Despite the rapid progress of organic solar cells based on non-fullerene acceptors, simultaneously achieving high power conversion efficiency and long-term stability for commercialization requires sustainable research effort. Here, we demonstrate stable devices by integrating a wide bandgap electron-donating polymer (namely PTzBI-dF) and two acceptors (namely L8BO and Y6) that feature similar structures yet different thermal and morphological properties. The organic solar cell based on PTzBI-dF:L8BO:Y6 could achieve a promising efficiency of 18.26% in the conventional device structure. In the inverted structure, excellent long-term thermal stability over 1400 h under 85 °C continuous heating is obtained. The improved performance can be ascribed to suppressed charge recombination along with appropriate charge transport. We find that the morphological features in terms of crystalline coherence length of fresh and aged films can be gradually regulated by the weight ratio of L8BO:Y6. Additionally, the occurrence of melting point decrease and reduced enthalpy in PTzBI-dF:L8BO:Y6 films could prohibit the amorphous phase to cluster, and consequently overcome the energetic traps accumulation aroused by thermal stress, which is a critical issue in high efficiency non-fullerene acceptors-based devices. This work provides insight into understanding non-fullerene acceptors-based organic solar cells for improved efficiency and stability.

Post-inflammatory erythema (PIE) and post-inflammatory hyperpigmentation (PIH) are the most common acne-related sequelae with no effective treatments. By combining different cut-off filters, intense pulsed light (IPL) therapy can effectively treat these conditions with few side effects. While the safety and effectiveness of IPL for treating post-burn hyperpigmentation is well known, there is little evidence for its benefits for acne-related PIH. In this article, we evaluate the efficacy and safety of IPL for the treatment of acne-related PIE and PIH.This retrospective study evaluated 60 patients with more than 6 months of PIE and PIH treated by the same IPL device and similar protocols. The treatment included three to seven sessions at 4-6-week intervals, and three cut-off filters (640 nm, 590 nm and 560 nm) were used sequentially in each session. Using the Global Aesthetic Improvement Scale (GAIS), Cardiff Acne Disability Index (CADI), and Erythema Assessment Scale (EAS), patients were evaluated on the basis of their facial photographs. The facial brown spots and red areas were visualised and analysed using the VISIA-CR system. No further corrections.On the basis of the GAIS, 49 of 60 patients (81.7%) showed complete or partial clearance of erythema and hyperpigmentation. The CADI and EAS scores showed significant improvement (p 0.01) after IPL treatment compared with pre-treatment. A significant reduction (p 0.01) in the facial brown spots and red areas was seen after IPL treatment. While no long-term side effects were reported, seven patients (11.7%) experienced acne relapse at follow-up.IPL is an effective and safe treatment for acne-related PIE and PIH.

Hepatocellular carcinoma (HCC) has emerged as the third cause of cancer-related death owing to lacking effective systemic therapies. Genomic DNA sequencing revealed the high frequency of loss-of-function mutations in ARID2, which encodes a subunit of SWI/SNF chromatin remodeling complex, however, the therapeutic strategy for the HCC patients with ARID2 mutations is still completely unclear. In this study, we first performed a high-throughput screening approach using a compound library consisting of 2 180 FDA-approved drugs and other compounds, to elicit the potential drugs for synthetic lethality to target ARID2-deficient HCC cells. Interestingly, JQ1, a selective inhibitor of bromodomain protein BRD4, uniquely suppressed the growth of ARID2- deficient HCC cells. Next JQ1 is further confirmed to predominantly induce cell lethality upon ARID2 depletion through exacerbating DNA damage, especially double strand breaks (DSBs). Functional assays demonstrated that both BRD4 inhibition and ARID2 deficiency synergistically impede two main DNA damage repair pathways, homologous recombination (HR) and non-homologous end-joining (NHEJ), through attenuating the transcription of BRCA1, RAD51, and 53BP1, which encode the core molecules responsible for DSB repair. Mechanistically, both ARID2 and BRD4 exert a synergistic effect for maintaining transcriptional enhancer-promoter loops of these genes within chromatin conformation. However, as both ARID2 and BRD4 are disrupted, the expression of these DNA repair-related genes in response to DNA damage are hindered, resulting in DSB accumulation and cell apoptosis. Taken together, this study discloses that BRD4 inhibition may induce synthetic lethality in ARID2-deficient HCC cells, which might provide a potential therapeutic strategy for HCC patients with ARID2 mutations.

Among the methods of detecting food adulteration, the PCR amplification techniques of the endogenous reference gene are increasingly significant. In this article, the improved SDS-CTAB is chosen for the DNA extraction of Flammulina velutipes, the tre6P Phosphatase gene (Tps2) is selected an endogenous reference gene suitable to identify agricultural important fungus F. velutipes and by-products via PCR methods. The Tps2 gene was assayed in 2 varieties of F. velutipes and 9 non-F. velutipes species by both qualitative and quantitative PCR, no amplicon was captured in the non-F. velutipes species. By SYBR Green quantitative PCR, the melting curve showed that the target Tps2 sequence displayed high homogeneity in different F. velutipes varieties. In Taqman probe quantitative PCR analysis, the detection limit was as low as 3 pg/uL of DNA template. From what is said in the foregoing, we can conclude that the Tps2 gene is a reliable, convenient, and accurate endogenous reference gene for detecting F. velutipes component, even in the processed food products, such as sauce and biscuits.

Regardless of whether the federal funds target rate has lifted off from the zero lower bound, the historical record of interest rates will be forever marred by the lack of variation in the aftermath of the great recession. This paper employs a method of indirect inference to analyze the interplay between unemployment rates, leading indicators and interest rates in an environment with near-zero interest rates. This method is used to estimate a decision tree for state-dependent Federal Reserve policy to estimate an alternative target rate series similar to those predicted by a shadow rate model.

A glucose (Glu) sensor was designed by introducing synthetic cyclic peptides (CPs) as recognition receptors and Au nanoparticles assisted graphitic carbon nitride (AuNPs/g-C3N4) for electrochemiluminescence (ECL) enhancement. The synthetic CP receptor (cyclo-[−CNDNHCRDNDC−]) with natural active center of Glu binding protein can mimic the interactions between Glu and Glu binding protein to specifically capture Glu. The AuNPs were reduced on g-C3N4 and formed a new nanohybrid that can be applied as an ECL emitter. The AuNPs/g-C3N4 effectively ameliorated the ECL response of bare g-C3N4. The ECL enhancement mechanism was theoretically speculated through computer simulation. Glu quantification was conducted by recording ECL shifts induced by the binding of Glu to CPs. The linear detection range of the fabricated CPs-based ECL sensor was 1 to 100 mmol L−1, and the detection limit (LOD) was 0.57 nmol L−1 (S / N = 3). The CP-based ECL sensor also showed good specificity, repeatability, stability, and favorable recoveries in sample analysis. This work offer a promising analytical method for Glu assay in clinical diagnostics and bioprocess monitoring.

Distributed acoustic sensing (DAS) technology plays an increasingly important role in the field of underwater acoustic detection because of its own advantages. To measure acoustic pressure sensitivity of seven kinds of optical cables in the same condition synchronously, the experimental setup based on DAS is proposed. Seven kinds of optical cables are connected in series to the DAS system, and the acoustic pressure sensitivity at different locations (different optical cables) is detected simultaneously via the DAS system. The highest acoustic pressure sensitivity of the sensitized optical cable is 69.9 mrad/Pa. The experimental results provide experimental data support for the selection of optical cables.

We demonstrate an intracavity-triggered passively Q-switched Nd:YVO4 laser within a diode-end-pumped configuration. We employ a Cr4+:YAG saturable absorber as the passive Q switch and an Nd:LiYF4 (YLF) laser as the laser triggering of the Q-switched laser. Since we use the same Cr4+:YAG crystal and output coupler with the Nd:YVO4 laser, the Cr4+:YAG Q switch is triggered inside the Nd:YLF laser cavity. As a result, the timing jitter in standard deviation of Nd:YVO4 laser can be reduced to 16 ns.

We demonstrate a diode-pumped passively Q-switched Nd:GdYAG mixed garnet laser at 1,123 nm. A Cr4+:YAG crystal with an initial transmission of 97% is used as the saturable absorber. The maximum average output power is 1.05 W at an absorbed pumping power of 8.12 W. A single-pulse energy can reach up to 78.9 μJ, with a corresponding pulse repetition rate of 13.3 kHz.

Licorice (root and rhizome of Glycyrrhiza uralensis Fisch.) is sometimes used as crude drug after processing. In this report, we prepared roasted licorice with or without honey using 3 lots of crude drug samples derived from wild G. uralensis, and analyzed the constituents in unprocessed, roasted, and honey-roasted licorice samples by high performance liquid chromatography–electrospray ionization-ion trap-time of flight mass spectrometry (HPLC–ESI-IT-TOF-MSn) with principal component analysis. We found that the areas of 41 peaks were noticeably changed by processing. Among them, the areas of 12 peaks, viz. isoliquiritin, isoliquiritigenin, glucoisoliquiritin, 6″-O-acetylisoliquiritin, 6″-O-acetylisoliquiritin apioside, glycyrrhetinic acid 3-O-glucuronide, 5 kinds of sugar-derivatives and one compound whose molecular weight was 386 Da were increased by roasting in all 3 lots, and those peak areas were increased by higher heating temperatures. Among the increased peaks, 3 kinds of sugar-derivatives had larger areas, and 6″-O-acetylisoliquiritin had lower areas than those in honey-roasted licorice. Those sugar-derivatives were the only characteristics differing between honey-roasted licorice and roasted licorice. Meanwhile, the areas of 9 peaks, four of them identified as 6″-O-acetylliquiritin, 6″-O-acetylliquiritin apioside, formononetin and gancaonin l, were decreased by roasting in all 3 lots, but there were no differences between roasted licorice with or without honey. Those compounds whose amounts were changed by processing could be used as markers for the quality control of roasted and honey-roasted licorice.

Background Community-acquired pneumonia (CAP) is a major cause of death worldwide and occurs with variable severity. There are few studies focused on the expression of soluble urokinase-type plasminogen activator receptor (suPAR) and syndecan-4 in patients with CAP. Methods A prospective, multi-centre study was conducted between January 2014 and December 2016. A total of 103 patients with severe CAP (SCAP), 149 patients with non-SCAP, and 30 healthy individuals were enrolled. Clinical data were recorded for all enrolled patients. Serum suPAR and syndecan-4 levels were determined by quantitative enzyme-linked immunosorbent assay. The t test and Mann–Whitney U test were used to compare between two groups; one-way analysis of variance and the Kruskal–Wallis test were used to compare multiple groups. Correlations were assessed using Pearson and Spearman tests. Area under the curve (AUCs), optimal threshold values, sensitivity, and specificity were calculated. Survival curves were constructed and compared by log-rank test. Regression analyses assessed the effect of multiple variables on 30-day survival. Results suPAR levels increased in all patients with CAP, especially in severe cases. Syndecan-4 levels decreased in patients with CAP, especially in non-survivors. suPAR and syndecan-4 levels were positively and negatively correlated with severity scores, respectively. suPAR exhibited high accuracy in predicting SCAP among patients with CAP with an AUC of 0.835 (p

In the field of oil well logging, real-time monitoring of the fluid flow parameter provides a scientific basis for oil and gas optimization exploration and increase in reservoir recovery, so a non-intrusive flow test method based on turbulent vibration was proposed. The specific length of the sensor fiber wound tightly around the outer wall of the pipe was connected with the optical fiber gratings at both ends, and the sensor fiber and the optical fiber gratings composed the flow sensing unit. The dynamic pressure was generated by the turbulence when fluid flows through the pipe, and the dynamic pressure resulted in the light phase shift of the sensor fiber. The phase information was demodulated by the fiber optic interferometer technology, time division multiplexing technology, and phase generated carrier modulation and demodulation techniques. The quadratic curve relationship between the phase change and flow rate was found by experimental data analysis, and the experiment confirmed the feasibility of the optical fiber flow test method with non-intrusion and achieved the real-time monitoring of the fluid flow.

The dynamics of polymer chains in layered silicate/polystyrene nanocomposites was studied by anelastic spectroscopy. Two thermal activated peaks (α and α′ peaks) appeared when the specimens were heated to a high temperature and they were related to glass transition and liquid-liquid transition, respectively. The activation energy was calculated based on Arrhenius equation and it showed that the activation energy of glass transition (E g) is much higher than that of liquid-liquid transition (E ll). Furthermore, the most interesting result for the activation energy was that there were two contrary trends for E g and E ll, E g decreased and E ll increased with the addition of clay platelets. The fragile parameter was analyzed and the variation of fragile parameters for the two transitions was also contrary to each other with the addition of clay platelets. All the results indicated that the confinement effect of clay platelets on the dynamics of polymer chain was scale dependent, and perhaps, the two transitions were produced by different mechanisms.

With the increasing development of genetically modified organisms (GMOs), labeling regulations have been introduced, which require appropriate detection methods. The polymerase chain reaction (PCR) technique has been the mainstay for GMO detection, especially for event-specific qualitative and quantitative PCR detection methods, which have become the internationally agreed state-of-art. This paper describes the character and event-specific quantitative detection method of DP-356043-5 (356043) soybean. In this research, the flanking regions were characterized by inverse PCR (I-PCR). Furthermore, the event-specific PCR primers and TaqMan probe were designed based on the discovered right and left flanking sequences. In the qualitative PCR assay, PCR systems were established with the species-specific and event-specific primers, respectively. And event-specific primers were established on both right and left flanking sequences; the limit of detection (LOD) was both 0.05% (approximates to 42 haploid genome copies). In the quantitative TaqMan real-time PCR assay, we obtained standard curves with good linearity and relatively high efficiency of PCR. All the results indicated that the established event-specific qualitative and quantitative PCR systems for 356043 soybean in this study were reliable and suitable for 356043 soybean detection in mixed samples. Besides, based on the flanking sequence information we obtained, not only the qualitative and quantitative PCR system for detecting 356043 soybean can be established, but also some other novel event-specific detection methods using gene microarray, biosensor, etc., with target sequence on them can also be developed, which have a good value for detecting 356043 soybean.

Dried whole herbs or roots and rhizomes of Asarum campaniflorum have been sold under the trade name Xixin and used as folk remedies in its producing areas for a long time. In order to avoid the misuse of A. campaniflorum as official species of Xixin (Asari radix et rhizoma), a comparative study based on the morphological and phytochemical analysis of the aerial and underground parts was carried out. The usual morphological methods and a microscopic imaging system were used. The results show that A. campaniflorum could be easily distinguished from two official species (Asarum sieboldii and A. heterotropoides var. mandshuricum) by the diameter of thicker roots (1.3-2.7 mm), distinct large parenchymatous cells in phloem of roots, and the size of oil cells in upper leaf epidermises [(40)80-140(174) microm in diameter, where numbers in parentheses are for rare cases], etc. Nine aristolochic acid analogues (AAAs) were identified and estimated by high-performance liquid chromatography-diode array detection (HPLC-DAD). The aerial and underground parts of A. campaniflorum contained 3-4 AAAs, i.e., aristololactam-II-N-beta-D-glucoside (AL-II-Glc), aristololactam I (AL-I), aristololactam II (AL-II), and aristolochic acid I (AA-I), while only AL-I was detected in underground parts of A. sieboldii and no AAAs were detected in underground parts of A. heterotropoides var. mandshuricum. The respective contents (in mg/g) in aerial and underground parts of A. campaniflorum, were as follows: AL-I, 0.06-0.12, 0.05-0.10; AL-II, 0.03-0.04, 0.01-0.03; AA-I, 0.01-0.02, 0.0-0.0. These data suggest A. campaniflorum has a high risk of causing aristolochic acid nephropathy. All these discoveries can contribute to not only the better understanding of this new resource species, but also the safe use of the crude drug Xixin.

Chemoresistance is a major cause of treatment failure and poor outcome in neuroblastoma. In this study, we investigated the expression and function of dual-specificity phosphatase 26 (DUSP26), also known as mitogen-activated protein kinase phophatase-8, in human neuroblastoma. We found that DUSP26 was expressed in a majority of neuroblastoma cell lines and tissue specimens. Importantly, we found that DUSP26 promotes the resistance of human neuroblastoma to doxorubicin-induced apoptosis by acting as a p53 phosphatase to downregulate p53 tumor suppressor function in neuroblastoma cells. Inhibiting DUSP26 expression in the IMR-32 neuroblastoma cell line enhanced doxorubicin-induced p53 phosphorylation at Ser20 and Ser37, p21, Puma, Bax expression as well as apoptosis. In contrast, DUSP26 overexpression in the SK-N-SH cell line inhibited doxorubicin-induced p53 phosphorylation at Ser20 and Ser37, p21, Puma, Bax expression and apoptosis. Using in vitro and in vivo assays, we found that DUSP26 binds to p53 and dephosphorylates p53 at Ser20 and Ser37. In this report, we show that DUSP26 functions as a p53 phosphatase, which suppresses downstream p53 activity in response to genotoxic stress. This suggests that inhibition of this phosphatase may increase neuroblastoma chemosensitivity and DUSP26 is a novel therapeutic target for this aggressive pediatric malignancy.

We analyzed 127 rDNA sequences (5S DNA units) obtained from 23 seed accession samples from more or less 10 taxa in wild and cultivated rye, genus Secale L. The sequences fell into two known groups, here assigned to two unit classes, viz. long R1 and short R1 (designations to reflect on R haplome of rye). The different taxa could not be fully differentiated based on the 5S DNA units. We searched for 5S DNA sequences from known unit classes most closely similar to the long R1 and the short R1. One set with the long R1 unit class contained sequences of the long P1 unit class from Agropyron (P haplome) and from Kengyilia (StYP haplome), long J1 from Thinopyrum (J haplome), whereas the set with the short R1 included the long S1 from Pseudoroegneria (St haplome) and Kengyilia (StYP haplome), the short J1 from Thinopyrum (J haplome) and the short V1 from Dasypyrum (V haplome). Each of the two sets was analyzed separately by maximum likelihood (ML) phylogenetic analysis from which we were able to infer that the 5S DNA units of Secale differentiated in a non-clock fashion and followed the HKY substitution model in the gene tree with the long R1 unit class and the HKY + G in the gene tree with the short R1 unit class. A complementary Bayesian analysis yielded identical tree topologies to the ML ones for each of the two sequence sets. In the tree with the long R1 units the long P1 and long J1 unit classes were closest to the long R1 unit class, whereas in the tree with the short R1 units the long S1 and short J1 unit classes were closest to the short R1 unit class, indicating possibly a close relationship between the St, J and R haplomes.

Three low-molecular-weight (LMW) glutenin-like genes (designated as Ssy1, Ssy2, and Ssy3) from Secale sylvestre Host were isolated and characterized. The three genes consist of a predicted highly conservative signal peptide with 20 amino acids, a short N-terminal region with 13 amino acids, a highly variable repetitive domain and a less variable C-terminal domain. The deduced amino acid sequences of the three genes were the LMW-m type due to a methionine residue at the N-terminus. The phylogenetic analysis indicated that the prolamin genes could be perfectly clustered into five groups, including HMW-GS, LMW-GS, α/β-, γ-, and κ-prolamin. The LMW glutenin-like genes of S. sylvestre were more orthologous with the LMW-GS genes of wheat and B hordein genes of barley, which also had been confirmed by the homology analysis with the LMW-GS of wheat at Glu-A3, Glu-B3, and Glu-D3 loci. These results indicated that a chromosome locus (designated as Glu-R3) might be located on the R genome of S. sylvestre with the functions similar to the Glu-3 locus in wheat and its related species.

Many genome walking methods based on polymerase chain reaction (PCR) are available, including those with and without restriction enzyme modification. Nevertheless, these methods suffer from low reproducibility, inefficiency, and non-specificity. Here, we present a traceable and efficient PCR strategy: randomly broken fragment PCR with 5′ end-directed adaptor for genome walking. The genome is first fragmented randomly. After blunting ends, the fragments are ligated to the 5′ end-directed adaptors. Semi-nested PCR is then performed. Thus, we can obtain an unknown sequence by cloning the fragments of interest, followed by sequencing. This method effectively bypasses the above-mentioned obstacles and offers the advances: 1) genome fragmentation without using restriction enzymes; 2) enhancement of primer specificity and the prevention of self-ligation between the adaptors by employing a 5′ end-directed adaptor. All of the steps in this new method are straightforward, and the unknown sequence can be definitively obtained by merely applying the method once.

One of the key processes of photosynthesis is to control the influx of atmospheric carbon dioxide (CO2). Ion channels fulfill this process by regulating the opening and closing of stomatal pores in plants’ leaves. Inspired by this natural process, we have developed an amidine-modified gas-responsive system that closely mimics stomatal pores: CO2 rather than the variation in the pH value directly modulates the conductance state of the channel. The CO2-activated chemical reaction of amidine groups is reversible and produces an excess surface charge on the pore walls of asymmetric nanochannels, which makes the ions pass preferentially through the nanochannels in one direction relative to the conductance in the other direction, resulting in a significant ion current rectification. Furthermore, the influence of the different molecular conformation of the amidine-containing molecules on the current is investigated and discussed. The conclusive simulation of our system based on the Poisson and Nernst–Planck (PNP) model is also in good agreement with the experimental results. Accordingly, we have successfully mimicked the mechanism of stomatal closure in plants with our gas-activated nanosystem. Inspired by stomatal pores in leaves, researchers in China have demonstrated artificial nanochannels that regulate the intake of CO2. Plants control CO2 intake by closing stomatal pores in leaves when the partial pressure of CO2 becomes high. Now, Jin Zhai and colleagues at Beihang University in Beijing have realized an artificial nanopore system that mimics this effect. Unlike plants, which respond indirectly to CO2 partial pressures via pH changes, the developed system responds directly to CO2 levels through a chemical reaction with amidine-containing molecules on the inner surfaces of the nanopores. This reaction is reversible and generates an excess surface charge on the nanopore walls so that ions pass preferentially in one direction depending on their charge. Simulations based on the Poisson-Nernst-Planck model agreed well with experimental results for this system. Inspired by photosynthesis in nature, an artificial smart system that has similar regulation mechanism is constructed, which is selective to anions after direct reactions with CO2.
