Objectives. Osteochondral defects (OCDs) are localized areas of damaged cartilage and underlying subchondral bone that can produce pain and seriously impair joint function. Literature reports indicated that icariin (ICA) has the effect of promoting cartilage repair. However, its mechanism remains unclear. Here, we explored the effects of icariin and extracellular vesicles (EVs) from rabbit synovial-derived mesenchymal stem cells (rSMSCs) on repairing of OCDs. Materials and Methods. Rabbit primary genicular chondrocytes (rPGCs), knee skeletal muscle cells (rSMCKs), and rSMSCs, and extracellular vesicles derived from the latter two cells (rSMCK-EVs and rSMSC-EVs) were isolated and identified. The rPGCs were stimulated with ICA, rSMSC-EVs either separately or in combination. The rSMCK-EVs were used as a control. After stimulation, chondrogenic-related markers were analyzed by quantitative RT-PCR and western blotting. Cell proliferation was determined by the CCK-8 assay. The preventative effects of ICA and SMSC-EVs in vivo were determined by H&E and toluidine blue staining. Immunohistochemical analyses were performed to evaluate the levels of COL2A1 and β-catenin in vivo. Results. In vitro, the proliferation of rPGCs was markedly increased by ICA treatment in a dose-dependent manner. When compared with ICA or rSMSC-EVs treatment alone, combined treatment with ICA and SMSC-EVs produced stronger stimulative effects on cell proliferation. Moreover, combined treatment with ICA and rSMSC-EVs promoted the expression of chondrogenic-related gene, including COL2A1, SOX-9, and RUNX2, which may be via the activation of the Wnt/β-catenin pathway. In vivo, combined treatment with rSMSC-EVs and ICA promoted cartilage repair in joint bone defects. Results also showed that ICA or rSMSC-EVs both promoted the COL2A1 and β-catenin protein accumulation in articular cartilage, and that was further enhanced by combined treatment with rSMSC-EVs and ICA. Conclusion. Our findings highlight the promising potential of using combined treatment with ICA and rSMSC-EVs for promoting osteochondral repair.

With the reinforcement project of a heavy-duty concrete-encased concrete-filled steel tube (CFST) column as the background, the mechanism of column reinforcement with supporting beams is proposed. The key mechanisms include the following: (1) designing the upper and lower corbel-shaped temporary support columns, (2) crack repairing the inner steel pipe using patch-shaped steel plates, and (3) replacing microwound concrete inside the steel pipe, which are all innovative and effective methods. Moreover, a system is developed for monitoring the safety during the reinforcement process and recovering the bearing capacity of the concrete-encased CFST column. The monitoring results indicate that the load conversion process of the temporary support columns lasts for 50 days, that the sum of the axial forces of the four supporting columns is 7,966.0 kN, accounting for 65% of the standard load, and that during the whole process, the structural components are safe and reliable. Finally, the construction simulation is in agreement with the field test results, which proves that the construction simulation developed based on the theory of slow time-dependent mechanics and the method of static nonlinear analysis is accurate.

The fatigue stress amplitude of the welded cross plate-hollow sphere joint (WCPHSJ) in a grid structure varies due to the random loading produced by suspending cranes. A total of 14 specimens considering three different types of WCPHSJs were prepared and tested using a specially designed test rig. Four typical loading conditions, “low-high,” “high-low,” “low-high-low,” and “high-low-high,” were first considered in the tests to investigate the fatigue behavior under variable load amplitudes, followed by metallographic analyses. The experimental and metallographic analysis results provide a fundamental understanding on the fatigue fracture form and fatigue mechanism of WCPHSJs. Based on the available data from constant-amplitude fatigue tests, the variable-amplitude fatigue life of the three types of WCPHSJs was estimated using the Miner rule and Corten-Dolan theory. Since both accumulative damage theories yield virtually same damaging results, the Miner rule is hence suggested to estimate the fatigue life of WCPHSJs.

Bone marrow-derived mesenchymal stem cells (BMSCs) are widely used in tissue engineering for regenerative medicine due to their multipotent differentiation potential. However, their poor migration ability limits repair effects. Icariin (ICA), a major component of the Chinese medical herb Herba Epimedii, has been reported to accelerate the proliferation, osteogenic, and chondrogenic differentiation of BMSCs. However, it remains unknown whether ICA can enhance BMSC migration, and the possible underlying mechanisms need to be elucidated. In this study, we found that ICA significantly increased the migration capacity of BMSCs, with an optimal concentration of 1 μmol/L. Moreover, we found that ICA stimulated actin stress fiber formation in BMSCs. Our work revealed that activation of the MAPK signaling pathway was required for ICA-induced migration and actin stress fiber formation. In vivo, ICA promoted the recruitment of BMSCs to the cartilage defect region. Taken together, these results show that ICA promotes BMSC migration in vivo and in vitro by inducing actin stress fiber formation via the MAPK signaling pathway. Thus, combined administration of ICA with BMSCs has great potential in cartilage defect therapy.

Tuberculosis (TB) remains one of the deadliest infectious diseases in the world. Previous genome-wide association studies suggested that single-nucleotide polymorphisms (SNPs) in some genes could indicate the susceptibility to TB in some populations. Herein, we studied the association of SNPs in the immunity-related genes, i.e., ASAP1 and SP110 genes with the susceptibility to TB in a Mongolian population in China. A case-control study was performed with 197 TB patients and 217 healthy controls. Six SNPs in ASAP1 and six SNPs in SP110 were selected for genotyping test by second-generation sequencing technique. A SNP in SP110 gene (rs722555) was identified to be associated with susceptibility to TB in the Mongolian population (p < 0.05). The T allele of rs722555 in SP110 gene was associated with a 36% increase of risk at TB (OR 1.36, 95% CI 1.03-1.81), and the CT+TT genotype of rs722555 was associated with a 74% increase of risk at TB (OR 1.74, 95% CI 1.16-2.60) in the dominant genetic model. None of SNPs in ASAP1 gene tested in this study were significantly associated with TB susceptibility, while some individuals with SNPs (rs10956514, rs4733781, rs2033059, rs12680942, rs1017281, rs1469288, and rs17285138) in the ASAP1 gene tended to have a reduced risk at TB. In conclusion, this study suggested that the rs722555 SNP in SP110 gene might be a risk factor for TB in a Mongolian population. [ABSTRACT FROM AUTHOR]

There has been a heated argument over self-incompatibilityof chrysanthemum (Chrysanthemum morifolium) among chrysanthemum breeders. In order to solve the argument, we investigated pistil receptivity, seed set, and compatible index of 24 chrysanthemum cultivars. It was found that the 24 cultivars averagely had 3.7–36.3 pollen grains germinating on stigmas at 24 hours after self-pollination through the fluorescence microscope using aniline blue staining method. However, only 10 of them produced self-pollinated seeds, and their seed sets and compatible indexes were 0.03–56.50% and 0.04–87.50, respectively. The cultivar “Q10-33-1” had the highest seed set (56.50%) and compatible index (87.50), but ten of its progeny had a wide range of separation in seed set (0–37.23%) and compatible index (0–68.65). The results indicated that most of chrysanthemum cultivars were self-incompatible, while a small proportion of cultivars were self-compatible. In addition, there is a comprehensive separation of self-incompatibility among progeny from the same self-pollinated self-compatible chrysanthemum cultivar. Therefore, it is better to emasculate inflorescences during chrysanthemum hybridization breeding when no information concerning its self-incompatibility characteristics is available. However, if it is self-incompatible and propagated by vegetative methods, it is unnecessary to carry out emasculation when it is used as a female plant during hybridization breeding.

The position and orientation system (POS) is a key equipment for airborne remote sensing systems, which provides high-precision position, velocity, and attitude information for various imaging payloads. Temperature error is the main source that affects the precision of POS. Traditional temperature error model is single temperature parameter linear function, which is not sufficient for the higher accuracy requirement of POS. The traditional compensation method based on neural network faces great problem in the repeatability error under different temperature conditions. In order to improve the precision and generalization ability of the temperature error compensation for POS, a nonlinear multiparameters temperature error modeling and compensation method based on Bayesian regularization neural network was proposed. The temperature error of POS was analyzed and a nonlinear multiparameters model was established. Bayesian regularization method was used as the evaluation criterion, which further optimized the coefficients of the temperature error. The experimental results show that the proposed method can improve temperature environmental adaptability and precision. The developed POS had been successfully applied in airborne TSMFTIS remote sensing system for the first time, which improved the accuracy of the reconstructed spectrum by 47.99%.

Hepatic ischemia/reperfusion injury (HIRI) is a common complication of liver surgery requiring hepatic disconnection, such as hepatectomy and liver transplantation. The aim of this study was to investigate the effects of cordycepin on HIRI and to elucidate the underlying mechanisms. Balb/c mice were randomly divided into six groups: a normal control group, sham group, H-cordycepin group, HIRI group, L-cordycepin (25 mg/kg) + HIRI group, and H-cordycepin (50 mg/kg) + HIRI group. Mice were subjected to I/R, and cordycepin was intragastrically administered for seven consecutive days before surgery. Orbital blood and liver specimens were collected at 6 and 24 h after HIRI. Serum levels of ALT and AST were decreased in the cordycepin pretreatment groups. Notably, cordycepin attenuated the inflammatory response and the production of proapoptosis proteins, while increasing expression of antiapoptosis proteins and decreasing expression of autophagy-linked proteins. Furthermore, cordycepin inhibited activation of the MAPK/NF-κB signaling pathway. Collectively, these results indicate that cordycepin pretreatment ameliorated hepatocyte injury caused by HIRI. As compared with the HIRI group, cordycepin pretreatment mitigated the inflammatory response and inhibited apoptosis and autophagy via regulation of the MAPK/NF-κB signaling pathway. Aims: The aim of this study is to investigate the situation and perceptions of nursing directors about emergency nursing staff deployment in designated hospitals during the pandemic of COVID‐19 in mainland China. Background: The pandemic of COVID‐19 has significantly depleted health care resources, leading to increased burden of nursing care and staffing and exacerbating the crisis in health care facilities. Currently, how to effectively plan and schedule nursing staffing in the pandemic still remains unknown. Methods: From 14 July 2020 to 8 September 2020, 62 nursing directors of designated hospitals in mainland China were invited to participate in a cross‐sectional online survey for their perceptions of nursing human‐resource allocation during the pandemic of COVID‐19. Results: A total of 55 valid questionnaires were collected, showing that 96.36% of the hospitals had emergency nursing organizations and management systems during the pandemic, 96.36% had well‐established scheduling principles for nursing human resources and 54.55% of hospitals had human‐resource scheduling platforms. All the hospitals had trained emergency nursing staff in infection control (55, 100%), work process (51, 92.73%) and emergency skills (50, 90.91%). Most of the participants were satisfied with the nursing staffing deployments at their institutions (52, 94.55%). However, more than two thirds of them believed that their human‐resource deployment plans need further improvements (39, 70.91%). Conclusions: Most of the designated hospitals investigated had established emergency nursing organizations, and management systems, and related regulations for the epidemic. However, the contents mentioned above still need to be further standardized. Implications for nursing management: The surge of patients in the epidemic was considerable challenge for the emergency capacity of hospitals. In the future, we should pay more attention to the following aspects: building emergency nursing staffing platforms, increasing emergency human‐resource reserves, establishing reliable communication channels for emergency response teams, improving the rules and regulations of emergency human‐resource management, offering more training and drills for emergency‐related knowledge and skills and giving more focus on bio‐psycho‐social wellbeing of nurses. The effects of CO2 enrichment on growth and development of Impatiens hawkeri, an important greenhouse flower, were investigated for the purpose of providing scientific basis for CO2 enrichment to this species in greenhouse. The plants were grown in CO2-controlled growth chambers with 380 (the control) and 760 (CO2 enrichment) μmol⋅mol-1, respectively. The changes in morphology, physiology, biochemistry, and leaf ultrastructure of Impatiens were examined. Results showed that CO2 enrichment increased flower number and relative leaf area compared with the control. In addition, CO2 enrichment significantly enhanced photosynthetic rate, contents of soluble sugars and starch, activities of peroxidase (POD), superoxide dismutase (SOD), and ascorbate peroxidase (APX), but reduced chlorophyll content and malondialdehyde (MDA) content. Furthermore, significant changes in chloroplast ultrastructure were observed at CO2 enrichment: an increased number of starch grains with an expanded size, and an increased ratio of stroma thylakoid to grana thylakoid. These results suggest that CO2 enrichment had positive effects on Impatiens, that is, it can improve the visual value, promote growth and development, and enhance antioxidant capacity.

Background. Many studies have reported an association between tea drinking and Parkinson's disease (PD). Our purpose is to summarize the available information and evaluate the risk of PD associated with tea drinking. Methods. We searched all publications in English language on the association of tea drinking and PD risk published up to December 2010. The pooled analysis was performed with Review Manager 5.0. Results. In total, eight articles including 1418 cases and 4250 controls were included in the meta-analysis. The pooled odds ratio (95% CI) was 0.85 (0.74–0.98), which suggests the protective effect of tea drinking in PD risks. Moreover, the summary OR (OR: 0.83, 95% CI = 0.69–0.99) for drinkers of ≤1 cup of tea per day versus nonconsumers and that (OR: 0.96, 95% CI = 0.73–1.27) for drinkers of >1 cups of tea per day versus nonconsumers showed that there was not an apparent dose-response relationship. No indication for publication bias was found. Conclusions. This meta-analysis showed that tea drinking can lower the risk of PD, while no apparent dose-response relationship was found. Further effort is needed to fully understand the mechanism underlying the beneficial effect of tea consumption in lowering PD risk.

This study optimized the storage conditions for fermented pork jerky via microbial fermentation. Lactobacillus bulogaricus was employed as starter culture, fermentation conditions, and baking conditions were optimized in study. Microbial indices reflecting parameters of preserved fermented pork jerky involved the detection of total plate count, Escherichia coli, Salmonella, Staphylococcus aureus, and Shigella. Once ideal conditions were determined, further optimization moved to prolonging the storage period of fermented pork jerky under all conditions. Our results revealed that optimal conditions for the storage of L. bulogaricus fermented pork jerky were a 10% inoculum, a 60‐hr fermentation period, a 30°C fermentation temperature, a 95°C baking temperature, and an 85‐min baking time. The storage period was determined to be 18 days at 30°C, 16 days longer than the non‐fermented pork jerky, and 14 days longer than the non‐optimized fermented pork jerky. This study provides key reference data for the industrial production of this foodstuff. Practical applications: In this work, we optimized the storage conditions for fermented pork jerky via microbial fermentation. Microorganisms play a vital role in the transformation of livestock and poultry meat into fermented meat products, with rich colors, unique flavors, and a longer shelf life.
