In the actual operation of urban rail transit (URT), the vibrations of steel-spring floating-slab tracks (SSFSTs) are amplified, and the track structure has strong low-frequency acoustic radiation; therefore, it is necessary to study the acoustic radiation of SSFSTs. In addition, multi-block short track structures are often laid within the URT lines; however, many researchers studying the reduction of vibration track service performance problems only select one or several block tracks to study. In reality, many short track structures will become sound sources when a train passes, and different sound sources will have various acoustic effects during the propagation process; therefore, it is necessary to study the rationality of any track acoustic model that analyzes the acoustic radiation problem. In order to more accurately predict the acoustic characteristics of steel-spring short floating-slab tracks (SSSFSTs) on a one-span bridge, train-track-bridge interaction theory and the acoustic boundary element method (BEM) were adopted to study the acoustical differences and mechanism of the float-slab number in the acoustic model. The results showed that with the increase in the floating-slab number in the acoustic model, the acoustic radiation ability of SSSFSTs and the sound pressure in the sound field increased; however, it was not a simple linear increase. Thus, the floating-slab number in the acoustic model not only affected the acoustic radiation ability but also caused acoustic effects during the propagation process, which affected the predicted results. The vibration characteristics of each floating-slab were different, and the acoustic input conditions of different numbers of floating-slabs in the acoustic model led to significant differences in the acoustic analysis. There was also obviously a different acoustic contribution of each floating-slab to the same sound field point, which led to the significant influence of the sound pressure at the sound field points when using different acoustic models. Therefore, using acoustic models with different floating-slab numbers had a significant effect on the acoustic analysis of SSSFSTs. In the context of noise reduction schemes for box-girder bridges (BGBs) used in high-speed railway, the thickened top plate design can effectively reduce the structural noise of the BGB, which has been widely recognized. However, it is difficult to obtain the optimum thickness of the top plate of the BGB without mastering the key mechanism of the noise reduction scheme. Therefore, this study took a 32 m simple-supported concrete BGB in the context of a high-speed railway as the research object and analyzed and compared the sound vibration characteristics of the entire thickened top plate versus the locally thickened top plate on BGB tracks, and the optimal noise reduction mechanism of the thickened top plate design scheme was studied in detail. The key issues of the thickened top plate noise reduction scheme are discussed. The results show that thickening the top plate can obviously reduce the bridge's structural noise when subjected to severe vibration and high frequency bands because the vibration of the BGB is reduced. However, in the low frequency band, acoustic radiation can occur as a result of the small amplitude vibration, and this phenomenon is closely related to the vibrational distribution of the BGB. Therefore, it is necessary to focus on the vibrational distribution of the BGB as a priority when carrying out noise reduction using a thickened top plate. This paper points out the most significant factors affecting the acoustic radiation ability of the BGB in different frequency bands, especially the key problem of the strong acoustic radiation ability caused by small vibrations in low frequency band. Good characterization of traffic interactions among urban roads can facilitate traffic-related applications, such as traffic control and short-term forecasting. Most studies measure the traffic interaction between two roads by their topological distance or the correlation between their traffic variables. However, the distance-based methods neglect the spatial heterogeneity of roads' traffic interactions, while the correlation-based methods cannot capture the non-linear dependency between two roads' traffic variables. In this paper, we propose a novel approach called Road2Vec to quantify the implicit traffic interactions among roads based on large-scale taxi operating route data using a Word2Vec model from the natural language processing (NLP) field. First, the analogy between transportation elements (i.e., road segment, travel route) and NLP terms (i.e., word, document) is established. Second, the real-valued vectors for road segments are trained from massive travel routes using the Word2Vec model. Third, the traffic interaction between any pair of roads is measured by the cosine similarity of their vectors. A case study on short-term traffic forecasting is conducted with artificial neural network (ANN) and support vector machine (SVM) algorithms to validate the advantages of the presented method. The results show that the forecasting achieves a higher accuracy with the support of the Road2Vec method than with the topological distance and traffic correlation based methods. We argue that the Road2Vec method can be effectively utilized for quantifying complex traffic interactions among roads and capturing underlying heterogeneous and non-linear properties. The middle and lower reaches of the Yangtze River basin (MLRYB) are prone to flooding because their orientation is parallel to the East Asian summer monsoon rain belt. Since the East Asian summer monsoon presents pronounced intraseasonal variability, the subseasonal prediction of summer precipitation anomalies in the MLRYB region is an imperative demand nationwide. Based on rotated empirical orthogonal function analysis, 48 stations over the MLRYB with coherent intraseasonal (10-80-day) rainfall variability are identified. Power spectrum analysis of the MLRYB rainfall index, defined as the 48-station-averaged intraseasonal rainfall anomaly, presents two dominant modes with periods of 20-30 days and 40-60 days, respectively. Therefore, the intraseasonal (10-80-day) rainfall variability is divided into 10-30-day and 30-80-day components, and their predictability sources are detected separately. Spatial-temporal projection models (STPM) are then conducted using these predictability sources. The forecast skill during the period 2003-2010 indicates that the STPM is able to capture the 30-80-day rainfall anomalies 5-30 days in advance, but unable to reproduce the 10-30-day rainfall anomalies over MLRYB. The year-to-year fluctuation in forecast skill might be related to the tropical Pacific sea surface temperature anomalies. High forecasting skill tends to appear after a strong El Niño or strong La Niña when the summer seasonal mean rainfall over the MLRYB is enhanced, whereas low skill is apparent after neutral conditions or a weak La Niña when the MLRYB summer seasonal mean rainfall is weakened. Given the feasibility of STPM, the application of this technique is recommended in the real-time operational forecasting of MLRYB rainfall anomalies during the summer flooding season. A strain-type three-component table dynamometer is presented in this paper, which reduces output errors produced by cutting forces imposed on the different milling positions of a workpiece. A sensor structure with eight parallel elastic beams is proposed, and sensitive regions and Wheastone measuring circuits are also designed in consideration of eliminating the influences of the eccentric forces. To evaluate the sensor decoupling performance, both of the static calibration and dynamic milling test were implemented in different positions of the workpiece. Static experiment results indicate that the maximal deviation between the measured forces and the standard inputs is 4.58%. Milling tests demonstrate that with same machining parameters, the differences of the measured forces between different milling positions derived by the developed sensor are no larger than 6.29%. In addition, the natural frequencies of the dynamometer are kept higher than 2585.5 Hz. All the measuring results show that as a strain-type dynamometer, the developed force sensor has an improved eccentric decoupling accuracy with natural frequencies not much decreased, which owns application potential in milling process monitoring. Pancreatic cancer (PC) is the most lethal type of gastrointestinal cancer, and early detection and monitoring is an urgent problem. Circulating tumor cells (CTCs) are emerging as a non-invasive biomarker for tumor detection. However, the low sensitivity is a main problem in the traditional CellSearch System for detecting CTCs, especially in patients with PC. In this study, we used negative enrichment (NE), immunofluorescence and in situ hybridization (FISH) of chromosome 8 (NE-iFISH) to capture and identify CTCs in PC patients. We showed that the NE-iFISH system exhibited a dramatically high detection rate of CTCs in PC patients (90%). The diagnostic rate of PC reached 97.5% when combining CTCs ≥ 2 and carbohydrate antigen 19-9 (CA19-9) > 37 µmol/L. The 1-year survival in the group of CTCs < 3 was significantly higher than that of CTCs ≥ 3 (p = 0.043). In addition, we analyzed the role of chromosomal instability in CTCs detection. The group of triploid (three hybridization signals of chromosome 8) CTCs ≥ 3 showed a shorter 1-year survival (p = 0.0279) and overall survival (p = 0.0188) than the group with triploid CTCs < 3. Importantly, the triploid CTC number but not the overall CTC counts could be a predictor of chemo-sensitivity. Moreover, circulating tumor microembolus (CTMs) were found in stage IV patients, and were positively related to the poor response to chemotherapy. In conclusion, the NE-iFISH system significantly improved the positive detection rate of CTCs and triploid CTC could be used to predict prognosis or the response to the chemotherapy of PC patients. Aspergillus fumigatus AFPAB1 is the ortholog of the Aspergillus oryzae cytoplasmic messenger ribonucleoprotein granules AOPAB1 that function to depress the initiation of translation during stress. A. fumigatus can regulate its cellular physiology in response to environmental stresses, but there has been no research on Pab1 in A. fumigatus. The associated gene afpab1 was replaced with a hygromycin-selectable marker to generate the strain Δafpab1. Phenotypic analysis showed that the Δafpab1 grew more weakly than the wild-type strain. Also the germination rate of Δafpab1 was decreased when exposed to oxidative stress. The morphology of Δafpab1 spores also showed great changes. The killing rate of Δafpab1 by RAW264.7 murine macrophage cells was increased, and the reactive oxygen species (ROS) scavenging ability of Δafpab1 was decreased. Pathogenicity testing showed that the deletion strain had decreased virulence. Therefore, we conclude that afpab1 activity is correlated with susceptibility to oxidative stress, and deletion of afpab1 from A. fumigatus possibly leads to observed hypovirulence in an immunosuppressed mouse model. The ability to identify hazardous traffic events is already considered as one of the most effective solutions for reducing the occurrence of crashes. Only certain particular hazardous traffic events have been studied in previous studies, which were mainly based on dedicated video stream data and GPS data. The objective of this study is twofold: (1) the Markov blanket (MB) algorithm is employed to extract the main factors associated with hazardous traffic events; (2) a model is developed to identify hazardous traffic event using driving characteristics, vehicle trajectory, and vehicle position data. Twenty-two licensed drivers were recruited to carry out a natural driving experiment inWuhan, China, and multi-sensor information data were collected for different types of traffic events. The results indicated that a vehicle’s speed, the standard deviation of speed, the standard deviation of skin conductance, the standard deviation of brake pressure, turn signal, the acceleration of steering, the standard deviation of acceleration, and the acceleration in Z (G) have significant influences on hazardous traffic events. The sequential minimal optimization (SMO) algorithm was adopted to build the identification model, and the accuracy of prediction was higher than 86%. Moreover, compared with other detection algorithms, the MB-SMO algorithm was ranked best in terms of the prediction accuracy. Mango seed kernel extract (MSKE) and its key components (gallic acid, GA; methyl gallate, MG; and pentagalloyl glucopyranose, PGG) have generated interest because of their pharmacological activities. To develop the potential use of the key components in MSKE as natural therapeutic agents, their pharmacokinetic data are necessary. Therefore, this study was performed to evaluate the factors affecting their oral bioavailability as pure compounds and as components in MSKE. The in vitro chemical stability, biological stability, and absorption were evaluated in Hanks' Balanced Salt Solution, Caco-2 cell and rat fecal lysates, and the Caco-2 cell model, respectively. The in vivo oral pharmacokinetic behavior was elucidated in Sprague-Dawley rats. The key components were unstable under alkaline conditions and in Caco-2 cell lysates or rat fecal lysates. The absorptive permeability coefficient followed the order MG > GA > PGG. The in vivo results exhibited similar pharmacokinetic trends to the in vitro studies. Additionally, the co-components in MSKE may affect the pharmacokinetic behaviors of the key components in MSKE. In conclusion, chemical degradation under alkaline conditions, biological degradation by intestinal cell and colonic microflora enzymes, and low absorptive permeability could be important factors underlying the oral bioavailability of these polyphenols. Aims: The aim of the present study is to investigate the differential expression of CD44, CD47 and c-met in ovarian clear cell carcinoma (OCCC), the correlation in their expression and their relationship with the biological behavior of OCCC. Methods: We used immunohistochemistry to examine the expression of CD44, CD47 and c-met in OCCC (86 cases) and investigated the effects of the expression and interaction of these molecules on the development of OCCC. Results: CD44, CD47 and c-met expression was significantly high in OCCC. Expression of CD44 and CD47 correlated with patient surgical stage, chemotherapy resistance and prognosis (all p < 0.05), and expression of c-met correlated with chemotherapy resistance and prognosis (all p < 0.05), but did not correlate with lymph node metastasis (all p > 0.05). The surgical stage, CD44, CD47 and c-met expression were independent risk factors for OCCC prognosis (all p < 0.05). Patients with low levels of CD44, CD47 and c-met showed better survival than those with high levels (all p < 0.05). There was a positive correlation between CD44 (or CD47) and c-met, as well as between CD44 and CD47 (the Spearman correlation coefficient rs was 0.783, 0.776 and 0.835, respectively, all p < 0.01). Additionally, pairwise correlation analysis of these three markers shows that the high expression of CD44/CD47, CD44/c-met and CD47/c-met were correlated with patient surgical stage, chemotherapy resistance and prognosis (all p < 0.05), but did not correlate with lymph node metastasis (all p > 0.05). Conclusions: Expression of CD44, CD47 and c-met was upregulated in OCCC and pairwise correlation. Nanostructured noble metal-semiconductor materials have been attracting increasing attention because of their broad application in the field of environmental remediation, sensing and photocatalysis. In this study, a facile approach for fabricating silver@mesoporousanataseTiO2 (Ag@mTiO2) core-shell nanoparticles employing sol-gel and hydrothermal reaction is demonstrated. The Ag@mTiO2nanoparticles display excellent surface-enhanced Raman scattering (SERS) sensitivity and they can detect the methylene blue (MB) molecules with the concentration of as low as 10−8 M. They also exhibit outstanding photocatalytic activity compared with mTiO2, due to the efficient separation and recombination restrain of electron–hole pairs under ultraviolet light. The Ag@mTiO2nanoparticles also present good stability and they can achieve recyclable photocatalytic degradation experiments for five times without loss of activity. Subsequently, the nanoparticles with dual functions were successfully used to in situ monitor the photodegradation process of MB aqueous solution. These results, demonstrating the multifunctional Ag@mTiO2 nanoparticles, hold promising applications for simultaneous SERS analysis and the removal of dye pollutants in environmental field. Objective: To measure Lewis y and integrin α5β1 expression in epithelial ovarian carcinoma and to correlate the levels of these molecules with ovarian carcinoma chemotherapy and prognosis. Methods: The study population included 34 ovarian carcinoma patients with chemotherapeutic drug-resistance, six partially drug-sensitive cases, and 52 drug-sensitive cases (92 total). Immunochemistry was used to determine expression of Lewis y antigen and integrin α5β1 in ovarian carcinoma tissues, and correlation of these molecules with chemotherapy resistance was further investigated, Multi-factor logistic regression analysis was applied to investigate: age, surgical stage, grade, subtype of patient cases, metastasis of lymph nodes, residual tumor size, expression levels of Lewis y antigen and integrin α5β1 correlation with ovarian carcinoma chemotherapy resistance. Results: The expression rates of Lewis y antigen and integrins α5 and β1 were significantly greater in the drug-resistant group (91.17%, 85.29%, 88.24%) than the partially sensitive (50.00%, 33.33%, 50.00%) or sensitive groups (61.54%, 57.69%, 55.77%). Binary logistic regression analysis revealed that surgical stage, residual tumor size, and expression of integrin α5 and Lewis y in ovarian carcinoma tissues were independent risk factors for chemotherapeutic drug resistance. Conclusions: Overexpression of Lewis y and integrin α5 are strong risk factors for chemotherapeutic drug resistance in ovarian carcinoma patients. [ABSTRACT FROM AUTHOR]

The biosynthesis of nanoparticles has received increasing attention due to the growing need to develop safe, cost-effective and environmentally friendly technologies for nano-materials synthesis. In this report, silver nanoparticles (AgNPs) were synthesized using a reduction of aqueous Ag+ ion with the culture supernatants of Aspergillus terreus. The reaction occurred at ambient temperature and in a few hours. The bioreduction of AgNPs was monitored by ultraviolet-visible spectroscopy, and the AgNPs obtained were characterized by transmission electron microscopy and X-ray diffraction. The synthesized AgNPs were polydispersed spherical particles ranging in size from 1 to 20 nm and stabilized in the solution. Reduced nicotinamide adenine dinucleotide (NADH) was found to be an important reducing agent for the biosynthesis, and the formation of AgNPs might be an enzyme-mediated extracellular reaction process. Furthermore, the antimicrobial potential of AgNPs was systematically evaluated. The synthesized AgNPs could efficiently inhibit various pathogenic organisms, including bacteria and fungi. Lewis (y) antigen is a difucosylated oligosaccharide present on the plasma membrane, and its overexpression is frequently found in human cancers and has been shown to be associated with poor prognosis. Our previous studies have shown that Lewis (y) antigen plays a positive role in the process of invasion and metastasis of ovarian cancer cells. However, the mechanisms by which Lewis (y) antigen enhances the invasion and tumor metastasis are still unknown. In this study, we established a stable cell line constitutively expressing Lewis (y) antigen (RMG-1-hFUT) by transfecting the cDNA encoding part of the human α1,2-fucosyltransferase (α1,2-FUT) gene into the ovarian cancer cell line RMG-1, and investigated whether Lewis (y) antigen regulates the expression of matrix metalloproteinase-2 (MMP-2) and MMP-9, and tissue inhibitors of metalloproteinases (TIMP-1) and TIMP-2. We found that RMG-1-hFUT cells exhibited higher invasive capacities than their control cells. In addition, expression of TIMP-1 and TIMP-2 was down-regulated and expression of MMP-2 and MMP-9 was up-regulated. Anti-Lewis (y) antigen antibody treatment significantly reversed the expression of TIMP-1, TIMP-2, MMP-2 and MMP-9. Taken together, we provide the first evidence that down-regulation of TIMP-1 and TIMP-2 and up-regulation of MMP-2 and MMP-9 represents one of the mechanisms by which Lewis (y) antigen promotes cell invasion. The non-cooperative or sequential events which occur during protein thermal denaturation are closely correlated with protein folding, stability, and physiological functions. In this research, the sequential events of human brain-type creatine kinase (hBBCK) thermal denaturation were studied by differential scanning calorimetry (DSC), CD, and intrinsic fluorescence spectroscopy. DSC experiments revealed that the thermal denaturation of hBBCK was calorimetrically irreversible. The existence of several endothermic peaks suggested that the denaturation involved stepwise conformational changes, which were further verified by the discrepancy in the transition curves obtained from various spectroscopic probes. During heating, the disruption of the active site structure occurred prior to the secondary and tertiary structural changes. The thermal unfolding and aggregation of hBBCK was found to occur through sequential events. This is quite different from that of muscle-type CK (MMCK). The results herein suggest that BBCK and MMCK undergo quite dissimilar thermal unfolding pathways, although they are highly conserved in the primary and tertiary structures. A minor difference in structure might endow the isoenzymes dissimilar local stabilities in structure, which further contribute to isoenzyme-specific thermal stabilities. [ABSTRACT FROM AUTHOR]

The objective of this study was to investigate the inhibitory effect of chlorogenin 3-O-β-chacotrioside derivatives against H5N1 subtype of the highly pathogenic avian influenza (HPAI) viruses and its molecular mechanism. A series of novel small molecule pentacyclic triterpene derivatives were designed and synthesized and their antiviral activities on HPAI H5N1 viruses were detected. The results displayed that the derivatives UA-Nu-ph-5, XC-27-1 and XC-27-2 strongly inhibited wild-type A/Duck/Guangdong/212/2004 H5N1 viruses with the IC50 values of 15.59 ± 2.4 μM, 16.83 ± 1.45 μM, and 12.45 ± 2.27 μM, respectively, and had the selectivity index (SI) > 3, which was consistent with the efficacy against A/Thailand/kan353/2004 pseudo-typed viruses. Four dealt patterns were compared via PRNT. The prevention dealt pattern showed the strongest inhibitory effects than other patterns, suggesting that these derivatives act on the entry process at the early stages of H5N1 viral infection, providing protection for cells against infection. Further studies through hemagglutinin inhibition (HI) and neuraminidase inhibitory (NAI) assay confirmed that these derivatives inhibited H5N1 virus replication by interfering with the viral hemagglutinin function. The derivatives could recognize specifically HA protein with binding affinity constant KD values of 2.57 × 10−4 M and 3.67 × 10−4 M. In addition, through site-directed mutagenesis combined with a pseudovirion system, we identified that the high-affinity docking sites underlying interaction were closely associated with amino acid residues I391 and T395 of HA. However, the potential binding sites of the derivatives with HA did not locate at HA1 sialic acids receptor binding domain (RBD). Taken together, these study data manifested that chlorogenin 3-O-β-chacotrioside derivatives generated antiviral effect against HPAI H5N1 viruses by targeting the hemagglutinin fusion machinery. The incidence of Aspergillus fumigatus infections has become more frequent as a consequence of widespread immunosuppression. At present, the number of available antifungal agents in the clinic is limited, and most of them, such as itraconazole (ICZ), are toxic and show resistance. Berberine (BER) is a plant alkaloid used in the clinic mainly for alimentary infections. We have used BER and ICZ to measure in vitro resistance in A. fumigatus isolated from clinical patients. The minimum inhibitory concentration ranges of BER and ICZ were 4-256 and 0.031-0.250 μg/mL, respectively. In addition, against A. fumigatus IFM 40808 strain, the MIC₅₀ values of BER and ICZ were 8 and 0.125 μg/mL. Using this strain, we compared the giant colonies with or without BER, and concluded that BER could restrain A. fumigatus mycelial growth and conidial pigment production. Combinations of the two drugs were also tested by the checkerboard assay to identify any functional interactions between them. Thirty-two out of 42 isolates had FICI values > 4.0, indicating that two drugs were mutually antagonistic. In conclusion, it is not advised that BER and ICZ be used in the clinic at the same time. Our results indicated that BER may inhibit A. fumigatus through the ergosterol biosynthesis pathway, like ICZ.
