Abandoned metal mines and refineries are considered environmentally vulnerable areas owing to high levels of exposure to heavy metals. This study examined the association between heavy metal exposure and renal function indicators. We studied a total of 298 participants, of which 74 and 68 resided in low- and high-exposure abandoned metal mine areas, respectively, with 121 in the refinery area and 35 in the control area. Blood and urine samples were collected from the participants to analyze the levels of blood lead, cadmium, and creatinine and urinary cadmium, NAG, and β2-MG. The estimated glomerular filtration rate, which is calculated using the Chronic Kidney Disease Epidemiology Collaboration equation, was used for assessments. The study participants comprised more females than males, and their mean age was 70.3 years. The blood lead and cadmium as well as urinary cadmium levels were 2.12 μg/dL, 1.89 μg/L, and 2.11 μg/L, respectively, in the heavy metal-exposure areas, and 1.18 μg/dL, 0.89 μg/L, and 1.11 μg/L, respectively, in the control area. The odds ratio (OR) for exceeding the reference value showed that blood cadmium in the refinery area was 38 times higher than that in the control area. Urinary cadmium was seven times higher in the low-exposure abandoned metal mine area than in the control area. NAG showed a positive correlation with urinary cadmium in all areas. In the refinery area, correlations were observed between β2-MG and urinary cadmium levels and the eGFR and blood cadmium level; in the high-exposure abandoned metal mine area, correlations were observed between NAG, β2-MG, and the eGFR and blood cadmium. In this study, the association between Cd exposure and some renal function indicators was observed. This study’s findings and the obtained biological samples can serve as a basis for future molecular biological research.

Background Cancer cells have developed molecular strategies to cope with evolutionary stressors in the dynamic tumor microenvironment. Peroxisome proliferator-activated receptor-γ coactivator-1α (PGC1α) is a metabolic rheostat that regulates diverse cellular adaptive behaviors, including growth and survival. However, the mechanistic role of PGC1α in regulating cancer cell viability under metabolic and genotoxic stress remains elusive. Methods We investigated the PGC1α-mediated survival mechanisms in metabolic stress (i.e., glucose deprivation-induced metabolic stress condition)-resistant cancer cells. We established glucose deprivation-induced metabolic stress-resistant cells (selected cells) from parental tumor cells and silenced or overexpressed PGC1α in selected and parental tumor cells. Results Several in vitro and in vivo mouse experiments were conducted to elucidate the contribution of PGC1α to cell viability in metabolic stress conditions. Interestingly, in the mouse xenograft model of patient-derived drug-resistant cancer cells, each group treated with an anti-cancer drug alone showed no drastic effects, whereas a group that was co-administered an anti-cancer drug and a specific PMCA inhibitor (caloxin or candidate 13) showed marked tumor shrinkage. Conclusions Our results suggest that PGC1α is a key regulator of anti-apoptosis in metabolic and genotoxic stress-resistant cells, inducing PMCA expression and allowing survival in glucose-deprived conditions. We have discovered a novel therapeutic target candidate that could be employed for the treatment of patients with refractory cancers.

Remote sensing and GIS technology were very helpful to determine an appropriate location of freshwater storage in Amhara, Ethiopia. The techniques were used to investigate the impact of lithology, surface geomorphology, slope parameters, drainage flow, drainage density, lineament density, land cover parameters on relief, and aerial and linear features and to understand their interrelationships. Morphometric parameters such as mean stream length (Lsm), stream length ratio (RL), bifurcation ratio (Rb), mean bifurcation ratio (Rbm), relief ratio (Rh), drainage density (Dd), stream frequency (Fs), drainage texture (Rt), form factor (Rf), circularity ratio (Rc), and elongation ratio (Re) were calculated. Spatial maps of morphometric parameters were produced by using AHP (analytical hierarchy process) of ArcGIS 10.3. Final priority map was generated by the overlay of those parameters with five categories of poor (16.6%), low (41.63%), moderate (29.61%), high (8.88%), and very high (3.28%) storage locations. The map showed that this study area belonged to the low to moderate storage location. The results exhibit precision-based assessment of the suitability for the dam construction sites of 6, 7, and 9 sub-basin zones. The outcome of this study strengthens the knowledge of geospatial analysis for water resources vulnerability and also allows policymakers in this drought-prone area to sustainably manage water supplies.

Hypoxia is the most frequently occurring adverse effect during endoscopic retrograde cholangiopancreatography (ERCP) under sedation; thus, oxygen must be properly supplied to prevent a reduction of oxygen saturation. In this study, we intend to verify the preventive effect for hypoxia during ERCP, using a high-flow nasal cannula (HFNC), in elderly patients. As a multicenter prospective randomized trial, patients who underwent ERCP with propofol-based sedation were randomly assigned into two groups: Patients in the HFNC group were supplied with oxygen via an HFNC, and those in the standard nasal cannula group were supplied with oxygen via a low-flow nasal cannula. The co-primary end points were the lowest oxygen saturation rate and hypoxia during the overall procedure. A total of 187 patients (HFNC group: 95; standard nasal cannula group: 92) were included in the analysis. Unexpected hypoxia events were more frequently observed among patients in the standard nasal cannula group than among patients in the HFNC group (13% vs. 4%, odds ratio 3.41, 95% confidence interval 1.06–11.00, p = 0.031). The mean of the lowest oxygen saturation rate during ERCP was significantly lower in the standard nasal cannula group than in the HFNC group (95% vs. 97%, p = 0.002). Oxygen supplementation with an HFNC can prevent oxygen desaturation and hypoxia events in patients undergoing ERCP under sedation. Exposure to diesel exhaust particles (DEPs) and urban particles (UPs) increases the incidence of degenerative brain diseases as well as respiratory diseases. However, there is limited evidence on the mechanism of neurotoxicity on exposure to these particles. In the present study, the damage to blood–brain barrier (BBB) function by DEP or UP exposure was evaluated in bEnd.3 cells, which are derived from the brain tissue of Balb/c mice. It was demonstrated that DEP and UP exposure may induce oxidative stress via increasing reactive oxygen species (ROS) and decreasing total antioxidant capacity (TAC) level in bEnd.3 cells. In addition, cells exposed to DEP and UP demonstrated a resistance value of about 50% each compared to the value noted prior to exposure; additionally, Claudin-5 and ZO-1 expression levels were significantly decreased compared to the corresponding levels in the control. It was inferred that DEP or UP exposure diminishes the expression of tight junction proteins in endothelial cells through ROS generation, thereby enhancing endothelial membrane permeability. This study showed that DEPs or UPs induced cell permeability and oxidative stress by increasing ROS generation in bEnd.3 cells. This suggests the possibility that exposure to DEPs or UPs may compromise the integrity of the BBB and induce adverse effects in the CNS.

The purpose of this study was to evaluate the metal concentrations in the Halda River in Bangladesh to determine the quality of the water and sediment in the natural spawning zone. Fe Zn Cr Cd Cu was the order of the metals in water, whereas Fe Zn Cd Cu was the order in sediments. Almost all of the heavy metals in the water and sediment had been found within the established limits, with the exception of Cr and Fe in the river and Cu in the sediment. In the case of water, Cr vs. Zn was found to have the strongest correlation (r = 0.96). Due to the coagulation and adsorption processes, it was shown that Fe and Zn had a substantial correlation of 0.96, Cu and Cd of 0.91, and Cr of 0.78 with Zn. Hazard quotient values of Cd show the not potable nature of Halda river surface water and might give adverse health effects for all age groups except Cu and Zn. Pollution load index values indicated the uncontaminated nature of the river bottom sediments. Natural and human activities were the key factors influencing the accumulation and movement of heavy metals in the water and sediments. Contamination sources are industrial effluents, garbage runoff, farming operations, and oil spills from fishing vessels which are comparable according to multivariate statistical analysis. Ion exchange, absorption, precipitation, complexation, filtration, bio-absorption, redox reaction, and reverse osmosis were considered to be effective for the degradation of metal concentrations. The feasibility of the suggested metal reduction procedures has to be studied to know which is optimally appropriate for this river region. It is expected that this study could provide a useful suggestion to decrease the metal pollution in the river.

We isolated S-formylglutathione hydrolase (scSFH) from Sphingobium chungbukense strain KCTC2955 and expressed it in Escherichia coli. The extracted scSFH showed activity toward p-nitrophenyl butyrate (p-NB), and a protein–substrate interaction was confirmed through 3D molecular docking simulation. The DNA sequence of the cloned scSFH gene (900 bp) has an open reading frame encoding 300 amino acids with a predicted molecular mass of 32.82 kDa; the sequence contains Ser-His-Asp conserved in the active site and a GHSMGG motif. The cloned scSFH gene successfully overproduced scSFH protein under the T7 promoter of the expression vectors pET21a and pGEX4T-1 in E. coli BL21(DE3). We analyzed E. coli-expressed scSFH protein for esterase activity using p-NB as a substrate. Furthermore, we confirmed the interaction between the scSFH protein and p-NB substrate. The scSFH protein of the SFH family was overexpressed in E. coli BL21(DE3) and purified using affinity Ni–NTA agarose resin. The activity of purified scSFH against p-NB was 6.87 mg L−1 for Km and 131.16 mg L−1 protein min−1 for Vmax. Additionally, we confirmed that the serine residue of the GHSMGG motif in the obtained protein interacts with p-NB through molecular docking prediction by the molecular operating environment. We identified a novel scSFH gene from S. chungbukense and overexpressed it in E. coli. The recombined scSFH protein showed activity toward p-NB, and the interaction was confirmed through 3D molecular docking prediction. This research could be applied to identify new genes and confirm their functions.

In this study, the medium requirements to increase the production of xylitol by using Candida tropicalis (CT) have been investigated. The technique of single addition or omission of medium components was applied to determine the nutritional requirements. The addition of amino acids such as Asp, Glu, Gln, Asn, Thr, and Gly had no significant effect on CT growth. However, in the absence of other metal ions, there was a higher concentration of cell growth and xylitol production when only Zn(2+) was present in the medium. The analysis of various vitamins unveiled that biotin and thiamine were the only vitamins required for the growth of CT. Surprisingly, when only biotin was present in the medium as a vitamin, there was less growth of CT than when the medium was complete, but the amount of xylitol released was significantly higher. Overall, this study will increase the xylitol production using the single omission or addtion technique.

Phenological shifts associated with climate and environmental change are evident in temperate forests. Further dense and comprehensive observations are necessary to understand the species-specific and regional variation in the responses of plant phenology to climate change. This study first introduces the national-scale phenology network (NPN) over South Korea. By having newly established phenology datasets for the period 2009–2018, we analyze spatial and temporal variations of phenology of 21 deciduous species in spring and autumn as well as 4 evergreen species in spring. For deciduous species, the phenological responses are examined using the dates of budburst, leaf unfolding, leaf coloring, and leaf fall, whereas the times of pollen start and pollen peak are adopted to investigate the responses of evergreen species. The average dates of budburst and leaf unfolding are changed by -1.1 and − 1.3 day/year, respectively, in agreement with the observed trends in temperature. The pollen activity (start and peak) of evergreen species is significantly advanced by 1.4 day/year throughout Korea, suggesting apparent phenological response of evergreen species to the warming. Especially, the shrub had up to 45 % larger temperature sensitivity than the subtree or tree in spring. By contrast, changes in the autumn phenology of deciduous species are not evident because of the large interannual variability in the dates of leaf coloring and leaf fall. Then, the large interannual variation in autumn phenology could constrain the interannual variability in the length of the growing season of temperate forest. We further expect on-going efforts on national-scale phenology monitoring will have an important contribution to understanding vegetation growing season changes related to warming.

A reliable assessment of the aquifer contamination vulnerability is essential for the conservation and management of groundwater resources. In this study, a recent technique in artificial intelligence modeling and computational optimization algorithms have been adopted to enhance the groundwater contamination vulnerability assessment. The original DRASTIC model (ODM) suffers from the inherited subjectivity and a lack of robustness to assess the final aquifer vulnerability to nitrate contamination. To overcome the drawbacks of the ODM, and to maximize the accuracy of the final contamination vulnerability index, two levels of modeling strategy were proposed. The first modeling strategy used particle swarm optimization (PSO) and differential evolution (DE) algorithms to determine the effective weights of DRASTIC parameters and to produce new indices of ODVI-PSO and ODVI-DE based on the ODM formula. For strategy-2, a deep learning neural networks (DLNN) model used two indices resulting from strategy-1 as the input data. The adjusted vulnerability index in strategy-2 using the DLNN model showed more superior performance compared to the other index models when it was validated for nitrate values. Study results affirmed the capability of the DLNN model in strategy-2 to extract the further information from ODVI-PSO and ODVI-DE indices. This research concluded that strategy-2 provided higher accuracy for modeling the aquifer contamination vulnerability in the study area and established the efficient applicability for the aquifer contamination vulnerability modeling.

People living near abandoned mines are at increased risk of exposure to toxic metals. We surveyed 4500 inhabitants with the mean age of 68.5 years old (male: 1768, female: 2732) living near 104 abandoned metal mines from 2013 to 2017 (the 2nd phase health survey in Korea). We conducted personal interviews, blood and urine sampling, and analyzed the concentrations of lead (Pb) and cadmium (Cd) in whole blood and Cd in urine using a graphite furnace atomic absorption spectrometer. The geometric means of blood Pb, blood Cd, and urine Cd were 2.27 μg/dL, 1.42 μg/L, and 1.66 μg/g creatinine, respectively. The level of metal exposure was lower than that reported from the first phase health survey in Korea (2008‒2011) but was higher than in the general population of Korea. Blood Pb was higher in males while blood Cd and urine Cd were significantly higher in females. Blood Pb was highest in the 40‒59 age group, while blood and urine Cd levels continuously increased until age 80 or older. The Cd levels in blood and urine were affected by consumption of locally produced rice and duration of residence near abandoned mines. Furthermore, negative correlations were observed between blood Pb and blood and urine Cd levels. Additionally, 252 of the 4500 subjects exceeded the thresholds of blood Cd or urine Cd levels. Together, these findings suggest that Cd has more sustainable and adverse health effects on the abandoned mine inhabitants, who are mostly aged. Therefore, continuous biomonitoring and risk assessment to environmental health risks are necessary for environmental pollution control and health promotion.

The direct current (DC) grid range is rapidly expanding according to the increase of power generation capacity for the DC demands. In order to stably operate the DC grid system, we need a suitable DC circuit breaker for DC characteristics. We propose the superconducting arc-induction type DC circuit breaker. Lorentz’s force is occurred by the strong magnetic field is occurring by the superconducting magnet and the arc is occurring by the between Anode and Cathode. If we exactly control that, the efficiency of the arc-induction can be higher. In this paper, the electric field and according to the cut-off operation of the superconducting arc-induction type DC circuit breaker were analyzed. It is very important because the electric field and Lorentz force are an indicator for confirming the movement of the arc caused by the opening contacts. As a result, we confirmed that the Lorentz force calculated from the FEM showed a maximum increase ratio of 1101 times.

In the present study, Rhus vernicifera laccase (RvLac) was immobilized through covalent methods on the magnetic nanoparticles. Fe(2)O(3) and Fe(3)O(4) nanoparticles activated by 3-aminopropyltriethoxysilane followed with glutaraldehyde showed maximum immobilization yields and relative activity up to 81.4 and 84.3% at optimum incubation and pH of 18 h and 5.8, respectively. The maximum RvLac loading of 156 mg/g of support was recorded on Fe(2)O(3) nanoparticles. A higher optimum pH and temperature of 4.0 and 45 °C were noted for immobilized enzyme compared to values of 3.5 and 40 °C for free form, respectively. Immobilized RvLac exhibited better relative activity profiles at various pH and temperature ranges. The immobilized enzyme showed up to 16-fold improvement in the thermal stability, when incubated at 60 °C, and retained up to 82.9% of residual activity after ten cycles of reuses. Immobilized RvLac exhibited up to 1.9-fold higher bisphenol A degradation efficiency potential over free enzyme. Previous reports have demonstrated the immobilization of RvLac on non-magnetic supports. This study has demonstrated that immobilization of RvLac on magnetic nanoparticles is very efficient especially for achieving high loading, better pH and temperature profiles, and thermal- and solvents-stability, high reusability, and higher degradation of bisphenol A.

We research team has proposed a current-limiting superconducting DC circuit breaker that combines a High-temperature superconducting wire (HTS) with a DC circuit breaker, and secured its operation performance in our previous study via simulation. In this paper, we designed and produced an actual current-limiting superconducting DC circuit breaker, as well as analyzed its current limiting and breaking performance. In addition, the operation characteristics at different lengths of the HTS with increasing voltage were analyzed. The operation reliability of the proposed DC circuit breaker was demonstrated by a simulation system that was built similar to the actual system.
