3. Tri-boson and WH production in the W+W+jj channel: predictions at full NLO accuracy and beyond.

- Abstract
In this work, we present the first full NLO predictions for the process pp → μ
+ νμ e+ νe jj at the LHC in a typical tri-boson phase space. The NLO corrections reach 50% at the level of the fiducial cross section and have a very different hierarchy with respect to vector-boson-scattering phase spaces. By comparing the cross section of the full off-shell process with the sum of contributing on-shell electroweak-boson production subchannels, we find that the process is dominated by WWW and WH production, while vector-boson-scattering topologies still play a non-negligible role. In addition, NLO QCD predictions matched to parton shower which are supplemented by approximate electroweak corrections are provided. For the fiducial cross section, the electroweak corrections turn out to be small but the QCD corrections reach 47%. For the inclusive cross section, matching to parton shower affects the predictions by 7%. Measurements of vector boson polarisation in vector boson production processes offer a powerful probe of the electroweak symmetry breaking mechanism, scrutinising the Standard Model and new physics scenarios alike. Since massive vector bosons can only be observed as intermediate particles, polarised cross section templates from simulation are necessary to extract their polarisation from measurable unpolarised distributions. In this work we present an extension of the Sherpa Monte-Carlo event generator allowing the simulation of polarised cross sections for vector boson production processes. Based on the narrow-width approximation, polarised cross sections of all possible polarisation combinations for an arbitrary number of intermediate vector bosons can be simulated in a single simulation run. In addition, it is possible to directly predict the interference between different intermediate polarisation states, and various differing polarisation definitions can be studied simultaneously. Besides the simulation of polarised cross sections at fixed LO and LO+PS accuracy as well as in multijet-merged calculations, we also present parton-shower-matched polarised cross sections with approximate NLO QCD corrections in the vector boson production processes. We demonstrate that the differences of this approximation to full NLO QCD predictions are small and it thus opens up the possibility for fully-simulated calculations at the hadron level including polarisation information and higher-order QCD effects for the first time. 5. Removal of endocrine disrupting chemicals from water through urethane functionalization of microfiltration membranes via electron beam irradiation.

- Abstract
Polyethersulphone (PES) membranes modified with urethane functional groups were prepared through an interfacial reaction using electron beam irradiation. The removal of eight endocrine disrupting chemicals (EDCs) was studied using both pristine and functionalized PES membranes. The prepared membranes underwent characterization using several techniques, including attenuated total reflectance-Fourier transform infrared (ATR-FTIR) spectroscopy, scanning electron microscopy, contact angle analysis, and measurements of pure water flux. Furthermore, dynamic adsorption experiments were conducted to evaluate the adsorption mechanism of the prepared membrane toward the eight EDCs. The urethane functionalized membranes were hydrophilic (52° contact angle) and maintained a high permeate flux (26000 L/h m
2 bar) throughout the filtration process. Dynamic adsorption results demonstrated that the introduction of urethane functional groups on the membranes significantly enhanced the removal efficiency of 17β-estradiol, estriol, bisphenol A, estrone, ethinylestradiol, and equilin. The adsorption loading of 17β-estradiol on the functionalized PES membrane was 6.7 ± 0.7 mg/m2 , exhibiting a 5-fold increase compared to the unmodified PES membrane. In this paper, we present an algorithm to construct the q
T distribution at NLO accuracy to arbitrary power precision, including the assembly of suitable zero-bin subtrahends, in a mathematically well-defined way for a generic choice of rapidity-divergence regularisation prescription. In its derivation, we divide the phase space into two sectors, the interior of the integration domain as well as the integration boundary, which we include here for the first time. To demonstrate the applicability and usefulness of our algorithm, we calculate the N2 LP corrections for Higgs hadroproduction for the first time. We observe that our approximate N2 LP-accurate qT spectra replicate the asymptotic behaviour of the full QCD calculation to a much better degree than the previously available results, both within the qT → 0 limit and in the large-qT domain for all the involved partonic processes. While playing a minor role at larger transverse momenta, we show that the newly incorporated boundary contribution plays a vital role in the qT → 0 limit, where any subleading power accuracy would be lost without them. In particular, our N2 LP-accurate qT expansion can approximate the exact qT distribution up to qT ≈ 30 GeV at the percent level for rapidities |YH | ≲ 3. [ABSTRACT FROM AUTHOR]- Published

With the advent of serial X-ray crystallography on microfocus beamlines at free-electron laser and synchrotron facilities, the demand for protein microcrystals has significantly risen in recent years. However, by in vitro crystallization extensive efforts are usually required to purify proteins and produce sufficiently homogeneous microcrystals. Here, we present InCellCryst, an advanced pipeline for producing homogeneous microcrystals directly within living insect cells. Our baculovirus-based cloning system enables the production of crystals from completely native proteins as well as the screening of different cellular compartments to maximize chances for protein crystallization. By optimizing cloning procedures, recombinant virus production, crystallization and crystal detection, X-ray diffraction data can be collected 24 days after the start of target gene cloning. Furthermore, improved strategies for serial synchrotron diffraction data collection directly from crystals within living cells abolish the need to purify the recombinant protein or the associated microcrystals. SX experiments require thousands of high-quality microcrystals. Here, authors present a pipeline for protein crystallization in insect cells, including compartment screening and identification of physiological ligands from data collection in cellulo. Mitochondrial DNA copy number (mtDNA-CN) is a biomarker for mitochondrial dysfunction associated with several diseases. Previous genome-wide association studies (GWAS) have been performed to unravel underlying mechanisms of mtDNA-CN regulation. However, the identified gene regions explain only a small fraction of mtDNA-CN variability. Most of this data has been estimated from microarrays based on various pipelines. In the present study we aimed to (1) identify genetic loci for qPCR-measured mtDNA-CN from three studies (16,130 participants) using GWAS, (2) identify potential systematic differences between our qPCR derived mtDNA-CN measurements compared to the published microarray intensity-based estimates, and (3) disentangle the nuclear from mitochondrial regulation of the mtDNA-CN phenotype. We identified two genome-wide significant autosomal loci associated with qPCR-measured mtDNA-CN: at HBS1L (rs4895440, p = 3.39 × 10
–13 ) and GSDMA (rs56030650, p = 4.85 × 10–08 ) genes. Moreover, 113/115 of the previously published SNPs identified by microarray-based analyses were significantly equivalent with our findings. In our study, the mitochondrial genome itself contributed only marginally to mtDNA-CN regulation as we only detected a single rare mitochondrial variant associated with mtDNA-CN. Furthermore, we incorporated mitochondrial haplogroups into our analyses to explore their potential impact on mtDNA-CN. We present a simple parton-shower model that replaces the explicit angular ordering of the coherent branching formalism with a differentially accurate simulation of soft-gluon radiation by means of a non-trivial dependence of the splitting functions on azimuthal angles. We introduce a global kinematics mapping and provide an analytic proof that it satisfies the criteria for next-to leading logarithmic accuracy. In the new algorithm, initial and final state evolution are treated on the same footing. 12. Photon splitting corrections to soft-photon resummation.

In this paper we present an algorithm to add photon-splitting corrections to the Yennie-Frautschi-Suura-style soft-photon resummation available in the Sherpa MonteCarlo event generator. Photon-splitting corrections enter at NNLO in QED and, as these effects are not incorporated in the standard QED FSR resummations, their size is larger than the pure hard photon-emission corrections at the same order. We introduce different lepton dressing strategies which incorporate further leptons and hadrons in addition to the customary photons, and discuss their sensitivity to dressing parameters such as the cone size. Finally, we quantify the effects of photon splittings into charged fermions and scalars under different such dressing strategies on Z → e+e− decays and find effects of up to 1% for suitably inclusive dressing strategies independent of the dressing cone size, and up to 9% if only photons are used in the dressing procedure with large dressing cones. - Abstract
Purpose: The aim of this study was to analyze the heart dose for left-sided breast cancer that can be achieved during daily practice in patients treated with multicatheter brachytherapy (MCBT) accelerated partial-breast irradiation (APBI) and deep-inspiration breath-hold (DIBH) whole-breast irradiation (WBI) using a simultaneous integrated tumor bed boost (SIB)—two different concepts which nonetheless share some patient overlap. Materials and methods: We analyzed the nominal average dose (Dmean) to the heart as well as the biologically effective dose (BED) and the equivalent dose in 2‑Gy fractions (EQD2) for an α/β of 3 in 30 MCBT-APBI patients and 22 patients treated with DIBH plus SIB. For further dosimetric comparison, we contoured the breast planning target volume (PTV) in each of the brachytherapy planning CTs according to the ESTRO guidelines and computed tangential field plans. Mean dose (Dmean), EQD2 Dmean, and BED Dmean for three dosing schemes were calculated: 50 Gy/25 fractions and two hypofractionated regimens, i.e., 40.05 Gy/15 fractions and 26 Gy/5 fractions. Furthermore, we calculated tangential field plans without a boost for the 22 cases treated with SIB with the standard dosing scheme of 40.05 Gy/15 fractions. Results: MCBT and DIBH radiation therapy both show low-dose exposure of the heart. As expected, hypofractionation leads to sparing of the heart dose. Although MCBT plans were not optimized regarding dose to the heart, Dmean differed significantly between MCBT and DIBH (1.28 Gy vs. 1.91 Gy, p < 0.001) in favor of MCBT, even if the Dmean in each group was very low. In MCBT radiation, the PTV–heart distance is significantly associated with the dose to the heart (p < 0.001), but it is not in DIBH radiotherapy using SIB. Conclusion: In daily practice, both DIBH radiation therapy as well as MCBT show a very low heart exposure and may thus reduce long term cardiac morbidity as compared to currently available long-term clinical data of patients treated with conventional tangential field plans in free breathing. Our analysis confirms particularly good cardiac sparing with MCBT-APBI, so that this technique should be offered to patients with left-sided breast cancer if the tumor-associated eligibility criteria are fulfilled. The availability of polygenic scores for type 2 diabetes (T2D) raises the question, whether assessing family history might become redundant. However, family history not only involves shared genetics, but also shared environment. It was the aim of this study to assess the independent and combined effects of one family risk score (FamRS) and a polygenic score (PGS) on prevalent and incident T2D risk in a population-based study from Germany (n = 3071). The study was conducted in 2004/2005 with up to 12 years of follow-up. The FamRS takes into account not only the number of diseased first grade relatives, but also age at onset of the relatives and age of participants. 256 prevalent and additional 163 incident T2D cases were recorded. Prevalence of T2D increased sharply for those within the top quantile of the PGS distribution resulting in an OR of 19.16 (p < 2 × 10
–16 ) for the top 20% compared to the remainder of the population, independent of age, sex, BMI, physical activity and FamRS. On the other hand, having a very strong family risk compared to average was still associated with an OR of 2.78 (p = 0.001), independent of the aforementioned factors and the PGS. The PGS and FamRS were only slightly correlated (r2 Spearman = 0.018). The combined contribution of both factors varied with varying age-groups, though, with decreasing influence of the PGS with increasing age. To conclude, both, genetic information and family history are relevant for the prediction of T2D risk and might be used for identification of high risk groups to personalize prevention measures. In discussing the ways in which we hold each other accountable for immoral conduct, philosophers have often focused on blame, aiming to specify adequate responses to wrongdoing. In contrast, theorizing about the ways we can appropriately respond to minor moral mistakes – i.e., criticizable conduct that is bad but not wrong – has largely been neglected. My first goal in this paper is, thus, to draw attention to this blind spot and argue that a separate account of blameless moral criticism is desirable. My second goal is to propose one way to explicate the contrast between blaming and blameless moral criticism in terms of the contrast between moral anger and moral disappointment: while moral anger, as many argue, is an appropriate response to moral wrongdoing, moral disappointment, but not moral anger, is an appropriate response to these minor moral mistakes. 16. Projected transverse momentum resummation in top-antitop pair production at LHC.

The transverse momentum distribution of the t t ¯ system is of both experimental and theoretical interest. In the presence of azimuthally asymmetric divergences, pursuing resummation at high logarithmic precision is rather demanding in general. In this paper, we propose the projected transverse momentum spectrum d σ t t ¯ / d q τ , which is derived from the classical q → T spectrum by integrating out the rejection component q τ ⊥ with respect to a reference unit vector τ → , to serve as an alternative solution to remove these asymmetric divergences, in addition to the azimuthally averaged case d σ t t ¯ / d ∣ q → T ∣ . In the context of the effective field theories, SCETII and HQET, we will demonstrate that in spite of the q τ ⊥ integrations, the leading asymptotic terms of d σ t t ¯ / d q τ still observe the factorisation pattern in terms of the hard, beam, and soft functions in the vicinity of qτ = 0 GeV. Then, with the help of the renormalisation group equation techniques, we carry out the resummation at NLL+NLO, N2LL+N2LO, and approximate N2LL′+N2LO accuracy on three observables of interest, d σ t t ¯ / d q T , in , d σ t t ¯ / d q T , out , and d σ t t ¯ / d Δ ϕ t t ¯ , within the domain M t t ¯ ≥ 400 GeV. The first two cases are obtained by choosing τ → parallel and perpendicular to the top quark transverse momentum, respectively. The azimuthal de-correlation Δ ϕ t t ¯ of the t t ¯ pair is evaluated through its kinematical connection to qT,out. This is the first time the azimuthal spectrum Δ ϕ t t ¯ is appraised at or beyond the N2LL level including a consistent treatment of both beam collinear and soft radiation. [ABSTRACT FROM AUTHOR]

17. Precise predictions for V + 2 jet backgrounds in searches for invisible Higgs decays.

We present next-to-leading order QCD and electroweak (EW) theory predictions for V + 2 jet production, with V = Z, W
± , considering both the QCD and EW production modes and their interference. We focus on phase-space regions where V + 2 jet production is dominated by vector-boson fusion, and where these processes yield the dominant irreducible backgrounds in searches for invisible Higgs boson decays. Predictions at parton level are provided together with detailed prescriptions for their implementation in experimental analyses based on the reweighting of Monte Carlo samples. The key idea is that, exploiting accurate data for W + 2 jet production in combination with a theory-driven extrapolation to the Z + 2 jet process can lead to a determination of the irreducible background at the few-percent level. Particular attention is devoted to the estimate of the residual theoretical uncertainties due to unknown higher-order QCD and EW effects and their correlation between the different V + 2 jet processes, which is key to improve the sensitivity to invisible Higgs decays. 19. Accelerating LHC event generation with simplified pilot runs and fast PDFs.

Poor computing efficiency of precision event generators for LHC physics has become a bottleneck for Monte-Carlo event simulation campaigns. We provide solutions to this problem by focusing on two major components of general-purpose event generators: The PDF evaluator and the matrix-element generator. For a typical production setup in the ATLAS experiment, we show that the two can consume about 80% of the total runtime. Using NLO simulations of p p → ℓ + ℓ - + jets and p p → t t ¯ + jets as an example, we demonstrate that the computing footprint of Lhapdf and Sherpa can be reduced by factors of order 10, while maintaining the formal accuracy of the event sample. Heme, an iron-protoporphyrin IX complex, is a cofactor bound to various hemoproteins and supports a broad range of functions, such as electron transfer, oxygen transport, signal transduction, and drug metabolism. 50 of about 4 nM under ambient and 63 nM under reducing conditions in a weakly voltage-dependent manner, favoring inhibition at resting potential. Functional studies on channel mutants and biochemical analysis of synthetic and recombinant channel fragments identified a heme-binding motif CxHx8 H in the C-linker region of the Kv10.1 C terminus, with cysteine 541 and histidines 543 and 552 being important for hemin binding. Binding of hemin to the C linker may induce a conformational constraint that interferes with channel gating. According to the standard view, a belief is based on a reason and doxastically justified—i.e., permissibly held—only if a causal relation obtains between a reason and the belief. In this paper, I argue that a belief can be doxastically justified by a reason’s mere disposition to sustain it. Such a disposition, however, wouldn’t establish a causal connection unless it were manifested. My argument is that, in the cases I have in mind, the manifestation of this disposition would add no positive epistemic feature to the belief: a belief that is justified after the manifestation of a reason’s causal powers must have already been justified before their manifestation. As a result, those who adhere to the standard causal view of the basing relation face a hard choice: they should either abandon the enormously popular view that doxastic justification has a basing requirement or modify their view of the basing relation. A new family of hybrid/mixed finite elements optimized for numerical stability is introduced. It comprises a linear hexahedral and quadratic hexahedral and tetrahedral elements. The element formulation is derived from a consistent linearization of a well-known three-field functional and related to Simo–Taylor–Pister (STP) elements. For the quadratic hexahedral and tetrahedral elements we derive (static reduced) discontinuous hybrid elements, as well as continuous mixed finite elements with additional primary unknowns for the hydrostatic pressure and the dilation, whereas the linear hexahedral element is of the discontinuous type. The elements can readily be used in combination with any isotropic, invariant-based hyperelastic material model and can be considered as being locking-free. In a representative numerical benchmark test the elements numerical stability is assessed and compared to STP-elements and the family of discontinuous hybrid elements implemented in the commercial finite element code Abaqus/Standard. We consider the production of a pair of Z bosons at the LHC and study the inclusion of EW corrections in theoretical predictions at fixed order and based on multijet-merged parton-shower simulations. To this end we present exact NLO EW results for pp → e
+ e− μ+ μ− , and, for the first time, for pp → e+ e− μ+ μ− j, and compare them to the EW virtual and NLL Sudakov approximation. We then match the exact NLO EW result to the resummed Sudakov logarithms to achieve an improved NLO EW + NLL EW sud exp result. Further, we discuss the inclusion of the above EW corrections in MePs@Nlo event simulations in the framework of the Sherpa event generator. Salience reasoning, many have argued, can help solve coordination problems, but only if such reasoning is supplemented by higher-order predictions, e.g. beliefs about what others believe yet others will choose. In this paper, I will argue that this line of reasoning is self-undermining. Higher-order behavioral predictions defeat salience-based behavioral predictions. To anchor my argument in the philosophical literature, I will develop it in response and opposition to the popular Lewisian model of salience reasoning in coordination games. This model imports the problematic higher-order beliefs by way of a 'symmetric reasoning' constraint. The data taken in Run II at the Large Hadron Collider have started to probe Higgs boson production at high transverse momentum. Future data will provide a large sample of events with boosted Higgs boson topologies, allowing for a detailed understanding of electroweak Higgs boson plus two-jet production, and in particular the vector-boson fusion mode (VBF). We perform a detailed comparison of precision calculations for Higgs boson production in this channel, with particular emphasis on large Higgs boson transverse momenta, and on the jet radius dependence of the cross section. We study fixed-order predictions at next-to-leading order and next-to-next-to-leading order QCD, and compare the results to NLO plus parton shower (NLOPS) matched calculations. The impact of the NNLO corrections on the central predictions is mild, with inclusive scale uncertainties of the order of a few percent, which can increase with the imposition of kinematic cuts. We find good agreement between the fixed-order and matched calculations in non-Sudakov regions, and the various NLOPS predictions also agree well in the Sudakov regime. We analyze backgrounds to VBF Higgs boson production stemming from associated production, and from gluon-gluon fusion. At high Higgs boson transverse momenta, the ∆y
jj and/or mjj cuts typically used to enhance the VBF signal over background lead to a reduced efficiency. We examine this effect as a function of the jet radius and using different definitions of the tagging jets. QCD radiative corrections increase for all Higgs production modes with increasing Higgs boson pT , but the proportionately larger increase in the gluon fusion channel results in a decrease of the gluon-gluon fusion background to electroweak Higgs plus two jet production upon requiring exclusive two-jet topologies. The cellular resting membrane potential (V
m ) not only determines electrical responsiveness of excitable cells but also plays pivotal roles in non-excitable cells, mediating membrane transport, cell-cycle progression, and tumorigenesis. Studying these processes requires estimation of Vm , ideally over long periods of time. Here, we introduce two ratiometric genetically encoded Vm indicators, rArc and rASAP, and imaging and analysis procedures for measuring differences in average resting Vm between cell groups. We investigated the influence of ectopic expression of K+ channels and their disease-causing mutations involved in Andersen-Tawil (Kir2.1) and Temple-Baraitser (KV 10.1) syndrome on median resting Vm of HEK293T cells. Real-time long-term monitoring of Vm changes allowed to estimate a 40–50 min latency from induction of transcription to functional Kir2.1 channels in HEK293T cells. The presented methodology is readily implemented with standard fluorescence microscopes and offers deeper insights into the role of the resting Vm in health and disease. Rühl et al. report the generation of ratiometric genetically encoded voltage indicators (GEVIs) and establish that they can be used in high-throughput automated imaging to measure compound membrane potential (Vm) in mammalian cells. The production of weak gauge bosons, W± and Z, are at the core of the LHC precision measurement program. Their transverse momentum spectra as well as their pairwise ratios are key theoretical inputs to many high-precision analyses, ranging from the W mass measurement to the determination of parton distribution functions. Owing to the different properties of the W and Z boson and the different accessible fiducial regions for their measurement, a simple one-dimensional correlation is insufficient to capture the differing vector and axial-vector dynamics of the produced lepton pair. We propose to correlate them in two observables, the transverse momentum qT of the lepton pair and its azimuthal separation ∆ϕ. Both quantities are purely transverse and therefore accessible in all three processes, either directly or by utilising the missing transverse momentum of the event. We calculate all the single-differential qT and ∆ϕ as well as the double-differential (qT, ∆ϕ) spectra for all three processes at N3LL′+N2LO accuracy, resumming small transverse momentum logarithms in the soft-collinear effective theory approach and including all singlet and non-singlet contributions. Using the double-differential cross sections we build the pairwise ratios ℛ W + / Z , ℛ W − / Z , and ℛ W + / W − and determine their uncertainties assuming fully correlated, partially correlated, and uncorrelated uncertainties in the respective numerators and denominators. In the preferred partially correlated case we find uncertainties of less than 1% in most phase space regions and up to 3% in the lowest qT region. [ABSTRACT FROM AUTHOR]

We present a comprehensive analysis of the cumulant lattice Boltzmann model with the three-dimensional Taylor–Green vortex benchmark at Reynolds number 1600. The cumulant model is investigated in several different variants, using regularization, fourth-order convergent diffusion and fourth-order convergent advection with and without limiters. In addition, a cumulant model combined with a WALE sub-grid scale model is being evaluated. The turbulence model is found to filter out the high wave number contributions from the energy spectrum and the enstrophy, while the non-filtered cumulant methods show good correspondence to spectral simulations even for the high wave numbers. The application of the WALE turbulence model appears to be counter productive for the Taylor–Green vortex at a Reynolds number of 1600. At much higher Reynolds numbers ( Re = 160 , 000 ) a deviation from the ideal Kolmogorov theory can be observed in the absence of an explicit turbulence model. The significance of the body-mind link and the contribution of nonverbal communication in the context of psychotherapy is enjoying increased interest. Yet, the main measurements used in psychotherapy studies rely mostly on verbal measures, missing other channels of communication that would allow rich and implicit, nonverbal information. The present study is a first step toward using the mirror game (MG) as a standardized mimicry task that involves the assessment of movement synchrony as a nonverbal outcome measure for psychotherapy research. The aims of the study were twofold: (a) to investigate the possibility of automated analysis of synchrony in the MG, detecting the different roles of leadership in the game, and (b) to validate the synchrony quantified by computer algorithms with a human-rated prosocial scale. All participants (N = 33) played the MG with a gender-matched expert player. Body motions of interacting pairs were assessed using motion energy analysis. Frequency of movement synchrony was computed by windowed cross-lagged correlation and a peak-picking algorithm. Independent observers rated items with respect to prosocial parameters, using the MG scales. The algorithm allowed to differentiate quite accurately between the various roles of leadership. Significant correlations were found between the MG human-rated scales and computer-detected synchrony. High frequency of synchronized movements was linked with high levels of having fun, shared affect, and reference to the other, and with low negative affect. The MG provides a standardized and flexible paradigm for the investigation of movement synchrony. We scrutinise the ability of the primary QED final-state resummation tools, combined with electroweak virtual corrections, to reproduce the exact next-to-leading order electroweak calculation in the four-charged-lepton final state. We further examine the dependence of the findings on the lepton-photon dressing-cone size as well as the resonance identification strategy. Overall we find excellent agreement with the fixed-order result, but partial differences not directly connected with resummation-induced higher-order effects at the few-percent level are observed in some cases, which are relevant for precision measurements. First, we present a combined analysis of pp → μ + v μ e − v ¯ e and pp → μ + v μ e − v ¯ e j at next-to-leading order, including both QCD and electroweak corrections. Second, we provide all-order predictions for pp → μ + v μ e − v ¯ e + jets using merged parton-shower simulations that also include approximate EW effects. A fully inclusive sample for WW production is compared to the fixed-order computations for exclusive zero- and one-jet selections. The various higher-order effects are studied in detail at the level of cross sections and differential distributions for realistic experimental set-ups. N-type inactivation of voltage-gated K
+ channels is conferred by the N-terminal "ball" domains of select pore-forming α subunits or of auxiliary β subunits, and influences electrical cellular excitability. Here, we show that hemin impairs inactivation of K+ channels formed by Kv3.4 α subunits as well as that induced by the subunits Kvβ1.1, Kvβ1.2, and Kvβ3.1 when coexpressed with α subunits of the Kv1 subfamily. In Kvβ1.1, hemin interacts with cysteine and histidine residues in the N terminus (C7 and H10) with high affinity (EC50 100 nM). Similarly, rapid inactivation of Kv4.2 channels induced by the dipeptidyl peptidase-like protein DPP6a is also sensitive to hemin, and the DPP6a mutation C13S eliminates this dependence. The results suggest a common mechanism for a dynamic regulation of Kv channel inactivation by heme/hemin in N-terminal ball domains of Kv α and auxiliary β subunits. Sleeping sickness is a fatal disease caused by the protozoan parasite Trypanosoma brucei (Tb). Inosine-5'-monophosphate dehydrogenase (IMPDH) has been proposed as a potential drug target, since it maintains the balance between guanylate deoxynucleotide and ribonucleotide levels that is pivotal for the parasite. Here we report the structure of TbIMPDH at room temperature utilizing free-electron laser radiation on crystals grown in living insect cells. The 2.80 Å resolution structure reveals the presence of ATP and GMP at the canonical sites of the Bateman domains, the latter in a so far unknown coordination mode. Consistent with previously reported IMPDH complexes harboring guanosine nucleotides at the second canonical site, TbIMPDH forms a compact oligomer structure, supporting a nucleotide-controlled conformational switch that allosterically modulates the catalytic activity. The oligomeric TbIMPDH structure we present here reveals the potential of in cellulo crystallization to identify genuine allosteric co-factors from a natural reservoir of specific compounds. Trypanosoma brucei inosine-5′-monophosphate dehydrogenase (IMPDH) is an enzyme in the guanine nucleotide biosynthesis pathway and of interest as a drug target. Here the authors present the 2.8 Å room temperature structure of TbIMPDH determined by utilizing X-ray free-electron laser radiation and crystals that were grown in insect cells and find that ATP and GMP are bound at the canonical sites of the Bateman domains. First, we've seen cases of forgiveness in which there was no such causal connection; but we haven't seen cases in which forgiveness did not require the appropriate form of endorsement. Photoresponsive hydrogels have been attractive because they can provide precise spatial and temporal control for molecule release, whereas the conventional preparation of photoresponsive hydrogels generally involves complex chemical synthesis steps or specific conditions which limits their practical applications. Herein, a new photoresponsive hydrogel is facilely prepared via co-assembly of two simple molecules, Fmoc-Phe-OH and Azp, without chemical synthesis. The co-assembly mechanism, morphology, and photoresponsiveness of (Fmoc-Phe-OH)-Azp hydrogel are investigated by circular dichroism (CD), ultraviolet-visible (UV-Vis), fluorescence, 1H nuclear magnetic resonance (
1 H-NMR), attenuated total internal reflection Fourier transform Infrared (ATR-FTIR) spectroscopy, and scanning electron microscopy (SEM). Furthermore, the enhanced release of encapsulated sulforhodamine B (SRB) dye molecules can be achieved via UV light irradiation. The enhanced dye release amount can be controlled by manipulating photoirradiation time. Fast N-type inactivation of voltage-gated K
+ (Kv) channels is important in fine-tuning of cellular excitability. To serve diverse cellular needs, N-type inactivation is regulated by numerous mechanisms. Here, we address how reactive sulfur species—the gaseous messenger H2 S and polysulfides—affect N-type inactivation of the mammalian Kv channels Kv1.4 and Kv3.4. In both channels, the H2 S donor NaHS slowed down inactivation with varying potency depending on the "aging" of NaHS solution. Polysulfides were > 1000 times more effective than NaHS with the potency increasing with the number of sulfur atoms (Na2 S2 < Na2 S3 < Na2 S4 ). In Kv1.4, C13 in the N-terminal ball domain mediates the slowing of inactivation. In recombinant protein exposed to NaHS or Na2 S4 , a sulfur atom is incorporated at C13 in the protein. In Kv3.4, the N terminus harbors two cysteine residues (C6, C24), and C6 is of primary importance for channel regulation by H2 S and polysulfides, with a minor contribution from C24. To fully eliminate the dependence of N-type inactivation on sulfhydration, both cysteine residues must be removed (C6S:C24S). Sulfhydration of a single cysteine residue in the ball-and-chain domain modulates the speed of inactivation but does not remove it entirely. In both Kv1.4 and Kv3.4, polysulfides affected the N-terminal cysteine residues when assayed in the whole-cell configuration; on-cell recordings confirmed that polysulfides also modulate K+ channel inactivation with undisturbed cytosol. These findings have collectively identified reactive sulfur species as potent modulators of N-type inactivation in mammalian Kv channels. [ABSTRACT FROM AUTHOR]- Published

Processes with three external electroweak gauge boson allow for a measurement of triple and quartic gauge couplings. They can be used to constrain anomalous gauge couplings, where new physics might predominantly couple to electroweak gauge bosons. In this paper we chose a class of such processes where we consider two photons and an additional vector boson in the final state. As additional vector boson we consider either a third photon or a W or Z boson. For the latter two cases we assume a leptonic decay of the boson. We calculate the next-to-leading order QCD and electroweak corrections to these processes with a particular emphasis on the until now unknown electroweak corrections. We find that the electroweak corrections to the total cross section are moderate in the range of a few per cent at most, but can reach several tens of per cent in regions of phase space that are particularly interesting in the context of new physics searches. In addition we investigate the difference between additive and multiplicative scheme when combining QCD and electroweak corrections and we assess the importance of photon induced contributions to these processes. We report on Scanning Thermal Lithography (SThL), a recently introduced lithographic tool, for local thermochemistry on tert-butyl acrylate based polymer films featuring chemical cross links. The tailored polymer films afford platforms for controlled high molecular density coupling and surface immobilization of biologically relevant molecules, such as proteins. The thermally labile tert-butyl ester groups in tert-butyl acrylate based polymer films can be cleaved in air at temperatures above 150 °C to yield carboxylic acid functional groups for further (bio)- conjugation. The films were optimized to avoid plastic deformation at the elevated temperatures used during SThL. Exploiting these properties patterns with length scales as small as 35 ± 6 nm have been successfully thermally activated with SThL. Hence SThL comprises an attractive approach for the development of e.g. (bio)sensors and platforms for cell surface interaction studies with nanoscale patterns.
