The sliding of roof snow may result in surcharges of snow load on lower roofs or the injury of pedestrians on the ground. It is therefore of great significance to study the mechanism of roof snow sliding, such that prevention or control measures can be developed to manage the risk. Considering four commonly used roofing materials, glass, steel, membrane, and concrete, two types of experiments were carried out in this study to possibly reveal the influence of roofing materials on the shear strength of the roof–snow interface: one is the critical angle tests where the angle at which the snow starts to slide off from the roof is tested, and the other is the shearing tests which aim to test the shear strength of the roof–snow interfaces at specific temperatures. The results showed that the critical angle for roof snow sliding, as well as the shear strength of the roof–snow interface for the four considered roofing materials, show a U-shape trend with the increase in surface roughness and that the shear strength of the roof–snow surface ranges from 0.15 kPa to 2 kPa for the cases considered, while the strength reaches its maximum at certain temperatures near −5 °C for a specific roofing material and snow thickness. Coastal wetland soil organic carbon (CW-SOC) is crucial for wetland ecosystem conservation and carbon cycling. The accurate prediction of CW-SOC content is significant for soil carbon sequestration. This study, which employed three machine learning (ML) methods, including random forest (RF), gradient boosting machine (GBM), and extreme gradient boosting (XGBoost), aimed to estimate CW-SOC content using 98 soil samples, SAR images, optical images, and climate and topographic data. Three statistical metrics and leave-one-out cross-validation were used to evaluate model performance. Optimal models using different ML methods were applied to predict the spatial distribution of CW-SOC content. The results showed the following: (1) The models built using optical images had higher predictive accuracy than models built using synthetic aperture radar (SAR) images. The model that combined SAR images, optical images, and climate data demonstrated the highest prediction accuracy. Compared to the model using only optical images and SAR images, the prediction accuracy was improved by 0.063 and 0.115, respectively. (2) Regardless of the combination of predictive variables, the XGBoost method achieved higher prediction accuracy than the RF and GBM methods. (3) Optical images were the main explanatory variables for predicting CW-SOC content, explaining more than 65% of the variability. (4) The CW-SOC content predicted by the three ML methods showed similar spatial distribution characteristics. The central part of the study area had higher CW-SOC content, while the southern and northern regions had lower levels. This study accurately predicted the spatial distribution of CW-SOC content, providing data support for ecological environmental protection and carbon neutrality of coastal wetlands. Soil total nitrogen (STN) is a crucial component of the ecosystem's nitrogen pool, and accurate prediction of STN content is essential for understanding global nitrogen cycling processes. This study utilized the measured STN content of 126 sample points and 40 extracted remote sensing variables to predict the STN content and map its spatial distribution in the northeastern coastal region of Hebei Province, China, employing the random forest (RF), gradient boosting machine (GBM), and extreme gradient boosting (XGBoost) methods. The purpose was to compare the ability of remote sensing images (Landsat-8, Sentinel-1, and Sentinel-2) with different machine learning methods for predicting STN content. The research results show the following: (1) The three machine learning methods accurately predicted the STN content and the optimal model provided by the XGBoost method, with an R2 of 0.627, RMSE of 0.127 g·kg−1, and MAE of 0.092 g·kg−1. (2) The combination of optical and synthetic aperture radar (SAR) images improved prediction accuracy, with the R2 improving by 45.5%. (3) The importance of optical images is higher than that of SAR images in the RF, GBM, and XGBoost methods, with optical images accounting for 87%, 76%, and 77% importance, respectively. (4) The spatial distribution of STN content predicted by the three methods is similar. Higher STN contents are distributed in the northern part of the study area, while lower STN contents are distributed in coastal areas. In the drilling process of Tarim Oilfield, a representative of ultra-deep oil and gas reservoirs, there are many problems of wellbore stability/instability caused by the development of a large number of micro-fractures. According to the nano-plugging mechanism, rigid nano-SiO2 and deformable nano-paraffin emulsion are added to the drilling fluid to improve the plugging rate. The effect of nanomaterials on the mechanical properties of limestone in the Karatal Formation was evaluated through a triaxial mechanical experiment, and it was found that rigid nano-SiO2 can have a better plugging effect under high formation pressure. It can increase the compressive strength of the limestone core by 10.32% and the cohesion of the core by 12.19%, and the internal friction angle of the core was increased from 25.67° to 26.39°. The internal structure of the core after nano-blocking was observed using CT scanning, and the fracture distribution state of the core before and after plugging and the fracture characteristics of the core under the pressure gradient were obtained, which confirmed that nano-SiO2 can effectively solve the fracture problem of deep limestone caused by micro-fractures. The science-practice gap has recently been discussed as a critical challenge restricting sustainable growth and development in all facets of our society, including explorations of Recreation Ecosystem Services (RES). To better explore how well the scientific study of RES and its application are connected, this paper aims to synthesize empirical evidence based on an in-depth and systematic literature review. We found that studies of RES have not effectively transformed into the decision-making and long-term planning of our cities. From 2005 to 2020, only 13% of studies referred to specific applications, and about 40% of papers mentioned no applications or practical implications for their research. However, RES research has many potential applications, which can be categorised into six main aspects. In terms of non-spatial improvement: Improved monetary benefits (40%), non-monetary benefits (30%); in terms of spatial improvement: space with high recreational potential or degradation (7%), the relation between supply and demand (7%); and Cross-service governance (16%). After combining the results of various studies, we developed a framework starting from applicable problems and their solutions, which can incorporate the outcomes of RES research while systematically narrowing down the research questions and methods. Roof structures are suffering serious threats caused by unbalanced snow distribution, especially long-span spatial structures, such as gable roofs. The formation of unbalanced snowdrifts on the gable roof is affected by the meteorological condition and the drifting snow. This study was conducted to explore the snowdrift characteristics on gable roofs under different snowfall conditions based on a new Eulerian–Eulerian multiphase approach. To consider the diffusive process of snow in different states, the governing equations of air and snow phases were modified separately according to the actual transport process. Additional terms based on the deposition/erosion process were inserted into the governing equations to consider the processes of snow particles being trapped or ejected by snow surface. The feasibility of the new model for the snowdrift was validated by comparing with a field observation. Then, the snowdrifts characteristics on typical gable roofs were investigated under different wind velocity and snowfall intensity conditions. The formation mechanism of snowdrifts and the influence of meteorological conditions on snowdrifts were clarified by analysis. The results show that the uneven distribution of snow on the gable roof becomes more significant with the increase in wind velocity. With the impact of urbanization and other factors, the vernacular architectural heritage faces the problem of how to carry on and innovate for sustainable development with originality and authenticity in the process of redesigning and planning. Therefore, this study uses the visualization method of quantitative analysis to analyze the research trends and hot spots of domestic vernacular architecture and explore new ways of coexistence between vernacular architecture, subject perception and cultural ecology. The critical challenge of this study is to analyze the theoretical coupling action rule between the socio-cultural structure of emotions and the cultural ecosystem of vernacular architecture, drawing on the concept of cultural ecology and the reasoning method of transmission relations in logical deduction. The main contribution of this study is to propose the concept of a subjective-cultural ecological design system for the sustainable development of vernacular architecture, to establish a three-dimensional structural analysis design paradigm and an evaluation analysis matrix, and to make vernacular architecture present the self-renewal ability of continuous exchange and revision in the dynamic cycle of this design system. The pathogenesis of type 2 diabetes (T2D) is highly related to the abnormal self-assembly of the human islet amyloid polypeptide (hIAPP) into amyloid aggregates. To inhibit hIAPP aggregation is considered a promising therapeutic strategy for T2D treatment. Melatonin (Mel) was reported to effectively impede the accumulation of hIAPP aggregates and dissolve preformed fibrils. However, the underlying mechanism at the atomic level remains elusive. Here, we performed replica-exchange molecular dynamics (REMD) simulations to investigate the inhibitory effect of Mel on hIAPP oligomerization by using hIAPP20–29 octamer as templates. The conformational ensemble shows that Mel molecules can significantly prevent the β-sheet and backbone hydrogen bond formation of hIAPP20–29 octamer and remodel hIAPP oligomers and transform them into less compact conformations with more disordered contents. The interaction analysis shows that the binding behavior of Mel is dominated by hydrogen bonding with a peptide backbone and strengthened by aromatic stacking and CH–π interactions with peptide sidechains. The strong hIAPP–Mel interaction disrupts the hIAPP20–29 association, which is supposed to inhibit amyloid aggregation and cytotoxicity. We also performed conventional MD simulations to investigate the influence and binding affinity of Mel on the preformed hIAPP1–37 fibrillar octamer. Mel was found to preferentially bind to the amyloidogenic region hIAPP20–29, whereas it has a slight influence on the structural stability of the preformed fibrils. Our findings illustrate a possible pathway by which Mel alleviates diabetes symptoms from the perspective of Mel inhibiting amyloid deposits. This work reveals the inhibitory mechanism of Mel against hIAPP20–29 oligomerization, which provides useful clues for the development of efficient anti-amyloid agents. [ABSTRACT FROM AUTHOR]

Bipedal models for walkers, originally developed in the research field of biomechanics, have been identified as potential candidates for modelling pedestrians in structural engineering applications. These models provide insight into both the kinetics and kinematics of walking locomotion and are considered to have a significant potential to improve the vibration serviceability assessment of civil engineering structures. Despite this notion, the ability of the bipedal models to represent the key features of the walking gait and natural variability within the pedestrian population are still under-researched. This paper critically evaluates the performance of two bipedal models with rigid legs to realistically both reproduce key features of an individual pedestrian's walking gait and represent a wide range of individuals. The evaluation is performed for walking on a rigid, rather than vibrating, structure due to the availability of experimental data and expectation that successful modelling on rigid surfaces is a necessary condition for progressing towards modelling on the vibrating structures. Ready-to-use equations are provided and the ability of the models to represent the kinematics and kinetics of individual pedestrians as well as the inter-subject variability typical of the human population is critically evaluated. It was found that the two models could generate realistic combinations of the gait parameters and their correlations, but are less successful in reproducing genuine kinetic and kinematics profiles. To explore the interference effects of a high-rise building on the snow load on a low-rise building with a flat roof, a series of wind tunnel tests were carried out with fine silica sand as a substitute for snow particles. The effects of the height of the interfering building and the distance between buildings on the snow distribution of the target building under three different wind directions were studied. The snow depth on the target building roof and the mass of particles blown off from the target building were measured during the wind tunnel tests, and the results showed that the snow distribution of the target building roof tends to be uniform when the interfering building is located upstream of the target building due to the shelter effect. When the interfering building is on the side of the target building, the snow distribution of the target building tends to be more uneven, because the interfering building increases the friction velocity on the target building roof near the interfering building. However, when the interfering building is located downstream of the target building, there will be an amplification effect of snow accumulation, and the snow distribution on the target building roof is nearly the same as that of the isolated condition. Under each wind direction, the interference effect of the snow load increases with the increase of the building height and the decrease of the building spacing. Therefore, the influence of the surrounding buildings on the snow distribution of the building roof cannot be ignored and should be considered in the structure design. [ABSTRACT FROM AUTHOR]

Featured Application: This study provides a simplified method that can be applied to help preliminarily estimate the snow load on a complex roof by combining the snow loads on several simple roofs, which have been provided in the load code or obtained by numerical studies. A considerable number of studies have been carried out for predicting snowdrifts on roofs over the years. However, few studies have focused on snowdrifts on complex long-span roofs, as the complex shape and fine structure pose significant challenges. In this study, to simplify the calculation requirements of snow load on such roofs, work was conducted to decompose the snowdrift on a complex roof into snowdrifts on several simple roofs. First, the snow–wind tunnel test similarity criteria were investigated based on a combined air–snow–wind experimental system. Thereafter, with reference to the validated experimental similarity criteria, a series of snow–wind tunnel tests were performed for snowdrifts on a complex long-span structure under the conditions of different inflow directions. Finally, based on empirical orthogonal function (EOF) analysis, the snowdrifts on the complex roof were decomposed into basic characteristic distribution modes, including snowdrifts caused by the local and overall roof forms. The snow distribution under a specific inflow direction could be derived from the weighted combination of the basic characteristic modes, based on the wind direction coefficients. Featured Application: A new approach for investigating snowdrifts around buildings and snow loads on building roofs based on a new facility named the "snow–wind combined experimental facility" is proposed in this paper. First, the method proposed in this paper is convenient and economical, and also the experimental results prove that the method is feasible and reliable. In this paper, only experiments on snowdrift around a cube were chosen for the verification for the sake of generality, but this method could also be adopted for the prediction of snowdrift around complex building environments and snow loads on various shapes of building roofs. Unbalanced, or non-uniform, snow loads caused by snow drifting or sliding in cold regions with heavy snowfalls, can be a serious problem for the building industry. However, the methods for predicting snow distribution still need to be improved. Field observation is the most direct and reliable method to study snow distribution, but because the natural environment is uncontrollable and varies dramatically, sometimes conclusions may be confused under the influence of the many variables in the investigation. This paper proposes a snowing experiment approach using an outdoor snow–wind combined experiment facility for the study of snow distribution. The facility can produce a stable and controllable wind field and snowfall environment. Experiments which focused on snowdrift around a building were conducted during the winter to make an evaluation of the repeatability and reliability of the new approach. Finally, from the analysis of results, it was demonstrated that the experimental facility was stable and that the similarity criterion adopted for the snowing pattern was reliable. Maize belongs to a tropical environment and is extremely sensitive to drought and chilling stress, particularly at early developmental stages. The present study investigated the individual and combined effects of drought (15% PEG-Solution) and chilling stress (15/12 °C) on morpho-physiological growth, osmolyte accumulation, production of reactive oxygen species (ROS), and activities/levels of enzymatic and non-enzymatic antioxidants in two maize hybrids (i.e., "XD889" and "XD319") and two inbred cultivars (i.e., "Yu13" and "Yu37"). Results revealed that individual and combined exposure of drought and chilling stresses hampered the morpho-physiological growth and oxidative status of maize cultivars, nevertheless, the interactive damage caused by drought + chilling was found to be more severe for all the studied traits. Between two individual stress factors, chilling-induced reductions in seedling length and biomass of maize cultivars were more compared with drought stress alone. Greater decrease in root length and biomass under chilling stress ultimately decreased the volume and surface area of the root system, and restricted the shoot growth. All the stress treatments, particularly chilling and drought + chilling, triggered the oxidative stress by higher accumulation of superoxide anion, hydrogen peroxide, hydroxyl ion, and malondialdehyde contents compared with the control. Variations in response of maize cultivars were also apparent against different stress treatments, and XD889 performed comparatively better than the rest of the cultivars. The better growth and greater stress tolerance of this cultivar was attributed to the vigorous root system architecture, as indicated by higher root biomass, root surface area, and root volume under drought and chilling stresses. Moreover, efficient antioxidant defense system in terms of higher total antioxidant capability, superoxide dismutase, peroxidase, catalase, and glutathione reductase activities also contributed in greater stress tolerance of XD889 over other cultivars. Digestate and biogas slurry (BS) are the byproduct of biogas engineering that could be used for elevating plant growth. However, the consequent emissions of ammonia from BS are considered a severe threat to the atmosphere. Herein, we conducted two consecutive field experiments with wheat–maize rotations to find out the optimum ratio of BS to combine with chemical fertilizer (CF) to reduce ammonia volatilization (AV) while keeping the stable crop yield. In maize season, 226.5 kg N/ha of CF was applied. In wheat season, 226.5 kg N/ha was applied at different ratios (100%, 80%, and 50%) between BS and CF. Our results found that the maximum yield of 6250 kg/ha was produced by CF, and this yield could be obtained through a combined application of 38% BS mixed with CF. Highest AV produced of 16.08 kg/ha by CF. BS treatments significantly reduced the emission from 18% to 32% in comparison to CF. The combined application of BS-CF produced the highest yield due to essential nutrients coming from both BS-CF. Subsequently, it reduced the AV depending on fertilizer type and fertilizer rate. An optimal ratio of 38% BS was recommended to produce the highest yield and lowest ammonia emissions. Inflammation is the body's self-protective response to multiple stimulus, from external harmful substances to internal danger signals released after trauma or cell dysfunction. Many diseases are considered to be related to inflammation, such as cancer, metabolic disorders, aging, and neurodegenerative diseases. Current therapeutic approaches include mainly non-steroidal anti-inflammatory drugs and glucocorticoids, which are generally of limited effectiveness and severe side-effects. Thus, it is urgent to develop novel effective anti-inflammatory therapeutic agents. Xanthones, a unique scaffold with a 9H-Xanthen-9-one core structure, widely exist in natural sources. Till now, over 250 xanthones were isolated and identified in plants from the families Gentianaceae and Hypericaceae. Many xanthones have been disclosed with anti-inflammatory properties on different models, either in vitro or in vivo. Herein, we provide a comprehensive and up-to-date review of xanthones with anti-inflammatory properties, and analyzed their drug likeness, which might be potential therapeutic agents to fight against inflammation-related diseases. Asymmetric triangulation is an interesting method combined with concentric pleating to obtain a 3D shape without stretching or tearing. There exists some geometric properties in the process of folding to help realize extension and contraction, which can be used in parametric modeling of different regular polygons. To facilitate design and modeling, adequate computational modeling methods are indispensable. This paper proposes a new mathematical idea and presents a feasible way to build the parameterized models in the digital environment of Rhinoceros, utilizing the Kangaroo plugin in Grasshopper. Designers can directly observe the models' kinematic deployment and calculate the folding efficiency. The pathology Alzheimer's disease (AD) is associated with the self-assembly of amyloid-β (Aβ) peptides into β-sheet enriched fibrillar aggregates. A promising treatment strategy is focused on the inhibition of amyloid fibrillization of Aβ peptide. Fullerene C60 is proved to effectively inhibit Aβ fibrillation while the poor water-solubility restricts its use as a biomedicine agent. In this work, we examined the interaction of fullerene C60 and water-soluble fullerenol C60(OH)6/C60(OH)12 (C60 carrying 6/12 hydroxyl groups) with preformed Aβ40/42 protofibrils by multiple molecular dynamics simulations. We found that when binding to the Aβ42 protofibril, C60, C60(OH)6 and C60(OH)12 exhibit distinct binding dynamics, binding sites and peptide interaction. The increased number of hydroxyl groups C60 carries leads to slower binding dynamics and weaker binding strength. Binding free energy analysis demonstrates that the C60/C60(OH)6 molecule primarily binds to the C-terminal residues 31–41, whereas C60(OH)12 favors to bind to N-terminal residues 4–14. The hydrophobic interaction plays a critical role in the interplay between Aβ and all the three nanoparticles, and the π-stacking interaction gets weakened as C60 carries more hydroxyls. In addition, the C60(OH)6 molecule has high affinity to form hydrogen bonds with protein backbones. The binding behaviors of C60/C60(OH)6/C60(OH)12 to the Aβ40 protofibril resemble with those to Aβ42. Our work provides a detailed picture of fullerene/fullerenols binding to Aβ protofibril, and is helpful to understand the underlying inhibitory mechanism. [ABSTRACT FROM AUTHOR]

Sloping farmland is an essential type of the farmland resource in China. In the Sichuan province, livelihood security and social development are particularly sensitive to changes in the sloping farmland, due to the region's large portion of hilly territory and its over-dense population. In this study, we focused on spatiotemporal change of the sloping farmland and its driving forces in the Sichuan province. Sloping farmland areas were extracted from geographic data from digital elevation model (DEM) and land use maps, and the driving forces of the spatiotemporal change were analyzed using a principal component analysis (PCA). The results indicated that, from 2000 to 2015, sloping farmland decreased by 3263 km2 in the Sichuan province. The area of gently sloping farmland (<10°) decreased dramatically by 1467 km2, especially in the capital city, Chengdu, and its surrounding areas. However, the steep sloping farmland (>25°) decreased by 302 km2, and was still the largest portion of total farmland in the area. The PCA analysis indicated that the main driving forces behind the changes were social and economic factors. The influence of agricultural intensification factors, such as the multiple cropping index and sown areas of crops, was relatively weak. Given the decrease in the overall slope cultivated area and the increased portion of moderately steep slope land (10–25°) in the cultivated area, special attention should be paid to the scientific conservation of sloping farmland during rapid social and economic development. The success of the first approved kinase inhibitor imatinib has spurred great interest in the development of type II inhibitors targeting the inactive DFG-out conformation, wherein the Phe of the DFG motif at the start of the activation loop points into the ATP binding site. Nevertheless, kinase inhibitors launched so far are heavily biased toward type I inhibitors targeting the active DFG-in conformation, wherein the Phe of the DFG motif flips by approximately 180° relative to the inactive conformation, resulting in Phe and Asp swapping their positions. Data recently obtained with structurally validated type II inhibitors supported the conclusion that type II inhibitors are more selective than type I inhibitors. In our type II BRAF V600E inhibitor lead discovery effort, we identified phenylaminopyrimidine (PAP) and unsymmetrically disubstituted urea as two fragments that are frequently presented in FDA-approved protein kinase inhibitors. We therefore defined PAP and unsymmetrically disubstituted urea as privileged fragments for kinase drug discovery. A pharmacophore for type II inhibitors, 4-phenylaminopyrimidine urea (4-PAPU), was assembled based upon these privileged fragments. Lead compound SI-046 with BRAF V600E inhibitory activity comparable to the template compound sorafenib was in turn obtained through preliminary structure-activity relationship (SAR) study. Molecular docking suggested that SI-046 is a bona fide type II kinase inhibitor binding to the structurally validated "classical DFG-out" conformation of BRAF V600E. Our privileged fragments-based approach was shown to efficiently deliver a bona fide type II kinase inhibitor lead. In essence, the theme of this article is to showcase the strategy and rationale of our approach.

Box-Behnken design (BBD), one of the most common response surface methodology (RSM) methods, was used to optimize the experimental conditions for ultrasound-assisted extraction of polysaccharides from Rhynchosia minima root (PRM). The antioxidant abilities and anticancer activity of purified polysaccharide fractions were also measured. The results showed that optimal extraction parameters were as follows: ultrasound exposure time, 21 min; ratio of water to material, 46 mL/g; ultrasound extraction temperature, 63 °C. Under these conditions, the maximum yield of PRM was 16.95%±0.07%. Furthermore, the main monosaccharides of purified fractions were Ara and Gal. PRM3 and PRM5 exhibited remarkable DPPH radical scavenging activities and reducing power in vitro. PRM3 showed strong inhibitory activities on the growth of MCF-7 cells in vitro. The above results indicate that polysaccharides from R. minima root have the potential to be developed as natural antioxidants and anticancer ingredients for the food and pharmaceutical industries.
