Full spectrum flow cytometry brings a breakthrough for minimal residual disease (MRD) detection in acute myeloid leukemia (AML). We aimed to explore the role of a new panel in MRD detection. We established a 24-color full-spectrum flow cytometry panel. A tube of 24-color antibodies included CD45, CD117, CD34, HLA-DR, CD15, CD64, CD14, CD11c, CD11b, CD13, CD33, CD371, CD7, CD56, CD19, CD4, CD2, CD123, CD200, CD38, CD96, CD71, CD36, and CD9. We discovered that when a tube meets 26 parameters (24 colors), these markers were not only limited to the observation of MRD in AML, but also could be used for fine clustering of bone marrow cells. Mast cells, basophils, myeloid dendritic cells, and plasmacoid dendritic cells were more clearly observed. In addition, immune checkpoint CD96 had the higher expression in CD117+ myeloid naive cells and CD56dimNK cells, while had the lower expression in CD56briNK cells in AML-MRD samples than in normal bone marrow samples. CD200 expression was remarkably enhanced in CD117+ myeloid naive cells, CD4+ T cells, T cells, activated T cells, CD56dimNK cells, and CD56briNK cells in AML-MRD samples. This study attempted to research the molecular mechanism underlying the inhibitory role of miR-1225-5p in the malignant progression of glioblastoma. Bioinformatics analyses based on the gene expression omnibus (GEO) and Chinese glioma genome atlas (CGGA) databases showed that miR-1225-5p, as a favorable prognostic factor, was expressed at low levels in glioblastoma, and its expression was also related to WHO grade and age. The subsequent CCK-8 assay indicated that miR-1225-5p might prevent the malignant progression of glioblastoma, which was represented by that miR-1225-5p mimic reduced the viability of glioblastoma cells. Then, we predicted that FNDC3B might be a potential target gene of miR-1225-5p, and it was negatively correlated with the level of miR-1225-5p, which were confirmed by dual-luciferase reporter assay, qRT-PCR and western blot assays. Moreover, based on the analyses of the cancer genome atlas (TCGA), Oncomine and CGGA databases, FNDC3B was enriched in glioblastoma and high expression of FNDC3B led to poor prognosis. Finally, CCK8 and transwell experiments showed that the ability of miR-1225-5p to inhibit glioblastoma cell viability, invasion and migration was at least partially achieved by targeting FNDC3B. In general, these results revealed that the miR-1225-5p/ FNDC3B axis contributes to inhibiting the malignant phenotype of glioblastoma cells, which lays a foundation for molecular diagnosis and treatment of glioblastoma. Introduction. This study aims to investigate the potential effects of regional hyperthermia combined with chemotherapy (RHCT) as a treatment strategy for advanced gastric cancer (AGC). Method. 118 AGC patients were randomly divided into treatment plans with chemotherapy (CT) alone or with RHCT. The prognostic value of clinicopathologic characteristics was assessed in terms of overall survival of AGC patients. Results. The disease control rate was determined to be 70.9% and 46.0% for the RHCT and CT group, respectively (P = 0.006). The median survival was determined to be 23.5 months for the RHCT group and 14.0 months for the CT group (P = 0.010). The 3-year survival rate for the RHCT group was 11.4% and 0% for the CT group (P = 0.018). No difference in grade 3 or 4 adverse events was observed between the two groups (P > 0.05). Multivariate analysis showed that hyperthermia, disease stage, Glasgow prognostic score, and abdominal metastasis were closely associated with the prognosis of these AGC patients. Conclusion. Objective. To investigate the effect of gastrointestinal function on the incidence of ventilator-associated pneumonia (VAP) in critically ill patients. Methods. From August 2012 to June 2016, 160 critically ill patients in the ICU (Intensive Care Unit) of our hospital were selected as the research group; patients were divided equally into an observation group and a control group, 80 patients in each group, based on the random draw envelope principle. The control group was given a nasogastric tube for gastric feeding, the observation group was given a dual lumen gastrointestinal enteral device for gastric feeding; the two groups’ enteral nutrition observation time was 7d; any changes in patient condition and prognosis were recorded. Results. The pH value of gastric juice in the control group and the observation group was 6.13±1.38 and 4.01±1.83, respectively: the pH for the observation group was significantly lower than that of the control group (t=4.982, P<0.05). The incidence of VAP in the observation group and the control group was 2.5% and 12.5%, respectively: the VAP for the observation group was significantly lower than that of the control group (P<0.05). The serum levels of pre-albumin and albumin after feeding in the two groups were significantly higher than before feeding (P<0.05); the serum levels of pre-albumin and albumin in the observation group after feeding were significantly higher than those in the control group (P<0.05). The mechanical ventilation time and ICU length of stay in the observation group were 9.12±2.13 days and 12.76±1.98 days, respectively, significantly lower than those of the control group of 10.56±2.89 days and 16.33±2.11 days (P<0.05). Conclusion. Obstacles to gastrointestinal function in critically ill ICU patients are common; enteral gastric feeding by dual lumen gastrointestinal for can improve the patient’s nutritional status, promote and maintain the normal pH value of gastric juice, thereby reducing the incidence of VAP through rehabilitation of patients. The effects of ultrasonication on the physicochemical properties of bovine serum albumin (BSA)-glucose and the galactose conjugates formed by glycation was investigated. Fourier transform ion cyclotron resonance mass spectra analysis showed that the conjugates had a higher molecular weight than the native and ultrasonicated BSA. Ultrasonicated BSA had significantly higher emulsifying and foaming properties than native BSA. The browning intensity, surface hydrophobicity, emulsifying property and foaming capacity of the conjugates were substantially improved while the free amino groups, intrinsic fluorescence emission and foaming stability were decreased compared to native and ultrasonicated BSA. The results of this study indicate that ultrasonication is an efficient technique to improve the physicochemical properties of proteins. The glycation between ultrasonicated BSA and monosaccharide can effectively improve the physicochemical properties of BSA, and the glycation rate order is galactose/gt glucose. Douchi is a traditional fermented soybean food in China. There are many types of Douchi in different regions of China. In this study, the effect of post-fermentation time and sodium chloride (NaCl) on Douchi fermented with Mucor wutungkiao were studied by measuring the color, texture and sensory attributes. The a* value and color differences (E) increased, while the hardness decreased during the post-fermentation. The results showed that post-fermentation time and NaCl content affected the color and texture significantly (p<0.05). The sensory attributes of Douchi with 12.5% NaCl content was more preferable than samples with 5%, 7.5% and 10% NaCl content in the post-fermentation, and 4 weeks of post-fermentation time was good for the formation of favorable sensory attributes in Douchi with 12.5% NaCl content. Ionic liquids (ILs) are desirable for use in a large number of applications because of their unique properties; however, compositions comprising only a single IL are expensive to synthesize and difficult to purify, and the widely used chloride-based ILs can be toxic and corrosive. Therefore, there is a need for new IL compositions that minimize common disadvantages encountered with single IL composition and synthetic methods which avoid halide intermediates. In this study, IL mixtures, which are chloride-free, were synthesized by a one-pot process, and the mixtures were used to dissolve biopolymers. This paper presents a new four-dimension autonomous system which shows extraordinary dynamical properties . Chaotic attractor and periodic attractor or hyper-chaotic attractor and quasi-periodic attractor, which are governed by different initial conditions instead of the system parameters, can coexist in the deterministic system. A sulfated polysaccharide H3-a1 [designated polysaccharide fraction, H3-a1, of Hydroclathrus clathratus (HCP)] was isolated and purified from an edible brown seaweed Hydroclathrus clathratus; HCP contained about 8.6% protein and a relatively high sulfur content (8.2%). HCP significantly affected the growth of human acute promyelocytic leukemia cells (HL-60), human breast carcinoma (MCF-7) and human hepatocellular carcinoma cancer cell lines. As revealed by flow cytometry, when HCP was co-incubated with HL-60 and MCF-7 cells for 72 h, it induced significant sub-G1 arrest (apoptosis) of these cells. HCP caused G1 phase arrest but did not induce terminal differentiation of these cancer cells. In vivo antitumor tests showed that HCP inhibited tumor growth at doses of 20 and 50 mg kg-1 in tumor-bearing BALB/c (supplied by the Laboratory Animal Services Centre of The Chinese University of Hong Kong) mice, but this effect was not due to an in vivo inhibitory effect on tumorigenesis. Nevertheless, HCP suppressed ascitic Sarcoma 180 tumor growth and prolonged the life span of the tumor-bearing mice by about 30–40%. HCP had no observed toxic side effects; it did not affect blood cells, or body, liver or spleen weights of mice 4, 7 and 10 days after treatment. This sulfated polysaccharide induced increased tumor necrosis factor-alpha (TNF-α) level in mouse serum. It also activated peritoneal macrophages to secrete TNF-α and nitric oxide. The article presents a study which described the metabolite spectrum of testosterone (T) in human fetal adrenal and liver in vitro and verified the roles of CYP3A and 2C isoforms in T metabolism. Researchers used the isocratic high performance liquid chromatography (HPLC) method of steroid separation and determination in order to demonstrate how human fetal adrenal microsomes can metabolize T. It is confirmed that isoforms did not play a significant function in metabolizing T in human fetal adrenal.

Abstract  A new ligand napaa (napaa = 7-acetamido-4-methyl-1,8-naphthyridin-2-carbaldehyde azine) and its two dinuclear copper(I) complexes, Cu2(napaa)(dppe)2(ClO4)2 (1) and Cu2(napaa)(PPh3)4(BF4)2 (2) (dppe = 1,2-bis(diphenylphosphino)ethane) and PPh3 = triphenylphosphine) were synthesized and characterized, and the structure of 1 was determined by X-ray crystal analysis. Each copper atom in 1 has a distorted tetrahedral geometry in which the metal center is associated to napaa and dppe ligands displaying chelating coordination modes and the naphthyridine rings of napaa are almost coplanar. The two complexes exhibit similar electronic absorption spectra with λmax at about 366 nm, which can be tentatively assigned to metal-to-ligand charge-transfer (MLCT) transition. Background: It has been shown that low concentrations of high-density lipoprotein cholesterol (HDL-C) are associated with non-alcoholic fatty liver disease (NAFLD) in patients with type 2 diabetes mellitus (T2DM). HDL2b, a major subfraction of high-density lipoprotein (HDL), is more significantly correlated with coronary heart disease (CHD) compared with total HDL. In this study, we analysed HDL2b in a cohort of Chinese T2DM subjects with or without NAFLD. Methods: A highly sensitive and reliable microfluidic chip method was adopted to measure HDL2b. In total, 48 T2DM patients with NAFLD diagnosed by a B-ultrasound were enrolled from our Beijing Community Pre-Diabetes (BCPD) study cohort. A total of 48 age and gender matched diabetic controls without NAFLD were selected from the same population. Results: Clinical characteristics and serum biochemical analyses demonstrated a significantly increased body mass index (BMI), waist circumference, homeostasis model assessment-insulin resistant index (HOMA-IR), total cholesterol (TC), and triglyceride (TG) concentrations in the NAFLD group. However, the concentrations of HDL2b and its ratio to total HDL in NAFLD patients was decreased, compared with controls (p<0.01). Significantly increased concentrations of high sensitive C-reactive protein (hs-CRP) (p<0.01) were also found. Multifactor logistic regression analysis showed that BMI and TG were the predominant risk factors for fatty liver, while HDL2b was a protective factor. Conclusions: T2DM patients with NAFLD have characteristics including obesity, marked insulin resistance, high TG, high hs-CRP, low HDL2b and a low HDL2b ratio to total HDL. We previously reported cloning of the Taenia solium annexin B1 gene from a metacestode cDNA expression library and demonstrated that it acts as a protective antigen for effective vaccine development against cysticercosis. In the present study we produced recombinant annexin B1 and antiserum against the protein to investigate its structural and functional properties. Western blotting of metacestode fractions indicated that T. solium annexin B1, similar to vertebrate annexins, associates with acid phospholipids in the presence of Ca2+. This property was confirmed by the recognition of apoptotic cells by labeled annexin B1. CD spectroscopy results demonstrated that α-helices are the main secondary structures of the protein. Ca2+ binding increases the α-helix content and causes significant thermal stabilization with a melting temperature increase of approximately 10°C. Functional Ca2+-dependent phospholipid binding sites of annexin B1 were investigated using mutant proteins. By changing a conserved acidic amino acid residue that putatively combines Ca2+ in each domain of annexin B1 singly or in combination, we found that Ca2+ binding in the first domain is more important than that at the other Ca2+ binding sites. Annexin B1 is a metacestode stage-specific antigen, with the protein being mainly localized in the teguments and surrounding cyst wall of T. solium metacestodes, suggesting a role in the parasite-host interaction. An invasive green alga, Caulerpa racemosa, collected from the South China Sea was tested for its antiviral properties. Hot water extracts and the n-butanol fraction obtained showed potent inhibition of herpes simplex virus type 1 (HSV-1) and Coxsackie virus B3 (Cox B3). A sulfoquinovosyldiacylglycerol (SQDG) compound was isolated and purified from the n-butanol fraction following further antiviral-guided fractionation. The SQDG compound exhibited an excellent antiviral effect against HSV-2, with a 50% inhibitory concentration (IC50) of 15.6 μg ml-1 against both standard and clinical strains of HSV-2, but showed only moderate antiviral effects against HSV-1 and Cox B3. The SQDG compound was chemically characterized using spectroscopic methods as (2 S)-1,2-di- O-palmitoyl-3- O-(6′-sulfo-α-D-quinovopyranosyl) glycerol. Molecular dynamics simulations have been used to examine the friction between commensurate selfassembled monolayers (SAMs) on Au (111). The kinetic friction is found to rise logarithmically with velocity. The analysis of friction force and discontinuous molecular movements reveals complex dynamics of the closedpacking chains under shearing. Molecules in the lower monolayer can be divided into two groups with a phase difference of p .The periodic motion of the molecules is not synchronous with the ftictional stick-slip loops, which result in second-order valley in friction curves. Protein-Chip as micro-assays for the determination of protein interaction, the analysis, the identification and the purification of proteins has large potential applications. The Optical Protein-Chip is able to detect the multi-interaction of proteins and multi-bio-activities of molecules directly and simultaneously with no labeling. The chip is a small matrix on solid substrate containing multi-micro-area prepared by microfabrication with photolithography or soft lithography for surface patterning, and processed with surface modification which includes the physical, chemical, and bio-chemical modifications, etc. The ligand immobilization, such as protein immobilization, especially the oriented immobilization with low steric hindrance and high bio-specific binding activity between ligand and receptor is used to form a sensing surface. Each area of the pattern is corresponding to only one bioactivity. The interval between the areas is non-bioactive and optically extinctive. The affinity between proteins is used to realize non-labeling microassays for the determination of protein identification and protein interaction. There exist many difficulties to fabricate a machine in nanometer-scale. One of the key technologies for nanofabrication is how to get a nanocomponent with definite sizes. In this paper, we explore a new method to fabricate nanorollers whose size can be controlled by cutting long carbon nanotube into pieces at appointed positions using the tip of atomic force microscope (AFM). Sample was prepared so that parts of carbon nanotubes are pressed by gold coating to keep them fixed while cutting. The relaxation process of a nanoroller while manipulating was observed. We prepared intercalated organic montmorillonite (OMMT) from a pristine MMT and long-alkyl-chain quaternary ammonium salts (LACQAS). X-ray diffraction, Fourier transform infrared spectroscopy, scanning electron microscopy, and thermogravimetric analyses revealed that the amount, carbon atoms, and alkyl chains of LACQAS influenced the large value of d(001) of OMMT significantly. The d(001) stabilized at 100–150 mmol·(100 g)-1 of LACQAS/MMT. The single LACQAS cations arranged in the form of a lateral layer or half-paraffin-type molecular structure between MMT layers, whereas the dual and triple LACQAS cations arranged in the form of a paraffin-type molecular structure. The pyrolysis temperature and maximum pyrolysis rate of OMMT increased greatly compared with those of MMT. The thermal weight loss of MMT was caused by the removed absorbed and structural water, whereas that of OMMT was by the pyrolysis of LACQAS. With the help of the Agrawal integral equation, the pyrolysis kinetics of MMT and OMMT were obtained using a trial-and-error method.
