Objective : Electroacupuncture (EA) in the treating principle of "soothing the liver and regulating the kidney" was applied to intervion the rats with post-traumatic stress disorder (PTSD), and its effect on anxiety-like behavior, spatial learning and memory ability, and expressions of synaptophysin (SYN) and postsynaptic dense 95 (PSD95) in hippocampus of rats were observed to explore the underlying mechanism. Methods : Twenty-four male SD rats were randomly divided into control group, model group, and EA group, with 8 rats in each group. There was no intervention in the control group. The rest 16 rats were prepared for modeling. The single-prolonged stress & shock (SPS&S) method was used to establish the PTSD models. There was no intervention in the model group after modeling. In the EA group, "Bǎihuì (百会GV20)"" Shéntíng (神庭GV24)""Gānshū (肝俞BL18)""Shènshū (肾俞BL23)" were manipulated with EA stimulation, intensity 1 mA, frequency 2/100 Hz, disperse-dense wave, and treatment was performed once a day, 20 min each time, for a total of 21 days. Open field test, elevated plus maze and Morris water maze tests were used to observe the behavioral differences of rats in each group. Immunohistochemical method was used to observe the differences of positive expressions of proteins SYN and PSD95 in hippocampus. Results : In the open field test, compared with the control group, the total traveling distance, the percentage of the time spent in the central cell and the numbers of the central cells crossing in the model group were all decreased (all P < 0.05). Compared with the model group, these indicators in the EA group were significantly all increased (all P < 0.05). In the elevated plus maze test, compared with the control group, the percentages of open arm staying time and entering times in the model group were both decreased (both P < 0.05). Compared with the model group, these indicators in the EA group were both significantly increased (both P < 0.05). The results of Morris water maze test showed that compared with the control group, the escape latency time and travelled distance of rats in the model group were all increased from day1 to day 4 (all P < 0.05), and the percentage of staying time in the target quadrant was decreased (P < 0.05). Compared with the model group, the escape latency time and travelled distance of EA group were both significantly shortened (both P < 0.05), and the percentage of staying time in the target quadrant was significantly increased (P < 0.05). The immunohistochemical results showed that, compared with the control group, the positive expressions of SYN and PSD95 in the hippocampus of the model group were significantly down-regulated (both P < 0.05). Compared with the model group, the positive expressions of SYN and PSD95 in the hippocampus of the EA group were significantly up-regulated (both P < 0.05). Conclusions : EA in the treating principle of "soothing the liver and regulating the kidney" can effectively relieve anxiety-like behavior and improve spatial learning and memory ability of rats with PTSD, and the mechanism is related to the up-regulation of the expressions of SYN and PSD95 in the hippocampus. Mucosal immunity is posed to constantly interact with commensal microbes and invading pathogens. As a fundamental cell biological pathway affecting immune response, autophagy regulates the interaction between mucosal immunity and microbes through multiple mechanisms, including direct elimination of microbes, control of inflammation, antigen presentation and lymphocyte homeostasis, and secretion of immune mediators. Some of these physiologically important functions do not involve canonical degradative autophagy but rely on certain autophagy genes and their 'autophagy gene-specific functions.' Here, we review the relationship between autophagy and important mucosal pathogens, including influenza virus, Mycobacterium tuberculosis , Salmonella enterica , Citrobacter rodentium , norovirus, and herpes simplex virus, with a particular focus on distinguishing the canonical versus gene-specific mechanisms of autophagy genes. Clearance of apoptotic cells, or "efferocytosis," is essential for diverse processes including embryonic development, tissue turnover, organ regeneration, and immune cell development. The human body is estimated to remove approximately 1% of its body mass via apoptotic cell clearance daily. This poses several intriguing cell metabolism problems. For instance, phagocytes such as macrophages must induce or suppress metabolic pathways to find, engulf, and digest apoptotic cells. Then, phagocytes must manage the potentially burdensome biomass of the engulfed apoptotic cell. Finally, phagocytes reside in complex tissue architectures that vary in nutrient availability, the types of dying cells or debris that require clearance, and the neighboring cells they interact with. Here, we review advances in our understanding of these three key areas of phagocyte metabolism. We end by proposing a model of efferocytosis that integrates recent findings and establishes a new paradigm for testing how efferocytosis prevents chronic inflammatory disease and autoimmunity. Efferocytosis is a complex, multi-step process that exerts a heavy metabolic burden on phagocytic cells. The structure of compound 1 was established based on extensive spectroscopic analysis. Compound 1 was the first example of 14-nordrimane-type sesquiterpene up to now. Apoptotic cell (AC) clearance (efferocytosis) is performed by phagocytes, such as macrophages, that inhabit harsh physiological environments. Here, we find that macrophages display enhanced efferocytosis under prolonged (chronic) physiological hypoxia, characterized by increased internalization and accelerated degradation of ACs. Transcriptional and translational analyses revealed that chronic physiological hypoxia induces two distinct but complimentary states. The first, "primed" state, consists of concomitant transcription and translation of metabolic programs in AC-naive macrophages that persist during efferocytosis. The second, "poised" state, consists of transcription, but not translation, of phagocyte function programs in AC-naive macrophages that are translated during efferocytosis. Mechanistically, macrophages efficiently flux glucose into a noncanonical pentose phosphate pathway (PPP) loop to enhance NADPH production. PPP-derived NADPH directly supports enhanced efferocytosis under physiological hypoxia by ensuring phagolysosomal maturation and redox homeostasis. Thus, macrophages residing under physiological hypoxia adopt states that support cell fitness and ensure performance of essential homeostatic functions rapidly and safely. Depression is one of the most debilitating and severe psychiatric disorders and a serious public health concern. Currently, many treatments are indicated for depression, including traditional Chinese medicinal formulae such as Xiao-Yao-San (XYS), which has effective antidepressant effects in clinical and animal studies. To summarize current evidence of XYS in terms of the preclinical and clinical studies and to identify the multi-level, multi-approach, and multi-target potential antidepressant mechanisms of XYS and active components of XYS by a comprehensive search of the related electronic databases. The following electronic databases were searched from the beginning to April 2022: PubMed, MEDLINE, Web of Science, Google Scholar, and China National Knowledge Infrastructure. This review summarizes the antidepressant mechanisms of XYS and its active ingredients, which are reportedly correlated with monoamine neurotransmitter regulation, synaptic plasticity, and hypothalamic–pituitary–adrenal axis, etc. XYS plays a critical role in the treatment of depression by the regulation of several factors, including the monoaminergic systems, hypothalamic–pituitary–adrenal axis, synaptic plasticity, inflammation, brain-derived neurotrophic factor levels, brain-gut axis, and other pathways. However, more clinical and animal studies should be conducted to further investigate the antidepressant function of XYS and provide more evidence and recommendations for its clinical application. Background: Parkinson's disease (PD) is the second most common neurodegenerative disease featured to mitochondrial dysfunction in neuronal cells. Dynamin-related protein 1 (Drp1) is an important regulator of mitochondrial fission and subsequent mitophagy. Mangiferin (MGF) is a glucosyl xanthone mainly derived from Mangifera indica L., possessing multifaceted properties, e.g., antioxidant, anti-inflammatory, and enhancement of cognitive ability. Besides, it can cross the blood-brain barrier, thereby exerting a neuroprotective effect. However, so far, MGF's effect in balancing mitochondrial homeostasis via regulation of Drp1 level and mitophagic pathway in PD remains rarely reported.Purpose: We aimed to investigate the neuroprotective effect of MGF against 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) mouse model of PD and examine the possible mechanisms.Methods: We utilized C57BL/6 mice exposed to 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP); Behavioral parameters, containing the open field test, balance beam, pole test, and rotarod test, assessed the locomotor activity; immunohistochemistry assessed the number of TH-positive neurons; transmission electron microscopy detected ultrastructural mitochondrial morphology in the dopaminergic neuron; complex I enzymatic activity microplate assay kit measured the mitochondrial complex I activity; ATP determination kit measured ATP levels in mitochondria isolated from cells or striatal tissues; western blot measured the levels of Drp1 and mitophagic proteins.Results: We observed that MGF could mitigate motor deficiency and improve the expression of tyrosine hydroxylase in the substantia nigra of MPTP-induced PD mice. Furthermore, MGF not only ameliorated mitochondrial ultrastructure, but also improved mitochondrial ATP content. Within mitochondria, MGF could reduce Drp1 expression and reverse the expressions of mitophagic proteins, including PINK1, Parkin, NIX, BNIP3, FUNDC1, and p62.Conclusion: Present study indicates that MGF benefits mitochondrial networks by recovering mitochondrial ultrastructure and ATP contents, reducing mitochondrial Drp1, and modulating mitophagic proteins in the MPTP-induced PD mice model, which revealed a novel acting mechanism of MGF in PD's treatment. Compost-based organic fertilizers made from animal manures may contain high levels of antibiotic resistance genes (ARGs). However, the factors affecting the abundance and profile of ARGs in organic fertilizers remain unclear. We conducted a national-wide survey in China to investigate the effect of material type and composting process on ARG abundance in commercial organic fertilizers and quantified the contributions of bacterial composition and mobile genetic elements (MGEs) to the structuring of ARGs, using quantitative PCR and Illumina sequencing of 16S rRNA gene amplicons. The tetracycline, sulfonamide, aminoglycoside and macrolide resistance genes were present at high levels in all organic fertilizers. Seven ARGs that confer resistance to clinically important antibiotics, including three β-lactam resistance genes, three quinolone resistance genes and the colistin resistance gene mcr-1 , were detected in 8 – 50% the compost samples, whereas the vancomycin resistance gene vanC was not detected. Raw material type had a significant (p < 0.001) effect on the ARG abundance, with composts made from animal feces except some cattle feces generally having higher loads of ARGs than those from non-animal raw materials. Composting process type showed no significant (p > 0.05) effect on ARG abundance in the organic fertilizers. MGEs exerted a greater influence on ARG composition than bacterial community, suggesting a strong mobility of ARGs in the organic fertilizers. Abstract: Capsules protect bacteria against phagocytic clearance. Capsular polysaccharides or polyglutamates have evolved also to resist antigen presentation by immune cells, thereby interfering with the production of opsonophagocytic antibodies. Linking capsular material to a carrier protein stimulates its presentation to the immune system. For many conjugate vaccines this is achieved by a process of random chemical cross-linking. Here we describe a new technology, designated sortase-conjugation, which generates a single amide bond between the C-terminal end of a carrier protein and the capsular material. Sortase-conjugation was used to link the poly-d-γ-glutamic acid (PDGA) capsule of Bacillus anthracis to the receptor binding domain (D4) of protective antigen (PagA). When used as a vaccine, PDGA–D4 conjugate elicited robust antibody responses against both capsule and D4. Background: Depression, as a prevalent and debilitating psychiatric disease, severely decreases the life quality of individuals and brings heavy burdens to the whole society. Currently, some antidepressants are applied in the treatment of severe depressive symptoms, while there are still some undesirable drawbacks. Paeoniflorin is a monoterpenoid glycoside that was firstly extracted from Paeonia lactiflora Pall, a traditional Chinese herb that is widely used in the Chinese herbal formulas for treating depression.Purpose: This review summarized the previous pre-clinical studies of paeoniflorin in treating depression and further discussed the potential anti-depressive mechanisms for that paeoniflorin to be further explored and utilized in the treatment of depression clinically.Methods: Some electronic databases, e.g., PubMed and China National Knowledge Infrastructure, were searched from inception until April 2021.Results: This review summarized the effective anti-depressive properties of paeoniflorin, which is related to its functions in the upregulation of the levels of monoaminergic neurotransmitters, inhibition of the hypothalamic-pituitary-adrenal axis hyperfunction, promotion of neuroprotection, promotion of hippocampus neurogenesis, and upregulation of brain-derived neurotrophic factor level, inhibition of inflammatory reaction, downregulation of nitric oxide level, etc. CONCLUSION: This review focused on the pre-clinical studies of paeoniflorin in depression and summarized the recent development of the anti-depressive mechanisms of paeoniflorin, which approves the role of paeoniflorin plays in anti-depression. Based on the high-throughput first-principles calculations with structural recognition, we have ystematically investigated the structural stabilities and optical properties of Si x Ge y H z nanocrystals(H-SiGeNCs), including various sizes, shapes and compositions. The total energies of H-SiGeNCs can be simply estimated by the bond energy model in high accuracy, where the error of test set is less than 0.5 meV per atom. According to the energy difference of Si/Ge in various bonding environments, we have determined the ground state structures by the geometry analysis, as is confirmed the convex hulls of formation enthalpy from the first-principles calculations. In addition, the energy gaps of H-SiGeNCs are modulated by the atomic distributions, as well as the vibrations of Si-H and Ge-H bonds at room temperature which is revealed by ab initio molecular dynamics simulations. Yeast is widely studied in producing biofuels and biochemicals using renewable biomass. Among various yeasts, Saccharomyces cerevisiae has been particularly recognized as an important yeast cell factory. However, economic bioproduction using S. cerevisiae is challenged by harsh environments during fermentation, among which inhibitory chemicals in the culture media or toxic products are common experiences. Understanding the stress-responsive mechanisms is conducive to developing robust yeast strains. Here, we review recent progress in mechanisms underlying yeast stress response, including regulation of cell wall integrity, membrane transport, antioxidative system, and gene transcription. We highlight epigenetic regulation of stress response and summarize manipulation of yeast stress tolerance for improved bioproduction. Prospects in the application of machine learning to improve production efficiency are also discussed. This study adopted the transformation approach to modify the structure of curriculum and instruction of a Taiwanese teacher-education program to weave in elements of culturally responsive teaching (CRT). Through observation, interviews, and data collection, the transformation of 15 candidates was analyzed. Their intrinsic motivation progressed from “establishing inclusion” to “developing a positive attitude,” “enhancing meaning,” and finally “engendering competence” through reflection. The findings can serve as the basis for deepening the theories and practices of culturally responsive teacher preparation, particularly in the context of Chinese cultural heritage. Depressive symptom is the prevailing non-motor symptom of Parkinson's disease (PD). Drug treatments for depressed PD (dPD) can mitigate the symptoms of patients. However, the results are discordant and need further analysis. This systematic review with network meta-analysis aims to evaluate the drug treatments for dPD. We included double-blind, randomized controlled trials to compare antidepressants with placebo or other antidepressants in dPD. We performed traditional pairwise analysis and network meta-analysis concerning the efficacy, acceptability, depression score, and adverse effect. The surface under the cumulative ranking curve was to assess the ranking probabilities of the enrolled agents. We enrolled 62 studies, including 12,353 subjects, to analyze these estimates. For the traditional pairwise meta-analysis, dopamine agonist (DOP; OR = 2.20 [95% CI, 1.46 to 3.33]) and selective serotonin reuptake inhibitor (SSRI; OR = 2.30 [95% CI, 1.15 to 4.60]) were observed to improve the efficacy compared with placebo. For network meta-analysis, DOP was observed to improve the efficacy compared with placebo (OR = −0.84 [95% CI, −1.20 to −0.48]). Both direct and indirect evidence showed that several treatments, e.g., DOP, monoamine-oxidase inhibitor, serotonin-norepinephrine reuptake inhibitors, SSRI, and tricyclic antidepressants, significantly improved depressive symptoms. Major depressive disorder (MDD) is a form of glial cell-based synaptic dysfunction disease in which glial cells interact closely with neuronal synapses and perform synaptic information processing. Glial cells, particularly astrocytes, are active components of the brain and are responsible for synaptic activity through the release gliotransmitters. A reduced density of astrocytes and astrocyte dysfunction have both been identified the brains of patients with MDD. Furthermore, gliotransmission, i.e., active information transfer mediated by gliotransmitters between astrocytes and neurons, is thought to be involved in the pathogenesis of MDD. However, the mechanism by which astrocyte-mediated gliotransmission contributes to depression remains unknown. This review therefore summarizes the alterations in astrocytes in MDD, including astrocyte marker, connexin 43 (Cx43) expression, Cx43 gap junctions, and Cx43 hemichannels, and describes the regulatory mechanisms of astrocytes involved in synaptic plasticity. Additionally, we investigate the mechanisms acting of the glutamatergic, gamma-aminobutyric acidergic, and purinergic systems that modulate synaptic function and the antidepressant mechanisms of the related receptor antagonists. Further, we summarize the roles of glutamate, gamma-aminobutyric acid, d -serine, and adenosine triphosphate in depression, providing a basis for the identification of diagnostic and therapeutic targets for MDD. This study examines the protective effects of gamma irradiation against postharvest insect damage and microbial contamination and its effect on the nutritive value of adlay (Coix lacryma-jobi L.). Adlay was treated with doses of 0–20kGy gamma irradiation and subsequently stored at ambient temperature. The number of insects, microbial quality and chemical properties of irradiated and non-irradiated adlay were evaluated immediately after treatment and after 6 months of storage. Before irradiation, total aerobic microbial counts ranged from 5.6×102 to 1.4×105 CFUg−1 and the mean total number of insects was 3.2±2.1 per 100g of polished kernel. Two kilograys was the lowest dose that provided 100% insect control. A radiation-resistant bacterium, Deinococcus radiodurans RC1, was found in 2 of 10 adlay samples. Four kilograys was a sufficient dose for Enterobacteriaceae inactivation, and 6kGy was a sufficient dose for yeast and fungi inactivation. Twenty and 8kGy sufficed for the inactivation of all mesophilic microbes in samples with and without D. radiodurans, separately. Moreover the moisture, ash, crude fiber, crude fat, crude protein and riboflavin content all remained constant. However, 8kGy irradiation reduced vitamin B1 concentration by 24.2%, but did not measurably reduce the amount of amino acids, except methionine and cysteine. Fatty acid contents did not alter after 8kGy irradiation, but changes were observed after the 6 months of storage. These changes caused by irradiation were no greater than those caused by the 6-month storage. Irradiation up to 8kGy did not markedly increase the acid value, but did increase the peroxide value to 13% of the initial value right after irradiation. However, significant changes in acid value and peroxide value were noted after 6 months of storage both in non-irradiated and post-irradiated storage samples. Additionally, 8kGy irradiation did not significantly change the adlay appearance. The improvement in the hygiene of this vital food source compensates for the small loss of some nutritional constituents. Hence 8kGy of gamma irradiation can be used in cold decontamination of adlay to prolong shelf-life, to improve postharvest quality, and to reduce the risk of food-borne disease. The chemical resistance of eight organic solvents in high density polyethylene (HDPE) geomembrane has been investigated using the ASTM F739 permeation method and the immersion test at different temperatures. The diffusion of the experimental organic solvents in HDPE geomembrane was non-Fickian kinetic, and the solubility coefficients can be consistent with the solubility parameter theory. The diffusion coefficients and solubility coefficients determined by the ASTM F739 method were significantly correlated to the immersion tests (p <0. 001). The steady state permeation rates also showed a good agreement between ASTM F739 and immersion experiments (r 2 =0. 973, p <0. 001). Using a one-dimensional diffusion equation based on Fick''s second law, the diffusion and solubility coefficients obtained by immersion test resulted in over estimates of the ASTM F739 permeation results. The modeling results indicated that the diffusion and solubility coefficients should be obtained using ASTM F739 method which closely simulates the practical application of HDPE as barriers in the field. The ubiquitous nature of microbes has made them the pioneers in radionuclides adsorption and transport. In this study, the radiation resistance and nuclide biosorption capacity of microbes isolated from the Lanyu low-level radioactive waste (LLRW) repository in Taiwan was assessed, the evaluation of the possibility of using the isolated strain as biosorbents for 60 Co and Co (II) from contaminated aqueous solution and the potential impact on radionuclides release. The microbial content of solidified waste and broken fragments of containers at the Lanyu LLRW repository reached 10 5 CFU/g. Two yeast strains, Candida guilliermondii (CT1) and Rhodotorula calyptogenae (RT1) were isolated. The radiation dose necessary to reduce the microbial count by one log cycle of CT1 and RT1 was 2.1 and 0.8 kGy, respectively. Both CT1 and RT1 can grow under a radiation field with dose rate of 6.8 Gy/h, about 100 times higher than that on the surface of the LLRW container in Lanyu repository. CT1 and RT1 had the maximum 60 Co biosorption efficiency of 99.7 ± 0.1% and 98.3 ± 0.2%, respectively in 60 Co aqueous solution (700 Bq/mL), and the 60 Co could stably retained for more than 30 days in CT 1. Nearly all of the Co was absorbed and reached equilibrium within 1 h by CT1 and RT1 in the 10 μg/g Co (II) aqueous solution. Biosorption efficiency test showed almost all of the Co (II) was adsorbed by CT1 in 20 μg/g Co (II) aqueous solution, the efficiency of biosorption by RT1 in 10 μg/g of Co (II) was lower. The maximum Co (II) sorption capacity of CT1 and RT1 was 5324.0 ± 349.0 μg/g (dry wt) and 3737.6 ± 86.5 μg/g (dry wt), respectively, in the 20 μg/g Co (II) aqueous solution. Experimental results show that microbial activity was high in the Lanyu LLRW repository in Taiwan.
