The effects of naloxone and beta-endorphin on LH secretion by pig pituitary cells were studied in primary cultures. On Day 4 of culture, cells (10(5) seeded/well) were challenged with 10(-9), 10(-8) or 10(-7) M gonadotrophin-releasing hormone (GnRH), 10(-10), 10(-9), 10(-8) or 10(-7) M-beta-endorphin or 10(-6) M-naloxone individually or in combinations. Secreted LH was measured at 4 h and 24 h after treatment and cellular content of LH was measured after 24 h. Basal LH secretion (control) was 23.5 +/- 7.6 and 36.9 +/- 10.3 ng/well at 4 h and 24 h, respectively. Relative to control at 4 h, 10(-9), 10(-8) or 10(-7) M-GnRH stimulated (P less than 0.05) LH secretion 140%, 210% and 250%, respectively. At 24 h, LH secretion was increased (P less than 0.05) by GnRH compared to control, but the dose-response to GnRH was absent. Naloxone increased (P less than 0.01) LH secretion 166 +/- 13% at 4 h and 141 +/- 13% (P less than 0.06) at 24 h. Secretion of LH after simultaneous addition of 10(-8) M-GnRH plus naloxone was greater (P less than 0.01) than after GnRH alone at 4 h but not at 24 h. beta-Endorphin at 10(-10), 10(-9), 10(-8) or 10(-7) M failed to alter basal LH secretion at 4 h but decreased secretion at 24 h, while cellular LH content was similar to control at 24 h.(ABSTRACT TRUNCATED AT 250 WORDS)

Glucocorticoids have been found to inhibit reproductive function in most domestic species studied but, in the ewe, preliminary reports suggest that glucocorticoids may have little or no effect. This study investigated the effects of the synthetic glucocorticoid dexamethasone on oestrus and ovulation rate in ewes during the breeding season and gonadotrophin secretion in the breeding and non-breeding seasons. In cyclic ewes, dexamethasone treatment at rates of up to 2 mg/day did not affect the natural or pregnant mare serum gonadotrophin-stimulated ovulation rate, or the timing and incidence of behavioural oestrus (P>0·05). Dexamethasone administration (2 mg/day) had no effect on LH secretion or the plasma LH response to a 1 μg injection of gonadotrophin-releasing hormone (GnRH) in ovariectomized ewes in the breeding and non-breeding seasons, and did not compromise the inhibition of plasma LH levels during chronic treatment with oestrogen. To investigate possible differential pituitary secretion of LH in breeding and non-breeding female naked mole-rats, the LH responses to administration of exogenous GnRH were measured in 55 females from 20 captive colonies. Single doses of 0.1, 0.5 or 1.0 micrograms GnRH produced a significant rise in plasma LH concentrations 20 min after s.c. injection in breeding and non-breeding females at all doses (P less than 0.001). While at the highest dose of 1.0 microgram there was no difference in the LH response between breeding and non-breeding females, as the dose was lowered there was a progressive decline in the LH response in non-breeding females such that, at the 0.1 microgram dose, GnRH produced only a small, but significant, increase in plasma LH (1.3 +/- 0.2 to 2.9 +/- 0.5 mi.u./ml, N = 5) compared with breeding females (3.4 +/- 0.8 to 9.6 +/- 2.0 mi.u./ml, N = 6). The LH responses of the latter were not significantly reduced at the lower doses of GnRH. The apparent lack of sensitivity to low doses of exogenous GnRH in non-breeding females was reversed by 4 consecutive 1-h injections of 0.1 microgram, which produced a rise in LH from 1.2 +/- 0.2 to 9.0 +/- 0.2 mi.u./ml (N = 4), comparable to that of breeding females given a single injection of 0.1 microgram GnRH. These results suggest that the anterior pituitary in non-breeding female naked mole-rats is less sensitive to low doses of exogenous GnRH than in breeding females, possibly due to a lack of priming by endogenous GnRH. Therefore, the socially-induced block to ovulation in non-breeding female naked mole-rats may be due to inhibition of hypothalamic GnRH secretion.

The mode of action of a recently isolated gonadal protein, termed FSH-suppressing protein (FSP) or follistatin, on basal and gonadotrophin-releasing hormone (GnRH)-stimulated release of FSH and LH and on pituitary cell content of FSH and LH was examined in rat pituitary cell cultures and compared with the previously reported effects of inhibin. Pituitary cells were cultured for 3–9 days in the presence of graded doses of FSP and the basal release rates and changes in cell contents of FSH and LH determined during this period. FSP suppressed both the basal release rate and the cell content of FSH with median inhibitory concentrations (IC50) of 135 and 161 pmol/l respectively. The corresponding effects of FSP on LH basal release rate and LH cell content (IC50 = 200 pmol/l) were limited compared with the effects on FSH. The effect of FSP on GnRH-stimulated release of FSH and LH during 4 h was determined in cells which had been preincubated with FSP for 3 days, and the GnRH-stimulated release of FSH and LH analysed as a percentage of the respective gonadotrophin available for release. FSP antagonized GnRH action with dose-related increases in the GnRH median effective (stimulatory) concentrations for FSH and LH release (EC50 values = 56 and 400 pmol/l respectively) and a suppression in the maximum release of FSH and LH by excess GnRH (IC50 values =142 and 150 pmol/l respectively). The effect of FSP on FSH cell content after 3 days in culture was insensitive to the neutralizing effects of an inhibin antiserum. The pattern of FSP-induced suppression of FSH basal release and cell content and its inhibition of GnRH-stimulated release of FSH and LH was similar to that observed with inhibin, although FSP is 10–20% as potent as inhibin. When inhibin and FSP were assayed either individually or as a 1:1 (v/v) mixture, the experimentally determined biological activity of the mixture did not differ significantly from that predicted from the individual activities. It was concluded from the similarity in pituitary responses to FSH and inhibin that FSP is a weak inhibin agonist. Its action can be discriminated from that of inhibin by an inhibin antiserum; however, FSP and inhibin may share a common underlying mechanism of action since their effects are additive. The pituitary response to exogenous GnRH was studied in 8 colts of Quarter Horse phenotype from 32 to 96 weeks of age. Colts were from dams treated daily from Day 20 to 325 of gestation with (1) 2 ml neobee oil per 50 kg body weight (controls); or (2) 2 ml altrenogest per 50 kg body weight. GnRH challenges (5 micrograms/kg body weight) were administered every 8 weeks from 32 to 96 weeks of age to estimate pituitary content of LH. Blood samples were collected every 20 min for 4 h before GnRH and 15, 30, 45, 60, 90, 120, 180, 240 and 360 min after GnRH. Serum concentrations of LH and FSH were determined for the 2 pre-GnRH and all post-GnRH samples. Baseline concentrations (mean of 2 pre-GnRH samples) of LH and FSH were not affected by treatment (P greater than 0.05). Serum concentrations of LH declined from 40 to 56 weeks and rose again between 72 and 80 weeks. Basal concentrations of FSH declined from 32 to 56 weeks, and varied widely after 56 weeks. The maximum LH response to GnRH (highest concentration after GnRH minus baseline) declined steadily in both groups for 48 to 64 weeks but remained relatively constant in both groups after 64 weeks. The maximum FSH response to GnRH declined from 32 to 64 weeks then remained relatively constant in both groups. The GnRH-induced gonadotrophin release remained low with a transient increase at 72 weeks for both hormones.(ABSTRACT TRUNCATED AT 250 WORDS)

Puberty was studied using 15 fillies of Quarter Horse phenotype. Fillies were from dams treated daily from Days 20 to 325 of gestation with: (1) 2 ml neobee oil per 50 kg body weight (controls); or (2) 2 ml altrenogest (2.2 mg/ml) per 50 kg body weight. The clitoris was measured at birth and approximately every 12 weeks until 84 weeks of age. Blood samples were collected from 9 fillies (5 treated, 4 controls) every 4 days over a 28-day period at 8-week intervals from 4 to 68 weeks of age; sampling continued every 4 days after 72 weeks of age until first oestrus. Blood samples were collected daily during oestrus (greater than or equal to 35 mm follicle) and on Days 4, 6, 10, and 14 after ovulation for the first 2 oestrous cycles. GnRH challenges (5 micrograms/kg) were administered every 8 weeks from 32 to 96 weeks of age. Puberty was defined as the first oestrus with ovulation. Beginning 1 February 1987, fillies were teased daily and their ovaries were examined by ultrasonography every 3 days (daily during oestrus). Fillies were inseminated with 500 x 10(6) motile spermatozoa from one stallion. Pregnancy was diagnosed by ultrasonography on Days 11, 12, 15 and every 5 days until Day 50 after ovulation. Prenatal altrenogest treatment caused clitoral enlargement (P less than 0.05) and increased serum concentrations of LH from 1 to 7 months of age. The amount of LH released in response to exogenous GnRH was greater (P less than 0.05) in treated fillies at 32, 64, and 72 weeks of age. Treated fillies had higher serum concentrations of FSH from 1 to 4 months (P less than 0.05), but FSH was lower (P less than 0.05) in treated fillies before and during first oestrus. Serum concentrations of LH and FSH peaked transiently at 10 months and LH was depressed from 64 to 88 weeks and began to rise 14 days before first oestrus. Concentrations of FSH began to decline 14 days before first oestrus. The median age at puberty was 90 weeks. Durations of oestrus, dioestrus, and the oestrous cycle were not different between groups and were similar to those for adult mares. First cycle pregnancy rates and overall rates were 100 and 82% and 100 and 91.7% for control and treated fillies, respectively (P greater than 0.05). Maternal treatment with altrenogest did alter gonadotrophin secretion before puberty, but had no effect on functional reproductive performance in fillies.

Anoestrous Romney Marsh ewes were treated with small-dose (250 ng) multiple injections of GnRH. Ewes in Groups 1 and 3 were hysterectomized 2 weeks before treatment, while those in Groups 2 and 4 were intact controls. Groups 1 and 2 were primed with progesterone (+P) and treated with 2 h injections of GnRH (250 ng) for 36 h, while Groups 3 and 4 were not pretreated (-P) but were given 2 h injections of GnRH (250 ng) for 18 h. Both treatment regimens were terminated with a bolus injection of GnRH (125 micrograms), given to synchronize the timing of the LH surge and subsequent luteal progesterone production. The plasma progesterone profiles of 5/5 animals in Group 2 (+P controls) and 2/5 animals in Group 4 (-P controls) were indicative of normal luteal function, while the remaining 3/5 animals in Group 4 produced plasma progesterone profiles typical of abnormal luteal function. However, in all the hysterectomized animals (Groups 1 and 3) peripheral plasma progesterone concentrations rose to reach a mean peak value of 1.3 ng/ml plasma on Day 8 which was maintained in all animals irrespective of progesterone pretreatment. The absence of a fall in progesterone concentrations precluded the identification of any animal in Group 4 showing abnormal luteal function. It was also noted that, after hysterectomy, although the corpus luteum was maintained, it was with reduced secretory capacity. The prevention of the expected proportion (70%) of -P animals from displaying a decline in plasma progesterone concentration after hysterectomy provides firm evidence that the uterus is involved in the premature regression of the short-cycle corpus luteum.

The release of LH from the pituitary of lactating ewes was studied. In Exp. 1, ewes were injected with 50 microng oestradiol benzoate (OB), 2-0 mg testosterone propionate (TP) or oil only (control) on days 5, 10, or 20 after lambing. LH was measured in peripheral plasma samples obtained 20-38 h after treatment, and the ovulations were recorded. The number of ewes in which an LH release was detected, and the amount released, declined between Day 5 and 20 after OB treatment but increased after TP treatment. The releases of LH were not always accompanied by ovulation and the incidence of ovulation was higher in ewes treated with TP. In Exp. 2, lactating ewes were injected with 1 or 5 (at 2-h intervals) doses of 50 microng Gn-RH, on Days 12 or 25 after lambing. LH was measured in peripheral plasma samples collected every 2 h for 10 h and every 3 h for a further 70 h. Release of LH occurred in all ewes, the amount being greater in ewes receiving multiple injections and in ewes treated on Day 25. The incidence of ovulation was higher after treatment on Day 25. Multiple injections of Gn-RH appeared to reduce the incidence of abnormal corpora lutea.

SUMMARY The experiment was designed to determine whether chronic administration of gonadotrophin releasing hormone (GnRH) plus thyrotrophin releasing hormone (TRH) to pubertal bulls would hasten sexual maturation. Nine bulls, three each at 4, 5 and 6 months of age were given i.m. injections of GnRH (0·4 μg/kg) plus TRH (2 μg/kg) three times daily (09.00, 16.00 and 23.00 h) for 30 days. Equivalent animals were given 0·9% saline vehicle as controls. At the outset of injections the 6-, 5- and 4-month-old bulls weighed 201, 157 and 169 kg respectively; equivalent weights at the final injection were 222, 186 and 198 kg. Plasma levels of LH, prolactin and testosterone were determined weekly around the injection given at 09.00 h. Twenty-four-hour plasma testosterone and LH profiles were obtained immediately after the last injection. The animals were unilaterally castrated at the end of this 24 h period and the second testis was removed 60 days later to assess testicular weights and epididymal sperm numbers. Surges of LH, testosterone and prolactin occurred throughout the experimental period in response to GnRH/TRH injection. The LH surge in response to the first injection was nearly fivefold larger than those measured at subsequent weekly intervals but prolactin and testosterone responses did not decline during the 28-day experimental period. Induced LH and prolactin peaks were larger than those occurring in the control bulls, but control animals produced an equivalent amount of testosterone. These bulls produced quasi-adult levels of testosterone even at 4 months of age. Chronic GnRH/TRH administration resulted in a depression of plasma testosterone, and a disruption of the normal episodic release pattern during the 24 h after the final treatment, but normal patterns returned within 60 days. Testicular weights and epididymal sperm numbers were not increased by chronic GnRH/TRH treatment.

Fourteen seasonally anoestrous, non-lactating fallow does were each treated with an intravaginal progesterone device for 14 days followed by a subcutaneously implanted osmotic minipump delivering synthetic GnRH at doses of 125 ng/h (7 does) or 250 ng/h (7 does) for up to 7 days, about 6 weeks before the natural breeding season. One doe (low delivery rate) lost its intravaginal device and 6 of the remaining does (46.2%) exhibited oestrus between 71 and 120 h after progesterone withdrawal/minipump insertion. Only one of these does received the low GnRH delivery rate and 5 received the high rate. Serum progesterone profiles indicated that an induced oestrus was followed by apparently normal luteal development. Does which did not exhibit oestrus failed to show a luteal response. Only 1 doe conceived at induced oestrus, producing a viable female fawn 6 weeks before the start of the natural fawning season. The remaining does returned to an anoestrous state until the onset of the natural breeding season.

Passive immunization of male lambs against oestradiol-17 beta from 2 to 16 weeks of age significantly elevated androgen concentrations in plasma and depressed the median eminence content of dopamine. Removal of endogenous oestrogens had no significant effects on plasma FSH, LH or prolactin concentrations or on testicular growth and hypothalamic content of GnRH. These results suggest that endogenous oestrogens may indirectly suppress testicular androgen secretion by exerting a stimulatory influence on hypothalamic dopaminergic neurones, which in turn may inhibit GnRH secretion by the median eminence.

Ovariectomized rats were treated with oestradiol benzoate and progesterone or GnRH. Prolonged exercise (running 4 days per week for 6 weeks) markedly potentiated the oestrogen/progesterone-induced release of LH and FSH, but the pituitary response to an injection of GnRH was unaffected. In contrast, at 24 h after a single exercise bout there was no apparent effect on steroid and GnRH stimulated LH and FSH responses although an acute exercise session given on the day of the LH surge inhibited steroid-induced LH release in some rats. We conclude that strenuous, prolonged exercise-training in the ovariectomized rat seems to modify the ability of the hypothalamus to release GnRH. The results were not attributable to a single bout of exercise since the gonadotrophin responses immediately or 24 h after such exercise did not parallel the results observed in the trained rats.

Plasma LH concentrations were monitored in 6 Hereford X Friesian suckled cows at about 80 days post partum, before and during a 14-day period of continuous s.c. infusion of GnRH (20 micrograms/h). Blood samples were collected at 10-min intervals on Days -2, -1, 1, 2, 3, 4, 7, 10, 13 and 14 (Day 1 = start of infusion). Plasma LH concentrations rose from mean pretreatment levels of 1.3 +/- 0.20 ng/ml to a maximum of 17.1 +/- 3.09 ng/ml within the first 8 h of GnRH infusion, but returned to pretreatment levels by Day 2 or 3. In 4/6 animals, the initial increase was of a magnitude characteristic of the preovulatory LH surge. In all animals, an i.v. injection of 10 micrograms GnRH, given before the start and again on the 14th day of continuous infusion, induced an increase in LH concentrations but the increase to the second injection was significantly (P less than 0.01) less (mean max. conc. 6.4 +/- 0.76 and 2.3 +/- 0.19 ng/ml). Mean LH concentrations (1.0 +/- 0.08, 1.1 +/- 0.08 and 0.9 +/- 0.06 ng/ml) and LH episode frequencies (3.3,4.3 and 3.2 episodes/6 h) did not differ significantly on Days -2,7 and 13. However, the mean amplitude of LH episodes was significantly lower (P less than 0.05) on Day 13 (1.3 +/- 0.10 ng/ml) than on Day -2 (1.8 +/- 0.16 ng/ml). Therefore, although the elevation in plasma LH concentrations that occurs in response to continuous administration of GnRH is short-lived and LH levels return to pre-infusion values within 48 h of the start of infusion, these results show that the pituitary is still capable of responding to exogenous GnRH, although the LH response to an i.v. bolus injection of GnRH is reduced. In addition, this change in pituitary sensitivity is not fully reflected in endogenous patterns of episodic LH secretion.

Ovulation was induced in seasonally anoestrous ewes by repeated 2-h injections of 250 ng Gn-RH, after 12 days (Group 1, N = 7; Group 2, N = 8), 2 days (Group 3, N = 8) or no (Group 4, N = 7) progesterone pretreatment. A preovulatory LH peak occurred spontaneously at a mean (+/- s.e.m.) time of 43.1 +/- 2.0 h, 38.5 +/- 3.1 h and 26.8 +/- 1.7 h after the start of Gn-RH treatment in Groups 1, 3 and 4 respectively, and was artificially induced in ewes in Group 2, after 24 h of treatment, by a single i.v. injection of 150 micrograms Gn-RH. Normal luteal function occurred in all progesterone-pretreated ewes, but in only 1/7 animals not treated with progesterone. These results demonstrate that, although normal luteal function in progesterone-primed ewes induced to ovulate with repeated injections of low doses of Gn-RH is associated with a delayed preovulatory LH peak, it is not this extended period of follicle development which is responsible for functional competence of the resultant corpus luteum. Since as little as 2 days of exposure to elevated plasma progesterone concentrations is effective, it is suggested that progesterone may act directly on the preovulatory follice.

When starlings (Sturnus vulgaris) are transferred from short to long days, hypothalamic content of gonadotrophin-releasing hormone (GnRH) and pituitary gonadotrophin content eventually decrease, as birds become photorefractory, to values lower than they were on short days. This implies that both should increase as photorefractoriness is terminated some time after transfer from long to short days. Further, since thyroidectomy causes termination of photorefractoriness in birds held on long days, this should also result in an increase in hypothalamic GnRH and pituitary gonadotrophin contents. This study tests these hypotheses. Male starlings were transferred from short to long days for 10 weeks, by which time they should have become photorefractory. One group of birds was then killed; blood was collected, hypothalami and pituitaries were excised, and the stage of moult and testicular weights recorded. The remaining birds were then kept on long days, transferred to short days or thyroidectomized and kept on long days. Groups of birds in each treatment group were killed and sampled 2, 8 and 14 weeks later. Hypothalamic content of GnRH, and pituitary and plasma FSH and prolactin concentrations, were measured by radioimmunoassay. In birds sampled after 10 weeks of long days, hypothalamic content of GnRH was quite low, pituitary and plasma levels of FSH were very low, testes were small and pituitary and plasma prolactin levels were high. In intact birds kept on long days, hypothalamic GnRH content decreased further and remained low. Pituitary and plasma FSH levels remained low, testes remained small and pituitary and plasma prolactin levels decreased slowly. In intact birds transferred to short days, hypothalamic GnRH content increased to values significantly (PP These results demonstrate that short days activate the hypothalamus to a state of 'readiness to function' and confirm that the thyroid glands are involved in this response. Crossbred boars were (a) immunized against GnRH conjugated to human serum globulin (200 micrograms GnRH-hSG) in Freund's adjuvant at 12 weeks of age and boosted at weeks 18 and 20 (N = 10), (b) served as controls and received hSG only in adjuvant (N = 10), or castrated at weaning (N = 10). At 24 weeks of age (immediately before slaughter), the boars were challenged with saline or pig LH (1 microgram/10 kg body weight). After slaughter, fresh testicular fragments were incubated with pig LH (0.05 and 0.2 ng/2 ml medium) to assess the effects of immunization on Leydig cell function. Pituitary contents of LH and FSH, and testicular LH receptor content were also measured. The results indicated that plasma LH and testosterone concentrations, pituitary LH content, testicular LH receptor content, testis and sex accessory organ weights were significantly reduced in GnRH-immunized boars compared to hSG-adjuvant controls. However, plasma and pituitary FSH content were not affected by high antibody titres generated against GnRH. The testicular testosterone response to exogenous LH in vivo and in vitro was significantly reduced (P less than 0.05) in GnRH-immunized boars. These results indicate that active immunization against GnRH impairs pituitary and Leydig cell functions in boars.

GnRH receptor values are 30-50% of normal in pituitaries of hpg male mice, and testicular LH receptors only 8% of normal (160.4 +/- 17.6 and 2013 +/- 208.1 fmol/testis respectively). In male hpg mice bearing fetal preoptic area (POA) hypothalamic implants for 10 days there was no change in pituitary GnRH receptors, pituitary gonadotrophin content, or seminal vesicle weight. However, testicular weights and LH receptors were doubled in 4/10 mice and 2 had increased serum FSH levels. Between 26 and 40 days after implantation pituitary GnRH receptors and pituitary LH increased to normal male levels, although at 40 days serum and pituitary FSH concentrations had reached only 50% of normal values. Testicular and seminal vesicle weights increased more than 10-fold by 40 days after implantation and LH receptors to 70% of normal. In hpg female mice bearing hypothalamic implants for 30-256 days pituitary gonadotrophin concentrations were normal, even though GnRH receptors reached only 60% of normal values (6.18 +/- 0.4 and 9.8 +/- 0.4 fmol/pituitary respectively). Serum FSH was substantially increased from values of less than 30 ng/ml in hpg mice to within the normal female range in hypothalamic implant recipients. Ovarian and uterine weights increased after hypothalamic grafting from only 4-5% to over 74% of normal values. LH receptors increased from 6.5 +/- 1.3 fmol/ovary for hpg mice to 566.9 +/- 39.2 fmol/ovary for implant recipients. Vaginal opening occurred about 23 days after implantation and these animals displayed prolonged periods of oestrus.(ABSTRACT TRUNCATED AT 250 WORDS)

Standard bred mares that were cycling normally were treated beginning on Days 9 or 10 of the oestrous cycle with repeated pulses of GnRH (20 micrograms/h) and/or a single injection of prostaglandin (PG)F-2 alpha (alfaprostol, 3 mg), and were subsequently bled and palpated daily until the next ovulation. GnRH treatment increased serum concentrations of LH and progesterone at 4 days after the start of treatment compared to controls. The combination of PGF-2 alpha + GnRH treatment resulted in an immediate decline in serum progesterone values, and subsequently decreased the interval to next ovulation by 4.5 days compared to controls. Mean serum concentrations of FSH were not different among treatment groups 4 days after the start of treatment, and there was a consistent trend among all treatment groups for decreasing concentrations of FSH within the 6 days before ovulation. We conclude that, under our experimental conditions, pulsatile administration of GnRH provides a short-term luteotrophic stimulus, probably by the elevation in serum LH, but that this stimulus cannot indefinitely prevent the luteolytic effects of exogenously administered PGF-2 alpha. Although GnRH treatment combined with PGF-2 alpha injection hastened the impending ovulation, this regimen was no more effective than PGF-2 alpha treatment alone.

Frequent blood samples were collected to study hormonal responses to GnRH in male and female leopards and tigers. Animals were anaesthetized with ketamine-HCl and blood samples were collected every 5 min for 15 min before and 160 min after i.v. administration of GnRH (1 micrograms/kg body weight) or saline. No differences in serum cortisol concentrations were observed between sexes within species, but mean cortisol was 2-fold greater in leopards than tigers. GnRH induced a rapid rise in LH in all animals (18.3 +/- 0.9 min to peak). Net LH peak height above pretreatment levels was 3-fold greater in males than conspecific females and was also greater in tigers than leopards. Serum FSH increased after GnRH, although the magnitude of response was less than that observed for LH. Basal LH and FSH and GnRH-stimulated FSH concentrations were not influenced by sex or species. Serum testosterone increased within 30-40 min after GnRH in 3/3 leopard and 1/3 tiger males. Basal testosterone was 3-fold greater in tiger than leopard males. LH pulses (1-2 pulses/3 h) were detected in 60% of saline-treated animals, suggesting pulsatile gonadotrophin secretion; however, in males concomitant testosterone pulses were not observed. These results indicate that there are marked sex and species differences in basal and GnRH-stimulated hormonal responses between felids of the genus Panthera which may be related to differences in adrenal activity.

SUMMARY The concentrations of luteinizing hormone releasing hormone (LH-RH) and luteinizing hormone (LH) were determined by radioimmunoassay in blood samples taken at intervals after the administration of different doses of synthetic LH-RH administered either intravenously or intranasally in healthy fertile men. Intranasal administration of LH-RH caused a dose-dependent increase of plasma LH with the peak occurring later than that after intravenous injection. The intravenous route was approximately 100 times more effective than the intranasal route in terms of the dose of LH-RH necessary to achieve an LH response of similar magnitude, but the route through the nasal mucosa seems a safe and convenient way for LH-RH administration. The characteristics of the disappearance curve, metabolic clearance rate and volumes of distribution of LH-RH in man are compared with those found by others.

Ovulation rate, median time to first ovulation, median time of all ovulations and median time from first to last ovulation were studied by repeated laparoscopy in Merino ewes. Treatments with FSH or PMSG significantly affected ovulation rate (8.4 +/- 0.81 and 7.3 +/- 1.21 respectively, P less than 0.05) and in median time of all ovulations (60 and 54 h respectively after progestagen sponge removal, P less than 0.05). Differences in the median time to first ovulation (60 and 48 h) and median time from first to last ovulation (6 and 6 h) for the respective treatments were not significant. The synchrony of ovulation after both treatments was adversely affected by (1) the occurrence of premature ovulations before the onset of superovulation, (2) variability in the time of commencement of superovulation, and (3) variability in the time from first to last ovulation. Administration of GnRH synchronized the timing of ovulation with both gonadotrophin treatments. This synchrony was due to a reduction in the period during which superovulation began and in the interval from first to last ovulation. The median time of all ovulations was significantly less with FSH + GnRH than with PMSG + GnRH (45 and 48 h after progestagen sponge removal, respectively, P less than 0.05). Administration of GnRH at 16, 20 or 24 h after progestagen sponge removal significantly affected all traits examined except ovulation rate. Administration at 20 and 24 h produced an equally good synchrony of ovulation which was better than that obtained at 16 h. We suggest that the use of GnRH in embryo collection programmes appears justified and is likely to improve embryo yields due to improved rates of fertilization.

Since the concomitant administration of a gonadotrophin-releasing hormone (GnRH) antagonist and testosterone suppresses sperm production only incompletely, the feasibility of treatment with a GnRH antagonist and delayed testosterone supplementation for sustained suppression of sperm production in a non-human primate model was investigated. Adult cynomolgus monkeys (Macaca fascicularis; five/group) received daily s.c. injections of the GnRH antagonist [N-acetyl-d-2-naphthyl-Ala1,d-4-chloro-Phe2,d-pyridyl-Ala3,nicotinyl-Lys5,d- nicotinyl - Lys6, isopropyl-Lys8,d-Ala10]-GnRH of either 450 or 900 μg/kg for 18 weeks. During week 6 of the GnRH antagonist treatment, all monkeys were given a single i.m. injection of 40 mg of a long-acting testosterone ester (testosterone-trans-4-n-butylcyclo-hexanecarboxylate; 20-Aet-1). Within 1 week, serum LH bioactivity was suppressed in both groups and remained low throughout the entire treatment period. Similarly, concentrations of serum testosterone declined precipitously. During week 6, substitution with testosterone restored concentrations of serum testosterone into the pretreatment range. Concentrations of serum inhibin declined within 1 week and remained suppressed during the period of treatment with the GnRH antagonist. Testicular volumes were reduced to approximately 25% of pretreatment values in both groups by week 8 and stayed in that range during the remaining period of administration of the GnRH antagonist. During the first 6 weeks of administration of the GnRH antagonist, the ejaculatory response to electrostimulation and the volume of the ejaculates diminished with time. Supplementation with testosterone during week 6 restored the ejaculatory responses within 2–3 weeks. From week 9 of GnRH antagonist treatment onwards, all monkeys given 450 μg/kg and four monkeys given 900 μg/kg produced azoospermic ejaculates. The fifth animal in the latter group became azoospermic during week 13. Azoospermia persisted throughout the entire period of treatment with the GnRH antagonist and for a further 7–13 weeks. All suppressive effects of administration of GnRH antagonist were reversible. During the recovery phase the increase in testicular volumes paralleled an increase in concentrations of serum inhibin. The suppression of inhibin levels during the period of administration of testosterone indicates that Sertoli cell activity was not restimulated by testosterone. In conclusion, GnRH antagonist treatment with delayed supplementation with testosterone might serve as a model for further research towards the development of an endocrine male contraceptive. The influence of low (5 mcM) and high (200 mcM) concentrations of indomethacin on gonadotropin release from rat pituitaries was studied in vitro. Low concentrations significantly (p less than .05) reduced the release of luteinizing hormone (LH) in comparison with controls whereas high concentrations significantly (p less than .05) increased the rate of release. The release of follicle stimulating hormone (FSH) was not affected. When pituitary tissue was stimulated by LH-releasing hormone (LH-RH) the high concentration of indomethacin significantly (p less than .05) increased LH release and produced a nearly significant (p less than .01) increase in FSH. The low concentration was without effect. The effect of the addition of prostaglandins (PGs) alone and in combination with indomethacin was also investigated. PGE-1 significantly (p less than .05) increased the release of LH. However there was no significant (p greater than .1) interaction between the 2 drugs. The effects on the release of FSH were similar. The addition of PGE-2 or PGF-2alpha slightly increased the release of LH and FSH (p greater than .1). It is suggested that high concentrations of indomethacin probably do not inhibit PG synthesis but may inhibit cyclic nucleotide phosphodiesterase.

This study investigated the importance of reorganization of cell components by cytoskeletal structures to the short-term dynamic changes in LH release from dispersed sheep pituitary cells in perifusion, when stimulated with different dynamic patterns of gonadotrophin-releasing hormone (GnRH). The changes in rate of LH release investigated were the initial response to GnRH, desensitization, change of dose–response during desensitization, and recovery of sensitivity between pulses of stimulation. Cytochalasin D and colchicine were used to modify microfilament and microtubule action respectively. To determine whether receptor movement after binding of agonist was involved in the altered responses, K+ and phorbol 12-myristate 13-acetate (PMA) were used as stimulants because they cause LH release independently of agonist-receptor interaction. After 3 and 48 h culture on dextran beads and 2–3 h incubation in the presence and absence of 2–48 μmol cytochalasin D/l, or 8 or 250 μmol colchicine/l, aliquots of collagenase-dispersed sheep pituitary cells were stimulated at 37 °C in tubes or in a multicolumn perifusion system with 850 pmol GnRH/l, 109 mmol K+/l or 10 nmol PMA/l. Fractions of supernatant or effluent were collected at intervals and LH concentrations measured by radioimmunoassay. Control samples were treated in the same way but without stimulation. Maximal, reversible enhancement of LH release over the first 20 min following stimulation with all secretogogues was observed after incubation of cells in 6 μmol cytochalasin/l. Desensitization behaviour, the supramaximal response, and the ability of cells to recover sensitivity to repeated pulses of GnRH were not altered by this modifier of microfilament polymerization at 6 or 24 μmol/ml. Colchicine at 8 μmol/l caused no changes in LH release. At 250 μmol/l, colchicine reduced the initial response of cells to GnRH stimulation but its action at this relatively high level may not be specific; there was no other major change in desensitization patterns, nor recovery of sensitivity to pulsed GnRH stimulation. Each treatment affected cellular responses similarly before and after culture. From studying the details of the dynamics of the short-term responses of gonadotrophs, we conclude that transport of cell components involving microfilaments and microtubules is unlikely to be a major limitation on the rate of LH release during desensitization, the supramaximal response, or the recovery of sensitivity between pulses of GnRH. This suggests that biochemical reactions rather than physical translocation may be rate-limiting in these processes. In addition, although inhibition of microfilament action does appear to enhance the earliest observed response to stimulation of the LH-release mechanism, this occurs after protein kinase C activation and is probably not related to impairment of processes such as polymerization and sequestration of agonist-bound GnRH receptors because the effects are also observed with K+ and PMA, stimulants acting independently of agonist-receptor interaction. Anoestrous ewes (N = 3) were treated with a 500 ng GnRH pulse administered via a jugular cannula every 2 h for 40 to 80 days. Plasma concentrations and therefore presumed ovarian activity changed cyclically with each progestational cycle (n = 10) lasting 14.0--18.5 days. It is concluded that, by increasing the frequency of GnRH secretory episodes from an apparent endogenous level of one episode per 3.6 h to at least one every 2.0 h, cyclic ovarian activity can be restored to seasonally anoestrous sheep.

In bulls there was no increase in plasma FSH and only a small increase in LH over the first 14 weeks of age. In steers (castrated) plasma LH and FSH were unchanged for the first 3 weeks but increased significantly at 7 and 14 weeks. After 100 micrograms Gn-RH, LH release in bulls was minimal until 7 and 14 weeks and there was no comparable rise for FSH. LH and FSH responded to Gn-RH throughout the trial in the steers. The neonatal calf testes selectively inhibited the release of FSH from the pituitary even when challenged with Gn-RH.

The effects of long-term treatment with physiological doses of oestradiol or oestradiol plus progesterone on plasma gonadotrophin levels and pituitary content of LH and gonadotrophin-releasing hormone (GnRH) receptors were studied in ovariectomized–hypothalamo-pituitary disconnected ewes given 250 ng pulses of GnRH every 2 h (i.v.). A pilot experiment showed that 3 cm long Silastic implants (s.c.) reduced both LH pulse frequency and pulse amplitude in long-term (> 6 months) ovariectomized ewes. The main experiment was conducted over 3 weeks in ovariectomized–hypothalamo-pituitary disconnected ewes that had received pulsatile GnRH replacement for 1 week after pituitary surgery. Group 1 (n = 5) received GnRH pulses alone throughout the study. Group 2 (n = 6) received oestradiol in week 2 and oestradiol plus progesterone in week 3 and in group 3 (n = 6) the steroid treatments were reversed. Oestradiol reduced (P < 0·05) the mean (± s.e.m.) amplitude of LH in pulses in group 2 (from 8·2 ± 1·6 to 5·0 ± 0·5 μg/l) and group 3 (from 11·6 ± 1·2 to 9·3 ±1·0 μg/l); an additional effect of progesterone was seen in group 2 but not group 3. The amplitudes of the LH pulses did not change in the control ewes. Plasma concentrations of FSH were reduced by approximately 50% by the oestradiol treatments with no additional effects of progesterone. There was no effect of steroidal treatment on pituitary content of LH or pituitary levels of GnRH receptors. We conclude that long-term oestradiol treatment, with or without progesterone, reduces plasma amplitudes of LH pulses by a direct pituitary effect, but the magnitude of this effect was less than that observed on GnRH secretion in short-term ovariectomized ewes in an earlier study. The microsphere technique was used to obtain estimates of ovarian capillary blood flow near ovulation, in 8 seasonally anoestrous ewes, which were induced to ovulate by GnRH therapy. Plasma progesterone concentrations were monitored in jugular blood sampled between Days 4 and 7 after the onset of the preovulatory LH surge. The ewes were then slaughtered. Three of the ewes were treated with a single injection of 20 mg progesterone before GnRH therapy. In these ewes and 1 other, plasma progesterone values increased after ovulation and reached 1.0 ng/ml on Day 7 following the preovulatory LH surge (normal, functional CL), whilst in the other 4 ewes progesterone concentrations increased initially then declined to 0.5 ng/ml by Day 7 (abnormal CL). In the ewes exhibiting normal luteal function, the mean ovarian capillary blood flow was significantly greater (P less than 0.01) than that for ewes having abnormal luteal function. Irrespective of the type of CL produced, capillary blood flow was significantly greater (P less than 0.05) in ovulatory ovaries than in non-ovulatory ovaries. These findings indicate that the rate of capillary blood flow in ovaries near ovulation may be a critical factor in normal development and maturation of preovulatory follicles and function of subsequently formed CL.

We have developed a non-surgical technique for longterm collection of pituitary venous blood which consists of slightly diluted hypophysial portal blood into which pituitary hormones have been secreted. In these experiments jugular and pituitary venous blood samples were collected from five unmedicated, ambulatory mares at 5-min intervals for 2–6 h on 11 occasions during the 6 days surrounding the ovulatory LH peak. Jugular blood only was collected from another five periovulatory mares without pituitary cannulae. The duration of oestrus was similar in mares with and without pituitary cannulae and all mares ovulated, showing that the procedure did not affect the reproductive axis. In all pituitary-cannulated mares the secretion of gonadotrophin-releasing hormone (GnRH), FSH and LH occurred almost continuously with broad, concurrent pulses of the three hormones superimposed upon this tonic background. Only 9% of the GnRH pulses appeared to be ineffective in inducing a rise in gonadotrophin levels. When measured in pituitary blood, gonadotrophin pulse frequency varied from 0·45 pulses/h early in the LH surge to 1·87 pulses/h at the time of ovulation. In contrast, mean pulse frequency measured in jugular blood did not exceed 1 pulse/h throughout the periovulatory period in cannulated or non-cannulated mares. The low amplitude of jugular pulses (< 50% fractional increase) may have caused problems in identifying the pulses. In the two mares in which pituitary venous blood was sampled during more than one period before ovulation, GnRH secretion tended to be lower on the day of ovulation (day 0) than earlier in oestrus (ratio day 0:day −1; mare WV = 0·58, mare LS = 0·66), whereas LH secretion rate was higher on the day of ovulation (ratio day 0:day −1; mare WV = 1·54, mare LS = 6·68). These studies show that the painless and non-invasive collection of pituitary venous blood, which is possible only in horses, can provide a useful tool for studying hypothalamic-pituitary interactions under completely physiological conditions. Anoestrous Romney Marsh ewes with and without progesterone treatment (+P, -P) were treated with small-dose (250 ng) multiple injections of GnRH at 2-h intervals for 48 h. Animals were slaughtered on Days 4, 5, 7 and 11 after the end of GnRH treatment and luteal function was assessed by the measurement of daily plasma progesterone concentrations. In all animals which ovulated (29/32, 91%) peripheral progesterone concentrations rose to 0.5-1.0 ng/ml within 3 days of the end of GnRH treatment. In 7/7 (100%) +P animals and 5/22 (23%) -P animals, progesterone concentrations continued to rise and were maintained at levels greater than 1.5 ng/ml until slaughter. In the remaining -P animals, plasma progesterone concentrations declined to reach basal levels by Day 5. Corpora lutea recovered from these animals showed signs of premature regression on Day 5 and were fully regressed by Day 7. Progesterone priming delayed the occurrence of the LH surge which occurred 39.1 +/- 3.6 h after the end of GnRH treatment in the +P animals compared to 20.2 +/- 1.74 h (P less than 0.001) in the -P animals in which luteal function was abnormal and 22.4 +/- 4.35 h in the -P animals in which luteal function was normal. These results show that abnormal luteal function occurs in the majority of GnRH-treated ewes in the absence of progesterone pretreatment.(ABSTRACT TRUNCATED AT 250 WORDS)

Four groups, each of 5 seasonally anoestrous ewes, were treated i.v with small doses (75, 125, 250 or 500 ng) of Gn-RH at 2-h intervals for 48 h. A further 15 ewes received 14 days pretreatment with progesterone and then the 250 ng Gn-RH treatment. Gn-RH injections induced an episodic pattern of LH secretion which differed significantly for the doses of Gn-RH used. A preovulatory LH surge occurred in all but 1 of the ewes during the period of Gn-RH treatment. Ovulation occurred in all 15 ewes pretreated with progesterone and in 19/20 ewes treated with Gn-RH alone. Although normal luteal function occurred in all ewes pretreated with progesterone, it was present in only 5 of the 20 ewes treated with Gn-RH alone. Oestrus, as shown by mating, occurred at a mean time of 34.7 +/- 2.6 h after the start of Gn-RH treatment only in those ewes receiving progesterone pretreatment. These results indicate that progesterone pretreatment has a marked effect on the ability of small doses of Gn-RH to induce ovulation and normal luteal function in seasonally anoestrous ewes.

Anoestrous Romney Marsh ewes with (+P) and without (-P) progesterone pretreatment were induced to ovulate by multiple low-dose injection of GnRH followed by a bolus injection of GnRH. Luteal function was assessed by twice daily measurement of plasma progesterone. Animals were slaughtered on Days 3 or 5 after the end of GnRH treatment and CL and endometrium were recovered. In all Day-5 ewes, blood samples were collected at 30-min intervals for 8 h on Days 3 and 5 for measurement of PGFM and oxytocin. At slaughter 92% of the Group +P ewes had ovulated compared with 54% of the Group -P ewes. The ovaries of some of the Group -P ewes only contained luteinized cysts either alone or in association with CL. In the ewes that ovulated, progesterone profiles were normal in all Group +P ewes, whereas Group -P ewes had 'normal' or 'abnormal' profiles in which plasma progesterone was declining prematurely. All of the CL from ewes with abnormal progesterone profiles were associated with follicular cysts, and were significantly smaller and with a lower progesterone content on Day 5. PGFM levels decreased (P less than 0.05) between Days 3 and 5 in ewes in Groups +P and -P with 'normal' CL but increased (P less than 0.01) in Group -P ewes with 'abnormal' CL. Oxytocin levels were lower in Group -P ewes with 'abnormal' CL on Day 5, than in 'normal' ewes in Groups -P (P less than 0.01) or +P (P less than 0.05). In 3/5 Day-5 ewes with 'abnormal' CL there was a clear association between a major peak of oxytocin and a rise in PGFM during the frequent sampling period on Day 3 or Day 5, and endometrial oxytocin binding sites were present at slaughter. This suggests that the premature regression of 'abnormal' CL occurs via the normal luteolytic mechanism. Although ewes in Groups +P and -P with 'normal' CL had similar progesterone profiles, plasma oxytocin was significantly higher (P less than 0.05) in the Group -P ewes and oxytocin binding sites were present only in this group, suggesting that progesterone pretreatment can influence the production of both oxytocin and its receptor.

Domperidone at a dose of 4 mg/kg significantly raised circulating prolactin levels in male and female rats although this increase was approximately 4-fold higher in females compared with males. No effect on LH secretion was observed. This LH and prolactin responses to Gn-RH and TRH respectively were investigated with isolated perfused pituitary glands obtained from domperidone-pretreated animals. For females, the basal release of prolactin was raised by domperidone pretreatment and the responsiveness of the pituitary to Gn-RH was markedly attenuated. When domperidone was added to the perfusing media, high concentrations (100 microgram/ml) reduced both the basal and TRH-stimulated prolactin secretion from pituitaries of untreated male and female rats although lower doses (1 microgram/ml and 10 ng/ml) were ineffective in altering prolactin release. The pituitary LH responses to Gn-RH were similarly reduced by the presence of 100 microgram domperidone/ml in the medium.

The effects of a single injection of estradiol-17beta and of synthetic luteinizing hormone-releasing hormone (LH-RH) on plasma LH levels in freemartin heifers were studied. Only 7 of 12 animals showed a surge in plasma LH levels within 24 hours of injection with estradiol-17beta. The increases were less than those reported for normal noncyclic heifers though some of the freemartins showed a delayed or more prolonged LH response. The LH response to estradiol was not related to the degree of chimerism or to plasma androstenedione levels. The same treatment 4 months later evoked a similar response even though plasma androstenedione levels had doubled within this period. Injection of LH-RH produced a greater release of LH in freemartins who responded to estradiol treatment than in nonresponders. It is suggested that this difference may involve pituitary dysfunction.

SUMMARY Release of luteinizing hormone (LH) and follicle-stimulating hormone (FSH) from organ cultures of human foetal pituitaries was stimulated by ovine hypothalamic extract (OHE), synthetic gonadotrophic hormone releasing factor and dibutyryl cyclic AMP. In each case, LH release was greater than that of FSH. Synthetic gonadotrophic hormone releasing factor produced an increase in the total FSH and LH content of the cultures. Oestradiol-17β stimulated release of FSH but not LH. Testosterone, progesterone and 5α-dihydrotestosterone were without effect on hormone release. Chronic exposure of cultures to progesterone prevented stimulation of hormone release by OHE. Long-term contact with oestradiol-17β did not modify tissue sensitivity to OHE.

Experiments were conducted in ovariectomized ewes after hypothalamo-pituitary disconnection (HPD) (a) to examine LH and FSH secretion during constant infusion of gonadotrophin-releasing hormone (GnRH) or physiological saline and (b) to determine whether or not a constant GnRH background enhances or diminishes pituitary responsiveness to GnRH pulses. Whereas pulsatile GnRH infusions maintained LH and FSH secretion, constant infusions (125 or 250 ng/h) led to the complete cessation of LH secretion and reduced FSH secretion. The rate of decline of plasma FSH concentrations was significantly (P J. Endocr. (1986) 111, 43–49

Significant uterine growth occurred in normal and hypogonadal (hpg) mice between Days 7 and 21 but thereafter no further growth was observed in hpg mice. The ovaries of hpg mice were significantly smaller than those of normals at all ages, but there was no significant difference between the number of non-growing follicles in the ovaries of mutants and their normal littermates at any age studied, and normal and hpg mice showed a marked reduction in the number of non-growing follicles during the first month of life. The size and composition of the growing follicle population in hpg mice, however, differed markedly from those in normal animals and by 21 days of age the number of growing follicles in mutants was significantly reduced. There was no significant difference in the number of Type 3b follicles before 60 days of age, but the number of all other follicle types was significantly less in hpg mice at all ages studied. Follicles in which the antrum is fully developed (Type 7 and 8) were never seen in the ovaries of mutants and corpora lutea were never observed. Interstitial tissue development was also very poor in hpg ovaries. The hypothalamic GnRH content in normal mice remained low until Day 20, before rising sharply to adult levels (approximately 800 pg) between Days 20 and 30. The pituitary FSH content increased over the first 10 days of life to reach a peak of about 5000 ng, before declining to the adult value of about 2000 ng by Day 30, whilst the plasma FSH concentration was high in the first 10 days, but fell to adult levels over the next 20 days. Pituitary LH content increased significantly between Days 5 and 10 to reach the adult level of about 600 ng. Hypothalamic GnRH was undetectable at all ages in hypogonadal mice, but the pituitary content of FSH and LH had risen to the attenuated mutant adult value by Day 15, and unlike normals, plasma FSH concentrations were not elevated during the neonatal period. These results suggest that minimal gonadotrophic stimulation of the ovary from birth has no effect on the total number of follicles but reduces the number of growing follicles and prevents follicle growth beyond the early antral stage. Gonadotrophins therefore appear to have a role in the initiation and continuance of follicle growth in the adult mouse.

Intact and chronically ovariectomized ewes were treated for 4 days with charcoal-treated bovine follicular fluid (FF) or charcoal-treated bovine serum during the late-anoestrous period, and the effects on basal and gonadotrophin-releasing hormone (GnRH)-induced secretion of LH and FSH observed. Subsequently, ewes received s.c. implants containing a sustained-release formulation of a potent GnRH agonist d-Ser(But)6-Azgly10-LHRH (ICI 118630) to desensitize pituitary gonadotrophs to hypothalamic stimulation, and the effects of bovine FF and bovine serum were re-assessed 2 weeks later. Chronic exposure (for 2–3 weeks) to ICI 118630 significantly reduced basal levels of LH and FSH in both intact and ovariectomized ewes and completely abolished both spontaneous LH pulses as well as exogenous GnRH-induced acute increases in plasma LH and FSH levels. Treatment with bovine FF significantly reduced plasma FSH levels, but not LH levels, in both intact and ovariectomized ewes before and after chronic exposure to ICI 118630. In intact ewes before exposure to ICI 118630, treatment with bovine FF actually enhanced pulsatile LH secretion and raised mean plasma LH levels by 240% (P in vivo, irrespective of whether a functionally intact hypothalamo-pituitary-ovarian axis is present. This article reviews the neuroendocrine factors which control the menstrual cycle in the macaque monkey. It describes the pulsatile characteristics of gonadotrophin secretion, the control of LH pulses by an arcuate neural Gn-RH oscillator and the significance of pulsatile Gn-RH secretion. The factors which may modulate the activity of the Gn-RH arcuate neural oscillator are anaesthesia, ovarian hormones and endogenous opiates, as well as the possible significance of changes in Gn-RH pulsatile characteristics. Finally, the oestrogen and progesterone feedback control of the mid-cycle gonadotrophin surge and the site of action (hypothalamic or hypophysial) of these steroids are contrasted in the monkey and rat.

An investigation of pituitary sensitivity, assessed in terms of increments in plasma LH and FSH concentrations, to stimulation with one or two injections of gonadotrophin releasing hormone (GnRH) was carried out on 26-day-old immature female rats which had received one of the following priming treatments: 10 μg oestradiol benzoate (OB) as a single injection on day 23 or day 25, or on both days; 10 i.u. pregnant mare serum gonadotrophin (PMSG) on day 24; an electrochemical brain lesion placed in the mediobasal hypothalamus on day 23; control animals received either vehicle alone or a sham lesion. Pituitary sensitivity assessed at 10.00 h on day 26, after one or two injections of GnRH (100 ng/100 g body weight, s.c.), was enhanced to a similar degree in the three groups treated with OB in terms of LH (P < 0-01). The FSH response also increased after OB treatment but was not statistically significant. In contrast, 48 h after the injection of PMSG (i.e. when the rats were in a 'pro-oestrous-like' condition) pituitary sensitivity in terms of both LH and FSH dropped sharply (P < 0·001). In lesioned animals, pituitary sensitivity to one injection of GnRH was unchanged. A second GnRH injection administered after a 60 min interval induced a slightly larger LH response in control animals. In contrast, the ratio of the second response to the first increased in animals treated with PMSG, despite the state of overall decrease in sensitivity, being 4·5:1 in PMSG-treated rats versus 1·4:1 in controls. In a second set of experiments, we investigated the variation of pituitary sensitivity in conjunction with an experimentally induced gonadotrophin surge. In animals treated with OB on day 23 and with 1 mg progesterone at 12·00 h on day 26, pituitary sensitivity was increased at both 14.00 and 17.00 h as compared with that in the day 23 OB-treated group at 10.00 h. The PMSG-treated animals maintained their state of decreased responsiveness at 14.00 h, but exhibited increased pituitary sensitivity at the time of the gonadotrophin surge (17.00 h). These results show that OB increases pituitary sensitivity to GnRH in 26-day-old female rats and that the induction of a gonadotrophin surge further increases this sensitivity. In contrast, PMSG-treated rats displayed a state of decreased responsiveness 48 and 52 h, but not 55 h, after the injection. Pituitary sensitivity on the second day after PMSG treatment thus clearly differs from that observed during pro-oestrus in the adult cyclic female rat.

The suppression of ovulation in subordinate female marmosets was associated with suppressed pituitary LH secretion and reduced pituitary LH response to gonadotrophin-releasing hormone (GnRH). In subordinate females, basal plasma LH concentrations were commonly below 2 IU/1 (n = 5) (maximum 10·7 IU/1). Plasma oestrogen concentrations were similarly low (maximum 0·62 nmol/l) and plasma progesterone concentrations of below 30 nmol/l confirmed the anovulatory condition. This infertility condition was rapidly reversed when subordinate females (n = 5) were removed from their social groups and housed singly, when plasma LH (maximum 140·0 IU/1) and oestrogen (maximum 7·84 nmol/l) concentrations increased preceding ovulation. Infertility was rapidly reimposed when these singly housed females were re-introduced to subordinate status in new social groups, when plasma LH concentrations fell to their previous low values within 4 days; no ovulation occurred thereafter. Plasma oestrogen levels also fell, but less dramatically. The luteal phases of three of the subordinate females were shortened following the re-instatement of subordinate status. The maximum LH response of subordinate females to the highest dose of GnRH (200 ng) was only 19·1±6·7 IU/1 (mean ± s.e.m.; n=8): this contrasted with that in dominant females in either the follicular phase (40·0±13·3 IU/1; n=6) or the luteal phase (126·7±24·9 IU/1; n=10) of the ovarian cycle. These results suggest that the social suppression of fertility in subordinate female marmosets is mediated by impaired hypothalamic GnRH secretion. Such an immediate and precise behavioural control of LH secretion and ovulation is without equal in anthropoid primates. J. Endocr. (1988) 117, 329–339

Prepubertal ewes can, under certain circumstances, be stimulated to ovulate by the novel introduction of a ram. The endocrine response to the presence of the ram is characterized by a rapid increase in the frequency of episodic release of LH. The purpose of this study was to investigate the effect of the presence of a ram on LH pulse frequency in vivo, gonadotrophin-releasing hormone (GnRH) and β-endorphin concentrations in the median eminence, and on the influence of the endogenous opioid peptide agonist [d-Ala2,N-Phe4,Met(0)ol5]-enkephalin (FK 33–824) on basal and depolarization-induced release of GnRH from median eminence tissue superfused in vitro. The study was performed at two prepubertal ages in August and September. In September, the introduction of a ram resulted in an increase in pulsatile release of LH, which was associated with an increase in the rate of basal release of GnRH from median eminence tissue superfused in vitro, and the development of a marked ability of FK 33–824 to suppress depolarization-induced release of GnRH. The concentration of β-endorphin in the median eminence was reduced in animals exposed to the ram at this time. In contrast, the introduction of a ram in August failed to stimulate an increase in LH pulse frequency, basal release of GnRH in vitro was not altered and FK 33–824 was ineffective in reducing depolarization-induced release of GnRH. These results suggest that the premature onset of reproductive activity induced by exposure to the ram may involve the participation of the endogenous opioid peptide system. J. Endocr. Pituitary–testicular relationships in mature male rats were investigated during the period of germinal involution after the induction of vitamin A deficiency (VAD). Vitamin A deficiency caused a decrease in testicular weight, a gradual increase in the incidence of delayed spermiation, increased phagocytosis of spermatids and pyknosis of germ cell nuclei in rats aged 80 to 110 days. Both basal and gonadotrophin releasing hormone (GnRH)-stimulated serum FSH concentrations were increased by 100 days of age. During the same period, the per cent increment in GnRH-stimulated FSH secretion, pituitary FSH concentration and LH secretion remained unchanged. These results suggest that the increased serum FSH may mark specifically an alteration in the germinal epithelium. By 140 days of age, spermatogenic activity in the rats with VAD was limited to the spermatogonial proliferations so that only Sertoli cells, spermatogonia and preleptotene spermatocytes remained. At this time hypersecretion of FSH persisted while the per cent increment of GnRH-stimulated FSH secretion decreased. Concomitantly, basal and GnRH-stimulated LH concentrations were also increased in the presence of normal serum testosterone. These results indicate that a complete cessation of spermatogenesis beyond preleptotene spermatocytes is associated with a change in the secretion of both FSH and LH. The relationship between serum LH and testosterone was normal until at least 110 days of age. By 140 days the ratio between basal LH and basal testosterone, and between total LH and total testosterone, after GnRH administration, increased in the rats with VAD. These changes may be caused by a hyporesponsiveness of the Leydig cells which may, in turn, be attributed to the cessation of spermatogenesis. J. Endocr. (1984) 100, 33–41

Acyclic beef cows received 1.0, 2.5 or 5.0 micrograms GnRH/2 h for 48 h as 24 X 2 h repeated i.v. injections or by continuous i.v. infusion. Preovulatory-type LH surges were detected in 9/18 injected and 8/15 infused cows and occurred 30.6 +/- 5.1 h and 3.3 +/- 0.7 h after the start of treatment respectively. Cows receiving the lowest infusion dose did not exhibit gonadotrophin surges. The LH response to individual injections increased with dose but the proportion of injected cows showing preovulatory-type surges at each dose level did not change. A total of 20 cows (10 injected and 10 infused) showed evidence of luteal activity within 7 days of the end of GnRH treatment, although this was transitory in most animals. Cows which exhibited preovulatory-type LH surges in response to treatment had significantly higher plasma oestradiol-17 beta concentrations and lower FSH concentrations before treatment than those which did not. The results suggest that the LH response to GnRH treatment is dependent on follicular status in the immediate pretreatment period.

Circhoral administration (250 ng/h, i.v.) of GnRH induced a preovulatory-like surge of LH and subsequent luteal function in 4 of 4 ewe lambs 1 month before expected date of puberty. Within 12h of the start of pulsatile delivery of GnRH, mean concentrations of immunoactive and bioactive LH increased significantly (P less than 0.05) and the LH surge occurred by 1.8 +/- 0.6 days of treatment. Mean concentrations of serum progesterone were elevated significantly (P less than 0.001) 3 days after the surge. The biopotency of LH (bioactive LH/immunoactive LH) before the GnRH-induced surge of LH did not differ from LH biopotency in ewe lambs receiving circhoral delivery of saline (0.41 +/- 0.05 and 0.46 +/- 0.04, respectively). Biopotency of LH declined markedly at the GnRH-induced LH surge (0.25 +/- 0.04), but biopotency of serum LH was significantly augmented (P less than 0.05) during the period of luteal activity (0.70 +/- 0.07). Regular oestrous cycles were observed in 3 of 4 ewe lambs after the 10-day GnRH treatment period. These results indicate that pulsatile delivery of GnRH is effective in inducing precocious puberty in ewe lambs. Increase in LH biopotency does not appear to be required in the pubertal transition to reproductive cyclicity in this species. Augmented LH biopotency may be important in support of luteal function after first ovulation.

A specific sheep LH radioimmunoassay was validated for the measurement of goat LH, and used to monitor luteal-phase LH episodes and the preavulatory LH surge in progestagen sponge-synchronized cycling goats. No luteal-phase LH episodes were detected during 12 h of frequent (15-min) blood sampling in 2 goats. A preovulatory LH surge was recorded in 5/5 goats, with a mean amplitude of 45.4 +/- 7.2 ng/ml and a mean time of onset of 38.4 +/- 1.2 h after removal of a progestagen-impregnated sponge. In anoestrous goats, single i.v. injections of 1000 and 2000 ng GnRH induced LH episodes with a mean amplitude of 2.04 +/- 0.11 and 3.67 +/- 0.06 ng/ml respectively, but injections of 250 or 500 ng did not consistently elevate LH concentrations. Progestagen-primed, seasonally anoestrous lactating goats were treated with repeated injections of 1500 ng GnRH (every 2 h for 52 or 78 h) in May 1985 or 1986. All 10 had kidded in March of the same year, and were consequently at peak lactation at the time of GnRH treatment. A preovulatory LH surge was detected in 9 goats with a mean time of onset of 59.5 +/- 2.9 h (1985) or 39.6 +/- 3.3 h (1986) after vaginal sponge removal. All animals displayed oestrus and ovulated, and 9 of the goats were mated: in 5 of these animals pregnancies were successfully carried to term. The results show episodic LH release in response to GnRH and indicate that ovulation can be induced in seasonally anoestrous goats, even at peak lactation, and normal pregnancies may result.

The effect of hyperprolactinaemia induced by dopamine-antagonist drugs on pituitary responsiveness to gonadotrophin releasing hormone (Gn-RH) and on plasma LH levels has been investigated in intact and ovariectomized rats. The animals were pretreated with haloperidol, pimozide or domperidone and the sensitivity of isolated pituitary glands to pulses of Gn-RH were tested using a perifusion system. Trunk blood, collected at the time of killing, was assayed for plasma LH and prolactin. In addition, a direct effect of the drugs on pituitary responsiveness to Gn-RH was investigated by perifusing pituitary glands taken from untreated, pro-oestrous rats in medium containing the dopamine antagonists. The pituitary responsiveness was significantly impaired in intact rats after treatment with each of the drugs whereas no effect was observed in pretreated ovariectomized rats. None of the drug treatments altered levels of circulating LH. High concentrations of the drugs present in the perifusion medium also inhibited pituitary responsiveness although it is not known whether the concentrations of the drugs present in the pretreated animals would exert a similar effect. The results suggest that short-term hyperprolactinaemia impairs pituitary responsiveness through a modulation of ovarian steroid secretion and that Gn-RH release is not altered. Treatment with domperidone exhibited similar effects on the parameters measured here to those caused by the two neuroleptics, indicating that this novel anti-dopaminergic drug is acting in a similar manner.

Mares at different stages of the oestrous cycle were given a single intravenous injection of 0.5 mg synthetic Gn-RH. The mean area of the induced LH peak was significantly less at mid-cycle (Day 10-11) than at any other time. The mean height of the LH peak above preinjection concentration was greater at late oestrus and early cycle (Day 5-6) than at mid-cycle and early oestrus. There were no significant different in mean FSH responses. The LH:FSH ratio for both height and area of induced peaks was significantly less at mid-cycle than at other times of injection. These results suggest that one releasing hormone could cause the release of both FSH and LH in the normal cyclic mare.

Blood samples were taken at 15-min intervals from prepubertal beef heifers, 6 aged 4 months and 6 aged 10 months, to determine plasma LH and FSH concentrations. Two animals of each age group were then subjected to 9 consecutive injections of 0.5, 2.0 or 5.0 micrograms Gn-RH at 2-h intervals. LH episodes were evident in all animals before treatment, and in 11/12 heifers occurred at a rate of between 1 and 4 per 24 h. One 10-month old heifer, later detected in oestrus, displayed 10 episodes/24 h. Mean FSH concentrations did not differ significantly between age groups. In all animals, Gn-RH injections induced an episodic pattern of LH secretion which was significantly different between Gn-RH doses, but not between age groups. A distinct episode-like response in FSH concentrations occurred to each Gn-RH injection in only one heifer. These results provide further evidence that the prepubertal period is associated with a lack of pituitary stimulation, rather than an inability to respond to Gn-RH.

Normal and abnormal corpora lutea were recovered from anoestrous Romney Marsh ewes on Days 3, 4, 5 and 6 after treatment with small-dose (250 ng) multiple injections of GnRH followed by a bolus injection (125 micrograms) with (+P) and without (-P) progesterone pretreatment and a study made of their characteristics in vitro. Plasma progesterone concentrations initially rose concurrently in all animals but abnormal luteal function occurred in 70% of the -P ewes and was defined on Day 5 when plasma progesterone concentrations declined relative to those in the +P ewes. All corpora lutea recovered on Days 3 and 4 appeared macroscopically similar and there were no significant differences between the +P and -P groups in terms of luteal weight, progesterone content and binding of 125I-labelled hCG on these days. However, corpora lutea from the -P animals only exhibited a decline in progesterone production in vitro on Day 4 (P less than 0.01), and morphological differences became apparent on Days 5 and 6 when the abnormal corpora lutea from the -P animals also decreased in weight (P less than 0.01) and progesterone content (P less than 0.001). Binding of 125I-labelled hCG increased on Day 5 in the normal corpora lutea only. These results show that, although abnormal luteal function induced by GnRH treatment of anoestrous ewes could not be distinguished from normal corpora lutea before Day 5 by measurement of progesterone in peripheral plasma, a significant decline in progesterone production in vitro occurred on Day 4 in the abnormal corpora lutea. This was followed by significant decreases in weight and progesterone content and a failure to increase 125I-labelled hCG binding. Abnormal corpora lutea are therefore capable of some initial growth and progesterone production, before undergoing a rapid and premature regression from Day 4, which has similar characteristics to natural luteolysis.
