The redistribution of baryonic matter in massive haloes through processes like active galactic nuclei feedback and star formation leads to a suppression of the matter power spectrum on small scales. This redistribution can be measured empirically via the gas and stellar mass fractions in galaxy clusters, and leaves imprints on their electron density profiles. We constrain two semi-analytical baryon correction models with a compilation of recent Bayesian population studies of galaxy groups and clusters sampling a mass range above ∼3 × 1013  M ⊙, and with cluster gas density profiles derived from deep, high-resolution X-ray observations. We are able to fit all the considered observational data, but highlight some anomalies in the observations. The constraints allow us to place precise, physically informed priors on the matter power spectrum suppression. At a scale of k  = 1  h  Mpc−1 we find a suppression of |$0.042^{+0.012}_{-0.014}$| (⁠|$0.049^{+0.016}_{-0.012}$|⁠), while at k  = 3  h  Mpc−1 we find |$0.184^{+0.026}_{-0.031}$| (⁠|$0.179^{+0.018}_{-0.020}$|⁠), depending on the model used. In our fiducial setting, we also predict at 97.5 per cent credibility, that at scales k < 0.37  h  Mpc−1 baryon feedback impacts the matter power less than 1 per cent. This puts into question if baryon feedback is the driving factor for the discrepancy between cosmic shear and primary CMB results. We independently confirm results on this suppression from small-scale cosmic shear studies, while we exclude some hydro-dynamical simulations with too strong and too weak baryonic feedback. Our empirical prediction of the power spectrum suppression shows that studies of galaxy groups and clusters will be instrumental in unlocking the cosmological constraining power of future cosmic shear experiments like Euclid and Rubin-LSST, and invites further investigation of the baryon correction models. The paper presents the analysis of optically selected GAMA groups and clusters in the SRG/eROSITA X-ray map of eROSITA Final Equatorial Depth Survey, in the halo mass range 1013−5 × 1014 M⊙ and at z < 0.2. All X-ray detections have a clear GAMA counterpart, but most of the GAMA groups in the halo mass range 1013−1014 M⊙ remain undetected. We compare the X-ray surface brightness profiles of the eROSITA detected groups with the mean stacked profile of the undetected low-mass haloes at fixed halo mass. Overall, we find that the undetected groups exhibit less concentrated X-ray surface brightness, dark matter, and galaxy distributions with respect to the X-ray-detected haloes. The mean gas mass fraction profiles are consistent in the two samples within 1.5σ, indicating that the gas follows the dark matter profile. The low-mass concentration and the magnitude gap indicate that these systems are young. They reside with a higher probability in filaments while X-ray-detected groups favour the nodes of the Cosmic Web. Because of the lower central emission, the undetected systems tend to be X-ray underluminous at fixed halo mass and to lie below the LX − M halo relation. Interestingly, the X-ray-detected systems inhabiting the nodes scatter the less around the relation, while those in filaments tend to lie below it. We do not observe any strong relationship between the system X-ray appearance and the active galactic nucleus (AGN) activity. We cannot exclude the role of the past AGN feedback in affecting the gas distribution over the halo lifetime. We present the first systematic follow-up of Planck Sunyaev–Zeldovich effect (SZE) selected candidates down to signal-to-noise (S/N) of 3 over the 5000 deg2 covered by the Dark Energy Survey. Using the MCMF cluster confirmation algorithm, we identify optical counterparts, determine photometric redshifts, and richnesses and assign a parameter, f cont, that reflects the probability that each SZE-optical pairing represents a random superposition of physically unassociated systems rather than a real cluster. The new PSZ-MCMF cluster catalogue consists of 853 MCMF confirmed clusters and has a purity of 90 per cent. We present the properties of subsamples of the PSZ-MCMF catalogue that have purities ranging from 90 per cent to 97.5 per cent, depending on the adopted f cont threshold. Halo mass estimates M 500, redshifts, richnesses, and optical centres are presented for all PSZ-MCMF clusters. The PSZ-MCMF catalogue adds 589 previously unknown Planck identified clusters over the DES footprint and provides redshifts for an additional 50 previously published Planck -selected clusters with S/N>4.5. Using the subsample with spectroscopic redshifts, we demonstrate excellent cluster photo- z performance with an RMS scatter in Δ z /(1 + z) of 0.47 per cent. Our MCMF based analysis allows us to infer the contamination fraction of the initial S/N>3 Planck -selected candidate list, which is ∼50 per cent. We present a method of estimating the completeness of the PSZ-MCMF cluster sample. In comparison to the previously published Planck cluster catalogues, this new S/N>3 MCMF confirmed cluster catalogue populates the lower mass regime at all redshifts and includes clusters up to z ∼1.3. HD 49798 is a hot subdwarf of O spectral type in a 1.55 d orbit with the X-ray source RX J0648.0−4418, a compact object with a spin period of |$13.2\,$| s. We use recent data from the Neutron Star Interior Composition Explorer instrument, joined with archival data from XMM–Newton and ROSAT , to obtain a phase-connected timing solution spanning ∼30 yr. Contrary to previous works, which relied on parameters determined through optical observations, the new timing solution could be derived using only X-ray data. We confirm that the compact object is steadily spinning up with |$\dot{P} = -2.28(2) \times 10^{-15}\,$| s s−1 and obtain a refined measure of the projected semimajor axis of the compact object a Xsin  i  = 9.60(5) light-second. This allows us to determine the inclination and masses of the system as |$i=84.5(7)\,$| deg, M X = 1.220(8)  |$\rm {M}_\odot$|⁠ , and |$M_{\rm opt}=1.41(2)\,$| |$\rm {M}_\odot$|⁠. We also study possible long-term (approximately years) and orbital variations of the soft X-ray pulsed flux, without finding evidence for variability. In the light of the new findings, we discuss the nature of the compact object, concluding that the possibility of a neutron star in the subsonic propeller regime is unlikely, while accretion of the subdwarf wind on to a massive white dwarf can explain the observed luminosity and spin-up rate for a wind velocity of ∼800 km s−1. We present the splashback radius measurements around the SRG/eROSITA eFEDS X-ray selected galaxy clusters by cross-correlating them with HSC S19A photometric galaxies. The X-ray selection is expected to be less affected by systematics related to projection that affects optical cluster finder algorithms. We use a nearly volume-limited sample of 109 galaxy clusters selected in 0.5–2.0 keV band having luminosity |$L_X \gt 10^{43.5}\, {\rm erg \, s}^{-1}\, h^{-2}$| within the redshift z < 0.75 and obtain measurements of the projected cross-correlation with a signal to noise of 17.43. We model our measurements to infer a 3D profile and find that the steepest slope is sharper than −3 and associate the location with the splashback radius. We infer the value of the 3D splashback radius |$r_{\rm sp} = 1.45^{+0.30}_{-0.26}\, h^{-1}\, {\rm Mpc}$|. We also measure the weak-lensing signal of the galaxy clusters and obtain halo mass |$\log [M_{\rm 200m}/ h^{-1}\, {\rm M_\odot }] = 14.52 \pm 0.06$| using the HSC-S16A shape catalogue data at the median redshift z  = 0.46 of our cluster sample. We compare our r sp values with the spherical overdensity boundary |$r_{\rm 200m} = 1.75 \pm 0.08\, h^{-1} \, {\rm Mpc}$| based on the halo mass, which is consistent within 1.2σ with the ΛCDM predictions. Further studies will play a pivotal role in the refinement of I ZBTB11 i -related neuropsychiatric features and the investigation of the potential role of I ZBTB11 i as a risk gene for Tourette syndrome. The soft X-ray pulsar RX J1856.5 − 3754 is the brightest member of a small class of thermally emitting, radio-silent, isolated neutron stars. Its X-ray spectrum is almost indistinguishable from a blackbody with |$kT^\infty \approx {60}\, {\rm eV}$|⁠ , but evidence of harder emission above |$\sim {1}\, {\rm keV}$| has been recently found. We report on a spectral and timing analysis of RX J1856.5 − 3754 based on the large amount of data collected by XMM-Newton in 2002–2022, complemented by a dense monitoring campaign carried out by NICER in 2019. Through a phase-coherent timing analysis we obtained an improved value of the spin-down rate |$\dot{\nu }=-6.042(4)\times 10^{-16}\, {\rm Hz\, s}^{-1}$|⁠ , reducing by more than one order magnitude the uncertainty of the previous measurement, and yielding a characteristic spin-down field of |$1.47\times 10^{13}\, {\rm G}$|⁠. We also detect two spectral components above |$\sim 1\, {\rm keV}$|⁠ : a blackbody-like one with |$kT^\infty =138\pm 13\,$| eV and emitting radius |$31_{-16}^{+8}\,$| m, and a power law with photon index |$\Gamma =1.4_{-0.4}^{+0.5}$|⁠. The power-law 2–8 keV flux, |$(2.5_{-0.6}^{+0.7})\times 10^{-15}\, {\rm erg}\, {\rm cm}^{-2}\, {\rm s}^{-1}$|⁠ , corresponds to an efficiency of 10−3, in line with that seen in other pulsars. We also reveal a small difference between the 0.1–0.3 keV and 0.3–1.2 keV pulse profiles, as well as some evidence for a modulation above 1.2 keV. These results show that, notwithstanding its simple spectrum, RX J1856.5 − 3754 still has a non-trivial thermal surface distribution and features non-thermal emission as seen in other pulsars with higher spin-down power. Cosmological constraints from current and upcoming galaxy cluster surveys are limited by the accuracy of cluster mass calibration. In particular, optically identified galaxy clusters are prone to selection effects that can bias the weak lensing mass calibration. We investigate the selection bias of the stacked cluster lensing signal associated with optically selected clusters, using clusters identified by the redMaPPer algorithm in the Buzzard simulations as a case study. We find that at a given cluster halo mass, the residuals of redMaPPer richness and weak lensing signal are positively correlated. As a result, for a given richness selection, the stacked lensing signal is biased high compared with what we would expect from the underlying halo mass probability distribution. The cluster lensing selection bias can thus lead to overestimated mean cluster mass and biased cosmology results. We show that the lensing selection bias exhibits a strong scale dependence and is approximately 20–60 per cent for ΔΣ at large scales. This selection bias largely originates from spurious member galaxies within ±20–60  |$h^{-1}\, \rm Mpc$| along the line of sight, highlighting the importance of quantifying projection effects associated with the broad redshift distribution of member galaxies in photometric cluster surveys. While our results qualitatively agree with those in the literature, accurate quantitative modelling of the selection bias is needed to achieve the goals of cluster lensing cosmology and will require synthetic catalogues covering a wide range of galaxy–halo connection models. Most leaf functional trait studies in the Amazon basin do not consider ontogenetic variations (leaf age), which may influence ecosystem productivity throughout the year. When leaf age is taken into account, it is generally considered discontinuous, and leaves are classified into age categories based on qualitative observations. Here, we quantified age-dependent changes in leaf functional traits such as the maximum carboxylation rate of ribulose-1,5-biphosphate carboxylase/oxygenase (Rubisco) (V cmax), stomatal control (Cgs %), leaf dry mass per area and leaf macronutrient concentrations for nine naturally growing Amazon tropical trees with variable phenological strategies. Leaf ages were assessed by monthly censuses of branch-level leaf demography; we also performed leaf trait measurements accounting for leaf chronological age based on days elapsed since the first inclusion in the leaf demography, not predetermined age classes. At the tree community scale, a nonlinear relationship between V cmax and leaf age existed: young, developing leaves showed the lowest mean photosynthetic capacity, increasing to a maximum at 45 days and then decreasing gradually with age in both continuous and categorical age group analyses. Maturation times among species and phenological habits differed substantially, from 8 ± 30 to 238 ± 30 days, and the rate of decline of V cmax varied from −0.003 to −0.065 μmol CO2 m−2 s−1 day−1. Stomatal control increased significantly in young leaves but remained constant after peaking. Mass-based phosphorus and potassium concentrations displayed negative relationships with leaf age, whereas nitrogen did not vary temporally. Differences in life strategies, leaf nutrient concentrations and phenological types, not the leaf age effect alone, may thus be important factors for understanding observed photosynthesis seasonality in Amazonian forests. Furthermore, assigning leaf age categories in diverse tree communities may not be the recommended method for studying carbon uptake seasonality in the Amazon, since the relationship between V cmax and leaf age could not be confirmed for all trees. Parametric modeling of galaxy cluster density profiles from weak lensing observations leads to a mass bias, whose detailed understanding is critical in deriving accurate mass-observable relations for constraining cosmological models. Drawing from existing methods, we develop a robust framework for calculating this mass bias in one-parameter fits to simulations of dark matter haloes. We show that our approach has the advantage of being independent of the absolute noise level, so that only the number of haloes in a given simulation and the representativeness of the simulated haloes for real clusters limit the accuracy of the bias estimation. While we model the bias as a lognormal distribution and the haloes with a Navarro–Frenk–White profile, our method can be generalized to any bias distribution and parametric model of the radial mass distribution. We find that the lognormal assumption is not strictly valid in the presence of miscentring of haloes. We investigate the use of cluster centres derived from weak lensing in the context of mass bias, and tentatively find that such centroids can yield sensible mass estimates if the convergence peak has a signal-to-noise ratio (SNR) approximately greater than 4. In this context we also find that the standard approach to estimating the positional uncertainty of weak lensing mass peaks using bootstrapping severely underestimates the true positional uncertainty for peaks with low SNRs. Though we determine the mass and redshift dependence of the bias distribution for a few experimental setups, our focus remains providing a general approach to computing such distributions. Cosmological inference from cluster number counts is systematically limited by the accuracy of the mass calibration, i.e. the empirical determination of the mapping between cluster selection observables and halo mass. In this work we demonstrate a method to quantitatively determine the bias and uncertainties in weak-lensing (WL) mass calibration. To this end, we extract a library of projected matter density profiles from hydrodynamical simulations. Accounting for shear bias and noise, photometric redshift uncertainties, mis-centreing, cluster member contamination, cluster morphological diversity, and line-of-sight projections, we produce a library of shear profiles. Fitting a one-parameter model to these profiles, we extract the so-called WL mass M WL. Relating the WL mass to the halo mass from gravity-only simulations with the same initial conditions as the hydrodynamical simulations allows us to estimate the impact of hydrodynamical effects on cluster number counts experiments. Creating new shear libraries for ∼1000 different realizations of the systematics provides a distribution of the parameters of the WL to halo mass relation, reflecting their systematic uncertainty. This result can be used as a prior for cosmological inference. We also discuss the impact of the inner fitting radius on the accuracy, and determine the outer fitting radius necessary to exclude the signal from neighbouring structures. We construct and validate the selection function of the MARD-Y3 galaxy cluster sample. This sample was selected through optical follow-up of the 2nd ROSAT faint source catalogue with Dark Energy Survey year 3 data. The selection function is modelled by combining an empirically constructed X-ray selection function with an incompleteness model for the optical follow-up. We validate the joint selection function by testing the consistency of the constraints on the X-ray flux–mass and richness–mass scaling relation parameters derived from different sources of mass information: (1) cross-calibration using South Pole Telescope Sunyaev-Zel'dovich (SPT-SZ) clusters, (2) calibration using number counts in X-ray, in optical and in both X-ray and optical while marginalizing over cosmological parameters, and (3) other published analyses. We find that the constraints on the scaling relation from the number counts and SPT-SZ cross-calibration agree, indicating that our modelling of the selection function is adequate. Furthermore, we apply a largely cosmology independent method to validate selection functions via the computation of the probability of finding each cluster in the SPT-SZ sample in the MARD-Y3 sample and vice versa. This test reveals no clear evidence for MARD-Y3 contamination, SPT-SZ incompleteness or outlier fraction. Finally, we discuss the prospects of the techniques presented here to limit systematic selection effects in future cluster cosmological studies. [ABSTRACT FROM AUTHOR]

Objectives The efficacy and safety of high-dose intravenous immunoglobulin (IVIG) in treatment-resistant diabetic painful polyneuropathy (DPN) were assessed. Design This was a randomized, double-blind, placebo-controlled, multicenter trial (EudraCT 2010–023883–42). Setting This trial was conducted at eight sites in Italy with a neurology specialist level of care. Subjects Twenty-six diabetic patients with DPN who reported baseline severity of pain >60 units (mm) on a VAS scale at enrollment and were resistant to antidepressants and antiepileptic drugs were enrolled; 23 were randomized (11 in the IVIG arm and 12 in the placebo arm). All patients completed the study and were evaluated. All patients were Caucasian, 15 were male, and 21 had a diagnosis of type II diabetes. Methods IVIG (0.4 g/kg/d) or placebo was given for five consecutive days. Pain intensity (visual analog scale, Neuropathic Pain Symptom Inventory) and quality of life (36-Item Short-Form Health Survey, Clinical/Patient Global Impression of Change questionnaires) assessments were performed at visits: baseline, start of therapy (one week later), end of therapy (five days later), and follow-up (four and eight weeks later). Results The study achieved its prespecified primary end point of ≥50% pain reduction at four weeks after IVIG, achieved in seven of 11 patients (63.6%) in the IVIG group vs zero of 12 in the placebo group (P  = 0.0013). Only two adverse events were reported during the study: one patient in the treatment arm reported a mild "dermatitis psoriasiform," whereas one patient from the placebo group reported a mild "influenza." Conclusions Treatment with IVIG at the dose given was efficacious and safe for patients with DPN resistant to standard therapies. [ABSTRACT FROM AUTHOR]

Background and Aims Cell wall disassembly occurs naturally in plants by the action of several glycosyl-hydrolases during different developmental processes such as lysigenous and constitutive aerenchyma formation in sugarcane roots. Wall degradation has been reported in aerenchyma development in different species, but little is known about the action of glycosyl-hydrolases in this process. Methods In this work, gene expression, protein levels and enzymatic activity of cell wall hydrolases were assessed. Since aerenchyma formation is constitutive in sugarcane roots, they were assessed in segments corresponding to the first 5 cm from the root tip where aerenchyma develops. Key Results Our results indicate that the wall degradation starts with a partial attack on pectins (by acetyl esterases, endopolygalacturonases, β-galactosidases and α-arabinofuranosidases) followed by the action of β-glucan-/callose-hydrolysing enzymes. At the same time, there are modifications in arabinoxylan (by α-arabinofuranosidases), xyloglucan (by XTH), xyloglucan–cellulose interactions (by expansins) and partial hydrolysis of cellulose. Saccharification revealed that access to the cell wall varies among segments, consistent with an increase in recalcitrance and composite formation during aerenchyma development. Conclusion Our findings corroborate the hypothesis that hydrolases are synchronically synthesized, leading to cell wall modifications that are modulated by the fine structure of cell wall polymers during aerenchyma formation in the cortex of sugarcane roots. We forecast the impact of weak lensing (WL) cluster mass calibration on the cosmological constraints from the X-ray-selected galaxy cluster counts in the upcoming eROSITA survey. We employ a prototype cosmology pipeline to analyse mock cluster catalogues. Each cluster is sampled from the mass function in a fiducial cosmology and given an eROSITA count rate and redshift, where count rates are modelled using the eROSITA effective area, a typical exposure time, Poisson noise, and the scatter and form of the observed X-ray luminosity – and temperature–mass–redshift relations. A subset of clusters have mock shear profiles to mimic either those from Dark Energy Survey (DES) and Hyper-Suprime-Cam (HSC) or from the future Euclid and Large Synoptic Survey Telescope (LSST) surveys. Using a count rate selection, we generate a baseline cluster cosmology catalogue that contains 13k clusters over 14 892 deg2 of extragalactic sky. Low-mass groups are excluded using raised count rate thresholds at low redshift. Forecast parameter uncertainties for ΩM, σ8, and w are 0.023 (0.016; 0.014), 0.017 (0.012; 0.010), and 0.085 (0.074; 0.071), respectively, when adopting DES+HSC WL (Euclid ; LSST), while marginalizing over the sum of the neutrino masses. A degeneracy between the distance–redshift relation and the parameters of the observable–mass scaling relation limits the impact of the WL calibration on the w constraints, but with baryonic acoustic oscillation measurements from Dark Energy Spectroscopy Instrument an improved determination of w to 0.043 becomes possible. With Planck cosmic microwave background priors, ΩM (σ8) can be determined to 0.005 (0.007), and the summed neutrino mass limited to ∑ m ν < 0.241 eV (at 95 per cent). If systematics on the group mass scale can be controlled, the eROSITA group and cluster sample with 43k objects and LSST WL could constrain ΩM and σ8 to 0.007 and w to 0.050. [ABSTRACT FROM AUTHOR]

We present the MARD-Y3 catalogue of between 1086 and 2171 galaxy clusters (52 per cent and 65 per cent new) produced using multicomponent matched filter (MCMF) follow-up in 5000 deg2 of DES-Y3 optical data of the ∼20 000 overlapping ROSAT All-Sky Survey source catalogue (2RXS) X-ray sources. Optical counterparts are identified as peaks in galaxy richness as a function of redshift along the line of sight towards each 2RXS source within a search region informed by an X-ray prior. All peaks are assigned a probability f cont of being a random superposition. The clusters lie at 0.02 < z < 1.1 with more than 100 clusters at z > 0.5. Residual contamination is 2.6 per cent and 9.6 per cent for the cuts adopted here. For each cluster we present the optical centre, redshift, rest frame X-ray luminosity, M 500 mass, coincidence with NWAY infrared sources, and estimators of dynamical state. About 2 per cent of MARD-Y3 clusters have multiple possible counterparts, the photo- z 's are high quality with σΔ z /(1 + z) = 0.0046, and ∼1 per cent of clusters exhibit evidence of X-ray luminosity boosting from emission by cluster active galactic nuclei. Comparison with other catalogues (MCXC, RM, SPT-SZ, Planck) is performed to test consistency of richness, luminosity, and mass estimates. We measure the MARD-Y3 X-ray luminosity function and compare it to the expectation from a fiducial cosmology and externally calibrated luminosity- and richness–mass relations. Agreement is good, providing evidence that MARD-Y3 has low contamination and can be understood as a simple two step selection – X-ray and then optical – of an underlying cluster population described by the halo mass function. We use galaxy dynamical information to calibrate the richness–mass scaling relation of a sample of 428 galaxy clusters that are members of the CODEX sample with redshifts up to z ∼ 0.7. These clusters were X-ray selected using the ROSAT All-Sky Survey (RASS) and then cross-matched to associated systems in the redMaPPer (the red sequence Matched-filter Probabilistic Percolation) catalogue from the Sloan Digital Sky Survey. The spectroscopic sample we analyse was obtained in the SPIDERS program and contains ∼7800 red member galaxies. Adopting NFW mass and galaxy density profiles and a broad range of orbital anisotropy profiles, we use the Jeans equation to calculate halo masses. Modelling the scaling relation as |$\lambda \propto \text{A}_{\lambda } {M_{\text{200c}}}^{\text{B}_{\lambda }} ({1+z})^{\gamma _{\lambda }}$|⁠ , we find the parameter constraints |$\text{A}_{\lambda }=38.6^{+3.1}_{-4.1}\pm 3.9$|⁠ , |$\text{B}_{\lambda }=0.99^{+0.06}_{-0.07}\pm 0.04$|⁠ , and |$\gamma _{\lambda }=-1.13^{+0.32}_{-0.34}\pm 0.49$|⁠ , where we present systematic uncertainties as a second component. We find good agreement with previously published mass trends with the exception of those from stacked weak lensing analyses. We note that although the lensing analyses failed to account for the Eddington bias, this is not enough to explain the differences. We suggest that differences in the levels of contamination between pure redMaPPer and RASS + redMaPPer samples could well contribute to these differences. The redshift trend we measure is more negative than but statistically consistent with previous results. We suggest that our measured redshift trend reflects a change in the cluster galaxy red sequence (RS) fraction with redshift, noting that the trend we measure is consistent with but somewhat stronger than an independently measured redshift trend in the RS fraction. Uncertainty in mass–observable scaling relations is currently the limiting factor for galaxy-cluster-based cosmology. Weak gravitational lensing can provide direct mass calibration and reduce the mass uncertainty. We present new ground-based weak lensing observations of 19 South Pole Telescope (SPT) selected clusters at redshifts 0.29 ≤  |$z$|  ≤ 0.61 and combine them with previously reported space-based observations of 13 galaxy clusters at redshifts 0.576 ≤  |$z$|  ≤ 1.132 to constrain the cluster mass scaling relations with the Sunyaev–Zel'dovich effect (SZE), the cluster gas mass M gas and Y X, the product of M gas and X-ray temperature. We extend a previously used framework for the analysis of scaling relations and cosmological constraints obtained from SPT-selected clusters to make use of weak lensing information. We introduce a new approach to estimate the effective average redshift distribution of background galaxies and quantify a number of systematic errors affecting the weak lensing modelling. These errors include a calibration of the bias incurred by fitting a Navarro–Frenk–White profile to the reduced shear using N -body simulations. We blind the analysis to avoid confirmation bias. We are able to limit the systematic uncertainties to 5.6% in cluster mass (68% confidence). Our constraints on the mass–X-ray observable scaling relation parameters are consistent with those obtained by earlier studies and our constraints for the mass–SZE scaling relation are consistent with the simulation-based prior used in the most recent SPT–SZ cosmology analysis. The galaxy phase-space distribution in galaxy clusters provides insights into the formation and evolution of cluster galaxies, and it can also be used to measure cluster mass profiles. We present a dynamical study based on ∼3000 passive, non-emission-line cluster galaxies drawn from 110 galaxy clusters. The galaxy clusters were selected using the Sunyaev–Zel'dovich effect (SZE) in the 2500 deg2 South Pole Telescope (SPT)–SZ survey and cover the redshift range 0.2 < |$z$| < 1.3. We model the clusters using the Jeans equation, while adopting NFW mass profiles and a broad range of velocity dispersion anisotropy profiles. The data prefer velocity dispersion anisotropy profiles that are approximately isotropic near the centre and increasingly radial toward the cluster virial radius, and this is true for all redshifts and masses we study. The pseudo-phase-space density profile of the passive galaxies is consistent with expectations for dark matter particles and subhaloes from cosmological N -body simulations. The dynamical mass constraints are in good agreement with external mass estimates of the SPT cluster sample from either weak lensing, velocity dispersions, or X-ray Y X measurements. However, the dynamical masses are lower (at the 2.2σ level) when compared to the mass calibration favoured when fitting the SPT cluster data to a Λcold dark matter model with external cosmological priors, including cosmic microwave background anisotropy data from Planck. The discrepancy grows with redshift, where in the highest redshift bin the ratio of dynamical to SPT + Planck masses is |$\eta = 0.63^{+0.13}_{-0.08}\pm 0.06$| (statistical and systematic), corresponding to a 2.6σ discrepancy. The development of lysigenous aerenchyma starts with cell expansion and degradation of pectin from the middle lamella, leading to cell wall modification, and culminating with cell separation. Here we report that nutritional starvation of sugarcane induced gene expression along sections of the first 5 cm of the root and between treatments. We selected two candidate genes: a RAV transcription factor, from the ethylene response factors superfamily, and an endopolygalacturonase (EPG), a glycosyl hydrolase related to homogalacturonan hydrolysis from the middle lamella. epg1 and rav1 transcriptional patterns suggest they are essential genes at the initial steps of pectin degradation during aerenchyma development in sugarcane. Due to the high complexity of the sugarcane genome, rav1 and epg1 were sequenced from 17 bacterial artificial chromosome clones containing hom(e)ologous genomic regions, and the sequences were compared with those of Sorghum bicolor. We used one hom(e)olog sequence from each gene for transactivation assays in tobacco. rav1 was shown to bind to the epg1 promoter, repressing β-glucuronidase activity. RAV repression upon epg1 transcription is the first reported link between ethylene regulation and pectin hydrolysis during aerenchyma formation. Background and Aims Aerenchyma develops in different plant organs and leads to the formation of intercellular spaces that can be used by the plant to transport volatile substances. Little is known about the role of cell walls in this process, although the mechanism of aerenchyma formation is known to involve programmed cell death and some cell wall modifications. We assessed the role that cell wall-related mechanisms might play in the formation of aerenchyma in sugarcane roots. Methods Sections of roots (5 cm) were subjected to microtomography analysis. These roots were divided into 1-cm segments and subjected to cell wall fractionation. We performed analyses of monosaccharides, oligosaccharides and lignin and glycome profiling. Sections were visualized by immunofluorescence and immunogold labelling using selected monoclonal antibodies against polysaccharide epitopes according to the glycome profiles. Key Results During aerenchyma formation, gas spaces occupied up to 40 % of the cortex cross-section within the first 5 cm of the root. As some of the cortex cells underwent dissolution of the middle lamellae, leading to cell separation, cell expansion took place along with cell death. Mixed-linkage β-glucan was degraded along with some homogalacturonan and galactan, culminating in the formation of cell wall composites made of xyloglucan, arabinoxylans, cellulose and possibly lignin. Conclusion The composites formed seem to play a role in the physical-chemical properties of the gas chambers, providing mechanical resistance to forces acting upon the root and at the same time decreasing permeability to gases. Recent measurements of the cosmic microwave background (CMB) by the Planck Collaboration have produced arguably the most powerful observational evidence in support of the standard model of cosmology, i.e. the spatially flat ΛCDM paradigm. In this work, we perform model selection tests to examine whether the base CMB temperature and large scale polarization anisotropy data from Planck 2015 (P15; Planck Collaboration XIII) prefer any of eight commonly used one-parameter model extensions with respect to flat ΛCDM. We find a clear preference for models with free curvature, ΩK, or free amplitude of the CMB lensing potential, AL. We also further develop statistical tools to measure tension between data sets. We use a Gaussianization scheme to compute tensions directly from the posterior samples using an entropy-based method, the surprise, as well as a calibrated evidence ratio presented here for the first time. We then proceed to investigate the consistency between the base P15 CMB data and six other CMB and distance data sets. In flat ΛCDM we find a 4.8σ tension between the base P15 CMB data and a distance ladder measurement, whereas the former are consistent with the other data sets. In the curved ΛCDM model we find significant tensions in most of the cases, arising from the well-known low power of the low-l multipoles of the CMB data. In the flat ΛCDM+AL model, however, all data sets are consistent with the base P15 CMB observations except for the CMB lensing measurement, which remains in significant tension. This tension is driven by the increased power of the CMB lensing potential derived from the base P15 CMB constraints in both models, pointing at either potentially unresolved systematic effects or the need for new physics beyond the standard flat ΛCDM model. Alternating hemiplegia of childhood is a rare disorder caused by de novo mutations in the ATP1A3 gene, expressed in neurons and cardiomyocytes. As affected individuals may survive into adulthood, we use the term 'alternating hemiplegia'. The disorder is characterized by early-onset, recurrent, often alternating, hemiplegic episodes; seizures and non-paroxysmal neurological features also occur. Dysautonomia may occur during hemiplegia or in isolation. Premature mortality can occur in this patient group and is not fully explained. Preventable cardiorespiratory arrest from underlying cardiac dysrhythmia may be a cause. We analysed ECG recordings of 52 patients with alternating hemiplegia from nine countries: all had whole-exome, whole-genome, or direct Sanger sequencing of ATP1A3. Data on autonomic dysfunction, cardiac symptoms, medication, and family history of cardiac disease or sudden death were collected. All had 12-lead electrocardiogram recordings available for cardiac axis, cardiac interval, repolarization pattern, and J-point analysis. Where available, historical and prolonged single-lead electrocardiogram recordings during electrocardiogram-videotelemetry were analysed. Half the cohort (26/52) had resting 12-lead electrocardiogram abnormalities: 25/26 had repolarization (T wave) abnormalities. These abnormalities were significantly more common in people with alternating hemiplegia than in an age-matched disease control group of 52 people with epilepsy. The average corrected QT interval was significantly shorter in people with alternating hemiplegia than in the disease control group. J wave or J-point changes were seen in six people with alternating hemiplegia. Over half the affected cohort (28/52) had intraventricular conduction delay, or incomplete right bundle branch block, a much higher proportion than in the normal population or disease control cohort (P = 0.0164). Abnormalities in alternating hemiplegia were more common in those ≥16 years old, compared with those <16 (P = 0.0095), even with a specific mutation (p.D801N; P = 0.045). Dynamic, beat-to-beat or electrocardiogram-to-electrocardiogram, changes were noted, suggesting the prevalence of abnormalities was underestimated. Electrocardiogram changes occurred independently of seizures or plegic episodes. Electrocardiogram abnormalities are common in alternating hemiplegia, have characteristics reflecting those of inherited cardiac channelopathies and most likely amount to impaired repolarization reserve. The dynamic electrocardiogram and neurological features point to periodic systemic decompensation in ATP1A3-expressing organs. Cardiac dysfunction may account for some of the unexplained premature mortality of alternating hemiplegia. Phytopathogens can co-opt plant primary metabolism to enhance pathogenesis and pathogen nutrition. In witches’ broom disease of cacao, sensing and modulation of compartmentalized carbon availability can also temporally regulate disease development.Witches’ broom disease (WBD) of cacao differs from other typical hemibiotrophic plant diseases by its unusually long biotrophic phase. Plant carbon sources have been proposed to regulate WBD developmental transitions; however, nothing is known about their availability at the plant–fungus interface, the apoplastic fluid of cacao. Data are provided supporting a role for the dynamics of soluble carbon in the apoplastic fluid in prompting the end of the biotrophic phase of infection. Carbon depletion and the consequent fungal sensing of starvation were identified as key signalling factors at the apoplast. MpNEP2, a fungal effector of host necrosis, was found to be up-regulated in an autophagic-like response to carbon starvation in vitro. In addition, the in vivo artificial manipulation of carbon availability in the apoplastic fluid considerably modulated both its expression and plant necrosis rate. Strikingly, infected cacao tissues accumulated intracellular hexoses, and showed stunted photosynthesis and the up-regulation of senescence markers immediately prior to the transition to the necrotrophic phase. These opposite findings of carbon depletion and accumulation in different host cell compartments are discussed within the frame of WBD development. A model is suggested to explain phase transition as a synergic outcome of fungal-related factors released upon sensing of extracellular carbon starvation, and an early senescence of infected tissues probably triggered by intracellular sugar accumulation. In spite of extensive criticisms, war metaphors are still widespread in medical discourse. In the domain of public health analogies between war and infectious diseases are rooted in the similar impacts they can have on political institutions and communities. This similarity has been emphasized by the recent trend of addressing infectious disease from the point of view of national security. Nevertheless, it is here argued that the analogy cannot be used to model normative principles for treating carriers of contagious diseases after the principles of ius in bello. Yet, the analogy may provide some useful practical and theoretical insights in tackling the growing problem of limiting the spread of drug-resistant pathogens: an issue that generates increasing worries and raises difficult ethical and political questions of rights, responsibilities and obligations. Head and neck cancer represents 3.3% of all new malignancies and 2.0% of cancer deaths in the USA, the majority of which are squamous in origin. The overall 5 year survival is 60% and worsens with increasing stage at diagnosis. Thus, novel biomarkers for early detection of squamous cell carcinoma of the head and neck (SCCHN) are needed. MicroRNA-137 (miR-137) plays a role in cell cycle control and seems to undergo promoter methylation in oral squamous cell carcinoma tissue. The main objectives of this study were to ascertain whether miR-137 promoter methylation is detectable in oral rinse samples, assess its association with SCCHN and identify potential risk factors for its occurrence. Oral rinse samples were collected from 99 SCCHN patients with no prior history of cancer and 99 cancer-free controls, frequency matched on gender; tumor tissue for 64 patients was also tested. Methylation of the miR-137 promoter, assessed using methylation-specific polymerase chain reaction, was detected in 21.2% oral rinses from SCCHN patients and 3.0% from controls [odds ratio (OR) = 4.80, 95% confidence interval (CI): 1.23–18.82]. Among cases, promoter methylation of miR-137 was associated with female gender (OR = 5.30, 95% CI: 1.20–23.44) and inversely associated with body mass index (BMI) (OR = 0.88, 95% CI: 0.77–0.99). Promoter methylation of miR-137 appears to be a relatively frequently detected event in oral rinse of SCCHN patients and may have future utility as a biomarker in DNA methylation panels. Treatment of human head and neck squamous cell carcinoma (HNSCC) cell lines with guggulsterone, a widely available, well-tolerated nutraceutical, demonstrated dose-dependent decreases in cell viability with EC50s ranging from 5 to 8 μM. Guggulsterone induced apoptosis and cell cycle arrest, inhibited invasion and enhanced the efficacy of erlotinib, cetuximab and cisplatin in HNSCC cell lines. Guggulsterone induced decreased expression of both phosphotyrosine and total signal transducer and activator of transcription (STAT)-3, which contributed to guggulsterone's growth inhibitory effect. Hypoxia-inducible factor (HIF)-1α was also decreased in response to guggulsterone treatment. In a xenograft model of HNSCC, guggulsterone treatment resulted in increased apoptosis and decreased expression of STAT3. In vivo treatment with a guggulsterone-containing natural product, Guggulipid, resulted in decreased rates of tumor growth and enhancement of cetuximab's activity. Our results suggest that guggulsterone-mediated inhibition of STAT3 and HIF-1α provide a biologic rationale for further clinical investigation of this compound in the treatment of HNSCC. Background: Dysregulation of signal transducers and activators of transcription (STATs) is associated with many cancers, but no role of STAT1 in human tumor progression has been demonstrated. Methods: We compared STAT1 protein expression in squamous cell carcinoma of the head and neck (SCCHN) tumors (n = 28) and normal oropharyngeal mucosa samples (n = 10) from patients without cancer as assessed by immunoblotting. Stable clones were established from SCCHN 1483 cells that were transfected with a STAT1 expression construct; cell growth and cisplatin-induced apoptosis of the clones and vector control-transfected 1483 cells were compared using trypan blue exclusion and Annexin V staining and expression of the cyclin-dependent kinase inhibitor p21 was assayed by immunoblotting. The growth of STAT1-overexpressing SCCHN 1483 xenograft tumors was compared with that of xenograft tumors derived from cells transfected with vector control DNA. DNA from SCCHN tumors (n = 16) and paired peripheral blood lymphocytes were analyzed for STAT1 mutations and promoter methylation using methylation-specific polymerase chain reaction and bisulfite sequencing. SCCHN cell lines (PCI-15b, 1483, and UM-22B) were treated with the demethylating agent azacytidine alone or in combination with the cytotoxic drug cisplatin, and expression of STAT1 and p21 were monitored by immunoblotting. All statistical tests were two-sided. Results: STAT1 levels were statistically significantly lower in the SCCHN tumors than normal mucosa (median = 0.8 relative units versus 2.4, difference = 1.6, 95% confidence interval [CI] = 1.3 to 2.0, P<.001). Overexpression of STAT1 abrogated the growth of SCCHN cells and xenograft tumors and increased p21 expression. STAT1 expression levels of the tumors with STAT1 promoter methylation (n = 12) were lower than those of tumors (n = 4) without promoter methylation of STAT1 (P = .008). Azacytidine treatment increased expression of STAT1 and p21 in SCCHN cell lines and increased apoptosis in cisplatin-treated 1483 cells compared with cisplatin treatment alone (mean = 61.3% versus 25.8%, difference = 35.5%, 95% CI = 24.5% to 43.4%; P = .028). Cruciferous vegetable-derived isothiocyanates (ITCs; chemical structure: R-N=C=S) are highly effective in affording protection against chemically induced cancers in animal models. Here, we studied the antitumor effects of benzyl isothiocyanate (BITC; Ph-CH2-N=C=S), the predominant ITC compound in broccoli, on head and neck squamous cell carcinoma (HNSCC) cell lines. Proliferation, apoptosis and immunoblotting assays were used to determine the effects and mechanism of several ITCs on HNSCC cells. The IC50 for BITC (24 h treatment) in two of the HNSCC cell lines was ∼22 and 17 µM, respectively. Interestingly, phenyl isothiocyanate (PITC; Ph-N=C=S), which is a close structural analog of BITC but lacks a -CH2- spacer that links the aromatic ring to N=C=S moiety, did not result in significant killing of the HNSCC cells in this dose range. BITC (but not PITC) caused activation of caspase 3 and PARP cleavage. Within 20 min of treatment, BITC (but not PITC) induced a rapid activation of p38 MAPK. In addition, BITC (but not PITC) treatment resulted in the activation of p44/42 MAPK. Co-treatment with a specific p38 MAPK inhibitor, SB203580, or an inhibitor of the MEK/MAPK pathway, U0126, partially rescued cells from BITC-induced killing. Our results show that minor structural differences in ITCs can be crucial for the antiproliferative activity of ITCs and that BITC may be a promising chemopreventive as well as therapeutic agent in HNSCC. Background: Gastrin-releasing peptide receptor (GRPR)-mediated autocrine growth appears to be an early marker of susceptibility to tobacco-related lung cancers. Because expression of GRPR, however, has not been reported in squamous cell carcinoma of the head and neck (SCCHN), we investigated its expression and that of its ligand GRP in normal mucosa and SCCHN tissues and the involvement of these proteins in the proliferation of SCCHN cells.Methods: We assessed GRPR messenger RNA (mRNA) expression in specimens from 25 patients with SCCHN, six control noncancer patients, and 14 SCCHN cell lines by use of quantitative reverse transcriptase-polymerase chain reaction. We used neutralizing GRP monoclonal antibody 2A11 to block the GRP-GRPR interaction in SCCHN cell lines and xenografts and assessed the antibody's effect on proliferation by counting cultured cells or measuring xenograft tumor volume in vivo. All statistical tests were two-sided.Results: Tumor and mucosa tissues, respectively, from SCCHN patients expressed sixfold and fourfold higher levels of GRPR mRNA than normal mucosa tissue from noncancer patients (P<.001). The levels of GRPR expression in the tumor and adjacent normal epithelium of individual patients with SCCHN were correlated (r =.652; P =.001), suggesting that increased GRPR expression is an early event in SCCHN formation. SCCHN cells expressed fivefold higher levels of GRPR mRNA than did cultured normal mucosal epithelial cells (P =.005). GRP stimulated proliferation of SCCHN cells in a dose-dependent fashion (P =.006). Neutralizing GRP monoclonal antibody 2A11 inhibited SCCHN cell proliferation in vitro and in vivo. Median survival was 54 months in patients with higher levels of GRPR mRNA and was not reached in those with lower levels.Conclusions: GRP and GRPR appear to participate in an autocrine regulatory pathway in SCCHN. Recent outbreaks of cryptosporidiosis from contaminated water supplies have led to a need for the detection of Cryptosporidium oocysts from various hosts and contaminating sources. The presence of nonpathogenic species or strains of Cryptosporidium is important for diagnostic purposes as there is a potential for false-positive detection of pathogenic parasites. The present review focuses on phenotypic differences and recent advances in genotypic analyses of the genus Cryptosporidium with an emphasis on detecting various isolates and identifying differences in Cryptosporidium parvum and other species in this genus. The information currently available demonstrates important patterns in DNA sequences of Cryptosporidium, and our understanding of macro- and microevolutionary patterns has increased in recent years. However, current knowledge of Cryptosporidium genetic diversity is far from complete, and the large amount of both phenotypic and genotypic data has led to problems in our understanding of the systematics of this genus. Background: The most accurate predictor of disease recurrence in patients treated for head and neck squamous cell carcinoma is, at present, the extent of regional lymph node metastasis. Since elevated levels of epidermal growth factor receptor (EGFR) and of its ligand, transforming growth factor-alpha (TGF-alpha), have been detected in primary tumors of patients with head and neck squamous cell carcinoma, we determined whether tumor levels of these proteins were of prognostic importance.Methods: Monoclonal antibodies specific for EGFR and TGF-alpha were used for immunohistochemical detection of each protein in tissue sections of primary tumors from 91 patients who were treated by surgical resection. Levels of immunoreactive EGFR and TGF-alpha were quantified by use of a computerized image analysis system and were normalized to appropriate standards. The logrank test and proportional hazards regression analysis were used to calculate the probability that EGFR and TGF-alpha levels were associated with disease-free survival (i.e., no recurrence of cancer) and cause-specific survival (i.e., patients do not die of their disease). All P values were two-sided.Results: When tumor levels of EGFR or TGF-alpha were analyzed as continuous variables, disease-free survival and cause-specific survival were reduced among patients with higher levels of EGFR (both P = .0001) or TGF-alpha (both P = .0001). In a multivariate analysis, tumor site, tumor level of EGFR, and tumor level of TGF-alpha were statistically significant predictors of disease-free survival; in a similar analysis, regional lymph node stage and tumor levels of EGFR and of TGF-alpha were significant predictors of cause-specific survival.Conclusion: Quantitation of EGFR and TGF-alpha protein levels in primary head and neck squamous cell carcinomas may be useful in identifying subgroups of patients at high risk of tumor recurrence and in guiding therapy. Amplified fragment length polymorphism (AFLP) is a novel molecular fingerprinting technique that can be applied to DNAs of any source or complexity. Total genomic DNA is digested using two restriction enzymes. Double-stranded nucleotide adapters are ligated to the DNA fragments to serve as primer binding sites for PCR amplification. Primers complementary to the adapter and restriction site sequence, with additional nucleotides at the 3'-end, are used as selective agents to amplify a subset of ligated fragments. Polymorphisms are identified by the presence or absence of DNA fragments following analysis on polyacrylamide gels. This technique has been extensively used with plant DNA for the development of high-resolution genetic maps and for the positional cloning of genes of interest. However, its application is rapidly expanding in bacteria and higher eukaryotes for determining genetic relationships and for epidemiological typing. From an autopsy series of 346 patients who died of acute myocardial infarction, we selected 36 cases for whom echocardiographic data preceding death were available: 17 cases died from a rupture of the left ventricular free wall (group A) and 19 from pump failure (group B). Our aim was to investigate whether any echocardiographic parameter could predict the final event. The total wall motion score, regional wall motion score index and percent of abnormally contracting myocardium were calculated. Diastolic and systolic volumes, ejection fraction and the eccentricity index, as a rough indicator of the left ventricular shape, were also estimated. Interventricular septum and posterior wall thicknesses were also measured. All measured parameters were similar in both groups except posterior wall thickness. This prospective, randomized trial was designed to determine the efficacy and mechanism of action of topical mouthwash versus parenterally administered perioperative prophylactic antibiotics in contaminated head and neck surgery. Patients were randomly assigned to 1 of 4 treatment groups: 1 day of parenteral clindamycin (standard prophylaxis), 1 day of topical clindamycin, 5 days of topical clindamycin, or 1 day of topical amoxicillin plus clavulanate/ticarcillin plus clavulanate. Patients who received the latter regimen had fewer bacteria postoperatively compared with the other 3 treatment groups. The number of gram-negative aerobic bacilli on postoperative oral cavity cultures was increased in all 3 clindamycin groups but not in the amoxicillin plus clavulanate/ticarcillin plus clavulanate group. Parenteral clindamycin appears to exert its effect by being in the neck tissues at the time of surgery; however, all 3 topical regimens were more effective at reducing the number of bacteria in the neck viscera. Background: Squamous cell carcinomas of the head and neck (SCCHN), unlike normal mucosal squamous epithelial cells, overexpress epidermal growth factor receptor (EGFR) messenger RNA and protein. EGFR protein is required to sustain the proliferation of SCCHN cells in vitro. To determine whether EGFR expression contributes to tumor growth, we investigated the effect of suppressing EGFR expression in tumor xenografts through in situ expression of antisense oligonucleotides.Methods: Intratumoral cationic liposome-mediated gene transfer was used to deliver plasmids capable of expressing sense or antisense EGFR sequences into human head and neck tumors, which were grown as subcutaneous xenografts in nude mice. The oligonucleotides were expressed under the control of the U6 RNA promoter.Results: Direct inoculation of the EGFR antisense (but not the corresponding sense) plasmid construct into established SCCHN xenografts resulted in inhibition of tumor growth, suppression of EGFR protein expression, and an increased rate of apoptosis (programmed cell death). Sustained antitumor effects were observed for up to 2 weeks after the treatments were discontinued.Conclusion: These results suggest that interference with EGFR expression, using an antisense-based gene therapy approach, may be an effective means of treating EGFR-overexpressing tumors, including SCCHN. This study was conducted with the objective of evaluating the performance, carcass and meat characteristics of castrated and intact Dorper male lambs slaughtered at three different body condition scores. Thirty-five lambs, approximately 3 months old, 21.6 ± 4.0 kg of body weight and 3.1 ± 0.5 of body condition score (BCS), were used. The BCS scale used varied from 1.0 (very thin) to 5.0 (very fat). Four weeks prior to the experiment, 17 lambs were castrated. Lambs were further divided into three groups, according to the criteria for slaughter: BCS of 3.5, 4.0 and 4.5. All animals received the same diet. BCS were achieved after 27, 84 and 130 days in feedlot, respectively. There were no interactions (P > 0.05) between sexual conditions and slaughter criteria. Average daily gain of weight (ADG) was different (P < 0.05) for the three BCS (average of 0.263). However, final body weight (28.6 ± 1.7; 42.7 ± 1.8 and 56.8 ± 1.7 kg) and back fat thickness (0.5 ± 0.5; 4.2 ± 0.5 and 7.3 ± 0.5 mm) were different (P < 0.05) among BCS criteria, panelists found that meats from lambs slaughtered with greater fat content were less (P < 0.05) desirable. We can conclude by the performance and meat results that it is better to slaughter Dorper lambs with BCS with no more than 4.0, or 4.2 mm of back fat cover, and for the best acceptability of the meat the lambs should be castrated.
