Staphylococcus pseudintermedius (S. pseudintermedius) is the main pathogen causing pyoderma of canines. With the emergence of drug-resistant bacteria, traditional antibiotic treatments are limited. As a potential antibacterial agent, NZ2114 was effective against S. pseudintermedius, including drug-resistant strains. Its bactericidal efficacy was superior to mupiroxacin, ofloxacin and lincomycin. To facilitate the transcutaneous delivery of NZ2114 for the treatment of superficial pyoderma, chemical permeation enhancers were added since water-soluble NZ2114 does not easily penetrate the skin lipid layer. Two different NZ2114 sprays were prepared by combining 1% Azone + 10% propylene glycol (PG) or 5% N-methylpyrrolidone (NMP) + 10% PG with NZ2114 after screening. The cumulative permeability of NZ2114 sprays were 244.149 and 405.245 μg/cm2 at 24 h with an in vitro percutaneous assay of mice skin, which showed a 244% and 405% increase in skin permeability than NZ2114, respectively. In addition, the efficacy of NZ2114 sprays in reducing skin bacteria colonisation was demonstrated in a mouse model of superficial pyoderma (24 mice, 3 mice/group) induced by S. pseudintermedius, and the 5% NMP + 10% PG + NZ2114 group had the best therapeutic effect compared to the other groups. This study aimed to isolate the proteolytic fraction from the silkworm thorn fruit (Cudrania tricuspidata) through ethanol precipitation at different ratios, and to determine its proteolytic activity and optimal activity conditions. Furthermore, the hydrolysis characteristics and antioxidant activity of soy protein isolate (SPI) and whey protein concentrate (WPC) hydrolyzates obtained through the enzymatic hydrolysis of freeze-dried silkworm thorn fruit powder (SF) were evaluated. For isolation and partial purification of proteolytic fraction, the water-solubilized fraction of the silkworm thorn fruit was purified through ethanol precipitation at four different ratios of 1:1, 1:2, 1:4, and 1:6 (v/v). The protein recovery rate, caseinolytic activity, protein pattern, and optimal activity (pH, temperature, and inhibitors) of fractional ethanol precipitate obtained from the silkworm thorn fruit (ESF) were evaluated. The proteolytic fraction obtained from silkworm thorn fruit exhibited a major protein band around 65–70 kDa and showed the highest proteolytic activity at a 1:4 ratio of ethanol precipitation (p < 0.05). The optimal activity of the measured enzyme fraction was determined to be at pH 9.0 and 50 °C, and the proteolytic activity of ESF was almost inhibited by phenyl methyl sulphonyl fluoride (PMSF, 2 mM), a serine protease inhibitor. Compared to Alcalase and papain, extensively used as commercial enzymes, the silkworm thorn fruit powder was less effective in hydrolyzing SPI and WPC. Nevertheless, SPI and WPC hydrolyzates mediated with silkworm thorn fruit powder showed even better antioxidant activities than those mediated with Alcalase and papain. This study investigated the interdecadal characteristics of the frequency of the winter single station extreme cold events (SSECEs) in North China and their relationship with sea surface temperature (SST). The results showed the following: (a) The SSECEs occurred frequently before 1991, but less thereafter, with an increase after 2018. The first two interdecadal modes of the SSECE frequency were east–west inverse and "n" patterns. (b) The interdecadal abrupt change of the "n" pattern occurred around 1997/1998. Before 1997/1998, the synergistic effects between the positive Interdecadal Pacific Oscillation (+IPO) and the negative North Atlantic Multidecadal Oscillation (−AMO) triggered the "two troughs and one ridge" anomalous circulation in Eurasia. The Rossby wave energy propagated downstream from the Atlantic, strengthening the Lake Baikal ridge. Furthermore, the Siberian High (SH) became weaker in the north and stronger in the south. With the favorable jet conditions, the cold air invaded North China along the northerly airflow in front of the Lake Baikal ridge, resulting in the frequent SSECE occurrence in central North China. Afterwards, the opposite occurred. (c) The cooperation of SST anomalies (SSTAs) led to the east–west inverse anomaly of the SSECE frequency. Before 1991, the high SSTAs in the central North Atlantic and low SSTAs in the equatorial Indian Ocean and the southwest Pacific triggered "+", "−", "+", and "−" wave trains at mid-latitudes from the Atlantic to the North Pacific. The Rossby wave energy propagated eastward from the Atlantic, resulting in the SH and Urals ridge strengthening, and the Aleutian Low and East Asian trough deepening. The northwestern airflow in front of the Urals ridge guided the cold air into North China, leading to frequent SSECEs in central and eastern North China before 1991. In this paper, we investigated the vertical distribution characteristics of surface soil moisture based on ISMN (International Soil Moisture Network) multilayer in situ data (5, 10, and 20 cm; 2, 4, and 8 in) and performed comparisons between the in situ data and four microwave satellite remote sensing products (SMOS L2, SMOS-IC, SMAP L2, and SMAP L4). The results showed that the mean soil moisture difference between layers can be −0.042~−0.024 (for the centimeter group)/−0.067~−0.044 (for the inch group) m3/m3 in negative terms and 0.020~0.028 (for the centimeter group)/0.036~0.040 (for the inch group) m3/m3 in positive terms. The surface soil moisture was found to have very significant stratification characteristics, and the interlayer difference was close to or beyond the SMOS and SMAP 0.04 m3/m3 nominal retrieval accuracy. Comparisons revealed that the satellite retrievals had a higher correlation with the field measurements of 5 cm/2 in, and SMAP L4 had the smallest difference with the in situ data. The mean difference caused by using 10 cm/4 in and 20 cm/8 in in situ data instead of the 5 cm/2 in data could be about −0.019~−0.018/−0.18~−0.015 m3/m3 and −0.026~−0.023/−0.043~−0.039 m3/m3, respectively, meaning that there would be a potential depth mismatch in the data validation. Here, we report a tunable electrochemical benzylic C−H functionalization of (thio)xanthenes with terminal alkynes and nitriles in the absence of any catalyst or external chemical oxidant. In this study, Precipitation Estimation from Remotely Sensed Information Using Artificial Neural Networks–Climate Data Record (PERSIANN-CDR) was evaluated for the assessment of long-term drought monitoring in Huaihe River Basin using daily gauge observation data for the period from 1983 to 2017. The evaluation results show that the PERSIANN-CDR algorithm has a good detection ability for small precipitation events over the whole basin, but a poor ability for extreme precipitation events (>50 mm/day). Daily PERSIANN-CDR estimates perform relatively better in areas with abundant precipitation, while the monthly and yearly PERSIANN-CDR estimates are highly consistent with gauge observations both in magnitude and space. The Standardized Precipitation Index (SPI) at various time scales (3, 6, and 12 months) was calculated based on PERSIANN-CDR and gauge observation, respectively. Grid-based values of statistics derived from those SPI values demonstrate that PERSIANN-CDR has a good ability to capture drought events of each time scale across the basin. However, caution should be applied when using PERSIANN-CDR estimates for basin-scale drought trend analysis. Furthermore, three drought events with long duration and large extent were selected to test the applicability of PERSIANN-CDR in drought monitoring. The results show that it has a good ability to capture when and where droughts occur and how far they spread. Due to the overestimation of small precipitation events, PERSIANN-CDR tends to overestimate the number of extreme droughts and their extents. The main osmoadaptive mechanisms of extremely halophilic archaea include the "salt-in" strategy and the "compatible solutes" strategy. Here we report the osmoadaptive mechanism of an extremely halophilic archaea H. kocurii 2020YC7, isolated from a high salt environment sample. Genomic data revealed that strain 2020YC7 harbors genes trkA, trkH, kch for K+ uptake, kefB for K+ output, treS for trehalose production from polysaccharide, and betaine/carnitine/choline transporter family gene for glycine betaine uptake. Strain 2020YC7 could accumulate 8.17 to 28.67 μmol/mg protein K+ in a defined medium, with its content increasing along with the increasing salinity from 100 to 200 g/L. When exogenous glycine betaine was added, glycine betaine functioned as the primary osmotic solute between 200 and 250 g/L NaCl, which was accumulated up to 15.27 mg/mg protein in 2020YC7 cells. RT-qPCR results completely confirmed these results. Notably, the concentrations of intracellular trehalose decreased from 5.26 to 2.61 mg/mg protein as the NaCl increased from 50 to 250 g/L. In combination with this result, the transcript level of gene treS, which catalyzes the production of trehalose from polysaccharide, was significantly up-regulated at 50–100 g/L NaCl. Therefore, trehalose does not act as an osmotic solute at high NaCl concentrations (more than 100 g/L) but at relatively low NaCl concentrations (50–100 g/L). And we propose that the degradation of cell wall polysaccharide, as a source of trehalose in a low-salt environment, may be one of the reasons for the obligate halophilic characteristics of strain 2020YC7. Wind shear has important implications for Kelvin–Helmholtz instability (KHI) and gravity waves (GWs) in the mesosphere–lower thermosphere (MLT) region where its momentum transport process is dominated by short-period (<1 h) GWs. However, the sub-hourly variation in wind shear is still not well quantified. This study aims to improve current understanding of vertical wind shear by analyzing multi-year meteor radar measurements at the Mohe (MH, 53.5°N, 122.3°E), Beijing (BJ, 40.3 ° N , 116.2 ° E ), Wuhan (WH, 30.5 ° N , 114.6 ° E ), and Fuke (FK, 19.5°N, 109.1°E) stations in China. The wind field is estimated by a new algorithm, e.g., the damped least squares fitting. Taking the wind shear estimated by normal products as a criterion, the shear produced by the new algorithm has more statistical convergence as compared to the traditional algorithm, e.g., the least squares fitting. Therefore, we argue that the 10 min DLSA wind probably produces a more reasonable vertical shear. Both intensive wind shears and GW kinetic energy can be simultaneously captured during the 0600–1600 UTs of May at MH and during the 1300–2400 UTs of March at FK, possibly implying that the up-propagation of GWs could contribute to the production of large wind shears. This study aims to investigate the features of the ionospheric mid-latitude trough over North America by using the MIT total electron content data obtained during three geomagnetic storms that occurred in August 2018, September 2017, and March 2015. The mid-latitude trough position sharply moves equatorward from the quiet-time subauroral latitude to mid-latitude with the decrease in SYM-H during geomagnetic storms. We find that the ionospheric behavior of TEC around the mid-latitude trough position displays three kinds of ionospheric storm effect: negative ionospheric storm effect, unchanged ionospheric behavior, and positive ionospheric storm effect. These ionospheric storm effects around the mid-latitude trough position are not always produced by the mid-latitude trough. The ionospheric storm effects produced by the mid-latitude trough are limited in the narrow mid-latitude trough regions, and are transmitted to other regions with the movement of the mid-latitude trough. CO2 is a major contributor to global warming, and considerable efforts have been undertaken to capture and utilise it. Herein, a nanomaterial based on ionic liquid (IL)–modified calcined magnesites was investigated for CO2 capture. The synthesised nanomaterial (magnesite modified using [APMIM]Br) exhibited the best adsorption performance of 1.34 mmol/g at 30% IL loading amount, 50 °C, 0.4 MPa and 150 mL/min. In particular, the obtained nanomaterial could be regenerated at a low temperature of 90 °C for 3 h, and its CO2 adsorption capacity of 0.81 mmol/g was retained after eight cycles. FT-IR results showed that the imidazole ring and C–N group are directly related to CO2 adsorption capacity. Moreover, improving the conjugative effect of the imidazole ring enhanced the adsorption performance. Further, CO2 was adsorbed on the adsorbent surface and incomplete desorption decreased the BET surface area and CO2 adsorption capacity. Additionally, four models were selected to fit the adsorption kinetics. The results show that the adsorption mechanism fits the pseudo-first-order model well. [ABSTRACT FROM AUTHOR]

Flue gas produced by biomass fuel combustion contains various chlorine-containing substances and is an important factor causing biomass boiler corrosion. The corrosion processes of chlorine, hydrogen chloride and water on iron covered with an intact/damaged oxide film were investigated under the high temperature of 1300 K through reactive molecular dynamics simulation. The results show that the diffusion processes of oxygen and chlorine are similar and can be divided into three stages: rapid diffusion, continuous diffusion, and no oxide film (stable). Oxygen diffusion in Fe2O3 into a pure iron layer is the main cause of gas corrosion in iron/iron oxide systems. A complete oxide film can hinder iron corrosion by chlorine and hydrogen chloride. Damage in an oxide film significantly affects oxygen and chlorine diffusion and iron corrosion. However, such influence is gradually reduced. The integrity of a protective film is the key to alleviating corrosion. Water facilitates the dissociation of chlorine and hydrogen chloride, and it reacts with iron at high temperatures to enhance corrosion. This study improves the understanding of the iron oxide/iron corrosion from chlorine-containing gases from a microscopic perspective and is of great significance to metal corrosion protection and biomass combustion technologies. Referring to the structural information of the "hit" compound A from the reported pharmacophore-based virtual screening, a series of novel thienylpyridyl- and thioether/sulfoxide/sulfone-containing acetamide derivatives have been designed and synthesized. The structures of new compounds were confirmed by 1H NMR, 13C NMR and HRMS. The single-crystal structure of A was firstly reported. All the new synthesized compounds were evaluated for insecticidal activities on Mythimna separata Walker and Plutella xylostella L. Through a step-by-step structural optimization, the high insecticidal agents, especially towards Plutella xylostella L., have been found, and thienylpyridyl- and sulfone/thioether-containing acetamides Iq, Io, Ib and A, which are comparable with the control insecticides cartap, triflumuron and chlorantraniliprole in the present study, can be used as novel lead structures for new insecticides innovation research. In addition, some of the compounds, e.g., A, Ih, Id, Io and Iq, also exhibited favourable fungicidal activities against Physalospora piricola, Rhizoctonia cerealis and Sclerotinia sclerotiorum and would provide useful guidance for the design and development of new fungicides. Cyanate ester resin (CER) is an excellent thermal stable polymer. However, its mechanical properties are not appropriate for its application, with brittle weakness, and it has poor functional properties, such as luminescence. This work innovatively combines the luminescence property and the improved mechanical properties with the inherent thermal property of cyanate ester. A novel nanocomposite, CER/uMWCNTs/Eu, with multi-functional properties, has been prepared. The results show that with the addition of 0.1 wt.% of uMWCNTs to the resin, the flexural strength and tensile strength increased 59.3% and 49.3%, respectively. As the curing process of the CER progresses, the injected luminescence signal becomes luminescence behind the visible (FBV). The luminescence intensity of CER/uMWCNTs/Eu was much stronger than that of CER/MWCNTs/Eu, and the luminescence lifetime of CER/MWCNTs/Eu and CER/uMWCNTs/Eu was 8.61 μs and 186.39 μs, respectively. FBV exhibited great potential in the embedment of photon quantum information. Therefore, it can be predicted that CER/uMWCNTs/Eu composites will not only have a wide range of applications in sensing, detection, and other aspects, but will also exhibit great potential in the embedding of photon quantum information. Understanding the stoichiometry of extracellular enzymes in soil, particularly in relation to nutrient acquisition (e.g., carbon, nitrogen, phosphorus), provides valuable insights into microorganisms' resource requirements. This study investigates the metabolic constraints of soil microorganisms in response to different growth stages of apple trees under various soil management practices. A 14-year long-term experiment with a split-plot design was conducted, where the main plots received different cover crop treatments (bare vs. cover crop), and subplots were subjected to four fertilizer treatments (CK, M, NPK, MNPK). The significant main and interactive effects of cover crops, fertilizer treatment, and growth period on soil nutrients were observed (p < 0.001). Both cover crop and fertilizer treatments significantly increased the soil organic matter content, with implications for orchard resilience to drought. However, the cover factor alone did not notably influence soil carbon–nitrogen ratios or microbial communities. Microbial carbon limitations were driven by soil water dynamics and microbial biomass, while microbial phosphorus limitations were closely linked to total nitrogen levels. The results underscore the combination of cover crops and MNPK fertilizer-enhanced soil nutrient levels and enzyme activities, mitigating microbial carbon and phosphorus limitations. Simultaneous measurements of atmospheric volatile organic compounds (VOCs), conventional gases and meteorological parameters were performed at an urban site in Jinan, East China, in June 2021 to explore the formation and evolution mechanisms of summertime ozone (O3) pollution events. O3 Episode Ⅰ, O3 Episode II, and non-O3 episodes were identified based on the China Ambient Air Quality Standards and the differences in precursor concentrations. The O3 concentrations in Episode I and Episode II were 145.4 μg/m3 and 166.4 μg/m3, respectively, which were significantly higher than that in non-O3 episode (90 μg/m3). For O3 precursors, VOCs and NOx concentrations increased by 48% and 34% in Episode I, and decreased by 21% and 27% in Episode II compared to non-O3 episode days. The analysis of the m,p-xylene to ethylbenzene ratio (X/E) and OH exposure demonstrated that the aging of the air masses in Episode II was significantly higher than the other two episodes, and the differences could not be explained by localized photochemical consumption. Therefore, we speculate that the high O3 concentrations in Episode II were driven by the regional transport of O3 and its precursors. Backward trajectory simulations indicated that the air masses during Episode II were concentrated from the south. In contrast, the combination of high precursor concentrations and favorable meteorological conditions (high temperatures and low humidity) led to an excess of O3 in Episode I. Positive matrix factorization (PMF) model results indicated that increased emissions from combustion and gasoline vehicle exhausts contributed to the elevated concentrations of VOCs in Episode I, and solvent usage may be an important contributor to O3 formation. The results of this study emphasize the importance of strengthening regional joint control of O3 and its precursors with neighboring cities, especially in the south, which is crucial for Jinan to mitigate O3 pollution. Psoriasis is a chronic inflammatory skin disease characterized by epidermal hyperproliferation, aberrant differentiation of keratinocytes, and dysregulated immune responses. WW domain-containing oxidoreductase (WWOX) is a non-classical tumor suppressor gene that regulates multiple cellular processes, including proliferation, apoptosis, and migration. This study aimed to explore the possible role of WWOX in the pathogenesis of psoriasis. Immunohistochemical analysis showed that the expression of WWOX was increased in epidermal keratinocytes of both human psoriatic lesions and imiquimod-induced mice psoriatic model. Immortalized human epidermal keratinocytes were transduced with a recombinant adenovirus expressing microRNA specific for WWOX to downregulate its expression. Inflammatory responses were detected using Western blotting, real-time quantitative reverse transcription polymerase chain reaction (PCR), and enzyme-linked immunosorbent assay. In human epidermal keratinocytes, WWOX knockdown reduced nuclear factor-kappa B signaling and levels of proinflammatory cytokines induced by polyinosinic: polycytidylic acid [(poly(I:C)] in vitro. Furthermore, calcium chelator and protein kinase C (PKC) inhibitors significantly reduced poly(I:C)-induced inflammatory reactions. WWOX plays a role in the inflammatory reaction of epidermal keratinocytes by regulating calcium and PKC signaling. Targeting WWOX could be a novel therapeutic approach for psoriasis in the future. Staphylococcus hyicus is recognized as a leading pathogen of exudative epidermitis in modern swine industry. Antimicrobial peptides are attractive candidates for development as potential therapeutics to combat the serious threats of the resistance of S. hyicus. In this study, a series of derivatives were designed based on the NZ2114 template with the aim of obtaining peptides with more potent antimicrobial activity through changing net positive charge or hydrophobicity. Among them, a variant designated as NZL was highly expressed in Pichia pastoris (P. pastoris) with total secreted protein of 1505 mg/L in a 5-L fermenter and exhibited enhanced antimicrobial activity relative to parent peptide NZ2114. Additionally, NZL could kill over 99% of S. hyicus NCTC10350 in vitro within 8 h and in Hacat cells. The results of membrane permeabilization assay, morphological observations, peptide localization assay showed that NZL had potent activity against S. hyicus, which maybe kill S. hyicus through action on the cell wall. NZL also showed an effective therapy in a mouse peritonitis model caused by S. hyicus, superior to NZ2114 or ceftriaxone. While air quality in China has improved significantly in recent years, the population is becoming increasingly vulnerable to air pollution due to the aging population. In this study, we assessed premature deaths attributable to long- and short-term exposures to PM2.5 and O3, as well as their driving forces in Shandong from 2014 to 2060 based on county-level near-real-time air pollutant concentration datasets and projected concentrations of PM2.5 and O3. We found that the concentrations of PM2.5 and O3 in most districts and counties from Shandong are still higher than the corresponding concentration limit of Grade II. Premature mortality caused by long- and short-term exposures to PM2.5 decreased by 13,045 and 8092 in 2020 compared with those in 2014, respectively. Furthermore, premature mortality attributable to short-term exposure to O3 was 36.08% higher than that due to short-term exposure to PM2.5 in 2020. The results of the driving force analysis indicate that the health benefits brought about by the improvement in air quality have been offset by the changes in population age structure. In the future, the increase in O3 concentration and population aging are the top two driving forces having adverse effects on the health burden. This study provides support for controlling the health risks of PM2.5 and O3 pollution, especially for the development of dual-pollutant concentration targets and synergistic control strategies. Bulk metallic glass (BMGs) is highly expected for applications in engineering structures due to their superior mechanical properties. The fracture toughness of some BMGs was investigated at cryogenic and at elevated temperatures. However, the mechanism of the temperature-dependence of BMG toughness still remains elusive. Here, we characterized the fracture toughness of Zr61Ti2Cu25Al12 BMG prepared with Zr elemental pieces with low Hf content at temperatures ranging from 134 to 623 K. The relaxation spectrum of the BMG was characterized by a dynamic mechanical analysis using the same temperature range. We found that the BMG is tougher at onset temperatures of the relaxation processes than at peak temperatures. Toxoplasma gondii is an obligate protozoon that can infect all warm-blooded animals including humans. T. gondii afflicts one-third of the human population and is a detriment to the health of livestock and wildlife. Thus far, traditional drugs such as pyrimethamine and sulfadiazine used to treat T. gondii infection are inadequate as therapeutics due to relapse, long treatment period, and low efficacy in parasite clearance. Novel, efficacious drugs have not been available. Lumefantrine, as an antimalarial, is effective in killing T. gondii but has no known mechanism of action. We combined metabolomics with transcriptomics to investigate how lumefantrine inhibits T. gondii growth. We identified significant alternations in transcripts and metabolites and their associated functional pathways that are attributed to lumefantrine treatment. RH tachyzoites were used to infect Vero cells for three hours and subsequently treated with 900 ng/mL lumefantrine. Twenty-four hours post-drug treatment, we observed significant changes in transcripts associated with five DNA replication and repair pathways. Metabolomic data acquired through liquid chromatography-tandem mass spectrometry (LC-MS) showed that lumefantrine mainly affected sugar and amino acid metabolism, especially galactose and arginine. To investigate whether lumefantrine damages T. gondii DNA, we conducted a terminal transferase assay (TUNEL). TUNEL results showed that lumefantrine significantly induced apoptosis in a dose-dependent manner. Taken together, lumefantrine effectively inhibited T. gondii growth by damaging DNA, interfering with DNA replication and repair, and altering energy and amino acid metabolisms. With the accelerating growth of antimicrobial resistance (AMR), there is an urgent need for new antimicrobial agents with low or no AMR. Antimicrobial peptides (AMPs) have been extensively studied as alternatives to antibiotics (ATAs). Coupled with the new generation of high-throughput technology for AMP mining, the number of derivatives has increased dramatically, but manual running is time-consuming and laborious. Therefore, it is necessary to establish databases that combine computer algorithms to summarize, analyze, and design new AMPs. A number of AMP databases have already been established, such as the Antimicrobial Peptides Database (APD), the Collection of Antimicrobial Peptides (CAMP), the Database of Antimicrobial Activity and Structure of Peptides (DBAASP), and the Database of Antimicrobial Peptides (dbAMPs). These four AMP databases are comprehensive and are widely used. This review aims to cover the construction, evolution, characteristic function, prediction, and design of these four AMP databases. It also offers ideas for the improvement and application of these databases based on merging the various advantages of these four peptide libraries. Atmospheric gravity waves (GWs) in the mesosphere-lower thermosphere (MLT) are crucial for the understanding of general circulation. However, their dynamical characteristics are hardly retrieved due to the difficulty in the high-resolution observation of wind. Therefore, this paper uses eight years (2013–2020) of meteor radar measurements in the MLT region at Mohe station (53.5°N, 122.3°E), China, to retrieve high-temporal-resolution mesospheric wind data and further evaluate the temporal variation of GW kinetic energy. As the detected meteor trails exceed 6, the wind velocity is recalculated using the least square algorithm method, significantly increasing the temporal resolution of wind from 1 h up to 5 min. This resolution is sufficiently high for the investigation of GW kinetic energy, which exhibits a high spatial-temporal variability. For instance, it is enhanced in the winter season during the period of 0200–1400 UT and in the spring season during the period of 0800–1300 UT. The similarity between the climatological characteristics of GWs in MLT and the seasonal variation of GW total energy in the troposphere, determined from high-resolution radiosondes near to Mohe station, suggests that the meteorology in the lower atmosphere could be an important source of GWs in the MLT region. Background: Desertification is one of the main obstacles to global sustainable development. Monitoring, evaluating and mastering its driving factors are very important for the prevention and control of desertification. As one of the largest deserts in China, the development of desertification in Otindag Sandy Land (OSL) resulted in the reduction in land productivity and serious ecological/environmental consequences. Although many ecological restoration projects have been carried out, the vegetation restoration of OSL and the impact mechanism of climate and human activities on desertification remain unclear. Methods: Taking OSL as the research area, this paper constructs the desertification index by using the remote sensing images and meteorological and socio-economic data, between 1986 and 2016, and analyzes the spatio-temporal evolution process and driving factors of desertification by using trend analysis and spearman rank correlation. Results: The results showed that: (1) Desertification in the OSL has fluctuated greatly during the past 30 years. Desertification recovered between 1986 and 1990, expanded and increased between 1990 and 2000, reduced between 2000 and 2004, developed rapidly between 2004 and 2007, and recovered again between 2007 and 2016; (2) The desertification of OSL is dominated by a non-significant change trend, accounting for 73.27%. In the significant change trend, the area of desertification rising trend is 20.32%, which is mainly located in the north and east, and the area of declining trend is 6.41%, which is mainly located in the southwest; (3) Desertification is the result of the superposition of climate and human activities. Climate change is the main influencing factor, followed by human activities, and the superposition effects of the two are spatio-temporal differences. Conclusions: These results shed light on the development of desertification in OSL and the relative importance and complex interrelationship between human activities and climate in regulating the process of desertification. Based on this, we suggest continuing to implement the ecological restoration policy and avoid the destruction of vegetation by large-scale animal husbandry in order to improve the situation of desertification. Mild cognitive impairment (MCI) is the prodromal stage and an important risk factor of Alzheimer's disease (AD). Interventions at the MCI stage are significant in reducing the occurrence of AD. However, there are still many obstacles to the screening of MCI, resulting in a large number of patients going undetected. Given the strong correlation between gastrointestinal function and neuropsychiatric disorders, the aim of this study is to develop a risk prediction model for MCI based on gastrointestinal myoelectrical activity. The Mini-Mental State Examination and electrogastroenterography were applied to 886 participants in western China. All participants were randomly assigned to the training and validation sets in a ratio of 7:3. In the training set, risk variables were screened using LASSO regression and logistic regression, and risk prediction models were built based on nomogram and decision curve analysis, then validation was performed. Eight predictors were selected in the training set, including four electrogastroenterography parameters (rhythm disturbance, dominant frequency and dominant power ratio of gastric channel after meal, and time difference of intestinal channel after meal). The area under the ROC curve for the prediction model was 0.74 in the training set and 0.75 in the validation set, both of which exhibited great prediction ability. Furthermore, decision curve analysis displayed that the net benefit was more desirable when the risk thresholds ranged from 15% to 35%, indicating that the nomogram was clinically usable. Simple Summary: Oligopeptide transporter 1 (PepT1) plays a role in the transportation and absorption of oligopeptides, which is an important part of protein nutrition and affects the growth of animals. Tributyrin (TB), the precursor of butyrate, exhibits similar functions to those of the butyrate in intestinal nutrients absorption. The analysis of TB on the growth of grass carp and its regulation pathway on PepT1 may help us to better understand the functions of TB and oligopeptide transportation via PepT1, which can be modulated by diet. In this study, we demonstrated that an appropriate level of tributyrin supplementation in the diet promoted the growth of juvenile grass carp and elevated the expressions of caudal type homeobox 2 (CDX2), specificity protein 1 (SP1), and PepT1 in the grass carp intestine and primary intestine cell. In addition, CDX2 and SP1 regulating the expression of PepT1 was investigated. Finally, CDX2/SP1-mediating tributyrin regulation on PepT1 was elucidated. This study verified the effect of tributyrin on the growth of juvenile grass carp and clarified the tributyrin regulation pathway on CDX2/SP1-PepT1. The nutritional functions of tributyrin (TB) have been extensively studied, but questions remain regarding its influence on the growth of juvenile grass carp (Ctenopharyngodon idellus) and the regulation pathway to PepT1 in the intestine of grass carp. To answer the remaining questions, feeding trials, cell trials, and peritoneal injection trials were conducted in this study. The results showed that an appropriate level of TB (0.5 g/kg and 1.0 g/kg) supplementation in feed significantly promoted the growth performance of juvenile grass carp. The expressions of intestine genes (CDX2, SP1 and PepT1) related to oligopeptide transportation increased in the 0.5 g/kg TB group of feeding trials and both the 5 mM and 10 mM TB groups of the intestine cell trials, respectively. Subsequently, the injection trials of inhibitors CDX2 and SP1 demonstrated that the inhibition of CDX2 or SP1 decreased the mRNA expression of PepT1. Finally, the results of independent or combined treatments of TB and the inhibitors suggested that CDX2/SP1 mediated TB regulation on PepT1. These findings may help us to better understand the functions of TB on growth and PepT1 oligopeptide transportation, which could be modulated by dietary TB through the CDX2/SP1-PepT1 pathway in juvenile grass carp. The catalytic conversion of CO2 to C2 products through the CO2 reduction reaction (CO2RR) offers the possibility of preparing carbon-based fuels and valuable chemicals in a sustainable way. Herein, various Fen and Co5 clusters are designed to screen out the good catalysts with reasonable stability, as well as high activity and selectivity for either C2H4 or CH3CH2OH generation through density functional theory (DFT) calculations. The binding energy and cohesive energy calculations show that both Fe5 and Co5 clusters can adsorb stably on the N-doped carbon (NC) with one metal atom anchored at the center of the defected hole via a classical MN4 structure. The proposed reaction pathway demonstrates that the Fe5-NC cluster has better activity than Co5-NC, since the carbon–carbon coupling reaction is the potential determining step (PDS), and the free energy change is 0.22 eV lower in the Fe5-NC cluster than that in Co5-NC. However, Co5-NC shows a better selectivity towards C2H4 since the hydrogenation of CH2CHO to CH3CHO becomes the PDS, and the free energy change is 1.08 eV, which is 0.07 eV higher than that in the C-C coupling step. The larger discrepancy of d band center and density of states (DOS) between the topmost Fe and sub-layer Fe may account for the lower free energy change in the C-C coupling reaction. Our theoretical insights propose an explicit indication for designing new catalysts based on the transition metal (TM) clusters supported on N-doped carbon for multi-hydrocarbon synthesis through systematically analyzing the stability of the metal clusters, the electronic structure of the critical intermediates and the energy profiles during the CO2RR. SMYD3 is a SET-domain-containing methyltransferase that catalyzes the transfer of methyl groups onto lysine residues of substrate proteins. Methylation of MAP3K2 by SMYD3 has been implicated in Ras-driven tumorigenesis, which makes SMYD3 a potential target for cancer therapy. Of all SMYD family proteins, SMYD3 adopt a closed conformation in a crystal structure. Several studies have suggested that the conformational changes between the open and closed forms may regulate the catalytic activity of SMYD3. In this work, we carried out extensive molecular dynamics simulations on a series of complexes with a total of 21 μs sampling to investigate the conformational changes of SMYD3 and unveil the molecular mechanisms. Based on the C-terminal domain movements, the simulated models could be depicted in three different conformational states: the closed, intermediate and open states. Only in the case that both the methyl donor binding pocket and the target lysine-binding channel had bound species did the simulations show SMYD3 maintaining its conformation in the closed state, indicative of a synergetic effect of the cofactors and target lysine on regulating the conformational change of SMYD3. In addition, we performed analyses in terms of structure and energy to shed light on how the two regions might regulate the C-terminal domain movement. This mechanistic study provided insights into the relationship between the conformational change and the methyltransferase activity of SMYD3. Bulk metallic glasses have application potential in engineering structures due to their exceptional strength and fracture toughness. Their fatigue resistance is very important for the application as well. We report the tension-tension fatigue damage behavior of a Zr61Ti2Cu25Al12 bulk metallic glass, which has the highest fracture toughness among BMGs. The Zr61Ti2Cu25Al12 glass exhibits a tension-tension fatigue endurance limit of 195 MPa, which is higher than that of high-toughness steels. The fracture morphology of the specimens depends on the applied stress amplitude. We found flocks of shear bands, which were perpendicular to the loading direction, on the surface of the fatigue test specimens with stress amplitude higher than the fatigue limit of the glass. The fatigue cracking of the glass initiated from a shear band in a shear band flock. Our work demonstrated that the Zr61Ti2Cu25Al12 glass is a competitive structural material and shed light on improving the fatigue resistance of bulk metallic glasses. [ABSTRACT FROM AUTHOR]

The nucleotide-binding site–leucine-rich repeat (NBS–LRR) gene family is the largest group of plant disease resistance (R) genes widespread in response to viruses, bacteria, and fungi usually involved in effector triggered immunity (ETI). Forty members of the Chinese cabbage CC type NBS–LRR family were investigated in this study. Gene and protein characteristics, such as distributed locations on chromosomes and gene structures, were explored through comprehensive analysis. CC–NBS–LRR proteins were classified according to their conserved domains, and the phylogenetic relationships of CC–NBS–LRR proteins in Brassica rapa, Arabidopsis thaliana, and Oryza sativa were compared. Moreover, the roles of BrCC–NBS–LRR genes involved in pathogenesis-related defense were studied and analyzed. First, the expression profiles of BrCC–NBS–LRR genes were detected by inoculating with downy mildew and black rot pathogens. Second, sensitive and resistant Chinese cabbage inbred lines were screened by downy mildew and black rot. Finally, the differential expression levels of BrCC–NBS–LRR genes were monitored at 0, 1, 3, 6, 12 and 24 h for short and 0, 3, 5, 7, 10 and 14 days for long inoculation time. Nanofiltration membrane technology is an effective method for secondary treated sewage purification. However, membrane fouling, which is inevitable in the membrane-separation process, can reduce membrane performance and shorten membrane life. Installing a turbulence promoter is a promising means of improving the hydraulic conditions inside the membrane chamber. In this study, the effect of turbulence promoter on prolonging membrane life was studied for the first time. Flat-sheet polyethersulfone nanofiltration membrane was used to filter humic acid solution, used for simulating secondary treated sewage. By comparing photographs and SEM images of the membrane before and after the simulated secondary treated sewage filtration, it was found that humic acid tended to be deposited on the low-velocity region, which was reflected by COMSOL simulation. After incorporating a turbulence promoter, the reduction of the humic acid deposition area and membrane fouling resistance indicated that the turbulence promoter could reduce membrane fouling due to the improved hydraulic conditions. Additionally, the turbulence promoter also increased the flux and reduced the flux decay rate. The turbulence promoter was then place in the crossflow flat-sheet membrane filtration module, and the variation of flux with time was tested in simulated secondary treated sewage with different concentrations. The results showed that the membrane life for the filtration of simulated secondary treated sewage comprising 50, 250, and 500 ppm humic acid increased by 23.1%, 80.4%, and 85.7%, respectively. The results of this article can serve as a reference for the prediction of membrane life and the performance enhancement mechanism of membranes containing a turbulence promoter. High-calorie diet, circadian rhythms and metabolic features are intimately linked. However, the mediator(s) between nutritional status, circadian rhythms and metabolism remain largely unknown. This article aims to clarify the key metabolic pathways bridging nutritional status and circadian rhythms based on a combination of metabolomics and molecular biological techniques. A mouse model of high-fat diet-induced obesity was established and serum samples were collected in obese and normal mice at different zeitgeber times. Gas chromatography/mass spectrometry, multivariate/univariate data analyses and metabolic pathway analysis were used to reveal changes in metabolism. Metabolites involved in the metabolism of purines, carbohydrates, fatty acids and amino acids were markedly perturbed in accordance with circadian related variations, among which purine catabolism showed a typical oscillation. What's more, the rhythmicity of purine catabolism dampened in the high-fat diet group. The expressions of clock genes and metabolic enzymes in the liver were measured. The mRNA expression of Xanthine oxidase (Xor) was highly correlated with the rhythmicity of Clock, Rev-erbα and Bmal1, as well as the metabolites involved in purine catabolism. These data showed that a high-fat diet altered the circadian rhythm of metabolic pathways, especially purine catabolism. It had an obvious circadian oscillation and a high-fat diet dampened its circadian rhythmicity. Major depressive disorder (MDD), also known as depression, is a state characterized by low mood and aversion to activity. Platycodins Folium (PF) is the dried leaf of Platycodon grandiflorum, with anti-inflammatory and antioxidative activities. Our previous research suggested that PF was rich in flavonoids, phenols, organic acids, triterpenoid saponins, coumarins and terpenoids. This study aimed to investigate the antidepressant effect of PF using lipopolysaccharide (LPS)-induced depressive mice. Several behavior tests (sucrose preference test (SPT), forced swimming test (FST) and tail suspension test (TST)) and biochemical parameters (IL-6, TNF-α and SOD levels) were used to evaluate the antidepressive effect of PF on LPS-induced depression model. Furthermore, a UPLC-Q/TOF-MS-based metabolomics approach was applied to explore the latent mechanism of PF in attenuating depression. As a result, a total of 21 and 11 metabolites that potentially contribute to MDD progress and PF treatment were identified in serum and hippocampus, respectively. The analysis of metabolic pathways revealed that lipid metabolism, amino acid metabolism, energy metabolism, arachidonic acid metabolism, glutathione metabolism and inositol phosphate metabolism were disturbed in a model of mice undergoing MDD and PF treatment. These results help us to understand the pathogenesis of depression in depth, and to discover targets for clinical diagnosis and treatment. Probable Maximum Precipitation (PMP) is an essential prerequisite in designing dams, spillways, and reservoirs in order to minimize the risk of overtopping infrastructure collapse, especially under today's changing climate. This study investigates conventional PMP estimation approach by using both scarce in-situ observations and mainstream satellite precipitation products in the Dadu River basin, where plenty of reservoirs and dams are being built. The satellite data include Climate Prediction Center (CPC) MORPHing algorithm (CMORPH), Precipitation Estimation from Remotely Sensed Information using Artificial Neural Networks-Climate Data Record (PERSIANN-CDR), and Tropic Rainfall Measuring Mission (TRMM) Multi-satellite Precipitation Analysis (TMPA) 3B42V7. The evaluation of satellite products shows that CMORPH and 3B42V7 agree well with gauge-based dataset for the period of 1998-2013 at both the grid and basin scales, also capturing the extreme precipitation events, with high Correlation Coefficients (CC) in terms of 0.68 and 0.71, respectively. Also, CMORPH and 3B42V7 show better performance for the magnitude and spatial distribution of 24-h PMP in such complex terrains. PERSIANN-CDR shows an overestimation in the upstream and an underestimation in the downstream. As among the first studies of satellite precipitation-based PMP estimation, this work sheds lights on the suitability of satellite precipitation in PMP estimation and could provide a reference for future extended spatially-distributed PMP estimation in vast ungauged regions. The spatial and temporal variations in global precipitable water (PW) derived from satellite and radiosonde observations during 2007–2016 are compared and analyzed in this study. The two observations show a good agreement in global spatiotemporal distributions and trends in PW. Generally, PW shows apparent latitudinal and seasonal variations. The averaged PW peaks in the tropical region, with the value of 40 mm or higher, and then decreases apparently to the polar region, less than 10 mm. PWs are larger in warm seasons and smaller in cold seasons for both hemispheres. It is indicated that the spatial and seasonal variabilities of PW are highly related to water vapor source and surface temperature as a direct heat source. The global PWs have changed in the past 60 years and, to some extent, strongly in the recent 10 years. It is found that PWs at Australian stations present statistically significant decreasing trends in last 60 years, while PWs at most stations in the United States have increasing trends. According to the global observations, it is found that PWs show uptrends over land and downtrends over the ocean in last 10 years, implying the widespread increase of water vapor in the troposphere over land. Laser pulses can be utilized to induce intermediate states of phase change materials between amorphous and crystalline phases, making phase change materials attractive and applicable for multi-level storage applications. In this paper, intermediate states of Ge2Sb2Te5 thin films induced via employing a nanosecond multi-pulse laser with different energy and pulse duration were performed by Raman spectroscopy, reflection measurement and thermal simulations. Upon laser-crystallized Ge2Sb2Te5 films, optical functions change drastically, leading to distinguishable reflectivity contrasts of intermediate states between amorphous and crystalline phases due to different crystallinity. The changes in optical intensity for laser-crystallized Ge2Sb2Te5 are also accompanied by micro-structure evolution, since high-energy and longer pulses result in higher-level intermediate states (corresponding to high reflection intensity) and largely contribute to the formation of stronger Raman peaks. By employing thermal analysis, we further demonstrated that the variations of both laser fluence and pulse duration play decisive roles in the degree of crystallinity of Ge2Sb2Te5 films. Laser fluence is mainly responsible for the variations in crystallization temperature, while the varying pulse duration has a great impact on the crystallization time. The combination of mode division multiplexing (MDM) based on orbital angular momentum (OAM) modes with wavelength division multiplexing (WDM) has attracted considerable attention due to its ability to increase optical transmission capacity. However, the switching of the multi-wavelength and multi-order OAM mode in an all-fiber structure has always been a challenge. As a solution, a thermally tunable dual-core photonic crystal fiber (DC-PCF) is proposed to achieve multi-order and multi-wavelength switching of the OAM mode. The results show that the OAM mode with topological charge m = ±1 can be excited with the linear polarization fundamental mode (LPFM) and circular polarization fundamental mode (CPFM). In addition, the device can effectively excite a high-purity ±1st order OAM mode with wavelengths ranging from 1520 to 1575 nm by thermal tuning. The purity of the mode is in excess of 99%, and the energy conversion efficiency (ECE) is above 95%. Unzipping of carbon nanotubes (CNTs) has been widely explored to obtain new nanocarbon structures with promising properties. In this work, we report that unzipping of CNTs according to the well-established modified Hummers method produces unzipped CNTs (uCNTs) that exhibit self-photoluminescence that depends on the diameter of pristine CNTs. The uCNTs were characterized using FTIR spectroscopy, XRD, XPS, and Raman spectroscopy indicating that unzipping is accompanied by the introduction of defects and oxygen-containing functional groups. The morphology of CNTs and uCNTs was determined by TEM showing longitude unzipping of CNTs. Our study shows that increasing the diameter of pristine CNTs results in decreasing the edge etching effect and decreasing the functionality of uCNTs. Based on the UV-Vis spectra, the band gap of uCNTs was calculated using the Kubelka–Munk function. The band gap of uCNTs increased with decreasing diameter of pristine CNTs. The uCNTs exhibited photoluminescence with a good emission in the visible light region. The uCNTs with the largest band gap and the highest oxygen content had the strongest fluorescence intensity. Moreover, different metal ions produced different degrees of fluorescence quenching for uCNT-15, which verified the self-photoluminescence of uCNTs. [ABSTRACT FROM AUTHOR]

Simple Summary: In cancer, CD8+ T cells enter a state of dysfunction within the tumor that prevents them from targeting and killing tumor cells. Our study aims to uncover how CD8+ T cells can be helped by CD4+ T cells or modified in order to improve their effector function against cancer. Thus, allowing them to better fight and control tumors. Our work shows that the protein Basic Leucine Zipper ATF-Like Transcription Factor (BATF) may be a key regulator of CD8+ T cells and their anti-tumor function. These findings can provide further insight for the development of novel therapeutic treatments for cancer patients. In cancer, CD8+ T cells enter a dysfunctional state which prevents them from effectively targeting and killing tumor cells. Tumor-infiltrating CD8+ T cells consist of a heterogeneous population of memory-like progenitor, effector, and terminally exhausted cells that exhibit differing functional and self-renewal capacities. Our recently published work has shown that interleukin (IL)-21-producing CD4+ T cells help to generate effector CD8+ T cells within the tumor, which results in enhanced tumor control. However, the molecular mechanisms by which CD4+ helper T cells regulate the differentiation of effector CD8+ T cells are not well understood. In this study, we found that Basic Leucine Zipper ATF-Like Transcription Factor (BATF), a transcription factor downstream of IL-21 signaling, is critical to maintain CD8+ T cell effector function within the tumor. Using mixed bone marrow chimeras, we demonstrated that CD8+ T cell-specific deletion of BATF resulted in impaired tumor control. In contrast, overexpressing BATF in CD8+ T cells enhanced effector function and resulted in improved tumor control, bypassing the need for CD4+ helper T cells. Transcriptomic analyses revealed that BATF-overexpressing CD8+ T cells had increased expression of costimulatory receptors, effector molecules, and transcriptional regulators, which may contribute to their enhanced activation and effector function. Taken together, our study unravels a previously unappreciated CD4+ T cell-derived IL-21–BATF axis that could provide therapeutic insights to enhance effector CD8+ T cell function to fight cancer. The 2011 Tohoku earthquake and the following enormous tsunami caused great disturbances in the ionosphere that were observed in various regions along the Pacific Ocean. In this study, the oblique-incidence ionosonde detection network located in North China was applied to investigate the inland ionospheric disturbances related to the 2011 tsunamigenic earthquake. The ionosonde network consists of five transmitters and 20 receivers and can monitor regional ionosphere disturbances continuously and effectively. Based on the recorded electron density variations along the horizontal plane, the planar middle-scale ionospheric disturbances (MSTIDs) associated with the 2011 Tohoku tsunamigenic earthquake were detected more than 2000 km west of the epicenter about six hours later. The MSTIDs captured by the Digisonde, high-frequency (HF) Doppler measurement, and Constellation Observing System for Meteorology, Ionosphere, and Climate (COSMIC) satellite provided more information about the far-field inland propagation characteristics of the westward propagating gravity waves. The results imply that the ionosonde network has the potential for remote sensing of ionospheric disturbances induced by tsunamigenic earthquakes and provide a perspective for investigating the propagation process of associated gravity waves. γ-Aminobutyric acid (GABA) is a widely distributed non-protein amino acid mediated the regulation of nitrate uptake and Al3+ tolerance in plants. However, there are few reports about the involvement of GABA in the regulation of iron (Fe) acquisition and translocation. Here, we show that GABA regulates Fe homeostasis in rice seedlings. Exogenous GABA decreased the chlorophyll concentration in leaves, with or without Fe supply. Over-expression of glutamate decarboxylase (GAD) gene, coding a crucial enzyme of GABA production, elevated endogenous GABA content and caused more leaf chlorosis than wild type (Nipponbare). GABA inhibited Fe transportation from roots to shoots and GABA application elevated the expression levels of Fe deficiency (FD)-related genes under conditions of Fe-sufficiency (FS), suggesting that GABA is a regulator of Fe translocation. Using Perls' blue staining, we found that more ferric iron (Fe3+) was deposited in the epidermal cells of roots treated with GABA compared with control roots. Anatomic section analysis showed that GABA treatment induced more aerenchyma formation compared with the control. Aerenchyma facilitated the oxidization of soluble ferrous iron (Fe2+) into insoluble Fe3+, resulted in Fe precipitation in the epidermis, and inhibited the transportation of Fe from roots to shoots. Pharmacokinetic and metabolism studies of 12-riboside-pseudoginsengenin DQ (RPDQ), a novel ginsenoside with an anti-cancer effect, were carried out, aiming at discussing the characteristics of the ginsenoside with glycosylation site at C-12. In the pharmacokinetic analysis, we developed and validated a method by UPLC-MS to quantify RPDQ in rat plasma. In the range of 5–1000 ng/mL, the assay was linear (R2 > 0.9966), with the LLOQ (lower limit of quantification) being 5 ng/mL. The LOD (limit of detection) was 1.5 ng/mL. The deviations of intra-day and inter-day, expressed as relative standard deviation (RSD), were ≤ 3.51% and ≤ 5.41% respectively. The accuracy, expressed as relative error (RE), was in the range –8.82~3.47% and –5.61~2.87%, respectively. The recoveries were in the range 85.66~92.90%. The method was then applied to a pharmacokinetic study in rats intragastrically administrated with 6, 12, and 24 mg/kg RPDQ. The results showed that RPDQ exhibited slow oral absorption (Tmax = 7.0 h, 7.5 h, and 7.0 h, respectively), low elimination (t1/2 = 12.59 h, 12.83 h, and 13.74 h, respectively) and poor absolute bioavailability (5.55, 5.15, and 6.08%, respectively). Moreover, the investigation of metabolites were carried out by UPLC-QTOF-MS. Thirteen metabolites of RPDQ were characterized from plasma, bile, urine, and feces of rats. Some metabolic pathways, including oxidation, acetylation, hydration, reduction, hydroxylation, glycine conjugation, sulfation, phosphorylation, glucuronidation, glutathione conjugation, and deglycosylation, were profiled. HnRNP A2/B1 has been found to be an oncogenic protein strongly related to the growth of human glioma cells. Herein, β-asarone, the main component in the volatile oil of Acori tatarinowii Rhizoma, inhibited the cell viability, proliferation, and colony formation ability of U251 cells. Moreover, β-asarone induced apoptosis and cell cycle arrest at the G1 phase. Notably, β-asarone suppressed the expression of hnRNP A2/B1 and hnRNPA2/B1 overexpression remarkably reversed β-asarone-mediated apoptosis and cell cycle arrest. Importantly, β-asarone promoted the alternative splicing of Bcl-x by enhancing the ratio of Bcl-xS/Bcl-xL. Meanwhile, hnRNPA2/B1 overexpression mitigated the promoting effect of β-asarone on the alternative splicing of Bcl-x. β-asarone also regulated the level of the key proteins involved in the death receptor pathway and mitochondrial apoptosis pathway. Additionally, β-asarone modulated the cell cycle-related proteins p21, p27, Cdc25A, cyclin D, cyclin E, and CDK2. Finally, β-asarone inhibited tumor growth and induced apoptosis in nude mice bearing U251 tumor xenografts. β-asarone also suppressed the hnRNP A2/B1 expression, enhanced the expression of cleaved-caspase 3 and p27 and the ratio of Bcl-xS/Bcl-xL, and reduced the expression of CDK2 in U251 xenografts. Together, β-asarone-induced apoptosis and cell cycle arrest of U251 cells may be related to the suppression of hnRNPA2/B1-mediated signaling pathway. [ABSTRACT FROM AUTHOR]
