisting of bioreducible poly(2-dimethylaminoethyl methacrylate) (rPDMAEMA) and DNA is examined with regard to rigidity, biodegradability, cell adhesion, and transfection activity using 1,5-diiodopentane (DIP) cross-linker. DIP chemically reacts with the tertiary amines of rPDMAEMA, altering the chemical composition of these LbL films. The result is a change in surface morphology, film swelling behavior, and film rigidity, measured with AFM and ellipsometry. It is found that the apparent Young's modulus is increased more than 4 times its original value upon cross-linking. Cross-linking mass is additionally confirmed with a quartz crystal microbalance with dissipation (QCM-D). Comprehensive analyses of these experimental values were investigated to calculate the degree of cross-linking using the rubber elasticity theory and the Flory-Rehner theory. Additionally, the Flory-Huggins parameter, chi, was calculated. Good agreement in the two methods yields a cross-linking density of approximately 0.82 mmol/cm(3). The Flory-Huggins parameter increased upon cross-linking from 1.07 to 1.2, indicating increased hydrophobicity of the network and formation of bulk water droplets within the films. In addition, the effects of cross-linking on film disassembly by 1,4-dithiothreitol (DTT) are found to be insignificant despite the alteration in film rigidity. Mouse fibroblast cells and smooth muscle cells are used to study the effect of cross-linking on cell adhesion and cell transfection activity. In vitro transfection activity up to seven days is quantified using secreted alkaline phosphatase (SEAP) DNA. Film cross-linking is found to enhance cell adhesion and prolong the duration of cellular transfection. These results contribute to the development of bioreducible polymer coatings for localized gene delivery.

The molecular disassembly and DNA release dynamics of bioreducible poly(amido amine) polyplexes were studied in real time by atomic force microscopy (AFM). Molecular disassembly of polyplexes constitutes a critical step along the nonviral gene delivery pathway. DNA release is triggered by the depolymerization of high-molecular-weight polycations into low-molecular-weight oligocations utilizing the thiol and disulfide exchange reaction mechanism. The research is motivated by low cytotoxicity and higher transfection of the poly(amido amine)s containing the disulfide bond than those without the disulfide linkage. AFM images were captured in simulated physiological reducing environment by employing dithiothreitol (DTT). Distinctive stages of polyplex disassembly common to the polyplexes with different disulfide content, molecular weight, and polymer architecture are revealed though the DNA release rate is dependent on the disulfide content. In stage one, upon the depolymerization, polyplexes evolve from metastable structures into the thermodynamically favorable toroid structure. In the second stage, toroids aggregate and sometimes fuse into larger ones. In the last stage DNA gradually unravels from the polyplex resulting in highly decondensed wormlike chains and loops that are held by a central compact core. The results affirm the promising aspect of bioreducible poly(amido amine)s as controlled DNA delivery vectors. The study offers new physical insights into the DNA release pathway including intermediate structures with a high degree of structural heterogeneity and disassembly-induced particle growth. The study identifies disassembly-induced colloidal and morphological instability as an important issue to be addressed in the development of successful polyelectrolyte-based gene delivery systems.

Controlled intracellular disassembly of polyelectrolyte complexes of polycations and DNA (polyplexes) is a crucial step for the success of nonviral gene delivery. Motivated by our previous observation of different gene delivery performances among multiblock reducible copolypeptide vectors (Manickam, D. S., and Oupický, D. Bioconjugate Chem. 2006, 17, 1395−1403), atomic force microscopy is used to visualize plasmid DNA in various decondensed states from reducible polypeptide polyplexes under simulated physiological reducing conditions. DNA decondensation is triggered by reductive degradation of disulfide-containing cationic polypeptides. Photodynamic therapy (PDT) is a promising treatment modality for cancer. PDT is based on the concept that photosensitizers, when exposed to light of specific wavelength, generate cytotoxic reactive oxygen species (ROS) capable of killing tumor cells. The effectiveness of PDT has been limited in part by the lack of photosensitizers that accumulate sufficiently in tumor cells and poor yield of ROS from existing photosensitizers. In this report, we investigated whether aerosol OT-alginate nanoparticles can be used as a carrier to enhance the therapeutic efficacy of a model photosensitizer, methylene blue. Methylene blue loaded nanoparticles were evaluated for PDT effectiveness in two cancer cell lines, MCF-7 and 4T1. Encapsulation of methylene blue in nanoparticles significantly enhanced intracellular ROS production, and the overall cytotoxicity following PDT. It also resulted in higher incidence of necrosis. Greater effectiveness of nanoparticles could be correlated with higher yield of ROS with nanoparticle-encapsulated methylene blue. Further, treatment of tumor cells with nanoparticle-encapsulated methylene blue resulted in significant nuclear localization of methylene blue while free drug treatment resulted in its accumulation mainly in the endolysosomal vesicles. In conclusion, encapsulation of methylene blue in aerosol OT-alginate nanoparticles enhanced its anticancer photodynamic efficacy in vitro. Increased ROS production and favorable alteration in the subcellular distribution contribute to the enhanced PDT efficacy of nanoparticle-encapsulated photosensitizer.

We report synthesis of temperature- and redox-responsive multiblock copolymers by reversible addition-fragmentation chain transfer (RAFT) polymerization. Well-defined α,ω-bis(dithioester)-functionalized poly(N-isopropylacrylamide) (PNIPAM) and poly(2-(dimethylamino) ethyl methacrylate) (PDMAEMA) were prepared using 1,4-bis(thiobenzoylthiomethyl)benzene and 1,4-bis(2-(thiobenzoylthio)prop-2-yl)benzene as RAFT agents, respectively. Dually responsive multiblock copolymers were synthesized in a single aminolysis/oxidation step from the α,ω-bis(dithioester)-terminated PNIPAM and PDMAEMA. The copolymers and their stimulus-responsive behavior were characterized by size exclusion chromatography, NMR, light scattering and atomic force microscopy. Due to the presence of redox-sensitive disulfide bonds between the blocks, the copolymers were readily reduced to the starting polymer blocks. The presence of temperature-responsive PNIPAM blocks provided the copolymers with the ability to assemble into core-shell nanostructures with hydrophobic PNIPAM as a core and cationic PDMAEMA as stabilizing shell when above the phase transition temperatures of PNIPAM. The temperature-induced assembly of the copolymers also showed substantial pH sensitivity. The phase transition temperature increased with decreasing pH, while molecular weight of the assemblies decreased.

The influence of solvents (alcohols and binary mixtures of alcohol and water) on the structure and permeability of self-assembled layer-by-layer polyelectrolyte multilayers (PEMs) has been investigated by in situ atomic force microscopy (AFM), cyclic voltammetry (CV), and confocal laser scanning microscopy (CLSM). A decrease in the dielectric constant of the solvent medium increases the strength of electrostatic interactions between the polyelectrolyte chains because the Coulombic force is inversely proportional to the dielectric constant. The stronger attractions as a result of increased alcohol volume percentage in water drive the polyelectrolyte chains to contract, collapse, and coagulate. Consequently, subtle changes in the PEM film structure are observed. AFM images show that the originally smooth surface of the PEM film becomes rougher with aggregates and holes developing with increasing alcohol amount. The Layer-by-layer deposition of positively and negatively charged macromolecular species is an ideal method for constructing thin films incorporating biological molecules. We investigate the adsorption of fibronectin onto polyelectrolyte multilayer (PEM) films using optical waveguide lightmode spectroscopy (OWLS) and atomic force microscopy (AFM). PEM films are formed by adsorption onto Si(Ti)O2 from alternately introduced flowing solutions of anionic poly(sodium 4-styrenesulfonate) (PSS) and cationic poly(allylamine hydrochloride) (PAH). Using OWLS, we find the initial rate and overall extent offibronectin adsorption to be greatest on PEM films terminated with a PAH layer. The polarizability density of the adsorbed protein layer, as measured by its refractive index, is virtually identical on both PAH- and PSS-terminated films; the higher adsorbed density on the PAH-terminated film is due to an adsorbed layer of roughly twice the thickness. The binding of monoclonal antibodies specific to the protein's cell binding site is considerably enhanced to fibronectin adsorbed to the PSS layer, indicating a more accessible adsorbed layer. With increased salt concentration, we find thicker PEM films but considerably thinner adsorbed fibronectin layers, owing to increased electrostatic screening. Using AFM, we find adsorbed fibronectin layers to contain clusters; these are more numerous and symmetric on the PSS-terminated film. By considering the electrostatic binding of a segmental model fibronectin molecule, we propose a picture of fibronectin adsorbed primarily in an end-on-oriented monolayer on a PAH-terminated film and as clusters plus side-on-oriented isolated molecules onto a PSS-terminated film.

The self-associating structures at the solid-liquid interface of three nonionic trisiloxane surfactants ((CH3)3SiO)2Si(CH3)(CH2)3(OCH2CH2)n OH (n = 6, 8, and 12), or BEn, are studied as a function of substrate properties by atomic force microscopy (AFM) imaging and force measurement. These trisiloxane surfactants are known as superwetters, which promote rapid spreading of dilute aqueous solutions on low-energy surfaces. This study also attempts to relate the BEn surface aggregate structures at the solid-liquid interface to their superwetting behavior. Four substrates are used in the study: muscovite mica, highly oriented pyrolytic graphite, and oxidized silicon wafer with and without a full monolayer of self-assembled n-octadecyltrichlorosilane (OTS). The concentration of BEn is fixed at 2 times the critical aggregation concentration (CAC). The BEn surfactants are only weakly attracted to hydrophilic surfaces, more on oxidized silicon than on mica. All three form ordinary planar monolayers on HOPG and OTS-covered oxidized silicon. The significance of surfactant adsorption on the AFM tip is investigated by comparing the force curves obtained by tips with and without thiol modification. The surface aggregate structures of the BEn surfactants correlate with their bulk structures and do not exhibit anomalous adsorption behavior. The adsorption behavior of the BEn superwetters is similar to that of the CmEn surfactants. Thus, our results confirm previous work showing that superwetting shares its main features with other classes of surfactants.

We investigate the making of semiconductor nanorods using well-defined molecular templates. The films are characterized by AFM and XPS. The spin-coated monolayer of arachidic acid assembles on graphite as 1D nanostripes with bilayer periodicity. The copper arachidate film shows curved stripes, which become straight after reacting with hydrogen sulfide. The new template scheme may facilitate the fabrication of nanorod arrays of a variety of inorganic and semiconducting materials.

Atomic force microscopy (AFM) was used to study the aggregation structure and kinetics of nonionic surfactant penta(oxyethylene) dodecyl ether (C12E5) on mica as a function of temperature. Surface forces and topographical images of surfactant aggregates at the liquid/solid interface were captured in the vicinity of 21 °C. The surfactant molecular aggregates were imaged by choosing an imaging force in the steric repulsion region so that the AFM tip was just outside the surface of the aggregates. At below 21 °C, C12E5 adsorbed as fragments of tens of nanometers in size. The fragments consist of 1−2 nm thick strongly adsorbed species and weakly adsorbed species extending as far as 15 nm from the substrate surface. The fragments gradually increased in number and attached to each other to form clusters connected in one dimension (strings) and two dimensions (networks). A recently developed method of spherulitic crystallization was used for the preparation of highly ordered 80 nm thin films of the dye 1.7-bis(dimethylamino)heptamethinium perchlorate (BDH+ClO4-). Depending on the crystallization temperature, the film color and surface topography varied widely, while the crystal structure and film thickness remained the same. At low undercooling, two homogeneous regions were obtained. One showed in-plane symmetrical and the other asymmetrical growth behaviors. At high undercooling, a banded spherulitic structure with rainbow-like colors developed, whose formation is attributed to the out-of-plane asymmetrical growth. Atomic force microscopy (AFM) studies of N-[4-[4-cyano-2-(2-furanylmethylene)-2,5-dihydro-5-oxo-3-furanyl]phenyl]-1-butanesulfonamide (1), a crystalline dye used in photographic film, are described. AFM of the largest crystal face of freshly cleaved blade-shaped crystals of 1 revealed molecular scale contrast with periodicity identical with the low-energy oleophilic (100) plane, and ledges oriented along [001] with step heights equal to the a lattice parameter, which corresponds to the height of single molecules of 1. These features are consistent with the solid-state structure of the dye, which reveals a low-energy oleophilic (100) plane and strong intermolecular π−π interactions along [001]. Real-time in situ AFM reveals that growth of 1 near equilibrium conditions proceeds by flow of the [001] ledges along the [010] direction, consistent with the relative surface energies of the crystal planes and a terrace−ledge−kink mechanism. Ultrathin organic multilayer films are appealing to scientists not only because they serve as molecular level models but also because they can be transformed into superlattices through successive deposition of alternating layers of different materials. Cationic bolaform amphiphile and anionic polyelectrolyte multilayers were self-assembled from their aqueous solutions via electrostatic attraction. The bolaform amphiphile layers are photopolymerizable by UV irradiation. The mechanical and surface electrical properties of the multilayer film were studied by surface force measurements, and the surface topography was studied by atomic force microscopy. We observed an average increase in film thickness of 75 with each amphiphile and polyelectrolyte double layer deposition, a molecularly smooth first anchoring amphiphile layer, and nonuniform polyelectrolyte layers with a surface roughness of 30 A. The surface roughness was preserved in the multilayer deposition process and was reduced by adding salt to the polyelectrolyte solution. Photopolymerization increases the integrity of the first anchoring layer as well as the subsequent multilayers. It took less than 5 min to dissolve a monomeric amphiphile film in chloroform, but immersion of the UV-irradiated film in chloroform for 2 h only resulted in partial removal of film materials. The multilayer film is quite fragile especially for the first few layers. Controlling of the monolayer structure by selective variation of the amphiphile chemical structure may help to optimize the photopolymerization process.

We have prepared self-assembled monolayers of novel cationic bolaform amphiphiles on negatively charged substrates. Most of these amphiphiles form smooth, defect-free monolayers which can be used to reverse the substrate surface charge and thus allow subsequent adsorption of anionic molecules and construction of multilayers. Atomic force microscopy, surface force measurement, and surface plasmon spectroscopy were combined to probe the molecular orientation and ordering, mechanical properties, and surface electrical properties of the monolayers. In addition, the amphiphile aggregation behavior at an air-water interface was studied by surface tension measurement, and lyotropic phase behavior was studied by polarization microscopy. Our study suggests that monolayer interfacial and bulk properties can be controlled to a certain degree by selective variation of amphiphile chemical structure, in particular, the alkyl chain length and the type of counterions. A n increase in alkyl chain length assists close-packing at the liquid-solid interface and self-assembly in a liquid medium due to a favorable hydrophobic free energy change. Exchange of halide ions with the strongly associating salicylate ions reduces electrostatic repulsion between head groups and also favors self-assembly and close-packing. Our study suggests that it is possible to overcome the dominance and limitation of the substrate electrostatic effect on monolayer structure by using amphiphiles with a strong inherent tendency for close-packing. Our observations contribute to the understanding of two-dimensional topochemical photopolymerization, multilayer deposition of alternating surface charges, modification of hydrophilic surface electrical properties, and in general, the dependence of monolayer architecture on molecular chemical structure and intermolecular forces.

Monoand multilayers of photopolymerizable bolaform amphiphiles and polyelectrolytes self-assembled onto mica from aqueous solution were studied by surface force measurement and atomic force microscopy. Multiple layers of positively charged amphiphiles self-assemble if a negatively charged polyelectrolyte is inserted between them. The layers can be photopolymerized with a high-pressure UV lamp. The atomic force microscopy images indicate that the photopolymerized amphiphile monolayer has a higher degree of order than the unpolymerized monolayer, although the latter possesses some long-range order. Mechanical properties of the system measured by the surface force apparatus indicate that a photopolymerized layer is more robust and demonstrate that the rigidity and integrity of the first anchoring layer greatly influence the stability of the multilayer. The interaction between polyelectrolyte layers in water can be either attractive or repulsive depending on the coverage of the polyelectrolyte on the amphiphile layer. The mica separation of a pair of opposed monolayers in contact in the presence of water is surprisingly small (25 A) compared to the thickness (78 A) of a bilayer composed of fully extended amphiphiles.

This communication describes a spin-coating method to nucleate organic molecular rods of uniform size from an inorganic nanoparticle at a solid surface. The particle-rod hybrid structure spontaneously forms when a film is spin coated from a mixed 2-propanol solution of arachidic acid (AA) and nanoparticles of cadmium selenide capped by mercaptoundecanoic acid (MUA-CdSe) on graphite. AFM images show that MUA-CdSe nanoparticles nucleate single crystalline rods of AA with a cross section of a single unit cell of the C-form. The solution-based process potentially allows the precise tuning of the wetting profile of the solution on the surface-attached nanoparticle, which provides the reservoir for the growth of the single crystalline rods. The results suggest that nanoparticles can be regarded as nanoseeds for the nucleation of guest crystals. It should be possible to further functionalize the AA rods by electrostatic complexation with metal or organic ions.
