Objective: The objective of this work was to assess the effect of physical therapy in patients with somatosensory tinnitus (ST) and explore the influence of physical therapy on clinical variables obtained before treatment. Methods: A total of 43 patients with ST were randomized to the immediate-start group (n = 20) and delayed-start group (n = 23). All patients received physical therapy for 1 week (seven sessions). Each session lasted 60 min. The Visual Analogue Scale (VAS), Tinnitus Handicap Inventory (THI), and numerical pain rating scale (NPRS) scores were documented at baseline and after treatment (week 1) for all patients. For subjects in the immediate-start group, the THI, VAS, and NPRS scores were measured after therapy (weeks 6, 9, and 12, respectively). Medical history characteristic functional activity scale (HCFA) scores were measured at baseline to assess the association between somatic symptoms and tinnitus. Results: At week 1, VAS, THI, and NPRS scores of patients in the immediate-start group were improved by 1.25 ± 1.59, 11.10 ± 15.10, and 0.95 ± 1.54 points, respectively, and were significantly higher than those in the delayed-start group (p < 0.05). The change in VAS, THI, and NPRS scores in the treatment group was significantly positively correlated with the scores of the HCFA before treatment (r = 0.786, p < 0.001; r = 0.680, p = 0.001; r = 0.796, p < 0.001). There was no significant difference in THI, VAS, and NPRS scores among patients in the immediate-start group between weeks 1, 6, 9, and 12 after treatment (p > 0.05). Conclusions: Although more participants were necessary in the further study, the study implies that physical therapy can reduce physical pain, improve tinnitus symptoms, and quality of life in ST patients without hearing loss, and the short-term curative effect is stable, especially for tinnitus patients with clear somatic symptoms. Simple Summary: Excessive white adipose tissue accumulation in farm animal results in a lower meat percentage of the carcass, which may attenuate economic benefits. Moreover, consumers prefer to purchase lean meat without excessive subcutaneous fat. Thus, developing strategies to enhance skeletal muscle growth and maintain or decrease adiposity in farm animals is desirable. Guanidinoacetic acid (GAA) has been officially registered as an animal feed additive to promote the performance of animals. Currently, little is known about its effects on sheep adipose tissue accumulation. We found that dietary GAA supplementation could attenuate adipose tissue growth in sheep. Moreover, GAA inhibited proliferation and differentiation of sheep stromal vascular fraction (SVF) cells in vitro. Thus, these data could potentiate the application of GAA to sheep meat production. Guanidinoacetic acid (GAA) is an amino acid derivative, previously described in the skeletal muscle of vertebrates, that serves as an important regulator of cellular bioenergetics and has been widely used as a feed additive. Nevertheless, the effect of GAA on adipose tissue growth remains unclear. Here, we hypothesized that dietary GAA negatively affected adipose tissue development in lambs. Lambs were individually fed diets with (0.09%) or without GAA for 70 d ad libitum, and the subcutaneous adipose tissues were sampled for analysis. The results showed that dietary GAA supplementation decreased the girth rib (GR) value (p < 0.01) of lamb carcasses. Both real-time PCR and Western blot analysis suggested that dietary GAA inhibited the expression of adipogenic markers, including peroxisome proliferator-activated receptor γ (PPARγ, p < 0.05), CCAAT/enhancer-binding protein α (C/EBPα, p < 0.01) and sterol-regulatory-element-binding protein 1c (SREBP1C, p < 0.01) in subcutaneous adipose tissue. In vitro, GAA inhibited sheep stromal vascular fraction (SVF) cell proliferation, which was associated with downregulation of proliferating cell nuclear antigen (PCNA, p < 0.05), cyclin-dependent kinase 4 (CDK 4, p < 0.05) and cyclin D1 (p < 0.01). GAA suppressed adipogenesis of SVF cells. Furthermore, miRNA sequencing revealed that GAA affected the miRNA expression profile, and real-time PCR analysis confirmed that miR-133a expression in both subcutaneous adipose tissue and SVF cell was downregulated by GAA. Meanwhile, miR-133a promoted adipogenic differentiation of SVF cells by targeting Sirt1. miR-133a mimics alleviated the inhibitory effect of GAA on SVF cells' adipogenic differentiation. Neurological diseases including stroke and neurodegenerative disorders cause a hefty burden on the healthcare system. Survivors experience significant impairment in mobility and daily activities, which requires extensive rehabilitative interventions to assist them to regain lost skills and restore independence. The advent of remote rehabilitation architecture and enabling technology mandates the elaboration of sensing mechanisms tailored to individual clinical needs. This study aims to review current trends in the application of sensing mechanisms in remote monitoring and rehabilitation in neurological diseases, and to provide clinical insights to develop bespoke sensing mechanisms. A systematic search was performed using the PubMED database to identify 16 papers published for the period between 2018 to 2022. Teleceptive sensors (56%) were utilized more often than wearable proximate sensors (50%). The most commonly used modality was infrared (38%) and acceleration force (38%), followed by RGB color, EMG, light and temperature, and radio signal. The strategy adopted to improve the sensing mechanism included a multimodal sensor, the application of multiple sensors, sensor fusion, and machine learning. Most of the stroke studies utilized biofeedback control systems (78%) while the majority of studies for neurodegenerative disorders used sensors for remote monitoring (57%). Functional assessment tools that the sensing mechanism may emulate to produce clinically valid information were proposed and factors affecting user adoption were described. Objective: To study the effect and mechanism of the Clostridium metabolite p-Cresol sulfate (PCS) in primary biliary cholangitis (PBC). Methods: Gas chromatography-mass spectrometry (GC-MS) was used to detect differences in tyrosine, phenylalanine, tryptophan, PCS, and p-Cresyl glucuronide (PCG) between the serum of PBC patients and healthy controls. In vivo experiments, mice were divided into the normal control, PBC group, and PBC tyrosine group. GC-MS was used to detect PCS and PCG. Serum and liver inflammatory factors were compared between groups along with the polarization of liver Kupffer cells. Additionally, PCS was cultured with normal bile duct epithelial cells and Kupffer cells, respectively. PCS-stimulated Kupffer cells were co-cultured with lipopolysaccharide-injured bile duct epithelial cells to detect changes in inflammatory factors. Results: Levels of tyrosine and phenylalanine were increased, but PCS level was reduced in PBC patients, with PCG showing a lower concentration distribution in both groups. PCS in PBC mice was also lower than those in normal control mice. After oral administration of tyrosine feed to PBC mice, PCS increased, liver inflammatory factors were decreased, and anti-inflammatory factors were increased. Furthermore, Kupffer cells in the liver polarized form M1 transitioned to M2. PCS can damage normal bile duct epithelial cells and suppress the immune response of Kupffer cells. But PCS protects bile duct epithelial cells damaged by LPS through Kupffer cells. Conclusions: PCS produced by Clostridium-metabolized tyrosine reduced PBC inflammation, suggesting that intervention by food, or supplementation with PCS might represent an effective clinical strategy for treating PBC. Dual thermo- and pH-responsive comb-type grafted hydrogels of poly(N,N-dimethy laminoethyl methacrylate) (PDMAEMA) and poly(N-isopropylacrylamide) (PNIPAM) with reversed network-graft architectures were synthesized by the combination of atom transfer radical polymerization (ATRP), reversible addition-fragmentation chain transfer (RAFT) polymerization and click chemistry. Two kinds of macro-cross-linkers with two azido groups at one chain-end and different chain length [PNIPAM--(N3)2 and PDMAEMA--(N3)2] were prepared with N,N-di(β-azidoethyl) 2-halocarboxylamide as the ATRP initiator. Through RAFT copolymerization of DMAEMA or NIPAM with propargyl acrylate (ProA) using dibenzyltrithiocarbonate as a chain transfer agent, two network precursors with different content of alkynyl side-groups [P(DMAEMA-co-ProA) and P(NIPAM-co-ProA)] were obtained. The subsequent azido-alkynyl click reaction of macro-cross-linkers and network precursors led to the formation of the network-graft hydrogels. These dual stimulus-sensitive hydrogels exhibited rapid response, high swelling ratio and reproducible swelling/de-swelling cycles under different temperatures and pH values. The influences of cross-linkage density and network-graft architecture on the properties of the hydrogels were investigated. The release of ceftriaxone sodium from these hydrogels showed both thermal- and pH-dependence, suggesting the feasibility of these hydrogels as thermo- and pH-dependent drug release devices. Food allergies are a serious food safety and public health issue. Soybean, dairy, aquatic, poultry, and nut products are common allergens inducing allergic reactions and adverse symptoms such as atopic dermatitis, allergic eczema, allergic asthma, and allergic rhinitis. Probiotics are assumed as an essential ingredient in maintaining intestinal microorganisms' composition. They have unique physiological roles and therapeutic effects in maintaining the mucosal barrier, immune function, and gastrointestinal tract, inhibiting the invasion of pathogenic bacteria, and preventing diarrhea and food allergies. Multiple pieces of evidence reveal a significant disruptive effect of probiotics on food allergy pathology and progression mechanisms. Thus, this review describes the allergenic proteins as an entry point and briefly describes the application of probiotics in allergenic foods. Then, the role of probiotics in preventing and curing allergic diseases by regulating human immunity through intestinal flora and intestinal barrier, modulating host immune active cells, and improving host amino acid metabolism are described in detail. The anti-allergic role of probiotics in the function and metabolism of the gastrointestinal tract has been comprehensively explored to furnish insights for relieving food allergy symptoms and preventing food allergy. China is the largest producer and exporter of coke globally, which means that it is very important to understand the characteristics of air pollutants and carbon emissions from China's independent coking industry. This study was the first to establish a bottom-up inventory of the air pollutants and carbon emissions of China's independent coking industry during 2001–2018 based on continuous emission monitoring system online monitoring data and unit-based corporate information. Based on the developed emission inventory, four scenarios were established to analyze potential emissions reduction of air pollutants and carbon dioxide (CO2) in future. The emissions of particulate matter (PM10 and PM2.5), sulfur dioxide (SO2), black carbon (BC) and organic carbon (OC) decreased by 62.11%, 63.41%, 72.85%, 63.41% and 63.41%, respectively. CO2, carbon monoxide (CO), volatile organic compounds (VOCs) and nitrogen oxides (NOX) emissions increased by 355.51%, 355.51%, 355.51% and 99.74%, respectively. In 2018, PM10, PM2.5, SO2, NOx, BC, OC, CO, VOCs and CO2 emissions were, respectively. 45.20, 16.91, 63.84, 117.71, 5.07, 5.92, 554.91, 1026.58 Gg, and 176.88 Tg. Shanxi province made the greatest contributions to the total emissions of air pollutants and CO2 from this industry by 25.01%. The emission source that contributed most to PM2.5 (SO2 and NOX) emissions was coke pushing (quenching and the coke oven chimney respectively) in 2018. Under the ULE scenario (2018–2035), PM2.5 and SO2 emissions will reduce by more than 30%. Under the PCP scenario, PM2.5 and SO2 emissions will reduce by more than 55%. Under the CBP scenario, CO2 emissions will peak at 197.99 Tg in 2025 and decrease to 70% of the peak in 2035. The results showed the emission characteristics of air pollutants and CO2, future emission with several scenarios and cooperative reduction potential in China's independent coking industry, which provides scientific support for the development of pollution control strategies. The excessive use or abuse of pesticides and veterinary drugs leads to residues in food, which can threaten human health. Therefore, there is an extremely urgent need for multi-analyte analysis techniques for the detection of pesticide and veterinary drug residues, which can be applied as screening techniques for food safety monitoring and detection. Recent developments related to rapid multi-residue detection methods for pesticide and veterinary drug residues are reviewed herein. Methods based on different recognition elements or the inherent characteristics of pesticides and veterinary drugs are described in detail. The preparation and application of three broadly specific recognition elements—antibodies, aptamers, and molecular imprinted polymers—are summarized. Furthermore, enzymatic inhibition-based sensors, near-infrared spectroscopy, and SERS spectroscopy based on the inherent characteristics are also discussed. Chlorophyll fluorescence (ChlF) parameters, especially the quantum efficiency of photosystem II (PSII) in dark- and light-adapted conditions (Fv/Fm and Fv'/Fm'), have been used extensively to indicate photosynthetic activity, physiological function, as well as healthy and early stress conditions. Previous studies have demonstrated the potential of applying hyperspectral data for the detection of ChlF parameters in vegetation. However, the performance of spectral features that have been documented to estimate ChlF is not ideal and is poorly understood. In this study, ChlF parameters and leaf reflectance were collected in two field experiments involving various wheat cultivars, nitrogen (N) applications, and plant densities, during the growing seasons of 2014 to 2015 and 2015 to 2016. Three types of spectral features, including vegetation indices (VIs), red edge position (REP), and wavelet features, were used to quantify ChlF parameters Fv/Fm and Fv'/Fm'. The results indicated that traditional chlorophyll fluorescence vegetation indices (ChlF VIs), such as the curvature index (CUR) and D705/D722 were capable of detecting Fv/Fm and Fv'/Fm' under various scenarios. However, the wavelet-based REP (WREP-S4) and the wavelet feature (WF) (704 nm, scale 4) yielded higher accuracy than other spectral features in calibration and validation datasets. Moreover, the bands used to calculate WREP-S4 and WF (704 nm, scale 4) were all centered in the red edge region (680 to 760 nm), which highlighted the role of the red edge region in tracking the change of active ChlF signal. Our results are supported by previous studies, which have shown that the red edge region is vital for estimating the chlorophyll content, and also the ChlF parameters. Clip domain serine proteases play vital roles in various innate immune functions and in embryonic development. Nilaparvata lugens proclotting enzymes (NlPCEs) belong to this protease family. NlPCE1 was reported to be involved in innate immunity, whereas the role of other NlPCEs is unclear. In the present study, N. lugens proclotting enzyme-3 (NlPCE3) was cloned and characterized. NlPCE3 contains a signal peptide, a clip domain, and a trypsin-like serine protease domain. NlPCE3 was expressed in all tissues examined (gut, fat body, and ovary), and at all developmental stages. Immunofluorescence staining showed that NlPCE3 was mainly expressed in the cytoplasm and cytomembrane of follicular cells. Double stranded NlPCE3 RNA interference clearly inhibited the expression of NlPCE3, resulting in abnormal egg formation and obstruction of ovulation. In recent decades, the increasing frequency and severity of cyanobacterial blooms in recreational lakes and water supply reservoirs have become a great concern to public health and a significant threat to the environment. Cyanobacterial bloom monitoring is the basis of early warning and treatment. Previous research efforts have always focused on monitoring blooms in a few specific lakes in China using moderate resolution imaging spectroradiometer (MODIS) images, which are available for the years 2000 onward. However, the lack of overall information on long-term cyanobacterial blooms in the lakes and reservoirs in the middle–lower Yangtze River (MLYR) basin is an obstacle to better understanding the dynamics of cyanobacterial blooms at a watershed scale. In this study, we extracted the yearly coverage area and frequency of cyanobacterial blooms that occurred from 1990 to 2016 in 30 large lakes and 10 reservoirs (inundation area >50 km2) by using time series Landsat satellite images from Google Earth Engine (GEE). Then, we calculated the cyanobacterial bloom area percentage (CAP) and the cyanobacterial bloom frequency index (CFI) and analyzed their inter-annual variation and trends. We also investigated the main driving forces of changes in the CAP and CFI in each lake and reservoir. We found that all reservoirs and more than 60% of lakes exhibited an increasing frequency and coverage area of cyanobacterial blooms under the pressures of climate change and anthropogenic interferences. Reservoirs were more prone to be affected by fertilizer consumption from their regional surroundings than lakes. High temperatures increased blooms of cyanobacteria, while precipitation in the lake and reservoir regions somewhat alleviated blooms. This study completes the data records of cyanobacterial blooms in large lakes and reservoirs located in hotspots of the MLYR basin and provides more baseline information before 2000, which will present references for water resource management and freshwater conservation. Whereas modern digital cameras use a pixelated detector array to capture images, single-pixel imaging reconstructs images by sampling a scene with a series of masks and associating the knowledge of these masks with the corresponding intensity measured with a single-pixel detector. Though not performing as well as digital cameras in conventional visible imaging, single-pixel imaging has been demonstrated to be advantageous in unconventional applications, such as multi-wavelength imaging, terahertz imaging, X-ray imaging, and three-dimensional imaging. The developments and working principles of single-pixel imaging are reviewed, a mathematical interpretation is given, and the key elements are analyzed. Precise detection of leaf nitrogen concentration (LNC) is helpful for nutrient diagnosis and fertilization guidance in farm crops. Numerous researchers have estimated LNC with techniques based on reflectance spectra or active chlorophyll fluorescence, which have limitations of low accuracy or small scale in the field. Given the correlation between chlorophyll and nitrogen contents, the response of sun-induced chlorophyll fluorescence (SIF) to chlorophyll (Chl) content reported in a few papers suggests the feasibility of quantifying LNC using SIF. Few studies have investigated the difference and power of the upward and downward SIF components on monitoring LNC in winter wheat. We conducted two field experiments to evaluate the capacity of SIF to monitor the LNC of winter wheat during the entire growth season and compare the differences of the upward and downward SIF for LNC detection. A FluoWat leaf clip coupled with a ASD spectrometer was used to measure the upward and downward SIF under sunlight. It was found that three (↓FY687, ↑FY687/↑FY739, and ↓FY687/↓FY739) out of the six SIF yield (FY) indices examined were significantly correlated to the LNC (R2 = 0.6, 0.51, 0.75, respectively). The downward SIF yield indices exhibited better performance than the upward FY indices in monitoring the LNC with the ↓FY687/↓FY739 being the best FY index. Moreover, the LNC models based on the three SIF yield indices are insensitive to the chlorophyll content and the leaf mass per area (LMA). These findings suggest the downward SIF should not be neglected for monitoring crop LNC at the leaf scale, although it is more difficult to measure with current instruments. The downward SIF could play an increasingly important role in understanding of the SIF emission for LNC detection at different scales. Pseudomonas aeruginosa-induced biofilm infection is difficult to treat and poses a significant threat to public health. Our previous study found a new coumarin derivative LP4C which exerted potent in vitro and in vivo anti-biofilm activity against Pseudomonas aeruginosa; however, the underlying molecular mechanism and drug-likeness of LP4C is unclear. In this study, we confirmed that LP4C could inhibit the biofilm in dose-dependent manner without bactericidal activity. The transcriptomic profiling and RT-PCR result revealed that bacterial pyrimidine mediated the inhibitory activity of LP4C. The cell viability was not affected in LP4C treatment groups with the concentration under 200 μg/mL, and no death or toxicity sign was observed in mice treated by 20, 40 and 80 mg/kg LP4C during the three-week test period. Ames test presented that LP4C had no effect on the bacterial reverse mutation. In additional, pharmacokinetic results showed that LP4C was likely to have the orally bioavailable properties. Salvianic acid A (SAA), as the main bioactive component of the traditional Chinese herb Salvia miltiorrhiza, has important application value in the treatment of cardiovascular diseases. In this study, a two-step bioprocess for the preparation of SAA from l-DOPA was developed. In the first step, l-DOPA was transformed to 3,4-dihydroxyphenylalanine (DHPPA) using engineered Escherichia coli cells expressing membrane-bound L-amino acid deaminase from Proteus vulgaris. After that, the unpurified DHPPA was directly converted into SAA by permeabilized recombinant E. coli cells co-expressing d-lactate dehydrogenase from Pediococcus acidilactici and formate dehydrogenase from Mycobacterium vaccae N10. Under optimized conditions, 48.3 mM of SAA could be prepared from 50 mM of l-DOPA, with a yield of 96.6%. Therefore, the bioprocess developed here was not only environmentally friendly, but also exhibited excellent production efficiency and, thus, is promising for industrial SAA production. Location-based services have permeated Smart academic institutions, enhancing the quality of higher education. Position information of people and objects can predict different potential requirements and provide relevant services to meet those needs. Indoor positioning system (IPS) research has attained robust location-based services in complex indoor structures. Unforeseeable propagation loss in complex indoor environments results in poor localization accuracy of the system. Various IPSs have been developed based on fingerprinting to precisely locate an object even in the presence of indoor artifacts such as multipath and unpredictable radio propagation losses. However, such methods are deleteriously affected by the vulnerability of fingerprint matching frameworks. In this paper, we propose a novel machine learning framework consisting of Bag-of-Features and followed by a k-nearest neighbor classifier to categorize the final features into their respective geographical coordinate data. BoF calculates the vocabulary set using k-mean clustering, where the frequency of the vocabulary in the raw fingerprint data represents the robust final features that improve localization accuracy. The effects of essential oil from Eucalyptus camaldulensis flowers oil on melanogenesis and the oil's antioxidant characteristics were investigated. Assays of mushroom and cellular tyrosinase activities and melanin content of mouse melanoma cells were performed spectrophotometrically, and the expression of melanogenesis-related proteins was determined by Western blotting. The possible signaling pathways involved in essential oil-mediated depigmentation were also investigated using specific protein kinase inhibitors. The results revealed that E. camaldulensis flower essential oil effectively suppresses intracellular tyrosinase activity and decreases melanin amount in B16F10 mouse melanoma cells. The essential oil also exhibits antioxidant properties and effectively decreases intracellular reactive oxygen species (ROS) levels. The volatile chemical composition of the essential oil was analyzed with gas chromatography-mass spectrometry (GC/MS). The chemical constituents in the essential oil are predominately oxygenated monoterpenes (34.9%), followed by oxygenated sesquiterpenes (31.8%), monoterpene hydrocarbons (29.0%) and sesquiterpene hydrocarbons (4.3%). Our results indicated that E. camaldulensis flower essential oil inhibits melanogenesis through its antioxidant properties and by down-regulating both mitogen-activated protein kinases (MAPK) and protein kinase A (PKA) signaling pathways. Four previously undescribed compounds, including three rarely occurring seco-dammarane triterpenoid glycosides and a pentacyclic triterpenic acid, were isolated from a 70% ethanol extract of the leaves of Cyclocarya paliurus (Juglandaceae), along with eleven known triterpenoids. Their structures were determined by spectroscopic techniques, including 2D NMR and HRESIMS, as well as chemical methods. Among them, several triterpenoids enhanced insulin stimulated glucose uptake in both 3T3-L1 adipocytes and C2C12 myotubes. Furthermore, compound 1 dose-dependently increased glucose uptake through activating AMP-activated protein kinase (AMPK)-p38 pathway. It is well-documented that CL316,243 (a β3 agonist) or rosiglitazone (a PPARγ agonist) can induce white adipocyte populations to brown-like adipocytes, thus increasing energy consumption and combating obesity. However, whether there is a combined effect remains unknown. In the present study, stromal vascular cells of inguinal white adipose tissue (iWAT-SVCs for short) from mice were cultured and induced into browning by CL316,243, rosiglitazone, or both. Results showed that a combination of CL316,243 and rosiglitazone significantly upregulated the expression of the core thermogenic gene Ucp1 as well as genes related with mitochondrial function (Cidea, Cox5b, Cox7a1, Cox8b, and Cycs), compared with the treatment of CL316,243 or rosiglitazone alone. Moreover, co-treatment with rosiglitazone could reverse the downregulation of Adiponectin resulting from CL316,243 stimuli alone. Taken together, a combination of rosiglitazone and CL316,243 can produce an additive effect of promoting thermogenic gene expression and an improvement of insulin sensitivity in mouse iWAT-SVCs. Tidal flats, which are non-vegetated land–sea transition areas, have an important ecological function in the global ecosystem. However, they have been shrinking in recent years due to natural and anthropogenic activities. Although many studies focus on tidal flats in the Yangtze River estuary (YRE) in China, how reclamation and plant invasion affect the expansion and erosion of tidal flats are still unclear. In this study, we analyzed all of the available Landsat TM/ETM+ /OLI imagery from the period 1996 to 2018 using the Google Earth Engine (GEE) cloud computing platform to obtain annual maps of coastal tidal flats of YRE at 30 m spatial resolution. We chose three sample tidal flats where severe Spartina alterniflora (S. alterniflora) invasion, reclamation, and control areas existed to explore the joint impact of plant invasion and reclamation on tidal flats. We also point out the main driving factor of tidal flat expansion of each island in YRE by multiple linear regression. Our results suggest that the tidal flats of YRE had obvious expansion from 1996 to 2018, and the speed of expansion is getting slower because of the decreasing deposits in the Yangtze River. Invasive S. alterniflora is effective at promoting silting, and tidal flats with S. alterniflora invasion expanded 2.54 times faster than the control group. Chongming and Hengsha Islands were mainly affected by sediment concentration, while Changxing and Jiuduansha Islands were affected by reclamation and S. alterniflora invasion, respectively. Cytochalasans have continuously aroused considerable attention among the chemistry and pharmacology communities due to their structural complexities and pharmacological significances. Sixteen structurally diverse chaetoglobosins, 10-(indol-3-yl)-[13]cytochalasans, including a new one, 6-O-methyl-chaetoglobosin Q (1), were isolated from the coral-associated fungus Chaetomium globosum C2F17. Their structures were accomplished by extensive spectroscopic analysis combined with single-crystal X-ray crystallography and ECD calculations. Meanwhile, the structures and absolute configurations of the previously reported compounds 6, 12, and 13 were confirmed by single-crystal X-ray analysis for the first time. Chaetoglobosins E (6) and Fex (11) showed significant cytotoxicity against a panel of cancer cell lines, K562, A549, Huh7, H1975, MCF-7, U937, BGC823, HL60, Hela, and MOLT-4, with the IC50 values ranging from 1.4 μM to 9.2 μM. We have previously reported that Trichoderma atroviride, an endophytic fungus isolated from S. miltiorrhiza, promotes S. miltiorrhiza hairy root growth and significantly stimulates the biosynthesis of tanshinones specifically the polysaccharide fraction (PSF). However, this study only focused exclusively on six metabolites whilst ignoring changes to the whole metabolite composition of the S. miltiorrhiza hairy roots. In the present study, the dynamic metabonomic responses of S. miltiorrhiza hairy roots were investigated using ultra-high-performance liquid chromatography-high resolution mass spectrometry (UHPLC-HRMSn). UHPLC-HRMS typical total ions chromatograms (TICs) of PSF-treated hairy root samples were different from the control. Moreover, the results of principal component analysis (PCA), partial least squares discriminant analysis (PLS-DA) and hierarchical clustering analysis (HCA) indicated that PSF-treated samples were significantly different from the control. Through the analysis of PLS-DA, a total of 114 and 99 differential metabolites were found from the positive and negative models respectively and a total of 33 differential metabolites were identified. Thus, S. miltiorrhiza hairy roots had been induced to regulate the metabolic profiling in response to PSF and the changes of the metabolic profiling contributed to promoting the biosynthesis of tanshinones notably whilst the biosynthesis of phenolic acids were slightly inhibited. The sensitive detection of Pb2+ is of significant importance for food safety, environmental monitoring, and human health care. To this end, a novel fluorescent biosensor, DNAzyme-functionalized R-phycoerythrin (DNAzyme-R-PE), was presented for Pb2+ analysis. The biosensor was prepared via the immobilization of Iowa Black® FQ-modified DNAzyme–substrate complex onto the surface of SPDP-functionalized R-PE. The biosensor produced a minimal fluorescence signal in the absence of Pb2+. However, Pb2+ recognition can induce the cleavage of substrate, resulting in a fluorescence restoration of R-PE. The fluorescence changes were used to measure sensitively Pb2+ and the limit of detection was 0.16 nM with a linear range from 0.5–75 nM. Furthermore, the proposed biosensor showed excellent selectivity towards Pb2+ even in the presence of other metal ions interferences and was demonstrated to successfully determine Pb2+ in spiked lake water samples.
