Inland water bodies, being the most active biogeochemical cycle reactors, play a pivotal role in the global carbon cycle and CO2 budget. This study integrates existing observation dataset of CO2 flux (fCO2) in rivers, lakes and reservoirs in China, to elucidate their spatial-temporal patterns and key influencing factors and to reappraise the significance for regional carbon balance. It showed that, the fCO2 in rivers, lakes and reservoirs in China presented significant variability with large range of −379.3–4947.6, −160.1–785.0 and −74.0–1603.1 mg CO2 m−2 h−1, respectively. The median of the fCO2 in rivers was of 228.5 mg CO2 m−2 h−1, observably higher than these in lakes and reservoirs (26.0 and 28.3 mg CO2 m−2 h−1, respectively). The fCO2 in rivers and reservoirs exhibited similar decreasing trend from south to north as a result of universal climate restraint, the averaged fCO2 in Pearl River and Yangtze River basins showed much higher than that in Northeastern rivers. While, the averaged fCO2 in the Mongolia-Xinjiang lake district and the Northeast lake district were higher than other lake districts, followed by the Qinghai-Xizang lake district, and the Eastern and Yungui lake district were generally low, contradicting the climatic restriction. The water primary production enhancement resulting from human activities was the main driver of spatial variation in the fCO2 in lakes. Meanwhile, the fCO2 in rivers presented seasonal pattern with higher wet season than dry season, while opposite patterns were found in lakes and reservoirs. Seasonal temperature, precipitation and water primary production were main factors. Furthermore, it showed pH was a key factor indicating the variability of the fCO2 levels either in rivers, lakes or in reservoirs. In lakes, the fCO2 is closely linked to chlorophyll a (Chl-a) and dissolved oxygen (DO), whereas, the fCO2 in rivers is primarily associated with organic carbon (OC) and total nitrogen (TN), highlighting the diverse controlling mechanisms of fCO2 in various inland water types. In addition, we found that water body sizes play an important role in regulating the fCO2 levels, and small waters act as hotspots of CO2 flux. Additionally, widespread urbanization and agricultural activities may enhance CO2 emissions from rivers but potentially mitigating that from lakes. Nevertheless, the comprehensive impact of these factors on CO2 emissions in inland water requires further evaluation. Based on the extrapolation method, we re-estimated that the total CO2 emission of inland waters in China is approximately 117.3 Tg yr−1, which could offset 4.6%–12.8% of the total land carbon sink in China. Vitamin A, also known as retinol, is a fat-soluble vitamin that plays crucial role in various physiological functions In vivo. However, factors such as light, oxygen, and others may impact the stability of VA. To enhance its stability. This study microencapsulated VA, Gelatin, carboxymethyl cellulose, and salt were mixed in a ratio of 5:1:0.1 as the shell material. Additionally, 12% TG and 3.5% sucrose ester were added with core–shell ratio of 1:8. The experimental results indicated that VA microcapsules exhibited an encapsulation efficiency of 81.12%, after 9 weeks of storage this rate decreased to 75.38%, and the encapsulated VA oil did not exhibit extravasation. The addition of an appropriate amount of salt to the shell material enhanced the mechanical properties of the shell material, compared to the shell material without added salt, the leakage of VA in the salt-added sample decreased by 5.8% for 30 min and 14.5% for 60 min. In vitro release experiments showed that after 3 h of incubation in simulated gastric fluid, the microcapsules had an 18.52% release rate. Background: Tumor microenvironment actually reduces antitumor effect against the immune attack by exclusion of CD8+T cells. Progranulin (PGRN) is a multifunctional growth factor with significant pathological effects in multiple tumors; however, its role in immunity evasion of breast cancer (BCa) is not completely understood. Methods: We depleted GRN (PGRN gene) genetically in mice or specifically in PY8119 murine BCa cell line, and mouse models of orthotopic or subcutaneous transplantation were used. Chimeric mice-deficient of PGRN (Grn−/−) in bone marrow (BM) compartment was also generated. Association of PGRN expression with chemokine production or BCa development was investigated by histological and immunological assays. Results: We found PGRN was involved in exhaustion of cytotoxic CD8+T cell in BCa with the increasing expressions of M2 markers and intercellular cell adhesion molecule-1 (ICAM-1) on macrophages. Specifically, ablation of PGRN in PY8119 cells reduced tumor burden, accompanied by the infiltrating of cytotoxic CD8+T cells into tumor nests. Moreover, our result revealed that blockade of PD-1 in PGRN-depleted tumors exhibited better antitumor effect in vivo and significantly decreased tumor burden. Conclusion: These findings suggest that inhibition of PGRN may act as a potential immune-therapeutic strategy by recovering infiltration of CD8+T cell in BCa tissue and thereby enhancing the response to anti-PD-1 therapy. Although structural family theory proposes that family boundaries play a significant role in marital relationships, limited empirical evidence supports this theoretical model. The present study tested structural family theory by examining the complex associations between intergenerational boundaries and adult children's marital satisfaction, as well as the indirect effect of family adaptability and cohesion. A nonclinical convenience sample of individuals (333 participants, 168 females) married for at least one year was recruited. Participants provided self-report data on demographic variables, family boundaries, family cohesion and adaptability, and marital satisfaction. The internal intrusion score (intrusion by families of origin on the nuclear family) of intergenerational family boundaries was negatively associated with marital satisfaction, while the external intrusion score (intrusion by the nuclear family on families of origin) was positively associated with marital satisfaction. The effect of intergenerational family boundaries on marital satisfaction was exerted indirectly through family cohesion and adaptability. There was a significant association between intergenerational family boundaries and adult children's marital satisfaction. This study contributes to the conceptual framework of structural family theory, enabling family therapy practitioners to better understand the pathways through which intergenerational family boundaries impact adult children's marital satisfaction. Sesame, a prized oil crop, is acknowledged for its distinctive aroma, health benefits, and copious antioxidants, including sesamin and sesamol. Despite the importance of features like wrinkled leaf and pollen sterility in sesame breeding, scant research into the former impedes progress in cultivation and production. This study endeavors to illuminate the etiology of wrinkled leaves in sesame by probing agronomic and biochemical factors like hydration, nutrient makeup, antioxidant enzymes, and hormones. The investigation found substantial disparities between mutant wrinkled leaves and normal ones; the mutants exhibited heightened water content (86.60% vs 84.99%), augmented dry leaf weight, greater stomata density per unit area (174.58 vs 146.32 cm–2), and a significantly amplified carotenoid content by 27.5%. The study deduced that deficient gibberellic acid (GA3) production in mutant leaves during the seedling stage (< 0.005 ng g−1) restricts leaf elongation, causing wrinkles. Moreover, abnormal auxin (IAA) production during the seedling stage (< 8.57 ng g−1) and deficient jasmonic acid (JA) in both central and flat leaves (< 12.0 ng g−1) were associated with wrinkling. Furthermore, excessive late-stage GA3 production (> 0.041 ng g−1) led to persistent wrinkling and postponed maturity. This research, the first of its kind, elucidates the biochemical origins of sesame's wrinkled leaf formation, offering insights into the mechanisms of this trait and potentially facilitating the creation of new sterile lines and endorsement of sesame as a fresh-leaf vegetable. Myocardial infarction (MI) is the leading cause of premature death. The death of cardiomyocytes (CMs) and the dysfunction of the remaining viable CMs are the main pathological factors contributing to heart failure (HF) following MI. This study aims to determine the transcriptional profile of CMs and investigate the heterogeneity among CMs under hypoxic conditions. Single-cell atlases of the heart in both the sham and MI groups were developed using single-cell data (GSE214611) downloaded from Gene Expression Omnibus (GEO) database (https://www.ncbi.nlm.nih.gov/geo/). The heterogeneity among CMs was explored through various analyses including enrichment, pseudo time, and intercellular communication analysis. The marker gene of C5 was identified using differential expression analysis (DEA). Real-time polymerase chain reaction (RT-PCR), bulk RNA-sequencing dataset analysis, western blotting, immunohistochemical and immunofluorescence staining, Mito-Tracker staining, TUNEL staining, and flow cytometry analysis were conducted to validate the impact of the marker gene on mitochondrial function and cell apoptosis of CMs under hypoxic conditions. We identified a cell subcluster named C5 that exhibited a close association with mitochondrial malfunction and cellular apoptosis characteristics, and identified Slc25a4 as a significant biomarker of C5. Furthermore, our findings indicated that the expression of Slc25a4 was increased in failing hearts, and the downregulation of Slc25a4 improved mitochondrial function and reduced cell apoptosis. Our study significantly identified a distinct subcluster of CMs that exhibited strong associations with ventricular remodeling following MI. Background: Within the tumor microenvironment, endothelial cells hold substantial sway over bladder cancer (BC) prognosis. Herein, we aim to elucidate the impact of endothelial cells on BC patient outcomes by employing an integration of single-cell and bulk RNA sequencing data. Methods: All data utilized in this study were procured from online databases. R version 3.6.3 and relevant packages were harnessed for the development and validation of an endothelial-associated prognostic index (EPI). Results: EPI was formulated, incorporating six genes (CYTL1, FAM43A, GSN, HSPG2, RBP7, and SLC2A3). EPI demonstrated significant prognostic value in both The Cancer Genome Atlas (TCGA) and externally validated dataset. Functional results revealed a profound association between EPI and endothelial cell functionality, as well as immune-related processes. Our findings suggest that patients with low-risk EPI scores are more likely to respond positively to immunotherapy, as indicated by immune checkpoint activity, immune infiltration, tumor mutational burden, stemness index, TIDE, and IMvigor210 analyses. Conversely, individuals with high-risk EPI scores exhibited heightened sensitivity to cisplatin, docetaxel, and gemcitabine treatment regimens. The eye, an anatomical extension of the central nervous system (CNS), exhibits many molecular and cellular parallels to the brain. Emerging research demonstrates that changes in the brain are often reflected in the eye, particularly in the retina1. Still, the possibility of an immunological nexus between the posterior eye and the rest of the CNS tissues remains unexplored. Here, studying immune responses to herpes simplex virus in the brain, we observed that intravitreal immunization protects mice against intracranial viral challenge. This protection extended to bacteria and even tumours, allowing therapeutic immune responses against glioblastoma through intravitreal immunization. We further show that the anterior and posterior compartments of the eye have distinct lymphatic drainage systems, with the latter draining to the deep cervical lymph nodes through lymphatic vasculature in the optic nerve sheath. This posterior lymphatic drainage, like that of meningeal lymphatics, could be modulated by the lymphatic stimulator VEGFC. Conversely, we show that inhibition of lymphatic signalling on the optic nerve could overcome a major limitation in gene therapy by diminishing the immune response to adeno-associated virus and ensuring continued efficacy after multiple doses. Deoxynivalenol (DON) poses significant challenges due to its frequent contamination of grains and associated products. Microbial strategies for mitigating DON toxicity showed application potential. Eight bacterial isolates with DON degradation activity over 5% were obtained from various samples of organic fertilizer in this study. One of the isolates emerged as a standout, demonstrating a substantial degradation capability, achieving a 99.21% reduction in DON levels. This isolate, underwent thorough morphological, biochemical, and molecular characterization to confirm its identity, and was identified as a new strain of Achromobacter spanius P-9. Subsequent evaluations revealed that the strain P-9 retains its degradation activity after a 24-h incubation, reaching optimal performance at 35 °C with a pH of 8.0. Further studies indicated that Ca2+ ions enhance the degradation process, whereas Zn2+ ions exert an inhibitory effect. Background and aims: T cells are master effectors of anti-tumor immunity in cancer. Recent studies suggest that altered lipid metabolism imposed by the tumor microenvironment constrains anti-tumor immunity. However, the tumor-associated lipid species changes that dampen T cell ability to control tumor progression are not fully understood. Here, we plan to clarify the influences of distinctly altered lipid components in hepatitis B virus (HBV)-related hepatocellular carcinoma (HCC) on T-cell function, aiming to seek lipid metabolic targets for improving T cell anti-tumor effects. Methods: Tumor tissues and non-tumor liver from HCC patients were collected for RNA-sequencing, lipid profiling and T cell characterizing, followed by correlation analysis. Additionally, the effects of significantly changed lipid components on anti-tumor potential of T cells were tested by in vitro cell experiments and/or in vivo tumor inoculated model. Results: Altered lipid metabolism coincides with impaired T cell response in HBV-related HCC. Characteristic lipid composition, significantly marked by accumulation of long-chain acylcarnitines (LCACs) and reduction of lysophosphatidylcholines (LPCs), are found in the tumor tissue. Notably, LCACs accumulated are associated with T cells exhaustion and deficient functionality, while LPCs correlate to anti-tumor effects of T cells. In particular, supplement of LPCs, including LPC (20:0) and LPC (22:0), directly promote the activation and IFN-γ secretion of T cells in vitro, and suppress tumor growth in vivo. Conclusions: Our study highlights the distinctly changed lipid components closely related to T cell dysregulation in HCC, and suggests a promising strategy by decreasing LCACs and increasing LPCs for anti-tumor immunotherapy. Owing to the specific electronic-redistribution and spatial proximity, diatomic catalysts (DACs) have been identified as principal interest for efficient photoconversion of CO2 into C2H4. However, the predominant bottom-up strategy for DACs synthesis has critically constrained the development of highly ordered DACs due to the random distribution of heteronuclear atoms, which hinders the optimization of catalytic performance and the exploration of actual reaction mechanism. Here, an up-bottom ion-cutting architecture is proposed to fabricate the well-defined DACs, and the superior spatial proximity of CuAu diatomics (DAs) decorated TiO2 (CuAu-DAs-TiO2) is successfully constructed due to the compact heteroatomic spacing (2-3 Å). Owing to the profoundly low C-C coupling energy barrier of CuAu-DAs-TiO2, a considerable C2H4 production with superior sustainability is achieved. Genetic variations in salt stress tolerance identified in rapeseed genotypes highlight the complicated regulatory mechanisms. Westar is ubiquitously used as a transgenic receptor cultivar, while ZS11 is widely grown as a high-production and good-quality cultivar. In this study, Westar was found to outperform ZS11 under salt stress. Through cell component isolation, non-invasive micro-test, X-ray energy spectrum analysis, and ionomic profile characterization, pectin demethylation-mediated cell wall Na+ retention was proposed to be a major regulator responsible for differential salt tolerance between Westar and ZS11. Integrated analyses of genome-wide DNA variations, differential expression profiling, and gene co-expression networks identified BnaC9.PME47, encoding a pectin methylesterase, as a positive regulator conferring salt tolerance in rapeseed. BnaC9.PME47, located in two reported QTL regions for salt tolerance, was strongly induced by salt stress and localized on the cell wall. Natural variation of the promoter regions conferred higher expression of BnaC9.PME47 in Westar than in several salt-sensitive rapeseed genotypes. Loss of function of AtPME47 resulted in the hypersensitivity of Arabidopsis plants to salt stress. The integrated multiomics analyses revealed novel insights into pectin demethylation-mediated cell wall Na+ retention in regulating differential salt tolerance in allotetraploid rapeseed genotypes. The presence of oral microbes in extra-oral sites is linked to gastrointestinal cancers. However, their potential ectopically colonization in the nasopharynx and impact on local cancer development remains uncertain. Our study involving paired nasopharyngeal-oral microbial samples from nasopharyngeal carcinoma (NPC) patients and controls unveils an aberrant oral-to-nasopharyngeal microbial translocation associated with increased NPC risk (OR = 4.51, P = 0.012). Thirteen species are classified as oral-translocated and enriched in NPC patients. Among these, Fusobacterium nucleatum and Prevotella intermedia are validated through culturomics and clonal strain identification. Nasopharyngeal biopsy meta-transcriptomes confirm these microbes within tumors, influencing local microenvironment and cytokine response. These microbes correlate significantly with the Epstein-Barr virus (EBV) loads in the nasopharynx, exhibiting an increased dose-response relationship. Collectively, our study identifies oral microbes migrating to the nasopharynx, infiltrating tumors, impacting microenvironments and linking with EBV infection. These results enhance our understanding of abnormal microbial communication and their roles in carcinogenesis. Oral microbes in non-oral locations are noted across various cancers. Epilepsy is a complex and multifaceted neurological disorder characterized by spontaneous and recurring seizures. It poses significant therapeutic challenges due to its diverse etiology and often-refractory nature. This comprehensive review highlights the pivotal role of AMP-activated protein kinase (AMPK), a key metabolic regulator involved in cellular energy homeostasis, which may be a promising therapeutic target for epilepsy. Current therapeutic strategies such as antiseizure medication (ASMs) can alleviate seizures (up to 70%). However, 30% of epileptic patients may develop refractory epilepsy. Due to the complicated nature of refractory epilepsy, other treatment options such as ketogenic dieting, adjunctive therapy, and in limited cases, surgical interventions are employed. These therapy options are only suitable for a select group of patients and have limitations of their own. Current treatment options for epilepsy need to be improved. Emerging evidence underscores a potential association between impaired AMPK functionality in the brain and the onset of epilepsy, prompting an in-depth examination of AMPK's influence on neural excitability and ion channel regulation, both critical factors implicated in epileptic seizures. AMPK activation through agents such as metformin has shown promising antiepileptic effects in various preclinical and clinical settings. These effects are primarily mediated through the inhibition of the mTOR signaling pathway, activation of the AMPK-PI3K-c-Jun pathway, and stimulation of the PGC-1α pathway. Despite the potential of AMPK-targeted therapies, several aspects warrant further exploration, including the detailed mechanisms of AMPK's role in different brain regions, the impact of AMPK under various conditional circumstances such as neural injury and zinc toxicity, the long-term safety and efficacy of chronic metformin use in epilepsy treatment, and the potential benefits of combination therapy involving AMPK activators. In contrast to rodents, the mechanisms underlying human trophectoderm and early placenta specification are understudied due to ethical barriers and the scarcity of embryos. Recent reports have shown that human pluripotent stem cells (PSCs) can differentiate into trophectoderm (TE)-like cells (TELCs) and trophoblast stem cells (TSCs), offering a valuable in vitro model to study early placenta specification. Here, we demonstrate that the VGLL1 (vestigial-like family member 1), which is highly expressed during human and non-human primate TE specification in vivo but is negligibly expressed in mouse, is a critical regulator of cell fate determination and self-renewal in human TELCs and TSCs derived from naïve PSCs. Parents and adolescents often hold discrepant perceptions of parental emotion socialization, which reflect misunderstandings in parent-adolescent communication on emotions and have potential detrimental effects on mental health of adolescents. The present study investigated the associations between parent-adolescent congruence and discrepancy in parental emotion socialization perception to two specific negative emotions (anger and sadness) and depressive symptoms in Chinese adolescents. A total of 372 adolescents (48.4% female, Mage = 13.43, SDage = 0.49) and their parents (79.6% mother, Mage = 41.15, SDage = 5.46) participated in this study. Both parents and adolescents reported perceived parental emotion socialization to anger and sadness, and adolescents reported depressive symptoms. Data were analyzed using polynomial regression and response surface analyses. Both congruence and discrepancy in parent and adolescent's reports were associated with adolescent depressive symptoms. A higher level of adolescent depressive symptoms was associated with higher parent-adolescent congruence in supportive responses to anger, sadness, and nonsupportive responses to anger. A higher level of depressive symptoms was associated with inconsistent reporting of supportive responses to sadness and nonsupportive responses to anger (only when parents had a more positive view than adolescents). This study highlights the significance of evaluating parent-child communication process by assessing perceived emotion socialization from both parents and adolescents and analyzing the reporting congruence and discrepancy. This paper presents a temperature-insensitive optical fiber strain sensor composed of single-mode fiber (SMF), no core fiber (NCF), and few-mode fiber (FMF). The NCF is fused to two FMFs with core-offset, exciting higher-order modes and generating mode interference. Experimental results demonstrate that the strain sensitivity of the optical fiber sensor can reach − 19.19 pm/με in a strain range from 0 to 600 με. Moreover, within a temperature range from 30℃ to 70℃, the strain-temperature crosstalk of the proposed sensor is remarkably low at 0.104 με/℃. Purpose: To develop and validate a machine learning (ML)-based prediction model for acute kidney injury (AKI) in patients with liver cirrhosis. Methods: Data on liver cirrhosis patients were extracted from the Medical Information Mart for Intensive Care III (MIMIC-III) and MIMIC-IV databases in this retrospective cohort study. ML algorithms, including random forest (RF), extreme gradient boosting (XGB), light gradient boosting machine (LGBM), and gradient boosting decision tree (GBDT) were applied to construct prediction models. Predictors were screened via univariate logistic regression, and then the models were developed with all data of the included patients. A bootstrap resampling method was adopted to validate the models. The predictive abilities of our final model were compared with those of the sequential organ failure assessment score (SOFA), simplified acute physiology score II (SAPS II), Model for End-stage Liver Disease (MELD), and MELD Na. Results: This study included 950 patients, of which 429 (45.16%) had AKI. Mechanical ventilation, vasopressor, international normalized ratio (INR), bilirubin, Charlson comorbidity index (CCI), prothrombin time (PT), estimated glomerular filtration rate (EGFR), partial thromboplastin time (PTT), and heart rate served as predictors. In the derivation set, the developed RF [area under curve (AUC) = 0.747], XGB (AUC = 0.832), LGBM (AUC = 0.785), and GBDT (AUC = 0.811) models exhibited significantly greater predictive performance than the logistic regression model (AUC = 0.699) (all P < 0.05). Among the ML-based models, the XGB model had the greatest AUC. In internal validation, the predictive capacity of the XGB model (AUC = 0.833) was significantly superior to that of the logistic regression model (AUC = 0.701) (P = 0.045). Hence, the XGB model was selected as the final model for AKI prediction. In contrast to the XGB model (AUC = 0.832), the SOFA (AUC = 0.609), MELD (AUC = 0.690), MELD Na (AUC = 0.690), and SAPS II (AUC = 0.641) had significantly lower predictive abilities in the derivation set (all P < 0.001). The XGB model was internally validated to have an AUC of 0.833, which was significantly higher than the SOFA (AUC = 0.609), MELD (AUC = 0.690), MELD Na (AUC = 0.688), and SAPS II (AUC = 0.641) (all P < 0.05). Conclusion: The XGB model had a better performance than the logistic regression model, SOFA, MELD, MELD Na, and SAPS II in AKI prediction for cirrhosis patients, which may help identify patients at a risk of AKI, and then provide timely interventions. Although considerable research has shown that attachment is a protective factor for cyberbullying, little research has investigated the mechanisms underlying this relationship among college students. This study examined whether loneliness mediates the association between parental attachment and cyberbullying and whether this process is moderated by interdependent self. A sample of 1125 college students (Mage = 19.14, SDage = 1.52 years) in China completed a questionnaire measuring parental attachment, cyberbullying, loneliness, and interdependent self. Loneliness partially mediated the relationship between parental attachment and cyberbullying. Moreover, the mediation effect of loneliness was moderated by interdependent self. A moderated mediation analysis further revealed that interdependent-self moderated the predictive effect of loneliness on cyberbullying. Specifically, the predictive effect of loneliness on cyberbullying was only significant among college students with low interdependent self. The study highlights the complex nature of the association between parental attachment and cyberbullying. Background: The utility of head computed tomography (CT) in predicting elevated intracranial pressure (ICP) is known to be limited in traumatic brain injury; however, few data exist in patients with spontaneous intracranial hemorrhage. Methods: We conducted a retrospective review of prospectively collected data in patients with nontraumatic intracranial hemorrhage (subarachnoid hemorrhage [SAH] or intraparenchymal hemorrhage [IPH]) who underwent external ventricular drain (EVD) placement. Head CT scans performed immediately prior to EVD placement were quantitatively reviewed for features suggestive of elevated ICP, including temporal horn diameter, bicaudate index, basal cistern effacement, midline shift, and global cerebral edema. The modified Fisher score (mFS), intraventricular hemorrhage score, and IPH volume were also measured, as applicable. We calculated the accuracy, positive predictive value (PPV), and negative predictive value (NPV) of these radiographic features for the coprimary outcomes of elevated ICP (> 20 mm Hg) at the time of EVD placement and at any time during the hospital stay. Multivariable backward stepwise logistic regression analysis was performed to identify significant radiographic factors associated with elevated ICP. Results: Of 608 patients with intracranial hemorrhages enrolled during the study time frame, 243 (40%) received an EVD and 165 (n = 107 SAH, n = 58 IPH) had a preplacement head CT scan available for rating. Elevated opening pressure and elevated ICP during hospitalization were recorded in 48 of 152 (29%) and 103 of 165 (62%), respectively. The presence of ≥ 1 radiographic feature had only 32% accuracy for identifying elevated opening pressure (PPV 30%, NPV 58%, area under the curve [AUC] 0.537, 95% asymptotic confidence interval [CI] 0.436–0.637, P = 0.466) and 59% accuracy for predicting elevated ICP during hospitalization (PPV 63%, NPV 40%, AUC 0.514, 95% asymptotic CI 0.391–0.638, P = 0.820). There was no significant association between the number of radiographic features and ICP elevation. Head CT scans without any features suggestive of elevated ICP occurred in 25 of 165 (15%) patients. However, 10 of 25 (40%) of these patients had elevated opening pressure, and 15 of 25 (60%) had elevated ICP during their hospital stay. In multivariable models, mFS (adjusted odds ratio [aOR] 1.36, 95% CI 1.10–1.68) and global cerebral edema (aOR 2.93, 95% CI 1.27–6.75) were significantly associated with elevated ICP; however, their accuracies were only 69% and 60%, respectively. All other individual radiographic features had accuracies between 38 and 58% for identifying intracranial hypertension. Conclusions: More than 50% of patients with spontaneous intracranial hemorrhage without radiographic features suggestive of elevated ICP actually had ICP > 20 mm Hg during EVD placement or their hospital stay. The changes in landscape ecological risk (LER) of the Qinghai–Tibet Plateau (QTP) profoundly affect the ecological environment of China and the world. We measured the evolution of the LER level and its driving factors through the past 40 years using meteorological data, population density information, and land use data acquired through remote sensing monitoring techniques spanning the years 1980 to 2020. Several key findings were derived: (1) The overall LER of the QTP was at a medium level during 1980–2020, with a fluctuating but decreasing overall trend. (2) Between 1980 and 2020, the spatial distribution of LER in the QTP was high in the west and low in the east; the LER level of the six provinces (districts) showed an overall decrease. (3) During 2000–2020, the LER of the QTP was influenced by a complex mechanism of action. The interactions between different influencing factors were mainly non-linear reinforcement and two-factor reinforcement, and factor interaction significantly enhanced the effect on LER. The minority people panmicrobial community database (MPPCD website: http://mppmcdb.cloudna.cn/) is the first microbe-disease association database of Chinese ethnic minorities. To research the relationships between intestinal microbes and diseases/health in the ethnic minorities, we collected the microbes of the Han people for comparison. Based on the data, such as age, among the different ethnic groups of the different regions of Sichuan Province, MPPCD not only provided the gut microbial composition but also presented the relative abundance value at the phylum, class, order, family and genus levels in different groups. In addition, differential analysis was performed in different microbes in the two different groups, which contributed to exploring the difference in intestinal microbe structures between the two groups. Meanwhile, a series of related factors, including age, sex, body mass index, ethnicity, physical condition, and living altitude, were included in the MPPCD, with special focus on living altitude. To date, this is the first intestinal microbe database to introduce altitude features. In conclusion, we hope that MPPCD will serve as a fundamental research support for the relationship between human gut microbes and host health and disease, especially in ethnic minorities. The leaf is the most important organ for photosynthesis; a moderately curly leaf is beneficial for improving the light energy utilization rate. Male sterility is another key trait that can facilitate hybrid seed production. Previously we have developed a mutant line JQA showing wrinkled leaves and male sterility in sesame (Sesamum indicum L.), which is an important genetic stock for the development of super variety with ideal plant type. Before it can be fully utilized in breeding practice, a better understanding of the cytological basis for leaf and anther development in JQA is required. In this study, we investigated the developmental processes in JQA at three cytological levels. Histological observation showed that the wrinkled leaf trait was expressed during the whole life cycle. Compared with normal leaves, the JQA wrinkled leaves have much fewer chloroplasts in the differentiated palisade tissue, which affect the photosynthesis efficiency by reducing the number of chloroplasts and delaying their development, and finally leads to leaf wrinkle. In addition, transmission electron microscope observation showed that most of the microspore mother cells in JQA could not divide evenly into dyads and tetrads during meiosis and that the tapetal cells could not disintegrate in time to provide adequate nutrition for the microspores. Consequently, pollens and pollen sacs were impeded or degraded due to a severe lack of nutrition. Background: Amyotrophic lateral sclerosis (ALS) is a devastating neurodegenerative disease. There is no cure currently. The discovery that mutations in the gene SOD1 are a cause of ALS marks a breakthrough in the search for effective treatments for ALS. SOD1 is an antioxidant that is highly expressed in motor neurons. Human SOD1 is prone to aberrant modifications. Familial ALS-linked SOD1 variants are particularly susceptible to aberrant modifications. Once modified, SOD1 undergoes conformational changes and becomes misfolded. This study aims to determine the effect of selective removal of misfolded SOD1 on the pathogenesis of ALS. Methods: Based on the chaperone-mediated protein degradation pathway, we designed a fusion peptide named CT4 and tested its efficiency in knocking down intracellularly misfolded SOD1 and its efficacy in modifying the pathogenesis of ALS. Results: Expression of the plasmid carrying the CT4 sequence in human HEK cells resulted in robust removal of misfolded SOD1 induced by serum deprivation. Co-transfection of the CT4 and the G93A-hSOD1 plasmids at various ratios demonstrated a dose-dependent knockdown efficiency on G93A-hSOD1, which could be further increased when misfolding of SOD1 was enhanced by serum deprivation. Application of the full-length CT4 peptide to primary cultures of neurons expressing the G93A variant of human SOD1 revealed a time course of the degradation of misfolded SOD1; misfolded SOD1 started to decrease by 2 h after the application of CT4 and disappeared by 7 h. Intravenous administration of the CT4 peptide at 10 mg/kg to the G93A-hSOD1 reduced human SOD1 in spinal cord tissue by 68% in 24 h and 54% in 48 h in presymptomatic ALS mice. Intraperitoneal administration of the CT4 peptide starting from 60 days of age significantly delayed the onset of ALS and prolonged the lifespan of the G93A-hSOD1 mice. Conclusions: The CT4 peptide directs the degradation of misfolded SOD1 in high efficiency and specificity. Selective removal of misfolded SOD1 significantly delays the onset of ALS, demonstrating that misfolded SOD1 is the toxic form of SOD1 that causes motor neuron death. Anaerobic digestion is a promising technology for energy recovery from secondary sludge, yet the presence of humic substances in wastewater limits anaerobic digestion. In particular, humic substances make secondary sludge denser and more compact, reducing the availability of organic matter for biodegradation. Here we review the impact of humic substances on the anaerobic process, with emphasis on humic substances properties, effect on sludge structure and composition, effect on hydrolysis, acidolysis and methanogenesis, evolution of humic substrances, and strategies to counteract negative impacts. Strategies include removing humic substances, pretreatment of secondary sludge prior anaerobic digestion, and addition of metal salts, enzymes and organisms. We observed that humic substances with a high E4/E6 ratio, representing the absorbance determined at 465 nm and 665 nm, with a low carbon/nitrogen ratio, and with a low aromaticity are easier to digest anaerobically. The liquid–solid phases distribution of humic substances influences the efficiency of anaerobic digestion, and the repolymerisation of humic substances during anaerobic digestion reduces sludge degradability. The traditional structural design process of rural buildings requires a lot of trial calculations and iterations of modelling work. However, rural buildings are subject to construction costs and cannot be professionally designed and checked by designers like urban buildings. Their safety and economy are difficult to meet the requirements. Therefore, an intelligent design concept of rural light steel frame structure was proposed, including intelligent modeling and intelligent optimization. Based on the automatic layer recognition method and optical character recognition technology, the BIM intelligent modeling method of rural light steel frame structure corresponding to the standard atlas of rural buildings was proposed, including layer recognition, axis text data extraction, primary selection and automatic arrangement of components. The intelligent modeling results basically met the requirements of practical application. Based on the proposed two-stage genetic algorithm, an intelligent optimization method of rural light steel frame structure was given. The optimization speed was fast and the optimization effect was good. The proposed intelligent design method was verified by practical case. The results showed that the proposed intelligent design concept of rural light steel frame structure was feasible. Austenitic S30408 stainless steel exhibits good low-temperature resistance and good welding performance. This steel is often used in liquefied natural gas stainless steel storage tanks. During the construction process, the tank wall is primarily connected by butt weld joints. Because welded joints are easily affected by temperature, low-temperature weld cracking can reduce the safety of structures. To study the cryogenic mechanical properties of austenitic S30408 stainless steel welded joints at low temperatures, the low-temperature mechanical properties of austenitic S30408 stainless steel base metal and welded joint components were studied by tensile tests from − 60 to 20 °C and scanning electron microscopy analysis of fractures at various temperatures. The results show that when the temperature decreases, the stress–strain curve of base metal components changes from a power function type to an inverted "s" type; in addition, secondary hardening occurs. The yield strength and tensile strength of the welded joint and base metal increased with decreasing temperature, and the elongation and reduction of area decreased. The plastic deformation capacity of the welded joint was significantly lower than that of the base metal, and there were obvious inclusions in the microstructure. [ABSTRACT FROM AUTHOR]

Unexpected benefits to the catalytic performance of materials often originate from the presence of surface defects. Here, novel D-penicillamine modified molybdenum oxide nanodots, with abundant oxygen vacancy defects, were fabrication by a mild, simple, and cost-effective method. Ultraviolet—visible (UV—Vis) absorption spectra analysis showed that the nanodots had peroxidase-like and catalase-like activities. The reactive oxygen species were probed by electronic paramagnetic resonance technique and spectroscopic methods, demonstrating that the nanodots also had oxidase-like activity. Interestingly, the peroxidase-like activity of nanodots was synergistically enhanced in the presence of ferrous ions or ferric ions. Remarkably, less than nanomolar levels of ferrous ions were required to display this phenomenon, meaning Fenton reagent acted as leverage. Based on this, a sensitive colorimetric and fluorescent dual-mode sensor for alendronate sodium was developed. The linear ranges for colorimetric and fluorescence analysis were 0.2–2.5 and 0.2–2.0 µM, with detection limits of 31.21 and 71.84 nM, correspondingly. The method has a simple large-scale material preparation process with higher sensitivity and shorter reaction time, which can inspire and enlighten the design of nanozyme sensors. Idiopathic pulmonary fibrosis (IPF) is a chronic progressive interstitial lung disease that lacks effective treatment modalities. Once patients are diagnosed with IPF, their median survival is approximately 3–5 years. PPARγ is an important target for the prevention and treatment of pulmonary fibrosis. Asarinin is a lignan compound that can be extracted from food plant Asarum heterotropoides. In this study, we investigated the therapeutic effects of asarinin in a pulmonary fibrosis model constructed using bleomycin in mice and explored the underlying mechanisms. Intraperitoneal administration of asarinin to mice with pulmonary fibrosis showed that asarinin effectively attenuated pulmonary fibrosis, and this effect was significantly inhibited by the PPARγ inhibitor GW9662. Asarinin inhibited TGF-β1-induced fibroblast-to-myofibroblast transition in vitro, while GW9662 and PPARγ gene silencing significantly inhibited this effect. In addition, asarinin inhibited not only the canonical Smad pathway of TGF-β but also the non-canonical AKT and MAPK pathways by activating PPARγ. Forest conversion is a common management practice in forestry, yet the effects of forest conversion on the transformation of phosphorus (P) fractions remain unclear. This work aims at assessing the changes in soil P fractions, including total P (TP), available P (AP), inorganic P (Pi), organic P (Po) and microbial biomass P (MBP) after natural conversion of Chinese fir plantations (CFP) to evergreen broad-leaved forests (EBLF) in subtropical China. Results showed the contents of soil TP and Po were not affected by the forest conversion, but soil AP and MBP concentrations and their proportion significantly increased with forest conversion from CFP to EBLF. The contents of soil P fractions were significantly accumulated in the topsoil (0–20 cm) in the studied forests. Additionally, soil P contents were higher in the winter–spring seasons than in the summer–fall seasons during the period of study. Soil AP content was positively correlated with TP, Po, Pi, labile organophosphorus (LOP), moderate labile organophosphorus (MLOP), moderate resistant organophosphorus (MROP), aluminum–bound P (Al–P), iron–bound P (Fe–P) and MBP contents in EBLF, while the content of soil AP was positively related to Po, LOP, MLOP, Al–P, Fe–P and MBP contents in CFP. Path analysis showed that the most important factor in affecting soil P availability was Po (LOP and HROP) in EBLF stands and was Al–P in CFP stands. Our study suggested that management practices in conserving soil Pi (especially in Al–P) in the CFP and in increasing Po inputs in the EBLF would improve soil P availability in subtropical forests. Fenton-like oxidation processes are widely used to degrade recalcitrant organic pollutants, but are limited by narrow application pH and low reaction efficiency. This study investigated the synchronous activation of H2O2 and persulfate (PDS) by sulfidated zero valent iron (S-nZVI) in ambient conditions for Fenton-like oxidation of bisphenol S (BPS), an estrogenic endocrine-disrupting chemical. The activation of S-nZVI induced H2O2 or PDS could be greatly enhanced with the assistance of PDS and H2O2, respectively, even across a wide range of pH value (3–11). The first-order rate constant of S-nZVI/H2O2/PDS, S-nZVI/PDS and S-nZVI/H2O2 systems was found to be 0.2766 min−1, 0.0436 min−1, and 0.0113 min−1, respectively. A significant synergy between H2O2 and PDS was achieved when the PDS-H2O2 molar ratio was above 1:1, and where sulfidation promoted iron corrosion and decreased solution pH were observed in the S-nZVI/H2O2/PDS system. Radical scavenging experiments and electron paramagnetic resonance (EPR) investigations suggest that both SO4•− and •OH were generated and that •OH played a crucial role in BPS removal. Furthermore, four BPS degradation intermediates were detected and three degradation pathways were proposed in line with the HPLC-Q-TOF–MS analysis. This study demonstrated that compared to the traditional Fenton-like system, the S-nZVI/H2O2/PDS system could be a more efficient, advanced oxidation technology capable of being used across a broad pH range for emerging pollutants' degradation. Tyrosinase is an enzyme in the process of melanin formation and relates to the occurrence of freckles, brown spots, or excessive deposition of melanin in human body and other diseases. In this work, a turn-on fluorescence detection toward tyrosinase was developed. Manganese dioxide nanosheets were conveniently synthesized as an oxidizing agent. These nanosheets converted non-fluorescent o-phenylenediamine (OPD) into a fluorescent product. This reaction and the related fluorescence could be inhibited by reductive dopamine. When tyrosinase was introduced, it catalyzed the oxidation of dopamine and thus led to a recovery of the fluorescence. This signaling provided enhanced fluorescence responses toward tyrosinase in the range 0.006–0.08 U/mL, with a detection limit at 0.003 U/mL. Direct, site-specific methods of protein functionalization are highly desirable for biotechnology. However, such methods are challenging due to the difficulty of chemically differentiating a single site within a large protein. Herein, we propose “metal binding targeting” strategy and develop a Copper Assisted Sequence-specific conjugation Tag (CAST) method to achieve rapid (second order rate 8.1 M−1 s−1), site-specific protein backbone chemical modification with pinpoint accuracy. We demonstrate the versatility of CAST conjugation by preparing various on-demand modified recombinant proteins, including a homogeneous antibody-drug conjugate with high plasma stability and potent efficacy in vitro and in vivo. The entanglements on various dual properties, such as the wave–particle duality, spin-up and spin-down, and so on, have attracted enormous attention, but how to obtain their entanglement states easily was not clear until now for the particle system. Herein, a method is found for constructing entanglement states on various dual properties of the N-particle system by using the basis vectors of the wave function before the entanglement for the N particles, where the N particles are independent of each other before forming the N-particle system. Moreover, the wave function on the dual property of any one of the mutually independent N particles can be written as a linear combination of the same basis vectors before forming the N-particle system. Chemoradiation-induced hearing loss (CRIHL) is one of the most devasting side effects for nasopharyngeal carcinoma (NPC) patients, which seriously affects survivors' long-term quality of life. However, few studies have comprehensively characterized the risk factors for CRIHL. In this study, we found that age at diagnosis, tumor stage, and concurrent cisplatin dose were positively associated with chemoradiation-induced hearing loss. We performed a genome-wide association study (GWAS) in 777 NPC patients and identified rs1050851 (within the exon 2 of NFKBIA), a variant with a high deleteriousness score, to be significantly associated with hearing loss risk (HR = 5.46, 95% CI 2.93–10.18, P = 9.51 × 10–08). The risk genotype of rs1050851 was associated with higher NFKBIA expression, which was correlated with lower cellular tolerance to cisplatin. According to permutation-based enrichment analysis, the variants mapping to 149 hereditary deafness genes were significantly enriched among GWAS top signals, which indicated the genetic similarity between hereditary deafness and CRIHL. Pathway analysis suggested that synaptic signaling was involved in the development of CRIHL. Additionally, the risk score integrating genetic and clinical factors can predict the risk of hearing loss with a relatively good performance in the test set. Collectively, this study shed new light on the etiology of chemoradiation-induced hearing loss, which facilitates high-risk individuals' identification for personalized prevention and treatment. Deoxynivalenol (DON) is one of the most prevalent mycotoxin contaminants, which posing a serious health threat to animals and humans. Previous studies have found that individually supplemented probiotic or glycyrrhinic acid (GA) could degrade DON and alleviate DON-induced cytotoxicity. The present study investigated the effect of combining GA with Saccharomyces cerevisiae (S. cerevisiae) and Enterococcus faecalis (E. faecalis) using orthogonal design on alleviating IPEC-J2 cell damage induced by DON. The results showed that the optimal counts of S. cerevisiae and E. faecalis significantly promoted cell viability. The optimal combination for increasing cell viability was 400 µg/mL GA, 1 × 106 CFU/mL S. cerevisiae and 1 × 106 CFU/mL E. faecalis to make GAP, which not only significantly alleviated the DON toxicity but also achieved the highest degradation rate of DON (34.7%). Moreover, DON exposure significantly increased IL-8, Caspase3 and NF-κB contents, and upregulated the mRNA expressions of Bax, Caspase 3, NF-κB and the protein expressions of Bax, TNF-α and COX-2. However, GAP addition significantly reduced aforementioned genes and proteins. Furthermore, GAP addition significantly increased the mRNA expressions of Claudin-1, Occludin, GLUT2 and ASCT2, and the protein expressions of ZO-1, Claudin-1 and PePT1. It was inferred that the combination of GA, S. cerevisiae, and E. faecalis had the synergistic effect on enhancing cell viability and DON degradation, which could protect cells from DON-induced damage by reducing DON cytotoxicity, alleviating cell apoptosis and inflammation via inhibiting NF-κB signaling pathway, improving intestinal barrier function, and regulating nutrient absorption and transport. Sensitive detection of cancer cells plays a critical role in early cancer diagnosis. Nucleolin, overexpressed on the surface of cancer cells, is regarded as a candidate biomarker for cancer diagnosis. Thus, cancer cells can be detected through the detection of membrane nucleolin. Herein, we designed a nucleolin-activated polyvalent aptamer nanoprobe (PAN) to detect cancer cells. In brief, a long single-stranded DNA with many repeated sequences was synthesized through rolling circle amplification (RCA). Then the RCA product acted as a scaffold chain to combine with multiple AS1411 sequences, which was doubly modified with fluorophore and quenching group, respectively. The fluorescence of PAN was initially quenched. Upon binding to target protein, the conformation of PAN changed, leading to the recovery of fluorescence. The fluorescence signal of cancer cells treated with PAN was much brighter compared with that of monovalent aptamer nanoprobes (MAN) at the same concentration. Furthermore, the binding affinity of PAN to B16 cells was proved to be 30 times higher than that of MAN by calculating the dissociation constants. Skeletal muscle wasting and atrophy is highly prevalent in chronic renal failure (CRF) and increases the risk of mortality. According to our previous study, we speculate that urotensin II (UII) can induce skeletal muscle atrophy by upregulating ubiquitin–proteasome system(UPS) in CRF. C2C12 mouse myoblast cells were differentiated into myotubes, and myotubes were exposed to different concentrations of UII. Myotube diameters, myosin heavy chain(MHC), p-Fxo03A, skeletal muscle-specific E3 ubiquitin ligases such as muscle RING finger 1 (MuRF1) and muscle atrophy F-box (MAFbx/atrogin1) were detected. Three animal models (the sham operation mice as normal control (NC) group, wild-type C57BL/6 mice with 5/6 nephrectomy (WT CRF) group, UII receptor gene knock out (UT KO) mice with 5/6 nephrectomy (UT KO CRF) group) were designed. Cross-sectional area (CSA) of skeletal muscle tissues in three animal models were measured, and western blot detected protein of UII, p-Fxo03A, MAFbx and MuRF1, and immunofluorescence assays explored the satellite cell marker of Myod1 and Pax7, and PCR arrays detected the muscle protein degradation genes, protein synthesis genes and the genes which were involved in muscle components. UII could decrease mouse myotube diameters, and upregulate dephosphorylated Fxo03A protein. MAFbx and MuRF1 were higher in WT CRF group than that in NC group, but after UII receptor gene was knocked out (UT KO CRF), their expressions were downregulated. UII could inhibit the expression of Myod1 but not Pax7 in animal study. We first demonstrate that skeletal muscle atrophy induced by UII associated with upregulating ubiquitin–proteasome system and inhibiting the differentiation of satellite cells in CRF mice. Background: Previous studies have proved that irisin is related to the development of chronic kidney disease. In this study, we aimed to compare serum irisin level in patients treated with peritoneal dialysis (PD) and hemodialysis (HD). Methods: Two hundred and fifty-two dialysis patients (146 PD patients and 106 HD patients) were included in the study. Levels of serum irisin and other parameters were compared between the two groups' patients. Results: There were higher serum irisin levels in PD patients than those in HD patients [113.10 (106.15 ~ 119.15) ng/ml vs. 45.72(21.67 ~ 79.71) ng/ml, P < 0.001]. Moreover, body fat mass, percent body fat, serum calcium, high-density lipoprotein, low-density lipoprotein, carbon dioxide combining power (CO2CP) and residual renal function were higher in patients on PD than that in those on HD, whereas levels of lean body mass, systolic blood pressure, albumin, serum uric acid, potassium, and phosphorus（It should be "were" replace are) are higher in HD patients in comparison to PD patients. Dialysis modality (PD/HD), serum CO2CP level, lean body mass, and percent body fat independently positively correlated with natural logarithm of irisin (lnirisin) by multivariate linear regression analysis. Conclusions: In this study, we prove that serum irisin level is significantly higher in patients treated with peritoneal dialysis than that with hemodialysis. Concrete-filled steel plate composite walls are increasingly used in high-rise buildings; their mechanical properties at room temperature have produced fruitful results. However, research on their mechanical properties in fire and at high temperatures is scarce. Moreover, published studies on the fire resistance of concrete-filled steel plate composite walls have not considered the impact of fire protection measures. Based on previous fire resistance research, a fire resistance test was designed in this study for four walls under the combined action of axial compression and a single-sided fire under the ISO-834 standard heating curve. Apart from that, the existing fire protections have the disadvantages of too thick, easy falling off and cracking or difficult to install. In order to eliminate the disadvantages of existing fire prevention measures, new composite fireproof structure using a rock wool board and a thin fire-retardant coating is proposed. The effects of no fireproof structures, autoclaved lightweight concrete plate fireproof structures, thick-coated fire-retardant structures, and new composite fireproof structures on the fire resistance of concrete-filled steel plate composite walls were studied experimentally. The results show that the fire resistance of the four test specimens meets the 3 h fire resistance limit of the primary load-bearing component in the specification. Based on the experimental results, reasonable fire protection methods are suggested for concrete-filled steel plate composite walls in actual projects, and an experimental basis is provided for further numerical simulation. A novel lytic Ralstonia phage, RPZH3, was isolated from the soil of a tobacco field via a double agar overlay plaque assay. The phage has an icosahedral head 75 ± 5 nm in diameter with a short tail about 15 ± 5 nm in length. It was able to infect 18 out of 30 tested strains of R. solanacearum isolated from tobacco, sweet potato, tomato, pepper, and eggplant. The latent period of the phage was 80 min, and the burst period was 60 min with a burst size of about 27 pfu/cell. The phage was stable at pH 4–12 at 28°C, and it was also stable at temperatures from 45°C to 60°C at pH 7.0. The complete genome of phage RPZH3 consists of 65,958 bp, with a GC content of 64.93%. The genome contains 93 open reading frames (ORFs) and encodes a tRNA for cysteine. Nucleotide sequence alignment and phylogenetic analysis indicated that RPZH3 is a new member of the genus Gervaisevirus belonging to the class Caudoviricetes. [ABSTRACT FROM AUTHOR]

The effects of interactions between the toxic and essential metal mixtures on cognitive function are poorly understood. This study aims to identify the joint association of arsenic (As), cadmium (Cd), and lead (Pb) with cognitive function in older adults and the moderating role of selenium (Se), zinc (Zn), and copper (Cu) in this association. This study included 1000 community-dwelling older adults. Cognitive function was assessed by the Mini-Mental State Examination (MMSE). Blood concentrations of As, Cd, Pb, Se, Zn, and Cu were measured using inductively coupled plasma mass spectrometry. Linear regression and Bayesian kernel machine regression (BKMR) models were applied to assess the individual and joint associations of As, Cd, and Pb with cognitive function and to examine whether Se, Zn, and Cu (individually and as a mixture) modified these associations. In the adjusted single-metal models, both Cd (β = − 0.37, 95% CI: − 0.73 to − 0.01) and Pb (β = − 0.44, 95% CI: − 0.86 to − 0.02) were associated with MMSE scores, while Se (β = 0.71, 95% CI: 0.30 to 1.13) exhibited a positive relationship with MMSE scores. Univariate exposure–response functions from BKMR models showed similar results. Moreover, the toxic metal mixture (As, Cd, and Pb) exhibited a significant negative association with MMSE scores in a dose–response pattern, with Pb being the greatest contributor within the mixture. The negative association of Pb alone or the toxic metal mixture with MMSE scores became weaker at higher concentrations of Se within its normal range, especially when Se levels were greater than the median (89.18 μg/L). Our findings support that Se can attenuate the negative associations of exposure to single Pb or the As, Cd, and Pb mixtures with cognitive function. A prediction interval (PI) method is developed to quantify the model uncertainty of embankment settlement prediction. Traditional PIs are constructed based on specific past period information and remain unchanged; hence, they neglect discrepancies between previous calculations and new monitoring data. In this paper, a real-time prediction interval correction method is proposed. Time-varying PIs are built by continuously incorporating new measurements into model uncertainty calculations. The method consists of trend identification, PI construction, and real-time correction. Primarily, trend identification is carried out by wavelet analysis to eliminate early unstable noise and determine the settlement trend. Then, the Delta method is applied to construct PIs based on the characterized trend, and a comprehensive evaluation index is introduced. The model output and the upper and lower bounds of the PIs are updated by the unscented Kalman filter (UKF). The effect of the UKF is compared with that of the Kalman filter (KF) and extended Kalman filter (EKF). The method was demonstrated in the Qingyuan power station dam. The results show that the time-varying PIs based on trend data are smoother than those based on original data with better evaluation index scores. Also, the PIs are not affected by local anomalies. The proposed PIs are consistent with the actual measurements, and the UKF performs better than the KF and EKF. The approach has the potential to provide more reliable embankment safety assessments. Various biomarkers targeting cell-free DNA (cfDNA) and circulating proteins have been tested for pan-cancer detection. Oncofetal chondroitin sulfate (ofCS), which distinctively modifies proteoglycans (PGs) of most cancer cells and binds specifically to the recombinant Plasmodium falciparum VAR2CSA proteins (rVAR2), is explored for its potential as a plasma biomarker in pan-cancer detection. To quantitate the plasma ofCS/ofCSPGs, we optimized an ELISA using different capture/detection pairs (rVAR2/anti-CD44, -SDC1, and -CSPG4) in a case-control study with six cancer types. We show that the plasma levels of ofCS/ofCSPGs are significantly higher in cancer patients (P values, 1.2 × 10−2 to 4.4 × 10−10). Validation studies are performed with two independent cohorts covering 11 malignant tumors. The individuals in the top decile of ofCS-CD44 have more than 27-fold cancer risk (OR = 27.8, 95%CI = 18.8–41.4, P = 2.72 × 10−62) compared with the lowest 20%. Moreover, the elevated plasma ofCS-CD44 could be detected at the early stage of pan-cancer with strong dose-dependent odds risk prediction. Developing multi-cancer early detection (MCED) tests based on cfDNA or protein markers holds great promise for effective cancer screening.
