A coordination polymer (CP) containing Co(II) was prepared via combination of 1,2-bis(4-pyridyl)ethylene (bpee) and semi-rigid, ether-based unsymmetrical tetracarboxylic acid 2,3,3′,4′-diphenyl ether tetracarboxylic acid (H4detc) under hydrothermal conditions, [Co2(detc)(bpee)2·5H2O]·1.5H2O. After construction of the postpartum hemorrhage animal model, the plasma was collected and prothrombin time (PT) was measured to evaluate the prothrombin activity of the compound. Then, the ELISA detection kit was used to measure the levels of the factor IX and factor XI in the plasma. After that, the content of inflammatory cytokines tumor necrosis factor-α (TNF-α) and Interleukin-1β (IL-1β) was determined and the prevention activity of the compound on the disseminated intravascular coagulation (DIC) during postpartum hemorrhage was assessed. Exosomes have been documented to function in human diseases, yet their transfer of microRNA (miRNA) in preeclampsia (PE) has seldom been reported. This study intends to discuss the role of miR-133b derived from exosomes in human umbilical cord mesenchymal stem cells (hUC-MSCs) in trophoblast cell development in PE. Placentas from PE patients and normal pregnant women were collected. The hUC-MSCs and their exosomes were obtained and identified. Trophoblast cell HPT-8 and HTR8-S/Vneo were obtained and co-cultured with hUC-MSCs-derived exosomes that had been transfected with different miR-133b plasmids. MiR-133b and glucocorticoid-regulated kinase 1 (SGK1) expression in placental tissues and HPT-8 and HTR8-S/Vneo cells was determined. HTR8-S/Vneo and HPT-8 cell proliferation, cell cycle distribution, apoptosis rate, migration and invasion were detected. MiR-133b was down-regulated and SGK1 was up-regulated in placental tissues of PE patients. MiR-133b expression was inversely related to SGK1 expression in HTR8-S/Vneo and HPT-8 cells co-cultured with hUC-MSC-derived exosomes. Exosomes promoted HTR8-S/Vneo and HPT-8 cell proliferation, migration and invasion abilities, cell cycle entry and inhibited apoptosis. Elevated exosome-derived miR-133b from hUC-MSCs boosted HTR8-S/Vneo and HPT-8 cell proliferation, cell cycle progression, migration and invasion and limited cell apoptosis. MiR-133b targeted SGK1. Collectively, we demonstrate that miR-133b is down-regulated and SGK1 is up-regulated in PE, and miR-133b derived from exosomes in hUM-MSCs facilitates trophoblast cell proliferation, migration and invasion in PE via constraining SGK1. Bleeding is a common problem during resection of a retroperitoneal mass. Massive bleeding may occur in case of injury of an adjacent major vessel or organ. This case report describes a successful anesthetic management of a patient with 10 l of blood loss within three hours surgery. A 44-year-old woman who underwent an operation for resection of a retroperitoneal mass, went to a hypovolemic shock, due to acute life-threatening intra-operative bleeding, and was successfully rescued with a combination of measures, including control of surgical bleeding, supportive treatment with rapid fluid infusion, massive transfusion of blood products and administration of intravenous vasoactive agents for maintaining tissue perfusion and oxygenation, utilizing intraoperative autologous blood salvaged via cell saver, as well as prevention and treatment of complications. The patient received a total of 22 units of Packet Red Blood Cells (PRBCs), 18 units of Fresh Frozen Plasma (FFP), 10 units of cryoprecipitate, 3750 ml of her own salvage blood. Postoperatively, she was transferred to the intensive care unit (ICU) with mechanical ventilator support, where she received another 5.4 units of FFP, 10 units of cryoprecipitate. The patient developed features of early acute lung injury such as fever and hypoxemia, and was managed successfully with mechanical ventilator support for a few days. At a three-month follow-up, the patient was doing very well. This paper explores the pathogenesia, implications, prevention and treatment of the transfusion-associated complications such as acidosis, hypothermia, electrolyte abnormalities, and transfusion-related acute lung injury (TRALI). Particular attention is given to the prevention of secondary coagulopathy of the patient requiring massive blood transfusion. In this article, we incorporated the organic phosphorus group on to a triazolothiadiazole ring to prepare the title compounds fused 1,2,4-triazolo[3,4-b][1,3,4]thiadiazole heterocyclic compounds. Novel pyrazolotriazolopyrimidine and dipyrazolopyrimidine fused tri-heterocyclic compounds were prepared in a convenient method and in good yields. The structures of the title compounds were confirmed by elemental analysis, 1H NMR, 13C NMR, IR and MS. In this article, we introduce organic phosphorus moiety into triazole ring to prepare O-dialkyl-dithiophosphoryl-S-acetoxy triazolo compounds and characterize their structures by elementary analysis and ¹HNMR and IR spectral data. From, the results of biological activity screening, we found that the O-dialkyl-dithiophosphoryl-S-acetoxy triazolo compounds show lower fungicidal activities and their structures need to be optimized for the better. The study on reaction of (1-phenyl-3-methylthio-4-cyanopyrazol-5-yl)iminomethylene ethyl ether with compounds containing amino group was discussed in order to understand its reactivity. AbstractThe preliminary study revealed that the ethyl acetate eluate of Youngia japonica (YJ-E) could inhibit the expression of key proteins of p-p65, p-IκBα, p-IKKα/β, and p-AKT in LPS stimulated BV2 cell. Further phytochemical study led to the isolation of eight compounds from YJ-E, including one new sesquiterpene lactone. Their structures were elucidated by several spectroscopic data, and comparing the NMR data of known compound. In addition, all of the isolates were evaluated for the anti-inflammatory effect.
