2. Laser-upgraded coal tar for smart pavements in road and bridge monitoring applications.

The implementation of an intelligent road network system requires many sensors for acquiring data from roads, bridges, and vehicles, thereby enabling comprehensive monitoring and regulation of road networks. Given this large number of required sensors, the sensors must be cost-effective, dependable, and environmentally friendly. Here, we show a laser upgrading strategy for coal tar, a low-value byproduct of coal distillation, to manufacture flexible strain-gauge sensors with maximum gauge factors of 15.20 and 254.17 for tension and compression respectively. Furthermore, we completely designed the supporting processes of sensor placement, data acquisition, processing, wireless communication, and information decoding to demonstrate the application of our sensors in traffic and bridge vibration monitoring. Our novel strategy of using lasers to upgrade coal tar for use as a sensor not only achieves the goal of turning waste into a resource but also provides an approach to satisfy large-scale application requirements for enabling intelligent road networks. 3. Exceptional points enhance sensing in silicon micromechanical resonators.

Exceptional points (EPs) have recently emerged as a new method for engineering the response of open physical systems, that is, systems that interact with the environment. The systems at the EPs exhibit a strong response to a small perturbation. Here, we show a method by which the sensitivity of silicon resonant sensors can be enhanced when operated at EPs. In our experiments, we use a pair of mechanically coupled silicon micromechanical resonators constituting a parity–time (PT)-symmetric dimer. Small perturbations introduced on the mechanically coupled spring cause the frequency to split from the EPs into the PT-symmetric regime without broadening the two spectrum linewidths, and this frequency splitting scales with the square root of the perturbation strength. The overall signal-to-noise ratio is still greatly enhanced, although the measured noise spectral density of the EP sensing scheme has a slight increase comparable to the traditional counterpart. 4. Construction of an oxidative stress-related lncRNAs signature to predict prognosis and the immune response in gastric cancer.

Oxidative stress, as a characteristic of cellular aerobic metabolism, plays a crucial regulatory role in the development and metastasis of gastric cancer (GC). Long noncoding RNAs (lncRNAs) are important regulators in GC development. However, research on the prognostic patterns of oxidative stress-related lncRNAs (OSRLs) and their functions in the immune microenvironment is currently insufficient. We identified the OSRLs signature (DIP2A-IT1, DUXAP8, TP53TG1, SNHG5, AC091057.1, AL355001.1, ARRDC1-AS1, and COLCA1) from 185 oxidative stress-related genes in The Cancer Genome Atlas (TCGA) cohort via random survival forest and Cox analyses, and the results were subsequently validated in the Gene Expression Omnibus (GEO) dataset. The patients were divided into high- and low-risk groups by the risk score of the OSRLs signature. Longer overall survival was detected in the low-risk group than in the high-risk group in both the TCGA cohort (P < 0. 001, HR = 0.43, 95% CI 0.31–0.62) and the GEO cohort (P = 0.014, HR = 0.67, 95% CI 0.48–0.93). Next, multivariate Cox analysis identified that the risk model was an independent prognostic characteristic (HR > 1, P = 0.005), and time-dependent receiver operating characteristic (ROC) curve analysis and nomogram analysis were utilized to evaluate the predictive ability of the risk model. Next, gene set enrichment analysis revealed that the immune-related pathway, Wnt/ β -catenin signature, mammalian target of rapamycin complex 1 signature, and cytokine‒cytokine receptor interaction was enriched. High-risk patients were more responsive to CD200, TNFSF4, TNFSF9, and BTNL2 immune checkpoint blockade. The results of qRT‒PCR further proved the accuracy of our bioinformatic analysis. Overall, our study identified a novel OSRLs signature that can serve as a promising biomarker and prognostic indicator, which provides a personalized predictive approach for patient prognosis evaluation and treatment. 5. Pan-cancer analysis reveals the prognostic and immunotherapeutic value of cytoskeleton-associated protein 2-like.

Cytoskeleton-associated protein 2-like (CKAP2L), a cell cycle-related protein, is correlated to tumor progression in some tumors. But there were no pan-cancer studies on CKAP2L, and its role in cancer immunotherapy is also unclear. The expression levels, expression activity, genomic alterations, DNA methylation and functions of CKAP2L in various tumors, as well as the associations between CKAP2L expression and patient prognosis, chemotherapy sensitivity, and tumor immune microenvironment, were all analyzed in a comprehensive pan-cancer analysis of CKAP2L by various databases, analysis websites, and R software. The experiments were also conducted to verify the analysis results. In the majority of cancers, CKAP2L expression and activity were markedly elevated. Elevated CKAP2L expression led to poor prognostic outcomes in patients, and is an independent risk factor for most tumors. Elevated CKAP2L causes decreased sensitivity to chemotherapeutic agents. Knockdown of CKAP2L significantly inhibited the proliferation and metastasis capacity of the KIRC cell lines and resulted in cell cycle G2/M arrest. In addition, CKAP2L was closely related to immune subtypes, immune cell infiltration, immunomodulators and immunotherapy markers (TMB, MSI), patients with high CKAP2L expression were more sensitive to immunotherapy in the IMvigor210 cohort. The results indicate that CKAP2L is a pro-cancer gene that serves as a potential biomarker for predicting patient outcomes. By inducing cells to transition from the G2 phase to the M phase, CKAP2L may promote cell proliferation and metastasis. The stability, low cost, non-toxicity, excellent pigment, and optical and electrical properties of TiO
2 have prompted its wide application. Moreover, the microwave is a new green heating technology that can promote crystal nucleation and growth; therefore, researchers have applied microwave heating to prepare rutile TiO2 . In this study, the phase transformation kinetics and enhancement mechanism of rutile TiO2 prepared by microwave-enhanced calcination of metatitanic acid-doped TiC were investigated. Microwave radiation and TiC doping effectively lowered the phase transition temperature point (450 °C) for the anatase-to-rutile transformation (ART) with a reduction of 600 °C under different calcination conditions. Subsequently, the isothermal phase transformation kinetics of the ART process of the calcination of metatitanic acid was studied by the Johnson–Mehl–Avrami–Kolmogorov model. The ART activation energy was 40.81 kJ/mol, which was only 0.43–0.0028 times that of commonly reported values. Moreover, the preparation process could generate a large number of oxygen vacancies and improve the optical properties of the products. - Abstract
Heat shock proteins (HSP) are molecular chaperones involved in many normal cellular processes and environmental stresses. At the genome-wide level, there were no reports on the diversity and phylogeny of the heat shock protein family in Procecidochares utilis. In this study, 43 HSPs were identified from the genome of P. utilis, including 12 small heat shock proteins (sHSPs), 23 heat shock protein 40 (DNAJs), 6 heat shock protein 70 (HSP70s), and 2 heat shock protein 90 (HSP90s). The characteristics of these candidates HSP genes were analyzed by BLAST, and then phylogenetic analysis was carried out. Quantitative real-time PCR (qRT-PCR) was used to analyze the spatiotemporal expression patterns of sHSPs and HSP70s in P. utilis after temperature stress. Results showed that most sHSPs could be induced under heat stress during the adult stage of P. utilis, while a few HSP70s could be induced at the larval stage. This study provides an information framework for the HSP family of P. utilis. Phosphogypsum (PG) is a solid waste product generated during wet-process phosphoric acid production. Various impurities considerably reduce the purity, whiteness, and application range of PG. This article analyzes the physical properties of PG in detail and systematically examines the content and distribution of impurities. Based on the obtained results, a simple process for the efficient removal of almost all impurities in the PG is proposed. The purity and whiteness of the purified gypsum (CaSO4) significantly increased to 99% and 92%, respectively. The migration of impurities and the material balance of this process were then analyzed. Most importantly, the purified gypsum showed high CO2 sequestration efficiency for CO2 mineral sequestration, through which a high value-added CaCO3 product was obtained. [ABSTRACT FROM AUTHOR]

Tetrahydroisoquinoline alkaloids (THIQAs) are ubiquitous compounds with important pharmaceutical and biological activity. Their key N-heterocyclic structural motifs are synthesised via Pictet–Spengler (P–S) reaction by norcoclaurine synthases (NCS) in plants. The synthesis of 1-aryl-tetrahydroisoquinoline alkaloids has attracted increasing attention due to their antitumor and antivirus activities. Herein, the L68T/M97V mutant of NCS from Thalictrum flavum with improved activity was developed by semi-rational design. This mutant not only showed higher catalytic performance (> 96% conversion) toward benzaldehyde and dopamine over the wild-type enzyme, but also catalysed the P–S reaction of the bulky substrate 4-biphenylaldehyde and dopamine with high conversion (> 99%) for the effective synthesis of 1-aryl-THIQA. In terms of stereoselectivity, all products synthesised by the L68T/M97V mutant showed high optical purity (92–99% enantiomeric excess). [ABSTRACT FROM AUTHOR]

10. Highly efficient generation of blastocyst-like structures from spliceosomes-repressed mouse totipotent blastomere-like cells.

- Abstract
Mammalian embryogenesis begins with a totipotent zygote. Blastocyst-like structures can be captured by aggregated cells with extended pluripotent properties in a three-dimensional (3D) culture system. However, the efficiency of generating blastoids is low, and it remains unclear whether other reported totipotent-like stem cells retain a similar capacity. In this study, we demonstrated that spliceosomal repression-induced totipotent blastomere-like cells (TBLCs) form blastocyst-like structures within around 80% of all microwells. In addition, we generated blastoids initiating from a single TBLC. TBLC-blastoids express specific markers of constituent cell lineages of a blastocyst and resemble blastocyst in cell-lineage allocation. Moreover, single-cell RNA sequencing revealed that TBLC-blastoids share a similar transcriptional profile to natural embryos, albeit composed of fewer primitive endoderm-like cells. Furthermore, TBLC-blastoids can develop beyond the implantation stage in vitro and induce decidualization in vivo. 11. Multiple habitat templates for phytoplankton indicators within the functional group system.

The functional group (FG) system sensu Colin S. Reynolds of freshwater phytoplankton has been widely applied in many aspects of freshwater ecological assessments. However, there is no current consensus as to how and to why some functional coda shows multiple habitat templates in different water bodies. To test whether there are multiple habitat templates for some functional coda in the FG system, we used two mesotrophic deep reservoirs in China as examples, analysing phytoplankton taxonomic and environmental data collected in 4 seasons over two years. Results showed that a large proportion of the indicators had similar habitat templates within the FG system, whilst some species had multiple or different habitat templates according to their niche differentiation. The random forest (RF) model filtered a minimum set of environmental conditions and provided two potential habitat templates for Oocystis lacustris. One explanatory habitat template of clear water provided by the FG system was verified in the Nanwan Reservoir, and the other habitat template of high dissolved inorganic nitrogen (DIN) was found in the Nianyushan Reservoir. 12. New seco-anthraquinone glucoside from the roots of Rumex crispus.

A new seco-anthraquinone, crispuside A (1), and three new 3,4-dihydronaphthalen-1(2H)-ones, napthalenones A-C (2–4), were isolated from the roots of Rumex crispus L., along with 10 known anthraquinones (6–14) and naphthalenone (5). Their structures were fully determined by extensive spectroscopic analyses, including ECD, and X-ray crystallography in case of compound 5, whose absolute configuration was determined for the first time. The isolates 1, 6–14 were evaluated for their anti-inflammatory and anti-fungal activity against three skin fungi, e.g., Epidermophyton floccosum, Trichophyton rubrum, and Microsporum gypseum. Most of the isolates showed weak anti-fungal and anti-inflammatory activity. Only compound 9 exhibited obvious anti-fungal activity against E. floccosum (MIC
50 = 2.467 ± 0.03 μM) and M. gypseum (MIC50 = 4.673 ± 0.077 μM), while the MIC50 values of the positive control terbinafine were 1.287 ± 0.012 and 0.077 ± 0.00258 μM, respectively. 6 -methyladenosine (m6 A) modification of mRNAs affects many biological processes. However, the function of m6 A in plant photosynthesis remains unknown. Here, we demonstrate that m6 A modification is crucial for photosynthesis during photodamage caused by high light stress in plants. The m6 A modification levels of numerous photosynthesis-related transcripts are changed after high light stress. We determine that the Arabidopsis m6 A writer VIRILIZER (VIR) positively regulates photosynthesis, as its genetic inactivation drastically lowers photosynthetic activity and photosystem protein abundance under high light conditions. The m6 A levels of numerous photosynthesis-related transcripts decrease in vir mutants, extensively reducing their transcript and translation levels, as revealed by multi-omics analyses. We demonstrate that VIR associates with the transcripts of genes encoding proteins with functions related to photoprotection (such as HHL1, MPH1, and STN8) and their regulatory proteins (such as regulators of transcript stability and translation), promoting their m6 A modification and maintaining their stability and translation efficiency. This study thus reveals an important mechanism for m6 A-dependent maintenance of photosynthetic efficiency in plants under high light stress conditions. Efficient photoprotection is vital for the maintenance of photosynthetic efficiency during photodamage. As the main immune checkpoint, PD-L1-PD-1 interaction plays a critical role in the dysregulation of effector T cells, which contributes to the failure of Chimeric Antigen Receptor T-cell (CAR-T) and other immunotherapies. Presently, most research focuses on the extracellular function of PD-L1. Membrane PD-L1 can interact with its receptor PD-1 and decrease T cell-induced cancer immunity. However, the function of PD-L1 in cancer cells is still unclear. Recent studies have shown the separated clinical significance of PD-L1 expression in various cancer types, showing the complexity of PD-L1 in cancer cell regulation. As a novel regulatory pathway, the nuclear translocation of PD-L1 in cancer cells receives more attention. Results of these preclinical studies demonstrated that nuclear PD-L1 has an essential role in cancer development and other immune checkpoint molecules transcription. Herein, we summarized the mechanisms involved in PD-L1 nuclear transportation and identify the key regulatory factors in this process. Furthermore, we also summarize the function of nuclear PD-L1 in cancer immunity. These findings suggested the novel PD-L1 regulation in cancer development, which showed that nuclear PD-L1 is a potential therapeutic target in future cancer therapy. [ABSTRACT FROM AUTHOR]

The present work comprehensively investigated the responses of submerged Ceratophyllum demersum to exudates and extracts of toxic Microcystis aeruginosa at the exponential (EU, ET) or declining phase (DU, DT). The highest amount of MC-LR was detected in medium with DT, the value of which reached 6.36 μg/l, and C. demersum exhibited the highest absorption of MC-LR with a value of 1.73 ng/g FW upon exposure to DU. Significant negative effects on C. demersum was detected under the treatment with EU of toxic M. aeruginosa, expressed with the reduction of freshweight, Chla/Chlb, and the Superoxide Dismutase (SOD) activity. Meanwhile, increased carotenoid/total chlorophyll ratio might be employed by submerged C. demersum as the defense strategy to EU of toxic M. aeruginosa. In addition, under EU treatment, the microorganisms Flavobacterium Bergey, and Chryseobacterium Vandamme, which exert negative effects on aquatic organisms, increased, whereas the Rhizobacter Goto & Kuwata, Limnothrix Meffert, and Pseudanabaena Lauterborn, which are beneficial to the growth of the plants, were lower. The research highlighted the significance of toxic cyanobacterial bloom development and different growth phases on submerged macrophytes, providing more insights for the allelopathic interaction between submerged macrophytes and toxic cyanobacteria in natural freshwater ecosystems. Myocardial ischemia/reperfusion (MI/R) injury is a pathological process that seriously affects the health of patients with coronary artery disease. Long non-coding RNAs (lncRNAs) represents a new class of regulators of diverse biological processes and disease conditions, the study aims to discover the pivotal lncRNA in MI/R injury. The microarray screening identifies a down-regulated heart-enriched lncRNA-CIRPIL (Cardiac ischemia reperfusion associated p53 interacting lncRNA, lncCIRPIL) from the hearts of I/R mice. LncCIRPIL inhibits apoptosis of cultured cardiomyocytes exposed to anoxia/reoxygenation (A/R). Cardiac-specific transgenic overexpression of lncCIRPIL alleviates I/R injury in mice, while knockout of lncCIRPIL exacerbates cardiac I/R injury. LncCIRPIL locates in the cytoplasm and physically interacts with p53, which leads to the cytoplasmic sequestration and the acceleration of ubiquitin-mediated degradation of p53 triggered by E3 ligases CHIP, COP1 and MDM2. p53 overexpression abrogates the protective effects of lncCIRPIL. Notably, the human fragment of conserved lncCIRPIL mimics the protective effects of the full-length lncCIRPIL on cultured human AC16 cells. Collectively, lncCIRPIL exerts its cardioprotective action via sequestering p53 in the cytoplasm and facilitating its ubiquitin-mediated degradation. The study highlights a unique mechanism in p53 signal pathway and broadens our understanding of the molecular mechanisms of MI/R injury. Overexpression of long non-coding RNA, lncCIRPIL protects the heart from ischemia/reperfusion (I/R) injury. LncCIRPIL binds and sequesters p53 in the cytoplasm to exert the cardioprotective action. Granulocyte colony-stimulating factor (G-CSF) has been widely used to mobilize bone marrow hematopoietic stem/progenitor cells for transplantation in the treatment of hematological malignancies for decades. Additionally, G-CSF is also accepted as an essential mediator in immune regulation, leading to reduced graft-versus-host disease following transplantation. Despite the important clinical roles of G-CSF, a comprehensive, unbiased, and high-resolution survey into the cellular and molecular ecosystem of the human G-CSF-primed bone marrow (G-BM) is lacking so far. Here, we employed single-cell RNA sequencing to profile hematopoietic cells in human bone marrow from two healthy donors before and after 5-day G-CSF administration. Through unbiased bioinformatics analysis, our data systematically showed the alterations in the transcriptional landscape of hematopoietic cells in G-BM, and revealed that G-CSF-induced myeloid-biased differentiation initiated from the stage of lymphoid-primed multipotent progenitors. We also illustrated the cellular and molecular basis of hyporesponsiveness of T cells and natural killer (NK) cells caused by G-CSF stimulation, including the potential direct mechanisms and indirect regulations mediated by ligand–receptor interactions. Taken together, our data extend the understanding of lymphomyeloid divergence and potential mechanisms involved in hyporesponsiveness of T and NK cells in human G-BM, which might provide basis for optimization of stem cell transplantation in hematological malignancy treatment. Molybdenum oxide (MoO
3 ) has become a potential anode material for lithium-ion batteries due to high theoretical capacity and environmental friendliness. Nonetheless, MoO3 suffers from serious capacity fading caused by severe pulverization and lower electronic conductivity. Herein, the carbon nanotubes (CNTs) are embedded in α-MoO3 nanoribbons via a facile one-step hydrothermal process. The synergistic effect of α-MoO3 and CNTs can effectively alleviate the volume expansion of α-MoO3 and dominated Li+ storage properties. As a result, the flexible α-MoO3 /CNTs composite exhibits excellent rate capability compared with the original α-MoO3 , as well as a reversible capacity of 270 mAh/g after 250 cycles. Multiple pluripotent states have been described in mouse and human stem cells. Here, we apply single-cell RNA-seq to a newly established BMP4 induced mouse primed to naïve transition (BiPNT) system and show that the reset is not a direct reversal of cell fate but goes through a primordial germ cell-like cells (PGCLCs) state. We first show that epiblast stem cells bifurcate into c-Kit
+ naïve and c-Kit− trophoblast-like cells, among which, the naïve branch undergoes further transition through a PGCLCs intermediate capable of spermatogenesis in vivo. Mechanistically, we show that DOT1L inhibition permits the transition from primed pluripotency to PGCLCs in part by facilitating the loss of H3K79me2 from Gata3/6. In addition, Prdm1/Blimp1 is required for PGCLCs and naïve cells, while Gata2 inhibits PGC-like state by promoting trophoblast-like fate. Our work not only reveals an alternative route for primed to naïve transition, but also gains insight into germ cell development. Amodiaquine (AQ) is a commonly used antimalarial drug, and N-desethyl-AQ (N-DEAQ) is an active metabolite of AQ. Given the significance of drug quality in the management of malaria cases, this study aims to develop antibody-based assays for the detection and quantitation of AQ without the need for sophisticated equipment. Two monoclonal antibodies (mAbs) against AQ, designated as JUN7 and TE7, were selected, which showed 72.7% and 9.5% cross-reactivity to N-DEAQ, respectively. These mAbs showed <0.1% cross-reactivity to other commonly used antimalarial drugs. An indirect competitive enzyme-linked immunosorbent assay (icELISA) based on JUN7 showed a 50% inhibitory concentration (IC
50 ) of 0.16 ng/mL and a working range of 0.06–0.46 ng/mL. A lateral flow immunoassay (LFIA) based on JUN7 was also developed with a working range of 2.58–30.86 ng/mL. The icELISA and LFIA were applied for the quantification of AQ in commercial drugs, and the results were comparable to those determined using high-performance liquid chromatography. In addition, a combination dipstick for simultaneous, qualitative analysis of AQ and artesunate was developed. All immunoassays based on JUN7 can be applied for quality control of AQ-containing artemisinin-based combination therapies. As TE7 showed low cross-reactivity to N-DEAQ, an icELISA based on TE7 was developed with an IC50 of 0.38 ng/mL and a working range of 0.14–1.67 ng/mL. The TE7 icELISA was applied for the study of pharmacokinetics of AQ in rat serum after intragastric administration, and the results were consistent with those of previous studies. 2 -coated zeolitic imidazoline framework-derived transition metal sulfides (MoS2 -coated S-doped ZIF-67@NF) are synthesized by hydrothermal method on nickel foam (NF). ZIF-derived transition metal sulfide and MoS2 as inner core and outer shell, respectively. Overall structure like a succulent plant, maintaining the unique skeleton structure of ZIF as an inner core, ZIF-derived transition metal sulfides enhances the charge transfer efficiency. The well-covered MoS2 protects the inner core and acts as a channel for electrolyte ion diffusion to the interior. At 1 mA cm−2 , the MoS2 coated S-doped ZIF-67@NF (MSZ@NF) electrode provides a specific capacity of 4840 mF cm−2 and good cycling stability (81.3% at 30 mA cm−2 after 5000 cycles). The easy loss of crosslinking ions in alginate can result in a structural collapse of the physiological environment, thereby losing its characteristics as a bone scaffold. Meanwhile, alginate lacks osteoconductive properties, which are necessary for ideal bone scaffolds. In this study, strontium (Sr) in combination with calcium (Ca) at different ratios were used as a crosslinking agent for the alginate to investigate the effect of Ca–Sr ratio on the physicochemical properties and biological preformation of alginate hydrogel. Here, Ca and Sr in different weight ratios (4:0, 3:1, 2:2, 1:3, and 0:4) were employed as crosslinking agents. The physicochemical properties of hydrogels, including pore size, elastic modulus, degradation rate and swelling ratio, could be effectively tuned by controlling the amount of Sr. The ion release experiment revealed a burst release of Sr
2+ in the first day after crosslinking. However, after 3 days, the amount of Sr2+ release had significantly declined and was proportional to the total strontium initially introduced into the alginate. Meanwhile, the live/dead results exhibited higher cell viability for alginate with 2:2 Ca–Sr weight ratio. The alginate with 2:2 Ca–Sr ratio not only improved osteoblastic attachment, but also up-regulated the alkaline phosphatase activity, the expression of osteogenic marker genes, and relative growth factors. Female germline cells in flowering plants differentiate from somatic cells to produce specialized reproductive organs, called ovules, embedded deep inside the flowers. We investigated the molecular basis of this distinctive developmental program by performing single-cell RNA sequencing (scRNA-seq) of 16,872 single cells of Arabidopsis thaliana ovule primordia at three developmental time points during female germline differentiation. This allowed us to identify the characteristic expression patterns of the main cell types, including the female germline and its surrounding nucellus. We then reconstructed the continuous trajectory of female germline differentiation and observed dynamic waves of gene expression along the developmental trajectory. A focused analysis revealed transcriptional cascades and identified key transcriptional factors that showed distinct expression patterns along the germline differentiation trajectory. Our study provides a valuable reference dataset of the transcriptional process during female germline differentiation at single-cell resolution, shedding light on the mechanisms underlying germline cell fate determination. Zhimin Hou, Yanhui Liu et al. used single cell RNA-seq to analyze the model organism, Arabidopsis thaliana, at three stages during female germline differentiation. The extensive chemical investigation on the branches and leaves of Terminalia chebula var. tomentella (Combretaceae) led to the isolation of two new lignan glucosides with a furofuran skeleton, termitomenins F (1) and G (2). In addition, 19 known compounds including five lignan glucosides (3–7), six hydrolyzable tannins (8–13) and eight simple phenolics (14–21) were also identified. Their structures were determined by comprehensive spectroscopic analyses. It is noted that 8 and 9 were C-glycosidic hydrolyzable tannins with one hexahydroxydiphenoyl and one gallagyl group linked to an open-chain glucosyl C-1/O-2/O-3 and O-4/O-6, respectively, which were rarely found in plants. Nine known compounds, 6–9, 13, and 18–21, were procured from the titled plant for the first time, while 3–5, 10–12 and 14–17 were also found in the fruits. Notably, the known hydrolyzable tannins 8–13 exhibited stronger α-glucosidase inhibitory activities with IC

Iris germanica, a species with very high ornamental value, exhibits the strongest drought resistance among the species in the genus Iris, but the molecular mechanism underlying its drought resistance has not been evaluated. To investigate the gene expression profile changes exhibited by high-drought-resistant I. germanica under drought stress, 10 cultivars with excellent characteristics were included in pot experiments under drought stress conditions, and the changes in the chlorophyll (Chl) content, plasma membrane relative permeability (RP), and superoxide dismutase (SOD), malondialdehyde (MDA), free proline (Pro), and soluble protein (SP) levels in leaves were compared among these cultivars. Based on their drought-resistance performance, the 10 cultivars were ordered as follows: 'Little Dream' > 'Music Box' > 'X'Brassie' > 'Blood Stone' > 'Cherry Garden' > 'Memory of Harvest' > 'Immortality' > 'White and Gold' > 'Tantara' > 'Clarence'. Using the high-drought-resistant cultivar 'Little Dream' as the experimental material, cDNA libraries from leaves and rhizomes treated for 0, 6, 12, 24, and 48 h with 20% polyethylene glycol (PEG)-6000 to simulate a drought environment were sequenced using the Illumina sequencing platform. We obtained 1, 976, 033 transcripts and 743, 982 unigenes (mean length of 716 bp) through a hierarchical clustering analysis of the resulting transcriptome data. The unigenes were compared against the Nr, Nt, Pfam, KOG/COG, Swiss-Prot, KEGG, and gene ontology (GO) databases for functional annotation, and the gene expression levels in leaves and rhizomes were compared between the 20% PEG-6000 stress treated (6, 12, 24, and 48 h) and control (0 h) groups using DESeq2. 7849 and 24,127 differentially expressed genes (DEGs) were obtained from leaves and rhizomes, respectively. GO and KEGG enrichment analyses of the DEGs revealed significantly enriched KEGG pathways, including ribosome, photosynthesis, hormone signal transduction, starch and sucrose metabolism, synthesis of secondary metabolites, and related genes, such as heat shock proteins (HSPs), transcription factors (TFs), and active oxygen scavengers. In conclusion, we conducted the first transcriptome sequencing analysis of the I. germanica cultivar 'Little Dream' under drought stress and generated a large amount of genetic information. This study lays the foundation for further exploration of the molecular mechanisms underlying the responses of I. germanica to drought stress and provides valuable genetic resources for the breeding of drought-resistant plants. 30. Resource and dependency based test case generation for RESTful Web services.

Nowadays, RESTful web services are widely used for building enterprise applications. REST is not a protocol, but rather it defines a set of guidelines on how to design APIs to access and manipulate resources using HTTP over a network. In this paper, we propose an enhanced search-based method for automated system test generation for RESTful web services, by exploiting domain knowledge on the handling of HTTP resources. The proposed techniques use domain knowledge specific to RESTful web services and a set of effective templates to structure test actions (i.e., ordered sequences of HTTP calls) within an individual in the evolutionary search. The action templates are developed based on the semantics of HTTP methods and are used to manipulate the web services’ resources. In addition, we propose five novel sampling strategies with four sampling methods (i.e., resource-based sampling) for the test cases that can use one or more of these templates. The strategies are further supported with a set of new, specialized mutation operators (i.e., resource-based mutation) in the evolutionary search that take into account the use of these resources in the generated test cases. Moreover, we propose a novel dependency handling to detect possible dependencies among the resources in the tested applications. The resource-based sampling and mutations are then enhanced by exploiting the information of these detected dependencies. To evaluate our approach, we implemented it as an extension to the EvoMaster tool, and conducted an empirical study with two selected baselines on 7 open-source and 12 synthetic RESTful web services. Results show that our novel resource-based approach with dependency handling obtains a significant improvement in performance over the baselines, e.g., up to + 130.7% relative improvement (growing from + 27.9% to + 64.3%) on line coverage. The RNA modification N
6 -methyladenosine (m6 A) has critical roles in many biological processes1,2. However, the function of m6 A in the early phase of mammalian development remains poorly understood. Here we show that the m6 A reader YT521-B homology-domain-containing protein 1 (YTHDC1) is required for the maintenance of mouse embryonic stem (ES) cells in an m6 A-dependent manner, and that its deletion initiates cellular reprogramming to a 2C-like state. Mechanistically, YTHDC1 binds to the transcripts of retrotransposons (such as intracisternal A particles, ERVK and LINE1) in mouse ES cells and its depletion results in the reactivation of these silenced retrotransposons, accompanied by a global decrease in SETDB1-mediated trimethylation at lysine 9 of histone H3 (H3K9me3). We further demonstrate that YTHDC1 and its target m6 A RNAs act upstream of SETDB1 to repress retrotransposons and Dux, the master inducer of the two-cell stage (2C)-like program. Within the mountain altitudinal vegetation belts, the shift of forest tree lines and subalpine steppe belts to high altitudes constitutes an obvious response to global climate change. However, whether or not similar changes occur in steppe belts (low altitude) and nival belts in different areas within mountain systems remain undetermined. It is also unknown if these, responses to climate change are consistent. Here, using Landsat remote sensing images from 1989 to 2015, we obtained the spatial distribution of altitudinal vegetation belts in different periods of the Tianshan Mountains in Northwestern China. We suggest that the responses from different altitudinal vegetation belts to global climate change are different. The changes in the vegetation belts at low altitudes are spatially different. In high-altitude regions (higher than the forest belts), however, the trend of different altitudinal belts is consistent. Specifically, we focused on analyses of the impact of changes in temperature and precipitation on the nival belts, desert steppe belts, and montane steppe belts. The results demonstrated that the temperature in the study area exhibited an increasing trend, and is the main factor of altitudinal vegetation belts change in the Tianshan Mountains. In the context of a significant increase in temperature, the upper limit of the montane steppe in the eastern and central parts will shift to lower altitudes, which may limit the development of local animal husbandry. The montane steppe in the west, however, exhibits the opposite trend, which may augment the carrying capacity of pastures and promote the development of local animal husbandry. It is crucial and desirable to develop green and high-efficient strategies to regulate solid-state structures and their related material properties. However, relative to solution, it is more difficult to break and generate chemical bonds in solid states. In this work, a rubbing-induced photoluminescence on the solid states of ortho-pyridinil phenol family was achieved. This rubbing response relied on an accurately designed topochemical tautomerism, where a negative charge, exactly provided by the triboelectric effect of a rubber, can induce a proton transfer in a double H-bonded dimeric structure. This process instantaneously led to a bright-form tautomer that can be stabilized in the solid-state settings, leading to an up to over 450-fold increase of the fluorescent quantum yield of the materials. The property can be repeatedly used due to the reversibility of the tautomerism, enabling encrypted applications. Moreover, a further modification to the structure can be accomplished to achieve different properties, opening up more possibilities for the design of new-generation smart materials. Changes in molecular properties due to stimuli response are critically important for the development of new materials. However, most processes are slow or inefficient in the solid state. Neuroendocrine prostate cancer is one of the most aggressive subtypes of prostate tumor. Although much progress has been made in understanding the development of neuroendocrine prostate cancer, the cellular architecture associated with neuroendocrine differentiation in human prostate cancer remain incompletely understood. Here, we use single-cell RNA sequencing to profile the transcriptomes of 21,292 cells from needle biopsies of 6 castration-resistant prostate cancers. Our analyses reveal that all neuroendocrine tumor cells display a luminal-like epithelial phenotype. In particular, lineage trajectory analysis suggests that focal neuroendocrine differentiation exclusively originate from luminal-like malignant cells rather than basal compartment. Further tissue microarray analysis validates the generality of the luminal phenotype of neuroendocrine cells. Moreover, we uncover neuroendocrine differentiation-associated gene signatures that may help us to further explore other intrinsic molecular mechanisms deriving neuroendocrine prostate cancer. In summary, our single-cell study provides direct evidence into the cellular states underlying neuroendocrine transdifferentiation in human prostate cancer. CoNi-contained nanosheets can be prepared by dealloying CoNiAl alloys in aqueous NaOH solution in the presence of H
2 O2 , and upon annealing sample exhibits dendritic NiCo2 O4 micro-nanostructure. The effect of H2 O2 solution on the structure, morphology, and electrochemical performances of the resulting products is studied systematically. These results indicate that the H2 O2 solution mainly influences the morphology of the NiCo2 O4 . When tested as anode materials for lithium-ion batteries (LIBs), the obtained NiCo2 O4 sample shows high specific capacity, excellent rate property, and superb cycling stability. A reversible capacity is still maintained at 1016.9 mAh/g after 100 cycles at a current density of 100 mA/g. Even at a current rate of 1000 mA/g, the capacity can reach to 691.4 mAh/g. The outstanding electrochemical properties of the NiCo2 O4 anode make them promising anode materials of LIBs and other energy storage applications. [ABSTRACT FROM AUTHOR]- Published

We have designed a polarization conversion coding metasurface (CM) for broadband radar cross-section reduction. The CM is made up of a kind of deformed bowtie-shaped particle and its mirror, named "0" and "1", respectively, which can realize cross-polarization conversion under the vertical incidence of a linearly polarized wave. According to the coding sequence optimized by the genetic algorithm, the two kinds of unit are arranged to form a design, called a 1-bit CM, which can scatter electromagnetic waves in various directions. The simulation results demonstrate that the two kinds of unit can achieve more than a 90% polarization conversion ratio, and the whole CM can realize more than a 10-dB reduction of monostatic radar cross-section (RCS) from 8.8 GHz to 20.9 GHz, while the maximum RCS reduction of 32.5 dB can be obtained at 14 GHz under the normal incidence of x- and y-polarized waves. Enterovirus B83 (EV-B83) is a new member of the enterovirus B group. Currently, there are only two full-length genomic sequences of EV-B83 in the GenBank database and few VP1 region sequences. The aetiology and epidemiology of EV-B83 is unclear. 24 stool specimens were collected from twelve AFP patients and 298 stool specimens were collected from 298 healthy children in support of polio eradication activities in Tibet in 1999. Two polioviruses (isolated by L20B cell) and one non-polio enterovirus (isolated by RD cell) were isolated from AFP patients and nine polioviruses (isolated by L20B cell) and 90 non-polio enteroviruses (isolated by RD cell) were isolated from health children. Through molecular typing, we confirmed that the six of non-polio enteroviruses belong to EV-B83. The sequence similarity between the VP1 region of the Tibet isolates and that of the EV-B83 prototype strain was 80%. The maximum-likelihood phylogenetic tree of the partial VP1 region in EV-B83 demonstrated that EV-B83 formed four genotypes globally during the evolution process. The six Tibet EV-B83 strains formed the D genotype alone. Recombination analysis of Tibet EV-B83 showed that CV-B4, CV-A9, EV-B80, and EV-B106 may act as recombinant donors in multiple regions. The serum neutralization test showed that the antibody-positive rate was 58.8% and GMT was 1:19.70, which was higher than the previously reported results of EV-B106 and EV-B80. Temperature sensitivity test results showed that the six Tibet EV-B83 strains were temperature-insensitive with stronger virulence and potential infectivity, which was consistent with the results of the serum neutralization test. This study enriched the genome-wide sequence, epidemiological characteristics, and provided basic data for the follow-up study of EV-B83. [ABSTRACT FROM AUTHOR]

Transition metal oxide materials prepared by dealloying method have attracted more and more attention and researches due to their high theoretical specific capacitance. In this study, nanoporous NiO@CoO composite electrodes were facilely synthesized by the melting and in situ corrosion, which exhibits an excellent electrochemical performance. The superior performance is ascribed to the reasonable pore size distribution (average pore diameter of 5.1 nm) and high specific surface area (50.75 m
2 g−1 ) of the NiO@CoO, which provides a fast channel for electrolyte ions, shorten the free path of ion transfer and provide sufficient active sites for redox reactions. Moreover, the as-prepared material demonstrates a highest specific capacitance of 18.4 F cm−2 at a current density of 10 mA cm−2 , and exhibits an excellent cycling stability with 93.9% retention after 5000 cycles at 30 mA cm−2 . In a word, owing to the porous structure and pseudocapacitance effect, the NiO@CoO composite electrode has an ultra-high performance, which proves that this study has a great research value in the field of supercapacitors. 42. Heme as a target for protection against doxorubicin-induced apoptosis in H9c2 cardiomyocytes.

Heme homeostasis is of vital importance to many biological processes associated with cell redox activity. However, the role of heme in the doxorubicin (DOX)-induced cardiotoxicity is still not clear. The aim of the present study was to test the hypothesis that heme is related to the DOX-induced oxidative stress and inhibition of heme expression may protect H9c2 cardiomyocytes against DOX-induced cardiotoxicity. For the evaluation of heme changing under doxorubicin treatment, H9c2 cells were treated with 0.5, 1, 2, and 4 mg/mL doxorubicin respectively. H9c2 cells were divided into 5 groups: Control group (cells were cultured without intervention), DOX group (cells were treated with 2 mg/mL doxorubicin for 6 h), Heme depletion+DOX group (cells were cultured with heme-depleted serum media, 0.5 mM succinylacetone and 2 mg/mL doxorubicin), Heme group (cells were treated with 30 μM heme), and Heme depletion+DOX+Heme group. Apoptotic cells were detected by flow cytometry with Annexin V-FITC/PI. The intracellular oxidant levels were measured by DCFH-DA fluorescence. The levels of heme were detected by ELISA. Doxorubicin significantly increased intracellular heme level from 5013 ± 187 ng/mL to the highest level of 11,720 ± 107 ng/mL, as well as the intracellular oxidants and cell apoptosis rate elevated by the increase of doxorubicin concentration. Heme depletion can significantly suppress the DOX-induced apoptosis from 39.8 ± 0.5% to 20.8 ± 0.5% (p < 0.001). Re-supplemented with exogenous heme partially but significantly restored the DOX-induced apoptosis. Heme plays an important role in doxorubicin toxicity–induced cardiomyocyte injury. By appropriate reduction in the accumulation of free heme in cardiomyocytes, doxorubicin-induced cardiotoxicity may be alleviated. [ABSTRACT FROM AUTHOR]

Emulsions, usually oil-in-water emulsions, have been widely applied to the preparation of hydrophobic drug carriers. However, the synthesis of safe, biocompatible and high-performance carriers for water-soluble drugs remains a challenge. So polyacrylamide (PAM) microspheres stabilized by carbon dots (CDs) were fabricated via a water-in-oil Pickering emulsion without the use of any surfactant. The PAM/CD microspheres were spherical with an even distribution of the CDs on their surfaces. The size of the microspheres can be controlled by changing the amount of CDs, which also influenced the drug loading properties. The hydrophilic chemotherapy drug, 5-fluorouracil (5-FU), was successfully loaded into the PAM/CD microspheres through incubation. The encapsulation efficiencies ranged from 68 to 82% with a maximum drug loading of 48%. The 5-FU release profiles depended on the concentration of the drug incubation solution, the concentration of the CD solution and the pH of the drug release environment. The release of 5-FU was relatively steady for the first 12 h and followed non-Fickian diffusion kinetics. This synthetic approach could be extrapolated to loading of other water-soluble drugs into hydrophilic polymers. [ABSTRACT FROM AUTHOR]

The denitrifier method is widely used as a novel pretreatment method for the determination of nitrogen and oxygen isotope ratios as it can provide quantitative and high-sensitivity measurements. Nevertheless, the method is limited by relatively low measurement accuracy for δ
18 O. In this study, we analyzed the factors influencing the accuracy of δ18 O determination, and then systematically investigated the effects of dissolved oxygen concentrations and nitrate sample sizes on estimates of the δ15 N and δ18 O of nitrate reference materials. The δ18 O contraction ratio was used to represent the relationship between the measured difference and true difference between two reference materials. We obtained the following main results: (1) a gas-liquid ratio of 3:10 (v/v) in ordinary triangular flasks and a shaking speed of 120 r/min produced an optimal range (1.9 to 2.6 mg/L) in the concentration of dissolved oxygen for accurately determining δ18 O, and (2) the δ18 O contraction ratio decreased as nitrate sample size decreased within a certain range (1.0 to 0.1 μmol). Our results suggested that δ18 O contraction is influenced mainly by dissolved oxygen concentrations in pure culture, and provided a model for improving the accuracy of oxygen isotope analysis. - Abstract
The successful segregation of germ cells from somatic lineages is vital for sexual reproduction and species survival. In the mouse, primordial germ cells (PGCs), precursors of all germ cells, are induced from the post-implantation epiblast1. Induction requires BMP4 signalling to prospective PGCs2 and the intrinsic action of PGC transcription factors3-6. However, the molecular mechanisms that connect BMP4 to induction of the PGC transcription factors that are responsible for segregating PGCs from somatic lineages are unknown. Here we show that the transcription factor OTX2 is a key regulator of these processes. Downregulation of Otx2 precedes the initiation of the PGC programme both in vitro and in vivo. Deletion of Otx2 in vitro markedly increases the efficiency of PGC-like cell differentiation and prolongs the period of PGC competence. In the absence of Otx2 activity, differentiation of PGC-like cells becomes independent of the otherwise essential cytokine signals, with germline entry initiating even in the absence of the PGC transcription factor BLIMP1. Deletion of Otx2 in vivo increases PGC numbers. These data demonstrate that OTX2 functions repressively upstream of PGC transcription factors, acting as a roadblock to limit entry of epiblast cells to the germline to a small window in space and time, thereby ensuring correct numerical segregation of germline cells from the soma. A high but normal fasting plasma glucose level in adults is a marker for insulin resistance and future development of type 2 diabetes mellitus. We aimed to investigate whether ADIPOR2 variants are associated with increased fasting plasma glucose (FPG) in non-diabetic Chinese Han subjects who had normal FPG levels. This study included 2038 subjects among the non-diabetic Chinese Han population. The ADIPOR2 polymorphisms were genotyped by the TaqMan method. For the analysis, participants were divided into low-normal FPG group (FPG < 4.88 mmol/L) and high-normal FPG group (6.1 mmol/L > FPG ≥ 4.88 mmol/L). We identified five single nucleotide polymorphisms (SNPs) of ADIPOR2 gene: rs10773989, rs2370055, rs9300298, rs10773983, and rs13611594. There was no significant difference in FPG levels between different genotypes of rs10773989, rs2370055, rs9300298, and rs13611594. Our data showed a significant association of rs10773983 with higher FPG levels, fasting insulin levels, and homeostatic model assessment of insulin resistance (HOMA-IR) in A-allele carriers (p = 0.006, p < 0.001, and p < 0.001, respectively). The subjects in high FPG group had higher median fasting insulin level (9.25 vs 8.16 μIU/mL, p = 0.003) and higher HOMA-IR (2.21 vs 1.58, p < 0.001) than those in normal FPG group. Subjects with the A-allele of rs10773983 compared to subjects with G-allele had 1.222-fold higher risk for high-normal FPG (OR = 1.222, 95%CI = 1.010-1.478, p = 0.039). Our findings suggest that ADIPOR2 rs10773983 polymorphism may be associated with an increased risk of high-normal FPG among non-diabetic Chinese Han subjects who have normal FPG levels. The radiation-induced bystander effect (RIBE) refers to a unique process in which factors released by irradiated cells or tissues exert effects on other parts of the animal not exposed to radiation, causing genomic instability, stress responses and altered apoptosis or cell proliferation. Although RIBEs have important implications for radioprotection, radiation safety and radiotherapy, the molecular identities of RIBE factors and their mechanisms of action remain poorly understood. Here we use Caenorhabditis elegans as a model in which to study RIBEs, and identify the cysteine protease CPR-4, a homologue of human cathepsin B, as the first RIBE factor in nematodes, to our knowledge. CPR-4 is secreted from animals irradiated with ultraviolet or ionizing gamma rays, and is the major factor in the conditioned medium that leads to the inhibition of cell death and increased embryonic lethality in unirradiated animals. Moreover, CPR-4 causes these effects and stress responses at unexposed sites distal to the irradiated tissue. The activity of CPR-4 is regulated by the p53 homologue CEP-1 in response to radiation, and CPR-4 seems to exert RIBEs by acting through the insulin-like growth factor receptor DAF-2. Our study provides crucial insights into RIBEs, and will facilitate the identification of additional RIBE factors and their mechanisms of action. [ABSTRACT FROM AUTHOR]

α-FeO particle supported on diatomite (α-FeO/diatomite composite) was prepared at room temperature by the chemical precipitation method using ferrous sulphate as the raw material and diatomite as supporter. α-FeO/diatomite composite was characterized by XRD, FE-SEM, FT-IR and DRS. The result showed that α-FeO was uniformly loaded on the diatomite. The photocatalytic activity was investigated under visible light toward degradation of Rhodamine 6G aqueous solution and the effects of various experimental factors on Rhodamine 6G degradation were investigated. Compared with the α-FeO, α-FeO/diatomite composite had significantly enhanced activity in the degradation of Rhodamine 6G under visible light irradiation. Besides, the photostability of catalysts was also investigated. The experimental results indicated that the prepared composite is a promising material for the wastewater treatment for its good catalytic performance property and long-term stability. Twelve salts of 3-nitro-1,2,4-triazol-5-one (NTO) (ammonium, hydrazinium, guanidinium, aminoguanidinium, diaminoguanidinium, triaminoguanidinium, N-carbamoylguanidinium, semicarbazidium, 1,5-diamino-1,2,4-tetrazolium, 3,4,5- triamino-1,2,4-triazolium, 3,6,7-triamino-7H-[1, 2, 4]triazolo[5,1-c][1,2,4]triazol-2- ium, and 4,4′,5,5′-tetraamino-3,3′-bi-1,2,4-triazolium) were synthesized. The new salts were fully characterized by H and C NMR spectroscopy, infrared spectroscopy, and elemental analysis. The crystal structures of salts 10 and 11 were determined by single-crystal X-ray diffraction. All energetic salts except salt 6 exhibit excellent thermal stabilities with decomposition temperatures ranging from 203 to 270 °C. The densities of salts ranged from 1.65 to 1.88 g cm as measured by a gas pycnometer. Theoretical performance calculations (Gaussian 03 and EXPLO5 v6.01) yielded detonation pressures and detonation velocities for the energetic salts, ranging from 24.4 to 38.1 GPa and 8136 to 9575 m s, respectively. In particular, salt 2 has an outstanding detonation performance ( P = 38.1 GPa, v = 9575 m s) with a satisfactory acidity compared to that of NTO ( pK = 5.63 versus pK = 2.37). Furthermore, the particles of salt 2 form two-dimensional blades of submicron size, as determined by scanning electron microscopy analysis. Meanwhile, salt 2 was compatible with TNAZ, TATB, TKX-50, Al, NHClO, CL-20, TNT, and F2603 fluororubber, as determined by differential scanning calorimetry or vacuum stability tests.
